Abstract:
The virus-inducible Type I interferon (IFN) genes serve as relevant models to examine the nature of cis-acting sequences and trans-acting protein factors involved in transcription control. The alpha and beta interferon genes are activated differentially depending on the nature of the inducer and the specific cellular background. A transient expression system was developed to analyze the activation of the interferon promoters in a homologous human cellular background. In epithelial cells, the IFN-$ beta$ but not the IFN-$ alpha$1 promoter was virus responsive with induction kinetics similar to

<p>Flaviviruses, family flaviviridae, are often associated with severe diseases and many of the viruses have been shown to affect the immune response. Interferons confer important ways of defending the host against viral infections because they provide a link between the innate and the adaptive immunity. Langat virus (LGTV), belonging to the Tick-Borne Encephalitis (TBE) complex of viruses, has recently been shown to provide interactions with interferon receptors and inhibit the interferon-mediated response. Similarly, it has been demonstrated that the TBE virus NS5 protein affects type 1 inte

Abstract:
BACKGROUND AND AIMS: Congenital Hepatic Fibrosis (CHF) is caused by mutations in PKHD1, a gene encoding for fibrocystin, a protein of unknown function, expressed in cholangiocyte cilia and centromers. In CHF, biliary dysgenesis is accompanied by severe progressive portal fibrosis and portal hypertension. The mechanisms responsible for portal fibrosis in CHF are unclear. The αvβ6 integrin mediates local activation of TGFβ1 and is expressed by reactive cholangiocytes during cholestasis. To understand the mechanisms of fibrosis in CHF we studied the expression of αvβ6 integrin and its regulation

Abstract:
Type I interferons (IFNs) function as the first line of defense against viral infections by modulating numerous biological processes to establish an antiviral state and influencing the activation of various immune cells. During influenza A infection, the NS1 encoded by the virus genome disrupts many cellular processes to block type I IFN responses. We show that expression of H5N1 NS1 in HeLa cells reduces IFN-inducible activation of STAT proteins and its subsequent binding to DNA complexes. Subsequent analysis suggests NS1 blocks IFN signaling by inhibiting expression of type I IFN receptor su
