Full textThe research for new therapeutic or diagnostic molecules targeting membrane receptors, including GPCRs, tyrosine kinase receptors and ion channels are the heart of the research invested by pharmaceutical companies. In this project we used the variable domain of llama antibody heavy chain also called single domain antibodies (sdAbs) that can bypass some limitations to the use of monoclonal antibodies or small molecules. Indeed, because of their structural features that make them particularly interesting in terms of stability, affinity and antigen recognition, sdAbs represent a very promising candidates that can be used in various fiels of application: as fluorescent probes , screening tools , or therapeutic molecules. In this manuscript are exposed the work performed on the tyrosine kinases receptor belonging to the ErbBs family and metabotropic glutamate receptors, mGluRs (GPCRs).We selected very highly specific sdAbs directed against antigen of interest, but these molecules have also shown very surprising and unexpected particularities. Indeed, most of the selected sdAbs exhibit specificity for a particular conformation of the receptor (active or inactive form). This very relevant feature opens an highly diversified application spectrum, because it allows to identify and analyze these receptors in different states of activation. Added to this, some sdAbs present an allosteric modulator activity, or even present an agonist effect. These encouraging results open up new perspectives, and these molecules represent a new approach for modulation and study of these membrane receptors

Full textThe aim of this thesis has been to study domain wall dynamics in magnetic thin films with perpendicular magnetic anisotropy embedded in a non centrosymmetric stack. In this kind of system the competition between the symmetric Heisenberg exchange and an antisymmetric exchange term, called the interfacial Dzyaloshinskii-Moriya interaction (DMI), favours non collinear magnetic textures with a fixed chirality, like chiral Néel domain walls and skyrmions. In this work we have been interested in the field and current-driven dynamics of chiral Néel walls in trilayer stacks, typically consisting of a thin Co film deposited on Pt and capped with an oxide.We have shown that the statics and dynamics of a domain wall (DW) is strongly affected by the DMI. The DMI favours Néel internal structure (rather than the Bloch structure usually found in symmetric systems) with a fixed chirality. By comparing DWs in Pt/Co/Pt (no DMI) and Pt/Co/AlOx (strong DMI), we have shown that in the presence of DMI, DWs can be moved more efficiently by a magnetic field. The stabilization of the internal DW structure by the DMI allows the precessional regime to be pushed to large magnetic fields and large velocities to be reached.Opposite to what is predicted by 1D models we show that in the presence of DMI, the DW velocity saturates after the Walker field, and that the saturation velocity is proportional to the ratio of the DMI strength and the saturation magnetization (D/Ms). The enhancement of the saturation velocity in systems with reduced Ms is shown by comparing DW dynamics in Pt/Co/GdOx and Pt/Co/Gd stacks. This also means that, knowing Ms, measuring the DW saturation velocity provides an original method to quantify the interfacial Dzyaloshinskii-Moriya interaction, as we show in this work.This method has been used to measure the DMI interaction in Pt/Co/AlOx trilayers in which the top Co interface presents a varying degree of oxidation. We show that besides the strong DMI at the Pt/Co interface, the Co/oxide interface also provides a DMI contribution of the same sign, whose strength depends on the degree of oxidation of the Co/AlOx interface. We also observe that this DMI scales with the perpendicular magnetic anisotropy, which suggest a common origin for the two effects. Finally we have shown preliminary results of field- and current-driven dynamics of DWs in a ferrimagnetic system (GdCo). While close to the compensation composition domain walls in Pt/GdCo/Ta trilayers can be moved to high velocities only by very high magnetic fields, the current driven dynamics is very efficient and depinning currents low. This effect is attributed to the 1/ Ms dependence of the spin-orbit torque acting on the DW magnetization

Full textIn the first part of this thesis, we study the magnetization reversal process of FePt nanowires with high magnetocrystalline anisotropy. When reducing the wire width below the mean dendrite width, the magnetization reversal favors a transition from the dendrite growth to the propagation of a single domain wall (DW). Further decreasing of the width towards the disorder length and/or the mean edge roughness leads to a large increase of coercivity, which finally results in a mix of DW propagation and nucleation in ultra-narrow wires. The second part focuses on the use of Magnon magnetoresistance (MMR), i.e., the magnon contribution to the resistivity, to study the magnetization reversal in nanostructures with either perpendicular (FePt) or planar magnetization (NiFe). We showed that MMR can be used in nanowires and nanomagnets, in particular to detect DW position in nanowires processed in a single layer. Finally, the dynamic of DW depinning under field and current in both FePt and NiFe systems has been studied. We observe three different modes of DW depinning, which depend on the nature of defects, or on the geometry of the constriction. Statistical analysis of the pinning time indeed shows that the depinning path can be described as simple path, serial paths or alternative paths. Additionally, the effect of DC current on all depinning mechanisms is found to be equivalent to the effect of applied field which, allow measuring the spin transfer efficiency in these systems

Full textThe P. M. Ti-6Al-4V alloy is foreseen in the cryogenic turbo-pump of Ariane V. Since local plastification are likely to occur under nominal condition, the plastic deformation and damage mechanisms at 20 K of a Ti-6Al-4V E. L. I. (Extra low interstitial) elaborated by Powder Metallurgy (PM) are studied. At liquid hydrogen temperature, half of the deformation is related to twinning. Crystallographic orientations, measured through EBSD, allow three different twinning modes to be identified. The "A" type twinning, with a {10-12} plane and a <10-1-1> direction , allows the deformation of grains with the c axis near the tensile axis. The "i" type twinning, with a {'5-61-3 '} plane and a <41-53> direction, has never been indexed in titanium. The "B" type twinning, with a {10-11} plane and a <10-1-2> direction, appears inside twins of "i" type. The damage of this alloy, strained at 20K, is a ductile damage, controlled by the cavities nucleation step. Cavities appear lately. Their growth and coalescence steps are very rapid. At a microscopic scale, the unique identified damage mechanism is the "i" type twin plane debonding, promoted by twins of "B" type. As soon as they appear after 10% of strain, large internal stresses, increasing with deformation, are generated. These internal stresses progressively reduce the applied stress needed to break the material. These results are used to build a damage and rupture map. A schematic process for cavity nucleation, based on these experimental observations, is proposed

Full textFrench engineering technicians in aeronautics have increasing contact with nationals of other countries and English has become prevalent in this industry. These changes inevitably affect the teaching of English to technicians as a second language in French Institutes of Technology (IUTs). The study focused on the didactics of the English language in the mechanical and production engineering department of the Toulouse IUT which has a bias towards aeronautical sciences and the use of English by technicians in French aerospace companies. A needs analysis was conducted in both environments using a triangulation of sources and methods based on tasks and situations. In the university context, questionnaires were sent to students and lecturers at local and national levels which enabled the current situation and the language needs of IUTs to be analyzed. The problems and difficulties faced by the technicians employed in the aerospace companies were identified using semi-structured interviews in the workplace. The results of the business surveys indicated that language anxiety could cause a mental block in the technicians’ ability to communicate in English and that the main problem was more emotional than purely linguistic. Recommendations for improving English language teaching in the Toulouse IUT were made, which included encouraging speaking, reading and writing e-mails, project-based learning (PBL) and the development of self-learning. Finally, the study concluded that the creation of a University Observatory of Languages in the Workplace would be of significant value in reducing the dichotomy between university and business which still appears to be present in the field of foreign languages

Full textIn this thesis, we aim to characterize a vehicle stable state domain, as well as vehicle state estimation, using interval methods.In the first part of this thesis, we are interested in the intelligent vehicle state estimation.The Bayesian approach is one of the most popular and used approaches of estimation. It is based on the calculated probability of the density function which is neither evident nor simple all the time, conditioned on the available measurements.Among the Bayesian approaches, we know the Kalman filter (KF) in its three forms(linear, non linear and unscented). All the Kalman filters assume unimodal Gaussian state and measurement distributions. As an alternative, the Particle Filter(PF) is a sequential Monte Carlo Bayesian estimator. Contrary to Kalman filter,PF is supposed to give more information about the posterior even when it has a multimodal shape or when the noise follows non-Gaussian distribution. However,the PF is very sensitive to the imprecision due by bias or noise, and its efficiency and accuracy depend mainly on the number of propagated particles which can easily and significantly increase as a result of this imprecision. In this part, we introduce the interval framework to deal with the problems of the non-white biased measurements and bounded errors. We use the Box Particle Filter (BPF), an estimator based simultaneously on the interval analysis and on the particle approach. We aim to estimate some immeasurable state from the vehicle dynamics using the bounded error Box Particle algorithm, like the roll angle and the lateral load transfer, which are two dynamic states of the vehicle. BPF gives a guaranteed estimation of the state vector. The box encountering the estimation is guaranteed to encounter thereal value of the estimated variable as well.In the second part of this thesis, we aim to compute a vehicle stable state domain.An algorithm, based on the set inversion principle and the constraints satisfaction,is used. Considering the longitudinal velocity and the side slip angle at the vehicle centre of gravity, we characterize the set of these two state variables that corresponds to a stable behaviour : neither roll-over nor sliding. Concerning the roll-over risk,we use the lateral transfer ratio LTR as a risk indicator. Concerning the sliding risk, we use the wheels side slip angles. All these variables are related geometrically to the longitudinal velocity and the side slip angle at the centre of gravity. Using these constraints, the set inversion principle is applied in order to define the set ofthe state variables where the two mentioned risks are avoided. The algorithm of Sivia is implemented. Knowing the vehicle trajectory, a maximal allowed velocityon every part of this trajectory is deduced

The Oseen model is a linearized version of the Navier-Stokes equations describing the flow of a viscous and incompressible fluid past a bounded body. To describe the behavior at infinity of solutions and to take into account the paraboloidal region, the so-called wake, which appears behind the body during the flow, we choose to set the problem in a functional framework which uses anisotropic weights. In a first step, we prove density results and Hardy inequalities. In a second step, we prove existence, uniqueness and regularity of solutions. Full textThis thesis aims at providing a quantitative version of the following theorem : there are only finitely many approximations with positive exponentof any subvariety of an abelian variety. This theorem was proved by Faltingsin 1991 using mostly the same tools as his proof of the Mordell-Langconjecture. A corollary is the following extension of Siegel's theorem,conjectured by Lang: any abelian variety has only finitely many integralpoints. We proceed with Vojta's method, following Rémond's work on a quantitativeversion of Mordell-Lang: the technical heart of the thesis is the proof of anexplicit version of a suitable variant of Vojta's inequality ; we thenestablish an inequality à la Mumford and an explicit bound for the number of exceptional approximations follows. However, we need an additional hypothesis to get an explicit bound when thevariety considered contains translates of a positive-dimensional abeliansubvariety. Indeed, in some cases, an explicit bound for the number of pointswould give an explicit bound for their height, which seems to be out of reach at the present time

Resumo: A leishmaniose é uma doença parasitária que foi considerada pela Organização Mundial de Saúde como uma doença de Categoria 1, pois para a mesma não existem formas de controle, diagnóstico ou terapia eficientes. Os parasitas do gênero Leishmania (família Trypanosomatidae) são agentes etiológicos da leishmaniose e possuem cromossomos lineares com extremidades teloméricas. Os telômeros são complexos nucleoproteicos essenciais para manuntenção da estabilidade do genoma e viabilidade celular. Devido à importância das proteínas teloméricas na manutenção dessas estruturas, elas têm sido considerados bons alvos para a terapia anticâncer e antiparasitária. Sendo assim, o objetivo deste trabalho foi identificar proteínas que se associam aos telômeros de Leishmania amazonensis. Três metodologias diferentes foram utilizadas para antigir este objetivo: (i) purificação de proteínas teloméricas a partir de extratos de L. amazonensis, (ii) busca no banco de dados de Leishmania por proteínas homólogas que apresentassem domínios de ligação ao DNA do tipo Myb, e (iii) seleção genética em levedura (sistema mono-híbrido). A purificação de proteínas teloméricas a partir de extratos nucleares de L. amazonensis resultou no isolamento da proteína LaRbp38. Ensaios in vitro de interação proteína-DNA e ensaios in vivo (imunoprecipitação de cromatina) comprovaram a interação de LaRbp38 com DNA telomérico, DNA rico em GT e DNA do cinetoplasto. LaRbp38 pode ser considerada um bom alvo para a terapia antiparasitária pois é uma proteína exclusiva de tripanosomatídeos e seu homólogo em T. brucei é essencial para a sobrevivência do parasita. Para dar início a experimentos de caracterização da estrutura dessa proteína, LaRbp38 foi expressa em bactérias. Como a proteína permaneceu na fração insolúvel, experimentos preliminares de renovelamento foram feitos mostrando que a proteína necessita da presença de DNA, ou de moléculas análogas, para se renovelar in vitro. As buscas no banco de dados de L. major resultaram no descobrimento de uma lista de proteínas que apresentam domínio de ligação ao DNA do tipo Myb. Uma proteína foi escolhida e sua homóloga em L. amazonensis foi denominada LaTBP1. O domínio Myb de LaTBP1 se localiza na porção central da proteína e apresenta alta similaridade de seqüência com domínios Myb encontrados em fatores de transcrição to tipo TFIIIB, proto-oncogene c-MYB e membros da família de proteínas teloméricas Rap1p. Ensaios de interação proteína-DNA e de imunoprecipitação de cromatina confirmaram que LaTBP1 interage tanto com o DNA telomérico quanto com DNAs ricos em GT. Prefências por estes dois tipos de DNAs são apresentadas pelas proteínas teloméricas Rap1, Taz1 e TEBP1, descritas em leveduras e eucariotos superiores. A utilização do sistema mono-híbrido para identificação de proteínas teloméricas de Leishmania resultou em uma lista de proteínas candidatas. A possível função telomérica das mesmas foi inferida com base na homologia de seqüência com proteínas já descritas e em testes de interação proteína-DNA in vitro utilizando-se extratos das leveduras recombinantes. Apesar desses resultados serem promissores, análises mais detalhadas são necessárias para confirmar estas interações. Concluindo, as três metodologias se mostraram eficazes para a identificação de proteínas teloméricas e a utilização das mesmas resultou na identificação de diversas proteínas teloméricas, algumas delas ainda hipotéticas
Abstract: Leishmaniasis is a parasitic disease that was classified by World Health Organization as a Category 1 disease because there are no effective control programs or therapeutics to this disease. Parasites from the Leishmania genus are the ethiologic agents of leishmaniasis and they possess linear chromosomes with telomeric ends. Telomeres are nucleoprotein specialized nucleoprotein complexes essential for maintaining chromosomal stability and cell viability. Since telomeric proteins are essential for the maintenance of telomeres, they could be considered good targets for anticancer and antiparasitic drugs. Therefore, the goal of this work was to identify Leishmania amazonensis proteins that interact with the telomeric DNA. Three methodologies were used the achive this goal: (i) purification of telomeric proteins from nuclear extracts of L. amazonensis, (ii) Leishmania genome database mining for protein containing a Myb-like domain, and (iii) use of One-hybrid system (Clontech). The search for telomeric proteins in nuclear extracts of L. amazonensis resulted in the identification of LaRbp38. EMSA and immunoprecipitation assays were used to attest LaRbp38 binding to telomeric, GT-rich and kinetoplast DNAs, both in vitro and in vivo. LaRbp38 could be considered a good drug target for antiparasitic therapy since it is exclusive of trypanosomatids and itshomologue in T. brucei is essential for parasite survival. In order to characterize structurally this protein, LaRbp38 was expressed in bacteria. The protein was present in the insoluble fraction of the bacterial lysate. Therefore, preliminary experiments of refolding were done. LaRbp38 seems to need DNA, or analog molecules, in order to correctly refold in vitro. Data-mining in the L. major genome resulted on a list of proteins bearing a Myb-like domain. One protein was chosen and its L. amazonensis homologue was termed LaTBP1. LaTBP1 Myb-like domain is centrally localized and shares sequence similarities with Myb-like domains found on transcription factors TFIIIB and c-MYB, and with the RAP1 telomeric proteins. Competition and chromatin immunoprecipitation assays confirmed the specificity of LaTBP1 for telomeric and GT-rich DNAs. This binding specifities are also found on the telomeric proteins Rap1, Taz1 and TEBP1, described in yeast and higher eukaryotes. A list of proteins with a putative telomeric function was generated after the use of the Onehybrid system. Sequence analysis (search for homologues in other organisms) and EMSA, done with protein extract of recombinant yeast, were used to infer a telomeric function for the proteins found by this methodology. Although the results are encouraging, detailed analysis are necessary to validate the interactions. In conclusion, the three methodologies were usefull for the identification of telomeric proteins. This work resulted in the identification of several telomeric candidate proteins

Full textBiobased raw materials are an interesting and promising option for the substitution of fossil resources in material design. Moreover, using green processes which limit environmental impact of the material conception can't be avoided nowadays. In this context, the L-lysine amino acid, a building block made by white biotechnologies and poorly described in material field has been evaluated. As photopolymerization is a green process in great expansion and allowing coating design, this building block has been modified into a photocurable macromonomer.L-lysine is hardly soluble in usual organic solvents, a transformation step is necessary in order to improve its processability. Thus, L-lysine polycondensation has been tackled and led to oligomers of poly-L-lysine with low molar mass and improved solubility. In addition, the structure determination has been undertaken by different analytic technics. These oligomers can thus be grafted with photocurable functional groups in mild conditions. The chosen photocurable functional groups are donor acceptor of electron. At last, the photopolymerization of L-lysine based oligomers grafted with donor acceptor functional groups has been done by UV waterborne technic which is innovative and environmentally friendly. The photopolymerization has been carried out in different conditions in order to optimize the process

Full textNous avons étudié l'expression des gènes LIM précédemment décrits chez le tournesol dans les protoplastes par RT-PCR. Nous avons détecté un transcrit pour le gène HaWLIM-1, mais pas pour les deux autres gènes HaPLIM-1 et HaPLIM-2. Des anticorps polyclonaux spécifiques de la protéine HaWLIM-1 reconnaissent en immunoblot deux polypeptides distincts, dont les masses moléculaires sont respectivement de 52 kDa et 78 kDa. Ces masses moléculaires sont nettement supérieures à la taille attendue pour la protéine HaWLIM-1. Ces résultats indiquent que la protéine n'est pas présente sous forme de monomères dans les protoplastes, mais qu'elle participe à la formation de complexes protéiques stables.
L'étude par immunocytologie de la localisation intracellulaire de la protéine HaWLIM-1 révèle que cette protéine est présente simultanément dans deux compartiments distincts : le noyau et le cytoplasme. Dans le noyau, elle s'accumule préférentiellement dans le nucléole, et pourrait jouer un rôle dans la régulation de la transcription des gènes des ARNr, ou dans l'assemblage des ribosomes. Dans le cytoplasme, différentes approches, incluant des expériences de double-marquage et de déstructuration de composants du cytosquelette, ont permis de mettre en évidence une très forte colocalisation de la protéine HaWLIM-1 avec les microtubules, ce qui suggère un rôle pour cette protéine dans l'organisation du cytosquelette.
Lors de la culture des protoplastes, le gène HaWLIM-1 s'exprime constamment, avec cependant des variations dans le niveau d'expression. Des expériences d'immunoblot utilisant les anticorps spécifiques de la protéine HaWLIM-1 indiquent que de nouveaux polypeptides, de masses moléculaires égales à 35 kDa, 42 kDa et 64 kDa, apparaissent au cours de la culture. Cette observation suggère que la protéine HaWLIM-1 possède la capacité de s'associer et de se dissocier avec de nouveaux complexes protéiques au cours du développement. La protéine HaWLIM-1 est associée aux microtubules pendant tous les stades de la division : elle est présente au niveau de la bande préprophasique à la fin de l'interphase, au niveau du fuseau mitotique pendant la mitose, et au niveau du phragmoplaste pendant la cytokinèse. Ces observations semblent indiquer que la protéine HaWLIM-1 occupe une fonction importante au niveau des microtubules.
