Este texto pretende ser una aproximación a la imagen que Octavio Paz tenía acerca de la cultura azteca. Así, se argumenta que el poeta mexicano tenía una visión negativa de dicha cultura, a la que consideraba como decadente. Esta visión negativa tiene su origen en la corriente denominada la «Nueva imagen del México antiguo», muy en boga durante los años 40´s y 50´s y que transformó el curso de los estudios mesoamericanos. Abstract:
Abstract Somatic point mutations in the active site of IDH (isocitrate dehydrogenase) 1 and IDH2 genes are observed in acute myeloid leukemia (AML). These mutations lead to the production and accumulation of R-2-hydroxyglutarate (2-HG) in the tumor blast cells as well as in the plasma of patients. High levels of 2-HG have been shown to inhibit alpha-ketoglutarate-dependent dioxygenases, including epigenetic regulators (i.e. histone or DNA demethylases), resulting in altered cellular differentiation. Ex vivo experiments in primary cells from AML IDH2 mutant patients have demonstrated that IDH2 mutant inhibitors are able to revert this phenotype. In order to test the biological activity of AG-221, an oral, reversible and selective inhibitor of mutated IDH2 currently in phase I trials, we developed primary human AML xenograft models. Primary blast cells from 3 AML patients with normal karyotype (n=3), NPM1 mutation (n=3) and FLT3ITD (n=1) were injected into immunodeficient NOD SCID IL2R gamma null (NSG) mice by intrafemoral injection after sub-lethal irradiation. Medullar and peripheral tumoral engraftment was monitored by flow cytometry (using species-specific antibodies) and serum 2-HG measurements. Engraftment of IDH2-R140Q human blast cells was not lethal, but these AML cells persisted over time. Twelve months after injection, IDH2-R140Q blast cell-engrafted mice were randomized into two groups, for treatment with AG-221 (30 mpk) or vehicle (0.5% methylcellulose / 0.2% Tween 80, in water) (n=10 mice; 5 mice per condition). Treatments were administered by oral gavage for 38 days twice per day (BID). Peripheral blood was collected at multiple time points for the determination of pharmacokinetics (PK) and pharmacodynamics (PD), and the assessment of the differentiation effects by AG-221 on human tumor cells. PK/PD analyses showed good plasma exposure of AG-221 and reduction of 2-HG. Furthermore, AG-221 administration also induced a burst of proliferation of human blasts followed by myeloid differentiation starting at day 20 in peripheral blood as measured by the expression of CD11b, CD14, CD15, CD24 and cell morphology. No effects on proliferation or differentiation were seen in the absence of AG-221 administration. Histological analyses of hematopoietic organs in treated animals showed a decrease of infiltrating human cells, as well as obvious morphological changes in the human cell population in AG-221-treated animals compared with vehicle-treated animals. Flow cytometry confirmed the differentiation of the human cells in the spleen and bone marrow of the AG-221-treated mice. Furthermore, cells undergoing differentiation retained the R140Q mutation, demonstrating the differentiating effect of the compound. In conclusion, AG-221 reduced serum 2-HG levels and triggered differentiation of leukemic blast cells engrafted in NSG mice. These results are consistent with what has been observed clinically in IDH2 mutant AML patients treated with AG-221 in a phase I dose escalation trial. Quant au dossier, il est consacré au débat qui entoure la féminisation de la langue française et à diverses façons de manifester la présence des femmes dans cette langue au Québec, en France et en Suisse romande.

Abstract:
Contamination from various sources has a huge impact on soil health and microbial community composition. Metal contamination of soil in mining scenarios is of concern and is not adequately addressed, particularly with respect to the microbial community. The mining industry is one of the largest contributors to heavy metal contamination of soil in South Africa, especially since the country is one of the major mining countries in the world. Platinum mining is of special importance, since the largest percentage of the world’s reserves of platinum group metals are found and mined in South Africa. Metals from mining activities become irreversibly immobilized in soil systems because they cannot be degraded and has a huge impact on soil systems. In this study, bacteria was isolated from soil samples collected from a platinum mine tailings dam outside Rustenburg. During the warm sampling season (March 2006) most isolates were found, especially in sites 3 and 4. During the colder and drier season (May 2006) there were less isolates. Most of the isolated cultures also displayed a wide temperature growth range, mostly between 24°C - 37°C. Paenibacillus lautus and Bacillus subtilus DN-10 had a growth range between 5°C - 40°C. Culturable metal tolerant bacteria were isolated, purified and identified using 16S rDNA sequences. Nine different species were found namely Paenibacillus lautus strain DS19, Paenibacillus lautus, Paenibacillus sp. C15, uncultured Paenibacillaceae, Bacillus subtilis strain DN-10, Bacillus sp. KDNB5, Bacillus cereus, Stenotrophomonas maltophilia and Alcaligenes sp. DJWH 146-2. The ability of these strains to tolerate metal concentrations were explored by determining their minimum inhibitory concentrations for a selection of metals e.g. aluminum, barium, cobalt, chromium, cadmium, copper, iron, lead, manganese, nickel and mercury. Most isolates were able to tolerate >5mM of the Al\Ni alloy and cobalt. Transmission electron microscopy was used to determine the location of metals inside bacterial cells and electron dispersive X-ray analysis was used to determine the levels of metals inside microbial cells. Bacillus subtilis DN-10 (LDK0306) showed a high MIC (>5mM) for most metals used, except Hg. This strain also had a high percentage (10.26%) of Pb detected in its cells by EDX. This was the highest percentage detected. Plasmids were extracted from the identified strains and can help gain a better understanding of metal tolerance mechanisms used by these isolates.

Depuis plusieurs années, de nombreux travaux ont étayé la fonction endocrine du cœur. L’existence de systèmes hormonaux dans le tissu cardiaque plus particulièrement celui des peptides natriurétiques, a conféré au cœur la propriété de glande endocrine en plus de sa simple fonction de pompe. Le BNP constitue le second membre de peptides natriurétiques. Il est principalement secrété par les ventricules en réponse à une surcharge de pression ou de volume. Grâce à ses propriétés diagnostiques, thérapeutiques et pronostiques, ce peptide est actuellement considéré comme le marqueur hormonal de la décompensation cardiaque. A l’échelle des cardiomyocytes, le BNP semble être impliqué dans la modulation de l’homéostasie calcique, régulant ainsi la fonction contractile du cœur. Le but de notre étude est d’évaluer sur des cardiomyocytes normaux et hypertrophiés, la concentration plasmatique de BNP, le rôle des canaux calciques ventriculaires dans sa sécrétion et le profil d’expression de ce peptide ainsi que celui de ces récepteurs (NPR-A, -B et -C); De même, nous avons étudié l’effet autocrine du BNP sur le taux de calcium intracellulaire, sur la densité du courant calcique de type L (ICaL) et sur le potentiel d’action (PA) dans des cardiomyocytes normaux. En utilisant plusieurs approches, combinant les techniques d’ELISA, de RT-PCR, d’immunohistochimie, de cytofluorimétrie laser et de patch-clamp, nous avons pu mettre en évidence que les canaux calciques transmembranaires de type L moduleraient la sécrétion du BNP, qu’il existe une surexpression des récepteurs NPR-A et NPR-C dans l’hypertrophie cardiaque, et finalement nous avons démontré que le BNP module la libération de calcium intracellulaire, modifie le PA et diminue la densité du ICaL dans les cardiomyocytes normaux. En effet, dans notre modèle d’hypertrophie, nous avons montré que la concentration de BNP dans le sang et le milieu de culture est significativement augmenté, que cette augmentation est contrebalancée par la présence d’antagonistes calciques dans le milieu de culture des cardiomyocytes normaux et hypertrophiés ; De plus, l’expression génique des récepteurs NPR-A et –C est significativement augmenté dans les cardiomyocytes hypertrophiés et elle est en corrélation avec l’augmentation de la concentration plasmatique de BNP et son degré d’expression dans le myocarde ; finalement le BNP diminue la concentration de calcium intracellulaire, augmente la durée et la phase II du PA, réduit l’amplitude et le temps d’inactivation du ICaL, , augmente le courant de fenêtre et modifie la disponibilité d’activation et d’inactivation du canal calcique de type L. En conclusion, cette étude nous a permis de mettre en évidence une relation réciproque entre le BNP et l’homéostasie calcique, qui pourrait être responsable des effets autocrines et paracrines du BNP sur la fonction cardiaque normale et dans le remodelage myocardique en réponse à un stress hémodynamique
The existence of hormonal systems in the heart tissue especially that of natriuretic peptides, has conferred to the heart its property of endocrine gland in addition to its function of pump. The BNP is the second member of natriuretic peptides, mainly secreted by the ventricles in response to pressure overload or volume. This peptide is currently regarded as the hormonal marker of cardiac decompensation. In the cardiomyocyte, the BNP appears to be involved in modulating calcium homeostasis, regulating the contractile function of the heart. The purpose of our study is to assess on normal and hypertrophied cardiomyocites, the plasma concentration of BNP, the role of ventricular calcium channels in its secretion and the expression of this peptide and that of its receptors(NPR-A,-B and C). Similarly, we studied the autocrine effect of BNP on the rate of intracellular calcium, the density of the L-type calcium current(lcal) and the action potential(AP) in normal cardiomyocytes. By using different approaches, combining the techniques of ELISA, RT-PCR, immunohistochemistry, cytofluorimetry and patch-clamp, we were able to show that L-type calcium channels modulates the secretion of BNP, that there is an over-expression of the NPR-A and NPR-C in cardiac hypertrophy, and that the BNP modulates the release of intracellular calcium, alters the AP and reduces the density of lcal in normal cardiomyocytes. In conclusion, this study has identified a reciprocal relationship between the BNP and calcium homeostasis, which could be responsible for autocrine and paracrine effects of BNP on normal cardiac function and myocardial remodelling in response to hemodynamic stress

Ce travail s'interroge sur la signification et la portée du concept de « souverain Bien » dans la philosophie pratique de Kant. Comment justifier l'affirmation kantienne selon laquelle le souverain Bien est l'objet nécessaire et a priori de la raison pratique pure ? L'objectif de la thèse est de chercher à rendre compte de ce moment téléologique qui surgit dans la « Dialectique de la raison pratique pure » et que la théorie normative exposée dans « l'Analytique » semble pourtant rendre inutile. L'aspiration au souverain Bien est nécessaire a priori parce qu'elle participe de l'inévitable et naturel élan métaphysique de la raison à l'inconditionné. On cherche ensuite à fonder la nécessité pratique du souverain Bien en montrant que si le souverain Bien ne fait pas partie des concepts fondamentaux de l'éthique, il fait cependant partie des concepts fondamentaux de l'application de l'éthique au contexte d'une volonté morale finie. Enfin, la nécessité morale de la tâche de promouvoir de souverain Bien vient ce que celle-ci est à rapporter à l'exigence immanente de la raison pratique de chercher à s'appliquer dans le monde
This study examines the role and the meaning of the concept of the « highest Good » in Kant's practical philosophy. It tries to give a justification for Kant's claim that the highest Good is the necessary and a priori object of pure practical reason, and to give an account for this teleological moment revealed into the « Dialectic of pure practical reason » even though the normative theory founded by the « Analytic » seems to make such a moment useless. At first, the highest Good appears as a necessary object because it refers to the ineluctable and natural striving of reason for the unconditioned. Then, the practical necessity of the highest Good is connected to the fact that this concept, which doesn't belong to the fundamental concepts of ethics, belongs nonetheless to the fundamental concepts of ethic's application to the situation of a finite moral volition. The aspiration to the highest Good is finally related to the moral requirement inherent to practical reason to have an application in the world of nature. For such application is a necessary and a priori duty, the practical task ordering to promote the highest Good inherits of this moral necessity

Abstract:
This research is devoted to semantic-stylistic and paradigmatic features of Nobel Prize laureates’ texts, implemented through public speech. The results of the study allowed to identify and clarify the basic pragmalinguistic principles and structural and functional features of banquet speech, as well as various speech strategies and tactics that are necessary to achieve a communicative intention. Laureates’ intention is to express a creative and philosophical worldview, as well as gratitude to the commission for the Nobel Prize awarding and to her or his colleagues for their assistance in the study.

The value of winter rape for the Central Federal district is described. The characteristics of winter rapeseed varieties approved for cultivation in the Central Federal district of Russia by the Federal Williams Research Center of Forage Production and Agroecology given. The best varieties for winter hardiness, environmental plasticity and productivity are Severyanin, Laureate, Garant, and Nord. The created varieties of winter rapeseed with a potential of 6.0–6.5 t/ha of seeds allowed to significantly expand the area of cultivation of the crop.
