95 %) still it is noteworthy because R. oryzae produces only the L (+) form of lactic acid which can be metabolized in the human body. The yield of an industrial process should be high for the feasibility of the production of a particular product. If a way can be found increase the flux through the glycolysis the yield of lactic acid may increase as well. Keeping this in mind we wanted to focus on the first step of glycolysis, hexokinase of R. oryzae. Hexokinase catalyzes the reaction that converts glucose to glucose-6-phosphate. In this study for the first time the two isoenzymes of hexokinase of R. oryzae were purified and characterized by biochemically and kinetically Hexokinase has two isoenzymes. The purified enzymes (isoenzymes1 &
isoenzymes2) obeyed Michealis-Menten Kinetics. The Km value of purified isoenzyme 1 is 0.16 mM and isoenzyme 2, 0.21 mM at pH 7.70 for glucose. The Km value of isoenzyme1 for fructose was 28.8 mM. Essentially isoenzyme 2 can not utilize fructose. None of the isoenzymes were inhibited by trehalose-6-phophate.The monomer moleculer weight of isoenzymes were estimated SDS PAGE analysis. Full textA high rate (70%) of adventitious shoot regeneration was observed from leaf-petiole explants of micropropagated shoot cultures of 'Comet' red raspberry cultured on modified Murashige-Skoog medium containing 1 mg/l thidiazuron (TDZ) and 0.5 mg/l 1H-indole-3-butanoic acid (IBA). Variation in the agar concentration or incubation temperature, orientation or scoring of the leaf-petiole explants and use of separate leaf or petiole explants had no effect on shoot regeneration while incubation in the dark for 1, 2, or 3 weeks prior to growth in the light depressed the number of adventitious shoots formed. Only 8 of 22 raspberry cultivars were capable of regenerating from leaf explants of greenhouse-grown plants.

Full textOwing to its ability to hydrolyze ATP and to the associated conformational changes, myosin is the most important protein of muscle contraction. Pyrophosphate gel electrophoresis under non denaturing conditions separates rat ventricular myosin in three isoforms V1, V2 and V3 in order of decreasing electrophoretic mobility and ATPase activity, V1 being the predominant form in the adult animal. During chronic hemodynamic overload of the rat ventricle, a myosin isoenzymic redistribution is observed in parallel with ventricular hypertrophy, with an accumulation of the V3 isoform and a parallel decrease in the V1 isoform. This redistribution is, at least in part, responsible for the decrease in the speed of muscle shortening observed with hypertrophy. Moreover, improving the efficiency of muscle contraction, it can be regarded as a truly adaptational mechanism. The myosin isoenzymic redistribution is also observed in chronic overload of the human auricle, also predominantly composed of a “V1-like” isoform. By contrast, the human ventricle already almost exclusively composed of a “V3-like” isoform, is scarcely concerned by this phenomenon. Myosin isoenzymic redistribution observed with chronic hemodynamic overload represents the first example of the regulation of myocardial muscle contractility by differential expression of a family of multigenic proteins

Full textTo obtain markers suitable for estimation of genetic variability of the basidiomycetes Agaricus bisporus and Pleurotus species, our study was focused on isozyme and genome polymorphism in this species. Analysis of nine activities (6 oxidoreductases and 3 hydrolases) in P. Cornupiae has showed that isozyme polymorphism (defined by the number of isozyme detected by activity) and isozyme variability (represented by the number of zymograms) were related for most of the activities. However, two oxidoreductases showing a low polymorphism have revealed a high variability in P. Cornupiae as in A. Bisporus. This result should be related in the involvement of these two activities in morphogenesis. Analysis of restriction fragment length polymorphism (RFLP) of ribosomal (rDNA) showed for the first time in basidiomycetes, existence of two types of rDNA units in the same species. Each type is defined by specific restriction sites. Length variations of the intergenic region between the 25S and 5S genes were evidenced for each type. Two rDNA units of the same type but having length variations can coexist in the same strain. We have showed that length variability could result from deletions of direct repeat sequences, constituting intergenic region. A specific probe of the unit rDNA of P. Cornucopiae showed a high level of heterogeneity of rDNA in 11 Pleurotus species and allowed to develop a numeral analysis for Pleurotus systematics. In A. Bisporus, study of nuclear and mitochondrial RFLP markers allowed to obtain (i) for the first time in this species evidence of genetic diversity by using a ribosomal marker, (ii) relationship between nuclear polymorphism and industrial strain phenotypes as well as detection of high level of heteromorphism between haploid nuclei of some varieties, and (iii) distinction of two mitochondrial types Isozyme and RFLP markers which have been characterized by this work constitute an important contribution to varietal characterization and genetic improvement of the cultivated species A. Bisporus and F. Cornucopiae

Full textEdible bananas are supposed to be originated from the two species M. Acuminata COLLA and M. Balbisiana COLLA. A long vegetative propagation has led to a wide diversity among cultivated forms. The genetic organization and the evolution of the crop is not completely elucidated through morphological and cytogenetical data. A chemotaxonomic study is undertaken : diversification in the genus Musa is analyzed by the use of two biochemical markers, flavonoids and isozymes. The polymorphism of flavonoids (anthocyanins and flavonol glycosides) is examined among 80 varieties of bananas and leads to the proposal of an evolutionary scheme. M. Acuminata and M. Balbisiana have choosen two distinct phenolic pathways. Observed results point out the high level of differentiation of acuminata species, and leads to recognize an evolutive hierarchical organization. The analysis of cultivars suggests that domestication did occur at several evolutive stages during differentiation of M. Acuminata. The analysis of enzyme diversity among 113 wild types and cultivars, through 8 polymorphie loci, displays the great divergence between M. Acuminata and M. Balbisiana species. The diversity within M. Acuminata species is high and the observed structure reveals a compartimentalization in all likelihood related to geographical isolation. These results are in good agreement with those obtained through flavonoid polymorphism. Comparison between wild acuminata and cultivars shows that domestication did act on a reduced genetic basis. Classifications obtained through the two kinds of markers globally corroborate previous classifications based on morphophysiological data. Isozyme electrophoresis applied to bananas turns out to be an efficient and promising tool, as well in genetic resources investigations as for classification and breeding
