Abstract:
Bádateľsky orientované vzdelávanie (BOV) patrí v poslednom období k najčastejšie skloňovaným pojmom v súvislosti s inováciou prírodovedného vzdelávania. Cieľom BOV je okrem konceptuálneho porozumenia aj rozvoj žiackych zručností realizovať bádanie a vyhodnocovať jeho výsledky. Prezentovaný výskum je zameraný na diagnostikovanie aktuálnej úrovne rozvoja vybraných bádateľských zručností žiakov gymnázia. Vychádzajúc z dostupných testov bol na základe klasifikácie bádateľských zručností vytvorený test bádateľských zručností. Testové úlohy určené na posúdenie úrovne zručnosti plánovať postup experimentu (identifikovať premenné a ich vzťah), transformovať výsledky do grafu, určovať vzťahy medzi premennými, identifikovať možné zdroje chýb a obhajovať výsledky a argumentovať, sú vsadené do matematického, fyzikálneho alebo informatického kontextu. Test bol zodpovedaný vzorkou 751 žiakov 1. a 2. ročníka niekoľkých gymnázií na Slovensku. Kvantitatívna analýza výsledkov testovania ukázala nízku priemernú úspešnosť žiakov v celom teste (32,5 %) ako aj v jednotlivých testových úlohách pohybujúcu sa približne v rozmedzí 8–50 %. Zo štatistického testovania vyplynulo, že medzi výsledkami žiakov 1. a 2. ročníka nebol významný rozdiel, zatiaľ čo výsledky chlapcov boli lepšie ako výsledky dievčat. Podrobná kvalitatívna analýza odhalila mnohé nedostatky, ktoré sa najvýraznejšie prejavili v oblasti analýzy a interpretácie výsledkov a žiackych argumentačných zručností.

Abstract:
Množstvo metodologických vplyvov zapríčinilo, že obsahová analýza nadobudla rôzne podoby a že jej súčasný stav je dosť komplikovaný. Cieľom tejto štúdie je opísať jej súčasné podoby opierajúc sa o zdôvodnený výber literatúry. Rôzne podoby obsahovej analýzy sú v jednotlivých kapitolkách štúdie postupne opísané podľa tzv. konštitučných komponentov. Týchto komponentov je šesť: typ analyzovaného obsahu, výskumná vzorka, hĺbka analýzy, smer postupu, druh kontroly kódovania a forma vyjadrenia výsledku výskumu. V štúdii budú podrobne charakterizované rôzne náplne týchto komponentov. Výsledkom štúdie je zistenie, aké konkrétne konfigurácie uvedených náplní existujú a v akom sú vzťahu. Celkový záver štúdie vedie k potvrdeniu, že rozdelenie obsahovej analýzy na kvantitatívnu a kvalitatívnu je nepresné, a preto nefunkčné a že lepšie je hodnotiť podobu obsahovej analýzy v konkrétnej výskumnej štúdii podľa špecifických náplní jej konštitučných komponentov.

Abstract:
This thesis examines, characterizes and quantifies how women and men are being presented on Czech and Slovak Wikipedia and further elaborates on their possible differences. The theoretical part of the thesis describes the current state of Wikipedia and media in general and draws attention to gender inequality within its editorial base, which is linked to the general trend in free culture communities, where there is also a visible inequality. Furthermore, the theoretical part of the thesis also deals with the issue of gender in general and summarizes current state of knowledge and methods of studying the issue of gender on Wikipedia. The practical part of this thesis partly replicates the study First Gender, Second Sex: Gender Bias on Wikipedia (Graells- Garrido, Lalmas and Menczer, 2015) using RStudio programme to do basic text mining and quantitative analysis of biographical texts of men and women in both languages, tracking word frequencies from selected word categories, namely Gender, Reference to the opposite sex, Family and family status and Career. Overall, we have analysed 24 510 Slovak biographical articles and 110 866 Czech biographical articles, and our findings have confirmed an imbalance and stereotyping in the presentation of women on Wikipedia in both languages.

Abstract:
Recent breakthroughs in the RNA quantification of single cells are rapidly transforming the view on biology and medicine. Flexibility and sensitivity of reverse transcription quantitative PCR (RT-qPCR) make it an ideal method for quantification of single-cell material, but its limits had not been yet fully explored. In this thesis, various factors influencing RT-qPCR performance in single-cell application have been assessed, including conditions of sample collection and processing, importance of quality control, performance of reverse transcription, preamplification and role of qPCR assays. We showed that prolonged time for single cell collection as well as repeated freeze-thaw cycles had negligible effect on RT-qPCR data quality. Direct lysis routinely applied for RNA extraction from single cells may be scaled up to 256 cells. The comprehensive comparison of 11 reverse transcriptases in low RNA input conditions identified 2 best-performing enzymes. Decrease in preamplification volume as well as poor primer design resulted in the loss of sensitivity. Finally, the established workflow has been applied to profile gene expression of astrocytes in mouse model of amyotrophic lateral sclerosis (ALS) identifying important components of ALS-induced changes to astrocyte transcriptome.
