Abstract:

Abstract:
Abstract Somatic point mutations in the active site of IDH (isocitrate dehydrogenase) 1 and IDH2 genes are observed in acute myeloid leukemia (AML). These mutations lead to the production and accumulation of R-2-hydroxyglutarate (2-HG) in the tumor blast cells as well as in the plasma of patients. High levels of 2-HG have been shown to inhibit alpha-ketoglutarate-dependent dioxygenases, including epigenetic regulators (i.e. histone or DNA demethylases), resulting in altered cellular differentiation. Ex vivo experiments in primary cells from AML IDH2 mutant patients have demonstrated that IDH2 mutant inhibitors are able to revert this phenotype. In order to test the biological activity of AG-221, an oral, reversible and selective inhibitor of mutated IDH2 currently in phase I trials, we developed primary human AML xenograft models. Primary blast cells from 3 AML patients with normal karyotype (n=3), NPM1 mutation (n=3) and FLT3ITD (n=1) were injected into immunodeficient NOD SCID IL2R gamma null (NSG) mice by intrafemoral injection after sub-lethal irradiation. Medullar and peripheral tumoral engraftment was monitored by flow cytometry (using species-specific antibodies) and serum 2-HG measurements. Engraftment of IDH2-R140Q human blast cells was not lethal, but these AML cells persisted over time. Twelve months after injection, IDH2-R140Q blast cell-engrafted mice were randomized into two groups, for treatment with AG-221 (30 mpk) or vehicle (0.5% methylcellulose / 0.2% Tween 80, in water) (n=10 mice; 5 mice per condition). Treatments were administered by oral gavage for 38 days twice per day (BID). Peripheral blood was collected at multiple time points for the determination of pharmacokinetics (PK) and pharmacodynamics (PD), and the assessment of the differentiation effects by AG-221 on human tumor cells. PK/PD analyses showed good plasma exposure of AG-221 and reduction of 2-HG. Furthermore, AG-221 administration also induced a burst of proliferation of human blasts followed by myeloid differentiation starting at day 20 in peripheral blood as measured by the expression of CD11b, CD14, CD15, CD24 and cell morphology. No effects on proliferation or differentiation were seen in the absence of AG-221 administration. Histological analyses of hematopoietic organs in treated animals showed a decrease of infiltrating human cells, as well as obvious morphological changes in the human cell population in AG-221-treated animals compared with vehicle-treated animals. Flow cytometry confirmed the differentiation of the human cells in the spleen and bone marrow of the AG-221-treated mice. Furthermore, cells undergoing differentiation retained the R140Q mutation, demonstrating the differentiating effect of the compound. In conclusion, AG-221 reduced serum 2-HG levels and triggered differentiation of leukemic blast cells engrafted in NSG mice. These results are consistent with what has been observed clinically in IDH2 mutant AML patients treated with AG-221 in a phase I dose escalation trial. Abstract:
Abstract Point mutations in isocitrate dehydrogenase (IDH) define distinct subsets of acute myelogenous leukemia (AML). IDH is a metabolic enzyme that interconverts isocitrate and α-ketoglutarate (α-KG), but cancer-associated point mutations in IDH1 and IDH2 confer a neomorphic activity that allows reduction of α-KG to the oncometabolite R-2-hydroxyglutarate (2-HG). High levels of 2-HG have been shown to inhibit α-KG-dependent dioxygenases including histone and DNA demethylases, which play a key role in regulating the epigenetic state of cells, but the relationship between 2-HG and oncogenesis is not completely understood. Consistent with 2-HG promoting cancer via an effect on chromatin structure, patients harboring IDH mutations display a CpG island methylator phenotype (CIMP) and several studies have shown that overexpression of IDH mutant enzymes can induce histone and DNA hypermethylation as well as block cellular differentiation. In addition, mice engineered to express IDH1-R132H in hematopoietic tissue have increased early hematopoietic progenitors, splenomegaly, anemia, hypermethylated histones and altered DNA methylation patterns similar to those found in AML patients harboring IDH1/2 mutations.[i] Taken together, these data suggest that cancer-associated IDH mutations may induce a block in cellular differentiation to promote tumorigenesis. To investigate whether selective pharmacological inhibition of the mutant IDH1 enzyme could provide an effective way to lower intracellular 2-HG levels and restore normal differentiation, we treated TF-1 cells or primary human AML patient samples expressing mutant IDH1 with AG-120, an oral, selective, first-in-class, potent IDH1 mutant inhibitor currently in phase I clinical trials. Treatment with AG-120 decreased intracellular 2-HG levels, inhibited growth factor independent proliferation and restored erythropoietin (EPO)-induced differentiation in TF-1 IDH1-R132H cells. Similarly, pharmacological inhibition of mutant IDH1 enzyme with AG-120 in primary human blast cells cultured ex vivo provided an effective way to lower intracellular 2-HG levels and induced myeloid differentiation. Taken together, these data demonstrate that AG-120 is effective at lowering 2-HG levels and restoring cellular differentiation, and support further clinical development of this compound. Occupying part of a single paragraph in the published autobiography, Memoirs of a Revolutionary, the Polish-Belgian novelist, journalist, and revolutionary, Victor Serge, almost casually speaks of his escape in 1941 to Mexico from Marseille in Vichy France:Some of us whose lives were in danger eventually made our exit. The Battle of the Visas which their friends have had to wage for their sake would stand some description: a single escape would provide material for a book of Balzacian proportions, packed with unexpected incidents and dark happenings behind the scenes.

Gender as political appeal: the strategic use of gender by Michelle Bachelet (Chile) and Laurette Onkelinx (Belgium)

This paper focuses on the strategic use of “gender” made by two salient female politic figures: Michelle Bachelet in Chile and Laurette Onkelinx in Belgium. Abstract:
Ahmad Shawqi was raised in the royal palace, where his maternal grandmother – who sponsored him after the death of his mother – was a favoured maid at Khedive Ismail. Shawqi studied law in Egypt and Paris, and when he returned to Egypt, he became poet Laurette for Khedive Abbas Helmy II. Although Shawqi was brought up in the royal palace, as a poet, he felt the pulse of the Egyptian people and felt their pain and dreams. After the First World War broke out, in 1915, Shawqi was exiled to Spain where he was swept away by longing for his homeland. During his exile, the 1919 revolution erupted in Egypt, and his longing for his homeland intensified, and obsessed his heart and soul. Exile was the greatest ordeal that Shawqi went through in his life. In exile, he did not find relief except when resorting to his poetry, to which he revealed the pains of his heart. He also visited the memorials of the Muslims and their reign and civilization in Seville, Cordoba, and Granada. This resulted in Shawqi composing his lengthy poem “Arab countries and the greats of Islam”. Shawqi’s poems are considered masterpieces for their sincerity of emotion and beauty of description. Perhaps the most famous of these is The Seeniya; rhyming with the letter S, entitled “The Journey to Andalusia”, and his other longing poem, “The Nouniya; rhyming with the letter N”, in which he opposed the famous medieval Arab Andalusian poet, Ibn Zaidoun.

Abstract:
The Cosmetic Ingredient Review (CIR) Expert Panel (Panel) reviewed the safety of alkyl polyethylene glycol (PEG) sulfosuccinates, which function in cosmetics mostly as surfactants/cleansing agents. Although these ingredients may cause ocular and skin irritation, dermal penetration is unlikely because of the substantial polarity and molecular size of these ingredients. The Panel considered the negative oral carcinogenicity and reproductive and developmental toxicity data on chemically related laureths (PEG lauryl ethers) and negative repeated dose toxicity and skin sensitization data on disodium laureth sulfosuccinate supported the safety of these alkyl PEG sulfosuccinates in cosmetic products, but. The CIR Expert Panel concluded that the alkyl PEG sulfosuccinates are safe in the present practices of use and concentration when formulated to be nonirritating.

Abstract:
In December 2019, Olga Tokarczuk, the Nobel Prize laureate in literature for 2018, delivered the Nobel lecture in her native Polish. It was therefore up to her English translators, Jennifer Croft and Antonia Lloyd-Jones, to relay the laureate’s message to the wider audience. Two linguists and translators, James W. Underhill and Adam Głaz, discuss this Nobel lecture in its broader historical, political, and social context, recognizing Olga Tokarczuk’s position on topical issues, the role she plays in contemporary Poland, as well as the controversies she arouses. But Tokarczuk is predominantly a writer: her lecture is concerned with literature and it is literature. In a masterly fashion, the lauretate champions the creative power of storytelling, explores her notion of the tender narrator, and constructs intriguing analogies. She weaves nuanced semantic networks around the Polish words tęsknić/tęsknota (‘miss/missing’ or ‘long/longing for’) and jestem (‘here I am’). Underhill and Głaz discuss the meanders of the English translation of the lecture, pointing out the challenges that the translators had to face and suggesting alternative ways of coping with them. Through dialogue, they inquire into the nature of translation as an endeavour that is profoundly communicative and interpersonal. They emphasize that Olga Tokarczuk is an important voice; the role of her translators is to make this voice heard worldwide.

Abstract:
This essay uses the 1960s, Gerald Wilson’s most prolific period, as a window into his life and work as a big band jazz trumpeter, soloist, arranger, conductor, and composer. This selective snapshot of Wilson’s career inserts him more fully into jazz—and California—history, while analyzing the influence of Latin music and Mexican culture on his creations. Tracing the black-brown connections in his Alta California art demonstrates an often-overlooked aspect of Wilson’s musical legacy: the fact that he wrote, arranged, recorded, and performed Latin-tinged tunes, especially several brassy homages to Mexican bullfighters, as well as Latin jazz originals. Wilson’s singular soul jazz reveals the drive and dedication of a disciplined artist—both student and teacher—who continually honed his craft and expanded his talents as part of his educational and musical philosophy. Wilson’s California story is that of an African American migrant who moves out west, where he meets a Chicana Angelena and starts a family—in the tradition of Cali-mestizaje—then stays for the higher quality of life, for the freedom to raise his children and live as an artist, further developing and fully expressing his style. However, because he never moved to New York, Wilson remains under-researched and underappreciated by academic jazz experts. Using cultural history and cultural studies research methods, this essay makes the case that Gerald Wilson should be more widely recognized and honored for his genius, greatness, and outstanding achievements in the field of modern jazz, from San Francisco to Monterey, Hollywood, and Hermosa Beach.

Abstract:
Purpose The purpose of this paper is to evaluate a recent training programme for graduates, implemented in Italy and entitled Work Experience Laureati and Laureate, i.e. Work Experience for Graduates. The aim of the programme was to increase the career prospects of unemployed graduates in the region of Umbria. Design/methodology/approach The authors rely on administrative data and matching methods to evaluate the effectiveness of the intervention in terms of employability of participants. Findings The results show that participants are more likely to be employed and to sign an apprenticeship contract within the region boundaries. The authors also find substantial differences in employability and type of contract by gender, with men having a higher probability of finding a job (permanent contract and apprenticeship). The authors show that this may be explained by the different choices in terms of field of study, with males being more prone to enrol in scientific areas and females in the humanities. Research limitations/implications It is an intervention implemented in one Italian region. Originality/value This is one of the few studies that analyses the effectiveness of active labour market policies targeting unemployed graduates, especially in the Italian context. The authors rely on different administrative data sources that allow them to evaluate the effectiveness of the programme.

Abstract:
Sympolymnia, a new genus of myrmecomorph jumping spider belonging to the tribe Simonellini Peckham, Peckham & Wheeler, 1889, is described. It comprises five species: the type species, Sympolymnia lucasi (Taczanowski, 1871), comb. nov., Sympolymnia lauretta (Peckham & Peckham, 1892), comb. nov., Sympolymnia edwardsi (Cutler, 1985), comb. nov. and Sympolymnia shinahotasp. nov. and S. cutlerisp. nov.Sympolymnia lauretta (Peckham & Peckham, 1892) is recorded from Bolivia for the first time. Ontogenetic shifts of ant-resemblance are observed: Juveniles of S. cutlerisp. nov. and S. lauretta mimic black ants of the genus Crematogaster Lund, 1831, but those of S. shinahotasp. nov. most closely resemble Pseudomyrmex ethicus (Forel, 1911). Adults of S. cutlerisp. nov., S. lauretta and S. shinahotasp. nov. resemble the ant Camponotus sanctaefidei Dalla Torre, 1892 and orange adults of S. shinahotasp. nov. are putative mimics of Camponotus latangulus Roger, 1863.

Abstract:
Abstract Introduction CD9 is a membrane protein, member of the tetraspanin family. Recent publications have reported the role of CD9 on engraftment of hematopoietic stem cells, and on cancer stem cell potential. The expression of CD9 has been correlated to the risk of metastases and to a poor clinical outcome in various types of cancer. Surprisingly, CD9 protein is downregulated in ETV6/RUNX1 pre-B acute lymphoblastic leukemia. The purpose of our study is to investigate the effect of CD9 expression on migration and engraftment abilities of pre-B lymphocytes. Materials and Methods The CD9-positive Nalm6 and REH (ETV6/RUNX1) pre-B cells were used. By lentiviral transduction of shRNA targeting mRNA, we generated Nalm6 and REH cell lines depleted in CD9 protein. Engraftment tests were performed in vivo using Nod Scid Gamma immunodeficient mice. REH and Nalm6 cells were detected in bone marrow by CD10 and respectively CD19 or HLA-DR labelling. Ability of the different cell lines to adhere on fibronectin and to migrate through double chambers system in response to CXCL12 were measured in vitro. We also investigated the presence of membrane villosities on REH and REH shCD9 cell surface by scanning electron microscopy. Finally, F-actin polymerization after CXCL12 stimulation was measured by rhodamin-phalloidin labelling. Results In vivo engrafments tests showed that the human cells detected in bone marrow is strongly enriched in CD9 positive cells compared to the initially injected population. This result suggests that CD9 facilitates pre-B lymphobasts homing. An in vitro analysis of adhesion on fibronectin demonstrated that cellular adhesion is dependent on membrane expression of CD9. As well, the more CD9 is expressed, the higher the migration rate in response to CXCL12 chemokine is. The analysis of membrane villosities on REH cell surface revealed that cells over expressing CD9 had longer villosity than shCD9 transducted cell lines. Moreover, F-actin labelling after CXCL12 stimulation showed an increased F-actin polymerization in CD9-positive cells and the formation of actin extensions. Conclusion We provide novel evidence that CD9 is a key player of pre-B lymphoblasts engraftment, adhesion and CXCL12 dependant migration. CD9 expression is related to actin remodelling. We are now investigating a potential link between CD9 and RAC1 activation in response to CXC12. Therefore, the expression level of CD9 could impact leukemic blasts abilities to spread and be responsible of relapses.
