Abstract:
Schistosoma mansoni infection leads to chronic inflammation and detrimental granulomatous pathology mediated largely by Th2 immune responses. Dendritic cells (DCs) provide an essential link between the innate and the adaptive immune response, and are critical for Th2 induction during S. mansoni infection. But the function of DCs in this process and the specific mechanisms they employ are not yet fully understood. Indeed, the role for individual DC subsets during the induction of Th2 responses is also unclear. Furthermore, the importance of DCs during the formation of granulomatous legions, and

The outcome of Leishmania infection depends both on host and pathogen factors. Macrophages, the specialized host cell for uptake and intracellular development of Leishmania parasites, play a central role in the control of infection. Underlying their effector and accessory functions is the activation of signalling pathways, which in turn are largely controlled by events of protein phosphorylation. Consequently, the regulation of protein kinase and phosphatase activities results critical for the sequential progression of the signalling cascade, and therefore for the control of antimicrobial and

Although many virulence factors are known to modulate macrophage signalling pathways to favour infection, the delivery mechanisms are largely unknown.

The enormous mortality burden exerted by P. falciparum malaria has evolutionarily selected for red blood cell (RBC) polymorphisms which confer protection against the severe manifestations of this disease. Although the epidemiological protection by these polymorphisms has been well-established for the past half-century, the mechanisms underlying this protection are still being uncovered. Recent studies implicate impaired cytoadherence to microvascular endothelial cells (MVECs) due to reduced surface levels and altered display of Plasmodium falciparum erythrocyte membrane protein 1 (PfEMP1) as a

The development of an efficacious P. falciparum malaria vaccine remains a top priority. Pre-erythrocytic vaccine efforts have traditionally focussed on two well- known antigens, CSP and TRAP, yet thousands of antigens are expressed throughout the liver-stage. The work described in this thesis aimed to assess the ability of other pre-erythrocytic antigens to induce an immune response and provide protective efficacy against transgenic parasites in a mouse model.
