Abstract:
Background Intramuscular rocuronium, in doses of 1,000 microg/kg in infants and 1,800 microg/kg in children, produces complete twitch depression in 5-6 min. To determine the rate and extent of absorption of rocuronium after intramuscular administration, blood was sampled at various intervals after rocuronium administration by both intramuscular and intravenous routes to determine plasma concentrations (Cp) of rocuronium. Methods Twenty-nine pediatric patients ages 3 months to 5 yr were anesthetized with N2O and halothane. The trachea was intubated, ventilation was controlled, and adductor pollicis twitch tension was measured. When anesthetic conditions were stable, rocuronium (1,000 microg/kg for infants and 1,800 microg/kg for children) was injected either intramuscularly (in the deltoid muscle) or intravenously. Four venous plasma samples were obtained from each child 2-240 min after rocuronium administration. A mixed-effects population pharmacokinetic analysis was applied to these values to determine bioavailability, absorption rate constant, and time to peak Cp with intramuscular administration. Results With intramuscular administration, rocuronium's bioavailability averaged 82.6% and its absorption rate constant was 0.105 min(-1). Simulation indicated that Cp peaked 13 min after rocuronium was given intramuscularly, and that 30 min after intramuscular administration, less than 4% of the administered dose remained to be absorbed from the intramuscular depot. Conclusions After rocuronium is administered into the deltoid muscle, plasma concentrations peak at 13 min, and approximately 80% of the administered drug is absorbed systemically.

Abstract:
The objectives of this study were twofold. Firstly, the present study was designed to examine susceptibility of the corpus luteum (CL) in early diestrus in jennies; and secondly, to investigate the effect of two commonly used hormonal agents in horses on the induction of ovulation in jennies. The oestrus cycles of eleven jennies were monitored by ultrasound every day. When the dominant follicle reached a diameter of 30 mm, the jennies were treated by intramuscular administration of gonadotropin-releasing hormone agonist lecirelin (GnRH, 50 µg pro toto) in the first oestrus cycle, and human chorionic gonadotropin (hCG, 1500 IU pro toto) intramuscularly in the second oestrus cycle. Prostaglandin F2α analogue cloprostenolum (PGF2α, 0.125 mg pro toto) was administered intramuscularly 2 days after the first ovulation and the interovulatory interval was monitored. This study showed that intramuscular administration of 50 µg of GnRH agonist lecirelin resulted in ovulation within 48 h in 73% of treated jennies. Intramuscular administration 1500 IU of hCG was found to be poorly effective to induce ovulation, with 36% of animals ovulating within 48 h. Intramuscular administration of PGF2α analogue cloprostenol 2 days after ovulation was unsuccessful in attempting to shorten the interovulatory interval in donkeys.

A cirrose hepática é uma das principais causas de sangramento espontâneo dado o desequilíbrio da hemostasia e hemodinâmica dos portadores da doença. Hematomas intramusculares espontâneos principalmente na região do dorso não são uma complicação comum da cirrose, portanto, apresentam prognóstico reservado. Além disso, a presença de hemotórax em pacientes cirróticos é também justificada pela coagulopatia da doença, sendo mais rara que o hematoma intramuscular. Neste relato será abordado o caso de um paciente com quadro de dor lombar e escapular direita que evoluiu com astenia, hiporexia e icterícia. Foi internado, colhido exames laboratoriais e feitos exames complementares, sendo diagnosticado com hematoma intramuscular espontâneo em dorso e hemotórax espontâneo, optou-se pela conduta conservadora. Os registros de hematomas intramusculares e hemotórax espontâneos são raros e, por esse motivo, o manejo de cada caso é individualizado. Com isso, é importante considerar o hematoma intramuscular como uma complicação grave associada a Cirrose Hepática Alcoólica.

Summary Background: the presence of intramuscular pin bones hinders the production and commercialization of fish fillet products; however, application of physical processes, such as thermal treatments, offers alternatives for the degradation of said bones. Objective: the present study aimed to conduct a microstructural and physicochemical analysis of Bocachico intramuscular pin bones subjected to a thermal treatment. Methods: collagen extracted from intramuscular pin bones of Bocachico fillets was analyzed using SDS-polyacrylamide gel electrophoresis and viscosity. Pin bones were subjected to 1.5, 2, and 3 minutes heating time and analyzed using electron microscopy and cutting force. Results: intramuscular pin bones contain type I collagen. Three-minute thermal treatment degraded collagen components present in the internal pin bone structure, coinciding with the lowest values of the cutting force test. Conclusions: according to our results, collagen degradation initiates in the internal structure of intramuscular pin bones and moves towards the external layer which does not show the effects of thermal treatment. Resumen Antecedentes: la presencia de espinas intramusculares en filetes de peces impide la obtención y posterior comercialización de estos productos, motivo por el cual la aplicación de procesos físicos antes del tratamiento térmico podría ser una alternativa en la degradación de estas espinas. Objetivo: el alcance del presente estudio fue realizar análisis fisicoquímico y microestructural de espinas intramusculares de Bocachico sometidas a proceso térmico. Métodos: las espinas intramusculares de filetes de Bocachico fueron analizadas a través de electroforesis en gel de SDS-poliacrilamida (SDS-PAGE), temperatura de desnaturalización del colágeno, microscopia electrónica de trasmisión y fuerza de corte. Fueron utilizados tres tiempos de duración del tratamiento térmico (1,5, 2 y 3 min). Resultados: el colágeno presente en espinas intramusculares es tipo l. El tratamiento térmico durante tres minutos degrada los componentes del colágeno en las estructuras internas de la espina, coincidiendo con los valores menores para la prueba de fuerza de corte. Conclusiones: los resultados permiten concluir que la degradación del colágeno es iniciada desde el interior de la estructura de la espina intramuscular hacia la capa externa, sin que esta capa muestre efecto del tratamiento térmico. Resumo Antecedentes: a presença de espinhos intramusculares impede a obtenção de filetes de peixe e posterior comercialização destes produtos, razão pela qual a aplicação de processos físicos antes do tratamento térmico pode ser uma alternativa na degradação desses espinhos. Objetivo: realizar analise físico-químico e microestrutural em espinhos intramusculares do peixe Bocachico submetidos a processo térmico. Métodos: os espinhos intramusculares de filetes do Bocachico foram analisados por eletroforese em gel de SDS poliacrilamida (SDS-PAGE), temperatura de desnaturação do colágeno, microscopia electrónica de transmissão e força de corte. Foram utilizados três tempos de duração do tratamento térmico (1,5, 2 e 3 min). Resultados: o colágeno presente em espinhos intramusculares é do tipo l. O tratamento térmico por três minutos degrada os componentes do colágeno nas estruturas internas do espinho, coincidindo com os menores valores para o teste de resistência ao corte. Conclusões: os resultados mostram que a degradação do colágeno é iniciado desde o interior do espinho intramuscular até a capa externa, sem que essa capa externa seja afetada pelo tratamento térmico. Abstract:
ABSTRACTDeveloping an effective anthrax vaccine that can induce a rapid and sustained immune response is a priority for the prevention of bioterrorism-associated anthrax infection. Here, we developed a recombinant replication-deficient adenovirus serotype 5-based vaccine expressing the humanized protective antigen (Ad5-PAopt). A single intramuscular injection of Ad5-PAopt resulted in rapid and robust humoral and cellular immune responses in Fisher 344 rats. Animals intramuscularly inoculated with a single dose of 108infectious units of Ad5-PAopt achieved 100% protection from challenge with 10 times the 50% lethal dose (LD50) of anthrax lethal toxin 7 days after vaccination. Although preexisting intranasally induced immunity to Ad5 slightly weakened the humoral and cellular immune responses to Ad5-PAopt via intramuscular inoculation, 100% protection was achieved 15 days after vaccination in Fisher 344 rats. The protective efficacy conferred by intramuscular vaccination in the presence of preexisting intranasally induced immunity was significantly better than that of intranasal delivery of Ad5-PAopt and intramuscular injection with recombinant PA and aluminum adjuvant without preexisting immunity. As natural Ad5 infection often occurs via the mucosal route, the work here largely illuminates that intramuscular inoculation with Ad5-PAopt can overcome the negative effects of immunity induced by prior adenovirus infection and represents an efficient approach for protecting against emerging anthrax.

Abstract:
Background A nondepolarizing muscle relaxant with an onset and offset profile similar to succinylcholine is desirable for pediatric anesthesia. The onset and offset of rapacuronium are rapid in children. In the current study, the authors determined its pharmacokinetic characteristics in children. In addition to administering rapacuronium by the usual intravenous route, the authors also gave rapacuronium intramuscularly to determine uptake characteristics and bioavailability. Methods Forty unpremedicated patients aged 2 months to 3 yr were anesthetized with halothane, 0.82-1.0% end-tidal concentration. When anesthetic conditions were stable, rapacuronium was injected either into a peripheral vein (2 mg/kg for infants, 3 mg/kg for children) or a deltoid muscle (2.8 mg/kg for infants, 4.8 mg/kg for children). Four venous plasma samples were obtained from each subject 2-240 min after rapacuronium administration. A mixed-effects population pharmacokinetic analysis was applied to these values to determine bioavailability, absorption rate constant, and time to peak plasma concentration with intramuscular administration. Results Plasma clearance was 4.77 ml x kg(-1) x min(-1) + 8.48 ml/min. Intramuscular bioavailability averaged 56%. Absorption from the intramuscular depot had two rate constants: 0.0491 min(-1) (72.4% of absorbed drug) and 0.0110 min(-1) (27.6% of the absorbed drug). Simulation indicated that plasma concentration peaks 4.0 and 5.0 min after intramuscular rapacuronium in infants and children, respectively, and that, at 30 min, less than 25% of the administered dose remains to be absorbed from the intramuscular depot. Conclusions In infants and children, rapacuronium's clearance and steady state distribution volume are less than in adults. After intramuscular administration, bioavailability is 56%, and plasma rapacuronium concentrations peak within 4 or 5 min.

Abstract:
We described 59-year-old men with history of secondary/tertiary hypogonadism, without primary hypothyroidism, elevated prolactin or hemochromatosis with structurally intact hypothalamo- hypophyseal (HT-HP) axis who was diagnosed in 2017. He was treated by different physicians with Testosterone intramuscular injections, hCG intramuscular injections and combination of hCG and FSH intramuscular injections. The patients wanted to have another child and this is why his therapy was changed from testosterone to hCG and later on to hCG and FSH injections three times a week by his urologist. Although his testosterone levels increased his semen analysis showed oligospermia. The patient was not very compliant with his Testosterone, hCG/hCG and FSH injections, because of the discomfort of intramuscular injections. This is why we started the patient from the therapy with hCG and FSH intramuscularly to Clomiphene citrate (CC) 25 mg per mouth every other day in 2021. After eight months therapy with CC the patient semen analysis normalized, Total and Free testosterone levels normalized, LH and FSH increased, his libido, morning erections improved and his erectile dysfunction resolved. Most importantly for the patient his wife become pregnant. With this case we emphasize the role of Clomiphene citrate in treating male hypogonadism if the patients desire fertility.

Abstract:
Introduction and aim. Osteoarthritis (OA) is generally a progressive disease that affects synovial joints, resulting in abnormalities to articular cartilage subchondral bone, synovium, and adjacent soft tissues. The purpose of this work was to investigate the specific activity of chondroitin sulfate (CS) in intra-articular and intramuscular administration to laboratory rabbits in experimental OA. Material and methods. OA was induced in rabbits by a single injection of mono-iodoacetate in knee joint. CS was administered intra-articularly and intramuscularly. The analysis of biochemical markers and macroscopic assessment of rabbit knee joints was performed. Results. Intramuscular and intra-articular injection of CS reduces the intensity of the degenerative-dystrophic process due to the impact on inflammatory and the activation of anabolic mechanisms. Intra-articular administration of CS leads to a greater increase in the level of factors of bone and cartilage formation and a greater decrease in the levels of factors of the acute phase of inflammation and factors that destroy the cartilage matrix. Conclusion. Intramuscular administration of CS revealed a lower intensity of destructive changes in the cartilaginous surface of the knee joint, and intramuscular – the absence of cartilage destruction and defects of the cartilaginous surface, which indicates the peculiarity of the topical effect of the CS.

En esta tesis se aborda el estudio de la grasa intramuscular como característica determinante de la calidad de la carne para ser utilizado en programas genéticos. El conejo se plantea no sólo por su interés como especie ganadera sino como modelo en otras especies. Este estudio se divide en tres experimentos, cada uno de ellos representa cada uno de los tres objetivos específicos de esta tesis: 1. Estudiar la variabilidad genética entre líneas de conejo de factores ligados directamente a la deposición de grasa tanto en músculo como en tejido adiposo, como son las enzimas que participan en la síntesis y degradación de la grasa y en la composición en ácidos grasos. 2. Puesta a punto de una calibración NIRS para estimar grasa intramuscular y la evaluación de su uso en programas de selección en conejo. 3. Selección divergente por grasa intramuscular. Estudio de las posibilidades de éxito de la selección por grasa intramuscular y conocer las relaciones genéticas entre caracteres examinando la respuesta correlacionada. Supone el inicio de una nueva línea de investigación en metabolismo lipídico, que puede servir tanto a la producción de conejo como animal de carne como al uso del conejo como modelo experimental. Es la primera vez que se propone un experimento de selección divergente por grasa intramuscular. Este experimento permitirá conocer mejor las relaciones genéticas entre la grasa intramuscular, la grasa de la canal, así como las relaciones con otros caracteres productivos. Abstract:
Background: Myofascial pain is a common musculoskeletal disorder characterized by muscles in a contracted state with increased tone and stiffness. It is one of the most common chronic pain syndromes that lead to disability and lower quality of life, creating a significant public health and economic burden. Intramuscular stimulation (IMS) is a dry needling technique that targets specifically chronic myofascial pain and is utilized in multidisciplinary pain centers around the world. Despite its wide use, the effect of IMS has been poorly studied. Methods: We conducted two chart reviews to generate information regarding IMS effectiveness and the feasibility of conducting a randomized clinical trial (RCT). Success rate of pain improvement (at least 1 unit reduction on pain score) was documented in percentage. In addition, an inter-rater reliability test was conducted to assess the consistency among IMS practitioners in examining patients. Intra-class correlation coefficient (ICC) and multiple-raters kappa were used to assess the agreement between practitioners in identifying number of taut bands and identifying taut bands in each muscle respectively. Results: From the two chart reviews, we found that a majority of patients (nearly 30%) had their pain for 10 or more years. Most of them had chronic low back pain. The percentage of success was at least 34% (95% CI: 25%, 43%) and 30% (95% CI: 22%, 38%) for worst and average pain (last week) respectively. For inter-rater reliability test, ICC values ranged from 0.64 to 0.77 (substantial agreement) but kappa values of identifying tender taut band in each muscle were ranged from -0.05 (poor agreement) to 0.46 (moderate agreement). Conclusion: Chart review showed positive results regarding short-term effect of IMS in relieving chronic pain and improving pain consequences. Given the difficulty in treating chronic pain, it is worth conducting a sham controlled RCT to further evaluate the effect of IMS. Population characteristics and success rate of pain improvement will be used in justifying the type and size of sample in the design of a RCT. Good consistency among practitioners in treating patients has to be achieved before conducting the RCT.

Aquesta tesi doctoral forma part d'una línia de recerca sobre la millora genètica de la qualitat de la carn en porcí. Un dels recents objectius inclòs en algunes línies paternes seleccionades per mercats de qualitat és aconseguir un nivell òptim de greix intramuscular i de composició en àcids grassos sense penalitzar el rendiment magre. Aquesta tesi està formada per quatre estudis realitzats en una línia pura de porcs Duroc destinats a la producció de càrnics d'alta qualitat. Els estudis es van dissenyar amb l'objectiu d'entendre millor la variabilitat genètica subjactent al contingut de greix i a la composició en àcids grassos i d’identificar marcadors potencials a la selecció. El primer estudi va examinar els paràmetres genètics de la ruta de l'àcid linoleic (C18:2) a l'àcid araquidònic (C20:4). En particular, es va demostrar que s’espera que la selecció pel valor absolut de C18:2 proporcioni una resposta a la selecció similar a la selecció pel greix intramuscular a greix dorsal restringit. Aquests resultats van conduir a investigar el gen de la desaturasa-2 dels àcids grassos (FADS2), com a gen candidat per a la ruta metabòlica de C18:2. Així, el segon estudi va avaluar els efectes d'una variant (el polimorfisme rs321384923 es va utilitzar com a marcador) al promotor del gen FADS2. Els resultats van demostrar que aquest polimorfisme afecta el perfil d'àcids grassos n-6 millorant l'eficiència de desaturació de C18:2 a C20:4. Addicionalment, es va avaluar l'associació dels gens de les perilipines (PLIN) i del gen de la proteína d’unió al guanilat 1 (GBP1) amb caràcters de creixement i de qualitat de la carn. Així, el tercer estudi va examinar els efectes de dos polimorfismes als gens PLIN1 i PLIN2, relacionats amb l'emmagatzematge i la mobilització de lípids. Els resultats indiquen que el polimorfisme rs333231747 a PLIN2 està associat al creixement primerenc i al pes magre. L'últim estudi va demostrar que GBP1 té dos senyals de poliadenilació actius i que el seu ús depèn del genotip rs80800372. Els porcs portadors de l'al·lel G, associat a una menor viremia després de la infecció pel virus reproductiu i respiratori porcí, van tenir transcripcions més llargues i una menor expressió gènica. Uno de los recientes objetivos incluido en algunas líneas paternas seleccionadas para mercados de calidad es lograr un nivel óptimo de grasa intramuscular y de composición en ácidos grasos sin penalizar el crecimiento magro. Esta tesis comprende cuatro estudios realizados en una línea pura de cerdos Duroc destinados a la producción de cárnicos de alta calidad. Los experimentos se diseñaron con el objetivo de comprender mejor la variabilidad genética subyacente en el contenido de grasa y la composición en ácidos grasos e identificar marcadores potenciales a la selección. El primer estudio examinó los parámetros genéticos de la ruta del ácido linoleico (C18:2) al ácido araquidónico (C20:4). En particular, se demostró que se espera que la selección del valor absoluto de C18:2 proporcione una respuesta similar a la selección por grasa intramuscular a grasa dorsal restringida. Estos hallazgos llevaron a investigar el gen de la desaturasa-2 de los ácidos grasos (FADS2), como gen candidato para la ruta del C18:2. Por lo tanto, el segundo estudio evaluó los efectos de una variante (el polimorfismo rs321384923 se usó como marcador) en el promotor del gen FADS2. Los resultados mostraron que este polimorfismo afecta el perfil de ácidos grasos n-6 al aumentar la eficiencia de desaturación de C18:2 a C20:4. Además, se evaluó la asociación de los genes de las perilipinas (PLIN) y el gen de la proteína de unión a guanilato 1 (GBP1) con caracteres de crecimiento y de calidad de la carne. De este modo, el tercer estudio examinó los efectos de dos polimorfismos en PLIN1 y PLIN2, relacionados con la deposición y la movilización de lípidos. Los resultados indicaron que el polimorfismo rs333231747 en PLIN2 se asocia con el crecimiento temprano y con el peso magro. El último estudio mostró que GBP1 tiene dos señales de poliadenilación activas y que su uso depende del genotipo rs80800372. Los cerdos portadores del alelo G, asociado con una menor viremia después de la infección por el virus reproductivo y respiratorio porcino, tenían transcritos más largos y una menor expresión génica. En condiciones no epidémicas, el alelo G aumentó el contenido de grasa intramuscular pero disminuyó el peso magro. El contenido de C18:2 y los marcadores genéticos investigados se pueden usar para diseñar estrategias de selección adecuadas para mejorar la calidad de la carne y el crecimiento magro.<br>This PhD dissertation is part of a research line on the genetic improvement of pork quality. One of the latest goals to be included in some sire lines selected for premium markets is to achieve an optimum level of intramuscular fat and fatty acid composition without penalizing lean growth performance. This thesis is comprised of four studies conducted on a purebred Duroc line used for producing high-quality meat products. The experiments were designed with the aim to better understand the genetic variability underlying fat content and fatty acid compostion and to identify potential markers for breeding. The first study examined the genetic parameters of the linoleic acid (C18:2) to arachidonic acid (C20:4) pathway. In particular, it was showed that selection for the absolute value of C18:2 is expected to deliver a similar response outcome as selection for intramuscular fat at restrained backfat thickness. These findings led to investigate the fatty acid desaturase-2 (FADS2) gene, as a candidate gene for C18:2 metabolism route. Thus, the second study evaluated the effects of a variant (rs321384923 was used as a tag single nucleotide polymorphism) in the promoter of the FADS2 gene. Results showed that this polymorphism affects the n-6 fatty acid profile by enhancing the desaturation efficiency of C18:2 to C20:4. Additionally, the association of perilipin (PLIN) genes and guanylate-binding protein-1 (GBP1) gene with growth and meat quality traits was assessed. Thus, the third study examined the effects of two polymorphisms in PLIN1 and PLIN2, which have been related to lipid storage and mobilization. Results indicated that the rs333231747 polymorphism on PLIN2 is associated to early growth and lean weight. The last study showed that GBP1 has two active polyadenylation signals and that their usage depends on the rs80800372 genotype. The pigs carrying the G allele, which has been associated with lower viraemia after porcine reproductive and respiratory virus infection, had longer transcripts and lower gene expression. In non-epidemic conditions, the G allele increased intramuscular fat content but decreased lean weight. Linoleic acid content and the investigated genetic markers can be used to design appropriate selection strategies to enhance meat quality and lean growth.

Beard<br>Pharmacokinetics of Intramuscular Morphine in the Horse Elizabeth Devine, DVM; Butch KuKanich, DVM, PhD, DACVCP; Warren Beard, DVM, MS, DACVS Objective - To determine the pharmacokinetics of morphine after intramuscular administration in a clinical population of horses Design – Prospective, clinical study Animals – Pilot study included 2 normal horses and the clinical study included 75 horses Procedures – Morphine was administered at 0.1mg/kg, IM and 2-3 blood samples were obtained from each horse at various times from 0-9 hours after administration. Plasma morphine concentrations were measured using liquid chromatography and mass spectrometry. Results – Data was analyzed using a naïve pooled pharmacokinetic model. The half-life for the elimination phase was approximately 1.5 hours, the volume of distribution (per bioavailability) was approximately 4.5 L/kg and the clearance (per bioavailability) was approximately 35 mL/kg/min. The peak plasma concentration was 21.6 ng/mL and occurred approximately 4 minutes after administration. Plasma concentrations of morphine were below the limit of quantification by 7 hours in 74 horses. Conclusions and Clinical Relevance – The relatively short half-life of morphine indicates the need for frequent dosing to maintain targeted plasma concentrations. Adverse effects were uncommon in this study and morphine was well tolerated at a dose of 0.1 mg/kg, IM. Morphine may be a useful adjunctive therapy in painful horses, but the variable plasma concentrations suggest the dose and dosing interval may need to be adjusted to the individual patient’s response.

A divergent selection experiment on intramuscular fat (IMF) content was performed in rabbits during eight generations. Selection criterion was the averaged phenotypic value of IMF measured in Longissimus dorsi (LD) muscle at 9 wk of age in two full-sibs of the candidate. The aim of this Thesis is to evaluate the direct response to selection for IMF and the correlated responses in other meat and carcass quality traits. Divergent selection for IMF was successful. Direct response to selection in the eight generation was 2.4 standard deviations (SD) of the trait, which represents a genetic progress of 5% of the mean per generation. Genetic trends were symmetrical for both lines. Correlated responses to selection were measured from fifth to eighth generations. Fatty acid composition of LD was affected by selection for IMF. High-IMF line showed greater monounsaturated (MUFA) but lower polyunsaturated (PUFA) fatty acids percentages, and their individual fatty acid percentages showed similar correlated responses, except for C18:3n-3 that was greater in the high-IMF line. We did not find differences between lines for saturated fatty acid (SFA) percentage, but C14:0 and C16:0 percentages were greater in the high-IMF line, and C18:0 was greater in the low-IMF line. A positive correlated response to selection for IMF was observed in the IMF content and fatty acid composition of other muscles with diverse oxidative pattern (Biceps femoris, Supraspinatus and Semimembranosus proprius). Protein content of LD that was greater in the high-IMF line, whereas we did not observed differences between lines in colour and pH meat quality traits. Instrumental firmness was 9.9% greater in the low-IMF line than in the high-IMF line, whereas we did not find differences between lines in other instrumental texture parameters and cooking loss. No effect of selection for IMF was observed in any sensory attribute. Regarding carcass quality traits, a positive correlated response to selection for IMF was observed in carcass fat depots, whereas we did not find differences between lines in other traits. Genetic parameters for IMF and some of the meat and carcass quality traits cited above were estimated and corrobated with the phenotypic responses to selection observed. In this Thesis, we also studied the lipid metabolism of the divergent lines. High-IMF line showed greater glucose-6-phosphate dehydrogenase (G6PDH), malic enzyme (EM) and fatty acid synthase (FAS) lipogenic activities in glycolytic LD muscle than low-IMF line, and greater G6PDH and FAS activities in the oxidative Semimembranosus proprius muscle and perirenal fat depot. However, differences between lines were observed only at 13 wk of age and not at 9 wk. Catabolic activities in muscle involved in the lipid metabolism did not explain the differences between lines for IMF. Liver size, which is the main lipogenic tissue in growing rabbits, was greater in the high-IMF line and showed greater G6PDH and EM activities than low-IMF line at 9 wk, although differences for EM were not relevant.<br>Se ha realizado un experimento de selección divergente por grasa intramuscular (GIM) en conejo durante ocho generaciones. El criterio de selección fue el promedio del valor fenotípico de GIM medido en el músculo Longissimus dorsi (LD) a las 9 semanas de edad en dos hermanos completos del candidato a la selección. El objetivo de esta tesis es evaluar la respuesta directa a la selección por GIM y las respuestas correlacionadas en otros carcateres de calidad de la canal y de la carne. La selección divergente por GIM ha tenido éxito. La respuesta directa en la octava generación de selección fue de 2.4 desviaciones típicas (DT) del carácter, lo que representa un progreso genético del 5% de la media en cada generación. Las líneas mostraron tendencias genéticas simétricas. Las respuestas correlacionadas a la selección en caracteres de calidad de la canal y de la carne se evaluaron desde la quinta hasta la octava generación. La composición de ácidos grasos de LD también se vió afectada por la selección. La línea de alta GIM mostró un mayor porcentaje de ácidos grasos moninstaurados (MUFA) y un menos procentaje de poliinsaturados (PUFA) que la línea de baja GIM, y los MUFA y PUFA individuales mostraron respuestas correlacionadas similares a los grupos, excepto el porcentaje de C18:3n-3 que fue mayor en la línea de alta GIM. No encontramos diferencias entre líneas en el porcentaje de ácidos grasos saturados (SFA), aunque los porcentajes de C14:0 y C16:0 fueron mayores en la línea de alta GIM, y el porcentaje de C18:0 fue mayor en la línea de baja GIM. Se observó una respuesta correlacionada positiva en el contenido de GIM y en la composición de ácidos grasos de otros músculos con un patrón oxidativo diferente al LD (Biceps femoris, Supraspinatus y Semimembranosus proprius). Se estimaron los parámetros genéticos de GIM y de otros caracteres de calidad de la carne y de la canal citados anteriormente, y se corroboraron con las respuestas correlacionadas a la selección observadas. En esta tesis también se ha estudiado el metabolismo lipídico de las líneas divergentes. La línea de alta GIM mostró mayor actividad de las enzimas lipogénicas 6-glucosa-fosfato deshidrogenasa (G6PDH), enzima málico (EM) y ácido graso sintasa (FAS) en el músculo glicolítico LD respecto de la línea de baja GIM, y mayor actividad de las enzimas G6PDH y FAS en el músculo oxidativo Semimembranosus proprius y en el depósito de grasa perirrenal. Sin embargo, estas diferencias solo fueron observadas a las 13 semanas de edad, y no a las 9 semanas. En los músculos, las actividades de las enzimas catabolicas implicadas en el metabolsimo de la grasa no mostraron diferencias entre líneas. El hígado, que es el principal tejido lipogénico en conejos en crecimiento, fué más grande en la línea de alta GIM, y mostró mayor actividad de las enzimas G6PDH y EM que la línea baja, aunque las diferencias en EM no fueron relevantes.<br>S'ha realitzat un experiment de selecció divergent per greix intramuscular (GIM) en conill durant huit generacions. L'objectiu d'esta tesi és avaluar la resposta directa a la selecció per GIM i les respostes correlacionades en altres carcateres de qualitat de la canal i de la carn. La selecció divergent per GIM ha tingut èxit. La resposta directa en la sèptima generació de selecció va ser de 2.4 desviacions típiques (DT) del caràcter, la qual cosa representa un progrés genètic del 5% de la mitjana en cada generació. Les respostes correlacionades a la selecció en caràcters de qualitat de la canal i de la carn es van avaluar des de la quinta fins a l'octava generació. En esta tesi també s'estudia en metabolisme lipídic de les línies divergents. La línia d'alta GIM va mostrar major activitat dels enzims lipogénics 6-glucosa-fosfato deshidrogenasa (G6PDH), enzim màlic (EM) i àcid gras sintasa (FAS) en el múscul glicolític LD respecte de la línia de baixa GIM, i major activitat dels enzims G6PDH i FAS en el múscul oxidatiu Semimembranosus proprius i en el depòsit de greix perirrenal. Abstract:
The steady decline in sheepmeat consumption throughout Australia over recent decades has been partly driven by inconsistencies in eating quality. Eating quality, and thus sensory perception (tenderness, juiciness, flavour, overall liking), is a key factor influencing consumer demand of sheepmeat. A strong positive driver of consumer sheepmeat sensory scores is intramuscular fat (IMF), and has therefore been proposed as a required measurement in future grading systems. IMF of the M. longissimus lumborum (loin) has been shown to be moderately correlated with IMF of muscles throughout the lamb carcass, indicating potential to predict eating quality of these muscles based on loin IMF. Thus, there is potential to predict the eating quality of these muscles based on the IMF content of the loin. Consumer sensory scores of untrained consumers were obtained for the grilled loin, M. semimembranosus (topside), M. gluteus medius (rump), M. biceps femoris (outside) and both the M. rectus femoris and M. vastus lateralis (knuckle) of 918 lambs. IMF was measured in all muscles using a lab-based near-infrared procedure. Linear mixed effects models (SAS Version 9.1) were used to analyse consumer scores for tenderness, juiciness, flavour and overall liking of all individual cuts. Two versions of these models were tested; firstly fitting loin IMF as a covariate, and secondly fitting IMF of each individual cut as a covariate. Both loin IMF and individual cut IMF predicted the eating quality of all cuts for tenderness and flavour. However, in the loin, outside and topside, only juiciness and overall liking scores could be predicted. In almost all cases, loin IMF accounted for more eating quality variation than individual cut IMF, describing between 8%-20% more variance. The results suggest that to predict eating quality, measuring of IMF within each cut is not needed, with only a single measurement of IMF taken from the loin required to predict eating quality. This is an important finding and supports current investment within the Australian sheepmeat industry to develop and implement an online objective measurement of IMF within the loin.

Abstract:
Muscle strain injuries affect a wide range of physically active people around the world and are reaching epidemic proportions. Despite the variety of treatment options available in rehabilitation, there are no clear guidelines for electrical stimulation that provide effective reproducible results that address the underlying cause of these injuries. For instance, electrotherapy is inefficient at stimulating muscles, because of imprecise parameters and an ability to target particular muscles. The difference between this study and previous research is the precise delivery of electrical stimulation (intramuscular) at two different frequencies (2 Hz and 50 Hz) and comparing it a control group. Objective: To determine the difference on muscle strength and functional status between three treatments modalities for sub acute and chronic hamstring strains. Design: A randomized experimental design was used to compare the effects of low (2 Hz), high (50 Hz) and no-electrical (control) intramuscular stimulation on muscle strength and mental and functional status (AMSMC HEALTH STATUS INDEX). Each group consisted of 18 subjects. Main Outcome: The difference in treatment modalities was evaluated by comparing the muscle strength test (Biodex Dynamometer) results and the AMSMC HEALTH STATUS INDEX results in pretest and post-test conditions. Results: The AMSMC HEALTH STATUS INDEX, but not muscle strength test (Biodex), changed significantly after 2-Hz electrical intramuscular stimulation (pre-test µ = 66.56, Std= 11.92, post-test µ= 92.89, Std= 6.25), whereas no statistically significant changes in health status index and muscle strength test occurred with 50-Hz (pre-test = 69.22, Std= 11.31, post-test µ= 70.22, Std= 12.27)) and no-electrical stimulation groups (pre-test µ= 69.11, post-test µ= 73.39, Std= 13.18).

Abstract:
"Os animais da raça Nelore podem apresentam um chifre, conhecido como chifre banana, pois não tem suporte ósseo no crânio, sendo este mole dando a impressão de quebrado. A descorna é um procedimento muito comum na pecuária, principalmente, em bovinos. Existem três técnicas de descorna a química, a térmica e a cirúrgica. A descorna por cirurgia é indicada para animais mais velhos, com o chifre já desenvolvido, sendo realizado um corte com bisturi na pele envolta dele, e depois com uma serra faz-se a sua retirada, entretanto, no chifre banana se usa apenas o bisturi. Após o término do procedimento é feita a sutura da pele e o curativo local. Este trabalho visa relatar um caso de descorna em bovino Nelore com “chifre banana”. Um bovino Nelore, macho, com aproximadamente 24 arrobas apresentando um chifre banana, foi contido no brete para descorna, pois, um dos seus chifres apresentava um crescimento tumoral na sua base. O animal foi sedado com 2,5ml de xilazina por via intramuscular, feita a tricotomia, assim como, a anestesia local, com 20ml de lidocaína 2% distribuído entre as bordas. Sendo um chifre banana, foi utilizado apenas o corte com o bisturi para retirada dos mesmos. Após a sutura da pele, foi feito o curativo local com unguento e spray prata. Como tratamento foi prescrito 50ml de AgroVet Plus® por via intramuscular, por cinco dias consecutivos. Depois da descorna, o animal ficou em observação e até o presente momento não apresentou nenhuma complicação evidente. Palavras-chave: Tratamento. Cirurgia. Ruminantes."

การทรานสเฟคชันโดยการนำพลาสมิดที่มีชิ้นยีนแทรกสอดเข้าสู่เซลล์เป็นขั้นตอนสำคัญในการประยุกต์ใช้งานด้านชีวเวชศาสตร์ เช่น การทำยีนบำบัด และวัคซีนชนิดดีเอ็นเอ เซลล์ส่วนใหญ่รับพลาสมิดเข้าสู่เซลล์ด้วยประสิทธิภาพที่ต่างกัน เซลล์นำเสนอแอนติเจน (Antigen Presenting Cell; APC) เช่น เซลล์เดนไดรติกและแมโครฟาจ เป็นหนึ่งในเซลล์ที่นำพลาสมิดเข้าสู่เซลล์ได้ยากที่สุด เซลล์เหล่านี้มีบทบาทสำคัญในการนำเสนอแอนติเจนต่อเฮลเปอร์ทีลิมโฟไซต์เพื่อชักนำให้มีการตอบสนองแบบ adaptive อย่างมีประสิทธิภาพ ในการที่จะกระตุ้นภูมิคุ้มกันได้อย่างมีประสิทธิภาพหลังจากการปลูกวัคซีนนั้น สามารถที่จะมุ่งเป้าแอนติเจนไปยังเซลล์นำเสนอแอนติเจน วัคซีนชนิดดีเอ็นเอเป็นวัคซีนชนิดใหม่ที่มีศักยภาพที่จะทดแทนวัคซีนที่มีใช้อยู่ในปัจจุบันได้ จากเหตุผลในด้านความง่ายในการเตรียม ความเสถียรและราคาต้นทุนที่ถูก โดยทั่วไปวัคซีนชนิดดีเอ็นเอจะให้ทาง intradermal หรือ intramuscular และเชื่อกันว่าเซลล์ใด ๆ ก็สามารถนำพลาสมิดเข้าสู่เซลล์ได้ การมุ่งเป้าวัคซีนชนิดดีเอ็นเอไปยังเซลล์จำเพาะชนิดแบบจำเพาะนั้นเป็นสิ่งที่พึงประสงค์ในการเหนี่ยวนำการตอบสนองของภูมิคุ้มกันที่ดีที่สุด ออโต้ฟาจีเป็นกลไกป้องกันการรุกรานของเชื้อก่อโรคในระดับเซลล์โดยจะกำจัดแอนติเจนภายในเซลล์ รวมถึงเชื้อก่อโรคแอนติเจนที่เป็นโปรตีนที่ผ่านการย่อยจะถูกนำเสนอแก่เฮลเปอร์ทีลิมโฟไซต์ ดังนั้น ในงานวิจัยนี้ผู้วิจัยจึงมีวัตถุประสงค์ในการพัฒนาระบบนำส่งดีเอ็นเอพลาสมิดที่มุ่งเป้าไปยัง APC โดยใช้อนุภาคนาโนจากไคโทซานและผนวกระบบการเหนี่ยวนำออโต้ฟาจีเมื่อให้นำส่งดีเอ็นเอแล้วโดยใช้พลาสมิดเหนี่ยวนำออโต้ฟาจี ก่อนอื่นผู้วิจัยได้หาภาวะที่เหมาะสมในการทรานสเฟคชันโดยใช้ไคโทซานละลายได้ในกรดในการห่อหุ้มพลาสมิดในระดับ in vitro ที่สัดส่วน N/P ที่ 8:1-10:1 ให้อัตราการทรานสเฟคชันเข้าเซลล์สูงสุดใน HEK293T ผู้วิจัยได้ดัดแปรไคโทชานให้มี strepavidin และใช้ในการห่อหุ้มพลาสมิด หลังจากนั้นนำไคโทซาน strepavidin-พลาสมิดไปบ่มกับ biotinylated anti-F4/80 antibody ซึ่งจำเพาะต่อโมเลกุลบนผิวเซลล์แมโครฟาจ ซึ่งพบว่าไคโทซานดัดแปรนี้มีบ่มร่วมกับ biotinylated anti-F4/80 antibody ไม่สามารถเพิ่มอัตราการทรานสเฟคชัยเข้าเซลล์ไลน์แมโครฟาจ RAW264.7 ได้ การศึกษานี้จึงบ่งชี้ว่าจำเป็นต้องมีการศึกษาเพิ่มเติมเพื่อหาภาวะที่เหมาะสมในการใช้ไคโทซานดัดแปรให้มี strepavidin เพื่อนำส่งแบบมุ่งเป้าดีเอ็นเอไปยังเซลล์เป้าหมาย อีกทั้งเพื่อเพิ่มการนำเสนอแอนติเจนต่อทีลิมโฟไซต์ผู้วิจัยจึงได้พัฒนาระบบพลาสมิดที่เหนี่ยวนำให้เซลล์เกิดออโต้ฟาจี จากการใช้พลาสมิดที่มียีน mTOR ที่มีการกลายพันธุ์ที่ทำให้ไคเนสของ mTOR เสียแอคติวิตี พบว่าสามารถตรวจหาการเกิดออโต้ฟาจีใน HEK293T และเซลล์ไลน์เดนไดรติก JAWSII ได้ ดังนั้น ระบบการเหนี่ยวนำออโต้ฟาจีจึงได้มีการพัฒนาขึ้น ดังนั้น จึงเป็นไปได้ว่าสามารถใช้วัคซีนชนิดดีเอ็นเอในการกระตุ้นการตอบสนองของภูมิคุ้มกันที่แรงโดยการนำเทคนิคนี้มาใช้ร่วมกัน
