Abstract:
Currently, the effect of fermented milk on the T-helper 17 response in inflammatory bowel diseases (IBDs) is unknown. The aim of the present study was to evaluate the effect of milks fermented with Lactobacillus fermentum on the Th1/Th17 response in a murine model of mild IBD. Exopolysaccharide (EPS), lactic acid (LA), and total protein (TP) contents and bacterial concentration were determined. Male C57Bl/6 mice intragastrically received either raw (FM) or pasteurized (PFM) fermented milk before and during a dextran sulfate infusion protocol. Blood, spleen, and colon samples were collected at Weeks 6 and 10. IL-6, IL-10, and TNFα were determined in serum, and IL-17, IL-23, and IFNγ were determined in intestinal mucosa and serum. The FM groups did not differ in cell concentration, LA, or TP content (p > 0.05); FM-J28 had the highest EPS content. Spleen weight and colon length did not differ among the FM groups (p > 0.05). In the FM-J20 and PFM-J20 groups, IL-17 and IFNγ decreased, and the IL-10 concentration was enhanced (p < 0.05) at Week 6. IL-6, TNFα, IL-23, and IFNγ did not differ in serum and mucosa (p > 0.05), and IL-17 was lowest in FM-J28 and FM-J20. Therefore, FM appears to potentially play a role in decreasing the Th17 response. However, further studies are needed to elucidate the FM-mediated anti-inflammatory mechanisms in IBD.

Abstract:
We previously showed that microglial keratan sulfate (KS) was induced in amyotrophic lateral sclerosis. However, the functional roles of the glycan and its synthetic enzyme in neurodegenerative diseases, such as Alzheimer’s disease (AD), a progressive disorder, are unclear. In our study, KS modified with sialic acids having a molecular mass of 125–220 kDa and the carbohydrate sulfotransferase GlcNAc6ST1 were up-regulated in the brains of two transgenic mouse models (J20 and Tg2576) and the brains of patients with AD. GlcNAc6ST1-deficient J20 (J20/GlcNAc6ST1−/−) mice demonstrated a complete absence of the microglial sialylated KS. J20/GlcNAc6ST1−/− primary microglia showed an increased level of amyloid-β phagocytosis and were hyperresponsive to interleukin 4, a potent antiinflammatory cytokine. Moreover, J20/GlcNAc6ST1−/− mice manifested reduced cerebral amyloid-β deposition. GlcNAc6ST1-synthesizing sialylated KS thus modulates AD pathology. Inhibition of KS synthesis by targeting GlcNAc6ST1 may therefore be beneficial for controlling AD pathogenesis.

Abstract:
Impaired neurovascular coupling has been suggested as an early pathogenic factor in Alzheimer’s disease (AD), which could serve as an early biomarker of cerebral pathology. We have established an anaesthetic regime to allow repeated measurements of neurovascular function over three months in the J20 mouse model of AD (J20-AD) and wild-type (WT) controls. Animals were 9–12 months old at the start of the experiment. Mice were chronically prepared with a cranial window through which 2-Dimensional optical imaging spectroscopy (2D-OIS) was used to generate functional maps of the cerebral blood volume and saturation changes evoked by whisker stimulation and vascular reactivity challenges. Unexpectedly, the hemodynamic responses were largely preserved in the J20-AD group. This result failed to confirm previous investigations using the J20-AD model. However, a final acute electrophysiology and 2D-OIS experiment was performed to measure both neural and hemodynamic responses concurrently. In this experiment, previously reported deficits in neurovascular coupling in the J20-AD model were observed. This suggests that J20-AD mice may be more susceptible to the physiologically stressing conditions of an acute experimental procedure compared to WT animals. These results therefore highlight the importance of experimental procedure when determining the characteristics of animal models of human disease.

Abstract:
Plasmalogens are alkenyl-acyl glycerophospholipids and decreased in post-mortem Alzheimer’s disease (AD) brains. The aim of this study is to investigate the time-dependent changes of plasmalogens in the hippocampus of an AD model mouse (J20). Plasmalogen levels at 3, 6, 9, 12 and 15 months were analyzed by liquid-chromatography-targeted-multiplexed-selected-reaction-monitoring-tandem-mass-spectrometry (LC-SRM/MS). Reactive oxygen species (ROS) levels were evaluated using dichlorofluorescein diacetate (DCF-DA). Plasmalogen synthesizing enzyme glycerone-phosphate O-acyltransferase (GNPAT) and late endosome marker Rab7 levels were quantified by Western blotting. GNPAT localization, changes of neuronal and glial cell numbers were evaluated by immunostaining. Compared to wild-type mice (WT), total plasmalogen-ethanolamine, but not plasmalogen-choline levels, were increased at 9 months and subsequently decreased at 15 months in J20 mice. A principal component analysis of plasmalogen-ethanolamine species could separate WT and J20 mice both at 9 and 15 months. Both GNPAT and Rab7 protein were increased in J20 mice at 9 months, whereas GNPAT was decreased at 15 months. ROS levels were increased in J20 mice except for 9 months. Our results suggest that increased plasmalogen-ethanolamine could counteract ROS levels and contribute to the phagocytosis process in J20 mice at 9 months. Such results might indicate a transient protective response of plasmalogen-ethanolamine in AD conditions.

This article investigates the impact of casualization on the labour market of organized manufacturing industries in India. In specific, this article analyses the impact of casualization on the labour demand and output elasticity of 15 manufacturing industries in the post-liberalization period for the direct production workers. Dynamic panel data analysis of system generalized methods of moment method is used here for the estimation of lagged labour demand for permanent production workers. Aggregate level analysis ensures that casualization has a significant positive impact on the labour demand and on output elasticity. Disaggregate level analysis also confirms the significant positive impact on the output elasticity at individual industry level. However, wage share of the workers falls along with the increase in the usage of contract workers across most of the industries. Output elasticity increases but at the cost of falling wage share. Casualization generates more job opportunity but reduces the wage share for the permanent production workers in post-liberalization period. JEL Codes: J20, J21, J30, J46, C23

Making a transgenic mouse involves random integration of exogenous DNA into the host genome that can have the effect of disrupting endogenous gene expression. The J20 mouse model of Alzheimer’s disease (AD) is a transgenic overexpresser of human APP with familial AD mutations and has been extensively utilised in preclinical studies and our aim was to determine the genomic location of the J20 transgene insertion. Methods: We used a combination of breeding strategy and Targeted Locus Amplification with deep sequencing to identify the insertion site of the J20 transgene array. To assess RNA and protein expression of Zbtb20, we used qRT-PCR and Western Blotting. Results: We demonstrate that the J20 transgene construct has inserted within the genetic locus of endogenous mouse gene Zbtb20 on chromosome 16 in an array, disrupting expression of mRNA from this gene in adult hippocampal tissue, while expression of Zbtb20 protein remains unchanged. We note that the endogenous mouse App gene also lies on chromosome 16, although 42 Mb from the Zbtb20 locus. Conclusions: These data will be useful for future studies utilising this popular model of AD, particularly those investigating gene interactions between the J20 APP transgene and other genes present on Mmu16 in the mouse.

Abstract Therapeutic interventions for Alzheimer’s disease (AD) remain limited due to an incomplete understanding of the molecular mechanisms of its onset and progression. Cerebrovascular dysfunction occurs early during disease development. Attenuation of mTOR, a key regulator of aging, attenuates and reverses cerebrovascular deficits by restoring cerebral blood flow, brain vascular density, neurovascular coupling, and vascular amyloid-β clearance in hAPP(J20) mice expressing human amyloid precursor protein carrying two FAD-associated mutations. The mechanisms by which mTOR attenuation alleviates AD pathology are poorly understood. This study defined changes in the microvascular proteome of hAPP(J20) mice arising from mTOR attenuation. At 7 months of age, hAPP(J20) mice were fed vehicle- or rapamycin-supplemented diet (2.24 mg/kg/day) for 4 months. Mass spectrometry of collected brain microvasculature identified significant changes in 840 of 3361 proteins (p<0.05). mTOR attenuation led to significant changes in 26 of these proteins, some of which are involved in pathways including tight junction regulation, calcium signaling, and actin cytoskeleton regulation. Candidate mediators of mTOR-driven cerebrovascular dysfunction were identified by selecting proteins that were aberrantly altered in hAPP(J20) microvasculature and normalized by rapamycin. Examples include members of the heterogeneous nuclear ribonucleoprotein family (hnRNPA/B, hnRNPD) which regulate the mRNA stability of genes related to cellular cycle arrest and inflammatory cytokines as well as localization of crucial mRNA involved in nitric oxide signaling. Also included are nucleoporin 54 and vacuolar ATPase assembly factor, both of which are altered in aging and neurodegeneration. Subsequent studies will elucidate the role of these proteins in mTOR-driven cerebrovascular dysfunction in AD.

Objective - Labor is one of the most important factors in production activities. Increased human effort in the production process will increase output, productivity and promote economic growth. This research aims to analyze the condition and potential of labor demand in Pontianak City, analyze job opportunities in Pontianak City and formulate a strategy of labor policy in Pontianak City. Methodology/Technique - The method used is descriptive with quantitative analysis which a qualitative interpretation. The data used is secondary data and related documents for data enrichment. Findings - The results show that: labor demand is showing an increasing trend year by year, increasing employment absorption from 233,788 in 2010 to 244,236 in 2014. The rate of absorption growth occurs primarily in the agriculture, building and transportation industries. Employment elasticity in Pontianak City is relatively low at only 0.02% which means the growth of labor absorption is smaller than the increase of economic growth. Novelty - This research shows that employment policy strategies in Pontianak City need to increase the role of Training Center (BLK), the enrichment of nutrition improvement and sustainability, encourage investment, increase competitiveness through increasing labor productivity and increase labor flexibility to the rules among other things. Abstract We measure U.S. listed companies’ skilled labor risk—that is, the potential failure in attracting and retaining skilled labor, by the intensity of discussions on this issue in 10-K filings. We show that this measure effectively captures firm risk due to the mobility of skilled labor. We find that an increase from the 25th to the 75th percentile in the skilled labor risk would increase the skilled labor wage by 22% (or $15,593) and also lead to higher equity-based incentive pay. The skilled labor risk also interacts with other corporate policies such as financial leverage, cash holdings, and M&As. A small molecule named ISRIB has recently been described to enhance memory in rodents. In this study we aimed to test whether ISRIB would reverse learning and memory deficits in the J20 mouse model of human amyloid precursor protein (hAPP) overexpression, a model that simulates many aspects of Alzheimer’s disease in which memory deficits are a hallmark feature. We did not observe a significant rescue effect with ISRIB treatment on spatial learning and memory as assessed in the Morris water maze in J20 mice. We also did not observe a significant enhancement of spatial learning or memory in nontransgenic mice with ISRIB treatment, although a trend emerged for memory enhancement in one cohort of mice. Future preclinical studies with ISRIB would benefit from additional robust markers of target engagement in the brain.

As economies transition from industrial to post-industrial, the types of jobs available and employment conditions change. Research indicates that youth employment has been negatively impacted by these changes. For young people seeking to enter the labour market, particularly those combining employment and study, precarious employment has become the norm. However, precarious employment is, for many, no longer a stepping stone on the path to permanent employment. Many young Australians, even those with higher education qualifications, experience prolonged periods of precarious employment. To examine how new employment landscapes are experienced by young workers, we conduct analysis of data collected by the Life Pattern Project, a longitudinal mixed-methods study. Our results show that precarious employment is related to lower levels of job satisfaction and autonomy in young adulthood. JEL Codes: J20, J28, J41

The increase in energy scarcity for all the vehicular needs with the increase in pollution leading to the environmental concerns especially in developing countries like India pushes the researchers towards finding a new source of energy with lesser emissions. This particular study was performed to improve the characteristics of 4 stroke, mono cylinder diesel engine by means of 20% and 40% mixtures of Jamun bio-diesel for two proportions of Rice husk Nano-additives (0.1% and 0.2%). Result illustrates that for J20 with 0.2% additive, Brake Thermal Efficiency reduces by 2.1% with Brake Specific Fuel Consumption increasing by 3.8%. In addition, emissions such as Hydro Carbon, Carbon monoxide and Smoke were reduced by 23.78%, 18.32%, 21.4% by a insignificant growth in oxides of Nitrogen by 11.32%. Result states J20 with 0.2% additive can be effectively used without any modifications and helps in reducing emissions with slight reduction in performance parameters.

Considering the fuel demands, lots of researches have been done in the field of alternate fuels. After attaining the saturation level in this research area, still we are in need to find some alternate ways for fuel demand. Previous researchers concluded that approximate addition of 0-20% of vegetable oil with conventional diesel fuel leads to closer performance of diesel oil and controlled carbon emission; additionally they proved that there is no engine modifications needed for this optimized blend characteristic usage. Likewise, Chemical additives also can be added directly with diesel fuel and fuel blends of vegetable oils in which various researches concluded that 0-10% addition of chemical additives providing closer performance to diesel fuel. If we specifically pointing about solvent ether say Di-ethyl ether, resulting preferable performance to use as alternate fuel up to 0-5% and accepted results have been provided on some vegetable oils such as POME, Neem oil. In case of Jatropha oil, Proper results and Experiments are not found to be used as better alternative solution with Di-ethyl ether blend. This article brings us to enhanced experiment details and results obtained on Di-ethyl ether with Jatropha oil blend in which optimized Bio-diesel blend (J20= 80% diesel and 20% Jatropha oil) is taken as base fuel throughout the experiment. Keeping J20 oil as base fuel, experiments are conducted with various proportions of Di-ethyl ether (0-12%). Additionally it provides, engine performance characteristics and emission parameters when compared to conventional diesels, Jatropha blend (J20), Di-ethyl ether blend.

Employment has an important role to support a certain economy. The number of female workers have increased every year, due to family welfare fulfillment and female role shifting as a head of households. This paper aims to observe the driving factors of female labor force participation in East Java-Indonesia. Logit model is incorporated to estimate the parameters which involved total sample of 23.218 women in productive age which consist of 13.473 working women and the rest of 9.745 non working women in East Java. On the basis of micro data level from National Labor Force Survey (Sakernas) 2018, the result shows that age, education level, marital status, head of the household status, household size, rural-urban location, and income have a substantial effect on FLFP in East Java. Abstract:
The purpose of the study is to analyze the primary incidence of conscript military personnel of the Navy and Ground Forces over the last 10 years from 2010–2019.Methodology. We conducted a selective statistical analysis of medical reports on the health status of personnel and the activities of the medical service in the form of 3/MED of military units, in which served about 80% of the total number of conscript military personnel of the Navy and Russian Ground Forces. The primary incidence of conscript military personnel was correlated with the chapters of the International Classification of Diseases, 10 th revision (ICD-10).Results and their discussion. The average annual incidence rate of conscript military personnel for the Navy amounted to 1143,4–59,1‰, of the Ground Forces was higher at the level of trends — 1345,9–86,1‰. The dynamics of the primary incidence of conscript military personnel of the Navy showed a decrease in data, the Ground Forces — resembled an inverted U-curve with maximum rates in 2013–2014. The leading chapters of diseases of the primary incidence of the Russian Navy and the Army were respiratory diseases (X chapter), skin and subcutaneous tissue diseases (XII chapter), some infectious and parasitic diseases (chapter I), digestive diseases (XI chapter) and bone disease -muscular system and connective tissue (XIII chapter). The total contribution of these classes to the structure was 83,9 and 84,5%, respectively. Indicators of primary morbidity of acute respiratory infections of the upper respiratory tract (J00–J06), infections of the skin and subcutaneous tissue (L00–L08), and other acute lower respiratory infections were of a different significance for the conscripts of the Russian Navy and the Ground Forces of rank 1–6 respiratory tract (J20–J22), flu and pneumonia (J10–J18), diseases of the esophagus, stomach and duodenum (K20–K31) and viral infections characterized by lesions of the skin and mucous membranes (B00–B09). The distinctive features of the primary morbidity of military personnel of the Navy and Ground Forces by disease classes and leading nosologies are revealed. The total contribution of the listed nosologies (groups in chapters) to the structure was 71,3 and 73,2%, respectively.

The Browse Basin is located offshore on Australia’s North West Shelf and is a proven hydrocarbon province, hosting gas with associated condensate in an area where oil reserves are typically small. The assessment of a basin’s oil potential traditionally focuses on the presence or absence of oil-prone source rocks. However, light oil can be found in basins where source rocks are gas-prone and the primary hydrocarbon type is gas-condensate. Oil rims form whenever such fluids migrate into reservoirs at pressures less than their dew point (saturation) pressure. By combining petroleum systems analysis with geochemical studies of source rocks and fluids (gases and liquids), four Mesozoic petroleum systems have been identified in the basin. This study applies petroleum systems analysis to understand the source of fluids and their phase behaviour in the Browse Basin. Source rock richness, thickness and quality are mapped from well control. Petroleum systems modelling that integrates source rock property maps, basin-specific kinetics, 1D burial history models and regional 3D surfaces, provides new insights into source rock maturity, generation and expelled fluid composition. The principal source rocks are Early–Middle Jurassic fluvio-deltaic coaly shales and shales within the J10–J20 supersequences (Plover Formation), Middle–Late Jurassic to Early Cretaceous sub-oxic marine shales within the J30–K10 supersequences (Vulcan and Montara formations) and K20–K30 supersequences (Echuca Shoals Formation). These source rocks contain significant contributions of terrestrial organic matter, and within the Caswell Sub-basin, have reached sufficient maturities to have transformed most of the kerogen into hydrocarbons, with the majority of expulsion occurring from the Late Cretaceous until present.

An altered form of the cellular prion protein, the PrPScor PrPRes, is implicated in the occurrence of the still untreatable transmissible spongiform encephalopathies. We have previously synthesized and characterized aromatic compounds that inhibit protease-resistant prion protein (PrPRes) accumulation in scrapie-infected cells. These compounds belong to different chemical classes, including acylhydrazones, chalcones and oxadiazoles. Some of the active compounds were non-toxic to neuroblastoma cells in culture and seem to possess drugable properties, since they are in agreement with the Lipinski´s rule of 5 and present desirable pharmacokinetic profiles as predicted in silico. Before the evaluation of the in vivo efficacy of the aromatic compounds in scrapie-infected mice, safety assessment in healthy mice is needed. Here we used Swiss mice to evaluate the acute toxicity profile of the six most promising anti-prionic compounds, the 2,4,5-trimethoxychalcones (J1, J8, J20 and J35) and the 1,3,4-oxadiazoles (Y13 and Y17). One single oral administration (300 mg/kg) of J1, J8, J20, J35, Y13 and Y17 or repeated intraperitoneal administration (10 mg/kg, 3 times a week, for 4 weeks) of J1, J8 and J35, did not elicit toxicity in mice. We strongly believe that the investigated trimethoxychalcones and oxadiazoles are interesting compounds to be further analyzed in vivo against prion diseases.

The Browse Basin hosts large gas accumulations, some of which are being developed for conventional liquefied natural gas (LNG). Extensive appraisal drilling has been focused in the central Caswell Sub-basin at Ichthys and Prelude, and along the extended Brecknock-Scott Reef Trend; whereas elsewhere the basin remains underexplored. To provide a better understanding of regional hydrocarbon prospectivity, the sequence stratigraphy of the Cretaceous succession and structural framework were analysed to determine the spatial relationship of reservoir and seal pairs, and those areas of enhanced source rock development. The sequence stratigraphic interpretation is based upon a common North West Shelf stratigraphic framework that has been developed in conjunction with industry, and aligned with the international time scale. Sixty key wells and 2D and 3D seismic data have been interpreted to produce palaeogeographic maps and depositional models for the Cretaceous succession. Geochemical analyses have characterised the molecular and stable isotopic signatures of fluids and correlated them with potential source rocks. The resultant petroleum systems model provides a more detailed understanding of source rock maturity, organic richness and hydrocarbon-generation potential in the basin. The model reveals that many accumulations have a complex charge history, with the mixing of hydrocarbon fluids from multiple Mesozoic source rocks, including the Lower–Middle Jurassic J10–J20 supersequences (Plover Formation), Upper Jurassic–Lowermost Cretaceous J30–K10 supersequences (Vulcan Formation), and Lower Cretaceous K20–K30 supersequences (Echuca Shoals Formation). Burial history and hydrocarbon expulsion models, applied to these Jurassic and Cretaceous supersequences, suggest that numerous petroleum systems are effective within the basin. For example, hydrocarbons are interpreted to have been generated from several source pods within the southern Caswell Sub-basin with migration continuing onto the Yampi Shelf, an area of renewed exploration interest.

The aim of this study was to evaluate the effect of milk fermented with Lactobacillus fermentum J20 (FMJ20) or J28 (FMJ28) on ameliorating indomethacin-induced inflammation. Twenty-eight male C57Bl/6 mice were divided into four experimental groups: indomethacin, indomethacin + FMJ20, indomethacin + FMJ28, and untreated (control). Groups were fed fermented milk for 15 days, followed by administration of indomethacin supplied in three sub-doses over experimental period. Body weight, and food consumption were recorded. Additionally, spleen, kidney, and liver were weighed, and the small intestine length was measured. The cytokines in serum (IL-2, IL-4, IL-6, IL-10, IL-17, IL-23 and TNFα) and in intestinal mucosa (IL-17 and IFNγ) were also determined. Compared to the control, all indomethacin-supplemented groups lost weight (~2.7 g; p < 0.05), but no changes were found in the organ-specific morphometry analysis. FMJ28 showed better results in attenuating serum and intestinal IL-17 levels. Furthermore, showed less epithelial cell loss and inflammatory infiltrates than the other indomethacin-treated groups. These results suggest that FMJ28 may be effective in reducing intestinal and systemic acute inflammation, specifically in mice.

The business process of the J20 production department is currently experiencing obstacles in terms of information delivery, production report, maintenance report, and problem information that causes decision making is often irrelevant. This is due to the use of paper media with a long bureaucratic flow so that the cycle time becomes inefficient. This study used business process reengineering, modeling business process in business process modeling notation (BPMN), and supporting research problem analysis used fishbone diagram. The result showed changes in a process flow, job desk, unit elimination, and proposed design of information systems in the form of application software as a solution to the problems studied, so that it is expected to obtain process time efficiency of 65.1% and cycle time of 99.2% of the previous total time. Proses bisnis departemen produksi J20 saat ini mengalami hambatan dalam hal penyampaian informasi, laporan hasil produksi, laporan maintenance, serta informasi problem, yang menyebabkan pengambilan keputusan sering kali terjadi bias. Hal ini di sebabkan oleh penggunaan media kertas dengan alur birokrasi yang panjang sehingga waktu siklus menjadi tidak efisien. Pada penelitian ini digunakan metode Business Process Reengineering, dengan memodelkan bisnis proses dalam Business Process Modelling Notation (BPMN), serta pendukung analisa masalah penelitian digunakan diagram tulang ikan. Hasil penelitian menunjukan adanya perubahan alur proses, jobdesk, eliminasi unit, serta usulan perancangan sistem informasi berupa software aplikasi sebagai solusi dari permasalahan yang diteliti, sehingga diharapkan dapat diperoleh efisiensi waktu proses sebesar 65,1% dan waktu siklus sebesar 99,2% dari total waktu sebelumnya.

The main aim of this work is the estimation of health risks arising from exposure to ozone or other air pollutants by different statistical models taking into account delayed health effects. This paper presents the risk of hospitalization due to bronchitis and asthma exacerbation in adult inhabitants of Silesian Voivodeship from 1 January 2016 to 31 August 2017. Data were obtained from the daily register of hospitalizations for acute bronchitis (code J20–J21, International Classification of Diseases, Tenth Revision – ICD-10) and asthma (J45–J46) which is governed by the National Health Fund. Meteorological data and data on tropospheric ozone concentrations were obtained from the regional environmental monitoring database of the Provincial Inspector of Environmental Protection in Katowice. The paper includes descriptive and analytical statistical methods used in the estimation of health risk with a delayed effect: Almon Distributed Lag Model, the Poisson Distributed Lag Model, and Distributed Lag Non-Linear Model (DLNM). A significant relationship has only been confirmed by DLNM for bronchitis and a relatively short period (1–3 days) from exposure above the limit value (120 µg/m3). The relative risk value was RR = 1.15 (95% CI 1.03–1.28) for a 2-day lag. However, conclusive findings require the continuation of the study over longer observation periods.

An intact blood-brain barrier (BBB) limits entry of proinflammatory and neurotoxic blood-derived factors into the brain parenchyma. The BBB is damaged in Alzheimer’s disease (AD), which contributes significantly to the progression of AD pathologies and cognitive decline. However, the mechanisms underlying BBB breakdown in AD remain elusive, and no interventions are available for treatment or prevention. We and others recently established that inhibition of the mammalian/mechanistic target of rapamycin (mTOR) pathway with rapamycin yields significant neuroprotective effects, improving cerebrovascular and cognitive function in mouse models of AD. To test whether mTOR inhibition protects the BBB in neurological diseases of aging, we treated hAPP(J20) mice modeling AD and low-density lipoprotein receptor-null (LDLR−/−) mice modeling vascular cognitive impairment with rapamycin. We found that inhibition of mTOR abrogates BBB breakdown in hAPP(J20) and LDLR−/− mice. Experiments using an in vitro BBB model indicated that mTOR attenuation preserves BBB integrity through upregulation of specific tight junction proteins and downregulation of matrix metalloproteinase-9 activity. Together, our data establish mTOR activity as a critical mediator of BBB breakdown in AD and, potentially, vascular cognitive impairment and suggest that rapamycin and/or rapalogs could be used for the restoration of BBB integrity. NEW & NOTEWORTHY This report establishes mammalian/mechanistic target of rapamycin as a critical mediator of blood-brain barrier breakdown in models of Alzheimer's disease and vascular cognitive impairment and suggests that drugs targeting the target of rapamycin pathway could be used for the restoration of blood-brain barrier integrity in disease states.

ABSTRACT This paper presents the calcareous nannofossil zonation of the Middle and Upper Jurassic of onshore Kuwait and formalizes current stratigraphic nomenclature. It also interprets the positions of the Jurassic Arabian Plate maximum flooding surfaces (MFS J10 to J110 of Sharland et al., 2001) and sequence boundaries in Kuwait, and correlates them to those in central Saudi Arabia outcrops. This study integrates data from about 400 core samples from 11 wells representing a nearly complete Middle to Upper Jurassic stratigraphic succession. Forty-two nannofossil species were identified using optical microscope techniques. The assemblage contains Tethyan nannofossil markers, which allow application of the Jurassic Tethyan nannofossil biozones. Six zones and five subzones, ranging in age from Middle Aalenian to Kimmeridgian, are established using first and last occurrence events of diagnostic calcareous nannofossil species. A chronostratigraphy of the studied formations is presented, using the revised formal stratigraphic nomenclature. The Marrat Formation is barren of nannofossils. Based on previous studies it is dated as Late Sinemurian–Early Aalenian and contains Middle Toarcian MFS J10. The overlying Dhruma Formation is Middle or Late Aalenian (Zone NJT 8c) or older, to Late Bajocian (Subzone NJT 10a), and contains Lower Bajocian MFS J20. The overlying Sargelu Formation consists of the Late Bajocian (Subzone NJT 10b) Sargelu-Dhruma Transition, and mostly barren Sargelu Limestone in which we place Lower Bathonian MFS J30 near its base. The lower part of the overlying Najmah Formation consists of the Najmah Shale, which is subdivided into three subunits: (1) barren Najmah-Sargelu Transition, (2) Late Bathonian to Middle Callovian (lower Zone NJT 12) Lower Najmah Shale, and (3) Middle Callovian to Middle Oxfordian (upper Zone NJT 12 to NJT 13b) Upper Najmah Shale. Middle Callovian MFS J40 and Middle Oxfordian MFS J50 are positioned near the base and top of the Upper Najmah Shale. The upper part of the Najmah Formation is represented by the Late Oxfordian (Subzone NJT 13b) Najmah Limestone, and is overlain by the Kimmeridgian (Zone NJT 14) Jubaila Formation. Early Kimmeridgian MFS J60 and Late Kimmeridgian MFS J70 are positioned near the base and top of the Jubaila Formation. The positions of Late Jurassic MFS J80, J90 and J100 are not constrained by our biostratigraphic data and are positioned in the Gotnia Formation. The Upper Tithonian MFS J110 and the Jurassic/Cretaceous boundary are positioned in the Makhul Formation.

We consider two life cycle models of labor supply that use nonconvexities to generate retirement. In each case we derive a link between hours worked prior to retirement, the intertemporal elasticity of substitution for labor (IES), and the size of the nonconvexities. Abstract We analyze the supply-side disruptions associated with COVID-19. We find that sectors in which a higher fraction of the workforce is not able to work remotely experienced greater declines in employment and expected revenue growth, worse stock market performance, and higher likelihood of default. The stock market overweights low-exposure industries. Thus, our findings cast light on the disconnect between stock market indices and aggregate outcomes. We combine these ex ante heterogeneous industry exposures with daily financial market data to create a stock return portfolio that tracks news about the supply-side disruptions resulting from the pandemic.
