Nuclear factor erythroid-2-related factor 1 (NRF1) and NRF2 are essential for maintaining redox homeostasis and coordinating cellular stress responses. They are highly homologous transcription factors that regulate the expression of genes bearing antioxidant-response elements (AREs). Genetic ablation of NRF1 or NRF2 results in vastly different phenotypic outcomes, implying that they play different roles and may be differentially regulated. Laminitis is a common and extremely painful hoof disease in horses. We know that it is caused by abnormally high levels of insulin, but the mechanism of insulin action is not known. One theory is that insulin over-stimulates the receptors for a related hormone, insulin-like growth factor-1, and that this leads to uncontrolled cell proliferation in the hoof, which ultimately causes the disease. Although signaling pathways promoting GLP-1 secretion are well characterized, the mechanism by which GLP-1 containing granules fuse to the L-cell membrane remain elusive.
