Abstract:

First part of the thesis is about literature research of optimization algorithms. Three of the algorithms were implemented and tested, concretely the ant colony algorithm, tabu search and simulated annealing. All three algorithms were implemented to solve the traveling salesman problem. In second part of the thesis the algorithms were tested and compared. In last part the influence of the ant colony parameters was evaluated.

Abstract:

This construction programme deals with a hermit colony located in the western part of the Vysočina Region. This hermit colony has been established to provide accommodation for women living in this area permanently while offering an opportunity of occasional relaxation for people who are living hectic lives in urban environment. The aim of my project is to design the remaining parts of the colony, i.e. two hermitages and the community’s chapel. A hermitage is a one-storey building intended for one person only. The load-bearing walls of the individual hermitages are designed to be built from different materials (wooden and masonry constructions) whereas the elevational structures of all the buildings are to be the same. The chapel’s wall system combines stone and wood. The chapel will serve solely the purposes of the community. The main objective of my project is to respect and maintain the natural features of the landscape to the maximum extent possible.

Abstract:

The diploma thesis deals with the Kamenna district with a specific area of housing. It describes its history, morphology, today's life and its virtues and shady pages. Approaching today's issues of this location and approach or view of stakeholders. In addition, eight properties are instantiated in the work. These properties are selected from the so-called „Upper Kamenka“ and „Lower Kamenka“ (four from each part) to ensure comparable technical advantages and disadvantages of individual parts. Technical disadvantages such as absence of sewerage and geological survey on the sloping slope in one of the mentioned objects. Comparing methodology to the comparison method according to Decree No. 441/2013 Coll. in current version, Comparison method-market price and cost method according to Decree No. 441/2013 Coll. in current version. At the end of the work, the author undertakes a discussion about negotiating the price and explains the great diversity of the results.

Ces "mythologemes" qui s'ancrent dans un fonds anthropologique plus profond et dont la resurgence est liee a la situation socio-politique de l'allemagne wilhelminienne s'organisent autour d'une utopie essentiellement "restauratrice" centree sur un retour a la "germanite" (deutschtum). Le colon, tenu pour l"'homme nouveau", adepte d'une mystique traditionnelle, pre-chretienne doit s'epanouir au sein d'une communaute du peuple (volksgemeinschaft). The creation of the myth of the german colonial - as it appears in the review "kolonie un heimat" - seems to be structured around certain "mythotyps" ("mythologemes", to use the terminilogy of g. Durand upon whose our approach is based) : "nostalgia of origins", "the struggle between good and evil" ans "germanic cohesion and unlimited progress". These "mythotypes", wich have profound anthropologic roots and owe their resurgence to the socio-political situation of the kaisers germany, are centred on an essentially backward-looking utopia which has as its axis a return to "germaness" ("deutschtuum"). The fulfilment of the settler - a "new man", subscribing to a traditional, prechristian mysticism-is to be achieved within a "community of the people" ("volksgemeinschaft"). The german colonies thas become a real alternative, an ultimate hope for a reich considered as embasked on the road of decadence and degeneration

Diplomová práce se zaměřuje na působení britské Východoindické obchodní společnosti v Indii. První kapitola shrnuje okolnosti vzniku a působení Společnosti na indickém území před rokem 1756. Následná kapitola popisuje přerod Společnosti jako monopolní firmy v suveréna na indickém subkontinentu. Teoretická část práce se věnuje přístupu předních britských ekonomů té doby k otázce kolonií a Společnosti samotné. Hlavní část práce analyzuje nositele, nástroje a financování hospodářské politiky Indie v době vlády Východoindické společnosti. Zabývá se vztahy britských politiků k indické otázce a jejich vlivem na formulaci hospodářské politiky. Analyzuje hlavní nástroje politiky Společnosti v Indii jako jsou obchodní monopol a armáda. Podrobněji sleduje financování, příjmy a výdaje Společnosti v době její vlády. V závěru nastiňuje některé okolnosti konce vlády Východoindické společnosti na subkontinentu.

Abstract:

The content of the thesis is a description of intracellular processes responsible for cell cycle regulation and reactions of cells to external and internal stimuli. Thoroughly described are important signaling pathways with appropriate methods, which can be used to simulate them in silico. From these cellular processes, a cell cycle model is created and implemented in a tool programmed in C ++ with OpenGL used for visualization. The model is then tested for various cell processes including HeLa cells growth. Finally, the results are compared with the behavior of living cells.

Abstract:

G-CSF besitzt direkte Effekte auf die Aktivierung bakterizider Eigenschaften neutrophiler Granulozyten und verbessert das Überleben bakteriell infizierter Tiere. Daher wurde in der hier vorliegenden Arbeit der Effekt einer prophylaktischen oder therapeutischen Gabe von G-CSF bei experimenteller Pneumonie durch Klebsiella pneumoniae in Mäusen untersucht. Unerwarteterweise verschlechterte aber eine prophylaktische G-CSF-Gabe das Überleben und führte dosisabhängig zu einer Steigerung der bakteriellen Dissemination von der Lunge in Leber und Milz. Im Gegensatz dazu konnte ein spezifisch gegen K2-Kapselpolysaccharid (K2-KPS) von K. pneumoniae gerichteter monoklonaler Antikörper signifikant die Vermehrung der Bakterien in Lunge, Leber und Milz reduzieren. Die Blockierung von TNF?? durch Pentoxifyllin hingegen verzögerte die Letalität nach Induktion der Pneumonie, verhinderte sie jedoch nicht. In vitro konnte hier nachgewiesen werden, daß G-CSF spezifisch an K. pneumoniae bindet und daß diese Bindung an mehrere Proteine mit einem Molekulargewicht von 41, 25 und 21 kDa erfolgt. Die Bindung von G-CSF an K. pneumoniae führte zu einer signifikant erhöhten Produktion des wichtigsten Virulenzfaktors, K2-KPS. Dies verminderte in vitro signifikant eine Phagozytose der Bakterien durch neutrophile Granulozyten. Damit gelang es zum ersten Mal, die Bindung von G-CSF an ein gram-negatives Bakterium, K. pneumoniae, nachzuweisen und zu zeigen, daß diese Bindung in vitro zu einer erhöhten Produktion des wichtigsten Virulenzfaktors und in vivo zur Verschlechterung einer experimentellen Pneumonie durch erhöhte bakterielle Disseminierung bei prophylaktischer Gabe von G-CSF vor Infektion führt. Die weitere Untersuchung dieser Phänomene hinsichtlich einer möglichen Bindung von G-CSF auch an andere Bakterien könnte zu einer differenzierten supportiven Therapie bakterieller Infektionen mit G-CSF in nicht neutropenischen Patienten führen.
Besides its well-established effects on granulocytopoiesis, granulocyte colony-stimulating factor (G-CSF) has been shown to have direct effects on the recruitment and bactericidal ability of neutrophils, resulting in improved survival of experimentally infected animals. The effect of G-CSF on the course of experimental pneumonia induced by Klebsiella pneumoniae was studied. Using a highly reproducible murine model, the paradoxical finding that mortality from infection was significantly increased when animals received G-CSF before induction of pneumonia could be demonstrated. Administration of G-CSF promoted replication of bacteria in the liver and spleen, thus indicating an impairment rather than an enhancement of antibacterial mechanisms. By contrast, a monoclonal antibody against Klebsiella K2 capsule significantly reduced bacterial multiplication in the lung, liver, and spleen, and abrogated the increased mortality caused by G-CSF. Blocking of TNF-? with pentoxifylline, however, could not prevent increased mortality caused by G-CSF. In vitro studies showed a direct effect of G-CSF on K pneumoniae resulting in inreased capsular polysaccharide (CPS) production. When bacteria were coincubated with therapeutically achievable concentrations of G-CSF, phagocytic uptake and killing by neutrophils was impaired. Western blot analysis showed three binding sites of G-CSF to K pneumoniae. Thus, in this model, the direct effect of G-CSF on a bacterial virulence factor, CPS production, outweighed any beneficial effect of G-CSF on recruitment and stimulation of leukocytes. Further investigations of possible binding of G-CSF to other bacteria might influence a differentiated supportive therapy of bacterial infections in non-neutropenic patients with this growth factor.

Abstract:

The diploma project is focused on screening the possibilities of increasing the inner area of the residences within the former workers-housing area Letná in Zlín. The targets of the project are: - To create an attractive residential district, composed of contemporarily re-designed working class spaces, - To support the district as a unit, which can be able to react on the actual demographical requests and conditions, and - To support the district, which should be able to compete with the new rising accommodation districts through its prevalent qualities. The result is a design consisting of 3 main variations of the default house extension. The added cubic elements present a solution for the problems most commonly seen in the district in its actual state; the house extension creates a transition between the private inner space of the house and the outer public space. The proposal allows maximal living space while minimizing construction size extension.

Abstract:

This master thesis deals with image processing of Petri dishes. The first part is about terms and procedures in laboratory practice. The following part is about devices and algorithms for image analysis of Petri dishes. The second part is about draft and implementation of own colony counting application, which was developed in cooperation with the microbiological laboratory. The application is developed for mobile devices running Android OS. The master thesis also includes the design and creation of two lighting units, which uses as an accessory for better results in the analysis of Petri dishes. In conclusion are presented results, compared lighting units and various types of mobile devices, on which the application was tested.

Abstract:

This diploma thesis focuses on meta-heuristic algorithms and their ability to solve difficult optimization problems in polynomial time. The thesis describes different kinds of meta-heuristic algorithms such as genetic algorithm, particle swarm optimization or ant colony optimization. The implemented application was written in Java and contains ant colony optimization for capacitated vehicle routing problem and particle swarm optimization which finds the best possible parameters for ant colonies.

Abstract:

Although the research on myrmecophilous insects is dated back to the 19th century, certain aspects of fascinating relationship between hosts and their social parasites are not satisfactorily understood. The latest studies focus mostly on the systematics of myrmecophilous taxa, and bionomy remains neglected. Most of the myrmecophilous insect can be found among beetles (Coleoptera). These myrmecophiles reveal a broad spectrum of adaptations that allow them to live in close association with their host ants. These adaptations significantly differ among the well-integrated species on one hand and poorly integrated or non-integrated species of myrmecophilous beetles on the other. This study focuses at the comparison of behaviour in two species of myrmecophilous beetles, Claviger testaceus (Staphylinidae: Pselaphinae) as the member of integrated myrmecophiles, and Pella spp. (Staphylinidae: Aleocharinae) as poorly integrated or non-integrated beetle. My work shows significant differences in behaviour taking place after encounter of a beetle and an ant. Ants are more aggressive towards the non-integrated species, and at the same time, these beetles are well-adapted for aggressive behaviour from the ants.
