Abstract:
Bakteri asam laktat (BAL) merupakan mikroflora alami yang terdapat di dalam saluran pencernaan organisme. Penelitian ini bertujuan untuk mengetahui isolat BAL yang mampu menghambat perkembangan bakteri V. harveyi. Metode penelitian terdiri dari isolasi bakteri, uji morfologi, uji biokimia dan uji fisiologi (uji ketahanan pH,uji daya hambat dan uji BSLT). Hasil penelitian mendapatkan total isolat sebanyak 10 isolat dengan 4 isolat terpilih. uji daya hambat mendapatkan aktivitas hambat tertinggi terhadap bakteri uji V. harveyi ditunjukkan oleh isolat B1.P4, B2.P4 dan B4.P4 sebesar 10 mm. uji BSLT mendapatkan isolate dengan nilai toksisitas terendah ditunjukkan oleh isolat B2.P4 dan B5.P4 dengan nilai regresi sebesar 880, 71. Hasil identifikasi menunjukkan isolat B1.P4 adalah jenis bakteri Leuconostoc sp, isolate B2.P4 adalah Bacillus sp. dan isolat B4.P4 dan B5,P4 adalah jenis Lactococcus sp. Berdasarkan hasil uji daya hambat dan uji BSLT dapat disimpulkan bahwa B4.P4 dan B5.P4 memiki potensi terbaik sebagai probiotik.

Abstract:
Marine water which has been polluted by heavy metals such as lead (Pb) from port activities can affect marine life, however several microorganisms are able to adapt in polluted waters due to their resistant capabilities. The research aimed to obtain isolates bacterial which are resistant to Pb and to test their resistant ability. Bacterial isolation was obtained from sediment and seawater taken in Paotere Port, Makassar. The isolation of the resistant bacteria was done using nutrient agar media which contained 10 ppm of Pb. The growth of the bacteria was initially observed macroscopically and microscopically to determine type of the colony. Resistant test for each type of isolate was done by inoculating them on nutrient broth media which contained different concentration of Pb: 5 ppm, 10 ppm, 15 ppm, 20 ppm, 25 ppm, and a control. The test resulted in eight different isolates, five isolates from sediment samples and three from seawater samples. The results showed that each isolate has different degree of resistant toward Pb. Isolate IA1 has the higher resistant ability, and it was followed by isolate IS5.---Perairan laut tercemar logam berat timbal (Pb), yang bersumber dari aktifitas pelabuhan, dapat mengganggu kehidupan biota laut, namun sejumlah mikrorganisme mampu beradaptasi karena memiliki kemampuan resistan. Penelitian ini bertujuan untuk memperoleh isolat bakteri yang resistan terhadap Pb dan menguji kemampuan resistansinya. Isolasi bakteri diperoleh dari sampel sedimen dan air laut dari Pelabuhan Paotere, Makassar.Untuk isolasi bakteri resistan menggunakan media nutrient agar yang megandung 10 ppm Pb. Bakteri yang tumbuh diamati secara makropskopis dan mikroskopis sebagai pendekatan awal untuk mengetahui jenis isolat. Uji resistansi dilakukan untuk setiap jenis isolat, yang diinokulasikan pada media nutrient broth pada konsentrasi Pb sebesar 5 ppm, 10 ppm, 15 ppm. 20 ppm, 25 ppm dan 0 ppm sebagai kontrol. Hasilnya, diperoleh delapan jenis isolat yang berbeda, yaitu lima jenis isolat dari sampel sedimen dan tiga dari sampel air laut. Dari hasil uji memperlihatkan, bahwa setiap isolat memiliki kemampuan resistansi berbeda-beda terhadap Pb. Isolat IA1 kemampuan resistansinya lebih tinggi, kemudian isolat IS5.

Abstract:
Abstrak. Penelitian ini bertujuan untuk mengisolasi dan mengkarakteristik fungi lignoselulolitik dari limbah sawit sebagai pendegradasi serat (senyawa polisakarida). Penelitian ini menggunakan metode eksplorasi melalui isolasi, karakteristik, uji degradasi lignoselulosa dan identifikasi. Isolasi menggunakan medium selektif yang dimodifikasi mengandung carboxy methyl cellulose (CMC), xylan, lignin dan manan. Dari hasil isolasi diperoleh 16 isolat fungi lignoselulolitik dan 4 diantaranya memiliki kemampuan tinggi dalam mendegradasi lignoselulosa yaitu isolate fungi YLF2, YLF3, YLF4 dan YLF8. Isolat fungi yang diperoleh memiliki karakteristik yang bervariasi meliputi bentuk, permukaan, tepi dan warna koloni. Hasil uji degradasi (hidrolitik) menunjukkan bahwa isolat fungi YLF8 menghasilkan indeks hidrolitik lebih tinggi dibanding fungi lainnya. Berdasarkan hasil dapat disimpulkan bahwa isolat fungi YLF8 termasuk pada strain Trichoderma sp berpotensi sebagai isolat pendegradasi serat dan dapat digunakan sebagai bioktivator dalam fermentasi pakan berserat.Isolation And Characteristic Of Lignocellulolitic Fungi Of Palm Waste As a Fiber Feed Degrading AgentAbstract. This study aims to isolate and characterize lignocellulolytic fungi from palm wastes as fiber degradation (polysaccharide compounds). This research uses exploration method through isolation, characteristic, lignocellulosic degradation test and identification. Isolation using modified selective medium contains carboxy methyl cellulose (CMC), xylan, lignin and manan. From isolation result obtained 16 isolates of lignocellulolytic fungi and 4 of them have high ability in degrading lignocellulose that is fungi YLF2, YLF3, YLF4 and YLF8. The obtained fungi isolates have varying characteristics including shape, surface, edges and colony color. The result of degradation test (hydrolytic) showed that YLF8 fungi isolates yielded higher hydrolytic index than other fungi. Based on the results it can be concluded that the isolates of YLF8 fungi belong to the Trichoderma sp strain potentially as fiber degrading isolates and can be used as bioctivators in fibrous fermentation feed.

ABSTRAK.Bakteri Pereduksi Sulfat (BPS) berperan dalam biodegradasi lahan tercemar. Identifikasi berdasarkan karakter fenotipik perlu dilakukan untuk mengetahui jenis bakteri ini. Penelitian ini bertujuan untuk mendapatkan beberapa isolat bakteri pereduksi sulfat di lokasi pemandian air panas Saronsong kota Tomohon. Identifikasi dilakukan dengan menggunakan metode profile matching. Bakteri termofilik pereduksi sulfat diisolasi dari lokasi pemandian air panas Sarongsong kota Tomohon menggunakan media cair postgate b yang telah dimodifikasi dan selanjutnya dilakukan pemurnian menggunakan metode pengenceran. Karakter fenotipik isolat bakteri termofilik pereduksi sulfat yang diuji meliputi morfologi koloni, morfologi sel dan karakter biokimia. Hasil isolasi pada kawasan air panas Sarongsong diperoleh 4 isolat bakteri termofilik pereduksi sulfat yang dilihat dari perubahan warna media selektif. Berdasarkan identifikasi dengan menggunakan metode profile matching menunjukkan bahwa keempat isolat tersebut diduga termasuk dalam anggota genus Desulfotomaculum (isolat AK4D3 dan L4D1), genus Desulfomicrobium (isolat AJ4D5) dan genus Desulfobulbus (isolat L5D2).Kata Kunci:, fenotipik, identifikasi, isolasi, karakterisasi ABSTRACT. Sulfate Reducing bacteria play a role in biodegradation of land contaminated. Identification based on phenotype character needs to be done to know this type of bacteria. The objective of this research is to get some isolate of sulfate reducing bacteria in clear water, cloudy water and mud in Tomohon hot spring water location. Identification is done by using profile matching method. The sulfate reducing thermophilic bacteria was isolated from the Tomohon Sarongsong hot spring location using the modified postgate b liquid medium and then purified by dilution method. Phenotypic characteristics of sulfate reducing thermophilic bacteria isolates tested included colony morphology, cell morphology and biochemical character. The result of isolation at Sarongsong hot bath obtained 4 isolates of sulfate reducing thermophilic bacteria seen from selective media color change. Based on the identification using the profile matching method showed that the four isolates were suspected to be included in the members of the genus Desulfotomaculum (isolates of AK4D3 and L4D1), genus Desulfomicrobium (isolate AJ4D5) and genus Desulfobulbus (isolate L5D2). <p>ABSTRACT</p><p><br />White spot syndrome virus (WSSV) is the most dangerous shrimp pathogen and give a very bad impact to the shrimp industry in many countries, including Indonesia. Furthermore, the vaccine usage is less effective since the local WSSV suspected to have some mutation and have differences in sequence from the one that used for recombinant vaccine production so far. Therefore, it is necessary to produce recombinant vaccine for immunization of local shrimp according to WSSV isolates from Indonesia. Viral protein 19 (VP19) is an important WSSV envelope proteins because of its involvement in the systemic infection of shrimp, so it can be developed as a recombinant vaccine. The objectives of this study are to obtain VP19-encoding gene of WSSV from Situbondo, Indonesia. This research conducted through several stages, DNA genome isolation of Penaeus monodon infected with WSSV, amplification of VP19-encoding gene, isolation of VP19-encoding gene, ligation of VP19-encoding gene into pGEM-T Easy vector, transformation to Escherichia coli, recombinant vector screening, vector isolation using mini preparation technique, vector verification, and sequencing. The result of cloning shows that the size of VP19-encoding gene from Situbondo isolates is similar to VP19-encoding gene from the abroad one, which is 366 bp. However, the sequence result shows that there are some differences on bases and amino acid. However, the result of sequencing shows there are difference of five base and three amino acid arranged VP19 sequence the Situbondo isolate with isolate from several countries. The similiarity of the Situbondo isolate sequence with the other isolate (from Indonesian and other countries) was only 98%. This difference indicates the importance of making a vaccine from a local WSSV genes especially Situbondo isolate.<br />Keywords: white spot syndrome virus, VP19, Penaeus monodon</p><p><br />ABSTRAK</p><p><br />White spot syndrome virus (WSSV) merupakan patogen yang paling serius menyerang udang windu dan telah menghancurkan industri udang windu di berbagai negara, termasuk di Indonesia. Penggunaan vaksin WSSV yang didesain berdasarkan isolat WSSV luar negeri kurang efektif karena diduga WSSV yang menyerang udang Indonesia telah mengalami mutasi dan memiliki perbedaan sekuen. Untuk itu, penggunaan isolat WSSV asal Indonesia, khususnya Situbondo sebagai cetakan dalam memproduksi vaksin rekombinan untuk imunisasi udang di Indonesia sangat perlu dilakukan. Viral protein-19 (VP19) merupakan protein pembungkus WSSV yang penting karena keterlibatannya dalam infeksi sistemik pada udang, sehingga dapat dikembangkan sebagai vaksin rekombinan. Tujuan penelitian ini adalah untuk mendapatkan gen penyandi protein permukaan VP19 virus WSSV isolat Situbondo. Tahapan penelitian yang dilakukan pada penelitian ini adalah isolasi DNA genom udang windu yang terinfeksi WSSV, amplifikasi gen penyandi protein VP19, isolasi gen penyandi protein VP19, ligasi gen penyandi protein VP19 ke dalam plasmid pGEM-T Easy, transformasi plasmid, skrining plasmid rekombinan, isolasi plasmid dengan teknik mini preparation, verifikasi plasmid dan pengurutan. Hasil pengklonan menunjukkan bahwa ukuran gen penyandi protein VP19 isolat Situbondo sama dengan ukuran gen penyandi protein VP19 isolat asing yaitu 366 pb. Namun hasil pengurutan menunjukkan adanya perbedaan lima basa dan tiga asam amino penyusun sekuen VP19 isolat Situbondo dengan isolat dari beberapa negara lain. Kemiripan sekuen isolat Situbondo dengan isolat lain (dari Indonesia sendiri maupun dari negara-negara lain) hanya sebesar 98%. Adanya perbedaan ini mengindikasikan pentingnya pembuatan vaksin dari gen WSSV lokal khususnya isolat Situbondo.</p><p>Kata kunci: white spot syndrome virus, VP19, Penaeus monodon</p>

Abstract:
Tujuan penelitian ini adalah menguji kemampuan sinergisme fungi selulolitik asal limbah jagung untuk dikonsorsiumkan sebagai bioaktivator pakan berserat. Isolate fungi selulolitik yang dikonsorsiumkan merupakan hasil isolasi dari limbah tanaman jagung yaitu isolate JB (Aspergillus sp.), JC (Fusarium sp.), JD (Aspergillus sp), JE (Rizoctonia sp.). Parameter yang diamati adalah sinergisme fungi selulolitik yang dikonsorsiumkan pada medium padat, pola interaksi antara fungi yang dikonsorsiumkan pada medium padat dan sinergisme fungi selulolitik yang dikonsorsiumkan pada medium cair (cocktail inokulum). Hasil penelitian menunjukkan bahwa dari konsorsium isolate fungi selulolitik diperoleh 5 pasangan isolat fungi selulolitik yang dapat bersinergi dan 1 pasangan tidak dapat bersinergi. Pola pertumbuhan yang terbentuk bersifat Mutual Intermingling (pasangan isolate JB vs JC dan JB vs JD), Partial Intermingling (pasangan isolate JB vs JE; JC vs JD; dan JD vs JE); dan Inhibition at touching point (pasangan isolate JC vs JE). Sinergisme fungi selulolitik yang dikonsorsiumkan pada medium cair (cocktail inokulum) menunjukkan tingkat pertumbuhan yang berfluktuatif seiring bertambahnya waktu fermentasi. Kesimpulan: dari 5 pasangan isolat fungi selulolitik yang dikonsorsiumkan diperoleh 1 pasang isolate yang tidak dapat bersinergis, dan sinegisme antara fungi membentuk pola interaksi yang bervariatif. Pertumbuhan fungi selulolitik pada medium cair berfluktuatif dan menunjukkan tingkat pertumbuhan optimum pada hari ke-3.

Abstract:
ABSTRAK Penelitian eksplorasi ini bertujuan untuk mengisolasi dan mengidentifikasi senyawa metabolit sekunder dari ekstrak etil asetat kulit batang mangrove Sonneratia caseolaris yang diperoleh dari daerah pinggiran Sungai Tallo Kelurahan Paccerakkang, Kota Makassar, Sulawesi Selatan. Isolasi dilakukan dalam beberapa tahap yaitu maserasi, partisi, fraksinasi, uji kemurnian dan identifikasi. Hasil penelitian berupa isolat murni berbentuk serbuk berwarna cokelat muda yang terdekomposisi pada suhu 140 0C. Pengujian dengan FeCl3 menunjukkan bahwa isolat positif flavonoid. Hal ini didukung oleh beberapa data spektrum FTIR pada isolat yang menunjukkan bilangan gelombang (cm- 1) yakni: 3380.93 (OH), 2954.95 (OH asam karboksilat), 1697.36 (C=O asam karboksilat), 1606.7 (C=C alkena); 2926.01 dan 2852.72 (CH3 dan CH2); 1359.82 (NO2); 1446.61 (C=C aromatik), 1211.3; 1188.15 dan 1037.7 (C-O alkohol) dan 1107.14 (C−O aril eter). Kata kunci: Isolasi, Etil Asetat, Sonneratia caseolaris, Flavonoid ABSTRACT This exploratory research have aim to isolate and identification the secondary metabolite compound contained in the etil acetate extract bark of Sonneratia caseolaris from Tallo River side, Paccerakkang district, Makassar City, South Sulawesi. Isolation were doned in several stages, were maceration, partitioning, fractionation, purity testing and identification. The result was obtained of pure isolate, it was light-brown powder with decompotition of 1400C. Identification result with FeCl3 test showed this isolate was flavanoid compound. Abstract:
Abstract Uncaria gambir Roxb., one of native plants of Indonesia contains high levels of catechins. Catechins are very potential to be used for medicinal raw materials because theirs effects are proven to be antibacterial, antiviral, and antidyslipidemic. Derivatization of catechin can be developed to produce drug compounds that are effective as antiviral agents for HIV. Derivatization process needs pure catechin isolate as a starting material in order to obtain maximum result so that the isolation process is one of the important stages. In this study, isolation and purification of catechin isolate from Uncaria gambir Roxb. extract was carried out, which will then be the starting material in the derivatization of catechin. Gambir extract used for catechin isolation was a quality 1 gambir extract obtained from Padang, West Sumatra. Gambier extract is characterized according to the standard method stated in Farmakope Herbal Indonesia. Isolation of catechin from gambir extract was done by percolation method using ethyl acetate solvents. Pure catechin isolate was obtained using a vacuum liquid chromatograpy method with a series of solvent hexane and ethyl acetate gradients. Catechin purification was monitored using Thin Layer Chromatography (TLC) method with eluent chloroform:ethyl acetate:formic acid (5:4:1, then identified using High Performance Liquid Chromatography (HPLC), Nuclear Magnetic Resonance (NMR) Spectroscopy, and Liquid Cromatography- Mass Spectroscopy (LC-MS). The purity of catechin isolate obtained was 99.80%± 0.132. Abstrak Uncaria gambir Roxb., salah satu tanaman asli Indonesia yang mengandung katekin dengan kadar yang tinggi. Katekin sangat potensial digunakan untuk bahan baku obat karena efeknya terbukti sebagai antibakteri, antivirus, dan antidislipidemia. Derivatisasi katekin dapat dikembangkan untuk menghasilkan senyawa obat yang efektif sebagai antivirus untuk HIV. Untuk derivatisasi ini diperlukan isolat katekin murni sebagai starting material agar diperoleh hasil yang maksimal sehingga proses purifikasi isolat merupakan salah satu tahap yang penting. Dalam penelitian ini dilakukan isolasi dan pemurnian isolat katekin dari ekstrak Uncaria gambir Roxb. yang selanjutnya akan menjadi bahan awal dalam derivatisasi katekin. Ekstrak gambir yang digunakan untuk isolasi katekin adalah ekstrak gambir kualitas 1 yang diperoleh dari Padang, Sumatera Barat. Ekstrak gambir dikarakterisasi sesuai dengan metode standar yang tertera dalam Farmakope Herbal Indonesia. Isolasi katekin dari ekstrak gambir dilakukan dengan metode perkolasi menggunakan pelarut etil asetat. Isolat katekin murni diperoleh menggunakan metode kromatografi cair vakum (KCV) dengan serangkaian gradien pelarut heksana dan etil asetat. Pemurnian katekin dimonitor menggunakan metode kromatografi lapis tipis (KLT) dengan eluen kloroform : etil asetat : asam format (5:4:1), kemudian diidentifikasi menggunakan High Performance Liquid Chromatography (HPLC), spektroskopi Nuclear Magnetic Resonance (NMR), dan Liquid Cromatography-Mass Spectroscopy (LC- MS). Kemurnian isolat katekin yang didapatkan 99,80%± 0,132.

Nudibranch has different of bacteria and bioactive compounds contained therein for protect theselves from predators.The aim of this study were to isolate symbiotic bacterial drived from Thurudilla lineolata and Phyllidiella pustulosa, and to observe the antibacterial activity of bacterial isolate against Escherichia coli and Staphylococcus aureus. Isolation and culture of the symbiotic bacterial were made on Nutrient Agar. Nudibranch memiliki berbagai jenis bakteri dan senyawa bioaktif yang terkandung di dalamnya untuk melidungi diri dari predator. Tujuan dari penelitian ini yaitu untuk mendapatkan isolat bakteri yang bersimbion dengan Thrudilla lineolata dan Phyllidiella pustulosa, kemudian mengamati aktivitas antibakteri dari isolat bakteri tersebut terhadap Escherichia coli, dan Staphylococcus aureus. Isolasi dan kultur bakteri yang bersimbion dengan Thrudilla lineolata dan Phyllidiella pustulosa, dibuat pada media Nutrient Agar. Skrining aktivitas antibakteri menunjukkan isolat bakteri yang bersimbion dengan Thrudilla lineolata dan Phyllidiella pustulosa mampu menghambat pertumbuhan organisme uji Staphylococcus aureus dan Escherichia coli. Abstract:
<p>Tujuan dari penelitian ini adalah melakukan uji aktivitas antifungal dari ekstrak kulit mangga yang telah disemprot dengan asam salisilat ke dalam kapang dan khamir perusak buah mangga yang diperoleh dari proses isolasi. Dari proses isolasi diperoleh tujuh isolat kapang dan khamir yang diberi kode isolat A, B, C, D, E, F, dan G, dan setelah dimurnikan diperoleh empat isolat (isolat A, B, D dan G). Perlakuannya adalah tingkat kepekatan ekstrak kulit mangga, ada 5 tingkatan yaitu ekstrak yang dievaporasi hingga volumenya ½ nya (ekstak A), dievaporasi hingga 1/3 nya (ekstrak B), dievaporasi hingga ¼ nya (ekstrak C), dievaporasi hingga 1/5 nya (esktrak D) dan dievaporasi hingga 1/6 nya (ekstrak E). Sebagai kontrol adalah etil asetat teknis dan larutan benomyl 500 ppm. Uji aktivitas menggunakan metode sumur, uji resorsinol menggunakan HPLC. Penelitian diulang sebanyak tiga kali. Hasil aplikasi ekstrak pada agar plate menunjukkan bahwa berbagai konsentrasi ekstrak kulit mangga mampu menghambat semua jenis isolat kapang dan khamir, kecuali isolat D. Daya hambat terbesar ditunjukkan pada isolat G, mencapai 31 mm zona hambat yakni pada ekstrak kulit mangga yang dievaporasi hingga 1/6 nya (ekstrak E). Nilai ini lebih besar dari daya hambat oleh benomyl 500 ppm yang hanya 10 mm. Kadar resorsinol pada ekstrak antifungal ini adalah 5.012 ppm. Seven earlier isolates have been obtained (isolates A, B, C, D, E, F and G), further purification was obtained 4 isolates (isolates A, B, D and G). The treatments were based on density of the extract, they were evaporated to ½ volume extract (Extract A), evaporation until to 1/3 volume extract (extract B), evaporation to ¼ volume (extract C), evaporation to 1/5 volumes (extract D) and evaporation to 1/6 volume (extract E). As the control were ethyl acetate pure solution and benomyl 500 ppm solution. The research method, antifungal assay using gel well diffusion, determination of resorcinol content using HPLC. This study was three times repeated. Applications on the plate showed that the mango peel extract at various levels of concentration can inhibit the growth of all mold and yeast isolates except isolate D. The greatest inhibition zone indicated on mold isolates G. The mango peel extract which evaporated until the 1/6 volume (extract E) showed of 31 mm zone inhibition, this value is greater than Benomyl 500 ppm solution that is 10 mm. Resorcinol content of this antifungal extract is 5,012 ppm. The end result of the process of identification has been obtained that were Aspergillus niger for isolates A, Fusarium solari for isolates B, Penicillium sp for isolate D and Rhodotorula sp. for isolate G.</p><p>Keywords :mango peel extract, antifungal, isolate, mold and yeast, resorcinol</p><p> </p>

Sono stati analizzati i diversi meccanismi di polarizzazione elettrica, polarizzabilità delle molecole e modelli di campo in un dipolo molecolare proposti da alcuni ricercatori ed analizzate le perdite dielettriche che compaiono negli oli isolanti elettrici principali, suddividendole per perdite dipolari, da cui il confronto fra diversi modelli teorici, e perdite dovute alla conduzione ionica di portatori di carica mobili.

La prima soluzione studiata è una struttura autoportante in c.a con isolante eps, mentre la seconda è una struttura di irrigidimento con putrelle HEA 160 con applicato un pannello sandwich ed isolante PIR. Abstract:
Nel progetto di tesi magistrale si sono trattate le problematiche relative agli azionamenti a velocità controllata, correlandole soprattutto allo stress elettrico che questa configurazione comportava sulle macchine elettriche. Si sono introdotti tali sistemi suddividendoli in motore asincrono trifase, cavo di connessione ed elettronica di potenza, analizzando ognuno di questi elementi in dettaglio. Il secondo passo è stato analizzare il fenomeno dell’invecchiamento elettrico nei sistemi isolanti. Facendo quindi riferimento ai sistemi isolanti presenti nelle macchine asincrone si sono analizzate le forme d’onda, l’invecchiamento elettrico e le varie sollecitazioni a cui esso viene sottoposto. Le sollecitazioni sono state suddivise in meccaniche, termiche ed elettriche con particolare attenzione proprio a quest’ ultime. In particolare si è studiato l’impatto della tensione applicata (ampiezza, frequenza, duty-cycle, polarità e rise time), dei fenomeni di risonanza e del fenomeno di innesco delle scariche parziali. Unitamente al lavoro di ricerca è stato svolto un lavoro di sperimentazioni su cavi elettrici che andranno a costituire gli avvolgimenti presenti in macchine elettriche. L’ultima parte dell’elaborato riporta quindi la descrizione degli impianti di prova utilizzati per svolgere le prove (prove di vita e prove di innesco delle scariche parziali), la descrizione delle prove e la tabulazione dei dati sperimentali ottenuti. I dati ottenuti sono poi stati confrontati con dati ottenuti in precedenza da altri laureandi e sono state elaborate delle conclusioni a scopo comparativo.

Abstract:
La valutazione delle condizioni di esercizio dei cavi elettrici di bassa tensione è una priorità nella diagnostica dei componenti delle centrali nucleari, in quanto i sistemi di sicurezza si basano proprio su questi componenti. Gran parte di questi cavi vengono installati all’interno del contenimento dei reattori dove le difficili condizioni ambientali, in termini di temperature relativamente elevate, radiazioni gamma, presenza di umidità e sollecitazioni meccaniche possono portare alla degradazione dei materiali. I materiali polimerici che costituiscono l’isolamento e il rivestimento del cavo subiscono i maggiori effetti, diventando sempre più fragili e sempre meno in grado di operare con affidabilità e sicurezza. Questa tesi mira a studiare gli effetti dell’invecchiamento sulle caratteristiche del cavo, al fine di comprendere l’evoluzione delle proprietà elettriche e chimiche associate alla loro degradazione. Il polimero in oggetto è costituito da polietilene reticolato (XLPE) e additivato con antiossidanti (a base fenolica e di tioetere) e ritardanti di fiamma (ATH), invecchiati attraverso un processo accelerato con radiazioni gamma (70 Gy/h) e a una temperatura di 50 °C per un tempo massimo di 168 giorni. L’impatto dell’invecchiamento e il ruolo che giocano gli antiossidanti sui questi materiali viene monitorato con tecniche diagnostiche di spettroscopia dielettrica, misure delle correnti di polarizzazione e depolarizzazione, tempo di induzione all’ossidazione, FTIR e allungamento percentuale a rottura.

Il materiale studiato è il polietilene crosslinkato (XLPE), additivato in diverse concentrazioni tra antiossidanti a base fenolica, antiossidanti a base tioeterica e ritardanti di fiamma (ATH). Le tecniche di misura impiegate sono state la spettroscopia dielettrica, la conducibilità e il voltage breakdown in AC.

Negli ultimi decenni, l’elettronica di potenza grazie ad un ampio sviluppo dato dall’introduzione del DSP (Digital Signal Processor) come regolatore, ed ai progressi nella tecnologia di fabbricazione dei semiconduttori, si è introdotta in modo massiccio nel campo degli ASD (Adjustable Speed Drives) ovvero degli azionamenti a velocità controllata, rivoluzionando sostanzialmente, la tipologia di alimentazione dei motori elettrici che costituiscono tali azionamenti. Si è infatti passati da tecniche di controllo tradizionali, che consistevano nell’alimentare i carichi direttamente con la rete sinusoidale alternata a 50 o 60 Hz, ad alimentazioni indirette, cioè interponendo tra la rete e la macchina un convertitore elettronico di potenza, cosiddetto PEC (Power Electronic Converter), che fornisce tensioni impulsive, cioè caratterizzate da rapidi fronti di salita e di discesa. La natura impulsiva e quindi distorta di tali forme d’onda ha fondamentalmente aggravato la sollecitazione elettrica a cui sono sottoposti i materiali isolanti impiegati negli avvolgimenti dei motori elettrici, nasce quindi la necessità di verificare l’adeguatezza dei sistemi isolanti delle macchine elettriche operanti all’interno di un ASD.

Lo scopo della presente tesi, svolta presso il Laboratorio LAPLACE di Tolosa, è quello di indagare sulle proprietà ottiche di campioni composti da un substrato di materiale polimerico (bi-axially oriented polyprophilene, BOPP) ricoperto con diversi tipi di elettrodi principalmente tramite misure di elettroluminescenza e di comprendere come queste siano legate al suo deterioramento e invecchiamento Nella prima parte della tesi, verranno illustrate le misure effettuate su due diverse strutture MIM (metal-insulator-metal), la prima ottenuta utilizzando oro e la seconda utilizzando ITO come elettrodi. Nella seconda parte, invece, le misure sono state effettuate su campioni multistrato contenenti nano compositi a base di argento, ricoperte con uno strato di ITO che fungerà da elettrodo. Le misure sulla prima tipologia di campioni hanno evidenziato la presenza di due componenti principali nelle emissioni dovute a elettroluminescenza, una prodotta dal materiale bulk (BOPP) e una scaturita dall’interazione tra il substrato e l’elettrodo metallico, grazie al coinvolgimento dei plasmoni di superficie. Lo scopo della seconda parte della tesi è quello di comprendere l’effetto che le particelle di argento potrebbero avere sui plasmoni superficiali e sulle emissioni luminose dovute a elettroluminescenza. I risultati ottenuti su questi campioni hanno evidenziato un livello di luce prodotto da elettroluminescenza incredibilmente maggiore rispetto ai campioni aventi ITO e oro come elettrodi. In conclusione l’impatto delle nano particelle di argento sulle emissioni per elettroluminescenza da BOPP possono essere molteplici e saranno necessari ulteriori studi per comprendere in modo dettagliato tale meccanismo. L’effetto mostrato qui potrebbe risultare molto utile per capire meglio l’elettroluminescenza nei materiali polimerici isolanti da un lato e dall’altro le proprietà foto-fisiche delle nanoparticelle metalliche.

Lo studio intrapreso si è posto come obiettivo la caratterizzazione dal punto di vista elettrico dei materiali coinvolti nella realizzazione di accessori per applicazioni in HVDC, in particolare mescole isolanti, semiconduttive e varioresistive. La necessità di un lavoro di questo tipo viene giustificata dalla costante espansione dei sistemi in DC nella trasmissione dell’energia elettrica, i quali presentano caratteristiche fisiche che si differenziano sensibilmente da quelle tipiche dei tradizionali sistemi in AC, dunque richiedono componenti e materiali opportunamente progettati per garantire condizioni di servizio sicure e affidabili. L’obiettivo della trattazione consiste nello studio di analogie e differenze tra le proprietà elettriche fornite da prove su diverse configurazioni di provini, nella fattispecie di tipo piano e cilindrico cavo. In primo luogo si studiano i provini di tipo piano al fine di ricavare informazioni basilari sul materiale e sulla mescola che lo costituisce e di prendere decisioni relative al proseguimento dei test. Dopo aver effettuato un sufficiente numero di test su varie tipologie di provini piani e aver riconosciuto le mescole più performanti dal punto di vista elettrico, meccanico e termico, si procede alla realizzazione di provini cilindrici stampati, su cui si intraprendono le medesime misure effettuate per la configurazione piana. Questa seconda fase di caratterizzazione è fondamentale, in quanto consente di verificare che le proprietà già studiate su piastra si conservino in una geometria molto più simile a quella assunta dal prodotto finale evitando di sostenere costi onerosi per la produzione di un accessorio full-size. Il lavoro è stato svolto nel laboratorio elettrico di alta tensione della divisione R&D del gruppo Prysmian di Milano, leader mondiale nella produzione di sistemi in cavo per alte e altissime tensioni.

Abstract:
Cendawan tanah memegang peran esensial dalam budidaya pertanian berkelanjutan. Kemampuannya dalam memproduksi berbagai senyawa metabolit menjadikan cendawan sebagai salah satu agens hayati yang potensial untuk mengendalikan patogen tanaman. Salah satu metabolit sekunder cendawan yang bersifat anti-patogen adalah enzim ekstraseluler protease. Enzim ini dapat mendegradasi dinding sel cendawan patogen, nematoda, dan telur nematoda. Penelitian ini bertujuan untuk mengisolasi cendawan penghasil enzim protease dari rhizosfer padi dan mengkarakterisasi pertumbuhannya. Sampel tanah diambil dari 5 titik pengambilan sampel pada lahan padi petani di Kelurahan Mamburungan, Kota Tarakan, Kalimantan Utara. Isolasi dilakukan menggunakan media potato dextrose agar (PDA) pada suhu 37°C di Laboratorium Perlindungan Tanaman, Universitas Borneo Tarakan. Cendawan yang tumbuh dikarakterisasi bentuk makroskopisnya secara visual. Pengujian aktivitas proteolitik dilakukan menggunakan media PDA yang ditambah dengan skim milk. Seluruh cendawan yang tumbuh kemudian dikarakterisasi kecepatan tumbuhnya selama 11 hari. Hasil penelitian menunjukkan terdapat 7 isolat cendawan yang berhasil diisolasi, dan 3 (48,85%) diantaranya mampu menghasilkan enzim protease. Isolat yang mampu menghasilkan enzim protease adalah isolat C5, C6, dan C7. Selanjutnya, berdasarkan karakter pertumbuhannya, tujuh isolat yang diperoleh menunjukkan respon yang beragam. Pada hari ke-11, pertumbuhan tertinggi ditunjukkan oleh isolat C1, C2, C5, dan C6 dengan diameter 8,5 cm. Pertumbuhan paling lambat ditunjukkan oleh isolat C7 dengan diameter 2,53 cm. Penelitian ini memberikan informasi bahwa terdapat cendawan proteolitik dari rhizosfer padi pada lokasi penelitian dengan karakter pertumbuhan yang beragam.
