In developing a vaccine against Rhipicephalus appendiculatus, an important tick ectoparasite of livestock in Africa, a necessary first step is identification of antigens which give protective immunity. Antigen profiles of extracts of unfed immature and adult Rh. appendiculatus ticks and their fractions were compared by immunoblotting with sera raised against tick infestation and against whole or fractionated extracts. Powassan (POW) virus, the cause of human encephalitis in the northeastern U.S. and Canada, is transmitted by tick bite. Since the geographic and host distribution patterns of Ixodes dammini Spielman, et al, 1979 and POW virus overlap, the potential of this tick species to transmit POW virus was explored. Transmission experiments were conducted with hamsters and rabbits which fed immature and adult ticks, respectively, from a POW-free colony. Surveys were conducted to compare the seasonal tick prevalence and loads, and sero-prevalence of tick-borne diseases (TBD) in Nguni and non-descript cattle on the sweet and sour communal rangelands of the Eastern Cape Province. The tick species observed on both rangeland types were Rhipicephalus appendiculatus (71.0 %), Rhipicephalus (Boophilus) species (29.2 %) and Rhipicephalus evertsi evertsi (40.2 %). In the first study, we date the origin and divergence time of the Flavivirus genus. It has been argued that the first flaviviruses originated after the last glacial maximum.
