AbstractIt took almost a century to develop electron microscopy into a powerful method for high-resolution structure determination of proteins. Technical improvements in microscopy, detector technology, and image processing software contributed to the exponential growth of high-resolution structures of protein complexes determined by cryo-electron microscopy in recent years. We now succeeded in breaking another resolution barrier in cryo-electron microscopy and for the first time in achieving true atomic resolution, where single atoms in the protein can indeed be visualized individually. These improvements in cryo-EM indicate that the method will continue to gain importance, not only as a method for structure determination but also in the development of new drugs in pharmaceutical research.

With the advent of `courier crystallography', the safe and reliable transportation of cryo-cooled crystals in dry Dewars has become an important factor in the outcome of an experiment. The results of measurements of the long-term temperature characteristics of a dry Dewar are presented, together with the variation in properties of a `cryo-vial', a `kryo-vial' and a Rigaku MSC ACTOR `puck'. The short-term effects of removing and replacing the top-hat (`cryo-cap') holder from the Dewar have also been investigated, and the implications for best practice on synchrotron trips are discussed.

Abstract:
A program of early postoperative rehabilitation (kinesy therapy and education) has been conducted since 1996 in our Department for Rehabilitation at the Institute of Oncology in Sremska Kamenica. The aim of this program is to prevent the appearance of secondary lymphedema of the arm and the contacture of the shoulder. Unless the patient, in further follow up, does not have any complication, a program of "late" rehabilitation is conducted. If secondary lymphedema of the arm (SLEA) is evolved, then the patient is submitted to a complex of decongestive physical therapy (CDP) or CDP and sequential pneumatic compression (SEPC). However if SLEA and local alterations on the skin exist, we immobilize the arm (mitela, plaster). In other cases without SLEA, e.g. periarthritis or the damage of brachial plexus, we perform physical procedures (kinesy therapy TENS, kryo-massage and acupuncture). With regard to the degree of complication of SLEA and consequences that might develop the starting point should be directed towards early detection of SLEA.

Abstract:
This paper proposed a liquid level measurement and classification system based on a fiber Bragg grating (FBG) temperature sensor array. For the oil classification, the fluids were dichotomized into oil and nonoil, i.e., water and emulsion. Due to the low variability of the classes, the random forest (RF) algorithm was chosen for the classification. Three different fluids, namely water, mineral oil, and silicone oil (Kryo 51), were identified by three FBGs located at 21.5 cm, 10.5 cm, and 3 cm from the bottom. The fluids were heated by a Peltier device placed at the bottom of the beaker and maintained at a temperature of 318.15 K during the entire experiment. The fluid identification by the RF algorithm achieved an accuracy of 100%. An average root mean squared error (RMSE) of 0.2603 cm, with a maximum RMSE lower than 0.4 cm, was obtained in the fluid level measurement also using the RF algorithm. Thus, the proposed method is a feasible tool for fluid identification and level estimation under temperature variation conditions and provides important benefits in practical applications due to its easy assembly and straightforward operation.

Abstract:
Despite its use by humans for thousands of years, the technology of cannabis usage and extraction is still evolving. Given that the primary pharmacological compounds of interest are cannabinoid and terpenoids found in greatest abundance in capitate glandular trichomes of unfertilized female inflorescences, it is surprising that older techniques of hashish making have received less technological advancement. The purpose of this study was to employ organically grown cannabis and to isolate pure trichomes from freshly picked flowers via exposure to vapor from solid CO2, commonly known as “dry ice”, followed by their isolation via sifting through a 150 µ screens while maintaining the cold chain. Biochemical analysis was undertaken on fresh flower, frozen-sifted flower by-products, treated trichomes (Kryo-Kief™), dried flower, dried sifted flower by-product and dried kief. The dry ice process successfully concentrated cannabinoid content as high as 60.7%, with corresponding concentration and preservation of monoterpenoids encountered in fresh flower that are usually lost during the conventional cannabis drying and curing process. The resulting dried sifted flower by-product after dry ice processing remains a usable commodity. This approach may be of interest to pharmaceutical companies and supplement producers pursuing cannabis-based medicine development with an eye toward full synergy of ingredients harnessing the entourage effect.

Particular attention is required to improve cryopreservation of embryonic stem cells (ESC) and study their characteristics. Stem cells were obtained from the inner cell mass of Day 5-6 blastocysts. The ESC were then cultured on mTeSR™1 medium (Stemcell Technologies, Cambridge, MA, USA). We studied the survival of ESC after slow freezing and vitrification. Slow freezing was carried out using a Planer Kryo 360-3.3 freezer (Planer plc, Sunbury-on-Thames, United Kingdom), using various cryoprotectants: 1.5 M dimethyl sulfoxide (Me2SO), 1.5 M ethylene glycol (EG), or 1.5 M propylene glycol (PG). Six vitrification solutions (VS) were used to vitrify ESC: VS1 = 20% Me2SO + 20% EG + 0.5 M sucrose; VS2 = 20% Me2SO + 20% PROH + 0.5 M sucrose; VS3 = 20% EG + 20% PG + 0.5 M sucrose; VS4 = 20% Me2SO + 20% EG + 0.5 M sucrose + 10% FCS; VS5 = 20% Me2SO + 20% PROH + 0.5 M sucrose + 10% FCS; and VS6 = 20% EG + 20% PG + 0.5 M sucrose + 10% FCS. For the dehydration of cells and the addition of vitrification solutions, a 3-step equilibration was used. The proliferative properties of the cells were determined using an Apel PD-303S spectrophotometer (Apel Co. Ltd., Kawaguchi, Japan), using an MTT test (staining with methylthiazolyl-diphenyl tetrazolium). After slow freezing, the highest percentage of frozen–thawed cells proliferating was observed when using 1.5 M EG (P > 0.05). At the same time, the highest cell doubling after thawing was observed when using 1.5 M EG, and 1.5 M Me2SO. After vitrification, the highest percentage of proliferation was observed in the VS2 and VS4 groups (49.7 ± 3.2% and 53.2 ± 3.8%, respectively). It should be noted that the addition of fetal calf serum to the vitrification solution also increased the proliferation of ESC after vitrification and thawing.

Kroy-processed and original wool fabric were successfully modified by TiO2/SiO2 70:30 sol with the stabilization of 60g/L 1,2,3,4-butanetetracarboxylic acid (BTCA). In order to explore the influence of Kroy-process on TiO2/SiO2 sol treatment, self-cleaning property of original and Kroy-processed wool samples coated with TiO2/SiO2 sol were investigated by a Color Measuring & Matching System. In addition, the contact angles of four kinds of wool fabric (original wool, Kroy-processed wool, TiO2/SiO2 coated original wool and TiO2/SiO2 coated Kroy-processed wool) were respectively evaluated to achieve the effect of Kroy-process on the hydrophilicity of wool fabric. The results revealed that the applying of Kroy-process is conducive to the coating of TiO2/SiO2 sols on wool fabric. The total color difference ∆E of coated wool increased 42% from 30.35 to 43.09 after the application of Kroy-process. Besides, the hydrophilicity of wool fabric was enhanced since the contact angle of Kroy-processed wool decreased to 133.5° while original wool was 140.3°. Furthermore, coated samples with Kroy-process had lower value of contact angle as compared with coated original wool fabric.

This study was aimed at investigating factors militating against the effective teaching and learning of Krio at the Junior Secondary School (JSS) Level in a sample of JSS pupils in selected schools in the Western Area of Freetown, Sierra Leone. Questionnaires were administered to both teachers of Krio and pupils offering the subject at the targeted level. These, alongside observation and focus group discussions, were used to determine the level of training and experience of the teachers; facilities which support the teaching and learning of Krio; attitudes of teachers, pupils and other stakeholders towards the teaching and learning of Krio; mode and frequency of assessment; provision of institutional support; and, the success rate of pupils at the Basic Educational Certificate Exams (BECE) in the selected schools. Three schools were targeted. In each of these schools, questionnaires were administered to 50 pupils, giving a total of 150 respondents. A total of eleven teachers were sampled. The study revealed that: The teachers of Krio in the selected schools were and trained and qualified. However, none of them studied krio as a major and only about one third read it as a minor. Teaching and learning materials are inadequate, and time allocated to the teaching of krio is insufficient. Attitude is a major factor which militates against the teaching and learning of krio in the selected schools. Written assignments, tests and exams are the main assessment tools. Institutional support (in-service trainings/workshops/seminars) plays a great role in building teachers’ capacity to teach the subject as all the teachers sampled did not study krio as a major subject. BECE results (krio -2010 and 2011) indicate that only a small percentage of the pupils at JSSIII opted for krio

Tujuan penelitian ini adalah: (1) Untuk mengetahui dampak pembangunan pemukiman di sekitar kota Semarang kerusakan lahan DAS Kreo; (2) Mengetahui cara mengkonservasi lahan di DAS Kreo supaya tidak rusak. Metode penelitian dilakukan metode survei. Lokasi penelitian dilakukan di DAS Kreo Kota Semarang dan sekitarnya. Teknik pengumpulan data observasi, pengukuran lapangan, dokumentasi, pemetaan sistem informasi geografis. Teknik analisis data analisis spasial berbasis SIG, analisis deskriptif kuantitatif, dan analisis metode tabel silang. Hasil penelitian menunjukkan bahwa tingkat kerusakan lahan yang ada di DAS Kreo termasuk dalam kriteria tinggi. Hal ini disebabkan karena pembangunan Waduk Jatibarang selain itu juga faktor geofisik seperti kemiringan lereng, geologi, tanah, penggunaan lahan, dan curah hujan yang tinggi. Abstrak. Telah dilakukan penelitian mengenai perilaku monyet ekor panjang (Macaca fascicularis Raffles) dan persepsi pengunjung di obyek wisata Goa Kreo yang terletak di Desa Kandri Kecamatan Gunung Pati Kota Semarang. Penelitian dilakukan pada bulan November 2020, dimana pada saat yang sama terjadi pandemi covid-19 yang melanda di hampir seluruh negara di dunia ini. Pandemi Covid-19 tersebut memaksa pengelola Obyek Wisata Goa Kreo untuk menutup operasional kunjungan wisata selama 3 bulan. Observasi dilakukan selama 150 menit dengan interval 15 menit, sementara itu responden merupakan pengunjung Goa Kreo yang berasal dari berbagai latar belakang. Hasil observasi MEP menunjukkan bahwa perilaku monyet ekor panjang di Goa Kreo menunjukkan 25% diam, 17% bergerak, 15% Tidur 13% Self Grooming, 11% grooming, main dan makan masing-masing 10% dan 9%. Sementara itu persepsi pengunjung menunjukkan Goa Kreo menunjukkan 76% tidak mengetahui status konservasi MEP, 12% tidak setuju MEP diberi makan tambahan. In February 2020, a unique publication came out under the title “Kryz-Azerbaijani-English dictionary”, prepared by I. A. Gummetov and M. Rind-Pavlovsky. The publication was presented on February 8, 2020 in the Khachmas ethnographic Museum in Azerbaijan with the invitation of the Kryz intelligence and representatives of the Khachmas city hall. Employees of the Tsadasa Institute of language, literature and art of the Russian Academy of Sciences were also invited to the presentation of the dictionary in Azerbaijan. The present dictionary is exceptional in its originality and completeness as far as the various lexical units are concerned from the point of their description and specification. The Kryz language is one of the non-written Lezgian languages of the Nakh-Dagestan language family. The Kryz language is used in the household sphere. The reviewed book with a description of the Kryz language lexemes consists of an introduction, two chapters, a list of abbreviations, and an Appendix. The description of the Kryz language lexemes is confirmed by thematic indexes from the Azerbaijani and English languages. The dictionary entry includes grammatical and stylistic characteristics of the three languages. Maintenance of population viability, especially endangered species, requires preservation of as much genetic variability as possible, therefore genetic resource banks are very important. For male gametes, preservation of all available sources (ejaculates and epididymal) are useful. Information regarding sperm characteristics of most wild ruminant species is limited compared to that from domestic species. The objective of this work was to characterize the freezability of electroejaculated and epididymal spermatozoa from a wildebeest (5 year old; housed in Cabárceno Nature Park, Cantabria, Spain) that was castrated because of behavioral problems. After general anesthesia (ethorfine + xilazine, 1.8 mL + 0.5 mg kg–1) with dart, semen was collected by electroejaculation (3 V and 75 mA). Sperm concentration was 250 × 106 mL–1 (total spermatozoa: 1128.6 × 106). After castration, epididymides were disected and spermatozoa were collected by making several incisions in the caudal epididymis. Concentration was 12 441 × 106 spermatozoa mL–1 (total spermatozoa: 24 882 × 106). Samples were diluted to 200 × 106 spermatozoa mL–1 (TesT-Fructose-Egg yolk-Glycerol-Antibiotics) and chilled to 5°C during 2 h. Diluted semen was packaged in 0.25-mL straws and frozen from 5°C to –100°C (–20°C min–1) in a programmable cell freezer (Kryo 10, Planer). Straws were plunged into liquid nitrogen until analysis and thawed in a water bath (65°C, 6 s). Fresh, pre-freezing and post-thawed samples were analysed for motility (total motility TM, %; progressive motility PM, %; path velocity VAP, μm s–1; track speed VCL, μm s–1; progressive velocity VSL, μm s–1) using a CASA (ISAS, Proiser, Valencia, Spain). Viability (VIAB %) (SYBR-14 and propidium iodide) and mitochondrial membrane potential (MIT %) (JC1) were assessed by flow cytometry. Post-thawing results for electroejaculated v. epididymal samples were, respectively: TM: 87.0 v. 64.6%; PM: 68.7 v. 33.4%; VAP: 95.9 v. 49.8 μm s–1; VCL: 108.3 v. 71.6 μm s–1; VSL: 86.7 v. 40.2 μm s–1; VIAB: 57.0 v. 73.9%; MIT: 59.5 v. 77.5%. Motility parameters were higher for the electroejaculated sample; however, viability was higher for the epididymal sample. Recovery rates (post-thawed value/pre-freezing value × 100) for electroejaculated v. epididymal samples were: TM: 97.2 v. 93.4%; PM: 113.3 v. 103.2%; VAP: 88.9 v. 122.0 μm s–1; VCL: 87.5 v. 126.0 μm s–1; VSL: 93.4 v. 125.3 μm s–1; VIAB: 75.0 v. 97.7%; MIT: 69.2 v. 95.9%). These rates suggest a good freezability of electroejaculated and epididymal spermatozoa in blue wildebeest. Kampung Hijau Mandiri is one of Tangerang Government’s Programs to increase public awareness for a clean, green, and beautiful surrounding. Ninety-eight of Kampung Hijau Mandiri were formed throughout Tangerang’s Sub-Districts, one of which is in RT 01 RW 06, Kreo Sub-District. Criteria for Kampung Hijau Mandiri include a lively, comfortable and clean village with waste sorting initiatives. Kreo, one of the densely populated villages in Tangerang, has 22,837 inhabitants. Waste generated in RT 01 RW 06, Kreo, Tangerang is around 2,040 liters/day, quite large and can be reduced. Presently, household wastes in RT 01 RW 06 are still managed conventionally using the collection and disposal system. The economic potential of household organic and inorganic waste has not been optimally utilized. Organic waste management through composting (Takakura baskets) and non-organic waste management through waste banks are considered appropriate for processing household wastes. Hence, the community develops a new habit of sorting and processing waste into productive means, reducing waste generation transported to landfills. The waste management educational mentoring program was attended by 14 people, especially housewives and Posyandu cadres, through presentations and question and answer. This activity has great potential to endure as housewives and Posyandu cadres in RT 01 RW 06 Kreo Sub-District are influential household stakeholders in their environment. Residents of RT 01 RW 06 Kreo Sub-District agreed to initiate waste bank establishments. This program is expected to increase citizen’s involvement in waste management. RT 01 RW 06 Kreo is an exemplary Kampung Hijau Mandiri.

Informasi penutup lahan merupakan data yang sangat penting dalam pengelolaan Daerah Aliran Sungai (DAS). Tantangan dalam penyediaan informasi penutup lahan di DAS Kreo adalah tutupan awan dan cangkupan areanya yang cukup luas. Hadirnya platform pengolahan data spasial berbasis cloud yaitu Google Earth Engine (GEE) bisa menjawab tantangan tersebut. Oleh karena itu penelitian ini bertujuan untuk memetakan penutup lahan di DAS Kreo menggunakan klasifikasi berbasis machine learning pada GEE. Proses pemetaan penutup lahan di DAS Kreo menggunakan citra satelit Landsat 8 dan DEM SRTM. Input data yang digunakan antara lain band 1 sampai 7 pada citra Landsat 8, transformasi NDVI dan NDBI serta nilai elevasi dari DEM SRTM. Adapun tahun yang dipilih adalah tahun 2015 dan 2020 dengan machine learning yang diujikan meliputi CART, Random forest dan Voting SVM. Hasil penelitian ini menunjukkan bahwa machine learning yang terbaik dalam memetakan penutup lahan di DAS Kreo adalah Random forest. Penelitian ini masih terdapat banyak keterbatasan terutama kelas penutup lahan yang dipetakan.

Seni tari Wanara Parisuka merupakan salah satu kesenian yang terdapat di dalam rangkaian ritual sesaji Rewanda di Goa Kreo Semarang. Metode yang digunakan dalam penelitian ini adalah kualitatif yang bersifat deskriptif dengan pendekatan etnografis dan teknik observasi, wawancara, dokumen belajar serta pengumpulan data. Tulisan ini membahas permasalahan tentang mitos dalam pertunjukan tari Wanara Parisuka pada ritual sesaji Rewanda di Goa Kreo Semarang, dengan pendekatan teori mitos Mircea Eliade sebagai pisau analisis. Hasil yang diperoleh dalam studi ini menunjukkan bahwa; pertama, tari Wanara Parisuka di Goa Kreo adalah jenis tarian sakral dengan estetika mistis religious. Kedua, tari Wanara Parisuka adalah bagian terpenting di akhir seluruh rangkaian Ritual Sesaji Rewanda, karena merupakan puncak ritual dalam masyarakat dusun Talunkacang Goa Kreo. Ketiga dari rangkaian acara ritual sesaji Rewanda, tari Wanara Parisuka adalah mewakili lima tipe mitos yaitu: 1) mitos kosmogoni, 2) mitos asal-usul, 3) mitos tentang dewa-dewa dan makhluk Illahi, 4) mitos androgoni, 5) mitos akhir dunia. Berdasarkan hasil penelitian, menempatkan meneruskan saran bahwa, pertama-tama, Departemen Pendidikan dan Budaya perlu memasukan seni tari Wanara Parisuka dalam pembelajaran di sekolah. Kedua, seniman perlu memperkenalkan lebih dalam tari Wanara Parisuka melalui studi. Ketiga, masyarakat Goa Kreo yang perlu memiliki kepedulian yang tinggi terhadap pentingnya mempertahankan, dan melestarikan tari Wanara Parisuka sebagai destinasi wisata unggulan di Semarang. Abstract:
The development of obesity is characterized by the metabolic overload of tissues and subsequent organ inflammation. The health effects of krill oil (KrO) on obesity-associated inflammation remain largely elusive, because long-term treatments with KrO have not been performed to date. Therefore, we examined the putative health effects of 28 weeks of 3% (w/w) KrO supplementation to an obesogenic diet (HFD) with fat derived mostly from lard. The HFD with KrO was compared to an HFD control group to evaluate the effects on fatty acid composition and associated inflammation in epididymal white adipose tissue (eWAT) and the liver during obesity development. KrO treatment increased the concentrations of EPA and DHA and associated oxylipins, including 18-HEPE, RvE2 and 14-HDHA in eWAT and the liver. Simultaneously, KrO decreased arachidonic acid concentrations and arachidonic-acid-derived oxylipins (e.g., HETEs, PGD2, PGE2, PGF2α, TXB2). In eWAT, KrO activated regulators of adipogenesis (e.g., PPARγ, CEBPα, KLF15, STAT5A), induced a shift towards smaller adipocytes and increased the total adipocyte numbers indicative for hyperplasia. KrO reduced crown-like structures in eWAT, and suppressed HFD-stimulated inflammatory pathways including TNFα and CCL2/MCP-1 signaling. The observed eWAT changes were accompanied by reduced plasma leptin and increased plasma adiponectin levels over time, and improved insulin resistance (HOMA-IR). In the liver, KrO suppressed inflammatory signaling pathways, including those controlled by IL-1β and M-CSF, without affecting liver histology. Furthermore, KrO deactivated hepatic REL-A/p65-NF-κB signaling, consistent with increased PPARα protein expression and a trend towards an increase in IkBα. In conclusion, long-term KrO treatment increased several anti-inflammatory PUFAs and oxylipins in WAT and the liver. These changes were accompanied by beneficial effects on general metabolism and inflammatory tone at the tissue level. The stimulation of adipogenesis by KrO allows for safe fat storage and may, together with more direct PPAR-mediated anti-inflammatory mechanisms, attenuate inflammation.

The aim of this study is to measure the influence of service quality, destination competitiveness, and previous experience toward revisit decision trough tourist satisfaction. The population of this resarch is visitors of Goa Kreo, while the sample is 100 visitor of Goa Kreo with some certain prerequirements. The data was analyzed using two-step analysis with SPSS 17.0 for windows where the validity test, reliability test,doefficient of correlation, coefficient of determination, test of significance (t-test and f-test) in advance has been done before.The result of this study is showing that service quality, destination competitiveness and previous experience has positive impact toward visitor satisfaction, where the biggest impact is own by previous experience. Based on the result of two-step analysis, tourist satisfaction is an intervening variable toward revisit decision in this study.Some recomendations are added based on those result. It’s recomended that hygine, security, and hospitality in Goa Kreo must be incerased. Moreover, it’s neccesary to increase destination competitiveness like facility’s maintenance and added up some general facility so that visitor will be satisfied with their experiences and willing to come back to Goa Kreo.

Watershed as rainwater catchment are is susceptible with erosion and sedimentaion problems. The calculation of erosion and sediment yield can be calculated with USLE and SDR (Sediment Delivery Ratio) method at each area unit. This research intends to find out the erosion value, distribution and prediction of sediment yield contained in Kreo Hulu Watershed in 2009-2014. Data accumulation methods for the research are observation, documentation, and field measurement. The erosion forecast result in this research obtained based on overlay few maps, that is rain erosivity map, soil erodibility map, length of slope map, management and conservation soil map. This research was construct with using rainfall data from 2009 until 2014. Based on the calculations model of erosion is known that the erosion in the watershed upstream Kreo fluctuation occurring annually. With the result of erosion from 2009 until 2014 are respectively 17.45 tons; 18.78 tons; 17.83 tons; 25.70 tons; 31.62 tons; 26.17 tons. Sediment yield estimates obtained by the relationship between the unit value erosion (based mapping unit), as well as the erosion area of the unit value of Sediment Delivery Ratio in watershed. By calculation of these factors, the value of watershed sediment Kreo Hulu forecast results for 2009-2014 are respectively 2,78 tons, 2.93 tons; 2,78 tons; 4.01 tons; 4.93 tons; 4,08 tons. Spatial distribution of erosion prediction Kreo upstream watershed of very high value erosion is largely rural area/village Gunungpati, Banjarejo, Karangmalang, Lebak and Cepoko. Weight and middle erosion are covering parts of Pasigitan, Branjang, Kalisidi, and Tinjomoyo. Then need to increase conservation of alternative methods land mechanically and vegetative at upstream Kreo watershed.

Abstract:
Whole-ovary cryopreservation followed by retransplantation with vascular anastomosis is a promising method to restore cancer-patient fertility. However, protocols need to be improved to minimize the damages related to an inadequate cooling rate caused by the large volume of whole organs. The aim of this study was to compare conventional slow-freezing (SF) versus directional-freezing (DF) techniques. Of 30 sheep ovaries collected at the local abattoir, 6 were used as fresh controls and 24 were perfused for 5 min with cryoprotectant solution made of Leibovitz L-15 medium, 10% FCS, and 1.5-M dimethyl sulfoxide. Samples were inserted in glass freezing tubes filled with the same solution. Twelve ovaries were frozen with a Kryo 560M apparatus (Planer, UK) using a cooling rate of 0.5°C min–1 from 4 to –40°C and 5°C min–1 from –40 to –100°C followed by direct plunging in liquid nitrogen. The other 12 ovaries were frozen using an MTG 1315 apparatus (Core Dynamics, Ness Ziona, Israel). Tubes were pushed along the thermal gradient (4 to –70°C) at 0.01 mm s–1 resulting in a cooling rate of 0.3°C min–1 and then plunged in liquid nitrogen. After 2 weeks, samples were thawed at 37°C for 2 min with gentle shaking. Morphological analysis showed that the rate of intact follicles was 94% in control samples, 87% in DF samples, and 58.3% in SF samples (P < 0.05). No significant differences were found in stromal cell density between fresh and DF ovaries (17 833 and 17 041 nuclei mm–2, respectively), whereas SF ovaries showed a lower density (10 875 nuclei mm–2; P < 0.001). To assess vessel integrity, thawed ovaries were perfused with 25% Indian ink/167 UI of heparin mL–1 saline solution and embedded in paraffin. Fresh and DF ovaries displayed a similar rate of perfused vessels (82.7 and 80.2%, respectively), whereas SF ovaries significantly lower number of perfused vessels (52.3%), indicating the occurrence of vascular injury. Follicle viability after freezing was evaluated by culturing individual cortical fragments (2 × 2 × 1 mm) for 7 days. A high rate of morphologically normal follicles was found in both fresh (85%) and DF ovaries (78%). The number of intermediate and primary follicles was significantly increased in both groups when compared to Day 0 (P < 0.001). By contrast, cortical slices obtained from SF ovaries showed a higher rate of degenerated follicles, oocytes with pyknotic nuclei, and a significant decrease of follicle number. Furthermore, qPCR and immunohistochemical assay for Ki-67 showed a similar cell proliferation in fresh and DF ovary but a significantly lower one in SF samples. We conclude that directional freezing allows a better preservation of both cortical and stromal tissue, as well as the vasculature, thus overcoming 2 of the main restrictions associated with whole-organ freezing. Both tissue and cell cryopreservation can be applied for biodiversity conservation. The proper preservation of tissues and cells from a wide range of animals of different species is of paramount importance, because these cell samples could be used to reintroduce lost genes back into the breeding pool by somatic cell cloning. The aim of this work was to investigate the effect of different cooling rates on viability of frozen–thawed sheep fibroblasts for conservation of biodiversity so that it might be used in the future to provide nuclear donors (Table 1). Skin samples collected from 10 adult sheep were cut on small pieces (1 × 1 mm), placed into culture Petri dishes containing DMEM supplemented with 20% (v/v) fetal bovine serum (FBS), and covered with coverslips followed by incubation at 5% CO2, 95% relative humidity, and 37°C. During culture, fibroblasts left skin samples and proliferated. Culture medium was changed every 4 days. After 21 to 22 days of incubation, a fibroblast monolayer was observed, culture medium was removed, and cells were incubated for 7 to 10 min in presence of Dulbecco’s phosphate buffered saline + 0.25% trypsin. Dissociated fibroblasts were washed with DMEM by centrifugation at 300g for 10 min. For cryoconservation, fibroblast samples were then diluted at a concentration of 2 × 106 cells mL–1 in DMEM + 20% FBS and 10% dimethyl sulfoxide or 10% ethylene glycol and placed into 0.25-mL plastic straws or 2-mL cryovials. Straws were sealed with modeling clay and maintained at +5°C for 120 min before freezing. Cryopreservation of fibroblasts was carried out by 2 procedures: (1) straws were frozen in programmable freezer Kryo Planer 360-3,5 using the following freezing regimen: +5°C to –40°C at –1°C min–1, –40°C to –85°C at –4°C min–1, and then plunged into liquid nitrogen; (2) cryovials were placed in a Styrofoam box and loaded into a freezer at –70°C for 24 h, and then samples were plunged into liquid nitrogen for storage. Samples were thawed for 1 min in a 37°C water bath. Frozen–thawed samples were diluted with DMEM (1 : 5) and centrifuged at 300g for 7 to 10 min. Supernatants were removed, and cells were diluted with DMEM at a concentration of 2 × 106 cells mL–1. Viability of frozen–thawed fibroblast samples was detected using the Trypan Blue staining method. The values obtained (Table 1) are expressed as mean standard error of the mean (SEM). Statistical analysis was done using Student’s test. Results indicated that there was a significant difference in viability between fresh and cryopreserved fibroblasts. However, there were no differences between the cooling procedures. Importantly, our data suggest that the use of 1.5-M ethylene glycol reduced the toxic elements contained in the cryopreservation solution while maintaining a similar ability to produce viable fibroblasts after cryoconservation. Abstract:
PHBSdilakukan dalam rangka mencegah timbulnya penyakit, menanggulangi penyakit dan masalah-masalah kesehatan lain. Di masa pandemi Covid-19, PHBS dapat diterapkan di masyarakat untuk memutus mata rantai penyebaran virus. Penerapan PHBS yang tepat oleh setiap masyarakat disebut sebagai cara yang paling efektif dalam mencegah infeksi dan penyebaran virus sehingga dapat menekan angka kejadian Covid-19. Berdasarkan observasi di awal waktu pandemi, masyarakat di sekitar Kampung Kroyo, Karangmalang, Sragen, belum semuanya menerapkan PHBS. Oleh karena itu, dilakukan kegiatan pengabdian kepada masyarakat melalui pemberdayaan masyarakat di Kampung Kroyo, Karangmalang, Sragen dalam penerapan PHBS sebagai upaya pencegahan dan penanggulangan covid-19. Metode kegiatan yang digunakan dalam kegiatan pengabdian masyarakat ini adalah pemberdayaan masyarakat dalam penerapan PHBS dan upaya penanggulangan covid-19. Dengan adanya kegiatan pengabdian kepada masyarakat ini warga dapat memahami dan menerapkan PHBS sehingga kasus penularan covid-19 di Wilayah Kampung Kroyo, Karangmalang, Sragen dapat dicegah dan dikendalikan.
