Abstract:
Type II nephronophthisis (NPHP2) is an autosomal recessive renal cystic disorder characterized by mutations in the inversin gene. Humans and mice with mutations in inversin have enlarged cystic kidneys that may be due to fluid accumulation resulting from altered ion transport. To address this, transepithelial ion transport was measured in shRNA-mediated inversin-depleted mouse cortical collecting duct (mCCD) cells. Loss of inversin decreased the basal ion flux in mCCD cells compared with controls. Depletion of inversin decreased vasopressin-induced Na+ absorption but did not alter Cl− secretion by mCCD cells. Addition of amiloride, a specific blocker of the epithelial sodium channel (ENaC), abolished basal ion transport in both inversin knockdown and control cells, indicating ENaC involvement. Transcript levels of ENaC β-subunit were reduced in inversin-knockdown cells consistent with decreased ENaC activity. Furthermore, Nedd4l (neural precursor cell expressed, developmentally downregulated 4 like), an upstream negative regulator of ENaC, was evaluated. The relative amount of the phosphorylated, inactive Nedd4l was decreased in inversin-depleted cells consistent with decreased ENaC activity. The protein levels of Sgk1 (serum and glucocorticoid-inducible kinase), which phosphorylates Nedd4l, remained unchanged although the transcript levels were increased in inversin-depleted cells. Interestingly, mRNA and protein levels of Crtc2 (Creb-regulated transcription coactivator) kinase, a positive regulator of Sgk1, were decreased in inversin-depleted cells. Together these results suggest that loss of inversin decreases Na+ transport via ENaC, mediated in part by transcriptional and posttranslational regulation of Crtc2/Sgk1/Nedd4l axis as a contributory mechanism for enlarged kidneys in NPHP2.

Abstract:
Mutations in inversin cause nephronophthisis type II, an autosomal recessive form of polycystic kidney disease associated with situs inversus, dilatation, and kidney cyst formation. Since cyst formation may represent a planar polarity defect, we investigated whether inversin plays a role in cell division. In developing nephrons from inv−/− mouse embryos we observed heterogeneity of nuclear size, increased cell membrane perimeters, cells with double cilia, and increased frequency of binuclear cells. Depletion of inversin by siRNA in cultured mammalian cells leads to an increase in bi- or multinucleated cells. While spindle assembly, contractile ring formation, or furrow ingression appears normal in the absence of inversin, mitotic cell rounding and the underlying rearrangement of the cortical actin cytoskeleton are perturbed. We find that inversin loss causes extensive filopodia formation in both interphase and mitotic cells. These cells also fail to round up in metaphase. The resultant spindle positioning defects lead to asymmetric division plane formation and cell division. In a cell motility assay, fibroblasts isolated from inv−/− mouse embryos migrate at half the speed of wild-type fibroblasts. Together these data suggest that inversin is a regulator of cortical actin required for cell rounding and spindle positioning during mitosis. Furthermore, cell division defects resulting from improper spindle position and perturbed actin organization contribute to altered nephron morphogenesis in the absence of inversin.

Penelitian ini berupa analisis inversi data seismik dengan menggunakan impedansi elastik dan lr (lambda-rho) untuk mengestimasi kandungan reservoir batupasir. Analisis Inversi Amplitude Versus Offset (AVO) dilakukan untuk mendapatkan reflektivitas impedansi elastik dan lambda-rho sehingga diperoleh kelas anomalinya. Dalam menentukan litologinya diperoleh sudut 30o sebagai sudut yang sensitif sehingga selanjutnya dilakukan proses inversi dengan sudut tersebut. Hasil dari analisis Anomali Inversi Amplitude Versus Offset (AVO) menetapkan kelas anomali respon terletak pada posisi IIp sehingga dengan menggunakan sudut 30o tersebut dilakukan pemetaan inversi impedansi elastik untuk mengetahui litologi batupasir yang diperkirakan mempunyai arah timurlaut-baratdaya. Selanjutnya untuk melakukan analisis untuk estimasi adanya fluida dengan menggunakan analisis inversi lambda-rho sehingga diperoleh nilai target 40-65 MPa*gr/cc pada litologi batupasir tersebut dengan arah orientasi utara-tenggara.

Skellefteå Kraft strävar, i likhet med andra elnätsbolag, efter att felbortkoppling ska ske så nära felstället som möjligt för att så få kunder som möjligt ska påverkas av strömavbrott. Linjereläbrytare (recloser eller autorecloser) på 10-20 kV-näten är ett effektivt sätt att begränsa antalet påverkade kunder vid fel eftersom sådana brytare kan placeras på valfri plats i nätet. Med en linjereläbrytare erhålls samma funktion som med ett linjeskydd, de är i likhet med dessa konstruerad för att bryta vid överbelastning, kortslutning och jordfel. Eftersom linjereläbrytarna installeras ute i friledningsnät kan därmed större nätavsnitt delas upp i mindre bitar rent skyddsmässigt. När dessa installeras i befintliga nät behöver dock reläskyddsinställningar i angränsande nätdelar ses över för bibehållen selektivitet. Skellefteå Kraft har idag 9 stycken linjereläbrytare installerade i nätet och har planer att inom de närmsta två åren installera ytterligare 19 stycken.     Målet med rapporten var att ta fram en uppdaterad selektivplan med skyddsinställningar för allt 10 kV-nät som under normal- och reservdrift matas från TT3215 Norsjö, där det installerats en recloser. Syftet med arbetet var sedan att utreda skillnad mellan felbortkopplingstider och möjlighet att uppnå selektivitet vid användning av konstanttid- och inverstidsfördröjning av överströmsskydd.   Examensarbetet har genomförts i flera steg. Första steget var att genom litteraturstudie skapa en teoretisk grund för vidare arbete samt ta reda på hur nuvarande praxis för skyddsinställning på Skellefteå Kraft ser ut. Andra steget var att genomföra den nätanalys och de beräkningar som var nödvändiga för att kunna ta fram en rekommendation av skyddsinställningar. I det tredje steget bestämdes skyddsinställningar enligt två olika strategier. Inställningsstrategi A enligt Skellefteå Krafts nuvarande praxis med konstanta tider för alla skydd och inställningsstrategi B med förändringen att istället använda inverstidsfördröjning av överströmsskydden (se avsnitt 4.2–4.3). Slutligen gjordes en analys av skillnaden mellan de två strategierna (se avsnitt 5).   Analysen visar att det finns en skillnad mellan felbortkopplingstider vid jämförelse mellan konstant- och inverstidsfördröjning. För de grupper där linjeskyddet vid normal- eller reservdrift sitter i serie med linjereläbrytare F-Arnberg-RS kommer det vid skyddsinställningar med inverstidskarakteristik (jämfört med konstanttid) innebära en snabbare felbortkopplingstid vid högsta förekommande felström. Felbortkopplingstiden reduceras med 0,26–0,30 s beroende på val av inverstidskarakteristik. För resterande linjeskydd (och linjereläbrytare) erhålls samma eller en långsammare felbortkopplingstid vid maximal felström. Analysen visar inte någon skillnad i möjlighet att uppnå selektivitet mellan inställningsstrategi A och B, detta eftersom de rekommenderade skyddsinställningarna är valda för att god selektivitet alltid ska uppnås.

Abstract:
Students enrolled in accounting courses often struggle because of the complexity of the topic. Accounting instructors have searched for effective means of fostering student success, but the learning process continues to change. Critical thinking and problem solving abilities are vital for students and future professionals. Thus, teaching should not be limited to the transmission of information. By moving the dissemination of basic knowledge outside the classroom inversed learning allows class time for deep dives into complex topics and hands-on activities. Students who are actively involved in learning tend to be more successful in the classroom. The purpose of this study was to determine how undergraduate students in an intermediate accounting course respond to an inversed classroom structure as it relates to financial accounting. A quasi-experimental, quantitative approach was used to investigate whether the academic performance of students who received instruction in a flipped classroom significantly differed from students who received instruction in a traditional classroom. Subgroups of students within the treatment group were examined to determine their response to the intervention. The study was completed over 2 semesters. Participants were determined by pre-existing groups. Students enrolled in an intermediate accounting course during the spring 2018 semester received instruction in a traditional manner. Students enrolled in the same course during the fall 2018 semester were taught using the inversed model. The researcher for this study also served as the instructor for both groups. Academic achievement was measured by student performance on four exams administered during each semester. Six research questions were addressed using MANOVA, ANOVA, and multiple regression analyses. The results indicated students generally perform better in the inversed learning environment than in the traditionally formatted classroom. Although the comparisons were not statistically significant, students in the flipped classroom did achieve higher scores on 3 of the 4 exams. No significant interaction was found between the classroom environment and gender or learning style. Both college GPA and gender were found to be significant predictors of academic performance. The findings from this study may support faculty in the enrichment of college curriculum by promoting active learning.

És sabut de fa molt temps que les inversions cromosòmiques son mantenides per selecció natural a les poblacions de Drosophila. No obstant, els mecanismes moleculars que generen aquest valor adaptatiu encara no es coneixen. A les poblacions naturals de D. buzzatii, la inversió 2j forma un polimorfisme equilibrat amb l’ordenació 2st, en què els individus 2j tenen una mida més gran y un temps de desenvolupament més llarg en comparació amb els 2st. En aquest treball hem posat a provat la hipòtesi de que un efecte de posició d’un dels punts de trencament podria ser la causa d’aquests canvis fenotípics. Per a fer-ho hem analitzat l’expressió d’un gen adjacent al punt de trencament proximal, CG13617, en línies de D. buzzatii amb i sense la inversió 2j. Hem trobat que els embrions 2j presenten un nivell d’expressió de CG13617 cinc vegades menor causat per un RNA antisense originat en una còpia d’un transposó de la família Galileo inserit al punt de trencament. Les conseqüències funcionals de la reducció de l’expressió de CG13617 s’han investigat utilitzant la tècnica de RNA interferència per reproduir aquest silenciament a D. melanogaster. Els experiments de microarrays i RT-PCR en temps real comparant larves de primer estadi amb i sense expressió de CG13617 han revelat que 41 gens mostren nivells d’expressió reduïts quan CG13617 és silenciat, mentre que cap gen presenta un increment. A més, hi ha un excés significatiu de gens implicats en la replicació del DNA i el cicle celular entre els afectats pel silenciament de CG13617. Nou de deu d’aquests gens van ser analitzats a D. buzzatii i també tenen un nivell d’expressió reduït en embrions 2j, però no en larves de primer estadi, una fase en què la diferència d’expressió de CG13617 entre ordenacions cromosòmiques és menor i el RNA antisense ja no es transcriu. Per a esbrinar la possible funció d’aquest gen hem dut a terme un exhaustiu anàlisi de seqüències nucleotídiques i proteiques en els 12 genomes de Drosophila disponibles i també en altres organismes. La proteïna CG13617 conté un dit de zinc tipus C2H2 molt conservat, tres regions que poden formar coiled coils, dues seqüències PEST i senyals de localització i exportació nuclear. També presenta similitud amb la proteïna humana DZIP1 i amb Iguana, un component de la via de senyalització Hedgehog al peix zebra, fet que indica que el seu paper dins la cèl·lula podria estar relacionat amb el transport de factors de transcripció cap a dins i cap a fora del nucli. Aquests resultats suggereixen que el gen CG13617 podria estar implicat en la regulació de la replicació del DNA i que l’efecte de posició en els portadors de la inversió 2j podria contribuir a explicar les diferències fenotípiques observades entre els individus 2st i 2j, així com el valor adaptatiu d’aquesta inversió.
Chromosomal inversions have been known for a long time to be maintained by natural selection in Drosophila populations. However, the molecular mechanisms underlying their adaptive value remain uncertain. In D. buzzatii natural populations, inversion 2j forms a balanced polymorphism with the 2st arrangement, in which 2j individuals have a larger size and a longer developmental time compared to 2st carriers. In this work we tested the hypothesis that a position effect of one of the inversion breakpoints could be the cause of these phenotypic changes by analyzing the expression of a gene adjacent to the proximal breakpoint, CG13617, in D. buzzatii lines with and without inversion 2j. We have found that in 2j embryos an antisense RNA originated in a copy of a Galileo family transposon inserted at the breakpoint causes a 5-fold decrease of the expression level of CG13617. In order to investigate the functional consequences of the reduction in CG13617 expression, we have used RNA interference to reproduce this silencing in D. melanogaster. Microarray and real-time RT-PCR experiments comparing first instar larvae with and without CG13617 expression revealed that 41 genes show reduced expression levels when CG13617 is silenced, while none is up-regulated. Interestingly, genes involved in DNA replication and cell cycle are significantly enriched among those affected by CG13617 silencing. Nine out of ten of these genes analyzed in D. buzzatii also show a reduced expression level in 2j embryos, but not in first instar larvae, a stage where the CG13617 expression difference between chromosomal arrangements is lower and the antisense RNA is no longer transcribed. To gain insight into the potential function of this gene we have carried out a comprehensive nucleotide and protein sequence analysis in the 12 available Drosophila genomes and also in other organisms. CG13617 protein contains a conserved C2H2 zinc finger, three coiled coil regions, two PEST sequences, and putative nuclear localization and export signals, and shows similarity to human DZIP1 and zebrafish Iguana (a component of the Hedgehog signaling pathway) proteins, which indicates that its cellular role could be related to the transport of transcription factors in and out of the nucleus. These results suggest that gene CG13617 could be involved in the regulation of DNA replication and that the position effect in 2j carriers might contribute to explain the phenotypic differences observed between 2st and 2j individuals as well as the adaptive value of the inversion.
