Abstract:
In this study, the performance of a new most probable number (MPN) test (Pseudalert®/Quanti-Tray®) for the enumeration of Pseudomonas aeruginosa from hospital waters was compared with both international and national membrane filtration-based culture methods for P. aeruginosa: ISO 16266:2006 and UK The Microbiology of Drinking Water – Part 8 (MoDW Part 8), which both use Pseudomonas CN agar. The comparison based on the calculation of mean relative differences between the two methods was conducted according to ISO 17994:2014. Using both routine hospital water samples (80 from six laboratories) and artificially contaminated samples (192 from five laboratories), paired counts from each sample and the enumeration method were analysed. For routine samples, there were insufficient data for a conclusive assessment, but the data do indicate at least equivalent performance of Pseudalert®/Quanti-Tray®. For the artificially contaminated samples, the data revealed higher counts of P. aeruginosa being recorded by Pseudalert®/Quanti-Tray®. The Pseudalert®/Quanti-Tray® method does not require confirmation testing for atypical strains of P. aeruginosa, saving up to 6 days of additional analysis, and has the added advantage of providing confirmed counts within 24–28 hours incubation compared to 40–48 hours or longer for the ISO 16266 and MoDW Part 8 methods.

Abstract:
Pseudomonas aeruginosa, a food- and water-borne opportunistic pathogen, constitutes a health risk mostly to immunocompromised patients, and also affects the taste, odour and turbidity of potable water. In order to detect P. aeruginosa in bottled water in Sri Lanka, 36 bottled water brands were collected randomly from retail and supermarkets island wide. P. aeruginosa was detected by the membrane filtration technique, using cetrimide agar supplemented with nalidixic acid. The isolates were subjected to confirmatory tests, viz: ISO 16266:2006 methods and growth at 4 and 42°C, and API 20NE followed by DNA sequencing. Presumptive isolates of P. aeruginosa were observed on cetrimide agar in 50% of brands. Among these isolates, ISO procedures confirmed 58% as P. aeruginosa. Thirty-nine randomly selected isolates were identified as P. aeruginosa by the API 20NE, three of which were further confirmed by DNA sequencing. The presence of P. aeruginosa in bottled water raises health concerns since P. aeruginosa is an indicator of inferior water quality. Therefore, strict regulations and regular monitoring of bottling plants are recommended in order to supply safe and acceptable bottled drinking water to the Sri Lankan market.

Abstract:
This study analyses issues concerning defects of passive paint coats. Four reasons of their occurrence have been identified: constructional, technological, operational, and caused by wear and tear. Standardized methods of paint coat assessment are discussed based on the impact of mechanical factors (PN-EN ISO 2409:2013-06, ASTM D3359-17, PN-EN ISO 1522:2008, PN-EN ISO 4624:2016-05, PN-EN ISO 1518-1:2011, PN-EN ISO 16276-2:2008, PN-EN ISO 6272:2011, PN-EN ISO 6860:2006, PN-EN ISO 2815:2004, PN-EN ISO 1519:2000). The original method of paint coat durability evaluation is presented with the consideration of synergetic impact of mechanical forces. This method permits carrying out testing in conditions close to those during normal service. The testing stand equipped with a rotating drum filled with mineral mass has been described as well as the optical method of assessing paint coat quality using original software developed by authors. Tests for three coating systems were carried out in order to present the possibilities of the developed method. In summary, the conclusions relating to the presented testing method are presented.

Abstract:
A qualidade da água de consumo é uma preocupação e uma questão de saúde pública, são cada vez mais os agentes patogénicos transmitidos através da água, sendo por isso do interesse público os requisitos cada vez mais específicos para o controlo da água de consumo. A água para o consumo humano é um recurso cada vez mais escasso que deve ser protegido e tratado como tal. Embora o acesso à água potável esteja a melhorar em todo o mundo, existe ainda um grande número de pessoas que não tem acesso à água tratada, como tal o risco de contaminação por parte de agentes patogénicos é muito maior. De acordo com a legislação portuguesa DL 306:2007, para água destinada ao consumo humano (rede de distribuição, fontanários, reservatórios, etc.) existem valores obrigatórios para Escherichia coli e Enterococcus, contudo em águas engarrafadas ou em outros recipientes existem valores paramétricos para mais microrganismos sendo, E. coli, Enterococcus, Pseudomonas aeruginosa, e o número total de colónias que se desenvolvem a 22 °C e a 37 °C. O presente trabalho foca-se na implementação da norma ISO 16266:2006, normativo, para a deteção e enumeração de Pseudomonas aeruginosas, através do método de filtração de membrana, incubada depois num meio especifico e quando necessário, a realização dos testes de confirmação. O estágio foi realizado na empresa SGS. Portugal, e teve também como objetivo a pesquisa de Pseudomonas aeruginosa em 10 fontes de águas para potencial consumo humano, com aplicação da ISO 16266:2006. Com base nos resultados obtidos, nas normas legislativas e na recomendação da ERSAR nº1/2017, do DL 306-2007, o método encontra-se validado e implementado, na SGS Portugal, S.A. Por outro lado, as águas analisadas demonstraram resultados negativos para a presença de P. aeruginosa, contudo não se pode afirmar segurança das mesmas para consumo humano.
