Abstract:
A emergência de resistência microbiana é um desafio para microbiologistas, médicos, órgãos de saúde pública e para a indústria farmacêutica. Passado três décadas, patógenos gram-positivos, principalmente Staphylococcus aureus, têm desenvolvido cada vez mais resistência a vários antibióticos devido ao uso extensivo nos hospitais e na comunidade. Este desenvolvimento aconteceu em um período em que poucas novas classes de antibióticos tenham sido identificadas, ressaltando a necessidade urgente de novos agentes antimicrobianos. A modificação molecular tem se mostrado como estratégia bastante promissora no planejamento e desenvolvimento de análogos com melhor biodisponibilidade, maior atividade intrínseca e menor toxicidade. Desta forma, foi planejada, neste trabalho, a síntese de série de derivados 5-metilsulfonil-2-tiofilidênicos e derivados 5(6)-benzofuroxânicos, análogos à nifuroxazida, buscando atividade bacteriostática e/ou bactericida frente a cepas resistentes de Staphylococcus aureus. A seleção dos grupos substituintes foi fundamentada na influência de suas propriedades físicoquímicas, como hidrofobicidade e efeito eletrônico. Para avaliação da atividade antibacteriana, este trabalho usou o método de determinação da concentração inibitória mínima, CIM, frente à Staphylococcus aureus, cepas ATCC25923, 3SP/R33 e VISA3. Os derivados 5-metilsulfonil-2-tiofilidênicos não apresentaram atividade antimicrobiana, enquanto que todos os derivados 5(6)-benzofuroxânicos foram ativos frente a cepa padrão ATCC25923. O composto mais ativo foi a 5(6)-benzofuroxano-4-nitrobenzidrazida (CIM= 16,80 µg/mL) e o menos ativo foi a 5(6)-benzofuroxano benzidrazida (CIM = 36,00 µg/mL). Ficou evidenciado que a atividade antimicrobiana destes compostos sofre forte influência da hidrofobicidade e do efeito eletrônico dos grupos substituintes. Os compostos 5(6)-benzofuroxano-4-nitrobenzidrazida e 5(6)-benzofuroxano-4-clorobenzidrazida foram testados frente às cepas com caráter de multi-resistência, 3SP/R33 e VISA3, e ambos mostraram atividade antimicrobiana similar quando comparados com a cepa padrão. Ressalta-se que foram sintetizados e identificados neste trabalho onze compostos que apresentam estruturas químicas inéditas.
Emergent antimicrobial resistance is a challenge for microbiologists, physicians, public health organizations, and pharmaceutical industry. Over the past three decades Gram-positive pathogens, most notably Staphylococcus aureus, have become increasingly resistant to multiple antibiotics due to their extensive hospital and community use. This has come at time when very few new antibiotic classes have been identified, and emphasizes the urge for new antibacterial agents. Molecular modification has been used as a quite promising strategy in the design and development of drug analogs with better bioavailability, higher intrinsic activity, and lesser toxicity. Thus, for this work a series of 5-methylsulfonyl-2-thiophylidene and 5(6)-benzofuroxans derivatives were synthetized and tested for antibacterial activity against resistant Staphylococcus aureus strains. The selection of the substituent groups was based on the influence of physical-chemical properties, such as hydrophobicity and eletronic effects. Antibacterial activity was evaluated through the determination of the minimum inhibitory concentration, MIC, against Staphylococcus aureus strains ATCC25923, 3SP/R33 and VISA3. 5-methylsulfonyl-2-thiophylidene derivatives did not show antibacterial activity, while all 5(6)-benzofuroxans derivatives were active against the standard strain ATCC25923. 4-nitro-benzoic acid[((5(6)benzo[c][1,2,5]oxadiazole N1-oxide)-2- yl)-methylene]-hydrazyde (MIC=16,80 µg/mL) was the most active compound, and the least active was benzoic acid[((5(6)benzo[c][1,2,5]oxadiazole N1-oxide)- 2-yl)-methylene]-hydrazyde (MIC = 36,00 µg/mL). It was evidenced that the antimicrobial activity of these compounds is influenced by the hydrofobicity and electronic effects of substituent groups. Compounds 4-nitro-benzoic acid[((5(6)benzo[c][1,2,5]oxadiazole N1-oxide)-2-yl)-methylene]-hydrazyde and 4-Chloro-benzoic acid[((5(6)benzo[c][1,2,5]oxadiazole N1-oxide)-2-yl)-methyl- ene]-hydrazyde were evaluated against the resistant strains 3SP/R33 and VISA3, and both showed similar antimicrobial activity when compared to the standard strain. It must be pointed out that eleven previously unknown compounds were synthetized and tested in this work.

Die Dissertation untersucht die Entwicklung der Rechtschreibfertigkeiten von Schülern mit Deutsch als Zweitsprache im Vergleich zu ihren Mitschülern mit Deutsch als Muttersprache in der Sekundarstufe I. Kern der Untersuchung bildet die Methode der Fehleranalyse. Mittels quer- und längsschnittlicher Analysen werden spezifische Vergleiche zwischen den Rechtschreibleistungen von 236 Schülern italienischer und deutscher Herkunft vorgenommen. Die Ergebnisse der Untersuchung präzisieren die empirisch bislang nur lückenhaft dokumentierten Rechtschreiberwerbsverläufe von ein- und zweisprachigen Schülern der Sekundarstufe I. Die Daten geben Auskunft über bestehende Leistungsrückstände der zweisprachigen Schüler und dokumentieren charakteristische Entwicklungsverläufe und Fehlerschwerpunkte. Für die Unterrichtspraxis geben die gewonnenen Erkenntnisse folglich konkrete Hinweise. Aufgezeigt werden nicht nur die Lerninhalte, für die vertiefende Instruktionen und Übungen im Unterricht der Sekundarstufe I nötig sind, sondern ebenfalls die Zeiträume, in denen sich die Schüler sichere Schreibfertigkeiten in den einzelnen orthographischen Problembereichen aneignen.

How may a reader who wishes to read the Christian Bible as scripture well today be formed; and how may interpretations of scripture inform such concern? The present work is an exploration of this under-considered question in the field of contemporary biblical scholarship via sustained exegetical engagement with three biblical texts, namely Nehemiah 9:6 – 37, Ezekiel 20:5 – 32 and Acts 7:2 – 60, which offer three different inner-canonical readings of scripture in the form of three distinctive recitals of Israel’s story. The purpose is to consider how these retellings read scriptural traditions in relation to the wider context of the Christian canon; and to reflect on their enduring and formative significance as scripture for readers seeking to appropriate the scripture faithfully today. Chapter one will indicate that the concern of the present work is not a recent one, but rather one that is integral to a Christian practice of reading scripture. This chapter will also consider how such a concern once under-explored in biblical scholarship is now receiving some renewed attention in the field of theological interpretation of scripture. An overview of selected works pertaining to such concern will be considered in chapter two as a means to set a context for articulating the approach and rationale of the present work. In chapters three through to five, each chapter will be devoted to each of the three biblical texts, Nehemiah 9:6 – 37, Ezekiel 20:5 – 31 and Acts 7:2 – 60, to consider how scriptural traditions are interpreted in these three texts in relation to the wider context of the Christian canon. The next step is to reflect on the implications of these three biblical texts as Christian scripture for readers seeking to interpret scripture faithfully today. For such concern, the three texts will be considered individually at the end of chapters three, four and five respectively and then in concert in chapter six.

Notre etude des chapitres 1 a 3, 4 1-5 7, 11 et 14 6-9 a permis de mettre en lumiere l'existence de quatre couches redactionnelles 1 la tradition osenne que nous situons vers le milieu du 8e siecle avant j. C. Applique l'image de la "mere" aux deesses asherah et anat. En outre, elle puise dans le riche materiau litteraire et iconographique des representations de la deesse nourriciere et du motif de la vache allaitant son veau. 2-nous avons repere les traces d'une tradition du nord qui se presente sous la forme d'une complainte sur la chute du royaume du nord (vers la fin du 8e siecle). 3- une redaction deuteronomiste a retravaille en profondeur les traditions preexistantes vers la fin du 7e siecle. The difficulty of interpreting the book of hosea has long been felt. In the same way, the identification of the "mother" has raised several problems. Is she the land (israel) or the people? in order to verify these hypothesis we have tried to study the texts. On the one hand. By the light of redactional criticism, and on the other hand, with the aid of iconographical materials based on the archaeological and epi graphical background as well as on the history of religions. We have located within chapters 1-3, 4 1-5 7 11 and 14 6-9 four redactional layers. 19 an hosean tradition (middle of 8th century) which associates the figure of the "mother" with the goddesses asherah and anat, yhwh's spouses. This tradition also uses literary and iconographical material from the feeding goddess and from the representations of a cow suckling her calf. 2) a nothern tradition in the shape of a lament after the destruction of the northern kingdom (end of 8th century) 3) a deuteronomistic redaction which concealed all mentions of goddesses according to her monotheistic theology (7th century). 4) a final editor, in the persian period, has recorded the old material and gathered into a big collection 1-3, 4-6, 11
