The article will focus on our personal experience in Russian translation of the reference book by French semioticians Algirdas Julien Greimas and Jacques Fontanille on the semiotics of passions. In particular, possible translative variations of terms relating to passions such as ‘umbrage’ with no exact analogue in Russian have been discussed with one of the authors, Jacques Fontanille. According to Umberto Eco, for a theory of translation, not only may it be necessary to examine many examples of translation, but also to have had at least one of the following three experiences: in checking translations by others, in translating, and in being translated - or better still, in being translated in collaboration with one’s translator. We will also present Yuri Lotman’s semiosphere, being especially interested in the French translation of concepts such as ‘unpredictability.’ The experience of written scientific translation, on the one hand, and experience in the international cultural sphere, on the other hand, will allow us to put forward some hypotheses about the importance of intersemiotic translation.

¿Hay máquinas pensantes? En este artículo consulto material histórico, filosófico y empírico, tratando de responder esta intrigante pregunta. Mi discusión histórica y filosófica revisa el pensamiento de cinco famosos franceses (Michele de Montaigne, René Descartes, Salomon de Caus, Julien Offray de La Mettrie y Jacques Vaucanson) y de un famoso americano (William James). Mi revisión de investigaciones empíricas se focaliza en cuatro temas contemporáneos en cognición comparada: aprendizaje asociativo/causal, memoria de corto plazo, discriminación numérica, y metacognición. Concluyo que existen máquinas pensantes, ellas son los humanos y los animales. El acto de pensar, podría ser dicho, media los complejos cambios en el comportamiento que muestran humanos y animales. Sin embargo, computadores y otros dispositivos mecánicos son pálidas réplicas que están construidas con "el material equivocado". Nunca lograrán el estatus de máquinas pensantes.

Este ensayo tiene como objetivo explorar cómo el poemario La tortuga ecuestre de César Moro, escrito entre 1938 y 1939, y el cortometraje Un perro andaluz de Luis Buñuel, que vio la luz en el año 1929, dos obras emblemáticas de la vanguardia surrealista, abordaron la representación icónica del cuerpo. Este fenómeno es analizado, por un lado, desde la perspectiva del quiasmo hallada en la obra del filósofo Maurice Merleau-Ponty; y, por otro, desde las semióticas generativa (Algirdas Julien Greimas y Joseph Courtés) y tensiva (Jacques Fontanille y Claude Zilberberg). La conclusión a la que se llega es que dichas obras confirman que el discurso surrealista en la poesía y el cine toma a través de la iconicidad una conciencia del quiasmo al enfocar el cuerpo y el mundo como una sola entidad, de modo íntegro y unitario.

Had anybody told me at the beginning of my university studies that I would end up as a historian of science, I would not only have shaken my head in disbelief, I would in all probability not even have understood the prophecy. When I left high school, my interests ranged from literary writing to the life sciences. After an initial attempt to study biochemistry, which I aborted after a year, my early university education was in philosophy, with an emphasis on continental philosophy, from Descartes to Spinoza, from Kant to Hegel, from Nietzsche to Heidegger, pretty much the curriculum that would dominate the vast majority of the philosophical institutes in German universities in the mid-1960s. Having moved from Tübingen to Berlin, in addition I absorbed early on, as part of a group of students around Jacob Taubes, what came to be subsumed under the label of structuralism and post-structuralism: linguistics and semiotics from the Prague School through Noam Chomsky to Julien Greimas, grammatology from Ignace Gelb to Jacques Derrida, historical epistemology in its latest, Canguilhemian and Foucauldian versions, literary theory as practiced by Roland Barthes, to mention just a few names that formed the horizon of our student reading circles.

The article establishes the source of Imitations of the Ancients, published in 1795 in Saint Petersburg. The translator and compiler of this book was Nikolai F. Emin (1767–1814), son of Fedor A. Emin (1735–1770), the famous Russian writer and adventurer of Turkish origin who died when his son was only three years old. Emin junior did not know ancient languages and therefore used a French anthology by Julien-Jacques Moutonnet de Clairfons (1740–1813), which was printed many times in the 18th century. For his book Emin choose poems by Sappho, Anacreon (to be precise, Anacreontic poetry), Theocritus, Bion, Moschus, Horace and Catullus. Emin also used the biographical preambles to their poems, which he shortened significantly. In the preface to his book, Emin expounded upon the theory of “imitation,” denying the possibility of precision in poetic translation. He also defended the necessity of rhyme in “imitations” of ancient Greek and Latin poetry. Despite these declarations, Emin generally followed quite closely Moutonnet de Clairfons’ French prose. Only once did Emin make fundamental changes. This was in his “imitation” of Theocritus’ eleventh idyll. In this case, he explained his logic in an extensive footnote. Emin changed the ugly cyclops Polyphemes into the gallant and handsome shepherd Menalk, substantiating his decision with the declaration that such a hero better suited “our present age.” In other cases, deviations from ancient texts can be explained not by Emin’s arbitrariness, but by changes made by Moutonnet de Clairfons in the French translation. For example, it was he who combined two poems by Catullus (XCII и LXXXV), a decision that Emin followed. To the famous lament for a sparrow, Moutonnet de Clairfons added an appeal to “sensitive lovers” which is also found in Emin’s text.

The article analyzes the phenomena of European art of the first half of the 20th century, which not only prepared the birth of electroacoustic music, but also contributed to the dialogue between this trend of academic art and film music. The new view on the phenomenon of sound was the result of scientific and technological progress, powerful social transformations, and a change in the worldview paradigm. The first part of the article discusses the idea of autonomous sound in the works of Luigi Russolo, the concept of “liberation of sound” in the music of Edgar Varèse, and the role of noise design in the creative actions of Dadaists and performances of Antonin Artaud’s “théâtre de la cruauté” (“theatre of cruelty”). The second part is devoted to an in-depth study of sound-noise experiments in the French film music of the 1930s and 1940s. Analyzing samples of musical and noise collages (collaborations of Jacques Tati and Jean Yatove in the film Jour de fête, René Clair and Georges Auric—in À nous la liberté, Jean Vigo and Maurice Jaubert — in L’Atalante, Jean Renoir and Joseph Kosma—in La Bête humaine); methods of manipulating the optical track and the “partition rétrograde” (the product of the “co-authorship” of directors Jean Grémillon and Julien Duvivier and composers Alexis Roland-Manuel and Maurice Jaubert); examples of enriching the timbre palette of film music through electronic instruments (ondes Martenot in the music of Arthur Honegger, Adolphe Borchard, Maurice Thiriet to the films of Abel Gance, Dimitri Kirsanoff, Sacha Guitry, Marcel L’Herbier) in the context of global trends in art and technology, the author comes to the conclusion that by the beginning of the 1950s, the ground for electroacoustic experiments and their immediate introduction into film music was fully prepared, and that in the previous years, filmmakers had realized the expressive power of noise and electronic sounds.

ISSN 0804–4643 We report the clinical history and results of endocrine investigations in two brothers born to consanguineous parents, who presented with hypokalemia and arterial hypertension when they were aged 2 and 6 years. The hormonal serum assay results, including extremely low values for aldosterone and plasma renin activity, favored the existence of apparent mineralocorticoid excess. A diagnosis of 11β-hydroxysteroid dehydrogenase (11β-HSD) deficiency was made, based on assays of the hydrogenated urinary metabolites of cortisol and cortisone, as well as of corticosterone and dehydrocorticosterone. Indeed we found a very low rate of urinary elimination of cortisone metabolites: tetrahydrogenated cortisone was reduced to between 0.10 and 30 μmol/24 h, which is 15–100 times lower than the normal rate; hexahydrogenated cortolones a and β were found to be 7-to 20-fold lower than normal levels; and the 11-keto-17-ketosteroid derivatives of cortisone were also reduced. Urinary elimination of the cortisol-reduced metabolites 5β- and 5α-tetrahydrogenated cortisol were slightly reduced or normal. These results argue in favor of a deficit in the enzyme 11β-HSD, which oxidizes cortisol into cortisone. A moderate defect in the conversion of cortisol into 5β-THF compared to normal conversion into 5α-THF was also found. With respect to corticosterone metabolism, we demonstrated the presence of a defect in the oxidation of that steroid into dehydrocorticosterone, also due to the deficit in 11β-HSD. Arterial hypertension and hypokalemia were corrected by treatment with dexamethasone, concomitantly with correction of the low aldosterone and plasma renin activity levels. On the other hand, during this treatment, urinary concentrations of the metabolites of cortisol. cortisone and corticosterone were only moderately affected. Abstract In non-small cell lung cancer (NSCLC), response to immune checkpoint blockade (ICB) is associated with programmed cell death ligand 1 expression that is induced by interferon-γ-produced by tumor-infiltrating CD8+ T cells. However, not all tumors with a PD-L1 expression and/or CD8+ T cell infiltrate respond to ICB, and some tumors without any PD-L1 expression respond to ICB. Moreover, little is known about all the mechanisms governing ICB resistance in NSCLC. The objective of the study was to investigate a circulating immunological signature (cytokines, chemokines and immune checkpoints) which could be predictive of resistance to ICB in patients with advanced NSCLC. We performed a multiplexed analysis on 23 TruCulture® (in vitro T cells activation system) and 41 plasma samples using the Luminex® platform (Bio-Techne, MN USA). We investigated the relationship between the levels at baseline of 30 circulating analytes and the response to ICB of advanced NSCLC patients. Through the TruCulture® samples analysis, we identified two types of responders depending on T cell functionality. The responders with a functional T cell activation had lower levels of neutrophil associated analytes (CXCL5/6; p-value<0.05) than non-responders. They had lower levels of IL-13 and higher levers of TNFα, respectively Th2 and Th1/CD8+/NK associated analytes. All responders had lower levels of CCL17 and higher levels of CXCL10 in plasma samples, respectively M2/N2 and M1/N1 associated analytes. This study highlighted two distinct profiles of ICB responders. The first group has a functional T cell response with a favorable orientation to antitumor cytotoxic action (Th1/CD8+) and few cytokines associated with neutrophils. The second group has a poor functional T cell response, whereas their favorable response to ICB is potentially linked to the activation of the innate immune response. The plasma study highlighted the potential role of polarization of the innate response in the context of the response to ICB for all patients. Doutor em Semiótica pela Universidade de Paris IV-Sorbonne e docente da Université de Limoges, onde fundou o Centre de Recherches Sémiotiques – CERES (Centro de Pesquisas Semióticas) e atuou como reitor. Fontanille é Professor Emérito do Institut Universitaire de France. Em seus trabalhos, mantém articulação com a semiótica fundadora, sem deixar de fazer importantes desdobramentos para as investigações atuais, sobretudo trazendo novos objetos de investigação a esse domínio de conhecimento. Abstract Purpose: Drug-tolerant “dormant” cells (DTC) have emerged as one of the major non-genetic mechanisms driving resistance to targeted therapy in lung cancer, although the sequence of events leading to entry and exit from dormancy remain poorly described. Here, we provide a step-by-step phenotypic and molecular characterization of the different processes involved during the adaptive response to osimertinib using several EGFR-mutated lung cancer models. This strategy led to the identification of a common vulnerability of drug-tolerant cells which could be efficiently and safely targeted by a clinical stage drug. Experimental design: We used the FUCCI (fluorescence ubiquitination cell cycle indicator) system to determine the cell cycle dynamics in real time during the adaptive response to osimertinib in a panel of EGFR-mutated lung cancer cell lines. We performed scRNAseq on untreated and osimertinib-treated G1 and S/G2 sorted cells during early relapse to determine the molecular mechanisms underlying entry and exit from dormancy. We validated our observations in several in vitro and in vivo models as well as in publicly available patient data. Results: FUCCI labelling allowed the identification of a rare population of S/G2 cycling cells (referred to as early escapers) that emerged in the first hours of treatment amongst a majority of stably arrested and progressively dying G1 cells. scRNAseq data revealed that early escapers emerged from a differentiated alveolar type 1 (AT1) phenotype which was invariably associated with an increase in contractile-related gene signatures, F-actin polymerization and Rho/ROCK pathway activation. Using a screen of Rho-pathway inhibitors, we found that tipifarnib, a farnesyltransferase inhibitor (FTi), induced a complete clearance of DTC in vitro. Co-treatment with tipifarnib, a clinically active FTi, safely and durably prevented relapse to osimertinib in a PC9-xenograft model as well as in a PDX model of EGFRL858R/T790M lung cancer for up to 6 months with no evidence of toxicity. Several farnesylated targets were identified in both G1 and S/G2 treated cells, which could explain the high efficiency of tipifarnib in preventing the adaptive response to osimertinib. Finally, we observed that osimertinib + tipifarnib co-treatment completely suppressed the emergence of the AT1 phenotype, prevented mitosis of S/G2-treated cells and increased the apoptotic response through activation of the unfolded protein response (UPR) pathway. Conclusion: Our data strongly support the use of tipifarnib in combination with osimertinib in the clinic to effectively, durably and safely prevent relapse. They provide examples of how individual freedom and restraint in the name of common good can be reconciled. Faced with the impossible choice between a life guided by the principle of love and that of virtue, women often choose self-sacrifice as a means of preserving a sense of individuality in the face of the demands of public universality. The epistolary novels, <em>Julie ou La Nouvelle H&eacute;lo&iuml;se </em>(1761) of Jean Jacques Rousseau and the <em>Delphine </em>(1802) of Germaine de Sta&euml;l, present two rather different treatments of this problem. Rousseau&rsquo;s Julie is a woman whose unquenchable desire transforms her into a prototype of female unreliability not worthy of societal recognition. Sta&euml;l&rsquo;s Delphine, in turn, unmasks a ruthless and unprincipled society which prohibits her from becoming its full-fledged member.</p>

The article traces the emergence of the new concept of communication by analysing Jean-Jacques Rousseau’s Confessions and Julie, or the New Heloise, and Polish letter-writing manuals from the eighteenth and early nineteenth centuries, putting emphasis on the premises of the new attitude (emergence of modern subjectivity, interpreting communication in referential rather than performative terms) and the wider socio-cultural context (emancipation of middle classes, transition from orality to literacy).

O presente artigo procura investigar as imagens de resistência às formas de violência no romance A Ocupação (2019), de Julián Fuks, que mescla três narrativas concomitantes, cujas experiências das personagens apresentam diversos sentidos de ocupação e resistência. Dentre os vários temas propostos neste romance escolheu-se o problema do sintoma como ocultamento de certas imagens que retornam muito tempo depois na condição de fantasmas à consciência. Utilizaremos o aporte teórico de Aby Warburg e Georges Didi-Huberman sobre o caráter fantasmático das imagens, bem como a noção de espectropoética, de Jacques Derrida. Os resultados dessa leitura evidenciam que a relação entre o que está perdido de maneira irremediável e o fantasma imagético da perda traz à tona as muitas maneiras em que a vida, e o circuito dos afetos que a envolve, pela política do passado e do presente, pode ser percebido como um modo de sublimação.

Abstract Background: Resistance to PD1/L1 immune checkpoint inhibitors (ICIs) in advanced NSCLC patients is observed in about 80% of individuals with no robust predictive biomarker yet. The PIONeeR trial (NCT03493581) aims to predict such resistances through a comprehensive multiparametric biomarkers analysis. Methodology: Among the &gt;300 advanced NSCLC patients (pts) recruited in PIONeeR, we focused on the first 137 ≥2nd line ECOG PS0-1 pts treated with single-agent nivolumab, pembrolizumab or atezolizumab. Tumor tissue was collected at baseline and pts were re-biopsied at 6 weeks, and blood-sampled every cycle throughout the 24 weeks post C1D1. Response to PD1/L1 ICIs was assessed by RECIST 1.1 every 6 weeks. Immune contexture was characterized in tumor & blood of each pt through FACS for circulating immune cell subtypes quantification and endothelial activation, blood soluble factors dosage, dual- & multiplex IHC/digital pathology to quantify immune cells infiltrating the tumor, WES for TMB & ICI plasma dosage, leading to 331 measured biomarkers in addition to routine clinical parameters. Multivariable (MV) logistic regression was used to examine the association of each biomarker (controlled by sex, age, smoking status, histological type & PDL1+ Tumor Cells) with the risk of Early Progression (EP), i.e. within 3.5 months of treatment. Multivariable Cox regression analysis was conducted for association with PFS and OS. Results: Overall, the 137 pts were mainly male (64%), smokers (92%) and &lt;70yrs (68%). Tumors were mainly non-squamous (79%) with &gt;1% PDL1+ TC in 36% of the cases, and 21% of pts were still on treatment at data cut-off. Archived samples were available for 80% of pts at inclusion and re-biopsy was available in 52.9% of these cases. The median follow up was 19.8 months, 22.5% of pts did not progress at data cut-off while 62% presented EP. Tumor Cytotoxic T-cells density, especially PD1+ were lower in EP (MV OR=0.45, p=0.022); conversely, higher proportions of circulating cytotoxic T-cells and activated T-cells (HLA-DR+) were observed in EP (MV OR=3.8, p&lt;0.001). Among other biomarkers, Tregs (MV OR=0.44, p=0.018), NK cell subsets (MV OR≤0.44, p&lt;0.05), albumin (MV OR=0.4, p&lt;0.01) and PDL1 TC % (MV OR=0.27, p&lt;0.01) were decreased whereas alkaline phosphatase was increased (OR=3, p=0.018). &gt;65% inter-pt variability was observed in plasma exposures for all ICIs, with 8-10% of pts displaying trough levels below the target engagement threshold. Data will be presented through unsupervised clustering algorithms & multi-modal supervised learning methods. Changes after 6 weeks of treatment will be analyzed to further investigate drugs mechanisms of action. Conclusion: The PIONeeR trial provides with the 1st comprehensive biomarkers’ analysis to establish predictive models of resistance in advanced NSCLC pts treated with PD1/L1 ICIs and highlights how tumor and circulating biomarkers are complementary. Abstract Rousseau, a philosopher of history? The suggestion may startle those who know him as an enemy of history, the founder of Counter-Enlightenment who rejected his century’s hope in progress and conjured quasi-utopias devoid of time. Alone, the political texts seem to justify this interpretation. Side by side with the Emile and Julie sagas, however, they disclose a new Rousseau, the weaver of a master plot that governs private and public history. This essay describes Jean-Jacques’ overarching narrative and the two main subnarratives that compose it by juxtaposing his political and fictional works. In doing so, it contests current conventions about his ideas on women, challenges assumptions about his educational ideals, retrieves new aspects of his debt to Fénelon, and foregrounds the pivotal role that the idea of ‘true love’ plays in his philosophy as the foundation of political community.

Abstract:
Na sociedade de mobilidade crescente, que vem substituindo a sociedade à dominante sedentária, o contato com estranhos/estrangeiros/desconhecidos aumenta velozmente, alterando instituições basilares da sociedade como casa, vizinhança, família, trabalho. A garantia de ser bem recebido em todos os lugares para onde vamos torna-se uma necessidade. A urbanidade, o tratamento polido, mas impessoal não é mais suficiente. Crescem as reivindicações por uma hospitalidade mais carregada de calor humano. Mesmo no comércio, termos como customização, fidelização são marcas da aspiração de pessoalidade no encontro. A discussão desses temas é o contexto para o propósito deste artigo de, com base nas reflexões de Jacques Derrida, Marcel Mauss, Julian Pitt-Rivers e Anne Gotman, desvelar as quatro leis básicas da hospitalidade: a incondicionalidade, a reciprocidade, a assimetria e a compensação. Adicionalmente, pretende levantar outras questões que surgem à margem da atual discussão, em especial o caso do turismo.

Abstract:
In Celine et Julie Vont en Bateau Jacques Rivette works through his discomfort with the theological function of the author, a discomfort stemming from the material effects of authorship on the bodies of his actors. Examples of bodily incision and bruising proliferate throughout the film, part of a process of violent characterization imposed by an authoring demiurge. The film explores several methods of escape from this process, starting with exotic travel and fairy tales, but culminates around repeated allusions to the crucifixion of Christ. The film advances a heretical Christology by positing God as a sadistic author and the wounded body of Christ as the paradigmatic example of being inscribed as a character against one’s will. As this characterization obviously engenders being inscribed in a narrative as well, the structure of the film probes at the notion of both Christianity and narrative cinema as means of escape.

Abstract:
Opinia Jean-Jacquesa Rousseau o powieści Daniela Defoe (pierwodruk: 1719) wpłynęła na późniejsze przekłady i adaptacje dzieła. W jej następstwie upowszechniło się mocniejsze niż w oryginale eksponowanie doświadczeń wyspiarskich w dziejach Robinsona Crusoe. Sukces powieści sprawił też, że jej bohater zaczął żyć jako postać emblematyczna dla kultury zachodniej (zwłaszcza angielskiej) ekspandującej poza granice Europy. Robinson-rozbitek, który przez wiele lat dawał sobie radę na bezludnej wyspie, uosabiał zespół cech gwarantujących sukces w podboju świata, połączenie indywidualizmu, przedsiębiorczości, protestanckiej religijności. Mimo że polska mentalność o rodowodzie szlacheckim miała charakter antymieszczański, powieść Defoe zdobyła popularność również wśród polskich czytelników. Wydaje się, że Robinson Crusoe był też sojusznikiem tych spośród polskich pisarzy, którzy działali najpierw na rzecz ratowania państwa pod koniec XVIII wieku, a kiedy to się nie udało, próbowali przynajmniej unowocześnić polską mentalność. Określa się ich zresztą niekiedy mianem „rozbitków”, nawiązując do popularnego toposu nawy państwowej jako okrętu. Jeden z nich, Julian Ursyn Niemcewicz, traktował Robinsona Crusoe jako postać z pogranicza fikcji i prawdy pozaliterackiej. Sam, jak Robinson, odbywał podróże do Nowego Świata i próbował tam przetrwać. W jego zapiskach dziennikowo-pamiętnikarskich można znaleźć wiele motywów, które rozważa się w związku z powieścią Defoe: migracje, etniczne uwarunkowania kolonizacji, stosunek przybyszów do rdzennych mieszkańców. Przez pryzmat Robinsona Crusoe warto również spojrzeć na powieść Niemcewicza Jan z Tęczyna. Jej bohater, podobnie jak Robinson, trafia na wyspę po katastrofie statku. Nie udaje mu się jednak na niej osiąść z powodu przeszkód natury wewnętrznej: idealistycznych wyobrażeń o miłości do kobiety oraz własnym statusie jako Polaka znakomitego rodu.
