Full textFrom the middle of the 1950s, traveling exhibitions of Contemporary Inuit Art have been staged across several continents. These exhibitions had been enthusiastically received, thanks in large part to the particular positioning put forth by James A. Houston, which established Contemporary Inuit Art within the artistic constructs of primitivism and modernism. This warm welcome was in marked contrast to France's own reception of Inuit art. Even though its collective imaginary has been largely shaped by figures from the North Pole, a view that can be traced back to the first apostolic and exploratory missions of the continent, France remained the only country that showed a certain distrust towards this artistic expression. This thesis will examine the conditions by which Contemporary Inuit Art has been re-interpreted within a series of evolving historical contexts, beginning with the domination of the primitivist paradigm until the 1970s, and moving towards the multiplication of individual and collective projects from the early 1980s, a context which prevails to this day. Throughout this examination, the question of Contemporary Inuit Art's ability to be seen as a symbol of artistic and cultural authenticity will be addressed. The final chapter will deal with the notion of aboriginal discourse and the 'double address' mode of communication that Inuit artists tend to employ

Full textShamanism in contemporary native art in Canada is becoming one of the most relevant ethnic markers. It is the sacred place, the major source for cultural resistance, with strong roots and political implications underneath. It works as a powerful identity referent. As we analyse the aesthetic phenomenon from a modern perspective,different issues are emerging : how shamanism is acknowledged, both as a concept and as a practice by the First Nations artists ? Which social functions and philosophical involvements are assumed by the contemporary “shamanic art”, considering its symbolic part as the keeper of identity and spirituality, or/and performed or seen as a healing process ?In the postmodern artistic context, shamanism may be considered as an essential source of power and inspiration, a sacred land that most of the native artists are now exploring, defining therefore a new ontology. From the deep roots of the traditional knowledge, their legacy, and through their own contemporary experiment, these artists are trying to bring back the original wisdom in order to reconnect themselves with the native cosmogony and consequently, to reduce the psychic schism between traditionalism’s nostalgy and contemporary indianness (nativeness). Therefore the status of “betweenness”, that is specifically attributed to the native artists, is shifted into a power of creative transformation. This thesis analyses the process of shamanistic inspiration in contemporary native art, through the philosophical and political issues as well as with an anthropological and aesthetics point of view. The historical and sociological contexts are explored before defining th! e two main missions of the “Shamen of Art” : the rewrighting of the History and the Conquest of Identity, drawing through the recognition of their alterity, the architecture of a new “Amerindia”

Full textThis doctoral research examines the themes of figurative practices, symbolic representations and the socio-cultural stakes specific to Inuit graphic arts in the communities of Kinngait (Cape Dorset) and Pangniqtuuq (Pangnirtung) in Nunavut (the Canadian Arctic). The notions of drawing (titiqtugaq-) and of speech (uqaq-) are central to the thesis, which is guided by an interdisciplinary approach within the perspective of ethno-history of Inuit sketch art. The thesis is organized into three parts. The first explores the configuration of Inuit thought associated with the concepts of graphic art, visual representation and creation, through their linguistic expression (Chapter II). In addition, ethnography of the local art scene looks at drawing and the socio-economic activities that are associated with it, in connection with the status of the artist (Chapters III and IV). The second part looks at figuration in relation to power words, from cosmogenesis and ancient graphic techniques (Chapter V). With this in hand, the second part then looks at the interactions between drawing and speaking from a symbolic perspective: through drawings, the thoughts and words are put into action (Chapters VI and VII). The last part of the dissertation continues the analysis by defining art as part of the socio-cultural and political dynamics of the Nunavummiut. Recourse to drawing, as a community project, is studied with reference to recent examples (Chapter VIII), prior to being placed, within an ontological dimension, at the centre of relational and socio-cosmic exchange dynamics (Chapter IX)

Full textThe thesis concerns the study of the fortifications of Ardea and Castrum Inui - the city harbour - which, being characteristic elements of the ancient town-planning, allowed a reinterpretation of the city and region transformations. The job consists of three sections. The first introductory section is focused on the geomorphology of the sites and on the retrace of the history of former studies and researches. In the second part were analyzed the transformation of the two sites from the earliest archaeological statements – dating back to Bronze Age in the urban area and to archaic age in the Castrum – to the imperial age. The analysis of each phase is introduced by a study on the historical context in order to determine what were the events, not only related to Ardea but more generally to the cities of Latium Vetus, which might have generated the urban transformation. From time to time were examined the issues relating to defence works and their relations with the organization of the city and its port. The opportunity to explore a formerly unpublished monument like the opus quadratum fortification of Castrum Inui has provided important elements of confrontation that led to recognize the existence of different construction phases even in the city walls, never identified up to now. Finally, the third section is the filing of the fortifications and includes the analysis of currently visible rests, made by describing the construction characteristics with particular emphasis on metrology data, which has been used for the chronological definition of the various construction phases. The study is therefore supplemented by tables containing measurements and information regarding the blocks and their installation. The thesis is finally accompanied by two appendices: the first one collects quotations pertaining Ardea coming from literary sources, the second one is a large photographic collection including both the documentation of the remains in its present state of preservation, and the archival records which helped to analyze the characteristics of destroyed traits.

Full textNon Small Cell Lung Cancer (NSCLC) is the leading cause of cancer death worldwide, with poor prognosis and a high rate of recurrence despite early surgical removal. It is therefore essential to identify new prognostic markers and new therapeutic targets. We are interested in gene regulation related to hypoxia, a factor associated with relapse of lung adenocarcinomas (LUAD). The roles of long non coding RNAs (incRNAs) in cancer development and hypoxic response are largely unexplored. A transcriptome profiling of early-stage LUAD samples indicated that a set of incRNAs was correlated to a metagene hypoxic signature. Some of these transcripts were also sensitive to hypoxia in LUAD cell lines. We focused on a new "hypoxaLinc", named NLUCAT1 that is strongly up-regulated by hypoxia in vitro and correlated to hypoxic markers and bad prognosis in LUAD samples. Full molecular charactherization of NLUCAT1 showed that LUCAT1 is mainly regulated by NF-kβ and NRF2 transcription factors. Targered deletion of NLUCAT using CRISPR/CAS9 in A549 LUAD cell line, revelated a decrase in proliferative and invasive properties, an increase in oxidative stress and a higher sensisivity to displatin-induced apoptosis. We identified genes of the NRF2-regulated and anti-oxidant response whose RNA interference partially mimicked the consequences of NLUCAT1 inactivation on ROS-dependent caspase activation. Overall, our data strongly demonstrate that NLUCAT1 exerts pro-tumoral activities in early stages hypoxic LUADs ans suggest it could represent a new potential therapeutic target in lung cancer

Full textCD95 belongs to the TNF-R superfamily, and it triggers an apoptotic signal. CD95 plays a key role in homeostasis of the immune system and in the elimination of infected and transformed cells. Upon CD95L binding, CD95 recruits FADD, which in turn aggregates initiator caspases (i.e., caspase-8 and -10). The complex CD95, FADD and caspase-8/10 is called DISC for Death Inducing Signaling Complex. At the DISC level, caspase aggregation leads to their activation and death of the cells through apoptosis. The CD95-mediated apoptotic signal is modulated by microdomains, or lipid rafts, which are plasma membrane sub-domains enriched in sphygolipids and cholesterol. Thereby, partition of CD95 into lipid rafts promotes the apoptotic signal. Activation of the phosphatidylinositol 3-kinase (PI3K)/Akt signaling pathway is known to prevent the CD95-mediated apoptotic signal in malignant cells, and to control lateral mobility of CD95 at the plasma membrane. Herein, we showed that inhibition of PI3K signal induced i) the distribution of CD95 into lipid rafts and ii) the subsequent induction of the CD95-mediated apoptotic signal through a CD95L independent manner. In conclusion, we pinpointed that PI3K/Akt signaling pathway inhibits the CD95 signal by acting upstream DISC formation and even upstream the CD95-CD95L interaction through the exclusion of the death receptor from the microdomains

Full textThe presence of DNA in the cytosol represents a danger signal. Mitochondrial DNA (mtDNA) has been recognized as a DAMP (damage-associated molecular-pattern molecule), able to induce the production of pro-inflammatory cytokines (interferons). The human cytidine deaminase APOBEC3A (A3A), upregulated by IFN, catalyzes the deamination of cytidine to uridine in single stranded DNA substrates leading to the catabolism of the mutated DNA sequence and the suppression of the signal. First, we demonstrated the role of the RNA polymerase III / RIG-I signaling in the upregulation of A3A expression in response to IFN production. We also confirmed the mtDNA catabolism induced by A3A. However, its overexpression leads to GC vers AT mutations and double-strand DNA breaks in the nuclear genome. The second part of my thesis highlighted the release of mtDNA within the cytosol upon Herpes Simplex Virus Type 1 (HSV-1) infection in a human cellular model, probably triggered by a fragmentation of the mitochondrial network. We demonstrated that this mtDNA release is associated with a strong production of IFN and the overexpression of A3A. While we confirmed the role of the RNA polymerase III / RIG-I signaling, the cGAS-STING pathway should be also involved. Finally, in this thesis, we have shown in a human model that the mitochondrial stress induced by HSV-1 contributes to the overexpression of A3A

Full textThe v-erbA oncogene transforms chicken erythroid progenitors by blocking their differentiation andpreventing them to exit a state of self-renewal. The transcriptome of primary avian erythroidprogenitors cells (T2EC) expressing either v-erbA or a non-transforming form of v-erbA werecompared by SAGE. Only some, but not all, mRNAs encoding ribosomal proteins were shown to beaffected. These results suggest that v-erbA could modulate the composition of ribosomes and/ormodulate the extraribosomal functions of specific ribosomal proteins. We therefore decided to analyzethe level of ribosomal proteins associated to ribosomes by 2D-DIGE performed on purified ribosomes.A statistical analysis performed on 4 independent flip-flop experiments demonstrated that the level ofRPL11 is significantly lower in T2EC expressing v-erbA as compared to the non-transforming form ofv-erbA. These data suggest the presence of ribosomes without RPL11 in T2EC expressing v-ErbA.Results obtained from immunoprecipitation experiments were strengthened this hypothesis. The set ofthese data evoke the involvement of ribosomal proteins, and specially RPL11, in the v-erbAtransformation process both at the translational level and possibly in its extra-ribosomal function.Overexpression of RPL11 in T2EC showed a decrease of cell proliferation
