Abstract:

Abstract:
Ecological conditions of the growing areas, growing practices as well as harvest and postharvest processing influence the yield and quality of chamomile. The aim of this research was to determine the influence of chamomile separation methods, as postharvest treatments, on the essential oil quality and content, with a view to improve current primary processing of this valuable medicinal plant. In order to explain the structure of laboratory data that would give deeper assessment of similarities among various samples of chamomile essential oil, PCA was employed. Tested results showed that separation of chamomile plant material, as postharvest and pre-drying treatment, had significant influence on the number of identified compounds in the chamomile essential oil. The highest content of individual essential oil compounds had chamomile flowers with short stems, especially a-bisabolol, chamazulene, Z-spiroether and E-b-famesene. In the essential oil obtained from chamomile flowers with long stems, content of a-bisabolol and chamazulene were significantly lower, while E-b-famesene and Z-spiroether contents were significantly higher. Furthermore, in the essential oil obtained from chamomile plant material without separation, the lowest content of abisabolol and Z-spiroether, and the highest content of E-b-farnesene were recorded. The correlation analysis was performed to investigate the likeness in the amounts of the active compounds of essential oil from differently processed chamomile samples. All these results indicate the importance of precise and controlled postharvest treatments, since it clearly affects the essential oil quality and content in the primary processing of this valuable medicinal plant.

Антимотилитетна активност обе групе екстраката(ферментисаних и неферментисаних цветова) одређена је уin vitro условима.<br>U okviru ove doktorske disertacije izvedeno jeispitivanje različitih ekstrakcionih postupaka zaizolovanje apigenina iz cveta kamilice, kao i evaluacijabiološke aktivnosti dobijenih ekstrakata. Starting plant samples included fermented andnonfermented (native) flowers.Extraction of fermented flowers was performed by usingultrasound-assisted extraction with ethanol. The concentrationof apigenin was high in obtained extracts. Optimization of theextraction procedures was performed by response surfacemethodology. Antiradical activity of observed extracts wasexamined by electron-spin resonance spectroscopy.Furthermore, pharmacological potential of obtained extractswas confirmed by determining their antimicrobial andantiproliferative activity.Native chamomile flowers were extracted by differentextraction techniques: microwave, ultrasound, Soxhlet andsubcritical water extraction. Subcritical water extractionshowed to be superior in comparison to other applied techniquesin respect to total phenols and flavonoids content. Optimizationof the subcritical water extraction was directed to maximizationof apigenin content. Isolation of pure apigenin from extractsobtained under optimal extraction conditions (sample-tosolventratio 1:30, agitation rate 3 Hz, temperature 115&ordm;C,pressure 45 bar, extraction time 30 min) was performed bypreparative chromatography. Chemical profiles and content ofindividual polyphenolic components in extracts obtained atdifferent pressures, temperatures, and with differentconcentrations of a modifier was determined by UHPLC-DADHESI-MS/MS. In all analyzed extracts the great number ofpolyphenolic components was detected while apigenin was thedominant compound in all extracts. Content of apigenin in theextract obtained under optimal extraction condition was1,700.34 mg/kg. Antioxidant and antiradical potential ofextracts was evaluated according to different mechanisms.Antimicrobial potential of extracts was determined against eightdifferent microbial strains. Ability of extracts to inhibit &alpha;-amylase, &alpha;-glucosidase and tyrosinase was determined by invitro assays. Antiproliferative activity of subcritical waterextracts was defined by testing their influence on the growth ofthree histologically different cell lines.Anti-intestinal motility activity of both group of extracts (nativeand fermented) was determined by in vivo experiments.

Abstract:
This thesis offers a comparative analysis of geographical themes in the works of two contemporary Pakistani women writers, Kamila Shamsie and Uzma Aslam Khan. In view of the importance of Pakistan’s geostrategic location, contemporary Pakistani writing in English has garnered critical attention in the context of its political perspectives. However, this thesis argues that given the inextricable intertwinement of geography with history and politics since the spatial turn, the geographical within the geopolitical deserves equal attention in the context of Pakistan. In the thesis, I apply a methodological framework derived from phenomenology and feminist geography in order to conduct a study of the representation of Pakistani terrestrial, marine and aerial geographies in Shamsie’s and Khan’s novels. More specifically, I evaluate Shamsie’s and Khan’s return to Pakistani ground reality, sea topography and aerial wildernesses via tropes associated with cartography and palaeontology: tropes of feminist transnational mobility that gauge the political, social and cultural implications of the spatial turn. The thesis concludes that a return to geography and geology in Shamsie’s and Khan’s fiction offers the possibility of a reorientation of Pakistani identity and a partial resolution to the contested notions of Pakistani belonging and citizenship in keeping with the inherent diversity and pluralistic dimensions of its geographical terrain. It also offers Pakistan possibilities of a more peaceful convergence and engagement with its international partners. This alternative rerouting/rerooting of identity foregrounds the potential benefits of a more intimate engagement with the geological and geographical aspects of Pakistani space.
