Abstract:
Od czasu Rewolucji Przemysłowej Ocean Światowy doświadcza gwałtownych zmian związanych przede wszystkim z ociepleniem wód ale również zjawisk temu towarzyszących jak zakwaszanie spowodowane nadmiernym absorbowaniem atmosferycznego CO2. Istotnym jest, iż przekształcenia w ekosystemach pelagialu zachodzą nie tylko w rejonach polarnych, ale również w innych rejonach świata. Wynika to z globalnie działającej cyrkulacji atmosferycznej, której oddziaływanie obserwowane jest w całym Oceanie Światowym. Zarówno w Arktyce jak i Antarktyce, zmniejszający się zasięg lodu morskiego i cielenie lodowców mają wpływ na przebieg produkcji pierwotnej, a co za tym idzie, na funkcjonowanie wyższych poziomów troficznych zaczynając od zooplanktonu a kończąc na ssakach morskich. Obserwacje zmian klimatu, najbardziej widoczne w ekosystemach polarnych, spowodowały wzrost zainteresowania biologów morskich reakcją organizmów morskich na zachodzące procesy, szczególnie zooplanktonu. Ich znaczenie w sieci troficznej oraz zwykle zawężone zakresy tolerancji na czynniki abiotyczne i biotyczne powodują, że są one kluczowym elementem monitoringu ekosystemowego. Skutkiem przemian środowiska jest, między innymi, przesuwanie zasięgów występowania wielu gatunków, szczególnie tych bytujących w rejonach polarnych, ale również liczne odpowiedzi organizmów reagujących na stopniowe zakwaszanie oceanów. Najbardziej narażone będą organizmy których tkanki zbudowane są częściowo z CaCO3 jak otwornice, kokolitofory, szkarłupnie czy ślimaki pelagiczne, ale również te które są stenobiontyczne jak polarne gatunki widłonogów czy kryl antarktyczny, stanowiące główny element łańcucha troficznego. W niniejszym artykule przedyskutowano konsekwencje procesów takich jak ocieplenie wód oraz ich zakwaszenie na liczne organizmy morskie oraz ich potencjalne reakcje na negatywne bodźce. Liczne badania pokazują, iż niewielki nawet spadek pH wód morskich i podwyższenie temperatury wód, powodują zaburzenia fizjologii organizmów, a także zmiany zasięgów ich występowania czy liczebności.

Abstract:
Aim: to evaluate the long-term clinical efficacy of the combined (laser plus surgical) minimally invasive technique for rhegmatogenous retinal detachment (RRD). Patients and Methods: treatment results of 41 patients (41 eyes) with local RRD resulting from horseshoe retinal tear were analyzed. All patients underwent wide-field optical coherence tomography (OCT) to localize and measure pathological vitreoretinal adhesions at the site of retinal tear. The next step was a combined laser surgical procedure that included Nd: YAG dissection of pathological vitreoretinal adhesion zone, pneumatic retinopexy (10% C3F8), and barrier laser retinal photocoagulation (LRP) after reattachment. Postoperatively, eye exams were performed after 3 and 7 days, 3, 6, 12, 18, and 24 months. Results: complete reattachment was achieved in 38 patients (92.7%) on day 2 or 3. No reattachment was seen in three patients (7.3%); of them, subretinal gas migration was observed in two patients (4.9%), and partial reattachment was detected in one patient (2.4%) with pseudophakia. Recurrent RRD occurred in 3 patients (7.9%) because of new inferior retinal breaks. In four patients (10.5%), postoperative wide-field OCT of the area of the initial retinal tear performed after reattachment revealed additional pathological vitreoretinal adhesions in the adjacent areas (that were not identified preoperatively). These patients underwent additional LRP around the areas of pathological vitreoretinal adhesions. No recurrent RRDs were seen during follow-up. Conclusion: combined minimally invasive laser surgical technique for local PPDs demonstrated high efficacy (92.7%). This technique was the effective in local retinal detachment resulting from a single superior horseshoe retinal tear in patients with crystalline lens. In some cases, pseudophakia is a technical obstacle to this procedure. Postoperative wide-field OCT is recommended to detect additional pathological vitreoretinal adhesions and to perform LRP around them to reduce the risks of recurrent RRDs greatly. Abstract:
Background: Sustainable Reserve Food Garden Area (KRPL) is program that to increase family dietary diversity, such as children. Children’s dietary diversity can affect to nutritional adequacy and can impact to growth process. The factor that can affect successful of KRPL program is parents’ education level.Objectives: The purpose of this study was to identify the difference between children’s dietary diversity level, household income level, and parents’ education level in sustainable reserve food garden area (KRPL) and non KRPL household in Kamal sub-district, Bangkalan district, Madura. Method: This was an observational study with case control comparative study design was conducted among 25 children in KRPL area and 25 children in non KRPL area with mother or other family member as respondents. Simple random sampling method was used to select the samples. Children and parents’ characteristic were asked to answer structured questionnaire. Children dietary diversity level was asked by Individual Dietary Diversity Score (IDDS) instrument. The data was analyzed with Mann Whitney statistic test. Result: Children that involved in this study, either in KRPL or non KRPL area, was less than the same 35 months old (52%). Children from KRPL area was mostly girls (64%), while children from non KPRL area was mostly boys (56%). Children dietary diversity condition in KRPL area was low (52%), while children dietary diversity level in non KRPL area was sufficient (64%). Children who lived in KRPL area not always have diverse intake. The result showed that there was no difference between children dietary diversity level in KRPL and non KRPL area (p=0.259). Household income level on KRPL area was higher (68%) if compared to household on non KRPL area (44%). Parents’ education level in KRPL area was classified high (father=88%, mother=88%), while that parents’ education level in non KRPL area classified low (father=88%, mother=100%) (p=0.000). Parents in KRPL area have higher education level compare to parents in non KPRL area. Conclusion: Children dietary diversity in KRPL area are not different with non KRPL area. Children who lived in KRPL area not always have diverse intake. There is difference between household’s income level and parents’ education level. Household in KRPL area have higher income and parents in KRPL area have higher education level compare to households and parents in non KPRL area. ABSTRAKLatar Belakang: Kawasan Rumah Pangan Lestari (KRPL) merupakan program yang bertujuan untuk memperbaiki dan meningkatkan keragaman pangan keluarga, salah satunya balita. Keragaman pangan pada balita dapat berpengaruh pada kecukupan kebutuhan gizi dan berdampak pada proses pertumbuhannya. Salah satu faktor yang dapat mempengaruhi keberhasilan program KPRL adalah tingkat pendidikan orang tua.Tujuan: Tujuan dari penelitian ini adalah untuk mengidentifikasi perbedaan tingkat keragaman pangan balita dan tingkat pendapatan keluarga, dan tingkat pendidikan orang tua di rumah tangga Kawasan Rumah Pangan Lestari (KRPL) dan non KRPL Kecamatan Kamal, Kabupaten Bangkalan, Madura.Metode: Penelitian observasional dengan rancang bangun penelitian potong lintang komparatif dilakukan pada subjek penelitian yang terdiri dari 25 balita di wilayah KRPL dan 25 balita di wilayah non KRPL dengan ibu atau anggota keluarga lainnya sebagai responden. Metode simple random sampling digunakan untuk memilih sampel. Karakteristik balita dan orang tua ditanyakan melalui kuesioner. Tingkat keragaman pangan balita ditanyakan melalui instrumen Individual Dietary Diversity Score (IDDS) Data dianalisis menggunakan uji statistik Mann Whitney.Hasil: Balita yang terlibat dalam penelitian ini, baik di area KPRL maupun non KPRL, sebagian besar berusia kurang dari sama dengan 35 bulan (52%). Balita yang berasal dari wilayah KRPL sebagian besar perempuan (64%), sedangkan dari wilayah non KPRL sebagian besar laki-laki (56%). Kondisi tingkat keragaman pangan balita di wilayah KRPL tergolong rendah (52%), sedangkan tingkat keragaman pangan balita di wilayah non KRPL tergolong cukup (64%). Balita yang tinggal di rumah tangga KRPL tidak selalu mengonsumsi makanan yang beragam. Hasil penelitian menunjukkan bahwa tidak terdapat perbedaan tingkat keragaman pangan balita di wilayah KRPL dan non KPRL (p=0,259). Tingkat pendapatan keluarga balita di wilayah KRPL lebih tinggi (68%) apabila dibandingkan dengan wilayah non KPRL (44%). Tingkat pendidikan orang tua di wilayah KRPL tergolong tinggi (ayah=88%, ibu=88%), sedangkan tingkat pendidikan orang tua di wilayah non KRPL tergolong rendah (ayah=88%, ibu=100%) (p=0,000).Kesimpulan: Tingkat keragaman pangan balita di area KRPL tidak berbeda dengan balita di area non KPRL. Terdapat perbedaan tingkat pendapatan keluarga dan tingkat pendidikan orang tua balita. Keluarga di wilayah KRPL memiliki tingkat pendapatan yang lebih tinggi dan orang tua di area KRPL memiliki tingkat pendidikan lebih tinggi apabila dibandingkan dengan rumah tangga dan orang tua di wilayah non KPRL.

<p><strong>English</strong><br />Food security issues deal with critical problem, namely food demand grows faster than that of production. To achieve food self-sufficiency and food security, the Ministry of Agriculture Indonesia through Indonesian Agency for Agricultural Research and Development develops the Model of Sustainable Food Houses Region (M-KRPL) and its replication, namely the Sustainable Food Houses Region (KRPL). The concept of M-KRPL and KRPL programs needs to be refined primarily due to program design, implementation period, implementing organizations, introduced technologies, and strengthened local institutions. Implementation of M-KRPL and KRPL should be carried out through excellent social process and stages of growth, i.e. growing, developing, maturation, and self-reliance. M-KRPL replication should take account the technology use as well as community empowerment. M-KRPL and KRPL is promising in terms of technical, economic and institutional aspects. Important policy implications are: (a) taking accounts the technical aspect and social-economic characteristics of the targeted groups, (b) program period must be at least three years along with the growth stages; (c) the main M-KRPL and KRPL implementing organizations are the Assessment Institute for Agricultural Technology (AIATs) and Regional governments, respectively; (d) the technology introduced consists of nursery, farm practice, post harvest, and processing; and (e) managerial and capital development.</p><p> </p><p><strong>Indonesian</strong><br />Pembangunan ketahanan pangan dihadapkan pada permasalahan pokok, dimana pertumbuhan permintaan pangan adalah lebih cepat dari pertumbuhan produksinya. Dalam rangka mewujudkan kemandirian pangan dan ketahanan pangan, Kementerian Pertanian melalui Badan Litbang Pertanian mengembangkan Model Kawasan Rumah pangan Lestari (M-KRPL) dan replikasinya menjadi Kawasan Rumah Pangan Lestari (KRPL). Konsep program M-KRPL dan KRPL perlu disempurnakan terutama terkait dengan rancangan program, jangka waktu pelaksanaan, organisasi pelaksana, serta teknologi yang diintroduksikan dan penguatan kelembagaan lokal. Payung hukum M-KRPL dan KRPL adalah : UU No.7 tahun 1966 tentang Pangan; PP No.68 tantang Ketahanan pangan; PP No. 22 tahun 2009 tentang Kebijakan Percepatan Penganekaragaman Konsumsi Pangan Berbasis Sumber Daya Lokal; serta PP No. 43 tahun 2009 tentang Gerakan Percepatan Penganekaragaman Konsumsi Pangan Berbasis Sumber Daya Lokal. Implementasi replikasi M-KRPL menjadi KRPL seyogyanya dilakukan melalui proses sosial yang matang melalui tahap penumbuhan, pengembangan, pematangan, dan kemandirian. Replikasi M-KRPL menggunakan entry point teknologi dan sekaligus kelembagaan, serta berdasarkan prinsip pemberdayaan masyarakat. Pengembangan M-KRPL dan KRPL memiliki prospek baik dan berlanjutan ditinjau dari aspek teknis, ekonomi dan kelembagaan. Implikasi kebijakan penting adalah : (a) rancangan program harus memperhatikan aspek teknis dan karakteristik sosial ekonomi kelompok sasaran; (b) jangka waktu program minimal 3 (tiga) tahun melalui tahap penumbuhan, pengembangan, dan kemandirian; (c) organisasi pelaksana utama M-KRPL adalah BPTP, sedangkan KRPL adalah Pemerintah Daerah; (d) teknologi yang diintroduksikan mencakup teknologi pembibitan, budidaya, serta pasca panen dan pengolahan hasil; dan (e) penguatan kelembagaan pengelola M-KRPL dan KRPL baik dari aspek manajemen, permodalan, dan partisipasi anggota.</p>

Abstract:
The subject of this thesis is the comprehensive research and design of costumes for the play Ubu Roi by Alfred Jarry, including documentation of the creative process from the initial analysis of the play to the final designs. This play was written in the late 19th century and is considered to be a precurser to Surrealism and Theatre of the Absurd. The story is about the brief and tyrannical reign of a hideous, backward individual named Ubu, who comes to power by murdering the Polish king. The play is notable for its rough playfulness, ruthlessness with regard to social and cultural traditions, its open-mindedness and unique use of language. On the surface it comes across as a silly farce, but it contains many deeper undertones that cause Jarry to be sometimes regarded as a symbolist writer. This work is divided into a theoretical and practical part. The theoretical part deals with the author of the play, its content and the context in which it was written, and studies how it was staged in the past. It is not the aim to offer a coherent overview of the life of Alfred Jarry, his work, or the history of past productions of King Ubu. Instead, the information contained here is intended only as a documentation of the historical and theoretical research necessary to understand this work, which ultimately had an impact on the final designs. Part two documents the process of developing the costume designs themselves. Initially it introduces the key considerations that arose from reading the play that affected the overall progress of the work. There follows an analysis of the characters and situations based on the text, and sources of inspiration. The main section describes the final designs of the set and costumes that are supported by attached images.

This diploma thesis aims at strategic strategic differentiation of two brands. The main goal of the thesis is to analyze the current marketing strategy of the brands that are competitive to each other in the category of white mold cheese. The brands have a common owner. They facing a continuous decline in market share. At the same time, the whole category of white mold cheese is growing. The thesis is divided into two parts.In the first part,there are described theoretical approaches of creating marketing strategy, brand management, brand differentiation and market segmentation. It contains various concepts of shaping marketing strategies and their realization. In the second part, there is an analysis of the brands, the market situation and the consumer behavior. In the final part, the author proposes recommendations of the future marketing strategy.

The subject of the diploma thesis is design a new building with a camping in the village Šoporňa. The building is situated near the river Váh on flat terrain. Emphasis was placed on the total visual impression of the building on its stand out from the surrounding landscape. The building will serve as a recreation building with the possibility of short and long-tern housing. There is a restaurant with outdoor seating and an accommodation capacity for 90 people, trade with sport equipment, service for vehicles and boats, garage for boats, a 11,6 metres high climbing wall, conference rooms and halls for exercise. The building has three floors above and no basement. The project includes a flat roof with an attic.

Abstract:
Die Kryo-Elektronentomographie (Kryo-ET) ermöglicht als einzigartige Methode die dreidimensionale Visualisierung der makromolekularen Struktur von Eis-eingebetteten Zellen in ihrem nativen Zustand [Baumeister 2005; Leis et al. 2009]. Ziel dieser Arbeit war es einen universellen Einsatz der Kryo-ET bei eukaryotischen Zellen zu ermöglichen. Dazu wurden neue Ansätze entwickelt bzw. bestehende Methoden optimiert. Da bei der Anwendung der Tomographie die Probendicke mit jedem Kippwinkel zunimmt, gilt momentan die Probendicke (< 1 µm) als limitierender Faktor bei der Kryo-ET. Während prokaryotische Zellen nahezu routinemäßig mittels Kryo-ET untersucht werden können, ist dies bei eukaryotischen Zellen nur partiell in peripheren Arealen oder Ausläufern der Zelle möglich. Dies konnte anhand der Untersuchung von intakten, pluripotenten Stammzellen (P19 Zellen) bestätigt werden. Aufgrund ihrer neuronen-ähnlichen dendritischen Morphologie können diese dünnen Bereiche mit dem Elektronenstrahl durchdrungen werden. So konnte in der 3D-Rekonstruktion eines Zellausläufers ein Mitochondrium in seiner nativen Umgebung visualisiert werden. Zudem wurden mehrere ATP-Synthasen in der Cristaemembran identifiziert, die erstmalig die Existenz von ATP-Synthasedimeren in situ bestätigen. Für die Untersuchung von zellmittigen (dicken) Bereichen müssen jedoch andere Methoden angewendet werden. So ermöglicht die Kryo-Ultramikrotomie das Herstellen von Dünnschnitten (< 100 nm) Eis-eingebetteter Proben. Mit Hilfe dieser Methode wurden in dieser Arbeit Kryo-Schnitte von HL-1 Kardiomyozyten erstellt und tomographisch analysiert. Da die Probe sehr heterogen verteilt ist, ist die Suche nach der Zielstruktur im Elektronenmikroskop sehr zeitaufwendig. Gleichzeitig ist die strahlenempfindliche Probe während der Suche dem Elektronenstrahl ausgesetzt, was die Struktur beeinträchtigen kann. Um die ‚Effizienz’ der Kryo-ET an Kryo-Schnitten zu erhöhen, wurden zwei neue Verfahren implementiert: Einerseits die korrelative Kryo-Fluoreszenzmikroskopie, welche sich zur Suche und Identifikation von Mitochondrien innerhalb des Dünnschnittes unter Flüssigstickstoff (LN2)-Temperatur eignet und andererseits eine neue Methode, die das Aufbringen von Goldkolloiden auf Kryo-Schnitten zum späteren Alignieren der Kippserie ermöglicht. Letztere setzt eine Synthese von 10 nm großem, kolloidalem Gold in Toluol voraus. Nach Zugabe von Isopentan werden die auf dem EM-Trägernetzchen (Grid) angehefteten Kryo-Schnitte bei einer Temperatur von -150°C in diese Suspension getaucht. Des Weiteren wurden verschiedene Trägermaterialien zur Kultivierung von Kardiomyozyten getestet und Osmolalitätsmessungen von unterschiedlichen Kryo-Schutz¬lösungen, welche für das Hochdruck-Verfahren notwendig sind, durchgeführt. Auch hier konnten in den 3D-Rekonstruktionen von Kardiomyozyten die ATP-Synthasen eindeutig in Kryo-Schnitten identifiziert werden. Darüber hinaus gelang die 3D-Visualisierung von zwei Mitochondrien, die sich in der Teilungs- oder Fusionsphase befanden. In einem dieser Mitochondrien sind inorganische Ablagerungen sichtbar. Im Verlauf dieser Dissertation wurde zusätzlich die Methode des Cryo-Planings entwickelt; eine Variante der Kryo-Ultramikrotomie. Bei dieser Technik wird die vitrifizierte Probe direkt auf dem EM-Grid gedünnt. Dieses Verfahren ermöglicht es, Material vom vitrifizierten Eisfilm mittels eines Diamantmessers zu entfernen. Dafür wurde ein spezieller Halter für das Kryo-Ultramikrotom konzipiert und hergestellt. Der Halter erlaubt das Zentrieren und Klemmen eines EM-Grids. Um das Abtragen kleinerer Bereiche der Eisoberfläche zu ermöglichen, wurde ein 1 mm breites Diamantmesser angefertigt. Die Analyse der gedünnten Proben mittels Kryo-Scanning Electron Microscopy (SEM) zeigte eine gleichmäßig abgetragene Oberfläche. Schneideartefakte, wie sie bei der Kryo-Ultramikrotomie auftreten, wurden nicht beobachtet. Zudem sind die zellulären Proben im gedünnten Bereich des Eisfilms sehr leicht identifizierbar. Brüche, die möglicherweise durch den Probeneinbau im Eisfilm entstehen, konnten mittels Kryo-SEM bis dato nicht beobachtet werden. Die theoretischen Betrachtungen ergaben, dass unter Verwendung des Cryo-Planings als alleinige Methode elektronentransparente Bereiche (< 1 µm Dicke) hergestellt werden können. Bisher konnte jedoch keine elektronentomographische Untersuchung einer geplanten Probe erfolgen, da sie sich als zu dick erwies. Dies ist darauf zurückzuführen, dass mit dem Stereomikroskop nur eine sehr grobe Abschätzung der tatsächlichen Dicke des abgetragenen Bereichs möglich ist.

Abstract:
Small data has been a barrier for many machine learning tasks, especially when applied in scientific domains. Fortunately, we can utilize domain knowledge to make up the lack of data. Hence, in this paper, we propose a hybrid model KRL that treats domain knowledge model as a weak learner and uses another neural net model to boost it. We prove that KRL is guaranteed to improve over pure domain knowledge model and pure neural net model under certain loss functions. Extensive experiments have shown the superior performance of KRL over baselines. In addition, several case studies have explained how the domain knowledge can assist the prediction.

Knowledge representation learning (KRL) aims at encoding components of a knowledge graph (KG) into a low-dimensional continuous space, which has brought considerable successes in applying deep learning to graph embedding. Most famous KGs contain only positive instances for space efficiency. Typical KRL techniques, especially translational distance-based models, are trained through discriminating positive and negative samples. Thus, negative sampling is unquestionably a non-trivial step in KG embedding. The quality of generated negative samples can directly influence the performance of final knowledge representations in downstream tasks, such as link prediction and triple classification. This review summarizes current negative sampling methods in KRL and we categorize them into three sorts, fixed distribution-based, generative adversarial net (GAN)-based and cluster sampling. Based on this categorization we discuss the most prevalent existing approaches and their characteristics.
