Full textStudy consist of: 50 printed pages, including 9 tables, 28 figures References: twenty eight of references were used. Objective: To identify the variations in circulating creatinine, urea and other substances - alanine aminotransferase, aspartate aminotransferase, and amylase concentration in cats suffering from chronic kidney failure. Methodics: In 2011 – 2013 years on „Kaivana“ small animal clinic a study was conducted on 50 cats who had chronic renal insufficiency. For those cats were made a blood chemistry analysis (creatinine, urea, alanine aminotransferase, aspartate aminotransferase, and amylase) with blood chemistry analyzer Refloctron Plus Roche. WinExcel program performed the statistical analysis, the assessment of feline creatinine stage, age, sex, breed and castration effect on patients with chronic renal failure. We also conducted a correlation analysis features, and one factor analysis of variance. Results, Conclusions: Cats with chronic renal failure suffer relatively rarely - 5%. For the incidence of CRF, animal age, breed, sex has no significant influence. However, the most receptive cats are from 7 to 9 years of life. Creatinine concentration accurately describes the severity of the disease and depends only on kidney damage and other factors it is not affected by fluctuations in volume. Sick-IV stage of a CRF cat was found mainly - 18 (36%). Urea levels directly correlated with the stage of CRF and creatinine. Also Urea is an informative setting... [to full text]

Full textIn the literature, there exists numerous studies of exercise – induced sports hematuria, proteinuria, acute renal failure following a marathon (Poortmans et al., 2001; Ayca et al., 2006; Steward, Pasen, 1980). But studies investigating the effects of exercise on blood indicators of renal function are quiet few. The aim of this study was to investigate the effects of submaximal veloergometric exercise on very important biochemical indicators of renal function – level nitrogen compounds in the blood. In venous blood samples was investigate concentrations of creatinine, urea, total protein and uric acid before and after submaximal (about 50 % VO2max) veloergometric exercise. This nitrogen compounds was studies in three groups of subjects. Methods. The study was performed on 10 trained (1 group), 10 untrained (2 group) and 10 subjects with I° hypertensive status (3 group). The age range was 20,5-21,3 yr, weight – 71,8-77,3 kg, height – 177-180 cm. All subjects volunteered to participate in the study after providing written informed consent. The study was approved in accordance with the Declaration of Helsinki. Blood samples were collected before and after the submaximal veloergometric test into vacationer tubes. Concentrations of creatinine, urea, total protein and uric acid in the serum were determined using Technician Auto Analyzer ADVIA 1650 system. Data from Submaximal ride compared using two-way repeated – measures ANOVA. Dependent t-tests were administered in the event of... [to full text]

Full textThe aim of this work is to explore the effect of ischemia/reperfusion on rat kidney mitochondrial function. The main objectives of this work are to: 1) evaluate the effect of ischemia (20, 40, 60 minutes) in vitro on kidney mitochondrial function; 2) evaluate the effect of ischemia (60 minutes) and reperfusion (30 minutes) in vivo on kidney mitochondrial function; 2) explore the effect of ischemia/reperfusion on kidney mitochondrial respiratory chain complex I activity. We chose two experimental models of ischemia for investigation of its effects on rat kidney mitochondrial functions, i.e. in vitro and in vivo models. The mitochondria were isolated by a differential centrifugation method, while the protein was determined using biuret method. Mitochondrial respiration rate was measured polarographically. Complex I activity was evaluated spectrophotometrically. The results showed that mitochondrial damage become apparent very early - after 20 minutes, i.e. the maximal (ADP- stimulated) mitochondrial respiration rate decreased with both, glutamate/malate and succinate. It is obvious that by increasing the duration of ischemia, the mitochondrial damage increased. The damage of mitochondrial outer membrane increased by increasing the duration of ischemia. We found that both, ischemia (60 minutes) and reperfusion (30 minutes) decreased the mitochondrial State 3 respiration rate. 60 minutes ischemia/30 minutes reperfusion had no effect on mitochondrial inner membrane permeability... Full textThe aim of investigation: to analyse the effect of caffeic acid phenethyl ester (CAPE) on kidney mitochodrial functions. Objectives: to evaluate direct in vitro effect of CAPE on kidney mitochondrial functions; the impact of CAPE on ischemia (20 min) in vitro affected kidney mitochondrial functions; on the mitochondrial respiratory chain complex I activity and on the mitochondria capability to accumulate Ca2+. Methods. Wistar rats were pretreated intraperitoneal with 22 mg/kg and 34 mg/kg of CAPE. Animals were divided into 4 groups: control group, 20 min of ischemia, CAPE 22 mg/kg group and CAPE 34 mg/kg group. Mitochondria were isolated by means of differential centrifugation. The mitochondrial respiration rate while oxidizing complex I and II dependent substrates was registered polarographically with Clark-type electrode. Ca2+ accumulation was measured fluorometrically. Activity of complex I was measured spectrophotometrically. Results: the results shown that CAPE 0,7 - 4,5 µM increases mitochondrial State 2 respiration rate by 15 - 34 % and has no effect on the State 3 respiration rate. Higher concentrations (5,2 - 6,5 µM) decreased mitochondrial State 3 respiration rate by 16% and 43%, respectively. Pretreatment with CAPE (22 mg/kg and 34 mg/kg) increased (by 20%) the ischemia suppressed mitochondrial State 3 respiration rate and respiratory control index. CAPE had no effect on succinate oxydation. Pretreatment with CAPE (22 mg/kg and 34 mg/kg) increased Ca2+... [to full text]

Full textIn this research work we investigated the effect of quercetin and its glycosides (rutin, hyperoside, isoquercitrin) on rat kidney mitochondrial oxidative phosphorylation system. In these studies we used Wistar rats. Eksperimental model – in vitro. Quercetin and its glycosides, i.e. rutin, hyperoside and isoquercitrin (3 - 48 nM) stimulated rat kidney mitochondrial respiration rate in the second metabolic state (uncoupled the mitochondrial respiration). Quercetin and rutin stimulates mitochondrial State 3 respiration rate, other tested compounds had no effect.

Full textThe aim of this study was to evaluate the sensitization with HLA antigens in kidney transplant recipients, who received induction therapy with monoclonal antibodies against IL-2R and in the group of patients, who were only under the triple drug therapy. This study comprises recipients, who received kidney transplant in the year 2000-2005, and who were tested for panel reactive antibody test before and after transplantation (Tx). The total number of 189 kidney transplant recipients takes part in this study. 83 received monoclonal antibodies against IL-2R (basiliximab or daclizumab), others (n=106) – did not. These groups were unequal in comparison to the main factors causing sensitization with HLA antigens. The group of patients, who received induction therapy with monoclonal antibodies had more blood transfuzions (72% vs. 57,3%), and previous transplantations (9,6% vs. 7,5%), in comparison with the other group. Only the number of pregnancies was higher in the group of patients who were only under the triple drug therapy (47,7% vs. 30,8%). Statistical analyses were caried out using chi-square test, differences were considered significant at p<0,05. 58% (110/189) of kidney transplant recipients were unsensitized (PRA 0-10%) before Tx, the rest 42% (79/189) were sensitized, from which 14% (11/189) were highly sensitized (PRA 50-100%). After Tx the number of medium sensitized (PRA 11-50%) kidney transplant recipients, who received induction therapy by monoclonal antibodies... [to full text]

Full textEnd-stage renal disease is highly prevalent worldwide, with more than 1 million patients undergoing renal replacement therapies, which are a life-saving treatment for such patients. Haemodialysis remains the most common treatment modality in all regions of the world. Despite many technical advances in the medical care and in the delivery of dialysis over the past years, mortality and morbidity of dialysis patients remains persistently high and their quality of life is rather poor. Survival rates differ among countries and registries. International differences in the mortality of dialysis patients are probably related to differences in population demographics, renal disease, lifestyle and socioeconomic status. Therefore, it is extremely important to investigate survival of such patients in each country and to identify characteristic risk factors in order to improve outcomes. In most countries, renal registries are responsible for end-stage renal disease data collection, analysis and reporting. There is no renal registry in Lithuania and regular information about patients on dialysis in Lithuania is not available. The aim of our study was to estimate the survival of patients on chronic haemodialysis in Lithuania and to identify associated risk factors. All patients who started chronic haemodialysis in Lithuania between 1 January, 1998, and 31 December, 2005, were enrolled. For the first time in Lithuania, survival of patients on chronic haemodialysis and associated risk factors... [to full text]

Full textThis study presents optimization of sirolimus and tacrolimus measurement in patients blood after renal transplantation by high–performance liquid chromatography with mass–spectrometry detection. In experiments with standart solutions of sirolimus and tacrolimus, using ascomycin as internal standart, chromatographical conditions were tested: type of eluation (isocratical and gradiental), properties of eluents (variable acidity of polar solvent and hydrophobity of nonpolar solvent), linearity of analysis. Patient blood extraction procedure was also tested. Sirolimus concentrations measured by HPLC-MS/MS method were 8-9 times lower as received by MEIA method. Both types of eluation were not sufficient for routine analysis in clinical laboratory, when eluents methanol and 0,2% acetic acid solvent were used. Gradiental eluation seamed to be better, but linearity analysis of sample series showed, that chromatographical system was not well-balanced. Methanol was changed to acetonitrile, but separation and retention time of analytes were even worse. Best results were obtained using mixture of methanol and tetrahydrofuran as nonpolar eluent.
