This paper originates from an address at the 8th International Symposium on Nitrogen Fixation with Non-Legumes, Sydney, NSW, December 2000 Detailed studies in field experiments have shown repeatedly that the transfer of 15N2 fixed by diazotrophic bacteria to wheat tissue is minimal. Here, a simple and convenient laboratory co-culture model was designed to assess important features of the association between Azospirillum brasilense and wheat, such as the rate of nitrogen fixation (acetylene reduction), ammonia excretion from the bacterium and the transfer of newly fixed 15N2 from the associative diazotroph to the shoot tissue of wheat plants. After 70 h, in this model, insignificant amounts of newly fixed N2 were transferred from an ammonia-excreting strain of A. brasilense to the shoot tissue of wheat. However, when malate was added to the co-culture the 15N enrichment of the shoot tissue increased 48-fold, indicating that 20% of shoot N had been derived from N2 fixation. Thus, the inability of the host plant to release carbon in the rhizosphere is a significant constraint in the development of associative N2-fixing systems. These specific results suggest that wheat plants with an increased release of photosynthate to the rhizosphere should be a priority for the future development of broad-acre agricultural systems that are more self-sufficient for nitrogen nutrition. The simplicity of the model for assessing the critical parameters of associative 15N2 fixation may allow large-scale surveys of plant–bacterial interactions to be conducted and a selection of improved associations for further study.

This paper originates from an address at the 8th International Symposium on Nitrogen Fixation with Non-Legumes, Sydney, NSW, December 2000 Ammonium uptake by cells has been studied for more than a century, but only recently a family of ammonium transporters (Mep/Amt) with 10–12 transmembrane domains has been defined. These proteins are probably ubiquitous, since homologues have been found in the major kingdoms of living organisms. Plants as well as yeast and some archaebacteria have multiple Mep/Amt paralogues, which can be distinguished by their affinity for ammonium and the ammonium analogue methylammonium. Most ammonium transporters are induced in nitrogen-starving conditions, both in prokaryotes and plants. In Saccharomyces cerevisiae, Escherichia coli and Azospirillum brasilense Mep/Amt proteins where shown to be necessary for growth when the external concentration of the diffusive ammonium form (NH3) becomes limiting. Ammonium transporters also play an important role in pseudohyphal differentiation in yeast and efficient symbiotic interaction between Rhizobium etli and its host plant. In most bacteria, NH4+ transport appears to be a uniport mechanism driven by the membrane potential, but, depending on the organism, a different mode of ammonium uptake may be operating. Current knowledge offers the basis to investigate further the physiological role of ammonium transporters in the natural habitat of organisms and their importance in plant–bacteria interactions.

Abstract By 1900, research identified urea, ammonia, and protein as the basic components of N metabolism in ruminants. Energy and protein metabolic interactions were outlined, amino acids were individually recognized as constituents of protein molecules, and the concept of enzyme-catalyzed reactions was established. Research stations were publishing research results and feeding recommendations. By 2000, the National Research Council built on the work of Henry and Morrison to create and revise publications of nutrient requirements for various classes of ruminants. Use of surgically altered animals, isotopically labelled molecules, and standardized laboratory analyses created quantitative information on ingestion, digestion, absorption, and metabolism of N-containing compounds. Protein composition and structure as well as the role of protein turnover to determine the concentration of cellular proteins were elucidated. Hypothesis-driven, statistically valid experimental designs created quantitative information on protein and amino acid requirements. Methionine was identified as the likely first-limiting amino acid for several ruminant species and production states. The internet and the advent of international symposia provided links among researchers around the globe. Today, multidisciplinary research teams are needed to enlarge the data base, to describe proteomics and metabolomics, and to integrate “big data” into insightful and useful models. Those models currently use or estimate rates and daily amounts of ruminal (in)degradability of dietary protein, ammonia production, urea recycling, microbial protein synthesis, postruminal protein digestion, metabolic fecal nitrogen, and amino acid absorption and metabolism. They predict use of metabolizable amino acids for maintenance, milk yield, growth, and fetal growth. Directions of future research include integration of functional roles of amino acids into recommended dietary supply and requirements, and focus on solutions to challenges presented by climate change, (in)sustainability of livestock production, and changes in public perception of humans’ use of ruminants and other animals.

Die heutige Welt der Menschen wird durch große Dynamik geprägt. Eine Vielzahl verschiedener Prozesse entfaltet sich parallel und teilweise auf unsichtbare Weise miteinander verbunden. Nimmt man z.B. den Prozess der Globalisierung: Hier erleben wir ein exponentielles Wachstum der internationalen Verknüpfungen von der Ebene einzelner Menschen und bis zur Ebene der Kulturen. Solche Verknüpfungen führen uns zum Begriff der Komplexität. Diese wird oft als Produkt der Anzahl der Elemente eines Systems mal Umfang der Verknüpfungen im System verstanden. In anderen Worten, die Welt wird zunehmend komplexer, denn die Verknüpfungen nehmen zu. Komplexität wiederum ist ein Begriff für etwas unverstandenes, unkontrollierbares, etwas unbestimmtes. Genau wie bei einem Menschen: Aus einer Zelle wächst ein Mensch, dessen Verhalten wir im Detail nur schwer vorhersagen können. Immerhin besitzt sein Gehirn 1011 Elemente (Zellen). Wenn also diese dynamischen sozialen Prozesse zu höherer Komplexität führen, müssen wir auch mehr Unbestimmtheit erwarten. Es bleibt zu Hoffen, dass die Unbestimmtheit nicht existenzielle Grundlagen betrifft. Was die Komplexität der Technik angeht, so versucht man hier im Gegensatz zu den gesellschaftlichen Unsicherheiten die Unsicherheiten zu erfassen und gezielt mit ihnen umzugehen. Das gilt für alle Bereiche, ob nun Naturgefahrenmanagement, beim Bau und Betrieb von Kernkraftwerken, im Bauwesen oder in der Schifffahrt. Und so verschieden diese Fachgebiete auch scheinen mögen, die an diesem Symposium teilnehmen: Sie haben erkannt, das verantwortungsvoller Umgang mit Technik einer Berücksichtigung der Unbestimmtheit bedarf. Soweit sind wir in gesellschaftlichen Prozessen noch nicht. Wünschenswert wäre, dass in einigen Jahren nicht nur Bauingenieure, Maschinenbauer, Mathematiker oder Schiffsbauer an einem solchen Probabilistik- Symposium teilnehmen, sondern auch Soziologen, Politiker oder Manager... PREFACE: The world today is shaped by high dynamics. Multitude of processes evolves parallel and partly connected invisible. For example, the globalisation is such a process. Here one can observe the exponential growing of connections form the level of single humans to the level of cultures. Such connections guide as to the term complexity. Complexity is often understood as product of the number of elements and the amount of connections in the system. In other words, the world is going more complex, if the connections increase. Complexity itself is a term for a system, which is not fully understood, which is partly uncontrollable and indeterminated: exactly as humans. Growing from a single cell, the humans will show latter a behaviour, which we can not predict in detail. After all, the human brain consists of 1011 elements (cells). If the social dynamical processes yield to more complexity, we have to accept more indetermination. Well, one has to hope, that such an indetermination does not affect the basic of human existence. If we look at the field of technology, we can detect, that here indetermination or uncertainty is often be dealt with explicitly. This is valid for natural risk management, for nuclear engineering, civil engineering or for the design of ships. And so different the fields are which contribute to this symposium for all is valid: People working in this field have realised, that a responsible usage of technology requires consideration of indetermination and uncertainty. This level is not yet reached in the social sciences. It is the wish of the organisers of this symposium, that not only civil engineers, mechanical engineers, mathematicians, ship builders take part in this symposium, but also sociologists, managers and even politicians. Therefore there is still a great opportunity to grow for this symposium. Indetermination does not have to be negative: it can also be seen as chance.

Variations in drug metabolism may alter drug efficacy and cause toxicity; better understanding of the mechanisms and risks shall help to practice precision medicine. At the 21st International Symposium on Microsomes and Drug Oxidations held in Davis, California, USA, in October 2-6, 2016, a number of speakers reported some new findings and ongoing studies on the regulation mechanisms behind variable drug metabolism and toxicity, and discussed potential implications to personalized medications. A considerably insightful overview was provided on genetic and epigenetic regulation of gene expression involved in drug absorption, distribution, metabolism, and excretion (ADME) and drug response. Altered drug metabolism and disposition as well as molecular mechanisms among diseased and special populations were presented. In addition, the roles of gut microbiota in drug metabolism and toxicology as well as long non-coding RNAs in liver functions and diseases were discussed. These findings may offer new insights into improved understanding of ADME regulatory mechanisms and advance drug metabolism research.
