The objective of this work was to compare the bacterial diversity found in soils under eucalyptus with those found under native forest. Six areas belonging to Celulose Nipo-Brasileira - CENIBRA were studied; three of them were eucalyptus plantations and three were preserved native forests. They were located in two different projects of the company, the Crystal Lake and the Jacinto Lake, bearing two classes of soils, Latosol and Fluvic Neosol. Sampling was performed in March 2008 and bacterial diversity was assessed for the α-Proteobacteria, β-Proteobacteria, γ-Proteobacteria, Actinobacteria and Firmicutes phyla. Denaturing Gradient Gel Electrophoresis (DGGE) was carried out for genetic diversity analysis and the resolved gel images were analyzed with the GELCOMPARII® software (Applied Maths, Inc - Texas USA). Shannon Weaver diversity indexes were calculated by means of the software Diversity of Species v. 2.0 (W.C Rodrigues - Lizaro Soft - Entomologists of Brazil - Brazil) The highest diversity indexes for total bacteria (Eubacteria), β-Proteobacteria, γ-Proteobacteria, and Actinobacteria was found in soils of native forests. Soils under eucalyptus forests displayed the highest diversity index for α- Proteobacteria and Firmicutes. The phylum α-Proteobacteria was the dominant one in Latosol and Neosol, both under eucalyptus and native forests. Diversity among Firmicutes and γ-Proteobacteria was also expressive; however, diversity of Actinobacteria and β-Proteobacteria was lowest in all areas studied. Diversity indexes averages among all phyla, when classes of soils were compared, were higher for Latosols, except for those of Firmicutes. The highest Eubacteria diversity index in soils under native forests as compared to those displayed under eucalyptus corroborates the hypothesis that plant coverage determines shifts in the microbial communities diversity.

Full textShigella invades cells of the intestinal epithelium by injecting effector proteins through a Type III Secretion System (T3SS). The type III effector IpaA binds to talin and vinculin through three Vinculin Binding Sites (VBSs), promoting cytoskeletal anchorage to promote bacterial internalization. As opposed to other invasive bacteria, Shigella does not show constitutive cell binding activity but triggers transient adhesion through T3SS activation. IpaA VBS3 was found to be structurally analogous to talin H5 helix, which, together with H1-H4, forms the R1 talin bundle. Functional analysis identified IpaA VBS3 is necessary for talin recruitment to Shigella entry foci and formation of nascent adhesions and filopodia. IpaA VBS3 also stimulates filopodial capture of bacteria and stabilizes cell adhesions in invaded cells. On the other hand, structural models of IpaA VBSs/Vinculin interactions indicate a novel mode of vinculin activation, involving conformational changes in vinculin head domain that leads to trimerization and stabilization of focal adhesions. IpaA was also observed to be injected by "kissing-and-running" bacteria, facilitating additional bacterial binding and invasion events. This IpaA-dependent priming was dependent on IpaA VBS3 but not on IpaA VBS1-2. Injected IpaA was observed to form clusters apposed to the cell apical side and to induce integrin clustering. Overall, the results indicate that IpaA triggers general changes in cell adhesive properties and promotes a cooperative mechanism of Shigella invasion
Itodo, Anthony Onoja. "An assessment of the effect of participation on sustainable development in a rural electrification project : a case study of the Ipari-Efugo project." Thesis, Stellenbosch : Stellenbosch University, 2008. http://hdl.handle.net/10019.1/20444.
Full textENGLISH ABSTRACT: Due to inadequate financial resources and lack of managerial skills, in this case study the local level of governance does not have appropriate mechanisms to manage their resources adequately. It is observed that the Nigerian government cannot provide all basic amenities for the people at the grass-root level of development. Following the above scenario, rural communities do not have access to basic amenities such as electricity and pipe borne water. In Nigeria, little research has been done with regard to sustainable development in rural electrification. This study is based on the impact of community and individual participation in a rural electrification project, and shows how sustainable development plays a leading role. The study is based on a rural electrification project initiated and facilitated in 1996, and was completed in 2000 by the Ipari-Efugo Otukpa community members in Benue State in the North Central part of Nigeria. In spite of prevailing poverty in rural areas, Ipari-Efugo community members were able to be responsible for their own development without government intervention. The research includes literature studies of rural electrification projects and demonstrates how participation and sustainable development can lead to a successful community project. Irrespective of obstacles such as poverty and lack of economic empowerment, Ipari- Efugo community members are resilient and were able to provide electricity. Access to electricity can serve as a prerequisite for economic development and growth. The benefits accruable from the use of electricity outweigh the costs of providing the amenity to the community. Participation through sustainable development remains the core instrument responsible for the completion of the electricity project in Ipari-Efugo. This study encourages rural communities to take responsibility for their own development.

Full textFragmentation and habitat loss are the two main causes of population and biodiversity decline, followed by overhunting. Such impacts made the Cracidae family the most endangered bird family in American continents. In Brazil, many reintroduction programs focused on cracids. The first one reintroduced the Red-billed Curassow (Crax blumenbachii Spix, 1825) in Ipaba, MG, Brazil. This work aimed to estimate the population size and density, the sex ratio and the landscape use of the Red-billed Curassow population reintroduced in Ipaba. We collected the data along 6 months through line-transects, totalizing 280 km of effort. The analysis followed distance sampling and occupancy protocols. We estimated the population size in 34 (10 111) birds and the density was 2.36 (1.58 7.79) birds/ha. However, we estimated the proportion of area effective used in 21%, what means that the density for such area might be higher. Apparently, the intensity of habitat use is mostly correlated to the understory density and to food supplementation. We estimated the population growth rate in -0.047 (-0.137 0.018), what indicates a decrease. Its possible that the population is declining due to its size, once small populations are more likely to suffer because of demographic, environmental and genetic stochasticity and Allee effect. The same factors might explain the skewed sex ratio, estimated in 0.35 (0.20 0.47).

Background: Post-operative pain has been identified by patients as being one of the most distressful experiences associated with surgery. Traditionally, post-operative pain was poorly controlled because of insufficient medical resources. Over the past decades, however, extensive efforts have been put forth to improve the quality of pain relief provided. Uncontrolled post-operative pain is harmful to patients, and optimal pain control can enhance recovery following surgery. Optimal pain control can result in reduced perioperative morbidity and facilitate an earlier return to function after surgery. In Taiwan, where one million operations are performed annually, over 12% of postoperative pain relief is managed by patient-controlled analgesia (PCA). In 1995, i-Pain was developed in collaboration with physicians and engineers as a total quality pain management solution on acute pain related service, technology, communication, finance, and research. Market and demand: In collaboration with i-Pain®, the PCA vendor (Hospira Inc.) has dramatically increased its Taiwanese PCA market share from 65% to 95% during 2002-2009 and has subsequently eliminated its competitors by setting up an insurmountable entry barrier. Currently, this clinician-driven service network is utilized in 10 medical centers throughout Taiwan and proves to be an extremely valuable data mine to continually improve the quality of post-op analgesia provided and to reduce analgesia-related side effects. The growing demand for value-added service in Taiwan and China has led to the global business model on i-Pain® system. Products and core value: Our core value is to provide a comprehensive product portfolio for patients, clinicians and vendors within the entire value chain. For patients, sentinel@i-Pain® accesses multiple charts and reports while guard@i-Pain® monitors the level of pain on a real-time basis and delivers the desirable drug on any given consumable application. For clinicians, desktop@i-Pain® automatically synchronizes documentation while mobile@i-Pain® communicates the bedside information with the cloud server anytime and anywhere. For vendors, web@i-Pain® is an efficient online package for logistical and financial management. Strategic positioning: Our core competence is the synergistic leverage of a clinician’s expertise, i-Pain’s extensive R&D capabilities, and the vast economy of scales provided by our vendors. We are capable of becoming the world’s leading portal design house in the acute pain industry. We will focus on innovating new consumable products while our i-Pain inside® will serve as the ERP system (Enterprise Resource Planning) for all customers. Business model: Our business model is to enhance customer’s demand through developing value-added information and consumables. The profitability of network service is primarily paid by B2B royalties that are based exclusively on the sale of the consumables and based on consumer utility. Furthermore, the design-house vertically integrates and efficiently facilitates the lengthy R&D process through rapid product design, fast prototyping and clinical trials, complete certification, and cost-effective manufacturing. Marketing Strategy: Our short-term plan is to enroll all PCA devices into the i-Pain® network in Taiwan. The academic and managerial benefits across the centers and platforms can be readily demonstrated by the advantages to automation, as well as the vast amount of data collection and processing afforded by the i-Pain® network. The intermediate plan is to form a strategic alliance with the PCA vendor’s marketing team. Our successful experience and launch in the Taiwan market could serve as a springboard into the Chinese market. Our long-term plan is to develop a globally oriented platform covering both network service and design. We would create value-added knowledge through data mining, processing and interpretation as well as define the standard features of PCA devices and associated consumables.

Exploring service quality attributions in coach services on national freeway by applying Kano model and IPA − a case of Kuo Kuang Motor Transport Company Student: Wan-Ru Pan Advisor: Dr. Hsin-Hung Wu Graduate School of Marketing and Logistics Management National Changua University of Education ABSTRACT This study chose Kuo Kuang Motor Transport (KKMT) Company as a case, applied SERVQUAL construct as a basis to develop the survey with 25 items, and incorporated Kano two-dimensional model to evaluate the service quality of coach services on national freeway. Each service quality characteristic is to beevaluated to examine if the two-dimensional quality characteristic exists and then each characteristic is to be classified into one of the five quality elementsfrom customers’ viewpoints. It attempts to segment markets and discuss the differences among different clusters by Kano two-dimentional model and analysis of variance. In addition, perceived importance-performance analysis (P-IPA) was proposed and developed based upon importance-performance analysis (IPA) and revised IPA with benchmarking philosophy to analyze the service performance and relative performance compared with Ubus of KKMT Company. Moreover, by integrating Kano model into P-IPA, the competitive advantages and weaknesses of service quality characteristics can be further prioritized and improved by effective decisions. By the findings, Kuo Kuang Motor Transport Company performs better than Ubus in ten items with high importance. However, Ubus outperforms Kuo Kuang Motor Transport Company by two items, which require immediate improvement by KKMT Company. According to Kano model analysis, there are 25 quality items, 17 of them are one-dimensional quality elements, and 4 items belong to must-be quality elements, and, finally, 4 items are classified as attractive quality elements. The results of this study can provide insights for KKMT Company to improve its service quality and enhance its competitiveness in the marketplace. Keywords: Kano model, importance-performance analysis, satisfaction, service quality, Kuo Kuang Motor Transport Company
