Abstract:
ABSTRACT We designed a new vector system for creating a random mutant library with multiple integrations of DNA fragments into the Rhodococcus genome in a single step. For this, we cotransformed two vectors into Rhodococcus by electroporation: pTip-istAB-sacB regulates the expression of the transposase (IstA) and its helper protein (IstB) under the influence of a thiostrepton-inducible promoter, and pRTSK-sacB provides the transposable-marker DNA. Both are multicopy vectors that are stable in the host cells; transposition of the transposable-marker DNA occurs only after the induction of IstA/IstB expression. With the addition of thiostrepton, all cultured cells harboring the two vectors, irrespective of the volume, can be mutated by random insertion of the transposable-marker DNA into their genome. Among the generated mutants examined, 30% showed multiple (two to five) insertion copies. The multiple integrated DNA copies were stable in the genome for more than 80 generations of serial growth without the addition of any selective antibiotics. This system can also be used for integrating various copy numbers of stably maintained protein expression cassettes in the host cell genome to modulate the expression level of biologically active recombinant proteins. We successfully applied this system to integrate multiple copies of expression cassettes for proline iminopeptidase and vitamin D3 hydroxylase into the Rhodococcus genome and verified that the clones containing double or multiple copies of the integrated cassettes produced higher levels and showed higher enzymatic activities of the target protein than clones with only a single copy of integration.

ABSTRACT Tandemly repeated insertion sequence IS21, located on a suicide plasmid, promoted replicon fusion with bacteriophage λ in vitro in the presence of ATP. This reaction was catalyzed in a cell extract containing the 45-kDa IstA protein (cointegrase) and the 30-kDa IstB helper protein of IS21 after both proteins had been overproduced in Escherichia coli. Without IstB, replicon fusion was inefficient and did not produce the 4-bp target duplications typical of IS21.

Abstract:
Clinical decisions regarding the stability and osseointegration of mandibular implants positioned using the bone expansion techniques are conflicting and limited. The objective was to evaluate the stability of implants placed using 2 surgical techniques, selected according to the initial width of the mandibular posterior edentulous ridge, with D3 bone density, during a 12-week period. Fifty-eight implants in 33 patients were evaluated. Thirty-two implants in 24 patients were positioned using the osteotome expansion technique, and 26 fixtures in 17 patients were installed using the conventional drilling technique. The implant stability quotient values were recorded at weeks 0, 1, 2, 3, 4, 6, 8, 10, and 12 postsurgery and evaluated using analysis of variance, independent, and paired t tests. Calibrated according to the stability reading of a 3.3-mm diameter implant, the osteotome expansion group was associated with a lower bone density than the conventional group (64.96 ± 6.25 vs 68.98 ± 5.06, P = .011). The osteotome expansion group achieved a comparable primary stability (ISQb-0, P = .124) and greater increases in secondary stability (ISQb-12, P = .07) than did the conventional technique. A D3 quality ridge with mild horizontal deficiency is expandable by using the osteotome expansion technique. Although the 2 groups presented similar implant stability quotient readings during the study period, the osteotome expansion technique showed significant improvement in secondary stability. The healing patterns for these techniques are therefore inconsistent.

ABSTRACT From Bradyrhizobium japonicum highly reiterated sequence-possessing (HRS) strains indigenous to Niigata and Tokachi in Japan with high copy numbers of the repeated sequences RSα and RSβ (K. Minamisawa, T. Isawa, Y. Nakatsuka, and N. Ichikawa, Appl. Environ. Microbiol. 64:1845–1851, 1998), several insertion sequence (IS)-like elements were isolated by using the formation of DNA duplexes by denaturation and renaturation of total DNA, followed by treatment with S1 nuclease. Most of these sequences showed structural features of bacterial IS elements, terminal inverted repeats, and homology with known IS elements and transposase genes. HRS and non-HRS strains ofB. japonicum differed markedly in the profiles obtained after hybridization with all the elements tested. In particular, HRS strains of B. japonicum contained many copies of IS1631, whereas non-HRS strains completely lacked this element. This association remained true even when many field isolates of B. japonicum were examined. Consequently, IS1631 occurrence was well correlated with B. japonicum HRS strains possessing high copy numbers of the repeated sequence RSα or RSβ. DNA sequence analysis indicated that IS1631 is 2,712 bp long. In addition, IS1631belongs to the IS21 family, as evidenced by its two open reading frames, which encode putative proteins homologous to IstA and IstB of IS21, and its terminal inverted repeat sequences with multiple short repeats.

Black box testing techniques are important to test the functionality of the system without knowing its inner detail which makes sure correct, consistent, complete and accurate behavior or function of a system. Black box testing strategies are used to test logical, data or behavioral dependencies, to generate test data and quality of test cases which have potential to guess more defects. Black box testing strategies play pivotal role to detect possible defects in system and can help in successful completion of system according to functionality. The studies of five companies regarding important black box testing strategies are presented in this thesis. This study explores the black box testing techniques which are present in literature and practiced in industry as well. Interview studies are conducted in companies of Pakistan providing solutions to finance industry, which is an attempt to find the usage of these techniques. The advantages and disadvantages of identified Black box testing strategies are discussed, along with it; the comparison of different techniques with respect to most defect guessing, dependencies, sophistication, effort, and cost is presented as well.

Cuando este proceso de pruebas no se encuentra definido, se realiza de una forma rústica, conlleva a que el tiempo planificado en el mismo no se cumpla, el número de incidencias encontradas incremente, el nivel de estrés entre los desarrolladores aumente y el usuario deba esperar más tiempo del proyectado, lo que resulta en una inconformidad del cliente. Ante ello, es imprescindible tener un proceso de pruebas de software correctamente precisado e implementado para que de esta manera se pueda asegurar la calidad del producto software desarrollado y de esta forma aumenta la satisfacción del cliente incrementando la confianza en la empresa proveedora.

Abstract:
There are many methodologies in the field of software engineering that focus on the entire software development process. The software development process aims to provide a quality product that meets user requirements, and to deliver it on time as well as within budget. Testing is a tool for verifying the quality of software and serves as feedback in software development. Most software development methodologies are available only in English, they emphasize the actual software development and address testing only marginally. An accurately designed testing process however, may contribute to higher software quality and avoid potential problems resulting from the entire information system's failure. Current Czech methodology that focuses exclusively on testing has been developed either inside individual companies and thus is not accessible to the public, or is contained in graduate theses, where it was typically created by adapting and expanding the existing software development methodologies for testing within a particular company or team. The main objective of this thesis is to establish a methodology for testing in Czech, which is based on international practices and standards, which is not tailored to a specific organization and which can be used as guidance in defining test processes in various organizations, regardless of their specific focus. The proposed methodology is also implemented in the tool EPF Composer that enables administration of methodology and its publication in the form of the website. The thesis also pursues additional objectives. In order to verify the objective of developing a standardized testing methodology is met, the thesis strives to produce a subsequent evaluation methodology using a standardized process framework in order to improve the testing process as well as a summary detailing which areas may be the subject of further expansion methodology. The theoretical part of the thesis aims to evaluate software development methodologies in terms of testing, to provide an overview of standards related to testing and to describe selected standards, to clarify the purpose of certification in testing and to analyze the possibilities of utilizing materials to prepare for the ISTQB certification. The objectives were achieved by analyzing the available materials for ISTQB certification, analyzing software development methodologies, standards, the TMMi procedural framework and other resources that deal with testing as well as its methodologies and standards. The main contribution of this work is a testing methodology, which is based on the testing process as defined by the international organization ISTQB. It provides a methodological framework by interconnecting and expanding individual methodological elements. Elements of the methodology include working products, as specified by the methodology with respect to the IEEE 829 standard. The methodology does not depend on the needs of a particular company and may be easily adopted, managed and customized. It may be used as a starting point in creating a testing strategy in Czech companies focused on software development which are interested in approaching testing in compliance with international practices.

presente estudio tuvo como objetivo conocer la relación del mobbing desde la concepción de Hidalgo y Piñuel, y el bienestar psicológico desde el modelo de Sánchez Canovas, considerando que en la medida que las organizaciones logren disminuir los estados psico emocionales negativos de sus colaboradores asociados al centro de trabajo debido a comportamientos agresivamente sistemáticos que destruyen la autoestima, generando sentimientos de culpabilidad acompañados de temor y ansiedad ocasionados por el mobbing y que según estudios en otras realidades disminuyen la productividad con repercusiones nocivas en la producción, es necesario que tanto quienes ocupan los roles jefaturales como los propios compañeros y los subordinados tengan la oportunidad de entrenar sus habilidades sociales de forma que contribuyan a disminuir la multicausalidad del mobbing, ya que su presencia, genera: miedo, altera el bienestar psicológico, con el riesgo de desencadenar males psicosomáticos con carácter de cronicidad, afectando a la productividad. Para este fin se trabajó con la metodología descriptiva correlacional, y diseño transversal y se aplicó a una muestra de 108 docentes del nivel superior dos cuestionarios verificados dentro del rigor de la psicometría. Entre los principales resultados se tiene que un 37% de la muestra examinada sufre de mobbing en algún grado, de allí que hay correlación significativa con el análisis nominal del Chi cuadrado y cuando se efectúa la inferencia a coeficientes ,resulta, inversa negativa entre el mobbing y la percepción general del bienestar psicológico, en el análisis de dimensiones se encontró resultado similar con el bienestar subjetivo, material y laboral, por tanto se afirma que cuando el colaborador se siente víctima de mobbing disminuye su bienestar psicológico. Estos resultados muestran la necesidad de asumir políticas organizacionales donde realmente se valore y reconozca al trabajador como persona y miembro efectivo de un equipo transformador en los docentes del ISTFB de Lima con la intención de generalizar los resultados.

Members of ISTB and ISTA have developed a list of issues regarding ISTB Pump Implementation that have been identified during IST Program Reviews, Day to Day operation of IST Programs, and site assessments of IST Programs (including issues found during updates). Implementation of the ISTB requirements for pumps in commercial US plants has presented challenges over the last few years with all the changes in ISTB since the issuance of the 1995 Edition of the ASME OM Code through the 2006 Addenda. This paper will discuss issues related to IST implementation. Good practices will be discussed to help IST engineers improve their programs. Abstract:
This paper describes actions being taken by the ASME (formerly the American Society of Mechanical Engineers) Operation and Maintenance of Nuclear Power Plants (OM Code) Subgroup on Pumps (ISTB) to revise pump test requirements to be more clearly stated and provide more flexibility in performance of tests. Specifically, this paper addresses two aspects of pump test requirements that are in the process of being changed. • Instrumentation requirements for measurement of hydraulic parameters (approved by ISTB subgroup). • Variance around the fixed reference value for establishing pump-test conditions (approved by ASME and published in the 2012 Edition of the OM Code). The OM Code changes discussed in this paper are currently in various stages of approval and endorsement. Therefore, the information provided in this paper is subject to change as a result of the OM Code approval and U.S. Nuclear Regulatory Commission (NRC) endorsement processes. Smooth Running Pumps have been an Industry issue since 1988. This caused many nuclear plants to obtain a Relief Request to use alternative requirements than those specified in the tables in subsections ISTB or ISTF of the ASME OM Code. Code Case OMN-22, Smooth Running Pumps, was approved in January 2017 for pumps with very low reference value vibration levels. This Code Case specifies the alternative requirements that may be used in lieu of the applicable Code subsections. Paper published with permission.
