Abstract:
Jellies for oral administration have been suggested as alternative dosage forms to conventional tablets for improved palatability and compliances for pediatric and geriatric patients. To evaluate the effect of jelly formulation on the bioavailability of cold medicines, two types of jellies were prepared for a fixed-dose combination of acetaminophen (AAP), chlorpheniramine maleate (CPM), dextromethorphan hydrobromide (DMH), and dl-methylephedrine hydrochloride (MEH). Jelly-S and Jelly-H were fabricated using carrageenan and locust bean gum in the absence and presence of xanthan gum, respectively. In vitro dissolution and in vivo absorption of the four drugs in the jellies were compared with other conventional formulations, a syrup and two types of immediate-release (IR) tablets with different hardness, Tablet-S (15 kPa) and Tablet-H (20 kPa). All the formulations exhibited more than 80% dissolution rate within 2 h even though the syrup, Jelly-S, and Tablet-S showed higher 30-min dissolution compared to Jelly-H and Tablet-H. The dissolution rates from the jellies decreased with increasing pH, which resulted in the slowest dissolution in pH 6.8 compared to the syrup and IR tablets. When administered orally to beagle dogs, all five formulations were determined not to be bioequivalent. However, Jelly-S and Jelly-H showed 0.82–1.05 of the geometric mean ratios (GMRs) of AUC0-t for all four drugs compared to the syrup suggesting comparable absorption. In two IR tablets, GMRs of AUC0-t were in a range of 0.55–0.95 indicating a tendency of lower absorption than the syrup and jellies. In conclusion, jelly can be a patient-centered formulation with comparable bioavailability to syrup.

Abstract:
The effects of reducing fat level (9% and 12%), substituting pork fat with olive oil and adding locust bean/xanthan gum on emulsion stability, jelly and fat separation, cook loss, and hardness of frankfurters were investigated and compared with control sample elaborated with 20% of fat content. Results showed that addition of locust bean/xanthan gum produced a significant increase in hydration/binding properties, characterised by lower cook losses, increasing yield, better emulsion stability and lower jelly and fat separation. The substitution of fat pork by olive oil did not affect these parameters. Multivariate comparison between elaborated low-fat products and commercial frankfurters (normal and low-fat) were carried out using a factorial analysis. Results showed that addition of locust bean/xanthan gum results in products similar to commercial frankfurters with higher fat contents.

Abstract:
The research features the technological challenge of producing jelly desserts by replacing gelatin by non-starch polysaccharides of plant, bacterial, and algal origin (separately or in binary mixtures) in order to overcome religious and ethical constraints. The authors focused on the texture which is responsible for gel network formation of desserts. Hydrocolloids are widely used in many food formulations to improve quality attributes and shelf-life. The polysaccharides under analysis included alginate, pectin, iota-carrageenan, konjac glucomannan, xanthan, and guar gum, separately or in binary mixtures. The experiment made it possible to determine polysaccharides and their optimal concentrations (konjac glucomannan 0.4 % : xanthan gum 0.6 %; locust bean gum 0.2 % : xanthan gum 0.8 %; iota-carrageenan 0.4 %) as gelling agents for the production of desserts without gelatin. Formation of a gel network in desserts occurs as a result of a complex interaction of hydrocolloids, milk fat, and sugar. Structure formation of jelly desserts with polysaccharides occurs already at 18 ± 2°C in 20–40 minutes. In contrast, the structure formation of the control sample occurred at 4 ± 2°C in 2–3 hours. Sensory evaluation of the products was conducted by panellists according to the following criteria: taste, colour, consistency, aroma, and appearance. The paper also describes an analysis of texture indicators. The shelf-life was assessed according to moisture content and water activity as 24 hours at 4 ± 2°C. Several economically-viable new formulations were identified: they successfully reproduced the essential attributes of well-established and widely-consumed traditional gelatin desserts.

Abstract:
The aim of this investigation is to address the technological challenge of producing jelly desserts by replacing gelatin (overcome religious and ethical constraints) by non-starch polysaccharides (PS) of plant, bacterial or algal origin - individually (PS-1) or in binary mixtures (PS-1 – PS-2), to study the texture of desserts, responsible for gel network formation of dessert. Hydrocolloids are widely used in many food formulations to improve quality attributes and shelf-life. The polysaccharides assessed included alginate, pectin, konjac glucomannan, xanthan and guar gum, singly or in binary mixtures. The polysaccharides and their optimal concentrations were determined (konjac glucomannan 0.4%: xanthan gum 0.6%; locust bean gum 0.2%: xanthan gum 0.8%; iota-carrageenan 0.4%) as structuring agents for the production of desserts without gelatin. Organoleptic quality of trial products was evaluated by subjective assessment of taste, consistency, aroma and appearance. Analysis of texture indicators of desserts was carry out. Probable shelf-life was assessed by measurements of moisture content and water activity. Several organoleptically-acceptable and economically-viable new formulations were identified; one of these successfully reproduces the essential attributes of a well-established and widely-consumed traditional dessert.

Abstract:
Abstract A polymerase chain reaction (PCR) was developed to differentiate the thickening agents locust bean gum (LBG) and the cheaper guar gum in finished food products. Universal primers for amplification of the intergenic spacer region between trnL 3’ (UAA) exon and trnF (GAA) gene in the chloroplast (cp) genome and subsequent restriction analysis were applied to differentiate guar gum and LBG. The presence of <5% (w/w) guar gum powder added to LBG powder was detectable. Based on data obtained from sequencing this intergenic spacer region, a second PCR method for the specific detection of guar gum DNA was also developed. This assay detected guar gum powder in LBG in amounts as low as 1% (w/w). Both methods successfully detected guar gum and/or LBG in ice cream stabilizers and in foodstuffs, such as dairy products, ice cream, dry seasoning mixes, a finished roasting sauce, and a fruit jelly product, but not in products with highly degraded DNA, such as tomato ketchup and sterilized chocolate cream. Both methods detected guar gum and LBG in ice cream and fresh cheese at levels <0.1%.

Abstract:
In the presented work the choice of starch syrup for the jelly products production has been substantiated. It was based on the analysis of the starch syrup carbohydrate profiles and recipes of jelly products. The influence of syrup on the rheological and structural-mechanical properties of jelly and aerated masses for a two-layer jelly has been determined. The expediency of polydextrose usage has been demonstrated. Its adding leads to the regulation of the jelly structure and foam layer for the samples with more than 50% of sugar replaced by starch syrup. Depending on the ratio of carbohydrates in jelly it was determined the amount of polydextrose adding of which provides the required firming and prebiotic properties of finish products. The change of physical-chemical, structural-mechanical, and organoleptic quality indices of two-layer jelly with modified carbohydrate composition has been investigated during storage.

Abstract:
Although the importance of endoscopic ultrasound (EUS) for esophageal varices (EVs) has been demonstrated, it is difficult to obtain sufficient EUS images with the water-filling method because of poor water stagnation in the esophagus. In this study on EVs, we aimed to evaluate the usefulness of the jelly-filling method for EUS. Consecutive patients who underwent EUS for EVs were included. The quality of EUS images, the diagnostic ability of the presence of blood vessels inside and outside the esophageal wall, and the procedure time were compared between the jelly-filling and water-filling methods. Thirty cases were analyzed (jelly-filling method in 13 and water-filling method in 17). The EUS image quality score was significantly higher in the jelly-filling method (jelly vs. water; three points vs. two points, p < 0.001). Additionally, EUS image quality scores in both nonexperts and experts were significantly higher in the jelly-filling method. The diagnostic ability of the presence of perforation veins was significantly higher in the jelly-filling method (jelly vs. water; 100% vs. 52.9%, p = 0.004). However, the procedure time was significantly longer in the jelly-filling method (p = 0.024). In conclusion, EUS using the jelly-filling method for EVs provided sufficient image quality.

Abstract:
Anthocyanin content in berries has been reported to promote antioxidant properties that mitigate the occurrence of noncommunicable diseases. However, only a few studies have investigated the benefits of anthocyanin-rich food products from mulberry fruit to reduce the cardiometabolic risk factor in dyslipidemia subjects. Anthocyanin-rich mulberry fruit jelly was formulated using mulberry fruit powder (MFP), and its activities on serum cardiometabolic risk factors in dyslipidemia subjects were studied. Morus alba var. Chiang Mai was used as the ingredient for MFP jelly containing 14 g MFP (191 mg anthocyanin) per serving size (170 g). To investigate the effect of MFP jelly on reduction of cardiometabolic risk factors, sixteen dyslipidemia subjects were given one serving of MFP jelly every day for seven days. After MFP jelly intervention, fasting blood cholesterol, low-density lipoprotein (LDL), and inflammatory markers including interleukin-6 levels of the subjects were significantly lower. Postprandial blood parameters were measured at 0–240 min after consuming a high-fat meal before and after MFP jelly intervention. Postprandial blood glucose at 30 min (p<0.05) and insulin at 60 and 90 min (p<0.01) were lower in MFP than in placebo jelly. The area under the curve of insulin in MFP jelly was smaller than in placebo by 31.2%. Therefore, MFP jelly intervention increased insulin sensitivity. For antioxidant activity markers, postprandial oxygen radical absorbance capacity after MFP jelly intervention gave a smaller decrease after high-fat meal intake compared to after placebo jelly intervention. Moreover, for the oxidative stress markers, postprandial malondialdehyde level was significantly lower in MFP jelly. Seven days of intervention by one serving size of MFP jelly containing 191 mg of anthocyanins reduced cardiometabolic risk factors by lowering blood total cholesterol, LDL, and inflammation, and improving insulin sensitivity and postprandial blood antioxidant-oxidative stress activity in dyslipidemia subjects. This trial is registered with TCTR20200415003.

Abstract:
Royal jelly has been demonstrated to have several physiological activities. However, in the literature, different reactions induced by royal jelly are reported. We describe a case of seven-year-old child that was referred to our observation for two episodes of oral allergy syndrome (OAS) that appeared ten minutes after ingestion of royal jelly. Skin prick test with standard panel of inhalant and food allergens, a prick-to-prick test using the royal jelly’s extract responsible for patient’s reactions, and royal jelly patch test with extemporaneous preparation were performed. The specific IgE by ImmunoCAP System method versus Hymenoptera venom, inhalant allergens, food allergens, and lipid transfer proteins was dosed. According to the positive reactions to royal jelly both by prick-by-prick test and by a first reading patch test, royal jelly immediate hypersensitivity was diagnosed. According to the positive response for almond in bothin vivoandin vitrotests we can think of the royal jelly contamination with almond pollen as possible cause of patient’s reaction. Moreover, from the results of specific IgE titers versus Compositae pollens, we have argued the possibility that this case of royal jelly allergy could be explained also by the mechanism of cross-reaction with Compositae pollens.

Abstract:
SummaryThe egg jelly of Discoglossus pictus contains sperm motility-activating activity, the molecular basis of which has not been studied. Discoglossus pictus sperm initiated motility immediately after immersion in egg-jelly extract, as well as after immersion in hyposmotic solution, which initiates sperm motility in the external fertilization of anuran amphibians. Sequential treatment of the D. pictus sperm with these two solutions revealed the predominant effect of hyposmolality in initiation of motility. The motility initiation induced by jelly extract was suppressed by a monoclonal antibody (mAb) that is specific for the 34 kDa sperm motility-initiating substance (SMIS) in the egg jelly of the newt, Cynops pyrrhogaster. Immunoblotting using the anti-SMIS mAb revealed several antigenic proteins that included major ones with sizes of 18- and 34-kDa in D. pictus jelly extract. Scanning electron microscopic observation revealed that granules of jelly matrix, in which SMIS localizes and which have a critical role in the internal fertilization of C. pyrrhogaster, were not observed near the surface of the D. pictus egg jelly. These results suggest that sperm motility-activating activity in egg jelly of D. pictus may be mediated by SMIS homologous proteins that act through a mechanism that is partially different from that of C. pyrrhogaster.

Abstract:
In the eusocial honey bee Apis mellifera, with reproductive queens and sterile workers, a female larva’s developmental fate depends on its diet; nurse bees feed queen-destined larvae exclusively royal jelly, a glandular secretion, but worker-destined larvae receive royal jelly for 3 days and subsequently jelly to which honey and beebread are added. RNA-Seq analysis demonstrated that p-coumaric acid, which is ubiquitous in honey and beebread, differentially regulates genes involved in caste determination. Rearing larvae in vitro on a royal jelly diet to which p-coumaric acid has been added produces adults with reduced ovary development. Thus, consuming royal jelly exclusively not only enriches the diet of queen-destined larvae but also may protect them from inhibitory effects of phytochemicals present in the honey and beebread fed to worker-destined larvae.

Pseudomonas aeruginosa (P. aeruginosa) is an opportunistic bacterium, which could aggressively infect immunocompromised patients and thus, cause high mortality rate. In addition, P. aeruginosa in oropharynx could be aspirated and cause ventilator associated pneumonia. Royal jelly is one of bee’s products that has been used for therapeutic needs including antibacteria. Adherence factor of P. aeruginosa were flagelum, pili and lectin. The aim of the study was to determine the effect of royal jelly to P. aeruginosa adhesion. Suspension of P. aeruginosa (ATCC® 27853) was incubated at 37 °C for 18 h. Treatment groups were exposed to royal jelly with several concentrations, 2%, 4%, 6%; while distilled water was being used as negative control. Bacterial adhesion test was determined using spectrophotometer λ = 600 nm to measure optical density values of adhered bacterial suspension in tubes. The result of one-way ANOVA showed significant differences (p<0.05) of optical density values among groups indicating that royal jelly affected the bacterial adhesion. LSD results showed significant difference of optical density values between 2%, 4%, and 6% royal jelly compared to distilled water. Six percent of royal jelly had the least optical density value compared to the other groups. In conclusion, royal jelly has the ability to decrease adhesion of P. aeruginosa. Six percent of royal jelly has better ability to decrease adhesion of P. aeruginosa than other concentrations.

Abstract:
Eggs from Xenopus laevis are surrounded by several layers of jelly that are needed for proper fertilization. Jelly coat is composed of high-molecular-mass glycoconjugates to which are bound many globular proteins. O-glycans released from the jelly coat of X. laevis have been partially described in previous studies. In this study, we compared the glycosylation pattern of the egg jelly coat isolated from six specimens of X. laevis. The O-glycans were released from jelly coats by alkali/borohydride treatment. Structural characterization was performed through a combination of one- and two-dimensional 1H-NMR and methylation analysis. This allowed the description of a new family of sulphated O-glycans present in jelly coats of all X. laevis. However, the jelly O-glycans showed a low extent of polymorphism between specimens. This intra-specific variability was restricted to the terminal substitution of O-linked oligosaccharides. The differential expression of two glycosyltransferase [an α-(1 → 4) galactosyltransferase and an α-(1 → 3) fucosyltransferase] activities resulted in the characterization of four phenotypes of X. laevis. Furthermore, electrophoretic analysis suggested that the high-molecular-mass fraction of jelly coat was mostly composed of mucin-type glycoproteins. Blot analysis with lectins confirmed that the glycan variability was borne by these mucin-type components. However, fertilization assays suggested that the glycan polymorphism had no repercussion on egg fertilizability.

Abstract:
Pseudomonas aeruginosais a Gram-negative bacterium and causes respiratory infection especially in elderly patients. Royal jelly has been used worldwide as a traditional remedy and as a nutrient; however, the effect againstP. aeruginosais unclear. The aim of this study was to analyze antibacterial, antiadherent, and anti-inflammatory effects of royal jelly againstP. aeruginosa. Wild-type strain PAO1 and clinical isolates ofP. aeruginosawere used for antibacterial assay and antiadherent assay to abiotic surface and epithelial cells, which are pharynx (Detroit 562) and lung (NCI-H292) epithelial cells. In anti-inflammatory assay, epithelial cells were pretreated with royal jelly before bacterial exposure to investigate its inhibitory effect on interleukin (IL-8) and macrophage inflammatory protein-3α/CCL20 overproduction. Although royal jelly did not have antibacterial activity at concentration of 50% w/v, antiadherent activity was confirmed on the abiotic surface and epithelial cells under concentration of 25%. Pretreatment with royal jelly significantly inhibited overproduction of IL-8 and CCL20 from both cells. These results demonstrated that royal jelly inhibitsP. aeruginosaadherence and protects epithelial cells from excessive inflammatory responses againstP. aeruginosainfection. Our findings suggested that royal jelly may be a useful supplement as complementary and alternative medicine for preventing respiratory infection caused byP. aeruginosa.

Abstract:
The article provides the results of 2016-2018 study of morphogenetic markers in honey bees producing royal jelly with different content of 10-hydroxy-2-decenoic acid (10-HDA). Working bees and royal jelly from Kirov region, Tomsk region, and Krasnodar Krai were used as the material for the research. Evaluation of honey bee origin was carried out according to morphometric and phenotypic characteristics with population and genetic analysis using PCR method with assessment of the intergenic locus COI-CОII mtDNA, microsatellite locus mrjp3. It has been established that honey bees having larger size of the abdomen (with the width of the third tergite and sternite of 4.8 and 4.7 mm, respectively) produced royal jelly with a higher content of 10-HDA (more than 3.0%), that is 1.2-1.4 times higher (P≤0.05). Working bees having yellowness on tergites produced royal jelly with 13.7% lower concentration of 10-HDA (P≤0.05) compared to the bees with no yellowness in the phenotype. Honey bees with locus mtDNA – PQQ or PQQQ in the genotype produced royal jelly with 10-HDA concentration on the average 38% higher (p≤0.01) than bees in genotype of which mtDNA – Q locus had been recorded. The change of 10-HDA concentration in royal jelly of honey bees having different allele registration frequency in the mrjp3 locus has been established. Maximum concentrated 10-HDA (3.45%) was recorded in the bees with domination of allele 392 with frequency R≥0,38 in the mrjp3 locus. Minimum 10-HDA content (2.39%) was observed in bees with domination of allele 406 registration frequency in the mrjp3 locus. Honey bees with domination of alleles 485 and 518 in the mrjp3 locus had no significant differences of 10-HDA content in royal jelly at 3.0% and 2.8% indexes respectively.

Abstract:
Objective. We conducted a single-center study to evaluate the usefulness of the magnetic resonance (MR) imaging jelly method for diagnosing endometriosis-associated adhesions in the Pouch of Douglas.Methods.Thirty women with menstrual pain, dyspareunia, and chronic pelvic pain were enrolled in the study. All had been scheduled for laparoscopic surgery on the basis of pelvic and/or ultrasonographic (US) evaluation. All underwent MR imaging both with and without application of US jelly to the vagina and rectum. The images were compared and analyzed postsurgically in a random and blinded fashion by a radiology specialist and a radiology fellow. The radiologists’ interpretations of the images were compared to the surgical findings recorded on DVDs.Results.Adhesions in the Pouch of Douglas were found in 21 patients. The sensitivity and specificity of MR imaging without jelly administration were 85.7% and 55.6%, respectively, for the specialist and 81.0% and 55.6%, respectively, for the fellow; with jelly administration, values were 95.2% and 88.9% for the specialist and 90.5% and 66.7% for the fellow. Opacity produced by the jelly increased the sensitivity and specificity for both radiologists.Conclusion.The MRI jelly method is a potentially useful, beneficial, and simple approach for diagnosing Pouch of Douglas adhesions.
