Abstract:
Syfte: Uppsatsens huvudsyfte är att kartlägga och beskriva hur modebranschen förhåller sig till fenomenet kopiering. Genom att kategorisera olika attityder gentemot kopiering samt de aktörer som står bakom attityderna, analyseras även varför vilka aktörer har specifika attityder kopplat till fenomenet.Design/Metod/Ansats: Uppsatsen presenterar, med hjälp av kategorier, de attityder och aktörer som är representerade i debatten om kopiering. Ett urval har gjorts i fråga om vilka media och texter som ska analyseras i ramen för studien. Dessa texter har sedan analyserats så till vida att kategorier av attityder kunnat identifieras i texterna. I ett andra steg har de aktörer som står bakom de identifierade attityderna kategoriserats. Slutligen har en analys gjorts gällande varför vissa attityder kan sammankopplas med vissa aktörer. Resultat: Studiens referensram och uppsatsens empiriska studies resultat har vid analys kunnat visa på vilka attityder gentemot kopiering som finns representerade i modebranschen, samt på vilka aspekter som påverkar hur, och förklarar varför, en aktör resonerar och förhåller sig till kopiering på det sätt som görs.Originalitet/Värde: Uppsatsen belyser nya typer av kopiering som är aktuella idag och kompletterar på så vis tidigare forsking inom området, vilken framför allt behandlat piratkopiering. Dessutom fokuseras modebranschens interna och professionella aktörer istället för konsumenter, vilket också syftar till att belysa ämnet kopiering utifrån en nytt perspektiv.Objective: The main objective of this paper is to identify and describe how the fashion industry is related to the phenomenon of copying. By categorizing different attitudes toward copying, and the actors behind the attitudes, analysis is made as to why the actors have specific attitudes associated with the phenomenon. Design/Methodology/Approach: The thesis presents, using categories, the attitudes and actors that are represented in the debate on copying. A selection has been made on what media and texts are to be analyzed in the study. These texts were then analyzed in the sense that the categories of attitudes were identified in the texts. In a second step, the actors behind the identified attitudes also were categorized. Finally, an analysis is made as to why certain attitudes are associated with certain actors.Results: When analyzed, the study of reference and the thesis empirical study results, show the attitudes toward copying that are represented in the fashion industry. The analysis also present the aspects that affect how, and explains why an actor think and relate to copying in the manner set out.Originality/Value: The thesis highlights new types of copying that are relevant today and the thesis is therefore complementary to previous research in the area, which has mainly focused on piracy. Furthermore, the thesis is focused on the fashion industry's professional actors rather than consumers, which also serves to illustrate the subject of copying from a new perspective.

Abstract:
Syftet med uppsatsen är att ta reda på hur åtta andraklassare väljer att bygga då de ställs inför två olika miljöer. Jag undersöker saken genom två observationer, och kompletterar med två gruppintervjuer. Observationerna genomförs i barngrupper om fyra barn där barngrupperna ges uppgiften att konstruera fritt med avklippta grillpinnar och häftmassa. När jag observerar barnen och deras val av bygge märker jag om de väljer att skapa efter egna idéer, kreativt, eller om de väljer att titta på andras byggen, det som jag i den här undersökningen benämner att tillämpa konformitet. Min strävan är att se vilka faktorer som påverkar barnen att välja det konstruktionssätt som de använder. Jag använder mig av tre teorier för att tydliggöra deras val: Vygotskijs sociokulturella teori där samtalets vikt för skapandet läggs fram, Piagets kognitiva utvecklingsteori där fokus ligger på de fysiska tingens påverkan, samt Banduras modellinlärningsteori som menar på att individer lär sig bäst genom att observera andra. Jag förser även undersökningen med en del forskning kring attityder, kreativitet, och konformitet. Analysen visar att samtliga teoriperspektiv påverkar på något sätt. Samtalet påverkar i dubbel bemärkelse då det antingen uppmuntrar eller hämmar barnet då barnets idé nämns. Den fysiska omgivningen påverkar i det avseendet att barnen konformerar i hög grad, och i ännu högre grad då barnen har fler modeller att inspireras ifrån. Jag märker att de barn som väljer att tillämpa konformitet gör det enligt vissa mönster. Jag ser också att attityderna påverkar genom barnens motivation att bygga, och deras uppfattning om vad materialet är till för. Kreativitetsavsnittet visar att nästan alla skapar något kreativt; sex av åtta barn skapar något unikt. Vad gäller konformiteten så visar det sig att nästan lika många barn väljer att bygga utefter en annan modell vid något tillfälle, fem av åtta. Sammanfattningsvis står det klart att barnen i undersökningen är något mer kreativa än konformativa, och även att det finns flera faktorer som påverkar barnens val av bygge.

Den här rapporten presenterar ett antibiotikafritt, stabilt och kromosombaserat expressionssystem för läkemedelsproduktion i Escherichia coli på beställning av företaget Affibody AB. E. coli-stammen BL21(DE3) valdes som värdorganism för expressionssystemet. Systemet består av en genkassett som innehåller en T7-promotor, en 5′-UTR från genen ompA och en terminatorsekvens från RNA-operonet rrnB. Fyra kopior av genkassetten ska integreras i pseudogenerna caiB, yjjM, hsdS och yjiV. En datormodell som modellerar det egentliga kopietalet i cellerna har skapats i mjukvaran MATLAB, vilket visar att det uppskattas vara maximalt 32 kopior av genkassetten per cell på grund av replikation av kromosomen. Ett högt pH i fermentorn; att använda fed-batch och blandade kolhydratkällor; och att använda stammen BL21(DE3) minskar acetatproduktionen i cellen. En lägre acetatproduktion kan leda till en högre produkthalt. En proteinutbytesmodell för mjukvaran MATLAB har konstruerats för att uppskatta koncentrationen av Affibody®-molekylen i en E. coli cell.

Abstract:
Humans are continuously challenged by harmful endogenous and xenobiotic substances. Detoxification is the ability to neutralise and remove these substances from the body. Glycine N-acyltransferase, EC 2.3.1.13 (GLYAT) is a key enzyme in detoxification. GLYAT catalyses an amino acid (glycine) conjugation reaction in phase II of detoxification. It is expected that, similar to what has been observed in the Cytochrome P450 enzymes, variations within the GLYAT gene may lead to altered enzyme activity that may affect the efficacy of detoxification. The aim of this study was to identify genetic variations within the GLYAT gene of a cohort of individuals whose GLYAT activity has been biochemically characterized. Biochemical profiles of phase I and II detoxification of a number of individuals was screened to select those with possible aberrant GLYAT activity. Eighteen selected individuals agreed to participate in the study. The 23.21 kb GLYAT gene of the participants was amplified in four fragments and sent for pyrosequencing (Roche GS FLX titanium) at Inqaba Biotec. The results were analysed with the Lasergene software package from DNAStar (Madison, Wisconsin, USA). A total of 94 variations were identified from the Next Generation Sequencing data. Of these three found in the exons were known variations and four variations located in the exons were novel. A total of 62 known and 25 novel variations were identified in the introns of the GLYAT gene. Sanger sequencing verified 70.29% (68 in total) of the variation, which included 12 novel variations, of which one is located in exon six. Real-time quantitative PCR (qPCR) experiments were conducted and the data analysed using CopyCaller software to identify copy number variations within the cohort. It was found that participant 17 may have multiple copies of parts of the 3-terminal end of the gene (exons five and six), which might have an effect on GLYAT activity. Variations could possibly affect GLYAT activity, but the data was inconclusive and must be confirmed. Some of the variations could possibly affect GLYAT activity, but no correlation could be made between the variations identified during this study and the cohort’s detoxification ability. Further studies needs to be conducted to establish the effect of the variations in combination with one another on GLYAT activity. If some of these variations affect GLYAT activity such data might shed some light on variations observed between the glycine conjugation ability of individuals. Such information could eventually be of value in treatment of inborn errors of metabolism.
