Abstract:
Regarding the accelerated innovation in the 21st century, the article suggests modifying the spatial principles of Research Institutes as isolated institutions. The 21st century Research Institute should reflect the increasing levels of openness and security in modern information exchange methods. Thus, the Institute reveals invisible information processes in the physical urban environment, becoming the urban centre of innovation, a new workplace and leisure centre, as well as a catalyst for enhancing the city’s sustainability. The article analyses the historical paradigm of effective methods for introducing innovation to the urban environment, as well as modern socio-economic needs of innovative research institutes in the city. As a result, a unique organizational structure and functional programme of a new Research Institute are suggested. The Institute directly participates in enhancing the urban environment and forming a new lifestyle of the city dwellers. New spatial principles of such buildings are also proposed, updating the architectural typology of Research Institutes in the 21st century. Due to the increased interaction with the urban environment, the Research Institute and the city are mutually transformed. This contributes both to increasing the Institute’s efficiency and raising the city’s economic potential and life quality.

Introduction. Acinetobacter spp has become an important cause of nosocomial infections due to its great ability to survive and spread in a hospital setting and to develop resistance to many antibiotics. The aim of this study was to examine the resistance to carbapenems and other commonly used antibiotics in strains of Acinetobacter isolated from wound swabs. Material and Methods. In the Laboratories of Microbiology Center at the Institute for Public Health in Novi Sad wound swabs were taken from the patients hospitalized at the Institutes and Departments of the Clinical Center of Vojvodina in Novi Sad. A total of 271 Acinetobacter spp strains were tested to susceptibility on carbapenems and other most commonly used antibiotics by disk diffusion method according to the recommendation of the Clinical and Laboratory Standards Institute. Results and Discussion. Acinetobacter spp (271 isolates) developed resistance to imipenem and meropenem (67.4% and 64.4%). The resistance to both cephalosporins III and IV generation and ciprofloxacin was 100%. The resistance to ampicillin-sulbactam was 71.4%. Conclusion. Our results show high resistance to carbapenems in Acinetobacter spp strains isolated from wound swabs. These facts suggest the need for continuous monitoring of susceptibility in order to take adequate measures to prevent and control spreading of resistant strains.

The clustering of isolates from both camps was confirmed by next-generation sequencing, as well as a secondary infection of a technician. The outbreak strain clustered with internationally isolated strains from patients with recent travel history to Central America. This report exemplifies enhanced surveillance and international collaboration between public health institutes by enabling to link local outbreaks to region-specific sublineages circulating abroad.

AbstractIn the rice-fallow system, dry root rot (DRR) is an emerging disease of mungbean (Vigna radiata (L.) R. Wilczek var. radiata) caused by the necrotrophic fungus Macrophomina phaseolina. The pathogen causes extensive production losses. In this study, the bioactivity of four Trichoderma harzianum isolates, namely Th-Dharwad, Th-Raichur, Th-Niphm, and Th-Udaipur procured from the Indian research institutes were evaluated against M. phaseolina of mungbean by the dual culture technique. The efficacy of these T. harzianum isolates were also compared with the effective fungicides such as thiram and carbendazim by the poison food method. Results showed that among the T. harzianum isolates, isolate of Th-Raichur was most effective, exhibiting 76.96% mycelial growth inhibition of the test pathogen. As compared to the thiram, carbendazim was more effective, and exhibited 100% mycelial growth inhibition of the test pathogen. In addition, carbendazim was also more effective than the isolate of Th-Raichur. In the sick pot experiment, mungbean seeds treated with Th-Raichur isolate showed a lower percent incidence of DRR (20%) than the untreated seeds (86.6%). The biological spectrum of Th-Raichur isolate was examined against M. phaseolina isolated from the different hosts such as urdbean and vegetable soybean, alongwith two other root pathogens, namely Fusarium solani of mungbean, and Sclerotium rolfsii of urdbean. The isolate of Th-Raichur showed maximum antagonistic activity against the pathogens M. phaseolina and F. solani of mungbean. Thus, Th-Raichur isolate can be used as a potential fungal biocontrol agent for the reduction of DRR in mungbean.

<p class="abstract"><strong>Background:</strong> Clavicle is one of the common bones to undergo fractures with incidence rate higher in second to third decade of one’s life as well as in elderly age. Management of these clavicle fractures have always been a subject of debate, where literature have been advocating both non-operative as well as operative methods.</p><p class="abstract"><strong>Methods:</strong> The present study was conceived to know the burden of these isolated clavicle fracture cases reporting to tertiary level healthcare institute of North India and to study about their management pattern.<strong></strong></p><p class="abstract"><strong>Results:</strong> During the five years study duration (2014-2018), a total of 38 patients had reported to the trauma centre of the institute. Out of total, males had more preponderance 77% (29) and incidences were reported more in younger population. All patients were managed well by opting conservative measures, besides deformity had been reported in 2 (5.26%) patients. 3 (7.9%) patients had reported with complaints of pain, weakness while doing work, fatigue and pain when lying on the affected shoulder None of the patient was managed by open reduction and fixation.</p><p class="abstract"><strong>Conclusions:</strong> The present study concludes that the number of patients having isolated clavicle fracture are not contributing to any sort of burden at tertiary level institutes and are not even time consuming.</p>

Background: Data about evolution of aphasia following stroke are rare and controversial especially following fibrinolysis. The aim of this study was to describe the early clinical patterns of isolated aphasia in consecutive stroke patients with or without thrombolysis. Methods: Clinical and radiological data of consecutive stroke patients were routinely entered in prospective registry. Patients were considered aphasic when NIHSS (National Institutes of Health Stroke Scale) item 9 >0. ‘Isolated aphasia' was defined by aphasic patients without motor limb deficit. We created a ‘composite language score' obtained by summing the NIHSS items 1b, 1c and 9, which reflects language-processing ability. Recovery of functions was evaluated as measured by global NIHSS, composite language score and language screening test (LAST) at baseline, H24 and day 7 (D7). ‘Mild deficit' was defined as global NIHSS <5. Results: A total of 100 consecutive patients met study criteria for isolated aphasia. Twenty-five underwent thrombolysis and 75 did not. There was no difference between the 2 groups concerning demographic characteristics, involved territories and presence of arterial occlusion, initial median NIHSS, composite language and LAST scores at entrance. Evolution was significantly better in thrombolysed patient for the 3 testings: NIHSS, composite language score and LAST at D7 (respective p = 0.0002; p = 0.01 and p = 0.004). Similar results were found when we focused on the subgroups of patients with initial ‘mild' deficits (p = 0.01; p = 0.0003 and p = 0.007). No symptomatic hemorrhagic transformation occurred following thrombolysis. Conclusion: These data strongly suggest that thrombolysis is safe and effective in patients with ‘isolated aphasia,' even if the global NIHSS score is <5.

<span>Handwriting recognition is a growing field of study in computer vision, artificial intelligence and pattern recognition technology aimed to recognizing texts and handwritings of hefty amount of produced official documents and paper works by institutes or governments. Using computer to distinguish and make these documents accessible and approachable is the goal of these efforts. Moreover, recognition of text has accomplished practically a major progress in many domains such as security sector and e-government structure and more. A system for recognition text’s handwriting was presented here relied on edge histogram descriptor (EHD), histogram of orientated gradients (HOG) features extraction and support vector machine (SVM) as a classifier is proposed in this paper. HOG and EHD give an optimal features of the Arabic hand-written text by extracting the directional properties of the text. Besides that, SVM is a most common machine learning classifier that obtaining an essential classification results within various kernel functions. The experimental evaluation is carried out for Arabic handwritten images from IESK-ArDB database using HOG, EHD features and proposed work provides 85% recognition rate.</span>

Nesse processo de expansão da educação de nível superior pelo interior paulista, convergiam fatores de ordem política, econômica e social vigentes no período, como também as mudanças trazidas pela nova conjuntura política instaurada no Brasil com o golpe militar de 1964 e a Reforma Universitária de 1968. Além da análise bibliográfica, fontes documentais, jornais, legislação e documentos oficiais, a pesquisa sustenta-se na interpretação de dados dos episódios selecionados da memória dos indivíduos participantes desta experiência, presentes nas entrevistas localizadas no CEDEM/UNESP. Os dados foram correlacionados a fim de evidenciar fragmentos da história dos Institutos Isolados de ensino superior no interior do estado de São Paulo, mais precisamente da Faculdade de Filosofia, Ciências e Letras de Marília. Com a criação da Faculdade de Filosofia, que tinha como proposta formar professores e pesquisadores unidos a uma tradição humanística, Marília conquista uma situação diferenciada com relação ao ensino superior no interior do estado de São Paulo como parte de um processo de interiorização da cultura. In addition to bibliographical analysis, documentary sources, newspapers, official legislation and documents, this research is based on interpretation of episodes data selected from participant individuals' memory who were involved in that experience, present in the interviews belonging to the CEDEM/UNESP. Data were correlated to make clear fragments of the history of the Isolated Institutes of higher education in the interior of the state of São Paulo, particularly those related to the Faculty of Philosophy, Sciences and Letters of Marília. Abstract:
The present study was aimed at extending our knowledge of the antioxidative properties of the milk protein products, whey protein isolate (WPI) and sodium caseinate (NaCas), in oil-in-water (O/W) emulsions rich in polyunsaturated fatty acids (PUFAs). In particular, the objective was to contribute to our understanding of the compositional and processing factors that influence the oxidative stability of protein-stabilised O/W emulsions. Linoleic acid (approximately 60 %) was used as the lipid for the oil phase (10.6 %). The emulsion samples were usually incubated at 50 °C to accelerate lipid oxidation. Lipid oxidation indicators were lipid hydroperoxides and headspace hexanal, determined by solid phase microextraction (SPME) combined with gas chromatography (GC). WPI- or NaCas-stabilised emulsions were prepared using a wide range of protein concentrations (0.5, 1.0, 2.0, 3.0, 4.0, 7.0 or 10.0 %) at two droplet sizes (d32 = 0.31 and 0.65 µm). In general, higher lipid oxidation levels were found for the larger droplet size. Increasing protein concentration led to a decrease in the lipid oxidation rate. The greatest decrease in lipid hydroperoxide levels (values after 4 h) occurred at up to 4.0 % protein concentration. The greatest decrease in hexanal levels (values after 24 h) occurred at up to 4.0 % protein concentration in WPI emulsions (0.31 µm). The hexanal levels were more independent of the protein concentration in the other emulsion types. The hexanal level decreased at protein concentrations > 4.0 % in NaCas emulsions (0.31 and 0.65 µm) and at protein concentrations > 7.0 % in WPI emulsions (0.65 µm). The difference between lipid hydroperoxide generation in emulsions with small and large droplet sizes decreased with increasing protein concentration. This effect was more pronounced in NaCas emulsions. In general, NaCas was a better inhibitor of lipid oxidation than WPI, but WPI appeared to be the better antioxidant at some droplet size/protein concentration combinations. The protein in the continuous phase, i.e. the unadsorbed protein, played an important role in lipid oxidation. In principal, the lipid hydroperoxide and hexanal levels showed the same development over the continuous phase protein concentration as over the protein concentration in WPI and NaCas emulsions (d32 = 0.31 µm). A low NaCas level in the continuous phase already led to a relatively low hexanal level, whereas a higher WPI level was required. When NaCas solution was added to a WPI emulsion or WPI solution was added to a NaCas emulsion, a synergistic antioxidative effect was observed. The high molecular weight fractions (molecular weight = 12000-14000) of WPI and NaCas contained pro-oxidative metal ions that contributed to lipid oxidation in the emulsions. An enrichment of NaCas emulsions with the low molecular weight fraction of NaCas (with a molecular weight = 12000-14000) notably inhibited lipid oxidation. An enrichment of WPI emulsions with the low molecular weight fraction of WPI (with a molecular weight = 12000-14000) also seemed to inhibit lipid oxidation, but the effect was not significant. The protein solutions were enriched with these fractions before emulsion preparation. Pure WPI solution or mixed WPI/NaCas (1:1, weight/weight) solution with 1.12 or 2.24 % protein concentration was heated at 84 °C for up to 40 min, cooled and then used to prepare emulsions. Lipid oxidation was generally not affected by the heat treatment or the degree of whey protein denaturation. However, at the lower WPI concentration, more hexanal was produced for the longer heating times (20, 30 and 40 min) and this appeared to be connected with the physical instability of the emulsions. Greater oxidative stability was found at the higher protein concentration and when the proteins were mixed, pointing to a possible synergistic antioxidative effect of WPI and NaCas. The addition of the free radical source 2,2’-azobis(2-amidinopropane) dihydrochloride (AAPH) greatly increased the oxygen uptake and the generation of lipid hydroperoxides in the emulsions. The oxidative stability increased with increasing protein concentration (1.0, 4.0 and 7.0 %). NaCas had a greater antioxidative effect than WPI. The inhibition of oxygen uptake appeared to be largely influenced by the free-radical-scavenging activity of the system, determined by the protein type and the protein concentration, as the radicals were produced linearly over time and oxygen was consumed linearly over time. It can therefore be concluded that free-radical-scavenging activity represents a major antioxidative mechanism of the milk proteins. Oxygen was consumed much faster in emulsions than in protein solutions when the same level of AAPH was incorporated. In a WPI (1.0 % protein) emulsion, much lower levels of protein hydroperoxides than of lipid hydroperoxides developed. This pointed to a much greater reactivity of linoleic acid than of the milk proteins with oxygen. In contrast, the exposure of WPI to oxidising linoleic acid in an emulsion (1.0 % protein) or to AAPH in aqueous solution led to oxidative damage of the whey proteins, indicated by the loss of amino acids. The loss of specific amino acids was different for proteins in the continuous phase or cream phase of an emulsion or in WPI solution. The present study confirms the antioxidative potential of WPI and NaCas and gives new insights into their functionality as oxidative stabilisers in O/W emulsions.

A utilização de vários materiais irá influenciar não só a acústica, mas também a sua ocupação, uma vez que estes podem ter características que não se adequam às atividades a desempenhar. A Sala de Exposições do Edifícios 2 do ISCTE-IUL é um elemento de estudo bastante complexo, uma vez que é necessária a introdução de materiais bastante porosos e pouco resistentes mecanicamente, difcultando a sua implementação e comprometendo a utilização do espaço. Deste modo, o trabalho desenvolvido estuda uma série de soluções possíveis na tentativa de encontrar a que melhor se adequa ao espaço, reduzindo ao máximo o impacto na arquitetura existente e garantindo o melhor conforto acústico possível dentro da legislação em vigor. Foram realizadas 14 soluções construtivas modeladas com recurso ao software Archicad e posteriormente analisadas pelo Ecotect para garantir a sua qualidade acústica. Em função das qualidades dos materiais propostos estes são implementados em diferentes áreas tentando retirar o máximo rendimento de cada um deles. Os resultados obtidos mostram a importância da utilização de absorção acústica em diferentes cotas, sendo bastante evidente a diminuição de conforto quando esta é feita em exclusivo a cotas mais elevadas. É possível também compreender que os materiais utilizados têm de ser aplicados em locais específcos para poderem ser preservados, pois a resistência ao impacto dos mesmos é bastante baixa.<br>The acoustic behavior of different spaces is related to their use. The use of various materials will not only influence acoustics, but also its occupation , since they may have characteristics that do not ft the activities to be performed. The Exhibition Hall of ISCTE-IUL Buildings 2 is a very complex element of study, since it is necessary to introduce very porous and mechanically poor materials, making them diffcult to implement and compromising the use of space. In this way, the work developed studies a series of possible solutions in the attempt to fnd the one that best suits the space, reducing as much as possible the impact on the existing architecture and ensuring the best acoustic comfort possible under current legislation. 14 constructive solutions were developed using Archicad software and subsequently analyzed by Ecotect to ensure its acoustic quality. Due to the qualities of the proposed materials these are implemented in different areas trying to extract the maximum yield of each of them. The results obtained show the importance of the use of an acoustic absorption at different levels, being very evident the decrease in comfort when this one is done in exclusive in higher levels. It is also possible to understand that the materials used have to be applied at specifc locations in order to be preserved because the resistance to impact is quite low.

When the decision is made to augment gene flow into an isolated population, managers must decide how to augment gene flow, when to start, from where to take the individuals or gametes to be added, how many, which individuals, how often and when to cease. Even without detailed genetic data, sound genetic management strategies for augmenting gene flow can be instituted by considering population genetics theory, and/or computer simulations. When detailed data are lacking, moving (translocating) some individuals into isolated inbred population fragments is better than moving none, as long as the risk of outbreeding depression is low.

Australia is quintessentially a maritime nation where sea travel and transportation have been vitally important. Despite being an island, Australia hasd never completely felt isolated, and the indigenous peoples were never cut off from the rest of the world. This article presents four case studies in order to provide insights into the types and extent of maritime archaeological research that has been conducted over more than three decades in Australia. One of the great influences of Australian maritime archaeology over the years has been the Australasian Institute for Maritime Archaeology. A drawback in Australian maritime archaeology is the lack of funding for academic research. Australia has developed legislation for the protection of the historic shipwreck component of its underwater cultural heritage.

The availability of choice of systemically active antifungal agents and the proliferation in the number of fungal species implicated in invasive disease have meant that clinicians are increasingly looking for guidance from clinical laboratory results to help select the most appropriate agent. There are now well-established and predictable patterns of innate in vitro resistance to one or more antifungal agents associated with many yeast and mould species. This chapter provides definitions for the most frequently used terminology and outlines some of the issues surrounding antifungal susceptibility testing with yeast and mould isolates. Reference methods published by the Clinical Laboratory Standards Institute (CLSI) and the European Committee on Antimicrobial Susceptibility Testing (EUCAST) are discussed. Both innate and emergent antifungal drug resistance are increasingly recognized as limiting factors in the selection of antifungal agents, and the epidemiology and mechanisms of resistance are described for each of the major classes of antifungal agent.

Rotor blades are the highest thermal-mechanical loaded components of gas turbines. Their service life is limited by interaction of creep, low cycle fatigue (LCF), high cycle fatigue (HCF) and surface attack. Because assurance of adequate HCF strength of the rotor blade is an important issue of the blade design the European project PREMECCY has been started by the European aircraft engine manufacturers and research institutes to enhance the predictive methods for combined cycle fatigue (CCF), as a superposition of HCF and LCF. Although today’s predictive methods ensure safe blade design, there are certain shortcomings of assessing fatigue life with Haigh or “modified Goodman diagrams”, such as isolated HCF assessment as well as uni-axial and off-resonant testing. HCF and LCF are considered without taking into account their interaction. PREMECCY is aimed to deliver new and improved CCF prediction methods for exploitation in the industrial design process. Beside development of predictive methods the authors are involved in the design and testing of advanced specimens representing rotor blade features. In this connection the paper presents a novel test specimen type and a unique hot gas rig for CCF feature test at mechanical and ambient representative conditions.

Glycoprotein Ib (GPIb) mediates von Willebrand factor-dependent platelet adhesion and participates in the resulting platelet activation process. In the present investigation, the primary structure of human platelet GPIb was studied. GPIb and its proteolytic fragment glycocalicin were purified to near homogeneity from human platelets by affinity chromatography using wheat germ agglutinin and anti-GPIb monoclonal antibody (D. Nugent, University of Washington) coupled to Sepharose. GPIba chain, β chain, and glycocalicin were isolated, reduced and carboxymethylated, and then fragmented by trypsin and S. aureus V8 protease. Peptides were isolated by HPLC and subjected to amino acid sequence analysis. Approximately 200 amino acid residues were identified. Affinity purified rabbit antibodies directed against the a chain, the ft chain, and glycocalicin were prepared and shown to be monospecific by Western blot analysis. Total RNA was prepared from human erythroleukemia cells grown in the presence of phorbol acetate. Poly(A)+ RNA was selected and used to prepare a cDNA library in λgt11. The library was screened with [125]I-labeled polyclonal antibody to glycocalicin. The clone with the largest cDNA insert was sequenced and shown to code for amino acid sequences corresponding to those determined by Edman degradation of glycocalicin. The predicted amino acid sequence contains at least six tandem repeats of 24 amino acids that are highly homologous with 13 repeats present in leucine rich α2 glycoprotein of human plasma. Another region in the protein contains a second repeat rich in threonine and serine, which shows some homology to a 9 amino acid repeat in the connecting region of human factor V. This region is probably the major site of attachment of clusters of O-linked carbohydrate in GPIbα. These results indicate that human platelet glycoprotein Ibα has a multi-domain structure composed of a number of repetitive sequences. Decision-making and judgement is at the centre of all academic activities and accordingly inherent risk will always exist, through the exercise of judgement, the operation of academic policies and procedures and through compliance. A normative model of academic risk assessment is proposed, based on three levels: isolated academic risk, repeated academic risk and systemic academic risk. This is followed by a proposed model for action according to the level of risk. Finally the operation of the model in our higher education institute is presented.

Bovine von Willebrand Factor (vWF) binds directly to human platelets and also to heparin-agarose. Cleavage of vWF with Protease I, a metalloenzyme isolated from the venom of the western diamondback rattlesnake, produces two major fragments with apparent Mr of 250 kD and 200 kD. The 200 kD fragment competes with native vWF for binding to the GPIb-associated vWF receptor on formalin-fixed human platelets and has weak platelet-agglutinating activity. It is composed of three polypeptide chains of apparent Mr of 97 kD, 61 kD, and 35 kD. Monoclonal antibodies #2 and H-9, which inhibit binding of vWF to a GPIb-associated receptor of platelets, recognize the 200 kD fragment.Modification of vWF with ^5x-la.beled. Bolton-Hunter reagent (I*-BHR) causes inhibition of platelet-agglutinating activity at very low levels of incorporation. Modification of less than 2% of the amino groups in vWF causes 50% loss of platelet agglutinating activity and a decreased affinity of vWF for binding to platelets. Labeling with I*-BHR does not block binding to heparin-agarose, even when 5-10% of the amino groups are modified. Differential labeling at pH 7.0 and pH 8.5, followed by proteolytic fragmentation with Protease I, suggests that it is the modification of amino groups on the 200 kD fragment which is responsible fpr the decrease in platelet binding activity. Modification of the 97 kD peptide chain is best correlated with this loss of platelet binding activity.Heparin inhibits the agglutination of human platelets by bovine vWF. The 200 kD fragment of vWF binds both to platelets and to heparin-agarose. These observations suggest that the heparin-binding and platelet-binding domains of vWF, although distinct from one another, reside in the same region of the vWF molecule. The platelet-binding domain contains a small number of very reactive amino groups which are required for vWF binding to human platelets.These studies were supported by a grant from the National Institutes of Health (#HL27993).

Abstract The Massachusetts Institute of Technology (MIT) Commission on Industrial Productivity, in their report Made in America, found that six recurring weaknesses were hampering American manufacturing industries. The two weaknesses most relevant to product development were 1) technological weakness in development and production, and 2) failures in cooperation. The remedies to these weaknesses are considered the essential twin pillars of CE: 1) improved development process, and 2) closer cooperation. In the MIT report, it is recognized that total cooperation among teams in a CE environment is rare in American industry, while the majority of the design research in mathematically modeling CE has assumed total cooperation. In this paper, we present mathematical constructs, based on game theoretic principles, to model degrees of collaboration characterized by approximate cooperation, sequential decision making and isolation. The design of a pressure vessel and a passenger aircraft are included as illustrative examples.

A large-scale base-isolated steel structural frame was tested at the shaking table laboratory of the National Research Institute for Earth Sciences and Disaster Prevention. These collaborative experiments featured auto-adaptive media and devices to enhance the performance of passive base isolation systems. The planning of these experiments involved determining appropriate device control methods, the development of a controllable damping device with fail-safe characteristics, and the evaluation of the performance of the controlled isolation system subjected to strong ground motion with pronounced near-field effects. The results of the planning study and their large-scale experimental confirmation provide guidelines for the development and implementation of auto-adaptive damping devices for full scale structures.

The UK Food Standards Agency (FSA) agreed with industry to reduce Campylobacter spp. contamination in raw chicken and issued a target to reduce the prevalence of the most contaminated chickens (those with more than 1000 cfu per g chicken neck skin) to below 10 % at the end of the slaughter process, initially by 2016. To help monitor progress, a series of UK-wide surveys were undertaken to determine the levels of Campylobacter spp. on whole UK-produced, fresh chicken at retail sale in the UK. The data obtained for the first four years was reported in FSA projects FS241044 (2014/15) and FS102121 (2015 to 2018). The FSA has indicated that the retail proxy target for the percentage of highly contaminated raw whole retail chickens should be less than 7% and while continued monitoring has demonstrated a sustained decline for chickens from major retailer stores, chicken on sale in other stores have yet to meet this target. This report presents results from testing chickens from non-major retailer stores (only) in a fifth survey year from 2018 to 2019. In line with previous practise, samples were collected from stores distributed throughout the UK (in proportion to the population size of each country). Testing was performed by two laboratories - a Public Health England (PHE) laboratory or the Agri-Food &amp; Biosciences Institute (AFBI), Belfast. Enumeration of Campylobacter spp. was performed using the ISO 10272-2 standard enumeration method applied with a detection limit of 10 colony forming units (cfu) per gram (g) of neck skin. Antimicrobial resistance (AMR) to selected antimicrobials in accordance with those advised in the EU harmonised monitoring protocol was predicted from genome sequence data in Campylobacter jejuni and Campylobacter coli isolates The percentage (10.8%) of fresh, whole chicken at retail sale in stores of smaller chains (for example, Iceland, McColl’s, Budgens, Nisa, Costcutter, One Stop), independents and butchers (collectively referred to as non-major retailer stores in this report) in the UK that are highly contaminated (at more than 1000 cfu per g) with Campylobacter spp. has decreased since the previous survey year but is still higher than that found in samples from major retailers. 8 whole fresh raw chickens from non-major retailer stores were collected from August 2018 to July 2019 (n = 1009). Campylobacter spp. were detected in 55.8% of the chicken skin samples obtained from non-major retailer shops, and 10.8% of the samples had counts above 1000 cfu per g chicken skin. Comparison among production plant approval codes showed significant differences of the percentages of chicken samples with more than 1000 cfu per g, ranging from 0% to 28.1%. The percentage of samples with more than 1000 cfu of Campylobacter spp. per g was significantly higher in the period May, June and July than in the period November to April. The percentage of highly contaminated samples was significantly higher for samples taken from larger compared to smaller chickens. There was no statistical difference in the percentage of highly contaminated samples between those obtained from chicken reared with access to range (for example, free-range and organic birds) and those reared under standard regime (for example, no access to range) but the small sample size for organic and to a lesser extent free-range chickens, may have limited the ability to detect important differences should they exist. Campylobacter species was determined for isolates from 93.4% of the positive samples. C. jejuni was isolated from the majority (72.6%) of samples while C. coli was identified in 22.1% of samples. A combination of both species was found in 5.3% of samples. C. coli was more frequently isolated from samples obtained from chicken reared with access to range in comparison to those reared as standard birds. C. jejuni was less prevalent during the summer months of June, July and August compared to the remaining months of the year. Resistance to ciprofloxacin (fluoroquinolone), erythromycin (macrolide), tetracycline, (tetracyclines), gentamicin and streptomycin (aminoglycosides) was predicted from WGS data by the detection of known antimicrobial resistance determinants. Resistance to ciprofloxacin was detected in 185 (51.7%) isolates of C. jejuni and 49 (42.1%) isolates of C. coli; while 220 (61.1%) isolates of C. jejuni and 73 (62.9%) isolates of C. coli isolates were resistant to tetracycline. Three C. coli (2.6%) but none of the C. jejuni isolates harboured 23S mutations predicting reduced susceptibility to erythromycin. Multidrug resistance (MDR), defined as harbouring genetic determinants for resistance to at least three unrelated antimicrobial classes, was found in 10 (8.6%) C. coli isolates but not in any C. jejuni isolates. Co-resistance to ciprofloxacin and erythromycin was predicted in 1.7% of C. coli isolates. 9 Overall, the percentages of isolates with genetic AMR determinants found in this study were similar to those reported in the previous survey year (August 2016 to July 2017) where testing was based on phenotypic break-point testing. Multi-drug resistance was similar to that found in the previous survey years. It is recommended that trends in AMR in Campylobacter spp. isolates from retail chickens continue to be monitored to realise any increasing resistance of concern, particulary to erythromycin (macrolide). Considering that the percentage of fresh, whole chicken from non-major retailer stores in the UK that are highly contaminated (at more than 1000 cfu per g) with Campylobacter spp. continues to be above that in samples from major retailers more action including consideration of interventions such as improved biosecurity and slaughterhouse measures is needed to achieve better control of Campylobacter spp. for this section of the industry. The FSA has indicated that the retail proxy target for the percentage of highly contaminated retail chickens should be less than 7% and while continued monitoring has demonstrated a sustained decline for chickens from major retailer stores, chicken on sale in other stores have yet to meet this target.
