Polynomial vector fields which admit a prescribed Darboux integrating factor are quite well understood when the geometry of the underlying curve is non-degenerate. In the general setting, morphisms of the affine plane may remove degeneracies of the curve, and thus allow more structural insight. In the present paper we establish some properties of integrating factors subjected to morphisms, and we discuss in detail one particular class of morphisms related to finite reflection groups. The results indicate that degeneracies for the underlying curve generally impose additional restrictions on vector fields admitting a given integrating factor.

We are interested in deepening the knowledge of methods based on formal power series applied to the nilpotent center problem of planar local analytic monodromic vector fields [Formula: see text]. Although by the existence of [Formula: see text] it is not possible to describe all nilpotent centers strata, we simplify, improve and also extend previous results on the relationship between these concepts. We use in the performed analysis the so-called Andreev number [Formula: see text] with [Formula: see text] associated to [Formula: see text] which is invariant under orbital equivalency of [Formula: see text]. In this work we continue the study of the centers which are limits of linear type centers. It is proved that if a degenerate center has an inverse integrating factor V(x, y) with V(0, 0) ≠ 0, then this degenerate center is also the limit of linear type centers. Moreover, we show that the degenerate centers with characteristic directions that are the limits of degenerate centers without characteristic directions are also detectable, from a theoretical point of view, with the Bautin method.

With the developing of high integrations in large scale systems, such as aircraft and other industrial systems, there are new challenges in safety analysis due to the complexity of the mission process and the more complicated coupling characteristic of multi-factors. Aiming at the evaluation of coupled factors as well as the risk of the mission, this paper proposes a combined technology based on the Decision Making Trial and Evaluation Laboratory (DEMATEL) model and the Bayesian network (BN). After identifying and classifying the risk factors from the perspectives of humans, machines, the environment, and management, the DEMATEL technique is adopted to assess their direct and/or indirect coupling relationships to determine the importance and causality of each factor; moreover, the relationship matrix in the DEMATEL model is used to generate the BN model, including its parameterization. The inverse reasoning theory is then implemented to derive the probability, and the risk of the coupled factors is evaluated by an assessment model integrating the probability and severity. Furthermore, the key risk factors are identified based on the risk radar diagram and the Pareto rule to support the preventive measurements. Finally, an application of the take-off process of aircraft is provided to demonstrate the proposed method.

AbstractThe Three-North Shelter/Protective Forest Programme (TNSFP), the largest ecological afforestation program in the world, was launched in 1978 and will last until 2050 to improve ecological conditions in the Three-North regions of China. To manage the shelter forests sustainably, it is necessary to accurately estimate air temperature on a large scale, but the spatial distribution of ground meteorological stations is limited. A hybrid method was established by combining stepwise regression modeling and spatial interpolation techniques (SRMSIT) to construct the monthly mean, minimum, and maximum air temperatures (Tmean, Tmin, and Tmax, respectively) at a 1 km × 1 km grid size in the Three-North regions. Stepwise regression modeling was applied to construct the relationship between air temperatures (Tmean, Tmin, and Tmax—the dependent variables) and geographical and Moderate Resolution Imaging Spectroradiometer (MODIS) variables (the independent variables). Spatial interpolation techniques were used to correct the residual values. According to the factor analysis, three geographic (altitude, latitude, and continentality) and two MODIS variables [nighttime land surface temperature (LST) and normalized difference vegetation index] were selected in stepwise regression modeling, and nighttime LST was the most powerful remote sensing variable. The SRMSIT method, in which the spatial interpolation of the residuals was done with inverse distance weighting, achieved average root-mean-square error values at 0.86°, 1.10°, and 1.13°C for Tmean, Tmin, and Tmax, respectively. Therefore, the simple regression algorithms derived from the combination of remote sensing and geographical variables, together with residual interpolation techniques, have the potential to accurately estimate monthly air temperature in large regions.

SummaryLinear quadratic regulator (LQR), a popular technique for designing optimal state feedback controller, is used to derive a mapping between continuous and discrete time inverse optimal equivalence of proportional integral derivative (PID) control problem via dominant pole placement. The aim is to derive transformation of the LQR weighting matrix for fixed weighting factor, using the discrete algebraic Riccati equation (DARE) to design a discrete time optimal PID controller producing similar time response to its continuous time counterpart. Continuous time LQR-based PID controller can be transformed to discrete time by establishing a relation between the respective LQR weighting matrices that will produce similar closed loop response, independent of the chosen sampling time. Simulation examples of first/second order and first-order integrating processes exhibiting stable/unstable and marginally stable open loop dynamics are provided, using the transformation of LQR weights. Time responses for set-point and disturbance inputs are compared for different sampling times as fraction of the desired closed loop time constant.

Glioblastoma is the most common brain tumor in adults in which recurrence has been attributed to the presence of cancer stem cells in a hypoxic microenvironment. On the basis of tumor formation in vivo and growth type in vitro, two published microarray gene expression profiling studies grouped nine glioblastoma stem-like (GS) cell lines into one of two groups: full (GSf) or restricted (GSr) stem-like phenotypes. Aquaporin-1 (AQP1) and aquaporin-4 (AQP4) are water transport proteins that are highly expressed in primary glial-derived tumors. However, the expression levels of AQP1 and AQP4 have not been previously described in a panel of 92 glioma samples. Therefore, we designed secondary data analytics methods to determine the expression levels of AQP1 and AQP4 in GS cell lines and glioblastoma neurospheres. Our investigation also included a total of 2,566 expression levels from 28 Affymetrix microarray probe sets encoding 13 human aquaporins (AQP0–AQP12); CXCR4 (the receptor for stromal cell derived factor-1 [SDF-1], a potential glioma stem cell therapeutic target]); and PROM1 (gene encoding CD133, the widely used glioma stem cell marker). Interactive visual representation designs for integrating phenotypic features and expression levels revealed that inverse expression levels of AQP1 and AQP4 correlate with distinct phenotypes in a set of cell lines grouped into full and restricted stem-like phenotypes. Discriminant function analysis further revealed that AQP1 and AQP4 expression are better predictors for tumor formation and growth types in glioblastoma stem-like cells than are CXCR4 and PROM1. Future investigations are needed to characterize the molecular mechanisms for inverse expression levels of AQP1 and AQP4 in the glioblastoma stem-like neurospheres.

Optical performance in terms of light efficiency, color crosstalk and ambient contrast ratio were analyzed for blue GaN-based micro-light emitting diodes (micro-LEDs) combined with red/green quantum dots (QDs)-polymethyl methacrylate (PMMA) films. The thickness and mass ratio of QDs films are two critical factors in affecting the performance of micro-LEDs. Firstly, the precise optical modeling of QDs-PMMA films is established based on the double integrating sphere (DIS) testing system and inverse adding doubling algorithm (IADA) theory. Red and green QDs-PMMA films are composed of ZnCdSe/ZnS QDs and green ZnCdSeS/ZnS QDs, respectively. The fundamental optical parameters of QDs-PMMA films, including scattering, absorption and anisotropy coefficients, are obtained successfully. Secondly, based on these optical parameters, the Monte Carlo ray tracing method is applied to analyze the effect of a QDs-PMMA film’s thickness and mass ratio on the optical performance of micro-LEDs. Results reveal that the light efficiency first increases and then decreases with the increase of a QDs film’s thickness or mass ratio, owing to the scattering characteristics of QDs. Different from the variation tendencies of light efficiency, the crosstalk between adjacent pixels increases as the QDs-PMMA film’s thickness or mass ratio increases, and the ambient contrast ratio is kept stable when the thickness increases. The mass ratio variation of QDs film can change the optical performance of micro-LEDs more effectively than thickness, which demonstrates that mass ratio is a more important factor affecting the optical performance of micro-LEDs.

This paper presents an improved inverse-time over-current protection method based on the compound fault acceleration factor and the beetle antennae search (BAS) optimization method for a microgrid. The proposed method can not only significantly increase the operation speed of the inverse-time over-current protection but also improve the protection coordination by considering the possible influential factors in terms of microgrid operation modes, distributed generation (DG) integration status, fault types, and positions, which are marked as the most challenging problems for over-current protection of a microgrid. In this paper, a new Time Dial Setting (TDS) of inverse-time protection is developed by applying a compound fault acceleration factor, which can notably accelerate the speed of protection by using low-voltage and short-circuit impedance during the fault. In order to improve the protection coordination, the BAS algorithm is then used to optimize the protection parameters of the pick-up current, TDS, and the inverse time curve shape coefficient. Finally, case studies and various evaluations based on DIgSILENT/Power Factory are carried out to illustrate the effectiveness of the proposed method.

Although genome-wide association studies have brought deep insights into SLE pathogenesis, the precise pathway from genome to metabolome has been largely unknown, and vice versa.Objectives:The aim of this study is to investigate metabolomic regulation in SLE in relation to gene expressions by integrating plasma metabolome data and transcriptome data.Methods:We collected plasma samples from patients with SLE (n=57) who met the 1997 American College of Rheumatology criteria for SLE. Gender- and age-matched healthy controls (HCs) (n=56) were recruited. Metabolic profiles focusing on 39 amino acids were analyzed with liquid chromatography (LC)-mass spectrometry. Transcriptome data of SLE patients were obtained from our RNA-sequencing data of each immune cell subset (total 19 subsets). Whole-genome sequencing was also performed.Results:Our previous experiment showed that about 160 peaks were detected from comprehensive LC-TOFMS and amino acids were useful for distinguishing SLE patients from HCs. Both partial least squares discriminant analysis (PLS-DA) and random forest, a machine learning algorithm, revealed the importance of histidine (His), one of the essential amino acids, to classify SLE patients from HCs, whose plasma level was lower in SLE patients. In addition, inverse correlation between His level and titer of ds-DNA as well as damage index (SDI) was detected. His level was correlated neither with PSL dosage nor with type I interferon (IFN) signature. Receiver operating characteristic (ROC) analysis showed the best predictability for SLE with the combination of specific amino acids including His. Our transcriptome analysis has revealed the significance of oxidative phosphorylation (OXPHOS) in B cells for SLE pathogenesis. Interestingly, OXPHOS signature was inversely correlated with His level in SLE B cells.Conclusion:His may be an important factor for SLE pathogenesis especially in B cells independently from IFN signal. SLC15A4, a transporter of His on lysosome, is one of the SLE GWAS SNPs and has been reported to play an important role in IFN production in B cells through regulation of TLR7/9 activation 2). We also identified that SLE patients with risk allele of SLC15A4 had tendency to show higher plasma His level, indicating His homeostasis could become a novel treatment target for SLE. Most previous obesity projections considered the short-term future only, focused on single non-European countries, and projected ongoing increases foremost. We comparatively project obesity prevalence into the long-term future for 18 European countries and the USA. <b><i>Data:</i></b> We used national age-specific (20–84 years) and sex-specific obesity prevalence estimates (1975–2016) from the NCD Risk Factor Collaboration (NCD-RisC) 2017 study, which are based on available measured height and weight data, supplemented with estimates from a Bayesian hierarchical model. <b><i>Methods:</i></b> We projected age- and sex-specific obesity prevalence up to the year 2100 by integrating the notion of a wave-shaped obesity epidemic into conventional age-period projections. <b><i>Results:</i></b> In 1990–2016, the increasing trends in obesity prevalence were decelerating. Obesity is expected to reach maximum levels between 2030 and 2052 among men, and between 2026 and 2054 among women. The maximum levels will likely be reached first in The Netherlands, USA, and UK, and last in Switzerland; and are expected to be highest in the USA and UK, and lowest in The Netherlands for men and Denmark for women. In 2060, obesity prevalence is expected to be lowest among Dutch men and highest among Swiss men. The projected age-specific obesity prevalence levels have an inverse U-shape, peaking at around the age of 60–69 years. <b><i>Discussion:</i></b> Applying our novel approach to the NCD-RisC 2017 data, obesity prevalence is expected to reach maximum levels between 2026 and 2054, with the USA and UK reaching the highest maximum levels first, followed by other European countries.

Abstract Gene transfer into peripheral blood lymphocytes has several potential applications including the correction of genetic diseases and therapeutic approaches for HIV-1 infection and cancer. Integrating gene transfer system based on murine oncoretroviruses are a convenient tool for such strategies. However, the recent occurrence of uncontrolled clonal T cell expansions in two patients treated with retroviral gene transfer for X-linked severe combined immune deficiency has raised the concern of the risk of insertional oncogenesis associated with the clinical use of integrating viral systems. In vitro studies have indicated that murine viral vectors tend to integrate in the vicinity of transcription start regions of the genome, thus providing a possible mechanism for oncogene activation, however, data from clinical gene transfer trials is lacking. We are following patients affected with adenosine deaminase (ADA) deficiency who have received T-lymphocyte-directed, retroviral-mediated gene transfer starting in 1990. The first treated patient received the last infusion of gene-corrected cells 12 years ago, has never shown any sign of lymphoproliferation and still carries ~20% of gene-corrected peripheral blood lymphocytes. We set out to study the integration sites in the cells of this patient with the aim of mapping the regions involved by retroviral integrations, determining their localization with respect to known genes, and assessing whether a preferred pattern could be defined. Genomic DNA was prepared from stored lymphocyte samples dating 1991, 1992, 1995, 1998, 2000, and 2003. By inverse PCR and ligation-mediated PCR, we have identified ~860 bona fide insertion sites. Search for homology within the human genome using BLAT returned ~330 unique hits that involved a variety of genes, including transcription factors and oncogenes (e.g. RUNX1, STAT5, FYN). To evaluate the distribution pattern of these integration sites, 2000 randomly generated data sets of genomic coordinates were assembled and their distribution relative to annotations of the human genome was analyzed. A preliminary comparison of the random distribution to our experimental samples showed that retroviral integrations in cells obtained from the patient were significantly skewed toward regions within 2 kb of genes (p<0.002) and CpG islands (p<0.001). These results suggest that, similar to what observed in murine fibroblast and human cancer cell lines, transcriptionally active regions of the genome may be preferred targets of retroviral vectors in human primary T lymphocytes. At the same time, our observations show that the resulting integration events are compatible with long-term, event-free in vivo survival of gene-modified cells in clinical settings.

Opticks is an open source project that accelerates optical photon simulation by integrating NVIDIA GPU ray tracing, accessed via NVIDIA OptiX, with Geant4 toolkit based simulations. A single NVIDIA Turing architecture GPU has been measured to provide optical photon simulation speedup factors exceeding 1500 times single threaded Geant4 with a full JUNO analytic GPU geometry automatically translated from the Geant4 geometry. Optical physics processes of scattering, absorption, scintillator reemission and boundary processes are implemented within CUDA OptiX programs based on the Geant4 implementations. Wavelength-dependent material and surface properties as well as inverse cumulative distribution functions for reemission are interleaved into GPU textures providing fast interpolated property lookup or wavelength generation. In this work we describe major recent developments to facilitate integration of Opticks with the JUNO simulation framework including on GPU collection effciency hit culling which substantially reduces both the CPU memory needed for photon hits and copying overheads. Also progress with the migration of Opticks to the all new NVIDIA OptiX 7 API is described.

A fast method called the “inverse approach” for sheet forming modeling is based on the assumptions of the proportional loading and simplified tool actions. To improve the stress estimation, the pseudo-inverse approach was recently developed: some realistic intermediate configurations are geometrically determined to consider the deformation paths; two new efficient algorithms of plastic integration are proposed to consider the loading history. In the direct scalar algorithm (DSA), an elastic unloading-reloading factor γ is introduced to deal with the bending-unbending effects; the equation in unknown stress vectors is transformed into a scalar equation using the notion of the equivalent stress, thus the plastic multiplier Δλ can be directly obtained without iterative resolution scheme. In the γ-return mapping algorithm, the equivalent plastic strain increment estimated by DSA is taken as the initial solution in Simo’s return mapping algorithm, leading to a stable, efficient, and accurate plastic integration scheme. The numerical experience has shown that these two algorithms give a considerable reduction of CPU time in the plastic integration.

The SBM is a strong-form boundary discretization collocation technique and is mathematically simple, easy-to-program, and free of mesh. The crucial step in the implementation of the SBM is to determine the OIFs which isolate the singularities of the fundamental solutions. Traditionally, the inverse interpolation technique (IIT) is adopted to calculate the OIFs on Dirichlet boundary, which is time consuming for large-scale simulation. In recent years, the new methodology has been developed to efficiently calculate the OIFs on Neumann boundary, but the Dirichlet problem remains an open issue. This study employs the subtracting and adding-back technique based on the integration of the fundamental solution over the whole boundary to develop a new formulation of the OIFs on 3D Dirichlet boundary. Several problems with varied domain shapes and boundary conditions are carried out to validate the effectiveness and feasibility of the proposed scheme in comparison with the SBM based on inverse interpolation technique, the method of fundamental solutions, and the boundary element method.

ABSTRACTChronic infection by hepatitis B virus (HBV) is the major contributor to liver disease worldwide. Though HBV replicates via a nuclear episomal DNA (covalently closed circular DNA [cccDNA]), integration of HBV DNA into the host cell genome is regularly observed in the liver in infected patients. While reported as a prooncogenic alteration, the mechanism(s) and timing of HBV DNA integration are not well understood, chiefly due to the lack ofin vitroinfection models that have detectable integration events. In this study, we have established anin vitrosystem in which integration can be reliably detected following HBV infection. We measured HBV DNA integration using inverse nested PCR in primary human hepatocytes, HepaRG-NTCP, HepG2-NTCP, and Huh7-NTCP cells after HBV infection. Integration was detected in all cell types at a rate of >1 per 10,000 cells, with the most consistent detection in Huh7-NTCP cells. The integration rate remained stable between 3 and 9 days postinfection. HBV DNA integration was efficiently blocked by treatment with a 200 nM concentration of the HBV entry inhibitor Myrcludex B, but not with 10 μM tenofovir, 100 U of interferon alpha, or a 1 μM concentration of the capsid assembly inhibitor GLS4. This suggests that integration of HBV DNA occurs immediately after infection of hepatocytes and is likely independent ofde novoHBV genome replication in this model. Site analysis revealed that HBV DNA integrations were distributed over the entire human genome. Further, integrated HBV DNA sequences were consistent with double-stranded linear HBV DNA being the major precursor. Thus, we have established anin vitrosystem to interrogate the mechanisms of HBV DNA integration.IMPORTANCEHepatitis B virus (HBV) is a common blood-borne pathogen and, following a chronic infection, can cause liver cancer and liver cirrhosis. Integration of HBV DNA into the host genome occurs in all known members of theHepadnaviridaefamily, despite this form not being necessary for viral replication. HBV DNA integration has been reported to drive liver cancer formation and persistence of virus infection. However, when and the mechanism(s) by which HBV DNA integration occurs are not clear. In this study, we have developed and characterized anin vitrosystem to reliably detect HBV DNA integrations that result from a true HBV infection event and that closely resemble those found in patient tissues. Using this model, we showed that integration occurs when the infection is first established. Importantly, we provide here a system to analyze molecular factors involved in HBV integration, which can be used to develop strategies to halt its formation.

Depressive symptoms and religious/spiritual (R/S) practices are widespread around the world, but their intersection has received relatively little attention from mainstream mental health professionals. This paper reviews and synthesizes quantitative research examining relationships between R/S involvement and depressive symptoms or disorders during the last 50 years (1962 to 2011). At least 444 studies have now quantitatively examined these relationships. Of those, over 60% report less depression and faster remission from depression in those more R/S or a reduction in depression severity in response to an R/S intervention. In contrast, only 6% report greater depression. Of the 178 most methodologically rigorous studies, 119 (67%) find inverse relationships between R/S and depression. Religious beliefs and practices may help people to cope better with stressful life circumstances, give meaning and hope, and surround depressed persons with a supportive community. In some populations or individuals, however, religious beliefs may increase guilt and lead to discouragement as people fail to live up to the high standards of their religious tradition. Understanding the role that R/S factors play in preventing depression, facilitating its resolution, or leading to greater depression will help clinicians determine whether this is a resource or a liability for individual patients.

Transition of grapevine buds from paradormancy to endodormancy is coordinated by changes in gene expression, phytohormones, transcription factors, and other molecular regulators, but the mechanisms involved in transcriptional and post-transcriptional regulation of dormancy stages are not well delineated. To identify potential regulatory targets, an integrative analysis of differential gene expression profiles and their inverse relationships with miRNA abundance was performed in paradormant (long day (LD) 15 h) or endodormant (short day (SD), 13 h) Vitis riparia buds. There were 400 up- and 936 downregulated differentially expressed genes in SD relative to LD buds. Gene set and gene ontology enrichment analysis indicated that hormone signaling and cell cycling genes were downregulated in SD relative to LD buds. miRNA abundance and inverse expression analyses of miRNA target genes indicated increased abundance of miRNAs that negatively regulate genes involved with cell cycle and meristem development in endodormant buds and miRNAs targeting starch metabolism related genes in paradormant buds. Analysis of interactions between abundant miRNAs and transcription factors identified a network with coinciding regulation of cell cycle and epigenetic regulation related genes in SD buds. This network provides evidence for cross regulation occurring between miRNA and transcription factors both upstream and downstream of MYB3R1.

AbstractInduced pluripotent stem cells (iPSCs), generated from somatic cells, not only possess similar characteristics with embryonic stem cells (ESCs), but also present more advantages than ESCs in medical applications. The classical induction method that utilizes the integration of exogenous genes into chromosomes may raise the potential risk of the safety of iPSCs. To investigate the potential correlation between the integration sites of exogenous transcription factors (TFs) and iPSCs’ pluripotency and safety, the integration of exogenous genes in three iPSC lines, which met the golden standard of murine developmental assay (tetraploid complementation), were analyzed. Twenty-two integration sites of exogenous TFs were identified by nested inverse polymerase chain reaction (iPCR) and 39 flanking genes’ functions were analyzed by gene ontology (GO). In the 22 integrated sites, 17 (77.3%) were located in the intergenic regions and the remainder were located in introns far from the transcription start sites. Microarray analysis of the flanking genes in these cells showed that there was no distinct difference in expression levels between the iPSCs, ESCs and mouse embryonic fibroblast (MEF), suggesting that the integration of exogenous TFs has no significant influence on the expression of flanking genes. Gene ontology analysis showed that although most of the flanking genes were housekeeping genes, which were necessary for basic life activity, none of these 39 flanking genes have correlation with tumorigenesis or embryogenesis, suggesting that the integration sites hold low risk of tumorigenesis.

To investigate the association between childhood obesity and neighborhood-level socio-economic and environmental factors in Japan. The prevalence of childhood obesity in 2008 was obtained from annual reports of the School Health Survey. The following 12 factors were publicly available: income per person, ratio of people completing up to college or university education, population density, total length of roads per square kilometer, number relative to the population of food and drink stores, restaurants, large-scale retail stores, convenience stores, passenger cars, traffic accidents, criminal offenses, and death by accidents. The ratio of people completing up to college or university education was inversely associated with obesity in boys and girls. No association was found between obesity and the other factors examined. An inverse association was shown between educational level and childhood obesity in both boys and girls. Further studies integrating associations between childhood obesity and neighborhood-level factors are needed.
