Abstract:
Several studies have been conducted in the past to clarify various aspects of species in the genus Juniperus L. One critical group is Juniperus oxycedrus L., especially from the taxonomical point of view. For this reason, we have studied the ecology, taxonomy and distribution of the taxa in the J. oxycedrus group. From an ecological and distribution standpoint, in this work we use the ombroedaphoxeric index (Ioex) to explain the presence of Juniperus populations in ombrotypes that are not optimum for these taxa. The controversy over the taxonomy of J. oxycedrus subsp. badia (H. Gay) Debeaux and J. oxycedrus subsp. lagunae (Pau ex C. Vicioso) Rivas Mart. is clarified, and it is accepted as a valid name, J. oxycedrus subsp. badia. The phytochemical differences in essential oils (EO) are addressed and their similarities analyzed; greater similarities are observed between oxycedrus and badia, and between navicularis Gand. and macrocarpa (Sm.) Ball. species. The phytochemical, molecular and distribution differences allow J. oxycedrus subsp. macrocarpa (Sm.) Ball and J. navicularis Gand. to be maintained as species. The results obtained make it possible to establish the rank to which the taxa belong and allow clear discrimination between species in groups that are difficult to interpret. Ecological, bioclimatic, phytochemical and morphometric similarities allow us to subordinate the subsp. macrocarpa to the species J. navicularis.

Experiments conducted in vitro and in vivo, as well as some preclinical trials for cancer therapeutics, support the antineoplastic properties of lectins. A screening of antitumoral activity on HT29 colon cancer cells, based on polypeptide characterization and specific lectin binding to HT29 cells membrane receptors, was performed in order to assess the bioactivities present in four Mediterranean plant species: Juniperus oxycedrus subsp. oxycedrus, Juniperus oxycedrus subsp. badia, Arbutus unedo and Corema album. Total leaf proteins from each species were evaluated with respect to cell viability and inhibitory activities on HT29 cells (cell migration, matrix metalloproteinase –MMP proteolytic activities). A discussion is presented on a possible mechanism justifying the specific binding of lectins to HT29 cell receptors. All species revealed the presence of proteins with affinity to HT29 cell glycosylated receptors, possibly explaining the differential antitumor activity exhibited by the two most promising species, Juniperus oxycedrus subsp. badia and Arbutus unedo.

Abstract:
We have studied Juniperus oxycedrus L. subsp. badia (H.Gay) Debeaux in the central and southern Iberian Peninsula, where the macrobioclimate ranges from Mediterranean-pluviseasonal-oceanic to Mediterranean-pluviseasonal-continental, and the thermotype from the thermo- to the supramediterranean. The relevés were taken following the Braun-Blanquet phytosociological methodology. A statistical treatment was applied to establish a separation among Juniperus communities. To understand the presence of Juniperus communities in territories dominated by species in the Quercus genus, we applied Thornthwaite’s formula to calculate potential evapotranspiration. The general cluster analysis clearly distinguishes two groups of plant communities and separates the different associations in each group. All the plant communities growing on rocky crests and in extremely steep sloping areas are significantly influenced by the soil. The ombroclimatic index does not explain the presence of plant communities influenced by substrate, so we proposed a new ombroedaphoxeric index which explains the presence of Juniperus communities in territories with a thermotype between the thermo- and supramediterranean. The areas of distribution of Juniperus species are expanding due to the spread of rocky areas; this phenomenon causes an increase in edaphoxerophilous areas and a decrease in climatophilous ones. We propose four new plant associations, with updated structures and floristic compositions. Efficient conservation is possible in both the territories studied (Spain and Portugal) through the implementation of specific cross-border cooperation projects.

Fatty acid compositions of seeds of five taxa of the Juniperus section of the genus Juniperus L. (Cupressaceae), i. e. J. drupacea Lab., J. communis L. var. communis, J. communis var. saxatilis Pall., J. oxycedrus L. subsp. oxycedrus, and J. oxycedrus subsp. macrocarpa (Sibth. & Sm.) Ball, were investigated. Methyl ester derivatized fatty acids of the lipophylic extracts of the five species were comparatively analyzed by capillary gas chromatography-mass spectrometry (GC-MS). Juniperus taxa showed uniform fatty acid patterns, among which linoleic (25.8–32.5%), pinolenic (11.9–24.1%) and oleic acids (12.4–17.2%) were determined to be the main fractions in the seed oils. Juniperonic acid was found to be remarkably high in J. communis var. saxatilis (11.4 %), J. oxycedrus subsp. oxycedrus (10.4 %), and J. communis var. communis (10.1 %). To the best of our knowledge, the present work discloses the first report on the fatty acid compositions of seeds of this Juniperus section grown in Turkey.

Abstract:
This study was conducted to examine chemical compositions, antioxidant and anti-inflammatory properties of methanolic and aqueous extracts from aerial parts of <em>Juniperus oxycedrus</em> subsp. <em>oxycedrus</em> growing in Mascara, Algeria. The quantitative assessment indicated that methanol extract was the most concentrated in phenolic, flavonoid and tannin contents (167.77&plusmn;5.12 mg GAE/g DW, 90.56&plusmn;2.23mg QE/g DE and 110.21&plusmn;2.38 mg CE/g DE respectively). The chromatographic analysis by HPLC showed quantitative differences in phenolic constituents, noting that Chlorogenic acid was the major compound of both extracts. Moreover, the methanolic extract exhibited the highest antioxidant activity than the aqueous extract when tested by the 1,1-diphenyl-2-picrylhydrazyl (IC50 4.45&plusmn;0.001 &mu;g/mL) and phosphomolybdenum (328.52&plusmn;0.071 mg of GAE/g DW) assays. Furthermore, the in vitro anti-inflammatory activity showed strong inhibition of albumin denaturation by the methanolic extract at different concentrations when compared to the standard drug diclofenac sodium. These findings confirm the richness of Algerian <em>Juniperus oxycedrus</em> extracts in bioactive compounds with antioxidant and anti-inflammatory capacities.

Tese de mestrado, Controlo da Qualidade e Toxicologia dos Alimentos, Universidade de Lisboa, Faculdade de Farmácia, 2015<br>O conhecimento sobre o valor terapêutico das plantas sempre tem acompanhado a evolução do homem através dos tempos. Várias plantas endémicas da flora Portuguesa, têm sido estudadas pela sua riqueza e elevada constituição em compostos bioactivos, não só com actividade anticancerígena, como também, na prevenção da doença. As lectinas são uns dos vários componentes extraídos das plantas e representam uma família de proteínas ubiquitárias, existentes na forma livre ou associadas às células, podendo ser encontradas na superfície ou no interior destas, exibem uma afinidade para hidratos de carbono, não apresentam actividade catalítica, nem são resultado de resposta imune. O estudo das lectinas justifica-se pela importância dos hidratos de carbono na identidade celular, promovendo mecanismos envolvidos na interacção entre células, assim como uma variedade de processos biológicos. São pontos cruciais na investigação actual, a descoberta de novas lectinas, assim como o estudo da sua especificidade de ligação ao hidrato de carbono. Para o efeito, neste estudo foram seleccionadas quatro espécies endémicas da flora portuguesa, Juniperus oxycedrus subsp. oxycedrus, Juniperus oxycedrus subsp. badia, Arbutus unedo e Corema album. A caracterização do perfil polipeptídico das espécies em estudo, a actividade hemaglutinante dos extractos obtidos, frente a grupos sanguíneos humanos e eritrócitos de coelho, além da especificidade de ligação aos hidratos de carbono, associada à detecção de proteínas glicosiladas, revelou a existência de potenciais proteínas do tipo lectina nos quatro extractos proteicos. A actividade antitumoral foi testada em linhas celulares tumorais humanas derivada de carcinoma do cólon (HT29). A incubação dos respectivos extractos com as membranas das células HT29, revelaram, por análise bidimensional (2D), ligação de péptidos com especificidade para epitopos glicosilados das membranas HT29. Esta ligação foi efectiva para as quatro espécies em estudo. O potencial antitumoral dos quatro extractos demonstraram actividades distintas na inibição da invasão celular em células HT29, ficando patente uma forte capacidade de inibição apresentada pelas espécies Juniperus oxycedrus subsp. badia e Arbutus unedo. Seguiu-se, para as duas espécies com maior poder de inibição da invasão celular, a avaliação da sua actividade inibitória para as metaloproteinases MMP9 e MMP2. Verificou-se uma inibição na produção destas enzimas, sendo um excelente prognóstico para a aplicação destas espécies na inibição da actividade tumoral.<br>The knowledge about the therapeutic value of plants always has followed the evolution of man through the ages. Several endemic plants of Portuguese flora, have been studied for their wealth and high constitution in bioactive compounds with anticancer activity, but also in the prevention of the disease. The lectins are some of the various components extracted from plants and represent a family of ubiquitous proteins, they exist in free form or associated to the cells, which can be found on the surface or inside of these, display an affinity for carbohydrates, do not exhibit catalytic activity, nor are they a result of immune response. The study of lectins is justified by the importance of carbohydrates in cell identity, promoting mechanisms involved in the interaction between cells, as well as a variety of biological processes. Are crucial points in the current investigation, the discovery of new lectins, as well as the study of its specificity of carbohydrate binding. To that end, in this study were selected four endemic species of Portuguese flora, Juniperus oxycedrus subsp. oxycedrus, Juniperus oxycedrus subsp. badia, Arbutus unedo and Corema album. The polypeptide profile characterization of the species under study, the haemagglutinating activity of the extracts obtained against the human blood groups and rabbit erythrocytes, in addition to the binding specificity to carbohydrates, coupled with the detection of glycosylated proteins, revealed the existence of potential proteins of type lectin in the four protein extracts. The antitumor activity was tested in human tumor cell lines derived from colon carcinoma (HT29). Incubation of extracts with HT29 cells membranes by two-dimensional (2D) analysis, revealed the binding of peptides with specificity for epitopes glycosylated HT29 membranes. This results was effective for the four species under study. The potential antitumor of four distinct extracts demonstrated different activities on inhibition of cellular invasion on HT29 cells, getting a strong inhibition capacity by Juniperus oxycedrus subsp. badia and Arbutus unedo species. This was followed, for the two species with greater power of inhibition of cell invasion, the evaluation of its inhibitory activity for the Metalloproteinases MMP9 and MMP2. There has been an inhibition on the production of these enzymes, being an excellent prognosis for the application of these species in the inhibition of tumoral activity.

30-летний опыт акклиматизации растений, накопленный Сахалинским филиалом Ботанического сада-института ДВО РАН, позволяет делать либо выводы, либо прогнозы о перспективности интродукции на юге Сахалина южных инорайонных видов, испытываемых на пределе их адаптивного потенциала – при условии, что к ним будут применяться особые приёмы агротехники. О перспективности видов в данном контексте можно говорить лишь условно, поскольку с классической точки зрения все они относятся к двум низшим, – 5 и 6, группам перспективности. Уже испытанные виды можно разделить на 5 групп. Первая – способные выживать, а некоторые, при определённых условиях, – цвести и плодоносить. Это Mespilus germanica, Pyracantha coccinea, Buxus sempervirens, Carya laciniosa, Ginkgo biloba, Staphylea colchica, Smilax excelsa, Aster albescens, Lonicera fragrantissima, L. etrusca, Elsholtzia fruticosa, Hedera helix, H. colchica. Вторая группа – растения, в первые лет 10 выживающие, но потом часто гибнущие после возвышения кроны над уровнем снежного покрова. Это Gleditsia triacanthos, Carya illinoinensis, Pinus wallichiana. Третья группа – растения, сталкивающиеся с проблемами из-за выпревания корневой шейки. Это виды, многие миллионы лет адаптировавшиеся к малоснежным районам Восточной Азии: Metasequoia glyptostroboides, Abelia chinensis, Viburnum fordiae. Четвёртая группа – способные какое-то время выживать при создании им условий, особых в сравнении даже с остальными упомянутыми видами. Это Ruscus aculeatus, Koelreuteria paniculata, Lonicera implexa, Diospyros virginiana, Jasminum fruticans, Viburnum propinquum, Calycanthus chinensis, Cedrus deodara, Davidia involucrata. Пятая группа – непригодные для акклиматизации. Это Diospyros kaki, Viburnum cylindricum, V. tinus, Juniperus oxycedrus, Pinus brutia subsp. pityusa, Firmiana simplex. Для многих видов пока можно дать лишь предварительный прогноз. Их можно разделить на 6 групп. Первая, наименее проблемная: Jasminum humile, Juniperus indica, Wisteria sinensis. Вторая, где возможна проблема выпревания, включает Cotoneaster microphyllus. Третья группа, в которой проблемы, вероятно, будут преодолимыми: Actinidia chinensis, Gleditsia × texana, Pistacia atlantica ssp. mutica, Campsis radicans. Четвёртая группа – с прогнозом высокой степени риска: Sequoiadendron giganteum, Cunninghamia lanceolata, Sarcococca confusa, Gleditsia sinensis, Cercis siliquastrum, Hibiscus syriacus. Пятая группа, где шансов на акклиматизацию в открытом грунте практически нет: Pittosporum napaulense, Nandina domestica, Callicarpa formosana, Pinus coulteri.
