Full textIn the frame of the Ecosystem Approach to Fishery (EAF) management, impact of the tropical tuna purse-seine fishery on targeted and incidentally captured species should be investigated. They may include megafauna species, such as sharks, rays, cetaceans, turtles or sea birds, which often are emblematic and vulnerable species. Tropical tuna purse-seiners actively search, at the surface of the sea, for clues indicating the presence of tuna schools (e.g. birds, floating objects, whales, dolphins or whale sharks). In the eastern Atlantic and western Indian Oceans, the main two fishing modes are sets on free swimming tuna schools and schools associated to natural or artificial floating objects, thereafter called Fish Aggregating Device (FAD). However dedicated studies on fishing sets associated to whale sharks and cetaceans are still lacking. The aim of this thesis is therefore, using logbook and scientific onboard observer data, to investigate the spatio-temporal co-occurrences and/or interactions between whale sharks, cetaceans and the tuna purse-seine fishery within an ecosystem conservation perspective. This work underlines that the megafauna/ fishery co-occurrence occurs in specific spatio-temporal strata: i) Gabon to Angola (April–September), ii) the Mozambique Channel (June–September), and iii) East of Seychelles (April–September). As baleen whales and whale sharks are filter feeders, the co-occurrence with the purse-seine fishery was mostly linked to highly productive environments (i.e. using proxys including chlorophyll-a concentration). In addition fishing sets involving megafauna were relatively high before 2000 (up to 20% of the sets), but are nowadays less frequent (AC3 and 1.5% of the sets associated to baleen whales and whale sharks). The purse-seine fishery appears to have a relatively low impact on megafauna species with mortality rates of 1.4% for whale sharks and 5.6% for cetaceans. Whale shark satellite tagging also confirms these results on the longer term, but the low sample size precludes any final conclusion. While dolphins are present in fishing areas, very few interactions with the fishery was detected, which highlights the striking difference with the eastern Pacific Ocean where half the sets are associated to dolphin herds. In addition, the diversity of targeted and bycatch species captured under whale shark and baleen whale sets was also investigated. Whale sharks are principally associated to skipjack and yellowfin (of various sizes) tunas and baleen whales mostly to large yellowfin tuna. In addition, bycatch species associated to these two megafauna groups was relatively low and dominated by the silky shark, and bycatch diversity was close to the one found for free swimming tuna schools. Finally, real and/or simulated encircled megafauna conservation measures or fishing effort management measures (especially FAD moratoria including larger ones) were investigated. The first ones were found to have limited consequences on tuna catch and bycatch. Conversely FAD moratoria had limited impacts on the number of megafauna associated fishing sets, due to the fact that the main spatio-temporal strata of megafauna and FAD sets differ. However larger and longer moratoria could be beneficial for juvenile tuna and some bycatch species. Overall, this thesis has lead to increase the knowledge on megafauna/ fishery interactions, essential in the general framework of setting up an EAF in the tropical tune purse-seine fishery

Atualmente, há uma crescente demanda pela disponibilidade de produtos fitoterápicos, para atender tanto o mercado privado quanto a inserção destes produtos no Sistema Único de Saúde. Para a indústria de medicamentos envolvida com a fabricação de fitoterápicos em escala, a estratégia de produção está hoje delineada pela utilização de extratos secos padronizados de espécies medicinais, sob a forma de cápsulas. As cápsulas duras comerciais mais disseminadas são elaboradas a partir de gelatina, que apresentam uma grande limitação de sua eficiência quando o material a ser encapsulado tem características higroscópicas, pois a gelatina é reconhecida por formar ligações cruzadas em condições de alta umidade e temperatura. Este fato tem sido um fator limitante para a indústria farmacêutica, especialmente no que tange à veiculação de extratos vegetais secos, desde que muitos fitoterápicos constituem pós extremamente higroscópicos, o que em última análise compromete a estabilidade do medicamento fitoterápico. O presente estudo teve como objetivo avaliar a eficiência de cápsulas de hipromelose e de gelatina contendo extrato padronizado comercial de Sene (Cassia angustifolia Vahl). As cápsulas de ambos os tipos foram submetidas ao estresse térmico acelerado a 35 graus Celsius (ºC) e 65% de umidade relativa (UR) e a 40ºC/75%U.R. por 6 meses. As medidas comparativas de estabilidade consistiram nas avaliações de (i) perda por dessecação, (ii) desintegração, (iii) suscetibilidade à quebra, (iv) atividade de água, (v) contaminação microbiana, e (vi) análise dos derivados hidroxiantracênicos e dos senosídeos A e B. Os testes físico-químicos foram realizados de acordo com os métodos farmacopeicos em vigor, e a comparação dos conteúdos químicos foi avaliada quantitativamente por Cromatografia Líquida de Alta Eficiência (CLAE), utilizando como marcadores os senosídeos A e B comerciais, cujas presenças nos extratos foram corroboradas com a CLAE acoplada à espectrometria de massas. Os resultados obtidos na caracterização física demonstraram que as cápsulas de hipromelose apresentaram maior estabilidade em todos os parâmetros estudados, quando comparadas as de gelatina. Os resultados da análise química apontaram uma queda maior no teor de senosídeo B para as cápsulas de gelatina em relação às de hipromelose: 0,021% e 0,022%, 30ºC/65%UR e 0,02% e 0,017% 40ºC/75% UR, respectivamente. Estes resultados demonstram preliminarmente que há vantagens no uso da cápsula hipromelose em relação à de gelatina para encapsular o extrato seco de Sene, em um estudo de estabilidade acelerada, no entanto outras avaliações serão necessárias, incluindo um estudo de estabilidade completo no produto.
Currently there is a growing demand for the availability of herbal products to meet both the private market and the inclusion of these products in the Unified Health System. For the pharmaceutical industry involved in the manufacture of herbal medicines in scale, the production strategy is outlined today by the use of standardized dry extracts of medicinal species in the form of capsules. The most widespread commercial hard capsules are made from gelatin, which present a major limitation of its efficiency when the material to be encapsulated has hygroscopic characteristics, because the gelatin is known to form cross-linking under conditions of high humidity and temperature. This has been a limiting factor for the pharmaceutical industry, especially in regard to the placement of dry plant extracts, since many herbal powders are highly hygroscopic, which ultimately compromises the stability of herbal medicine. The present study aimed to evaluate the efficiency of capsules of hypromellose and gelatin containing commercially standardized Senna dry extract (Cassia angustifolia Vahl.). The capsules of both types were subjected to accelerated heat stress at 35 Celsius degrees (°C) / 65% relative humidity (RH) and at 40ºC / 75%RH for 6 months. The comparative measures of stability in the evaluations consisted of (i) loss on drying, (ii) disintegration, (iii) susceptibility to breakage, (iv) water activity, (v) microbial contamination, and (vi) analysis of hydroxyanthracene derivatives and senosídes A and B. The physicochemical tests were performed according to the pharmacopoeial methods in place, and the comparison of chemical content was assessed quantitatively by High Performance Liquid Chromatography (HPLC), using commercial senosídeos A and B as chemical markers, whose presence in the extracts were corroborated by HPLC coupled with mass spectrometry. The results of the physical characterization of the capsules showed that hypromellose capsules showed a higher stability in all these parameters, when compared with gelatin. The chemical analysis results showed a greater decrease in the concentration of senoside B for gelatin compared to hypromellose capsules: 0.021% and 0.022%, 30 ºC/65% R.H. and 0.02% and 0.017% 40 ºC/75% R.H., respectively. These preliminary results show that there are advantages in the use of hypromellose capsule in relation to the gelatin to encapsulate the Senna dry extract in an accelerated stability study, but further evaluation will be needed, including a complete study of stability in the product.

Listening to lung sounds, feeling the pulse, observing posture and gait-these are just a few of the examinations that doctors perform on their patients. A physical exam exists for every organ, from the brain to the bones of the feet, each carried out with the physician's senses. For thousands of years, humans had been solely responsible for this exam ritual, until the emergence of diagnostic equipment-CT scans, MRI scans, ultrasounds, echocardiograms, mammograms, and more. In some cases, these devices replaced the physical exam. But in areas of the world where technology is unavailable, and even in places where it exists, many physicians and healthcare professionals cannot or will not to cede their tasks to tools. Their goal: to maintain an environment in which technology and the learned senses can coexist; an environment in which the physical exam remains an integral part of medicine.

Full textThe experience of causing and controlling one s own actions and their consequences is a major aspect of our self-awareness, which has been termed sense of agency or experience of authorship. The underlying neural and cognitive mechanisms are still not well understood. In two consecutive experiments on action awareness, the electroencephalogram (EEG) of healthy participants was recorded to measure brain activity related to the perception of visual feedback. Reduced neuronal activity (sensory attenuation) in response to self- versus externally generated feedback was taken as an implicit measure for the sense of agency. The results showed that the amount of sensory attenuation was primarily determined by the mere presence of an action and external cues independent of highly precise predictions based on specific contingency rules. Explicit judgments of agency, in contrast, were mainly determined by high degrees of contingency between action and feedback, and external cues had an influence only in ambiguous contexts where contingency information was not reliable enough. A third EEG study investigated patients suffering from obsessive-compulsive disorder (OCD). OCD is characterized by lacking a sense of completion and agency for action outcomes. The results of this study revealed that OCD patients fail to suppress the consequences of their own action. Embodied signals were used less by patients for making specific predictions of the action feedback. In conclusion, the present work offers validation of sensory attenuation as an implicit measure of non-conceptual agency experience, and provides evidence that the sense of agency is based on an optimal integration of multiple embodied and external, contextual cues. Moreover, the present research reveals for the first time, to our knowledge - reduced gating of extracorporeal sensory action consequences in patients suffering from OCD, which may explain aberrant feelings of action completion and agency in these patients.

O uso de produtos naturais na medicina popular como agente terapêutico é conhecido por praticamente todas as civilizações antigas. O gênero Senna Mill, é uma importante fonte de substâncias com grande diversidade estrutural, sendo atualmente utilizada como laxantes e purgativas. Diante disso, este trabalho teve como objetivo caracterização química e atividades biológicas do extrato bruto metanólico das folhas de Senna alata L. Roxb e Senna siamea (Lam.). O perfil fitoquímico foi realizado em cromatografia de camada delgada, com diferentes fases móveis e reveladores. A extração do óleo essencial foi utilizada por hidrodestilação e analizado em cromatografia gasosa acoplada ao espectro de massa (CG/MS). A atividade larvicida do óleo essencial foi avaliada segundo o método recomendado pela Organização Mundial de Saúde, adaptado. A atividade antimicrobiana foi realizada pelo método de difusão em Agar, e identificação da concetração inibitória mínima. Os métodos de Mosman (1983) e Costa-Lotufo et al. (2002) foram utilizados para analisar a atividade hemolítica e citotóxica, respectivamente. O potencial de toxicidade aguda foi avaliado segundo a OECD 423. A metodologia de Rao et al., 1997, modificada, foi utilizado para análise da atividade laxante. A caracterização fitoquímica evidenciou os seguintes metabólitos: açucares redutores; flavonóides; antraquinonas, triterpenos, cumarinas, monoterpenos e sesquiterpenos, proantocianidinas e leucoantocianidina. Após análise em CG/MS, 03 (três) compostos foram identificados: ácido n-hexadecanóico, fitol e 9,12,15- ácido octadecatrienóico (Z,Z,Z). O óleo essencial da S. siamea não apresentou atividade larvicida. Não foi evidenciada atividade antimicrobiana do extrato bruto metanólico das folhas (EBMF) de S. siamea. Entretanto o EBMF de S. alata apresentou atividade moderada contra B. subitilis (AM04), e baixa atividade contra S. aureus (AM103) e S. aureus (AM1210). Ambos os extratos se mostraram inativos frente à atividade hemolítica e citotóxica. Na toxicidade aguda não houve óbito dos animais após administração da dose 2000mg/kg, com isso, ambos os extratos são considerados atóxicos ou baixa toxicidade. O Extrato metanólico bruto das folhas (EMBF) de S. siamea e S. alata, nas doses de 62,5mg/kg, 125mg/kg e 250mg/kg, não apresentaram atividade laxante. Estudos mais aprofundados dos extratos são necessários para garantir a segurança e eficácia terapêutica dos mesmos.
The use of natural products in folk medicine as a therapeutic agent is known by virtually all ancient civilizations. The genus Senna Mill, is an important source of substances with great structural diversity, being currently used as laxatives and purgatives. The objective of this work was to characterize the chemical and biological activities of the crude methanolic extract of the leaves of Senna alata L. Roxb and Senna siamea (Lam.). The phytochemical profile was performed in thin layer chromatography, with different mobile phases and developers. The extraction of the essential oil was used by hydrodistillation and analyzed by gas chromatography coupled to the mass spectrum (CG / MS). The larvicidal activity of the essential oil was evaluated according to the method recommended by the World Health Organization, adapted. The antimicrobial activity was performed by the agar diffusion method, and identification of minimal inhibitory concentration. The methods of Mosman (1983) and Costa-Lotufo et al. (2002) were used to analyze the hemolytic and cytotoxic activity, respectively. The acute toxicity potential was evaluated according to OECD 423. The methodology of Rao et al., 1997, modified, was used to analyze the laxative activity. The phytochemical characterization showed the following metabolites: reducing sugars; Flavonoids; Anthraquinones, triterpenes, coumarins, monoterpenes and sesquiterpenes, proanthocyanidins and leucoanthocyanidin. After analysis in CG / MS, 03 (three) compounds were identified: n-hexadecanoic acid, phytol and 9,12,15-octadecatrienoic acid (Z, Z, Z). The essential oil of S. siamea showed no larvicidal activity. There was no evidence of antimicrobial activity of the crude methanolic leaf extract (EBMF) of S. siamea. However, S. alata EBMF showed moderate activity against B. subitilis (AM04), and low activity against S. aureus (AM103) and S. aureus (AM1210). Both extracts were inactive against hemolytic and cytotoxic activity. In acute toxicity, the animals did not die after administration of the 2000mg / kg dose, with both extracts being considered nontoxic or low toxicity. The crude methanolic extract of the leaves (EMBF) of S. siamea and S. alata, at doses of 62.5mg / kg, 125mg / kg and 250mg / kg, presented no laxative activity. Further studies of extracts are necessary to ensure the therapeutic safety and efficacy of the extracts.
