Aim: 4-pyridoxic acid (PDA) has been proposed as an endogenous biomarker for renal organic anion transporter 1/3 (OAT1/3) inhibition. Clinical data are needed to support the proposal. Materials & methods: A hydrophilic interaction chromatography (HILIC)–LC/MS/MS assay was developed and characterized to support clinical drug–drug interaction (DDI) studies. Results: A HILIC–LC/MS/MS assay was successfully developed. PDA was measured in two clinical DDI studies; one where no significant OAT1/3 inhibition was observed and a second where a known inhibitor of the transporter was dosed. In both c

Anthocyanin extracts of two blueberries,Vaccinium myrtillus(bilberry) andVaccinium ashei(rabbiteye blueberry), and of three other berries,Ribes nigrum(black currant),Aronia melanocarpa(chokeberry), andSambucus nigra(elderberry), were analyzed by high-performance liquid chromatography coupled with photodiode array detection and electrospray ionization - mass spectrometry (LC/PDA/ESI-MS). Both bilberry and rabbiteye blueberry contained 15 identical anthocyanins with different distribution patterns. A stability-indicating LC method was developed for quantification of linagliptin (LGT) and three synthetic impurities. The method utilizes a Thermo Scientific® RP-8 column (100 mm × 4.6 mm; 5 μm) with the PDA detector for quantitation of impurities. A mixture of 0.1% formic acid with pH 3.5 (A) and acetonitrile (B) was used as the mobile phase at a flow rate of 0.6 mL·min−1 with gradient elution. The percentage of mobile phase B increases from 30% to 70% over 5 min and decreases from 70% to 30% between 5 and 8 min. The method was validated according to International Council for Harmonization (

An in-house strategy to dereplicate colchicinoid alkaloids was recently developed by our team. It aimed at quickly identifying Colchicum constituents using LC-MS (liquid chromatography- mass spectroscopy) and LC-UV/Vis PDA (liquid chromatography-ultraviolet/ visible photodiode array) techniques. In this project, our goal was to validate the developed method through analysing the alkaloid-rich fractions of three Colchicum species that had been previously studied phytochemically using the traditional bioactivity-guided fractionation methodology. The analysed species were Colchicum tauri Siehe ex
