Abstract:
The aim of this diploma thesis is to describe historical development of Kenya, analyze its current socioeconomic and political situation and characterize major problems and comparative advantages, which are crucial for its future development. The main contribution of the thesis are my own findings and experience gained during cultural internship in Kenya. The first chapter describes historical development from the beginning of the first settlement on its territory to post colonial era. The second part focuses on demographic indicators, sectorial economic structure, development of macroeconomic indicators in the context of east Africa, political situation and on governmental program called Vision 2030. The last part characterizes key Kenyan advantages and problems with focus on tourism, education and political and religious conflicts. The thesis is closed with future economic prognoses for Kenya.

The aim of this final thesis is to evaluate the impact of British colonialism on Nigeria and Kenya, to describe positives and negatives. Attention is paid to relations between countries in the period of colonial rule, the development of nationalist movements, the transition to independence, and contemporary relations between the countries. The aim is also to identify differences in the colonization of Nigeria and Kenya. For each country there are mentioned important factors affecting the development of political and socio-economic relations.

Abstract:
The thesis discusses economic development and fight against poverty in Kenya.The aim is to prove that the development assistance is not the the main incentive for the economic development in Kenya and the kenyan government should focus on alternative development paths. The paper tries to analyze development assistance and to find arguments against it, describes other forms of capital inflow, such as foreign trade, foreign investment and microfinance. The first chapter defines the basic concepts and puts the arguments for and against development aid. The second chapter discusses alternative paths of development. In the third chapter the paper starts with the analytical section where political and economic situation in Kenya, development aid and business environment are analyzed. The importance of foreign trade, foreign investment and microfinance is demonstrated. The last chapter analyzes the selected microfinance institutions in Kenya and it contains a case study from the field.

Abstract:
The aim of my Thesis is to analyze the way how Kenya gained independence, how the process of democratization had conducted and the development in the 21th century. The reason for the focus on Kenya is that it is a country which I am interested in and I want to get the new information. The work is divided into five chapters. The first chapter gives information about the current economic, geographic, demographic and political situation in the country. The second chapter focuses on the historical development till 1944. Includes description of the oldest inhabitants of the planet Earth, the original inhabitants of the country, settlers and colonizers. Year 1944 is chosen deliberately, this is the year when first Kenya had been elected to the National Assembly. The third chapter deals with the situation in the country until 1963, when the country was granted independence. This chapter deals with the rebellion Mau - Mau, congresses at which was discussed the constitution of an independent country, important personal proces sof independence - the first and the second president Jomo Kennyat and Daniel Arap Moi's regime. The fourth chapter gives information about the process of democratization. Here I describe how the process of democratization in Kenya had conducted and all the things that happened in the country since independence. The fifth chapter seeks to answer the question of why we can not say that Kenya is a democratic state. Describe here the notion of tribalism, which has a major impact on what is happening in the country. The members of the nation are loyal to their ethnicity in any case, and therefore it is not possible to develop democracy in the country in the literal sense.

Abstract:
This thesis is focused on the elimination of slums in Kenya. It is currently facing a huge challenge - improving the quality of life of slum dwellers. Despite the large amount of efforts developed to promote the slums in Kenya, the situation remains problematic and local slums has already grown to such proportions that it was not easy efficient and sustainable. Thesis is divided into three subsequent chapters. The first chapter focuses on the economic, political and social situation in Kenya. In the second chapter I explain term slum, it's history, the life of slum dwellers and I also think about causes of their origin and subsequent dynamic urban sprawl. The third chapter is focused on the actual Kenyan slums and projects dedicated to tackle the challenges of their elimination. My goal is to identify the causes that lead to the extension of slums. Further I want to analyze how the issue of slums in Kenya is solved and whether these projects are effective and sustainable.

The first part of this paper presents insight in contemporary academic discussion about deeper determinants of economic growth. In the second theoretical part, the role of state in economic development is discussed from five perspectives -- the perspective of neoclassical development economics, neoliberal approaches, the perspective of providing public services, integration in international trade and the perspective of institutional factors with impact on state structure and functioning. The analytical part compares development experience of Kenya, Uganda and Tanzania (from the mid 60s) from the above mention five perspectives. Then it is concluded which policies and institutions in case of Kenya, Uganda and Tanzania were yielding unsatisfactory results in terms of economic development.

Hlavním cílem mé diplomové práce je analyzovat současný stav cestovního ruchu v Keni, jeho význam v národním hospodářství a zjistit, zda má země dostatek předpokladů být destinací cestovního ruchu. Stanovila jsem si dva dílčí cíle: 1. Navrhnout, jakou novou strategii by měla Keňa uplatňovat v rozvoji cestovního ruchu, aby z části dokázala zmírnit chudobu svých obyvatel, tzn. jak by cestovní ruch mohl pozitivně ovlivnit životní úroveň keňských obyvatel. Budu se věnovat především alternativním formám cestovního ruchu. 2. Zjistit, jak si Keňa stojí v rámci cestovního ruchu mezi ostatními africkými zeměmi. V diplomové práci pracuji s následujícími hypotézami, které se pokusím potvrdit či případně vyvrátit. Hypotéza 1: Keňa má dostatek předpokladů být destinací cestovního ruchu. Hypotéza 2: Rozvoji cestovního ruchu nejvíce brání nedostatečná suprastruktura a nevzdělané obyvatelstvo. Hypotéza 3: Keňa disponuje dobrými podmínkami pro rozvoj alternativních forem cestovního ruchu. Hypotéza 4: Keňa má před sebou velkou budoucnost v oblasti cestovního ruchu v rámci afrických zemí. Hypotéza 5: Keňa se začíná orientovat na nové formy cestovního ruchu. Záměrům a cíli diplomové práce odpovídá struktura členění.

Abstract:
This diploma thesis focuses on the analysis of the "phenomenon" of politicization of etnicity in Kenya, its impacts on temporary situation and future development of the country. Through the analysis of the sources and development of the phenomenon it makes an effort to explain the motives of the post-election violence in 2007 and review the temporary constitutional reform process focused on its impacts on political ethnization and minimalization of the social conflict. However for a long time, Kenya used to be perceived as a politically stable and prosperous African country, where many European tourists spent their holidays, the post-election violence in 2007 harshly damaged the positive international reputation of the country. Fortunately, an agreement between the two main presidential candidates was promptly reached and finally in August 2010 Kenyans peacefully passed the constitutional reform. There are doubts, whether they manage to reconcile the latent grievances of the past and settle ethnic conflicts of the Kenyan society to become a model to other multiethnical countries in Africa.

The thesis analyses development cooperation in Kenya, including an example of personally realized development project. The goal of the thesis is to find out what is the current state and direction of development cooperation in Kenya, demonstrate some of the problems of Kenya and their possible solutions using an example of personally visited region and realized development project and to determine whether this project corresponds to the current trend of development cooperation in the world and in Kenya. The thesis examines the Kenya's cut-flower industry in the wider context of global commodity networks. The Kenya's floriculture sector as a part of these networks has been influenced by notions of environmental and social responsibility in the past two decades. The first part of the thesis describes floriculture in Kenya and the cut flower supply chain. The second half analyses current ethical trading practice in the flower industry in Kenya. In this respect codes of conduct which embody notions of responsible business in practice are discussed here. In view of the fact that codes can't guarantee adequate working conditions to flower farm workers in many cases the thesis tries to explain why this concept of ethical trade failed.

Abstract:
In this study, a number of experiments were undertaken to investigate possible methods for effective improvement of the texture and flavour of canned late-run chum salmon. These included removal of the skin and bones from the fish, processing of the boneless-skinless steaks in retort pouches, brine treatment using two washes with an 8% salt solution for one hour each time, and a precanning treatment in which the boneless-skinless steaks were soaked in a solution of 10% tripolyphosphate and 2% brine for two minutes. Only fish of advanced sexual maturity were used. The canned salmon was steam processed at 120°C for 65 minutes in an FMC laboratory retort. This was based on a known commercial process for 307 x 115 cans. Heat penetration studies were carried out to design the process schedules for the pouched samples. It was found that the pouched product required 48% less thermal processing time than the canned product to achieve similar lethality. Sensory results showed that the removal of the skin and bones did not produce any significant improvement in the flavour and acceptability of the fish. There was no significant difference between the polyphosphate/brine samples and the untreated (control) samples for all attributes tested. The brine treatment also did not improve the texture of the samples. However, there was less detection of late-run flavour in the brine treated samples when compared to the control. Comments offered by panelists described these samples as having a salty/briny flavour. Pouched samples had a firmer, drier and more fibrous texture than the canned product. They also scored better in terms of late-run flavour. Acceptance of the fish however was only moderate. As a consequence, although this study demonstrated an improvement in the texture and flavour of the pouched late-run chum in comparison to the canned product, it was concluded that a more acceptable pouched product could probably be obtained by using late-run salmon of less advanced sexual maturity. Results of linear regression analysis showed that significant relationships were obtained between sensory firmness, fibrousness and chewiness and instrumental hardness, maximum slope and chewiness. However, none of the sensory parameters were well predicted by the instrumental results.

Abstract:
To test the hypothesis that temporal isolation due to differences in season of breeding could result in genetic divergence among chum salmon populations, I compared the reproductive environments, phenotypes and genotypic performances of early and late season breeding stocks of chum salmon, Oncorhynchus keta. These were autumn (AB) and winter (WB) spawners from one Vancouver Island stream (Bush Creek) and another winter spawning stock (W) from a nearby stream (Walker Creek). All stocks had similar time of downstream migration of the fry. No differences were found among the W and AB stocks in age at maturity, length composition of spawners, egg size, vertebral count of adults and fry and time of fry migration. The WB stock differed from the other stocks in egg size and vertebral number of adults and fry. In 1981, WB spawners were larger than AB spawners. In 1982, WB spawners were younger than AB spawners. Analysis of ten external morphological features of the fry revealed that there was considerable overlap in body form of the stocks. To test the hypothesis that phenotypic similarities observed among seasonal ecotypes in the wild were due to genetic differences, sample populations were reared under controlled conditions in the laboratory. When reared at 6°C, under the autumn - winter - spring progression and the winter -spring progression, the early spawning population in Bush Creek took significantly more time to hatch and emerge than the spatially and temporally isolated population of Walker Creek, and the combined average of the late spawning populations. At 10°C the populations had similar incubation rates. Temperature regime, population and temperature regime by population interaction all had significant effects on vertebral number. AB progrency were morphologically distinct from WB and W. These results indicate that the early spawning population differs genetically from the late spawning populations. To test the hypothesis that selection on the speed of development from fertilization to hatch and emergence would reduce the additive genetic variation in these traits, individual families were reared from AB, WB and W. Heritabilities were found to be between 0.27 and 0.54. The high heritabilities suggest that some countervailing force is opposing selection on incubation rate. Analysis of diversity using biochemical genetics techniques suggest that genetic migration goes from the WB stock to the AB stock. Genetic distance measures and a Wagner-Tree analysis indicate that biochemically AB and WB are more closely related than W. It is postulated that the genetically determined program for incubation which results in synchronous downstream migration has survival value. Survival could be enhanced by predator satiation or synchrony with food resources in the estuary.

In order to develop a stable strain of Bacillus megaterium for Keratinase production, the Keratinase gene (KerA) of Bacillus lichiniformis ATCC 53757 and SPlipA gene from plasmid pHIS1525.SPlipA (Bacillus megaterium origin) were PCR amplified and constructed to give a gene cassette called SPK . Then the gene cassette SPK was cloned into the Integration vector, pMUTIN-GFP+ 6192bps and transformed in Bacillus megaterium ATCC 14945. The chromosomal integration was created using homologous single crossing over mechanism. The strong natural promoter from the chromosomal locus of the SPK not only produced the increased extracellular enzyme, but also functions as a non inducible promoter which does not require any inducer for the production of the enzyme in the new integrant strain. The integrant strain was subjected to feather degradation test and found that it could totally digest the feather meal in complete seven days, resulting in a rich fermentation broth.

Coastal indigenous communities in Keta, Ghana, are experiencing resettlement as a result of slow-onset, climate-induced flooding and erosion. Previous researchers have documented the risk of relocation from rapid-onset events, but little is known about the effectiveness of policies developed in response to slow-onset changes. This phenomenological study investigated the ongoing lived experiences of adult household members in Keta who were relocated by the government. Jun's critical theory provided a constructionist interpretive framework to determine whether Ghana's national policy on climate change resettlement adequately meets Rawls's criteria for distributive social justice. Policy documents and transcriptions of interviews with a purposeful sample of 17 family members were thematically coded and categorized into essence descriptions. Results revealed aligned perceptions of an absence of justice or fairness in the allocation of resources to households relocated by the government. Negative experiences characterized all families' resettlement processes. The government's commitment to ensuring basic community welfare was perceived to be poor. Findings highlight the need for social justice to be the primary policy consideration for future allocation of benefits to resettled households. To avoid reaching a tipping point at which prompt governmental intervention will be either compelled or impossible, quantitative studies are needed to guide policymakers in considering the real costs of relocation and the cumulative effects on families and communities. This study provides evidence for public consideration of the severe consequences of injustice in relocation and the need to prevent human rights abuse in the formulation of social, economic, and cultural policies associated with climate-induced resettlement.

Abstract:
Interactions between fish utilizing nearshore habitats of the Fraser River estuary were investigated by field observations and laboratory experiments. Chum salmon fry (Oncorhynchus keta) were the most abundant salmonid captured between April and June. Non-salmonid species captured included: threespine stickleback (Gasterosteus aculeatus), prickly sculpin (Coitus asper), and peamouth chub (Mylocheilus caurinus). Potential predators included: prickly sculpin, and northern squawfish (Ptychocheilus oregonensis), but few had been feeding on fish. Chum fry fed predominantly on surface insects but the proportion of benthic prey in the diet increased over time. Stickleback shared the greatest diet overlap with chum fry. Interactions between two dissimilar prey, chum fry and threespine stickleback, and a predator, cutthroat trout (Oncorhynchus clarki) were investigated in the laboratory. Prey response to hungry and satiated predators was related to the degree of risk. Although attack rates by trout on chum and stickleback were similar, trout captured more stickleback than chum, but consumed both prey at similar rates. I tested the hypotheses that prey foraging efficiency is reduced in the presence of a predator and increased in the presence of alternate prey. When alone, chum fed on surface Drosophila and mid-water Daphnia, while stickleback fed on benthic Tubifex and Daphnia. The feeding efficiency of chum increased in the presence of stickleback and decreased in the presence of trout. Hatchery and wild chum showed opposite dietary shifts in the presence of trout. Hatchery chum shifted from surface to mid-water feeding and the number of fish feeding significantly decreased. Wild chum fed at the surface, at significantly decreased feeding rates. In the presence of stickleback and trout the feeding behaviour of chum was similar to that when chum were alone. Stickleback feeding behaviour was not affected by presence of trout or chum. Chum and stickleback detected Daphnia faster than Drosophila or Tubifex, and chum responded to Daphnia significantly faster than stickleback. Foraging time per item was significantly less for chum than stickleback. Habitat use by fish prey was investigated in the presence and absence of trout and alternate prey. Wild chum shifted from mid-water to the surface in the presence of trout, but returned to mid-water when stickleback were present. Stickleback fed in bottom habitats regardless of the presence of trout or chum. When prey were confined to specific depths in the water column, trout attacked chum more frequently than stickleback in all locations and attacked both prey more frequently within 24 cm of the substrate. Movement by prey did not affect the attack rate. When given a choice between a food-rich open water habitat and a food-deficient vegetated habitat in the presence of trout and alternate prey, chum and stickleback used vegetated refugia significantly more in the presence of trout. Alternate prey presence decreased the proportion of chum but increased the proportion of stickleback using vegetation. Behavioural responses to avoid predation significantly reduced the foraging efficiency of prey. Chum showed stronger responses to trout than stickleback. The presence of stickleback reduced the effect of predation on foraging efficiency. Possible explanations for the positive effect of stickleback on chum feeding efficiency were experimentally examined including: social facilitation, reduced intraspecific competition, and the calming influence of stickleback on chum behaviour ("dither"). The results suggest that stickleback have a calming influence on chum behaviour and that mixed species feeding groups may reduce intraspecific competition.

The performance of astronomical interferometer arrays operating at (sub) millimeter wave-lengths is seriously compromised by rapid variations of atmospheric water vapour content that distort the phase coherence of incoming celestial signals. Unless corrected, these phase distortions, which vary rapidly with time and from antenna to antenna, seriously compromise the sensitivity and image quality of these arrays. Building on the success of a prototype infrared radiometer for millimeter astronomy (IRMA I), which was ued to measure atmospheric water vapour column abundance, this thesis presents results from a second generation radiometer (IRMA II) operating at the James Clerk Maxwell Telescope (JCMT) on Mauna Kea, Hawaii from December, 2000 to March, 2001. These results include comparisons with other measures of water vapour abundance available on the summit of Mauna Kea and a comparison with a theorteical curve-of-growth calculated from a new radiative transfer model, ULTRAM, developed specifically for the purpose. Plans for a third generation radiometer (IRMA III) are also be discussed.

Abstract:
The kea is the only parrot species in the world to include the true alpine environment as part of their habitat. Survival in these harsh alpine conditions has been hypothesised to be the cause of the generalist behaviour of kea, leading to their heightened explorative behaviour and curiosity. Kea are also widely regarded as being extraordinarily intelligent. It is their intelligence that suggests that kea may possess a sophisticated communication system. I conducted a study exploring the potentially complex vocal repertoire of the kea. My study was conducted with wild population of banded juvenile and fledgling kea in Aoraki/Mount Cook National Park between February 2003 and April 2004. I obtained 449 vocalisations from 16 identified and several unidentified kea. The results of this study confirmed that the vocal repertoire of the kea is exceptionally large for a parrot species, encompassing over 17 vocalisations. This study revealed five vocalisations previously undescribed in the kea repertoire and showed for the first time that vocal repertoire of immature kea may be different to the repertoire of adult kea. Two possible gender specific vocalisations were also revealed. The study of apparent vocal responses revealed that kea appear to be able to identify vocalisation types and respond accordingly using combinations of increasingly complex vocalisations. This is also the first study to take advantage of the similarity between human and parrot vocal systems for the kea, by utilizing powerful human speech analysis software. The results of this analysis allowed the identification of subtle differences in kea vocalisations, including the presence of graded signals, not identifiable by use of spectrogram analysis.

The central premise of this dissertation is that a pastorally sympathetic assessment of cultural bereavement healing approaches in the light of a biblically informed understanding of death, bereavement and afterlife provide a basis for the bereaved to cope with the angst associated with bereavement through death and engenders pastoral care among the Abaluyia. It is noted that although many Abaluyia Christians consciously or otherwise lurch back to cultural approaches for consolation and healing during bereavement, there is a lack of meaningful engagement between the Abaluyia culture and the gospel. The result of this lack of engagement has not been syncretism as commonly assumed but rather parallelism whereby, on the one hand, the gospel is held as though it were alien and whimsical in contrast with traditional beliefs and practices which are perceived as practical and consequential. On the other hand, cultural approaches are dismissed as being part of ancestor worship or superstition and thus contrary to the gospel. In view of these extreme perceptions, Abaluyia Christians often feel guilty for either engaging in or shunning cultural approaches thus raising a pastoral concern in relation to caring for the bereaved. In response to the aforementioned pastoral concern, this dissertation firstly examines the role of cultural beliefs and practices in mediating healing for the bereaved among the Abaluyia. It is suggested that the persistence of cultural models is informed largely by their therapeutic value. Consequently, attention is put on the efficacious beliefs and practices employed by the Abaluyia which include community based approach to bereavement, rituals, systems of inheritance and care, conduct of the bereaved, procedures for forgiveness and reconciliation, hospitality, acts of benevolence and performances. Secondly, the identified cultural approaches are reflected upon in the light of the normative biblical teachings. The reflection is done in order to expose and isolate cultural beliefs and practices that are at odds with biblical teachings. The reflection follows a systematic analysis undertaken to establish the biblical understanding of death and the afterlife as well as a biblical hermeneutical analysis of pertinent biblical passages in order to draw paradigmatic biblical practices in bereavement. It is argued that the biblical ministry of healing in bereavement is rooted in the hope of resurrection and the conception of God as compassionate in his character. Lastly, an integrated pastoral care model, namely, Circle for Pastoral Concern, is proposed as a tool for bereavement healing among the Abaluyia. The proposed integrated model is cognisant of local cultural milieu in engaging in pastoral care for the bereaved. According to the Circle for Pastoral Concern model, the community of believers in their local setting, under the Triune God, form the context for healing.

Abstract:
The spawning migration of chum salmon results in a deterioration of the eating quality of the flesh. There is a loss in the colour of the muscle from red-pink to white-grey, a distinct off-flavour develops and the firm texture becomes soft and watery. The reduced appeal of the canned and fresh product results in an economic loss of this important B.C. commodity. Chum salmon were sampled at four stages during their spawning migration in order to describe the physical, chemical and structural changes which occurred as the fish matured. The fish were categorized into four grades based on their external characteristics. Grade one, silver-brights, were the least mature fish, grade two, intermediates, and grade three, darks, were progressively more mature. Grade four, spawning, were the most mature. Proximate analysis of the raw muscle revealed a significant decrease in protein and lipid and increase in moisture. The level of astaxanthin and the Hunter values a/L, a and a/b decreased significantly with increasing grade. The texture of the canned fish was measured instrumentally with an Instron Universal Testing Machine using the texture profile analysis (TPA) method. A panel of semi-trained judges also evaluated the canned flesh based on its firmness, fibrousness, dryness, and chewiness. The sensory panel determined that the silver-bright fish were significantly more firm, fibrous, dry, and chewy than the spawning fish. Of the ten instrumental TPA parameters, cohesiveness and slope 2 were able to distinguish between grade one and four. Poor correlation between the instrumental and sensory measure of texture suggested that the softening characterized by the sensory panel was not being measured by the Instron. Thin and ultrathin sections of the muscle were photographed at the light and electron microscopic level. The light micrographs clearly showed an apparent loss in the myofibrillar diameter or number within muscle fibres as the fish became more mature. Focal degeneration of myofibrils was evident in the electron micrographs. Changes were described by four criteria, smearing, the loss of a distinct appearance of the actin and myosin, splitting, the tearing of myofibril bundles, thinning, a loss in myosin or actin filaments, and an increase in intermyofibril space. Proteinase activity of the dorsal muscle was assayed at pH 3.5, 6.2 and 7.0. There was a significant increase in activity at pH 3.5 but no change at pH 6.2 or 7.0. Acid phosphatase, a lysosomal marker, also increased significantly with grade. These findings suggested that lysosomal enzymes largely contributed to the degradation in the muscle.

Abstract:
This thesis uses acoustic measurements as a basis for the phonological analysis of the interaction of tone with voicing and foot structure in Kera (a Chadic language). In both tone spreading and vowel harmony, the iambic foot acts as a domain for spreading. Further evidence for the foot comes from measurements of duration, intensity and vowel quality. Kera is unusual in combining a tone system with a partially independent metrical system based on iambs. In words containing more than one foot, the foot is the tone bearing unit (TBU), but in shorter words, the TBU is the syllable. In perception and production experiments, results show that Kera speakers, unlike English and French, use the fundamental frequency as the principle cue to 'Voicing" contrast. Voice onset time (VOT) has only a minor role. Historically, tones probably developed from voicing through a process of tonogenesis, but synchronically, the feature voice is no longer contrastive and VOT is used in an enhancing role. Some linguists have claimed that Kera is a key example for their controversial theory of long-distance voicing spread. But as voice is not part of Kera phonology, this thesis gives counter-evidence to the voice spreading claim. An important finding from the experiments is that the phonological grammars are different between village women, men moving to town and town men. These differences are attributed to French contact. The interaction between Kera tone and voicing and contact with French have produced changes from a 2-way voicing contrast, through a 3-way tonal contrast, to a 2-way voicing contrast plus another contrast with short VOT. These diachronic and synchronic tone/voicing facts are analysed using laryngeal features and Optimality Theory. This thesis provides a body of new data, detailed acoustic measurements, and an analysis incorporating current theoretical issues in phonology, which make it of interest to Africanists and theoreticians alike.

A YakA é uma proteína quinase necessária para a regulação da resposta a diversos estresses em Dictyostelium e é uma efetora chave da transição do crescimento para desenvolvimento nesse organismo. O gene keaA foi isolado como um supressor do mutante yakA- em uma busca para revelar genes envolvidos na sobrevivência a estresse nitrosoativo. O gene keaA codifica uma proteína com seis repetições kelch na porção C-terminal, um domínio zf-C3HC4 na porção N-terminal, também chamado RING-finger, e uma sequência rica em cisteínas localizada na porção mediana da proteína. Mutantes deficientes em keaA foram avaliados revelando-se um papel para esse gene também no processo de desenvolvimento. A expressão de mRNA de keaA é induzida quando células selvagens crescem e a fonte de alimento começa a esgotar. A indução do mRNA de keaA também ocorre durante o desenvolvimento. Essa indução não é observada em células yakA- indicando que YakA regula KeaA. Células deficientes em keaA expressam baixos níveis de mRNA de pkaC, acaA e carA durante a agregação em baixa densidade celular, o que pode explicar porque as células deficientes em keaA apresentam o processo de desenvolvimento mais lento em baixa densidade celular. Células deficientes em keaA são mais resistentes a estresses nitrosoativo e oxidativo e keaA é necessário para a produção e detecção de AMPc. A análise da agregação de células deficientes em keaA durante o desenvolvimento multicelular indica que KeaA é necessário para que as células participem eficientemente desse processo. Células que super-expressam o domínio rico em cisteínas levam o mesmo tempo que as células selvagens para atingir o estágio de agregação. No entanto, essas células apresentam agregados, culminantes e corpos de frutificação menores. Células que super-expressam o domínio Kelch expressam altos níveis de acaA e carA depois de 8 horas de desenvolvimento, mas os níveis de pkaC são similares aos observados em células selvagens. Isso poderia indicar que o domínio Kelch induz a ativação da PKA. No entanto, essa interação não foi observada em experimentos de duplo híbrido. The YakA is a protein kinase required for the regulation of several stress responses in Dictyostelium and is a key effector of the transition from growth to development in this microorganism. The gene keaA was isolated as suppressor of the yakA- mutant in a screen targeted to reveal genes involved in the survival to nitrosoative stress. The keaA gene codes a protein with six kelch domain repeats at the C-terminus, a zf-C3HC4 domain at the N-terminus, also called RING-finger, and a cysteine-rich sequence located in the mid-portion of the protein. The analysis of mutants deficient in keaA revealed a role for this gene also in the development process. keaA mRNA expression is induced when wild type cells grow and the food source becomes scarce. An induction of keaA mRNA expression also occurs during development. This induction is not observed in yakA- cells, indicating that YakA regulates KeaA. keaA deficient cells express low levels of pkaC, acaA and carA mRNA during aggregation in low cell densities, which may explain why the keaA deficient cells present a delay in development in low cell densities. keaA deficient cells are more resistant to nitrosoative and oxidative stress and keaA is necessary for the production and detection of cAMP. The analysis of agreggation of keaA deficient cells during multicellular development indicated that KeaA is required for the cells to efficiently participate in the process. Cells over-expressing the cisteine-rich domain took the same time as the wild-type cells to reach the aggregation stage. However, these cells present smaller aggregates, culminants and fruiting bodies. Cells over-expressing the Kelch domain express high levels of acaA and carA after 8 hours of development, but the levels of pkaC are kept similar to those in wild-type cells. This could indicate that the Kelch domain induces the activation of PKA. However, this interaction was not observed when we conducted tests in a two-hybrid system. Additionally, gene expression in response to compounds that generate redox stresses was studied using cDNA microarrays. This minithesis seeks to understand why infrastructure projects fail to contribute effectively to poverty (rural) reduction. The thesis assumes that though infrastructure provision can impact positively on rural poverty reduction, the same infrastructure provision has worsened or put people in worse conditions of poverty. Therefore it is not automatic that infrastructure provision would reduce rural poverty as often held. The thesis goes on to postulate that a positive relationship between infrastructure and rural poverty reduction is best achieved within a broad or generic policy which provides the framework for providing such infrastructure. The thesis assesses these assertions empirically by first, testing the relationships between infrastructure and rural poverty reduction. Here a large scale infrastructure project in Ghana known as the Keta sea defence project serves as the case study. Secondly the thesis assesses Ghana&rsquo

O ciclo de vida de Dictyostelium discoideum é composto de duas fases independentes. Durante o crescimento vegetativo, as amebas crescem isoladas até que a fonte de nutrientes seja esgotada. A carência nutricional induz sua entrada num processo de desenvolvimento, que inclui a parada do crescimento, a agregação das células e a formação de um organismo multicelular onde as células se diferenciam em esporos que sobrevivem as condições desfavoráveis. A proteína quinase YakA é requerida para a transição entre o crescimento e o desenvolvimento. YakA regula os níveis da PKA. Mutantes yakA- apresentam o crescimento acelerado, são deficientes no processo de agregação e são hiper-sensíveis a estresse oxidativo e nitrosoativo. Uma mutação em um segundo sítio em keaA, suprime a morte induzida por SNP (um gerador de óxido nítrico) no mutante yakA-. O papel de keaA foi determinado em resposta a estresse oxidativo, nitrosoativo e carênica nutricional. O gene keaA é necessário para o crescimento e desenvolvimento. Uma mutação em keaA confere resistência a estresse nitrosoativoloxidativo confirmando que uma mutação em keaA confere resistência a estresse. Um segundo supressor da morte induzida por SNP no mutante yakA- foi isolado pela mesma técnica de REMI e identificado como pkaC um regulador da resposta a estresse. YakA e PKA integradam a resposta a vários estresse em Dictyostelium. Dictyostelium discoideum\'s life cycle is composed of two phases. During the vegetative phase, amoebae grow as single cells until the nutrients are depleted. Starvation induces a developmental program where isolated amoebae adopt a multicellular mode of living and differentiate into spores to survive the harsh conditions. The protein Kinase YakA is required for the growth to development transition. YakA regulates the leveis of PKA. yakA null cells have a faster cell cycle, are aggregation deficient and are hypersensitive to nitrosoative/oxidative stress. A second-site mutation in keaA, supresses the death induced by SNP, a generator of nitric oxide. The role for keaA has been determined in response to oxidative, nitrosoative and nutrient starvation stresses. keaA is required for proper growth and development. The keaA- cells are more resistant to nitrosoative and oxidative stress confirming that a mutation in keaA confers stress resistance. A second supressor of the death induced by SNP in the mutant yakA- was isolated with REMI and identified pkaC as a regulator of the nitrosoative stress response. YakA and PKA may integrate the responses to several stresses in Dictyostelium. The results indicate that yakA regulates the cell cycle arrest in response to stress through the modulation of keaA. KeaA regulates the expression of pkaC, a key regulator of cAMP prodution and development. Genetic interactions among these elements is complex and must be finely tuned in the many environmental changes cells endure during the life cycle.

Abstract:
Within the environmental approach for the study of fluctuations in fish population abundance, factors that may regulate the overlap in time and space (phasing) of fishes, food supply, and predators, are sought. This trophic phasing analysis is based on the recognition that production of food is a process at least partially independent of the production of consumers. Trophic phasing analysis was applied in investigating production of chum salmon in the Fraser River estuary. Juvenile chum salmon were captured near a tidal flat; the abundance of salmon near the flat was highest in late May in 1985 and in early June in 1986. These salmon relied heavily on harpacticoid copepods as a food source. Individual taxa as well as the assemblage of main prey harpacticoids also had periods of highest abundance in the water column. The blooming period of the prey harpacticoid assemblage coincided with the appearance in the sediment of warming episodes. These warming episodes result from interactions between the daily heat cycle and specific tide patterns. Variations in the degree of overlap of the periods of highest abundance of salmon and harpacticoids could affect the survival of the fishes. The degree of overlap of those periods was hindcasted using indices for the temporal patterns of abundance of salmon and harpacticoids on the flat. The median date of downstream migration at a counting station upstream was used for the salmon; the timing of the second annual occurrence of tide conditions giving rise to a warming event in the sediment was used for harpacticoids. Difference in time between the two events was taken as a phasing index accounting for two degrees of freedom in the process of fish production. There is suggestion of a non-monotonic relationship between the index of survival of even broodyear chum salmon and the hindcasted phasing index.

Abstract:
The project Kera-Plast aims to re-loop humans and nature by questioning the current systems and ethics through materiality. Human hair, currently considered as waste, functions as the base for the material exploration fabricated through thermo-compression molding. The flexible, short and opaque keratin-fibers get glued together with heat, pressure and water, acting as a plasticizer during the compression molding process. The results are stiff and remind on plastic due to shine and translucency. Aesthetics and function of the resulting material are controlled and designed by traditional textile techniques as knitting, weaving and non-woven processes. The material samples display the potential of Kera-Plast in the categories of 3D surface structures, patterns, shapeability and the influence of light. The findings also provide information about the parameters for designing with keratin fibers through the thermo-compression process. It can be concluded that despite all ethical and cultural factors, Kera-Plast and its fabrication method has the potential to add a sustainable, functional and aesthetical value to the design field and our future material consumption.

Abstract:
In response to the increasing need for ecosystem services throughout the Southeast Alaska region, decision makers are tasked with balancing the need for natural resources with salmon conservation. However, accurate historical and current information on salmonid population abundance, freshwater distribution, and habitat quality are sparse with limited resolution for large portions of this remote and rugged landscape. Here, I created Intrinsic Potential (IP) models for chum and pink salmon to predict the potential for portions of coastal rivers to provide high-quality spawning habitat. I developed IP models for both species from field redd surveys and synthetic habitat variables derived from 1-m resolution digital elevation models. The surveys were performed at 49 study reaches in five coastal drainage basins on the north end of Chichagof Island, Southeast Alaska. I used a spatially balanced random sampling design that included field surveys for redds during two field seasons with contrasting precipitation patterns and disparate adult salmon escapements. The IP models predict probable spawning habitat for both species based on persistent landform characteristics and hydrologic processes that control the formation and distribution of spawning habitat across the landscape. Selection of persistent reach variables for both species IP models was informed by principal component analysis (PCA), resource selection ratios, random forest modeling, and regression models of field and synthetic variable comparisons. I observed primarily one spawning strategy by chum salmon associated with mainstem channels, and two distinct spawning strategies for pink salmon related to small moderate-gradient channels and tributaries, and lower drainage basin mainstem channels. The relationships suggest that chum and pink salmon primarily selected for unconstrained channel types in large-and small-size channels, with chum salmon being more selective toward the larger mainstem channels, and pink salmon selecting for smaller channels and tributaries. The prediction of chum salmon redd presence within a specific reach for both high and low streamflow regimes was explained by channel gradient, floodplain width, and mean annual flow in order of importance. In general, chum salmon redds were observed in larger unconstrained low-gradient floodplain reaches where accumulation of deposited gravels and adequate flow produce habitat heterogeneity suitable for spawning. Pink salmon redd presence for both survey years was explained by channel gradient, reach elevation, and mean annual flow, in order of importance. Specifically, when flows allowed upstream access, spawning pink salmon utilized smaller moderate-gradient channels where substrate size and flows were better suited to their smaller body size. Remotely sensed persistent fish habitat data is valuable information for helping understand fish population distributions across the landscape. These synthetic metrics enabled the identification and evaluation of persistent landscape features as probable predictors of IP. Validation of LiDAR-derived channel characteristics indicated channel lengths measured from the DEM were 12% longer than field measured channel length, primarily for channels wider than 10 meters. Thus, understanding the limitations of the data is important so that decision makers do not unintentionally set unrealistic objectives. This research highlights the utility of using IP models with high resolution remote sensing to expand known distributions and quality of spawning habitat for these two species in Southeast Alaska coastal streams.

Hoy en día, la cantidad de información electrónica en forma de texto ha aumentado considerablemente por lo que una búsqueda de información puede traer consigo varios documentos recuperados. Posteriormente, los documentos recuperados se tienen que revisar para saber si contienen lo que realmente se busca. Una manera de simplificar una búsqueda es el empleo de palabras o frases clave ya que actúan como filtro en un campo de búsqueda. Las palabras o frases clave se utilizan en muchas áreas, por ejemplo, la mercadotecnia y publicidad, en donde el objetivo es capturar la atención del público. De igual forma, en todo aquello que se quiere dar a conocer hacia el público en general. Ya sean documentos de textos impresos o electrónicos, las frases clave ayudan al lector mostrándole las ideas principales del texto. En esta tesis, se comparan los sistemas de extracción automática de frases clave sobre un conjunto de artículos científicos utilizados en la tarea 5 del SemEval-2010, con el objetivo de conocer qué sistemas pueden encontrar las frases clave que fueron propuestas por un ser humano. En la experimentación se presentan los resultados de la comparación entre los sistemas instalables y en línea. Por último, los resultados de la evaluación se comparan con los de la tarea 5 del SemEval-2010.

Le Big Data est maintenant la nouvelle ressource naturelle. Les architectures actuelles des environnements d'analyse des données massives sont constituées de trois couches: les flux de données sont acquis par la couche d’ingestion (e.g., Kafka) pour ensuite circuler à travers la couche de traitement (e.g., Flink) qui s’appuie sur la couche de stockage (e.g., HDFS) pour stocker des données agrégées ou pour archiver les flux pour un traitement ultérieur. Malheureusement, malgré les bénéfices potentiels apportés par les couches spécialisées (e.g., une mise en oeuvre simplifiée), déplacer des quantités importantes de données à travers ces couches spécialisées s’avère peu efficace: les données devraient être acquises, traitées et stockées en minimisant le nombre de copies. Cette thèse propose la conception et la mise en oeuvre d’une architecture unifiée pour l’ingestion et le stockage de flux de données, capable d'améliorer le traitement des applications Big Data. Cette approche minimise le déplacement des données à travers l’architecture d'analyse, menant ainsi à une amélioration de l’utilisation des ressources. Nous identifions un ensemble de critères de qualité pour un moteur dédié d’ingestion des flux et stockage. Nous expliquons l’impact des différents choix architecturaux Big Data sur la performance de bout en bout. Nous proposons un ensemble de principes de conception d’une architecture unifiée et efficace pour l’ingestion et le stockage des données. Nous mettons en oeuvre et évaluons le prototype KerA dans le but de gérer efficacement divers modèles d’accès: accès à latence faible aux flux et/ou accès à débit élevé aux flux et/ou objets
Big Data is now the new natural resource. Current state-of-the-art Big Data analytics architectures are built on top of a three layer stack:data streams are first acquired by the ingestion layer (e.g., Kafka) and then they flow through the processing layer (e.g., Flink) which relies on the storage layer (e.g., HDFS) for storing aggregated data or for archiving streams for later processing. Unfortunately, in spite of potential benefits brought by specialized layers (e.g., simplified implementation), moving large quantities of data through specialized layers is not efficient: instead, data should be acquired, processed and stored while minimizing the number of copies. This dissertation argues that a plausible path to follow to alleviate from previous limitations is the careful design and implementation of a unified architecture for stream ingestion and storage, which can lead to the optimization of the processing of Big Data applications. This approach minimizes data movement within the analytics architecture, finally leading to better utilized resources. We identify a set of requirements for a dedicated stream ingestion/storage engine. We explain the impact of the different Big Data architectural choices on end-to-end performance. We propose a set of design principles for a scalable, unified architecture for data ingestion and storage. We implement and evaluate the KerA prototype with the goal of efficiently handling diverse access patterns: low-latency access to streams and/or high throughput access to streams and/or objects
