In that short period, the concepts and techniques of gene therapy have progressed from being entirely fanciful to experimental clinical application. A major stumbling block for gene therapy is the inefficiency of gene transfer and the transient nature of therapeutic gene expression. Attempts to deliver therapeutic genes using replication-defective adenoviruses have been hampered by a strong host immune response to the vector, leading to clearance of transduced cells and loss of transgene expression. Abstract:
Thesis (MSc)--Stellenbosch University, 2015.<br>ENGLISH ABSTRACT: Late Embryogenesis Abundant (LEA) genes have been irrefutably linked to the osmotic stress response since their initial discovery in maturing cotton seeds. They have since been reported from a multitude of other organism where their occurrence is often associated with general responses to abiotic stress. Many studies have been conducted using LEA genes in over expression strategies to improve abiotic stress resistance. Of the known classes of LEAs, the group 1 LEAs have been widely reported, in plants, to only occur in see

Abstract:
Viruses are infectious agents that must infect the cells of living organisms in order to reproduce. They have relatively simple genomes which encode few proteins but can compensate for their simplicity by hijacking components of their cellular hosts. Arenaviruses, a family of zoonotic viruses carried by rodents, encode only 4 proteins. One of these proteins, Z, is responsible for several functions during the virus life cycle including driving the formation and release of new virus particles at the plasma membrane of infected cells. Relatively little is known about how this viral protein is reg

Abstract:
The efficient processing of replication-dependent histone mRNA requires the Stem-Loop Binding Protein (SLBP). SLBP is also involved in regulating histone mRNA half-life, their nucleocytoplasmic transport, and their translation. Unlike somatic cells, where SLBP protein accumulates only in S-phase, SLBP protein is present throughout the first two embryonic cell cycles in mice. We report here that in late 2-cell mouse embryos there is a substantial, proteasome-dependent decrease in SLBP throughout the cell. Based on chromosome morphology, the degradation of SLBP protein in late 2-cell embr
