Abstract:

Första gången jag använde mig av elever för att lära andra elever var det på grund av tidsbrist. Jag skulle ha en lektion i biologi och hade inte tid att göra en genomtänkt planering av lektionen. Jag delade därför upp eleverna i två grupper och gav dem två olika avsnitt i läroboken att läsa i läxa. När de kom tillbaka fick de först diskutera med andra som läst samma avsnitt och sedan fick de två och två redovisa för varandra. Jag upptäckte då att detta var en väl fungerande lärandeprocess för många av eleverna.

Abstract:

This article focus on how pupils and teachers understand preschool class, school and Leisure Time Centre (LTC) in relation to place. To understand how place is used the concepts of spatial integration and separation are used. The study builds on participant observations - following a group of children during their transition from preschool class to school - observed situations where children and adults talk about LTC are analyzed. The results show that the spatial integration of LTC and preschool class made the children understand them as more alike than the spatial separated LTC and first grade. The results also show that in addition to place, time was significante and that children and adults related time to place in different ways. Children primarily understood different school activities in relation to place and mobility, whereas adults more often refered to clock time in the meaning making processes.

Abstract:

Discussions about books and ways of reading play an important role in the curriculum for the Swedish secondary school system. In an intervention study based on a series of book discussions of an easy reader novel, I investigated the cooperation between a group of Swedish language learners aged 16 to 19, their teacher in the introduction program for newcomers, and the school librarian. Based on my function as a teacher educator, and drawing on the current research in literature didactics, I planned and carried out six discussions where different reading strategies and methods were used. Using the concept of third space (Gutiérrez, 2008; Skerrett, 2010b) and interpersonal space(Cummins, 2000), I discuss the students’cognitive and emotional responses to the story we shared, and reflect upon the way these discussions have developed. Semi-structured interviews with five students, a teacher, and a school librarian constitute the data for this study. The analysis of the data indicates that the leader of the group discussion is instrumental in helping the participants shift between different perspectives, try out new roles, and transgress different boundaries, thereby offering the group different emotional, cognitive and linguistic challenges. Abstract:

I den här artikeln argumenterar jag för praktikbaserade inslag i icke-konfessionell utbildning i religionsvetenskap. Det görs utifrån några konkreta exempel på religiös praktik som jag har utarbetat inom ramen för den undervisning som jag bedriver. Förslaget gäller inte övningar i varje situation eller sammanhang, och det indikerar inte heller att religiös praktik är det enda sättet att uppnå målen med religionsvetenskaplig utbildning. Det finns även etiska implikationer av att införa religiös praktik i en icke-konfessionell utbildning som jag menar bör tas i beaktande. Jag argumenterar för att religiös praktik kan vara ett sätt att åstadkomma ett djupinriktat lärande och tillägna sig kunskap om religion som skiljer sig från andra typer av färdighetsträning som övas inom ramen för en icke-konfessionell utbildning. Religiös praktik i utbildningen kan även, menar jag, vara ett sätt att hantera de problem som det så kallade världsreligionsparadigmet är förknippat med, som har dominerat religionsvetenskaplig utbildning, läromedelsproduktion och kursplaner. Dessa problem handlar om försanttaganden, föreställningar och definitioner som präglar paradigmets religionsförståelse.

Abstract:

Despite exerting a considerable influence over contemporary constitutional thought, there is a very real sense in which the political constitutionalism of JAG Griffith has been neglected. This paper seeks a genuine engagement with Griffith's work and, in doing so, offers a reconstruction and partial defence of Griffith's political constitutionalism. The article characterises Griffith's political constitutionalism as ‘agonistic’– that is to say, issuing from a belief in the inescapably conflictual nature of human association. Central to this agonistic political constitutionalism is the relationship between conflict and politics – or what might be termed the political management of conflict. This paper argues that Griffith's account of conflict, although central to his constitutional thought, is underdeveloped, not least in its failure to elucidate precisely what is to be understood by ‘politics’. It is thus that the paper draws on the work of Michael Oakeshott to propose an understanding of politics as the pursuit of intimations that, it is argued, can help draw out an important blend of conflict, continuity and contingency in Griffith's political constitutionalism.

Abstract:

Notch signaling plays a critical role in controlling proliferation and differentiation of pulmonary arterial smooth muscle cells (PASMC). Upregulated Notch ligands and Notch3 receptors in PASMC have been reported to promote the development of pulmonary vascular remodeling in patients with pulmonary arterial hypertension (PAH) and in animals with experimental pulmonary hypertension. Activation of Notch receptors by their ligands leads to the cleavage of the Notch intracellular domain (NICD) to the cytosol by γ-secretase; NICD then translocates into the nucleus to regulate gene transcription. In this study, we examined whether short-term activation of Notch functionally regulates store-operated Ca2+ entry (SOCE) in human PASMC. Treatment of PASMC with the active fragment of human Jagged-1 protein (Jag-1) for 15–60 min significantly increased the amplitude of SOCE induced by passive deletion of Ca2+ from the intracellular stores, the sarcoplasmic reticulum (SR). The Jag-1-induced enhancement of SOCE was time dependent: the amplitude was maximized at 30 min of treatment with Jag-1, which was closely correlated with the time course of Jag-1-mediated increase in NICD protein level. The scrambled peptide of Jag-1 active fragment had no effect on SOCE. Inhibition of γ-secretase by N-[ N-(3,5-difluorophenacetyl-l-alanyl)]- S-phenylglycine t-butyl ester (DAPT) significantly attenuated the Jag-1-induced augmentation of SOCE. In addition to the short-term effect, prolonged treatment of PASMC with Jag-1 for 48 h also markedly enhanced the amplitude of SOCE. These data demonstrate that short-term activation of Notch signaling enhances SOCE in PASMC; the NICD-mediated functional interaction with store-operated Ca2+ channels (SOC) may be involved in the Jag-1-mediated enhancement of SOCE in human PASMC.

Abstract:

The Joint Advisory Group on Gastrointestinal Endoscopy (JAG) was initially established in 1994 to standardise endoscopy training across specialties. Over the last two decades, the position of JAG has evolved to meet its current role of quality assuring all aspects of endoscopy in the UK to provide the highest quality, patient-centred care. Drivers such as changes to healthcare agenda, national audits, advances in research and technology and the advent of population-based cancer screening have underpinned this shift in priority. Over this period, JAG has spearheaded various quality assurance initiatives with support from national stakeholders. These have led to the achievement of notable milestones in endoscopy quality assurance, particularly in the three major areas of: (1) endoscopy training, (2) accreditation of endoscopy services (including the Global Rating Scale), and (3) accreditation of screening endoscopists. These developments have changed the landscape of UK practice, serving as a model to promote excellence in endoscopy. This review provides a summary of JAG initiatives and assesses the impact of JAG on training and endoscopy services within the UK and beyond.

Abstract:

För drygt hundra år sedan frigjordes de pedagogiska frågorna från den filosofiska domänen. 1910 tillträdde Bertil Hammer som vårt lands förste professor i pedagogik. Ett av de skäl som angavs för denna professur var lärarutbildningens behov av vetenskaplig förankring. Hammer skrev fram ett ambitiöst program, där pedagogiken skulle omfatta kunskaper om lärande (psykologi), kunskaper om utbildning och samhälle (historia/sociologi) samt kunskaper om vad som kan anses rätt och gott (filosofi). Det skulle dock gå många år innan hela detta program realiserades fullt ut. Pedagogiken förblev fram till 1950-talet förankrad i psykologin. På 1960-talet diskuterades de sociala aspekterna allt livligare (en diskussion som inom disciplinen började med Urban Dahllöfs arbeten och sedan utvidgades, med delvis annan riktning, av Ulf P. Lundgren), och först på 1990-talet slog de filosofiska aspekterna igenom (med början i Tomas Englunds pragmatiska inriktning, följt av bland andra Klas Roth). 1996 inrättades landets första professur i didaktik, intressant nog med liknande motivering som den första professuren i pedagogik – i syfte att stärka lärarutbildningens vetenskapliga grund. Då jag tillträdde denna professur var det pedagogiska landskapet som ett slagfält med åsiktslinjer och skyttegravar, och jag fick diverse frågor om vilken sida jag stod på: pedagogikens eller didaktikens, didaktikens eller ämnesdidaktikens, det pedagogiska arbetets eller utbildningssociologins sida och så vidare. Men min uppgift var inte, som jag såg det, att välja sida. Snarare ville jag som ämnesföreträdare utveckla något nytt. Det vid den tiden nyvaknade intresset för ämnesdidaktiska frågor (som Ference Marton redan under 1980-talet hade diskuterat i termer av fack-didaktik), liksom de professionsorienterade frågorna, utgjorde en ny fond för mitt intresse. Härutöver utgjorde frågor om språk, social-semiotik och multimodalitet ett intressant område för att förstå kunskapsrepresentationer och lärande, något som också utgjorde ett nytt perspektiv på frågan om lärande: synen på lärande som teckenskapande och meningsskapande kommunikation i stället för en fråga om individuell begåvning, kapacitet eller förmåga. Men ämnesdidaktiken bidrog även med kunskaper kring lärande av olika kunskapsområden, vilket i sin tur gav upphov till nya frågor jämfört med synen på lärande som en generell förmåga. Och allt detta påverkade i sin tur synen på lärararbetet. Så småningom sammanlänkades allt detta för mig i ett design-orienterat perspektiv på undervisning och lärande. Jag vill berätta denna historia ur ett högst personligt perspektiv. Hela det didaktiska fältet är mångfacetterat och fyllt av allehanda traditioner. Här vill jag berätta om min egen väg – i termer av tre olika ”vändningar”. Dessa vändningar är förstås påverkade av sin tid, men i viss mån har jag också bidragit till dessa vändningar.

Abstract:

Pod shattering resistance is an essential component to achieving a high yield, which is a substantial objective in polyploid rapeseed cultivation. Previous studies have suggested that the Arabidopsis JAGGED (JAG) gene is a key factor implicated in the regulatory web of dehiscence fruit. However, its role in controlling pod shattering resistance in oilseed rape is still unknown. In this study, multiplex genome editing was carried out by the CRISPR/Cas9 system on five homoeologs (BnJAG.A02, BnJAG.C02, BnJAG.C06, BnJAG.A07, and BnJAG.A08) of the JAG gene. Knockout mutagenesis of all homoeologs drastically affected the development of the lateral organs in organizing pod shape and size. The cylindrical body of the pod comprised a number of undifferentiated cells like a callus, without distinctive valves, replum, septum, and valve margins. Pseudoseeds were produced, which were divided into two halves with an incomplete layer of cells (probably septum) that separated the undifferentiated cells. These mutants were not capable of generating any productive seeds for further generations. However, one mutant line was identified in which only a BnJAG.A08-NUB-Like paralog of the JAG gene was mutated. Knockout mutagenesis in BnJAG.A08-NUB gene caused significant changes in the pod dehiscence zone. The replum region of the mutant was increased to a great extent, resulting in enlarged cell size, bumpy fruit, and reduced length compared with the wild type. A higher replum–valve joint area may have increased the resistance to pod shattering by ~2-fold in JAG mutants compared with wild type. Our results offer a basis for understanding variations in Brassica napus fruit by mutating JAG genes and providing a way forward for other Brassicaceae species.

Abstract:

I artikeln analyseras en diskussion om vargen och dess existensberättigande som pågick i Österbottens Tidning under år 2018 i samband med att vargar rörde sig i trakten. Utgående från Paul Slovic analyseras hur tankegångar om risk byggs upp i materialet. Syftet för artikeln är att undersöka hur varg som risk produceras och hanteras i ett tidningsmaterial, med ett särskilt fokus på de roller som barnen tilldelas. Jag intresserar mig i synnerhet för tiden, och för vad som händer med den i dessa processer. En sammansmältning av Slovics tankar om risk och Heta Lähdesmäkis och Outi Ratamäkis analys av den farliga vargen som en diskursiv resurs i det kulturella minnet i Finland ger ett underlag för en analys av ett material som i sin helhet handlar om en konstruktion av risk för vargangrepp på barn. Jag föreslår termen ”maskhål” som ett alternativ till det som Lähdesmäki och Ratamäki kallar för tidshopp, för att visa på den cirkulära rörelsen i den nutida ständiga återkomsten till händelser som man själv inte upplevt – dödliga vargangrepp på barn i äldre tid. Maskhålet ger tillgång till vargangreppen oberoende av tid och plats, och jag ser de känslor som berättelserna genererar som den kraft som upprätthåller existensen av maskhålet. I processen synliggörs även vad jag kallar för barnets inneboende moraliska imperativ.

Abstract:

I följande artikel undersöks vilka diskurser och positioneringar som framträder hos studenter angående deras val att läsa en lärarutbildning. Positioneringar handlar i denna studie om grad av säkerhet, generalisering och attityd, som här har att göra med känslor och värderingar. Att få syn på diskurserna och positioneringarna är relevant för lärarutbildare för att kunna utveckla adekvata strategier för undervisning inom lärarutbildningen. Emedan tidigare forskning genomlyst motiven med en innehållslig synvinkel presenteras i den här artikeln en motsvarande genomlysning av hur innehållet, det vill säga motiven, faktiskt formuleras. Vi menar att i sättet att formulera motiven finns en problematisk aspekt som lärarutbildare bör bemöta för att undervisningen ska leda till att lärarstudenten får en utbildning som är hållbar över tid i ett samhälle i ständig förändring. Resultatet av den studie vi presenterar visar att studenterna främst uttrycker en social diskurs och positioneringar som förmedlar en syn på lärar- och elevidentiteter som förhållandevis ensidiga. I mötet med ett akademiskt förhållningssätt där perspektivtagande och utveckling står i fokus kan en dylik hållning behöva luckras upp för att ge utrymme för förståelse av kunskapande och undervisningskontexter.

Med posthumanistisk ingång har jag i min masteruppsats i samtida dansdidaktik på Uniarts Stockholm (Hultenius 2019) undersökt hur de materialiteter som danselever under en föreställningsprocess i Kulturskolan möter, bidrar till kunskapande i och om dans. Studiens syfte var att undersöka hur materialiteterna som används i en föreställningsprocess i kulturskolans dansundervisning hade dansdidaktisk betydelse. Utifrån studiens resultat argumenterar jag för att dansdidaktiken i Kulturskolan kan utvecklas med en ökad insikt om materialiteternas betydelse.

Hur upprättas och upprätthålls naturalisering av statliga könsklassificeringspraktiker? Vilka vägar till naturalisering aktualiseras när ambitionen är att inkludera trans- och intersexpersoner i statlig administration och reglering? Dessa frågor ligger till grund för analysen i denna artikel om betydelsebildning i den offentliga utredningen Transpersoner i Sverige (SOU 2017:92) – en utredning som på många sätt skiljer sig från tidigare utredningar som behandlar trans- och intersexpersoner och statlig reglering. I analysen behandlar jag utsagor om kunskap samt det arbete dessa utsagor gör i förhållande till betydelsebildning om inkludering i statlig reglering. Jag redogör för vägar till naturalisering av statliga könsklassificeringspraktiker som handlar om möjligheter till kategoribaserad mätning och sortering i befolkningsstyrningsprogram. Jag vill med artikeln bidra till ett akademiskt och politiskt samtal som tar sin utgångspunkt i att naturaliserad statlig reglering leder till ökade negativa konsekvenser för marginaliserade grupper. Det är därför av stor vikt att studera hur naturalisering upprättas, upprätthålls och utmanas.

Abstract:

Denna artikel tar avstamp i den socialsemiotiska multimodalitetsforskningen som utgår från att betydelser skapas i samspel mellan olika semiotiska system, som skrift och bild. I artikeln besvarar jag frågan hur djur, växter och människans relation till dem representeras i texter som handlar om ekologisk produktion och konsumtion inom den finska dagligvaru- och alkoholhandeln. Jag diskuterar även likheter och skillnader mellan representationerna i skrift och bild. Mitt material består av multimodala texter från tre svenspråkiga kundtidningar i Finland: Etiketten, K-Mat och Samarbete. I analysen redogör jag för olika semiotiska resurser och deras betydelsepotentialer i representationerna. Resultaten visar att representationer av det lyckliga livet av korna har en framträdande roll i marknadsföringen. Dessutom framkallar representationerna minnen från det förflutna och väcker konnotationer av den äkta och traditionella, finska matkulturen. I texterna betonas även den ömsesidiga relationen mellan människor och djur och människor och växter. Min analys visar att kundtidningarnas texter som helhet orienterar sig mot en verklighet där känslor och det sinnliga har betydelse.

Abstract It has previously been shown that an activating mutation in β-catenin in osteoblasts leads to the development of myelodysplasia (MDS) with rapid progression to AML in mice through upregulation of Jagged1 expression in osteoblasts and subsequent activation of Notch signaling in hematopoietic cells. The disease is cell autonomous, because it can be transferred to healthy, lethally irradiated recipients. Moreover, the AML phenotype is associated with clonal evolution at the cytogenetic level since common clonal abnormalities are detected in leukemic blasts from all leukemic mice examined. Revealing the relevance of these observations to human disease, activated β-catenin/Notch signaling was identified in 30% of patients with MDS or AML arising from previous MDS. In the current study we examined the efficacy of a monoclonal mouse anti-human Jagged1 ab (JAG1 ab) as a therapeutic agent for osteoblast-induced MDS/AML. JAG1 ab, which recognizes EGF2/EGF4 domains of human, mouse and rat JAG-1, efficiently bound to them with an EC50 ≤ 0.066 nM (11ng/ml) and dose dependently inhibited Jagged-1 induced Notch signaling with an IC50 ≤ 6nM (1ug/ml) in a Notch reporter assay. Single administration pharmacokinetic (PK) analysis in mice for 3 mg/kg BW showed maximum blood concentration (6.26 µg/ml) in 24 hr, and a terminal half-life of 36 hrs. Organ dysfunction or toxicity was not observed at concentrations as high as 30 mg/kg BW for 5 weeks. The efficacy of weekly subcutaneous administration of 3 mg/kg BW was examined in the AML model of activated β-catenin in osteoblasts (Ctnnb1CAosb mice). Administration of JAG1 ab atpost natal day 8, a time at which MDS/AML has fully developed, prevented anemia, neutrophilia and lymphocytopenia of Ctnnb1CAosb mice. Infiltration of blood with leukemia blasts was reversed within 2 weeks following JAG1 ab administration (2 doses). JAG1 ab reversed the increase in the LSK+/CD150+/CD48- subset of long term repopulating HSC progenitors, the leukemia initiating population in Ctnnb1CAosb mice. The percentage of mature myeloid population (CD11b+/Gr1+) increased in the JAG1 ab treated mice suggesting that blocking Jagged-1 signaling promotes myeloid maturation. Indeed, JAG ab abolished myeloid block as shown by the decrease in the ckit+/CD11b+/Gr1 cells that define immature myeloid progenitors. Dysplastic features seen in the marrow, spleen and liver of Ctnnb1CAosb mice were absent in JAG ab-treated animals and normal trilineage hematopoiesis was established. As a result, JAG1 ab prevented lethality of Ctnnb1CAosb mice and progressively increased body weight for a period of at least 12 weeks, the entire time that the treatment was maintained. Upon termination of the treatments, blast infiltration in the blood appeared within 5 weeks with subsequent development of full blown AML phenotype leading to lethality within 23 weeks. These results suggest that blocking Jagged-1 signaling between osteoblasts and HSCs efficiently treats osteoblast-induced MDS/AML in mice and may have a therapeutic application in patients with MDS and MDS transformed to AML. Abstract Abstract 642 HSCs are rare immature cells capable of reconstituting all blood cell lineages throughout the life of an individual. We have previously shown that intermittent treatment with PTH is sufficient to increase the number of HSCs in the marrow of mice. This PTH effect is blocked in vitro with inhibition of gamma-secretase, the mediator of a required step in Notch signaling. Osteoblastic cells are a critical component of the HSC niche and are likely mediators of the PTH-induced increase in HSCs. Specifically the Notch ligand Jag 1 is expressed on osteoblasts and is therefore implicated as a mechanism through which PTH acts on HSCs. Therefore we investigated in vivo the role of osteoblastic Jag 1 in the PTH-dependent increase in HSCs. We utilized the 2.3kb collagen 1 promoter driven cre recombinase to specifically excise Jag 1 from osteoblastic cells in mice (OBJag1 mice). As we previously reported treatment of wild type (WT) controls with PTH 3 times daily for 10 days resulted in a significant increase in phenotypic HSC populations including Lin-Sca1+cKit+CD48-CD150- short-term HSCs (ST-HSCs) (VEH/PTH 0.0405±0.001 vs 0.0650±0.0038, p≤0.0001) and Lin-Sca1+cKit+CD48-CD150+ long-term HSCs (LT-HSCs) (VEH/PTH 0.0077±0.0008 vs 0.0125±0.00096, p≤0.01) as determined by flow cytometric analysis. In contrast treatment of OBJag1 mice did not result in a phenotypic increase in these populations. Despite the lack of a phenotypic increase in HSCs in OBJag1 mice, when HSC function was assessed by competitive repopulation assay, OBJag1 marrow cells demonstrated the same increased repopulating ability as WT mice (WT: VEH/PTH 12.16±2.7 vs 22.32±2.4, p≤0.01, OBJag1: VEH/PTH 13.6±1.8 vs 31.6±5.9, p≤0.01). Upon secondary transplantation however, HSCs from OBJag1 donors treated with PTH resulted in a lower engraftment rate than VEH treated controls (VEH/PTH 14.61±3.8 vs 4.38±0.9, p≤0.05). This result suggests that osteoblastic Jag 1 is necessary for the increase in phenotypic HSCs resulting from PTH treatment and is required to maintain LT-HSC self-renewal. However these data also suggest an osteoblastic Jag 1 independent mechanism that mediates a transient increase in repopulating ability. Decreased apoptosis is a potential mechanism by which PTH may functionally increase HSCs in the absence of increased self-renewal. To determine if PTH treatment decreases the apoptosis rate of HSCs, WT mice were treated intermittently with PTH once a day for 7 days. Despite a lack of increased HSCs by phenotypic analysis at 7 days, marrow from PTH treated mice displayed an increase in LT-HSC function as measured by competitive transplantation. We determined to measure the effect of PTH on apoptotic rates of HSCs using Annexin V membrane expression. By the 7th day of PTH treatment, LT-HSC apoptotic rates were decreased in the PTH treated group (VEH/PTH 10.482±2.25 vs. 6.27±1.93, p≤0.01) suggesting that changes in apoptotic rate of LT-HSCs precedes the HSC increase. These results were confirmed by flow cytometric measurement of activated caspase 3. PTH treatment decreased the percentage of LT-HSCs that were positive for activated caspase 3 (VEH/PTH 4.3±0.5 vs. 2.4±0.3, p≤0.01). PTH induced micro-architectural changes in trabecular bone at day 7 of treatment suggesting bone involvement despite the lack of an increase in bone volume. These results suggest for the first time that PTH may exert its beneficial effect on bone marrow reconstitution through both Jag 1 dependent and independent effects. Additionally, HSCs demonstrate decreased apoptotic rates and increased reconstitution ability prior to a demonstrable phenotypic increase, mimicking the effect seen in the absence of osteoblastic Jag 1. Together these results suggest that the decreased apoptotic rate may be mediated by an osteoblastic Jag 1 independent mechanism. Whether osteoblasts are required for the observed osteoblastic Jag 1 independent effects remains to be seen as these effects could be mediated by a Jag 1 independent osteoblastic mechanism or by an altogether different cellular component of the HSC niche. Further, since stressful manipulation of HSCs ex vivo is essential for their use in transplantation, defining factors regulating and decreasing their apoptosis may improve their engraftment efficiency, expanding their clinical use when their numbers are limited. Detta görs dels i den dagliga verksamheten genom ett stationsinriktat arbetssätt och dels i de olika projekt vi utför. Vårt arbete med stationer och projekt har lett till att vi idag har lugnare barngrupper. Det har också lett till att vi har utvecklats som arbetsgrupp till följd av de diskussioner vi fört och det stöd vi givit varandra, skriver Åsa Berglund, Jenny Harman och Jessica Iversen.

Redaktionen för tidskriften Pedagogisk forskning i Sverige har bjudit in till en nationell debatt över temat Praktiknära forskning – ett paradigmskifte på gång? Ett lovvärt initiativ där det i inledningen till debatten mycket riktigt anges att detta inte är en ny fråga för pedagogisk forskning i Sverige (Debatt, 2020). Dock har den under de senaste decennierna fått ny aktualitet genom reformer, betänkanden, utredningar, ny myndighet och en försöksverksamhet om modeller för samverkan, ULF. Jag har (tillsammans med andra) blivit inbjuden att kommentera de inlägg som publicerats, vilket jag tackar för.
