Abstract:
This diploma thesis focuses on the design and production technology of making pattern and core box by milling. The theoretical part summarizes the issue of production models, CAD / CAM technology and milling technology. The practical part is elaborates engineering design of machined parts focused on the specific requirements for application in the foundry. For designing of milling technology was used CAM system. This milling technology was programmed and optimized. Parts are finally produced on 3-axis milling machine. In the end there is technical-economic evaluation of production and discussion of results.

Abstract:
This master thesis is devoted to different possibilities of dealing with consequences of nuclear risks through a commercial insurance. It deals with nuclear energy in the world and in the Czech Republic, international liability conventions, Czech regularization of liability principles, possibilities of insuring nuclear risks through nuclear pools and alternative risk transfer methods. The thesis analyzes the causes, process, consequences and financing the damage of two selected nuclear power plant accidents. Based on available data, the thesis tries to evaluate the ability of the existing system of nuclear risk insurance to eliminate potential negative consequences and suggest possibilities to strengthen the role of commercial insurance.

Abstract:
Otázka jaderného odpadu je více než aktuální. Nejenom Česká republika, ale i ostatní státy řeší otázku, jak izolovat nebezpečný odpad. V ČR je v současné době jediná reálná varianta vybudování hlubinného úložiště, které musí zajistit bezpečnost po sto tisíc let. Zajištěna musí být nejenom dostatečná izolace, ale i ochrana před ať již chtěnou či nechtěnou lidskou činností. Tento technický problém, ale není ten největší, který je třeba v souvislosti s tímto tématem řešit. Ještě větší problém než právní nepřehlednost je váznoucí komunikace. Vláda vybrala lokalitu pro vybudování úložiště sama, bez souhlasu dotčených obyvatel. Veřejnost nedostala včas a dostatek informací a proto se teď bouří a požaduje nápravu. Nedostatek informací vyvolalo panický strach, který je podpořen katastrofami z minulosti. Jak vláda reagovala na protest veřejnosti a jak problém řeší? V současné době vláda vyhlásila pětiletou pauzu kvůli vzrůstajícím protestům veřejnosti. Bylo chování státu v zahraničí jiné? V chování vlády ani občanů však nelze vidět nic odlišného, než co se odehrávalo v okolních státech. Vláda by měla svůj postup plánování úložiště ze základu přehodnotit a pokud tak neučiní, měly by se obce připravit na to, jak využít všechny možné právní způsoby k obraně svého území. Vyhořelý odpad bude ovlivňovat život několika dalších generací a proto by problém nemělo řešit jen několik úředníků za zavřenými dveřmi. Nejedná se o věc jen ekologickou, technickou a ekonomickou, ale i morální.

Thesis focuses on a detailed technical description of all important parts of secondary circuit in Dukovany power plant and its connection to other systems of power plant. In thesis are analyzed significant adjustments which have been made during the entire operation of power plant including in particular the actions associated with project called Utilization of project reserves of units EDU. In the main part of the thesis were carried out calculations of the power plant's power for states before and after the modernization and there is also analyzed the impact of individual changes on the whole power plant. These changes are with the entire secondary circuit drawn in the T-s diagram.

Abstract:
This thesis introduces reader to current knowledge of nanomaterials and their usage. It summarises production methods and usage of different materials in nuclear power plants, nuclear research and nuclear medicine. Theoretical part of this thesis is dedicated to possible usage of carbon nanotubes for neutron beam collimation and guides. In experimental part different materials were tested in measuring box connected to horizontal radial channel of VR-1 nuclear reactor and their influence on neutron flux was measured. Tested samples were non-oriented carbon nanotubes, carbon nanofibers, alumina nanowires, oriented carbon nanotubes with several angles of rotation and these samples were compared with results of graphite.

Abstract:
This thesis deals with the topic of nuclear energy. The Czech Republic is currently a net electricity exporter however this status will probably change in next few years because of growing electricity consumption and decommissioning of obsolete coal-fired power plants. Moreover, contemporary government declared that it won't approve any construction of new reactors stressing the importance of renewable sources of energy (in spite of their limited potential in the Czech Republic). The goal of this thesis is to answer the question whether nuclear energy can compete with other sources of energy and whether is it possible to satisfy growing electricity consumption under the conditions of the Czech Republic by any other source of electricity besides nuclear energy taking into account environmental aspects.

This work deals with the potential of nuclear energy in general and in our country. Nuclear energy offers as almost a necessary alternative to fill growing demand for electricity in our country and the world. Increased attention is paid to the electricity market in our country, the role of nuclear energy in the Czech energy mix and the possibilities of its use in the future as an alternative to other energy sources (renewable, fossil resources), and savings and imports from abroad. The author summarizes the main benefits, but also the risk that nuclear energy brings. Does not forget the attitude of environmentalists, opponents and the public to the nuclear issue, which improves long-term, giving scope for the use of nuclear energy.

The thesis deals with the communication of the Temelín Nuclear Power Plant. Nuclear sources of energy are currently a sensitive issue. The planned extension of the Temelín NPP is also a frequently discussed topic in the Czech Republic; it affects many stakeholders and faces increased public and media interest. The goal of the thesis is to create a communication strategy of the Temelín NPP targeted to the public. The draft of the strategy is based on the conclusions of the SWOT analysis focused on the Temelín NPP communication, furthermore, on the analysis of selected online communication tools used by the Temelín NPP, and mainly on the quantitative survey conducted with the purpose to find out the actual public perception of the Temelín NPP and the public attitude to nuclear energy in general. The strategy also uses the knowledge of the company ČEZ media analysis and the findings of the sociological survey among inhabitants of the area close to Temelín compiled by STEM/MARK research agency.

Abstract:
This master´s thesis deals with the design of a steam turbine for a small nuclear power plant with a power of 50 MWe. In the first part of the work is proposed thermal and mass balance. The turbine has seven unregulated steam extractions, which are divided: two are for high-pressure feedwater heater, four are for low-pressure condensate heater and one is for degassing. The next chapter deals with the calculation of the regulation stage which is designed like an A-wheel and here is also strength control. Then, a pre-design and a detailed design of the stage part with the reaction blading. The stage part is consisted of eight cones with a total number of 27 stages. In the following chapters, is calculated the compensating piston, seals system and bearings. In the end, is made the consumption diagram. Part of the work is a conceptual drawing of a steam turbine section. The resulting steam turbine has a power output 50,197 MW and an internal thermodynamic efficiency of 73,9 %.

This diploma thesis deals with nuclear energetics and its impact to the environment. Present state of the energetics, mainly nuclear energetics, in the Czech Republic is discussed here. Also perspective of nuclear energetics is given. The thesis describes nuclear power plant waste handling and also spent nuclear fuel handling. Nuclear power plant is compared with other sources of electric energy, which are counted to the energetic mix. The author focuses on fossil fuels and also on the renewable energy sources. The amount of produced waste to the unit of produced energy, built up area, safety and economic indicators and other parameters are followed for each source of energy.

Abstract:
Fatique life extension of nuclear powerplants lies in the search for project reserves. This work deals with the evaluation of low-cycle fatigue of nuclear installations of the VVER type and the assessment of the influence of the computational model level. Fatigue tests of austenitic steel using optical method of digital image correlation for which the evaluation procedure is designed and used is performed. Selected model of plasticity with kimenatic (Chaboche) and combinated hardening (Chaboche, Voce) are calibrated from the obtained data. Subsequently, the durability of the test specimen is determined by computational modeling for different material models. From the comparison of the results of fatigue tests with the calculation, the material models suitable for the description of fatigue life and their validity are determined.

The second part defines the goals to be achieved. The third part deals with the methods that are used in the fourth part of the document to analyze the company, its surroundings and project risk analysis. Part of this chapter is the identification and evaluation of risks using the RIPRAN method. In the last part of the document, measures are proposed to reduce the probability of occurrence or impact of risks on the project.

This diploma thesis is dedicated to the future development of nuclear energy in the EU area and its incorporation into the current EU energy policy. The work also focuses on a detailed examination of EU energy policy in terms of energy security of the Member States and throughout the text deals with the role of nuclear power as a determining factor of energy security. The work is divided into two parts - theoretical part (Chapter one) and research part (Chapter two and three). The first chapter of this thesis is devoted to current theoretical approaches to the issue of energy security. The issues of energy security are then examined in the final part of this chapter in terms of integration into the all-embracing EU energy policy. The second chapter provides an analysis of the current energy situation in the European Union as a whole and in individual Member States. All findings are applied with emphasis on the status of nuclear energy in the energy mix. The final chapter summarizes current nuclear energy trends in the EU. In addition, analysis of the opinion polls and government interventions in the field of nuclear energy are provided.

The entire diploma thesis analyses causes, consequences and repercussions of an electricity grid network blackout and its impact to the respective consumers. Remarkable blackouts recorded since 1965 are briefly described and basic available information is considered. The grid standard fixing measures and electricity delivery priorities are characterized. The thesis also pays attention to entire Czech Republic grid availability and its blackout endurance as well as to its robustness within last several years. Indirect Fukushima event influence to the Czech Republic electricity system is mentioned. The main part of the thesis comprises of four big European grid blackouts causes and consequences, their comparison to each other and to the Dukovany nuclear power plant grid blackout drill scenario. The preventive and standard corrective measures against blackouts are pointed out.

Abstract:
Nuclear power plants provide about 11 percent of the world's electricity production. For fission process is uranium fuels used with varying percentage of enrichment 235U for most of nuclear reactors. Uranium reserves are reducing and their mining cost increases. Therefore, the thorium fuel is discussed as revolution fuel for current and future nuclear power plants. This diploma thesis deals with possibility of thorium fuel utilization at various types of nuclear reactors with a focus on light water reactors. The practical part of the thesis is focused on simulation and calculations of various uranium dioxide and thorium dioxide layers at the fuel rods. Model of WWER 440 reactor was developed for the calculations with the addition of thorium fuel. The model simulates burning out of fuel for 5 years, with monitoring of fuel behavior and tracking changes of each material. The thesis tries to define the suitable ratio and parameters of layers combination of uranium and thorium fuel. For these ratios and parameters the thesis tries to give sufficient amount of computational analyzes.

Affordable energy in sufficient quantities is essential for the melioration of the world and especially developing countries. Nuclear energy has the potential to provide long-term solution for growing energy consumption in an economically and environmentally sustainable manner. Problematic factors include the financial magnitude of the initial investment, the extensive requirements for technological and institutional establishment, issues related to safety and security, and the disposal of radioactive material. This thesis analyzes the position of nuclear power in the world. Based on an analysis of advantages and disadvantages of nuclear energy, it then identifies the potential for utilizing nuclear energy in emerging countries. Five developing countries, with very different starting positions, have been selected to investigate how they can secure and develop their energy requirements. The selected countries include China, Chile, Ukraine, United Arab Emirates and Bangladesh.

Abstract:
The objective of the thesis is to characterize the nuclear energy in the european area with the focus on nuclear energy in France. The sector of nuclear energy is now experiencing a new period, so-called "nuclear renaissance". Many european countries decided to rebuild the present nuclear power sites or to construct new ones. Nuclear energy is the most advantageous solutions for many european countries because it decreases external energy dependence and it helps to diminish the carbon dioxide emissions. In the connection with the Czech Presidency in the European Union, it was made proposal for the "Nuclear Security Community" which has to unify the security level of member states of the EU and to intensify the mutual cooperation in the domain of nuclear energy. France represents world nuclear leader thanks to efficiently operating oragnizations, excellent results in nuclear research, realization of nuclear fuel recycling and strong government support. Nucler energy in France can serve as an example for four recommendations to other european countries: government support, efficiently working research institutions, global cooperation between all subjects participating on the formation of nuclear energy and active involvement in the international cooperation in nuclear energy. In spite of constantly high initial investment in construction of nuclear power sites, the nuclear energy seems to experience a positive future.

Abstract:
The Master Thesis deals with the use of thorium nuclear fuel in accelerator driven systems. Basic principle of ADS, present situation and future possibilities are described in this work. The main goal of the work is determination of neutron flux in spallation target QUINTA. In December 2013, an experiment was performed at Joint Institute for Nuclear Research, Dubna. Samples of cobalt, situated at different positions in QUINTA target, were irradiated in secondary neutron field generated by deuteron beam of energies 2 AGeV and 4 AGeV and beam of C-12 with energy 2 AGeV. The samples were measured with the use of germanium semiconductor detectors and analysed using gamma-ray spectrometry. Reaction rates of Co-59 products were determined. Neutron flux was determined in setup QUINTA on the base of experimental reaction rates. Experimental reaction rates were compared with calcula1tion of MCNPX code.

Abstract:
This dissertation focuses on irradiations of a spallation set-up consisting of more than half a ton of natural uranium that were executed by a 660 MeV proton beam at the Joint Institute for Nuclear Reserch in Dubna. Two types of irradiations were arranged: with and without lead shielding. Both types were arranged with threshold activation detectors (Al-27, Mn-55, Co-59, and In-nat) located throughout the whole set-up both in horizontal and vertical positions and activated by secondary neutrons produced by spallation reaction. The threshold activation detectors were analysed by the method of gamma-ray spectroscopy. Radionuclides found in the threshold detectors were analysed and reaction rates were determined for each radionuclide. Ratios of the reaction rates were determined from irradiation of the set-up with and without lead shielding. Subsequently, the neutron spectra generated inside the spallation target at different positions were calculated using Co-59 detector. The experimental results were compared with Monte Carlo simulations performed using MCNPX 2.7.0.

Abstract:
This dissertation covers the application of heat accumulation systems in nuclear power engineering, namely in nuclear power plants. It is mainly a case of passive emergency systems, whose task is to accumulate the heat produced in the reactor’s active zone and in spent fuel pools during DBA (design-basis accidents) or beyond DBA. A particular example of heat accumulation is steam condensation after LOCA (loss of coolant accident). The primary circuit steam leakage increases containment pressure and has to be decreased by the steam condensation. This thesis deals with a theoretical substitute for ice condensers, which are used as a passive safety measure in some nuclear power plants. The substitute involves a choice of an alternative material, whose melting temperature (for heat accumulation) is closer to nuclear power plant operating temperatures. The other part of the dissertation discusses heat accumulation in spent fuel pools in case of all cooling systems failure.

Abstract:
The main target of the master´s thesis is reviewing the generation III nuclear reactors in term of the nuclear safety. At first we have to learn some theory of the nuclear safety in order to understand safety systems of the generation III nuclear reactors. Therefore the thesis is divided into two parts. Legislative and technical approaches to nuclear safety are mentioned in the first part. Regulatory bodies, whose task is to supervise nuclear safety in the nuclear power plants, belongs to the legislative approaches. There are defined terms such as defence in depth, redundancy, diversity, etc. There are mentioned methods to assessing nuclear safety – deterministic and probabilistic methods, especially probabilistic methods, for which a simple example is provided. There are also mentioned active and passive safety systems and their significance for nuclear safety and inherent safety too. There is an example of the function of the active and passive safety systems of the EDU nuclear power plant in conclusion of this issue. The second part deals with description of the selected nuclear reactors in context of the construction of the new units of nuclear power plant in Temelín. The nuclear reactors from companies, which applied for the public tender opened by ČEZ, a. s., for the construction of the ETE 3+4. Thus, the nuclear reactor MIR-1200 by ATOMSTROYEXPORT (Russian Federation), the nuclear reactor AP1000 by WESTINGHOUSE (USA) and the nuclear reactor EPR by AREVA (France) are taken into account . Comparison of the generation II and these generation III+ nuclear reactors necessarily belongs to this master´s thesis. These the generation III+ nuclear reactors are compared with the nuclear reactor VVER 440 (EDU) and in particular with the nuclear reactor VVER 1000, which is operated in the nuclear power plant Temelín. The final chapter contains generally appraisal of the whole problem.

The VVER-1200 reactor became the model for the new nuclear power plant. There was designed a heat diagram containing primary components in this work. After that, there was the balance calculation and calculation of parameters of the main devices of the proposed scheme such as condenser, low-pressure and high-pressure regeneration exchangers and steam generator.

This master‘s thesis deals with radiation damage of reactor pressure vessels, specifically of NPP Dukovany Unit No. 3. In general damage mechanisms of reactor steels are described and possibilities of monitoring of material degradation and its recovery used at NPP’s all over the world are mentioned as well. A practical part of the thesis is focused on interpretation of analyses carried out with the assistance of MOBY DICK code. The ground of these analyses is a neutron fluence value development on different locations of RPV for the whole life of operation up to 24th cycle. The analyses results are put into context with performed in-service inspections. The thesis follows up with neutron fluence computation for the future cycles containing new types of nuclear fuel up to 34th cycle. The outcome of practical part of the master‘s thesis is a comparison between new types of nuclear fuel with respect to radiation damage of RPV’s.

Oxid uraničitý (UO2) je v současnosti nejčastěji používaným materiálem jakožto palivo v komerčních jaderných reaktorech. Největší nevýhodou UO2 je jeho velmi nízká tepelná vodivost, a protože se při štěpení UO2 v jaderném reaktoru vytváří velké množství tepla, vzniká v UO2 peletě velký teplotní gradient. Tento teplotní gradient způsobuje vznik velkého tepelného napětí uvnitř pelety, což následně vede k tvorbě trhlin. Tyto trhliny napomáhají k šíření štěpných plynů při vysoké míře vyhoření paliva. Tvorba trhlin a zvýšený vývin štěpného plynu posléze vede ke značnému snížení odolnosti jaderného paliva. Tato práce se zabývá problematikou zvyšování tepelné vodivosti jaderného paliva na modelu materiálu (CeO2). V této práci jsou studovány podobnosti chování CeO2 a UO2 při konvenčním slinováním a při „spark plasma sintering.“ Způsob jak zvýšit tepelnou vodivost použitý v této práci je včlenění vysoce tepelně vodivého materiálu, karbidu křemíku (SiC), do struktury CeO2 pelet. Od karbidu křemíku je očekáváno, že zvýší tok tepla z jádra pelety, a tím zvýší tepelnou vodivost CeO2. V této práci je také porovnávána podobnost chování SiC v CeO2 matrici s chováním SiC v UO2, které bylo popsáno v literatuře.

The aim of the dissertation is proposing and solving energy convection systems for nuc-lear power plants with a sodium fast reactor of the 4th generation. The first part of the dissertation deals with collection and evaluation of information available about nuclear power plants with sodium fast reactor which use nuclear or non-nuclear reheating to increase thermal efficiency. On the basis of the acquired information, thermal schemes are developed and thermal effi-ciency is determined for the systems working in both Rankine thermal cycle and Brayton thermal cycle. In the further part of the dissertation thermal calculation of the reheater for nuclear and non-nuclear reheating is made for the systems working in Rankine thermal cycle. At the end of this dissertation, an apparatus suitable for these systems is suggested and the systems are evaluated in terms of technical implementation and nuclear safety.

This dissertation deals with the idea of annexing a new nuclear resource in the locality of the Dukovany Nuclear Power Plant and the way of its connection to the power system. The main aim of this dissertation is to design concrete solution of the connection of the new resource to a certain point of the power system and complete this design by means of chosen calculation. According to the task, this dissertation consists of three parts. The first part describes the current engineering solution of the main one line diagram of our two existing nuclear power plants and generally of chosen types of power plants abroad, too. There is also a design of the internal consumption of the nuclear power plants. The objective of the second part of this dissertation is to present the survey of the power and dispositional variants of the new resource in view and mainly assess general possibilities of its connection. Using all information from the preceding part, the third part presents the concrete design of the engineering solution of the connection of the new resource to the power system and also the calculation of parameters of block transmission line. Another dissertation named “Study of Slavětice Distribution Substation Enlargement for EDU New Resources“ concurs this part or more precisely, my dissertation as a whole . For my dissertation I have used some materials and information from the ČEZ Corporation which are confidential. I have also used some knowledge from the analysis from the EGÚ Brno Corporation. The aim of the thesis is to analyze the state of preparedness of nuclear power plants (NPP) for long term operation (LTO) based on the IAEA SALTO (Safety Aspects of Long Term Operation) peer review service, analysis of the most significant failures, accidents and operational experience with type reactors PWR/VVER focusing on cases caused by equipment ageing and identification of major structures and components limiting life time of PWR/VVER-type nuclear power plants, and possible measures to ensure their required service life. Based on the results of the IAEA SALTO peer review service, an analysis of the main deficiencies and measures of NPPs in preparation for a safe LTO was performed, focusing on topics whose deeper knowledge is important for the future more precise determination of technical factors limiting the lifetime of NPPs. The main deficiencies and measures in the preparatory phase for LTO and the most important technical measures are summarized in chapter 4.5. The main deficiencies and the most important technical corrective measures in the area of ageing management of structures and components are discussed separately. The history of major failures and operational experience of nuclear power plants with PWR/VVER reactors from the point of view of ageing of structures and components is analyzed in chapter 6.2. The result is a statistic analysis of ageing-related events, an overview of the most significant PWR/VVER reactor failures with an impact on their service life, a statistical overview and discussion of the most important degradation mechanisms, and other important findings from the history of major failures and operational experience. Chapter 6.3 analyzes factors limiting the operation of nuclear power plants with PWR/VVER reactors with focus on structures and components potentially limiting the life of PWR/VVER reactors and possible measures to ensure their required life. In conclusion, the main reasons of permanent shut down of NPPs (actual and potential) for 40, 60 and 80 years of operation and the measures to ensure their required life are summarized.

This thesis describes the provision of the data communication of the new protection relays via 6 kV switch gears to the electro monitoring system at the Dukovany Nuclear Power Station. These protection relays will replace the current analog protection. The replacement process will start in 2015 and will finish in 2018. Given the overall complexity of the project, this thesis deals only with the following 6 kV switch gears 9BB and 9BD. The thesis is divided into a theoretical and a practical part. The theoretical part concerns the actual consumption and output of the Dukovany Nuclear Power Station. The following chapters describe the electro monitoring system and GRAF and LOGA software programs that are closely connected with it and that were used in the practical part. The last chapter of the theoretical part deals with the actual replacement of the protection relays of the 6 kV switch gears. Also, it describes a new optical network, which will be set up at the Dukovany Nuclear Power Station as a part of the process of replacement. The segment of the practical part concerns laboratory verification of the data communication of the new protection relays. Next, it describes actions that had to be taken in order to ensure and verify correct functionality of the data communication. This part is followed up by a closely related practical activity, which describes examination and verification of the protection functions of each of the 9BB and 9BD switch gears protection relays.

This diploma thesis focuses on the changes in the media image of nuclear power in Ceska Televize, which is the only public television in Czech Republic. The study uses a quantitative research method, content analysis, to find out the changes of nuclear power representation in media and to compare changes of news construction mechanisms in the period of two years - one year before and one year after the Japanese nuclear power plant Fukushima Daiichi disaster. The accident started on 11th March 2011 and there is a short description of the situation in Japan at the beginning of the thesis. The theoretical part of thesis serves as an overview of basic information on principles of the media studies theory and mechanisms of news construction (work with information sources, the news value concept, agenda setting, the framing concept), which is also shown from the environmental point of view. The study also focuses on the nuclear power image in the socio-political field and there are some more information about the nuclear power development and status in Czech Republic. This Master thesis compares the reactions of the neighbouring countries to first nuclear tests carried out by their neighbour. It deals with first nuclear tests of eight countries (USA, Soviet Union, United Kingdom, France, China, India, Pakistan and North Korea) and with the reactions of states that are or were at the time of the first nuclear test neighbours of concerned country. In the end, forty-seven reactions of neighbouring countries were examined in this thesis. The author relies to a large extent on qualitative research when she compares individual reactions of the countries and creates eight case studies, each focused on one nuclear state. The main output of this thesis is a clear and well-arranged typology of countries' reactions to the first nuclear test carried out in the state right across their borders. The types of reactions that have been identified in this thesis vary all the way from active support and passive coming to terms with the situations, through neutral reaction all the way to passive disapproval and active rejection. This typology can then in the future contribute to a better predictability of the neighbours' reactions and therefore also to a better assessment of how such event might affect the stability of the region.

Tubulin is the basic building block of microtubules which ensure intracellular transport, morphological changes, the connection of proteins of metabolic pathways, the segregation of chromosomes during mitosis and many other essential processes in plant cells. The localization of tubulin regarding nucleus is strictly controlled and during the interphase, tubulin is usually not present in the nucleus. As we previously showed, one of the exceptions of the presence of tubulin in the nucleus is the cold stress condition. Also, we discovered several plant-specific nuclear export sequences (NES) in tubulin molecules: one in α-tubulin and two in β-tubulin. In this work I found that double mutation of both functional β-tubulin NESes cause slight accumulation of the protein in the nuclei. However, the phenotype of the transgenic plants expressing β-tubulin with mutations is probably not affected by the presence of the mutated protein. The effect of the expression of the mutated β-tubulins was observed in Nicotiana tabacum BY-2 cells as well. The expression of β-tubulin carrying a single NES3 mutation or double mutation of both NESes bring changes of the division activity of the cells, while the NES2 mutation does not have any effect. History of peaceful use of nuclear energy, despite of all necessary emphasis on operation safety, is accompanied by a series of accidents, some of which left indelible trace in minds of wide public and so led to changes in the comprehension of nuclear safety and in the access to its preservation. Because the consequences of possible nuclear power plant accident have always serious social impacts, it is most desirable to define all possibly risks, to quantify probability of their formation and in this way to gain the possibility to prevent them effectively and efficiently. This work is dedicated to approve or to disapprove that the KARS method is practically applicable for evaluation of nuclear power station operation safety. The work objective is to evaluate events and risks associated with the operation of a particular nuclear facility by the KARS method and critically evaluate obtained results and the used method as such. The work is divided into theoretical and practical part. Theoretical part includes basic information necessary for correct understanding of the chosen data processing procedure and subsequent interpretation of the gained results. A brief overview of basic concepts relating to the nuclear safety, the safety culture and the emergency preparedness, including their definitions is given in this section of the work. For the evaluation of obtained results it is useful to make familiar with basic concepts of the NPP safety evaluation, PSA and the stress tests. Obviously, it is necessary to describe the KARS method and to define the concept of quality of human performance with regard to the obtained results. The risk analysis by the KARS method was carried out in the four consecutive steps: 1. Data collection for the risk analysis. 2. Data processing by the use of the KARS method. 3. Interpretation of the obtained results. 4. Comparison of the obtained results with the available data. The particular risks for the construction of the risk correlation table were based on the analysis of activities of individual members of the emergency and technical support center on the basis of managing documentation for each function. In the emergency instructions these risks are either named or the action to manage them are determined. Subsequently these risks are further more specify according to the operational documentation used for solving abnormal and emergency conditions or solving of severe accidents. Determination of the risk correlation seems to be subjective and requires a considerable degree of professional insight into the analyzed issues. The risk analysis was processed according to the procedure that was laid down by Ing. Stefan Pacinda, Ph.D. Each risk taken into consideration in this analysis is briefly described within the interpretation of obtained results. The coefficients of activity with the biggest value were found for these risks: human error, earthquake, plane crash, terrorist threat, errors in operating and managing documentation and fire. The coefficients of passivity with the biggest value were found for these risks: human failure, serious damage to health, ecological disaster, components malfunctions, radiation accident, nuclear material integrity damage and control system malfunctions. Comparison of the risks that were postulated in the stress tests and the risks that were considered in this analysis indicate that the ETE NPP emergency response was prepared to be able to cope these risks. Results of this analysis showed that the essential element for the safe operation of nuclear power plants is qualified, well selected and systematically trained staff. In light of the experience acquired during elaboration of this work I have concluded that the KARS method can be used for nuclear safety evaluation only in a limited way.

Interleukin-1alpha (IL-1alpha) is a well-known proinflammatory mediator acting as a secreted molecule. However, in addition to its ability to activate its membrane-bound receptor, there is growing evidence on its noncanonical nuclear function, which classifies IL-1alpha as a "dual function cytokine". This nuclear action depends on the evolutionary conserved N-terminal domain of IL-1alpha. After proteolytic processing, the N-terminal domain of IL-1alpha translocates into nucleus. Histone acetyltransferase (HAT) complexes were previously identified as nuclear targets of IL-1alpha precursor. However, the specific protein which is responsible for the interaction between IL-1alpha and HAT complexes has not been identified yet. This paper deals with aesthetic evaluation of nuclear power plant Temelín (located in South Bohemia, Czech Republic). Interdisciplinary culturological approach embraces ideas of aesthetics, historiography and landscape ecology. Theoretical part begins with definition of aesthetical paradigma as biologically, culturally and historically influenced perception. Historical excursion into history of aesthetics and landscape painting shows that appreciation of landscape and nature beauty is influenced by history and culture. Paper mentions some aesthetical approaches (to interpret datas with), preferes contextual apporach. Practical part of this paper begins with geographical, demographical and historical description of chosen locality. It is followed by history of construction of Temelín nuclear power plant and summary of its influence on the landscape. Paper sums up results of previous studies and defines research's theory and methodology. Final part contains results and discussion. This master thesis called ''Nuclear devices in the view of law'', deals with the current legal arrangement of civil nuclear facilities (devices) in the Czech Republic. Due to the fact that the Czech Republic is bound by the international commitments and its membership in the EU, the master thesis also covers the international and European regulations. The master thesis is composed of four parts, which are subdivided into chapters and subchapters. The first part is devoted to the concept of the nuclear facilities, its kinds, position in the legal system and the relevant legal instruments. The second part deals with a particular legal framework of the nuclear law, which governs the issue, including the international, European and Czech regulations. A separate chapter discusses the principles, which are common to the all relevant branch of law. The third part is concerned with the life cycle of the operation of the nuclear facility and the conditions for execution of its particular stages. The final part deals with the safety and security of nuclear facility and its realization as the key issue of the nuclear facility legal regulation.

Abstract:
The theme of the dissertation is a change in an approach to securing fire protection in the Temelín Nuclear Power Plant after the accident in the Fukushima Nuclear Power Plant. The theoretical part is an introduction to fire protection. It especially points out the statutory obligations applicable for both the public and the private sector, which are transferred to the companys fire rescue corps too. At the same time, fire protection is interconnected with legislation and specified in greater detail in EU countries. The theoretical part describes the process of building a uniform information system, safety-related issues and prevention of dangerous situations. The theoretical part summarizes the history of the nuclear energy industry, from the discovery of the disintegration of the uranium nucleus in 1938 and the first controlled reaction of uranium liberation by splitting a nucleus in 1942, when, at the same time, the first reactor was built. Unfortunately, this discovery did not only familiarize mankind with positive aspects associated with a controlled reaction, but it also showed the dark side of nuclear energy in the form of atomic bombs used mainly in the course of the World War II. After the World War II, civil defence organizations were gradually founded in the world. The main goal of all these organizations was to prevent nuclear weapons, nuclear power plants and nuclear fuel from being spread. At the present time there are three strong international organizations dealing with nuclear fission. Despite all the efforts to build a system of radiation protection, environmental protection and impacts on living organisms were not elaborated fully. First, passive attitudes aimed at protective equipment were the central theme, and these passive attitudes have been reflecting in an active approach to environmental protection in the past few years, the goal being to eliminate phenomena giving rise to threats to the environment and life as such. The actual history of nuclear power plants is very complex and complicated. The principle of all these power plants, no matter what type of reactor their operation is based on, is always nuclear fission the trouble is that in this case any wrong handling or storage has immense consequences for the whole world's population. The development of the nuclear energy industry in the Czech Republic was slow, which was caused by the occupation of the Jáchymov uranium mines by the Soviet Army after the World War II. Based on an agreement with Czechoslovakia, the Soviet Army exported the uranium extracted to the Soviet Union and left just 10% of it in the territory of Czechoslovakia. At that time, nuclear power plants began to be built in Czechoslovakia, such as the Jaslovské Bohunice Nuclear Power Plant, Dukovany Nuclear Power Plant and the Temelín Nuclear Power Plant as the last one, whose construction was started in times of Czechoslovakia, but which was completed and put into operation in times of the existence of the Czech Republic. The research is focused on emergency events which were the cause of the accident in the Fukushima Nuclear Power Plant. Based on an FMEA (failure mode and effects analysis), the issues related to the activities of the company's fire rescue corps were specified. Another analysis is focused on the evaluation of outside and inside factors and evaluation of strengths, weaknesses, opportunities and threats to the company's fire rescue corps. Utilizing an FMEA, possible risks and failures are detected. The goal of this analysis is to eliminate risks and increase safety. The result is objective evaluation and increase in safety within the Temelín Nuclear Power Plant. A considerable negative aspect of this analysis is that it is very time consuming and varies in time due to the changeability of the factors monitored. The result of all the work is the evaluation of the existing state of the company's fire rescue corps through a SWOT analysiS.

1. ABSTRACT Myosins are proteins that convert chemical energy stored in ATP into mechanical force that is applied on an actin filament. Nuclear myosin 1 (NM1) was the first myosin detected in the cell nucleus. Together with nuclear actin they were shown to play important roles in DNA transcription and chromatin remodeling. However, the molecular details of the NM1 functions are largely unknown. To expand our knowledge about this molecular motor we studied tissue expression, mechanism of nuclear localization and molecular interactions of this myosin motor. In the first part we examined the expression pattern of NM1 in various mouse tissues. We demonstrated that NM1 is present in cell nuclei of all mouse tissues examined except for cells in terminal stages of spermatogenesis. Quantitative PCR and western blots demonstrated that the expression of NM1 in tissues varies, with the highest levels in the lungs. NM1 is a nuclear isoform of earlier identified myosin 1c (Myo1c), which was described initially as a cytosolic, and plasma membrane associated protein. The only known difference between these two proteins was the presence of additional 16 amino acids at the N-terminus of NM1. Abstract: Magnetically ordered iron oxides, namely magnetite, maghemite, hexagonal ferrite M or system Fe-B, with nano or submicron dimensions of particles are the main subject of study of the presented thesis. The materials were investigated mainly by 57 Fe nuclear magnetic resonance (NMR). The results were thoroughly analyzed and compared with the results obtained by other methods (Mössbauer spectroscopy, ZFC/FC magnetic measurement, X-ray diffraction or TEM). In case of the maghemite nanoparticles a regular distribution of vacancies in octahedral positions was verified by the help of NMR in external magnetic fields and at various temperatures. The experi- mental results were also compared with ab-initio calculations. In thin layers of barium M type hexaferrite, effects of reduced particle size on 57 Fe NMR spectra were observed. The NMR methods were also successfully applied to investigation of system FeMoCuB of amorphous and nanocrystalline ribbons, where 57 Fe NMR was able to resolve formation of different phases in depen- dence on the process of preparation.
