Full textThis paper presents a new multimedia CD ROM course on Telemetry that has is just being developed for DoD by the Defense Test and Evaluation Professional Institute (DTEPI). The paper will discuss the Commercial Off the Shelf (COTS) software packages that were used to develop the course. It will discuss the methodology used to develop the course and lessons learned in its development. During the presentation of this paper a computer and VGA projector will be used to show some of the material in the course. This is the second CD ROM course developed by DTEPI, the first one was on Time, Space-Position Information (TSPI). The TSPI course has been completed, passed Beta testing at most of the National Ranges, and has been released. About 800 CD ROM disks have been distributed to the Ranges and other qualified users. The Telemetry course will be similarly completed and distributed. The course is intended to be an introduction to the subject of telemetry for use by engineering professionals just entering the workforce, by professionals cross training into T&E, and by others with a need or desire to understand telemetry. The value of developing an interactive course using audio narration, animations, as well as still pictures and video of actual instrumentation and equipment cannot be overemphasized. This multimedia environment makes the explanation of concepts like an optical encoder easily understandable as the student can "see" a simulation of the encoder in operation. The course is designed to be self paced with students controlling their own progress and choosing the topics they want to cover. The student also has the option to print a hard copy of the page narration or read them on screen. The course facilitates the rapid learning of the jargon of telemetry, all the essential acronyms, the way telemetry systems work, what they look like, and many of the limitations of telemetering systems. The course covers the History of Telemetering, Telemetry Subsystems, Range Applications, Telemetry Schemes, Theory of Operation, Telemetry Processing Systems, Testing Telemetry Systems, and other Miscellaneous Topics. It will afford the student a lot of insight into telemetry without the mathematics and detail required of a telemetry design engineer.

Full textWith the current major development in computer science, new methodologies have been used to improve the process of information dissemination. A perspective that cannot be ignored is the question of how to use these technologies to improve the teaching and learning process. One of these methodologies is the multimedia. This study, conducted in the laboratories of the Federal Institute of Informatics Sul de Minas, Campus Machado, MG, and in the José Rosário Vellano University, Campus Alfenas, MG, aims at demonstrating that the use of multimedia techniques, in parallel with all the resources it aggregates, may be a usefull and efficient tool, planned and developed to be a practical source of information collected in a sigle application of study on biodiesel. The Menu strategy, was used for navigation, where one chooses the option of acessing the contents without having to obey any predetermined order. Thus the users will free to access the contents in a dynamic, interactive and pleasant way, according to their interest or need, using audiovisual resources of easy operation and assimilation. The software was available for some students and teachers to evaluate the usability, readability, learning, teaching, multimedia elements, layout and interactivity. In general the software was assessed and satisfactorily met the user s expectations. In a scale of 2-10 points, for all variables studied, the average was above than 8.5 points. The CD-ROM of the biodiesel multimedia is a didactic material developed for educators and high school and college students.
Atualmente, com a grande evolução na informática, novas metodologias vêm sendo utilizadas para melhorar o processo de disseminação das informações. Uma perspectiva que não pode mais ser ignorada é a questão de como utilizar tais tecnologias para melhorar os processos de ensino e aprendizagem. Uma dessas metodologias é o que se chama de multimídia. O presente trabalho pretende demonstrar que a utilização de técnicas multimídia, paralelamente com todos os recursos a ela agregados, poderá ser uma ferramenta útil, eficaz, que foi planejada e desenvolvida para ser uma fonte prática de informações reunidas em uma única aplicação de estudo sobre o tema Biodiesel. O trabalho foi desenvolvido nos Laboratórios de Informática do Instituto Federal Sul de Minas , campus Machado MG, e da Universidade José do Rosário Vellano, campus Alfenas MG. Para navegação foi usada a estratégia de Menu, onde se escolhe a opção de acessar o conteúdo, não obedecendo a nenhuma ordem pré-determinada. Com isso os usuários não se sentirão aprisionados, pois o conteúdo é acessado de forma dinâmica, interativa e prazerosa, de acordo com o seu interesse ou necessidade, utilizando-se de recursos audiovisuais, de fácil operação e assimilação. O software foi disponibilizado para alguns alunos e professores para avaliação quanto à usabilidade, legibilidade, aprendizado, didática, elementos multimídia, leiaute e interatividade. De maneira geral, o software foi avaliado satisfatoriamente e atendeu às expectativas dos usuários. Em uma escala de notas de 2 a 10 pontos, para todas as variáveis estudadas, a média foi superior a 8,5 pontos. O CD-ROM do multimídia Biodiesel é material didático para educadores e alunos do ensino médio e superior.

Full textCD1e belongs to the family of antigenic presenting molecules CD1 but CD1e is very particular. Indeed, CD1e remains surprisingly intracellular, accumulating mainly in the Golgi compartments of immature dendritic cells (DCs) into a complete transmembrane associated form and in the late endosomes of mature DCs. In the latter compartments, CD1e is cleaved and becomes soluble due to a cleavage between the luminal and transmembrane domains. We studied the distribution of CD1e in immature and maturing DCs. In immature DCs, CD1e accumulates in Golgi compartments, is not expressed on the plasma membrane but reaches CD1b+ late multilamellar compartments. During maturation, CD1e molecules accumulate in mature endosomes CD1b+. Its cytoplasmic domain does not contain known targeting motifs. CD1e reaches late endosomes in AP-1 and AP-3 deficient cell lines. Interestingly, the cleavage of CD1e occurs in AP-3-, but not in AP-1- cells. Deletion and substitution mutants were engineered and expressed in an HLA-DR+ melanoma cell line. Experiments showed that different parts of the cytoplasmic tail control the localization of CD1e in the golgi compartments and its absence on the cell surface, facilitating the routing of CD1e to late endosomes where it is cleaved. The replacement of the cytoplasmic domain of CD1e by that of CD1b blocked its cleavage. Thus, CD1e reaches late endosomes using a cellular pathway not shared by CD1b. Finally, we show that CD1e contains a propeptide which is cleaved in endosomes. Our data strongly suggest that the function of the intracellular transport of CD1e is the generation of soluble endosomal molecules, which might play a role in the presentation of antigens by other CD1 molecules transiting in CD1e+ endosomes

Full textSeptins comprise a conserved protein family that binds guanidine nucleotide and forms heterofilaments. In structural terms they have a common organization: a central GTPase domain, a N-terminal domain and a C-terminal domain, this last one is predicted to form coiled coil structures. Currently, the human septin heterocomplex best characterized (SEPT2/SEPT6/SEPT7) reveals the importance of the GTPase domain in filament assembly, however the absence of electron density for the C-terminal domains makes its function still unknown. Studies with mammals septins, and of others organisms like C. elegans and S. cerevisea suggests that some septins groups (e.g. II e IV in mammals) interact via its C-terminal domains and this could act in a determinative way to correct filament assembly. In this way, this project aimed to study the homo/heterotypical affinity for the C-terminal domains of human septins belonging to groups II (SEPT6C/8C/10C/11C) e IV (SEPT7C), investigating whether this domain contributes with the preference of septins to interact with proteins of different groups during assembly of the heterofilament. The C-terminal domains were expressed in E. coli and purificated. It was carried out studies using analytical ultracentrifugation and circular dichroism spectropolarimetry tecniques which allowed identification of major affinity and stability in the heterotypical association compared to homotypical. It was measured apparent dissociation constants for homodimers of low µM range while for heterodimers our group\'s data shows dissociation constants in the nM range. To understand at atomic level the factors responsible for this significant preference in the C-terminal domains interaction between groups II and IV was performed molecular modelling studies and analysis of the primary sequence. These analysis suggests the presence of a high number of charged residues in position a of the coiled coil as responsible for selectivity. Consequently, the heterodimer would be therefore favoured because of the minor repulsive effect coming from the staggered of charged residues in a. Thus, these results indicate the crucial or cooperative action of C-terminal domains in preferential organization of septins during filament assembly, favouring the NC interface between septins of groups II and IV.

Full textCell membranes are complex systems composed of variety of lipids that interacts with proteins to trigger cellular function. The delivery of these lipids to the right compartment is crucial for cells to work efficiently. The coat protein (COP) complex vesicles are involved in lipids traffic in the early stages of the secretory pathway. Recently, a highly specific interaction has been found between the transmembrane domain of p24 protein (p24TMD) abundant in COPI membrane and sphingomyelin C18:0. As such highly specific interaction have been reported for protein-protein and protein-nucleic acid interactions to be involved in regulation of cell functions, we decide to investigate this specific interaction. Full textOral infection with Toxoplasma gondii (T. Gondii) triggers a robust intestinal immune response associated with a number of well-described immunologic events. The immune response involves inflammatory and regulatory components, which control both the parasite dissemination and the inflammatory process. In synergy with intestinal epithelial cells, lamina proprié CD4+ T cells are an important componant of this intestinal process. CD40-CD154 interaction and IL-12 expression appear to be essential for the induction of this intestinal immune response. The inflammatory process limits the parasite proliferation and leads to cyst formation into the host tissues. In order to survive against its own inflammatory response, the host generates several intestinal immunoregulatory events, which control the intestinal homostasis. TGF-β-producing intraepithelial lymphocytes modulate the inflammation by controlling both la mina propria CD4+ T cell and epithelial cell activation. At day 7 after oral infection with T. Gondii cysts, unmanipulated C57BL/6 mice fail to regulate this inflammatory process and evelop an acute lethal ileitis. It is the collapse of gut homeostasis that is also the major cause of the chronic intestinal inflammatory bowel disease (IBD) in human. One hypothesis for the pathogenesis of IBD is thas disease is due to an abnormal response of the gut immune system to some antigens from the normal mucosal microflora. Thus, this pattern-driven ileitis model has many histological and immunological characteristics of human Crohn's disease and may be particularly useful for a better understanding of the immunopathogenesis of this human pathology

Full textIonic liquids (ILs) are salts that are liquid at temperatures smaller than 100 ° C and are gaining prominence in the so-called green chemistry, promising: replace harmful solvents to the environment, improve electronic components, and favor biocatalysis, among others. Its high stability and low toxicity are often asserted; nevertheless, they are ascribed to ILs due to its small volatility. With the aim of improving the understanding of the interaction of ILs with biological relevant systems, we conducted a systematic study of the interaction of three different ionic liquids of the same polar head and different paraffinic tails ([C10mim][Cl], [C12mim][Cl] and [C14mim][Cl]) with three different model proteins, through the techniques of optical absorption, fluorescence, circular dicrhoism (CD) and small angle X-ray scattering (SAXS). To do so, we use BSA and HSA proteins (Bovine Serum Albumin and the Human Serum Albumin, respectively) and lysozyme. We observed fluorescence quenching, of all studied proteins, where the decrease in the fluorescence was (for BSA, HAS and lysozyme, respectively): (55 ± 3)%, (16.1 ± 0.8)% to (4.1 ± 0.2 )% in the presence of 0.6mm [C14mim][Cl], (38 ± 2)%, (13.2 ± 0.7)% to (0.6 ± 0.1)% in the presence of 0.6mm [C12mim][Cl] and ( 11.0 ± 0.5)% (9.2 ± 0.5)% and (0.0 ± 0.1)% in the presence of 0.6mm [C10mim][Cl]. UV-vis absorbance spectra and fluorescence indicate all systems in a contact interaction between proteins and ionic liquids. We also note the shift of the fluorescent peak of BSA and HSA proteins for shorter wavelengths (blue-shift), as the IL content was increased. The maximum shift () achieved corresponded to (21 ± 1) nm for both albumins, whereas no significant displacement was observed for lysozyme. The blue-shift can be explained by the approach of carbon chains and formation of hydrogen bonds in the vicinity of tryptophan. SAXS data indicate an increasing in the proteins radius of gyration value as ILs was added in the solution. The turning radius of BSA, HSA and lysozyme in the absence of IL are (29 ± 1) Å, (30 ± 1) Å and (15 ± 1) Å, respectively, and go to (46 ± 1) Å, ( 44 ± 1) Å and (20 ± 1) Å, respectively, in the presence of 0.6mm [C14mim][Cl]. The SAXS curves also show evidence of the formation of micellar structures from a given concentration. Besides the change in its tertiary structure, the CD data indicates a slight loss of secondary structure of both albumins (BSA and HSA), from 80 to 65% of -helix in the absence and presence of 0.6mm [C14mim][Cl], respectively. We suggest that the interactions of the protein with the ionic liquid, although initially driven by electrostatic forces, have a major factor hydrophobic effect and thus the higher the carbon chain of greater IL is its interaction with the protein. This interaction causes unfolding of the protein and formation of a micellar structures at high concentrations of IL. We believe this work provides new information about the interaction of ILs with model proteins, indicating its ability to alter the conformation of the same.
