Abstract:
Contamination from various sources has a huge impact on soil health and microbial community composition. Metal contamination of soil in mining scenarios is of concern and is not adequately addressed, particularly with respect to the microbial community. The mining industry is one of the largest contributors to heavy metal contamination of soil in South Africa, especially since the country is one of the major mining countries in the world. Platinum mining is of special importance, since the largest percentage of the world’s reserves of platinum group metals are found and mined in South Africa. Metals from mining activities become irreversibly immobilized in soil systems because they cannot be degraded and has a huge impact on soil systems. In this study, bacteria was isolated from soil samples collected from a platinum mine tailings dam outside Rustenburg. During the warm sampling season (March 2006) most isolates were found, especially in sites 3 and 4. During the colder and drier season (May 2006) there were less isolates. Most of the isolated cultures also displayed a wide temperature growth range, mostly between 24°C - 37°C. Paenibacillus lautus and Bacillus subtilus DN-10 had a growth range between 5°C - 40°C. Culturable metal tolerant bacteria were isolated, purified and identified using 16S rDNA sequences. Nine different species were found namely Paenibacillus lautus strain DS19, Paenibacillus lautus, Paenibacillus sp. C15, uncultured Paenibacillaceae, Bacillus subtilis strain DN-10, Bacillus sp. KDNB5, Bacillus cereus, Stenotrophomonas maltophilia and Alcaligenes sp. DJWH 146-2. The ability of these strains to tolerate metal concentrations were explored by determining their minimum inhibitory concentrations for a selection of metals e.g. aluminum, barium, cobalt, chromium, cadmium, copper, iron, lead, manganese, nickel and mercury. Most isolates were able to tolerate >5mM of the Al\Ni alloy and cobalt. Transmission electron microscopy was used to determine the location of metals inside bacterial cells and electron dispersive X-ray analysis was used to determine the levels of metals inside microbial cells. Bacillus subtilis DN-10 (LDK0306) showed a high MIC (>5mM) for most metals used, except Hg. This strain also had a high percentage (10.26%) of Pb detected in its cells by EDX. This was the highest percentage detected. Plasmids were extracted from the identified strains and can help gain a better understanding of metal tolerance mechanisms used by these isolates.

Abstract:
The existence of hormonal systems in the heart tissue especially that of natriuretic peptides, has conferred to the heart its property of endocrine gland in addition to its function of pump. The BNP is the second member of natriuretic peptides, mainly secreted by the ventricles in response to pressure overload or volume. This peptide is currently regarded as the hormonal marker of cardiac decompensation. In the cardiomyocyte, the BNP appears to be involved in modulating calcium homeostasis, regulating the contractile function of the heart. The purpose of our study is to assess on normal and hypertrophied cardiomyocites, the plasma concentration of BNP, the role of ventricular calcium channels in its secretion and the expression of this peptide and that of its receptors(NPR-A,-B and C). Similarly, we studied the autocrine effect of BNP on the rate of intracellular calcium, the density of the L-type calcium current(lcal) and the action potential(AP) in normal cardiomyocytes. By using different approaches, combining the techniques of ELISA, RT-PCR, immunohistochemistry, cytofluorimetry and patch-clamp, we were able to show that L-type calcium channels modulates the secretion of BNP, that there is an over-expression of the NPR-A and NPR-C in cardiac hypertrophy, and that the BNP modulates the release of intracellular calcium, alters the AP and reduces the density of lcal in normal cardiomyocytes. In conclusion, this study has identified a reciprocal relationship between the BNP and calcium homeostasis, which could be responsible for autocrine and paracrine effects of BNP on normal cardiac function and myocardial remodelling in response to hemodynamic stress

Abstract:
This study examines the role and the meaning of the concept of the « highest Good » in Kant's practical philosophy. It tries to give a justification for Kant's claim that the highest Good is the necessary and a priori object of pure practical reason, and to give an account for this teleological moment revealed into the « Dialectic of pure practical reason » even though the normative theory founded by the « Analytic » seems to make such a moment useless. At first, the highest Good appears as a necessary object because it refers to the ineluctable and natural striving of reason for the unconditioned. Then, the practical necessity of the highest Good is connected to the fact that this concept, which doesn't belong to the fundamental concepts of ethics, belongs nonetheless to the fundamental concepts of ethic's application to the situation of a finite moral volition. The aspiration to the highest Good is finally related to the moral requirement inherent to practical reason to have an application in the world of nature. For such application is a necessary and a priori duty, the practical task ordering to promote the highest Good inherits of this moral necessity

Abstract:
Marker Assisted Selection (MAS) is a widely studied tool for the creation of line and hybrid varieties but few authors analyzed its implementation for the creation of synthetic varieties. Synthetic varieties are selected from polycross progenies which are biparental connected populations. The objective of this thesis consisted in developping methodologies to implement MAS in the breeding scheme of perennial ryegrass, taking plant height and crown rust resistance as example traits. In the first part, based on three biparental populations connected by a common parent, the multipopulation connected analysis from MCQTL software was more powerful for the identification of favorable alleles than the detection within each population. Numerous QTL (Quantitative Trait Loci) were detected by using this method, in spite of a narrow genetic basis of the “élite” studied material. In the second part, the same multipopulation connected analysis enabled us to detect QTL directly in breeding material, while revealing the constraints by using this type of material. The use of QTL in breeding schemes raises the question of their stability across different genetic backgrounds. Thus, in the last part, it was shown that only QTL with strong effects were stable over different genetic backgrounds. As a result of this thesis, a strategy, based on QTL detection directly in the breeding material is suggested in order to implement MAS in species commercialized as synthetic varieties

Kim, Chunghee. "Laurence Sterne and an ethics of pleasure." Thesis, Birkbeck (University of London), 1997. http://ethos.bl.uk/OrderDetails.do?uin=uk.bl.ethos.267532.

Full text
Abstract:
This thesis proposes that an ethics of pleasure informs the destabilising elements of Sterne's texts, delineated through the peculiar energies and vitality in his jesting narrative voice. I demonstrate the Sternean synthesis of two problematic principles, ethics and pleasure, incompatible for the modem mind but complementary for Eighteenth century moralists. Sterne's configuration of ethical pleasure can be differentiated from those of major moral discourses of the seventeenth and eighteenth century: 1) by his distinctive ethical position which enfranchises laughter, gaiety, and mirth which his contemporaries underplay; 2) through a Rabelaisian humour which Bakhtin sees as exemplifying signs of Renaissance folk and peasant culture, so creating a distance from the progressivistic zeal of the Enlightenment; 3) in his conjunction of Fideistic scepticism with Rabelaisian grotesque, which absorbs and transfigures a sense of the plenitude and the inexplicability of nature. The first chapter examines Sterne's incorporation of a "heteroclite" perspective into orthodox latitudinarian theism in his sermons, in the specific forms of his selectiveness in adaptations or borrowings from various divine homilies. The second chapter compares his ethics of pleasure with major contemporaneous moral discourses, focusing on the production of different moral frameworks through differing perceptions of man's place in nature. The third chapter locates Sterne's formulation of text and narrative in a material presence of communion and fellow-feeling among disparate sUbjectivities. The fourth chapter investigates the ethical significance of Sterne's particular use of laughter, in which he tries to re-enact the laughing spirit of Rabelaisian grotesque with partial success, thus suggesting his comic spirit be identified as Romantic grotesque. A fmal chapter concludes with Sterne's attempt to test his ethics through interlocution and engagement with strangers in A Sentimental Journey, charting the pragmatic obstacles to a praxis of love for the concretised being.

Considering the appreciation Luiz Ruffato showed for Laurence Sterne. We intend to understand the similitaries between the authors works. Therefore, we proceed to a bibliographical analysis of the novels “The life and opinions of Tristram Shandy, gentleman” and “They were many horses” in comparison to the menippean satire and the cinema, poetry, music and drama. With the theoretical groundwork of Mikhail Bakhtin, Northrop Frye, José Guilherme Merquior and Enylton José de Sá Rego we take notice of the mennipean satire influence in the analysed novels to discover correlations and dissimilarities between them. It is necessary to point out that this literary gender brings a taste for social criticism by discussing also the relation between literature and reality. Which is further questioned by the intense use of techniques derived from other arts. Because of this usage “Tristram Shandy” and “They were many horse” are considered hybrid novels. To work with hybridization we approach the thought of Néstor Garcia Canclini. It is also important the concept of performance, because in the making of the novels there are a personalized form of mediation with the reader and a typographical style. This concept is based in Paul Zumthor’s theory and other authors like Renato Cohen and Regina Melim. In the end, we conclude that in spite of the centuries apart there is an intense dialogue between the authors and their works that demonstrate the usefulness of tradition in contemporary brazilian literature vanguard.

Abstract:
The novels of Jacques Laurent (1919-2000), a writer linked with the literary group the "Hussards", in the 1950's, portray a composite landscape of textual practices. A versatile, prolific writer, who was subversive and libertine, he explores the possibilities of narrative both under his own name and the pseudonym of Cecil Saint-Laurent. A harsh polemist, he enlists in the fight against sartrian commitment and promotes an anarchistic aesthetics, based on the singularity of the author. Jacques Laurent's novels, which represent a hazardous and changeable authority, unfold within the metamorphic dynamics of travesty. His poetics, which are hostile to normativeness, turn writing into the unpredictable and deviant experience of the perversion of narrative forms. They strive to free themselves from mimetic referentiality, whilst moving towards a specular narrative, which is an exploratory space of autoreflexive poeticism

The thesis activities are made on two different test rigs. On the first test rig, the dynamic characteristics of a hybrid aerostatic bearing are experimentally investigated in a rigid rotor configuration. The rotor is driven up to 60 krpm by an impulse turbine and is horizontally mounted on two identical aerostatic bearings fed with are pressures up to 11 bar. Dynamic loads consist either of impacts provided by a hammer or of added unbalance masses. The test rig can measure the bearing feeding pressures and flow, the rotation speed, the impact force, the displacements of the two bearings and the bearing housing accelerations. This experimental information together with the equations of motion of the rotor enables the identification of the dynamic coefficients of the bearings. A validation of these coefficients is providing by the unbalanced response of the rotor. The second part, on the second test rig, presents the results of the experimental analysis of static and dynamic characteristics of a generation 1 foil bearing. The test rig is of floating bearing type, and can reach the speed of 40 krpm. Two different casings are used for start up and for measurement of dynamic coefficients. In its first configuration, the test rig is designed to measure the start up torque. In its second configuration, a different casing is used to measure the impedances of the foil bearing. Stiffness and damping coefficients are identified from the complex impedances and enable the calculation of natural frequencies. The experimental results show that the dynamic characteristics of the tested bearing have a weak dependence on the rotation speed but vary with the excitation frequency

Gourdon, G. L. "Time and space in 'Tristram Shandy' and other eighteenth century novels : the issues of progression and continuity." Thesis, University of Sheffield, 2003. http://etheses.whiterose.ac.uk/14724/.

Full text
Abstract:
The thesis argues that the narratives of the eighteenth-century novels selected for this study demonstrate a conscious manipulation of time and space, and that the consequence of this manipulation is to provide the reader with a unique literary journey through the text. The thesis, in its analysis and comparison of these distinctive journeys, chooses to focus on the narrative techniques which facilitate or hamper progression and continuity within the texts. It particularly concentrates on the impact of these narrative techniques on the reading experience. The first chapter studies and compares texts resorting mainly to the present tense with those predominantly written in the past tense. It examines the effects of the tense used in the narration on the reader's engrossment in the fiction. The second chapter concentrates on the repercussions of the author's choice of a beginning and an ending for his story on the nature of the progression of the narrative. The third chapter is devoted to the destabilising reading journey offered by Tristram Shandy. It examines the numerous techniques which react against continuity and progression in time and in space, and the narrator's motivation behind their use. It shows how the narrative choices of Tristram Shandy place the reader face to face with his own act of reading. The fourth and final chapter is concerned with the role and the status of fictional footnotes in some eighteenth-century prose fictions. It demonstrates the fictional nature of the footnotes in Tom Jones. It argues that fictional footnotes affect the reader's progression across the text in time and in space as well as his understanding of the work of fiction, and this in a fundamental way.

The study deals with the various aspects of space and time : space and time of history as related to the major characters of the novels : space and time of narration ; space of the reader. It also deals with such technical aspects of the enunciation as focalization, narration (narrative technique) and narratee. The continuity and changes that the author's narrative technique undergoes throughout her career as a novelist are pointed out in the process before they are then picked up in the conclusive part of the study

Gray, Sophie. "Conjurer laureates : reading early modern magicians with Derrida." Thesis, University of Liverpool, 2013. http://livrepository.liverpool.ac.uk/18193/.

Full text
Abstract:
This thesis uses the philosophy of Jacques Derrida to propose a performative method of close reading, which it uses to explore the relationship between language and identity in early modern literary representations of magicians. It addresses the common assumptions we make about the authority and stability of language by exploring the tensions that arise as the magicians attempt to use performative language to realise impossible ambitions of absolute power, knowledge and self presence. The texts studied are a combination of prose and drama from 1512 to 1607. They are read in light of Derrida’s engagement with speech act theory in ‘Signature Event Context’ and ‘Limited Inc a b c . . .’ , and also other related work of his on the performative foundations of literature and the law, including ‘Before the Law’, ‘The Law of Genre’, and ‘Force of Law: The ‘Mystical Foundations’ of Authority’. The final chapter also takes in Aporias. The magicians’ trajectories are followed in three chapters. The first is on the founding violences of their authoritative identities. Working with Derrida’s accounts of the law, it begins by describing how the foundation of authority is self-authorizing and therefore performative. It then explores how performative pacts, deals and contracts with the devil are used to establish and authorize the magicians and their supernatural worlds. This metatextual authority is reflected in the magician’s own ambitions for the certainty and stability of absolute power and knowledge. Using the early modern sense of ‘perform’ as to complete, carry out or make something, the second chapter focuses upon materiality and embodiment as attempts to stabilise or fix performative utterances. Fake suicide notes and contracts written in blood are analysed to demonstrate the slip and drift equally at work in the structures of writing and signatures. In addition, Friar Bacon’s enormous brass head with its promise of ‘sound aphorisms’ is discussed as an alternative example of the misinformed urge to embody certainty. The final chapter addresses why almost all the stories of magicians, good and bad, end with their deaths. Following Derrida’s Aporias, death is represented as the ultimate opening limit, which undermines all attempts at certainty and self-presence. This leads to discussion of the other’s role in identity and, it follows, death. The magicians’ ends are compared, with particular focus on the sense of a coming event in the final scene of Doctor Faustus. The use of Derrida’s work to engage with early modern texts responds to a significant gap in the field. It offers original, contemporary insight into traditional themes of power, language and identity that are usually approached from a historicist perspective. The critical-creative close reading describes and carries out by the thesis suggests an exciting way to performatively respond to literature of the past, bearing witness to it but also transforming it into something new.

Abstract:
It has been conjectured that Fano manifolds correspond to certain Laurent polynomials under Mirror Symmetry. This correspondence predicts that the regularized quantum period of a Fano manifold coincides with the classical period of a Laurent polynomial mirror. This correspondence is not one-to-one, as many different Laurent polynomials can have the same classical period; it should become one-to-one after imposing the correct equivalence relation on Laurent polynomials. In this thesis we introduce what we believe to be the correct notion of equivalence: this is algebraic mutation of Laurent polynomials. We also consider combinatorial mutation, which is the transformation of lattice polytopes induced by algebraic mutation of Laurent polynomials supported on them. We establish the basic properties of algebraic and combinatorial mutations and give applications to algebraic geometry, most notably to the classification of Fano manifolds up to deformation. Our main focus is on the surface case, where the theory is particularly rich.

Abstract:

In 1789, lovoisier is 45. Famous as a chemist all over europe, he is an expert in finance and banking. The aim of this thesis is to determine the importance of his contribution to the history of economics. The first part des cribes the different schools of thought who influenced his mind : mercantilism with forbonnais and the ferme generale; liberalism with the physiocrats, turgot and du pont de nemours; social sciences with condorcet; political and social reform with necker. The result is a new approach of political economics, together liberal, practical, scientific and generous. The second part analyses lavoisier's activities. In charge of accounts and controls at the highest levels with the ferme generale, executive at the manufactures of tobacco and powder and salpter, landowner encouraging the improvement of agriculture, daring entrepreneur, he is at the same time concerned by public welfare and social problems. He is responsible for many far reaching proposals at orleans provincial assembly, at the government's committee on agriculture, as an organizer of ballooning, hydrogen production or the establishment of the metric system of weights and measures. He shows great talent when he is in charge of financial resources during the french revolution : discount bank, assignats, government debt, taxes, budget, statistics and accounts department, national treasury are the main missions in which he takes an active part. The importance and actuality of these accomplishments should merit to lavoisier a title of precursor of modern economists

Abstract:

Abstract:
Paul Laurence Dunbar was one of the most popular poets of his day. He was highly regarded for his black dialect poetry, which earned him the title, ?poet laureate of his race.? Dunbar?s second book of poetry, Majors and Minors, was even reviewed by the famous critic William Dean Howells. However, despite Dunbar?s popularity, he has also been widely criticized for his black dialect poetry. Many scholars and African-Americans have argued that it is an unsympathetic portrait of blackness meant to appease his paying white readership. This thesis discusses the conditions and circumstances that influenced Dunbar to write black dialect poetry. It places the poet?s life and career in the social, economic, and critical context of the mid-to-late nineteenth century. My research concludes that Dunbar?s intentions for his dialect poetry were misconstrued by William Dean Howells? racially-biased interpretation of Majors and Minors.

Cotton, Christopher Adrian. "War and the body in the work of Laurence Sterne." Thesis, University of Cambridge, 2006. http://ethos.bl.uk/OrderDetails.do?uin=uk.bl.ethos.598059.

Full text
Abstract:
The military theme epitomizes Tristram Shandy’s intolerance of systematic interpretation. Sterne recognizes the heterogeneity of war and is preoccupied with various military strategies, technologies, and paradigms, all of which have specific implications. Sterne renders the spatial and physical properties of these elements of war by displaying their effects on the body. He also manipulates narrative form to express this spatiality. In Part 1, an interplay between generic and military developments is theorized and the unique suitability of the novel for rendering modern warfare is postulated. The numerous wars mentioned in Tristram Shandy are then surveyed. Besides the provision of historical detail, the arrangement of this material, its tendency to reflect issues pertaining to Sterne’s conceptual treatment of war, is examined. Part 2 concerns Sterne’s handling of the aspects of war that are a product of rationalism. In Tristram Shandy, Sterne demonstrates how the body is often restrained and victimized in the course of attempts to assimilate it to military systems. An implicit linkage between rationalism itself and bellicosity is apparent in Walter Shandy’s theories and projects. Sterne also the matrixes the corollaries of military rationalism, in particular the development of long-range projectile warfare and the action of military discipline and mechanization. The counter-rational aspects of war are explored in Part 3. Sterne regularly depicts war as chaotic and labyrinthine. The amorphous parts of war are more congenial to the body than the rationalistic facets, admitting of its fluidity and reanimating it. Tristram’s narrative is also informed by principles of nonlinear warfare. In conclusion, the effect of the dissipation of the pronounced physical and spatial presence of war is considered. In A Sentimental Journey, Sterne presents a series of fragile veterans and suggests, through Yorick, a restitution of the virtues of chivalry and gallantry that had been largely negated by modern war.
