Abstract:
We have previously shown that dairy cows carrying the ‘fertil−’ haplotype for one quantitative trait locus affecting female fertility located on the bovine chromosome three (QTL-F-Fert-BTA3) have a significantly lower conception rate and body weight after calving than cows carrying the ‘fertil+’ haplotype. Here, we compared by Tiling Array the expression of genes included in the QTL-F-Fert-BTA3 in ‘fertil+’ and ‘fertil−’ adipose tissue one week after calving when plasma non-esterified fatty acid concentrations were greater in ‘fertil−’ animals. We observed that thirty-one genes were overexpressed whereas twelve were under-expressed in ‘fertil+’ as compared to ‘fertil−’ cows (P < 0.05). By quantitative PCR and immunoblot we confirmed that adipose tissue KIRREL mRNA and protein were significantly greater expressed in ‘fertil+’ than in ‘fertil−’. KIRREL mRNA is abundant in bovine kidney, adipose tissue, pituitary, and ovary and detectable in hypothalamus and mammary gland. Its expression (mRNA and protein) is greater in kidney of ‘fertil+’ than ‘fertil−’ cows (P < 0.05). KIRREL (mRNA and protein) is also present in the different ovarian cells with a greater expression in granulosa cells of ‘fertil+’ than ‘fertil−’ cows. In cultured granulosa cells, recombinant KIRREL halved steroid secretion in basal state (P < 0.05). It also decreased cell proliferation (P < 0.05) and in vitro oocyte maturation (P < 0.05). These results were associated to a rapid increase in MAPK1/3 and MAPK14 phosphorylation in granulosa cells and to a decrease in MAPK1/3 phosphorylation in oocyte. Thus, KIRREL could be a potential metabolic messenger linking body composition and fertility.

Grundtvigianisme og danskere i AmerikaAf Thorvald HansenArtiklen beskæftiger sig med to forbundne emner: den »Grundtvigianerskræk«, der i sidste århundrede fandtes inden for protestantiske kredse i USA, samt den kirkelige splittelse blandt de danske immigranter. Da det danske udvalg for kirkeligt arbejde blandt udvandrere i 1867 sendte repræsentanter til USA, konstateredes en udtalt biblicisme blandt immigranterne, kombineret med en voldsom afstandtagen fra grundtvigianismen, den nævnte »Grundtvigianer-skræk«. Dette var resultatet af en længere udvikling, hvor Grundtvigs kirkelige synspunkter oprindelig var blevet vel modtaget blandt norske immigranter og lagt til grund for det første forsøg på en norsk-dansk synodedannelse, i hvilken også den danskfødte præst Claus L. Clausen medvirkede. I 1850-erne skiftede det teologiske miljø i Norge imidlertid den oprindelig positive holdning til Grundtvig ud med en afvisning, immigranterne fulgte moderkirken og op gennem 1860-erne voksede den amerikanske modstand mod Grundtvig. Da den danske kirke som et resultat af det førnævnte udvalgs arbejde i 1871 indledte sin virksomhed, havde den derfor særdeles vanskelige vilkår.En konkret sag forværrede den i forvejen spændte situation. Den danske præst Adam Dan, - der ikke var grundtvigianer, men heller ikke i opposition til Grundtvig, - var blevet præst inden for den norsk-danske kirkedannelse.Han kætterdømtes under medvirken af den tyske Missouri Synodes lærestol. Sagen medvirkede til at umuliggøre et samarbejde mellem den norsk-danske synode og den danske kirke. Da de danskere, der hørte til den norsk-danske kirke, i 1884 i fuld forståelse med nordmændene gjorde sig selvstændige, tilsluttede de sig ikke den danske kirke, men dannede en ny gruppe the Danish Evangelical Lutheran Association, der skabte et præsteseminarium i Blair, Nebraska.Til disse den første danske kirkedannelses vanskeligheder føjedes den almindelige amerikanske puritanisme og tendensen til uniformitet inden for de protestantiske immigrantkirker. Den danske kirke prægedes af en liberal holdning og rummede både pietistiske kredse, grundtvigske miljøer og grupper uden retningspræg og havde det derfor ikke nemt, hverken udadtil eller indadtil.I 1892 førte spændinger mellem de pietistiske og ikke-pietistiske grupper til nedlæggelsen af kirkens præsteseminarium i Luck, Wisconcin. Seminariet havde været ledet af grundtvigianeren Thorvald Helveg i samarbejde med den indremissionske Peder Sørensen Vig. Nedlæggelsen af seminariet grundede sig på disses uenighed om bibelsyn og luthersk konfessionalisme, og indtil oprettelsen af Grand View College i Des Moines, Iowa, i 1897 havde kirken da ingen præsteskole, men måtte klare sig med de præster, der uddannedes på Askov Folkehøjskole i Danmark.Andre stridsemner inden for kirken var spørgsmålet om holdningen til hemmelige selskaber, eksempelvis the Danish Brotherhood, idet Grundtvigs søn, Frederik Lange Grundtvig, præst i Clinton, Iowa, markerede sig som modstander af disse; endvidere diskussionen om, hvorvidt det hørte til kirkens arbejdsområde at oprette danske kolonier, samt striden om det af F.L.Grundtvig stiftede Dansk Folkesamfund. En række præster og lægfolk følte, at Dansk Folkesamfund fremstod som et konkurrenceforetagende i forhold til kirken.I 1890-erne markerede en pietistisk orienteret gruppe sig med tidsskriftet Danskeren, og efter en lang og opslidende debat splittedes kirken i to i 1894. En tid var der således tre danske kirker i USA, men i 1896 forenede den pietistiske udbryderkirke sig med den danske kirke, der drev præsteseminariet i Blair, Nebraska. Et vist samarbejde fandt i årenes løb sted mellem kirkerne, bl.a. om udgivelsen af en salmebog. Først i 1960-erne og senere i 1980-erne er de to kirker igen forenet i den samme kirke, men da som resultat af foreningen af to større lutherske kirker i USA, hvortil de to danske kirker tidligere havde sluttet sig, og ikke som en følge af de to danske kirkers selvstændige bestræbelser.

Abstract:
Background Gastric cancer (GC) is one of the most common cancers with high mortality globally. However, the molecular mechanisms of GC are unclear, and the prognosis of GC is poor. Therefore, it is important to explore the underlying mechanisms and screen for novel prognostic markers and treatment targets. Methods The genetic and clinical data of GC patients in The Cancer Genome Atlas (TCGA) was analyzed by weighted gene co-expression network analysis (WGCNA). Modules with clinical significance and preservation were distinguished, and gene ontology and pathway enrichment analysis were performed. Hub genes of these modules were validated in the TCGA dataset and another independent dataset from the Gene Expression Omnibus (GEO) database by t-test. Furthermore, the significance of these genes was confirmed via survival analysis. Results We found a preserved module consisting of 506 genes was associated with clinical traits including pathologic T stage and histologic grade. PDGFRB, COL8A1, EFEMP2, FBN1, EMILIN1, FSTL1 and KIRREL were identified as candidate genes in the module. Their expression levels were correlated with pathologic T stage and histologic grade, also affected overall survival of GC patients. Conclusion These candidate genes may be involved in proliferation and differentiation of GC cells. They may serve as novel prognostic markers and treatment targets. Moreover, most of them were first reported in GC and deserved further research.

National identitet og udfordringen for den lettiske kirke idagAf Guntis DislersForfatteren er præst i den lutherske kirke i Letland og har gennem de seneste år været ansvarlig for at pleje forbindelsen til kirker i udlandet, nu hvor sådanne relationer kan etableres og plejes.Guntis Dislers gennemgår i en række punkter hovedtræk af den aktuelle religiøse situation i Letland. Det er hans hovedsynspunkt, at livsanskuelsessituationen er broget, om ikke ligefrem uoverskuelig. Det skyldes, at de religiøse kræfter, der var bundet under kommunismens totalit.re undertrykkelsessystem, nu slippes fri, med en løbsk aktivitet til følge. Ud over de kirker og trosretninger, der traditionelt har spillet en rolle i Letland, drejer det sig også om en form for nyhedenskab med rødder i urgammel, indoeuroæiske mytologi. Desuden går ny-religiøse grupperinger, både af indisk og af amerikansk oprindelse, sig kraftigt gældende. Ikke mindst den frie adgang til at gøre brug af de elektroniske medier indebærer, at der har åbnet sig et kolossalt virkefelt for nye religiøse vækkelsesgrupper, der kan lukrere p. det, der har nyhedens interesse, og som samtidig besidder de penge, der skal til for at lancere deres program.Forfatteren inddrager i sin redegørelse de træk i Letlands nationale historie, der forklarer landets kirkehistoriske udvikling. Ikke mindst har påvirkningen fra Tyskland spillet en rolle, i perioder sammen med en politisk afhængighed. Til den modsatte side har naboskabet med Rusland og forholdet til den ortodokse kirke været en mindst lige så afgørende faktor. Den lutherske kirke i Letland har i dette perspektiv været knyttet til den nationale selvstændighedstanke, et forhold, der også har gjort sig gældende under den allerseneste udvikling.Afsluttende omtaler Guntis Dislers i hovedtræk den aktuelle situation. Han går specielt ind p. behovet for at uddanne teologer og præster. Nu er der adgang for en forsvarlig uddannelse, efter at et teologisk fakultet fik officiel status i 1990. Endvidere nævnes behovet for kirkelig undervisning af lægfolket, kirkelige tidsskrifter og aviser, samt det ikke mindre udtalte behov for at opbygge internationale, økumeniske kontakter.

Abstract:
Somatic cell fusion is essential for the in vivo function of skeletal muscle, osteoclasts and trophoblasts. A diverse range of additional tissues including brain, heart, liver, prostate and intestinal epithelium are also subject to somatic cell fusion events in vivo. Fusion of embryonic stem cells has been reported and it has been hypothesised that fusion of cancer cells with macrophages may enable metastases. Furthermore, somatic cell fusion of Dystrophin positive cells with Dystrophin negative muscle cells may be therapeutically beneficial for the treatment of muscular dystrophy. In Drosophila, the type 1 transmembrane proteins Kin of Irre (Kirre), Roughest (Rst), Hibris and Sticks and Stones (SnS) are involved in regulating the muscle cell fusion process during larval development. The mammalian orthologues of Kirre and Rst include: Kirrel, Kirrel2 and Kirrel3 while Nephrin is the mammalian orthologue to Hibris and SnS. It is currently unclear whether the Kirrel family and Nephrin are expressed in mammalian skeletal muscle cells and whether they regulate the cell fusion process in these cells, as is the case in Drosophila. It was therefore hypothesised that all Kirrel family members and Nephrin would be temporally expressed in mammalian skeletal muscle cells as they undergo cell fusion events and hence may regulate the somatic cell fusion process. It was further hypothesised that inhibition of fusion would alter their expression patterns. Presented within this thesis are novel findings regarding the detection and expression profiling of all Kirrel family members and Nephrin during in vitro myogenesis of the C2C12 murine skeletal muscle cell line. Previously unreported splice variants of Kirrel and Kirrel3 have also been identified in C2C12 cells and in additional murine tissues. Variable expression patterns among family members are reported. The presence of multiple Kirrel family members and Nephrin in C2C12 cells suggest that redundancy and/or compensatory mechanisms are in place, as is the case in Drosophila, with regards mammalian muscle cell fusion in vivo. Results presented within this thesis lay the foundations for improving understanding of the molecular mechanisms underpinning mammalian somatic cell fusion events during development, injury repair and hypertrophy and also may enable increased efficacy of cellular therapies for diseases such as muscular dystrophy and the prevention of cancer metastasis.

Au cours des dernières décennies, une dégradation de la fertilité des vaches laitières, parallèlement à une augmentation de la production laitière a été observée. Des régions du génome, les QTL, affectent la fertilité femelle. Le but de ce travail de thèse est d’étudier la fertilité et certains paramètres zootechniques chez des vaches Prim'Hosltein en première lactation choisies pour leur haplotype favorable "fertil+" ou défavorable "fertil-" pour un QTL de fertilité situé sur le chromosome 3. Ce phénotypage a montré une meilleure fertilité et un meilleur bilan énergétique dans la première semaine de lactation pour les vaches "fertil+" par rapport aux vaches "fertil-". De plus, les vaches "fertil-" ont un flux d’ingestion plus rapide. Au pic de mobilisation, certains gènes du QTL étaient différentiellement exprimés dans le tissu adipeux des deux haplotypes. Dans les cellules de la granulosa, un de ces gènes, nommé Kirrel, est plus exprimé chez les vaches "fertil+" et sa protéine recombinante inhibe la sécrétion de progestérone in vitro. Notre travail a permis d'affiner les interactions génotype-phénotype liées à un QTL de fertilité et de mettre en avant un des possibles rôles d'un gène de ce QTL dans la fonction de reproduction chez la vache laitière<br>In recent decades, the dairy cow fertility has declined, in parallel with an increase in milk production. Some regions of the genome, named QTL, affect female fertility. The purpose of this thesis is to study fertility and some zootechnical parameters in Prim'Hosltein cows in first lactation chosen for their favorable haplotype "fertil+" or unfavorable haplotype "fertil-" for one fertility QTL on chromosome 3. This phenotyping showed better fertility and energy balance in the first week of lactation for "fertil+" than for "fertil-" cows. In addition, "fertil-" cows had a higher eating rate. At the peak of mobilization, the QTL genes are differentially expressed in adipose tissue of "fertil+" and "fertil-" cows. In granulosa cells, one of these genes, named Kirrel, is higher expressed in "fertil+" cows and its recombinant protein inhibits the secretion of progesterone in vitro. Our work has contributed to refine interactions genotype-phenotype linked to one fertility QTL and highlighted one of the possible roles of a gene which belongs to this QTL in the reproductive function in dairy cows

Abstract:
The aim of the studies included in the thesis is to better understand the interplay between neuronal activity-dependent gene regulation and the bioactive vitamin A metabolite all-trans-retinoic acid (RA) during postnatal development, refinement and maintenance of precise neuronal connectivity using the olfactory sensory neuron (OSN) in the olfactory epithelium (OE) of genetically modified mice as a model. We show that: Inhibition of RA receptor (RAR)-mediated transcription in OSNs reduces expression of the olfactory cyclic nucleotide-gated (CNG) ion channel, which is required for odorant receptor (OR)-mediated stimulus transduction. This, results in increased OSN death and errors in precise connectivity. The increased cell death may be a consequence of reduced intrinsic excitability and/or reduced influx of Ca2+ ions while the errors in connectivity may be due to altered OR-dependent expression of axonal guidance proteins, such as Kirrel-2 and Neuropilin-1. Expression of the RA catabolic enzyme Cyp26B1 in OSNs is positively regulated by RAR-mediated transcription as well as sensory stimulation in a CNG channel-dependent manner. This shows that neuronal activity and local vitamin A metabolism are parts of novel regulatory feedback loop controlling precise connectivity and neuronal survival. The feedback loop may be a form of homeostatic plasticity in response to global changes in neuronal activity. BACE1, an enzyme is implicated in Alzheimer´s disease, and Cyp26B1 are inversely regulated by CNG channel-dependent sensory stimulation. Cyp26B1 expression is switched on at birth, forms a topographic expression gradient in OE and inhibits BACE1 expression into an inverse counter gradient. Taken together these results reveal a novel neuronal activity-dependent mechanism by which sensory stimuli can shape spatial gene expression via altered RA bioavailability. Increased Cyp26B1 expression stimulates turnover of OSNs during adult neurogenesis by a non-cell-autonomous mechanism. The gradient of Cyp26B1 expression correlates with spatially-regulated diversification of OSNs into subpopulations that express different subsets of OR genes. Cyp26B1 expression influences spatial OR diversification of OSNs by two different mechanisms. In the ventrolateral OE, Cyp26B1 inhibits OR expression by blocking OSN differentiation at a stage that may be associated with the cell intrinsic mechanism regulating OR gene choice. In the dorsomedial OE the expression frequency of some ORs is unaltered while other increases, presumably as a consequence of neuronal activity-dependent competition. A probable function of graded and activity-dependent Cyp26B1 expression is to form a topographic partitioning of the olfactory sensory map into functional domains, which gradually differ from each other with regard to experience-driven plasticity and neurogenic potential along the dorsomedial-ventrolateral axis of OE.

Mais la vision de kireeev est aussi une vision de loin : le personnage se fait allegorique, la psychologie schematique. L'individu qui naissait grace a la vision de pres est etouffe par divers procedes de rabaissement et par sa dissolution dans des modeles mythiques ou litteraires. Dans cette bipolarisation de la vision resident l'originalite de la poetique kireevienne et l'expression d'une epoque qui d'un cote revalorise l'individu, mais de l'autre le brise insidieusement. Sans passe de par la brisure du stalinisme, sans present de par l'"eclipse de l'epoque" que represente la stagnation, et sans avenir de par le manque de foi et d'ideal, le personnage kireevien est a l'image de l'homme sovietique double coince dans le trou noir de la stagnation, mais qui pourtant resiste a l'ecrasement du moi. Cette dualite de l'oeuvre de kireev s'avere ainsi l'expression poetique par excellence de la stagnation<br>Ruslan kireev, who belongs to the generation of the forty-year-old writers, rejects the cliches of the socialist realism. His characters appear both matter-of-fact and ordinary. The author refuses the didactism. He scrutinizes the individual's inner world of which he unerlines the profound moral and psychological ambivalence. However, kireev's vision is also a distancing vision. The individual who had taken shape by means of the closest vision is shatterd by a wide range of belittlement devices and by its dissolution into mythical and literary models. In this bipolarization of the vision lie the originality of kireev's poetics as well as the expression of an era in which the individual sound himself reasserted but also insidiously cruched. Without a past, because of the stalinian break, without a present, because of the "eclipse of the epoch" (the stagnation), and without a future, due to the lack of faith and ideal, kireev's character is the reflection of the soviet, a double-faced man, trapped in the black hole of stagnation, but who nevertheless resists the hammering of the self. This duality in kireev's work can be considered as the poetic expression of stagnation. 1
