Abstract:
Abstract Purpose: T-DM1, an antibody-drug conjugate, has significant antitumor activity in patients with HER2-positive metastatic breast cancer (mBC) patients who had progressed after trastuzumab-based chemotherapy. This study was conducted to investigate the clinical practice and factors related with outcomes of T-DM1 use for HER2-positive mBC patients in the nation-wide real-world setting. Method: This complete enumeration study included the patients with HER2-positive mBC who received T-DM1 as palliative therapy from August 2017 to December 2018 under the registry of Health Insurance Review & Assessment Service in Korea. Safety and outcomes of T-DM1 including overall response rate (ORR), progression-free survival (PFS), and overall survival were evaluated. Factors significant in univariate analysis were analyzed in multivariate model. Result: From the sixty institutions, a total of 824 patients were enrolled. Mean age was 58 years-old, 818 patients (99.3%) were female and 516 patients (62.6%) had relapsed after curative treatment. About 40% patients received T-DM1 as first or second line treatment, 21.5% received it as third line and 37.3% as fourth or over line. During a median follow-up of 16.8 months, the ORR was 32.8%, median PFS was 7.2 months and median OS was not reached. In multivariate analysis, clinical factors associated with the lower PFS were age (< 65 year-old, hazard ratio[HR] 1.53, 95% confidence interval[CI]: 1.214-1.919, p < 0.001), poor ECOG performance status (PS ≥ 2, HR 1.98, 95% CI: 1.493-2.626, p < 0.001), previous pertuzumab use (HR 1.40, 95% CI: 1.118-1.742, p = 0.003) and previous lapatinib use (HR 1.29, 95% CI: 1.047-1.586, p =0.017). The common grade 3-4 adverse events were thrombocytopenia (13.0%), neutropenia (2.9%), and elevation of liver enzyme (2.5%). Hypokalemia (≤ 3.0 mmol/L) and any-grade bleeding event such as epistaxis and gum-bleeding occurred in 25 (3.1%) and 94 patients (11.4%), respectively. Conclusion: This is the first, nationwide, real-world data about T-DM1 use of the HER2-positive metastatic breast cancer patients in Korea. The efficacy and toxicity profile of T-DM1 in the real-world practice were comparable with those of randomized trials. Patients’ factors and previous anti-HER2 therapy could predict the outcomes of T-DM1. Further studies to reveal the subtypes of good responders to T-DM1 and the sequence of anti-HER2 therapy are warranted. Abstract:
Abstract Purpose Capecitabine is one of a treatment option in anthracycline and taxane pre-treated metastatic breast cancer (MBC) patients. We have performed miRNA microarray of durable and poor responders to capcitabine, and compared with cell line miRNA expression profile (MCF-7, tamoxifen-resistant MCF-7 [MCF-TAM] cell line treated with 5-FU. Methods Between Jan 2006 to Jan 2021, 66 patients treated with capecitabine after anthracycline and taxane failure were enrolled. Twenty-three archival tumor tissues (11 durables and 12 poor responders, each) were collected and went through nCounter miRNA expression assay. MCF-7, tamoxifen resistant MCF-7 (MCF-TAM) cell line was treated with tamoxifen or 5-FU for proliferation assay and also went through nCounter miRNA expression assay. Results HR positive patients who received anti-estrogen treatment before capecitabine showed longer PFS (median 7.7 vs. 4.03 months, P=0.006). MCF-7 treated with tamoxifen and 5-FU showed decreased proliferation compared to 5-FU treated MCF-7. There was significant difference of miR-579-3p expression between durable and poor responders (log2FC -3.02, P=0.004). Tamoxifen-treated MCF-7 cell line showed upregulation of miR-579-3p compared to MCF-7. Tamoxifen + 5-FU treated MCF-7 cell line showed decreased expression of miR-579-3p compared to 5-FU treated MCF-7 cell line. MCF-7 cell line was treated with 5-FU and miR-579-3p. Compared to 5-FU only treated cell line, 5-FU + miR-579-3p treated cell line showed trends for decreased cell proliferation, suggesting that miR-579-3p may have chemo-sensitizing role during systemic chemotherapy. Based on miRNA database, authors hypothesized that MDM2 may be target protein of miR-579-3p and planning to perform further analysis. Conclusions Anti-estrogen treatment before capecitabine administration was associated to better survival in HR positive MBC. Anti-estrogen potentiated the effect of 5-FU in MCF-7. MiR-579-3p was upregulated in durable responders and tamoxifen treated MCF-7 cell line. Further study is planned to analyze the role of miR-579-3p and its potential target, MDM2 in breast cancer. Abstract:
Abstract Background: Prognosis of patients with HER-2 positive metastatic breast cancer (MBC) has been revolutionized with the development of monoclonal antibodies targeting HER-2 and antibody-drug conjugate, but resistance to anti-HER-2 therapy is inevitable ultimately. PI3K-AKT-mTOR pathway aberration is known to be one of the key resistance mechanisms. BR18-13(KM-10A) study is a phase 2 clinical trial evaluating efficacy and safety of Herzuma® (trastuzumab biosimilar) plus Gedatolisib (dual PI3K/mTORC inhibitor) in patients with HER-2 positive MBC who progressed after multiple lines of therapy. Methods: Patients with HER-2 positive MBC with known PIK3CA pathologic mutation or amplification whose disease progressed after more than 2 HER-2 directed therapy were enrolled in the study. They received Herzuma® (8mg/kg IV for 1st cycle loading dose, and then 6mg/kg IV every 3 weeks) plus Gedatolisib (180mg on D1, 8, 15 of every 21 days). We evaluated efficacy of the combination treatment as interim analysis. The data cutoff of this interim analysis was June 8, 2021. Results: 17 patients were enrolled and followed for a median of 6.2 months. At data cutoff, 17 patients were eligible for response assessment. All patients were confirmed to have pathologic PIK3CA aberrations: 9 kinase mutations (H1047X), 5 helical mutations (E545X), 2 other point mutations, and 1 amplification. Overall, response rate was 64.7% and disease control rate was 82.4%. Eleven patients reached partial response (PR) as their best response, three were stable disease (SD), and three had progressive disease (PD). Two patients who have reached PR remain on investigational treatment until the data cutoff point, and the longest one is on treatment for 12.0 months. The median progression-free survival assessed in data cutoff time was 5.9 months. One patient ended treatment due to CNS disease progression, but her visceral metastatic lesions were decreased with experimental treatment. No fatal adverse events related to trial medication were reported. Conclusion: In this phase 2 study, Trastuzumab biosimilar plus Gedatolisib presented 64.7% of response rate with manageable toxicity in patients with HER-2 positive MBC with PIK3CA mutation. Abstract:
These papers are originally published in the proceedings of Korea fuzzy logic and intelligent systems society (KFIS) fall conference in 1999. Eight papers are selected for this special issue. Major topics of them are fuzzy theory, neural network, inference system, intelligent controller, etc. In this issue, Seihwan Park and Hyung Lee-Kwang extend the concept of fuzzy hypergraph to type-2 fuzzy hypergraph using type-2 fuzzy sets. It has not only the same properties of hypergraphs but also the extended properties of them. It is also shown that interval valued fuzzy hypergraph is a special case of type-2 fuzzy hypergraph. Jung-Heum Yon, Yong-Taek Kim, Jae-Yong Seo and Hong-Tae Jeon design an efficient neural network called dynamic multidimensional wavelet neural network. It can perform an effective dynamic mapping with less dimensions of the input signal. These features show one way to compensate the weakness of the diagonal recurrent neural network and feedforward wavelet neural network. Yigon Kim, Yang Hee Jung and Young Chel Bae propose a new method for diagnosis of insulation aging using wavelet. It measures the partial discharge on-line from data acquisition system and analyses it using wavelet to acquire 21) patterns. They design a neuro-fuzzy model that diagnoses an electrical equipment using the data. Byung-Jae Choi, Seong-Woo Kwak and Byung Kook Kim develop an adaptive fuzzy logic controller. A sole input fuzzy variable is used to simplify the design procedure and the switching hyperplane of sliding mode control is used to improve the adaptability. Myung-Geun Chun, Keun-Chang Kwak and Jeong-Woong Ryu show an efficient fuzzy rule generation scheme for adaptive network-based fuzzy inference system using the conditional fuzzy c-means and fuzzy equalization methods. They apply this method to the truck backer-upper control and Box-Jenkins modeling problem. Daijin Kim proposes a new data classification method based on the tolerant rough set that extends the existing equivalent rough set. Twostage classification method is used. All data are classified by using the lower approximation at the first stage and then the non-classified data at the first stage are classified again by using the rough membership functions obtained from the upper approximation set. Min-Soeng Kim, Sun-Gi Hong and Ju-Jang Lee incorporate the Q-learning algorithm into the fuzzy logic controller. Modified fuzzy rule is used for the incorporation. As a result, a fuzzy logic controller is obtained that can learn through experience. Dong Hwa Kim designs a new 2-DOF PID controller and applies it to the operating data based transfer function of Gun-san Gas turbine in Korea. Abstract:
Abstract Background: Cyclin-dependent kinase 4/6 inhibitors (CDK4/6i) are the standard of care for hormone receptor positive (HR+) metastatic breast cancer (MBC). Mechanisms of intrinsic and acquired resistances have been proposed but are poorly understood in the real-world setting. The current study aims to study the association of genomic biomarkers and CDK 4/6i resistance mechanisms utilizing genomic data acquired through routine clinical practice. Methods: Tumor genomic data from patients with HR+ MBC who received FDA-approved CDK4/6i (palbociclib, ribociclib, and abemaciclib) were obtained by chart review. Progression free survival (PFS) was determined from the first day of treatment until date of disease progression. Patients were divided by number of lines of treatment (first line vs. ≥ 2 lines of therapy) and subcategorized into early progressors (< 6 months PFS), intermediate progressors (6-24 months PFS for first line and 6-9 months PFS for ≥ 2 lines of therapy), and late progressors (>24 months PFS for first line and > 9 months PFS for ≥ 2 lines of therapy). Genomic alterations from next-generation sequencing (NGS) were analyzed with PFS. PFS stratified by first vs. >2 lines of treatment were performed using a Cox proportional hazards model with hazard ratios and 95% confidence intervals reported. Results: Among 795 patients with HR+ MBC treated with CDK 4/6i between 2015 and 2020 including 673 patients from a main campus and 122 patients from community sites, a total of 131 patients had genomic data available for the analysis. The obtaining rate of genomic data was 18% (122 out of 673 patients) from the main campus and 7% (9 out of 122 patients) from community sites. Seventy-one patients were treated with CDK4/6i as the first line of MBC, and 60 were treated as second or sequential lines. 35 patients had early progression, 40 patients had intermediate progression, 44 patients had late progression, and 12 patients were indeterminate. The distribution of genomic alterations was not significantly different between each group. The genomic alterations that were associated with impaired PFS were ZNF703(hazard ratio, HR 2.1, N=13), ERBB2 (HR 1.9, N=12), MDM2 (HR 3.3, N=6), PALB2 (HR 3.8, N=5), ARFRP1 (HR 3.1, N=4), FRS1 (HR 2.9, N=4), IRS2 (HR 6.9, N=2), and JAK2 (HR 6, N=2). RNA seq analysis is currently underway. Conclusion: Tumor genomic tests are not routinely performed for patients with HR+ MBC, particularly less performed from community practices. In this single institution retrospective analysis, we identified genomic alterations, such as ZNF703, ERBB2, PALB2, and MDM2, as potential resistance mechanisms of CDK4/6i. Tumor genomics through standard clinical practice are essential to understand resistance mechanisms to CDK4/6i. Over the past two decades, there have been a series of major solar space missions, namely Yohkoh, SOHO, TRACE, and in the past 5 years, STEREO, Hinode and SDO, studying various aspects of the Sun and providing images and spectroscopic data with amazing temporal, spatial and spectral resolution. Over the same period, the type and nature of numerical models in solar physics have been completely revolutionized as a result of widespread accessibility to parallel computers. These unprecedented advances on both observational and theoretical fronts have led to significant improvements in our understanding of many aspects of the Sun's behaviour and furthered our knowledge of plasma physics processes that govern solar and other astrophysical phenomena. In this Theme Issue, the current perspectives on the main astrophysical processes that shape our Sun are reviewed. In this Introduction, they are discussed briefly to help set the scene.

Levels of some antinutrients, namely, cyanide, nitrate, and oxalate and some micronutrients including vitamin C, β‐carotene (provitamin A) and some mineral elements (Fe, Mg, Cu, Zn, Ca Na and K) were determined in fresh and processed samples of Telfairia occidentalis Hook.f. as a step towards establishing processing techniques that would reduce or eliminate unacceptable levels of antinutrients and the associated health problems. The processing methods used were cooking (5 and 10 min boiling) and sun drying. There was significant (p< 0.05) reduction in the levels of these phytotoxins. The cyanide levels in the variously processed samples were: fresh sample (170.83mg/kg), 5 min decoction (36.98mg/kg), 10 min decoction (44.87mg/kg), leaves boiled for 5 min (49.88mg/kg), and leaves boiled for 10 minutes (34.68mg/kg), and sundried leaves (94.74mg/kg). Vitamin C content was significantly (p< 0.05) reduced, the levels being 192.28mg/100g, 1.51mg/100g, 2.16mg/100g, 53.70mg/100g, 35.60mg/100g and 13.15mg/100g in fresh sample, 5 min decoction, 10 min decoction, leaves boiled for 5 min, leaves boiled for 10 min, and sundried sample respectively. β‐carotene content was elevated in the cooked vegetable, while its content decreased in sundried samples. However, cooking beyond 5 min led to significant (p < 0.05) reduction in β‐ carotene levels in the vegetable. Sun drying had no significant effect on Fe, Cu, Mg, Na and K in the vegetable. Cooking however, led to decreases in the levels of these minerals. It is concluded that moderate cooking and sun drying reduced the levels of the studied antinutrients in T. occidentalis without adversely compromising the micronutrient contents.

This triennial report (1996-1999) reviews the subject from a somewhat personal angle, mostly related to binary star formation and young binary star populations – a subject whose time had come in the early 1990s and is now in full swing.Many astronomers have searched for binary systems among main-sequence stars, and two large-scale surveys published in 1991 and 1992 have already become classics. Well before they became famous for finding extrasolar planets (see below), observing teams led by Michel Mayor (Geneva Observatory) and Geoffrey Marcy (San Francisco State Univ., now Univ. of Calif, at Berkeley) spent many years searching for low-mass stellar companions of nearby stars. The late Antoine Duquennoy and Mayor surveyed all solar-type dwarfs (spectral types F7 through G9) within 20 pc of the Sun, while Debra Fischer and Marcy studied stars with somewhat lower mass (M dwarfs) slightly nearer to the Sun.

Korean dramas are very popular with audiences from various mass media and social media. With a very interesting storyline, it is able to hypnotize Indonesian audiences to become active viewers and bring up various assumptions about Korean drama content. The Korean drama adopted in this study is entitled The World of the Married. The drama that was broadcast by Trans TV a few months ago was very successful in anesthetizing the general public and making viewers adopt drama in their daily lives. This study aims to describe the female self-image of the main character, Ji Sun Woo’s in the Korean drama entitled The World of the Married which raises the issue of divorce and shows the value of feminism as shown by Ji Sun Woo as a strong figure and a brave female character who can make her own decisions to seek justice. By using qualitative research, data was collected using reading and note-taking techniques. The Korean drama The World of the Married discusses gender inequality, work, and social group problems and indirectly harms society as a whole, such as the social inequality on Lee Tae Oh's part between men and women in terms of leading and working..

This article looks into the postcolonial Arabic narrative of Ghassan Kanafani to examine its underplayed existential and naturalistic aspects. Postcolonial texts (and their exegeses) deal with the effects of colonization/imperialism. They are expected to be political and are judged accordingly. Drawing on Kanafani’s Men in the Sun (1963), I argue that the intersection among existentialism and naturalism, on the one hand, and postcolonialism, on the other, intensifies the political relevance of the latter theory and better establishes the politically committed nature of Kanafani’s fiction of resistance. In the novella, the sun and the desert are a pivotal existential symbol juxtaposed against the despicable life led by three Palestinian refugees. The gruesome death we encounter testifies to the absurdity of life after attempts at self-definition through making choices. The gritty existence characteristic of Kanafani's work makes his representation of the lives of alienated characters more accurate and more visceral. Kanafani uses philosophical and sociological theories to augment the political nature of his protest fiction, one acting within postcolonial parameters of dispossession to object to different forms of imperialism and diaspora. Therefore, this article explores how global critical frameworks (naturalism and existentialism) enrich the localized contexts essential to any study of postcolonial literature and equally move the traditional national allegory of Kanafani to a more realist/unidealistic level of political indictment against oppression.

Since few foods are naturally rich in vitamin D, novel food products with a high content of vitamin D are needed to decrease the prevalence of vitamin D deficiency. Pork cracklings are Danish snacks with high contents of protein and fat. They are consumed mostly during wintertime when sun exposure cannot fulfil human needs for vitamin D3. Pork cracklings were produced in an industrially friendly manner from UVB LED illuminated pork rind, using a combination of sous vide (85 °C, 60 min) and roasting in the oven (200 °C, 20 min). Thermal processing resulted in a significant loss of vitamin D3 (>90%). Thus, the process was optimized by the UVB exposure of pork cracklings, i.e., after thermal processing. The produced pork cracklings had a vitamin D3 level of ~10 µg/100 g, with a possibility of tailoring its final content. Furthermore, the fat content at 15–20% was a reduction of 50% compared to marketed products in 2021. No significant difference was found in the content of vitamin D3 during 31 days of storage in the air. A consumer preference test (n = 53) indicated that >80% of participants liked the product and saw its potential as a new food source of vitamin D3.

Water differentiation of the bodies owing to 26A1 decay and their internal thermal evolution might have created conditions for water soluble organics and prebiotic compounds formation. Subsequent longtime periodic changing temperature and other physico-chemical parameters (due to spinning and moving around the Sun) in the near-surface layers of primitive asteroids have led probably to formation of more complex organic compounds of astrobiological significance.

For this reason, cardiac biomarkers are used in European 2015 and Mayo clinic 2012, two representative staging systems for AL amyloidosis. Since the NT-proBNP levels of the existing AL amyloidosis staging systems are different, we tried to find the appropriate NT-proBNP level in our 16-year AL amyloidosis patient cohort. Newly diagnoded AL amylodosis patients between August 2004 and July 2020 were included in this study (n=401). Patients who did not have laboratory results for staging had been exclude (n=86). Among them, 86 patients of stage III and 145 patients of stage IV patients (according to Mayo clinic 2012 stage) were analyzed (n=231). Of the 231 stage III, IV patients, 25, 82, 47, and 77 patients were classified as a group of NT-proBNP ≤1800, 1800 &lt; NT-proBNP ≤5000, 5000&lt; NT-proBNP ≤8000, and NT-proBNP &gt;8000 (ng/L), respectively. The characteristics and overall survival of each group were investigated through statistical analysis. Age at diagnosis (p=0.016), ECOG (p=0.046), serum creatinine(p=0.001), and Estimated glomerular filtration rate (eGFR) (p=0.003) had statistically significant differences in the groups divided by the NT-proBNP criteria. With 54.4 months of median follow up, the overall survivals analyzed by Mayo clinic 2012 were stage I: not reached, stage II: 49.6 months, stage III: 46.8 months, and stage IV: 11.9months, respectively. As a result of European 2015 analysis, stage I: not reached, stage II: 65.9 months, stage IIIa: 41.4 months, stage IIIb: 4.3 months.) In our analysis according to NT-proBNP (ng/L) in stage III and IV patients, the overall survival of NT-proBNP ≤1800 group has not yet been reached. The median OS of group 1,800&lt;NT-proBNP ≤5000, 5000&lt; NT-proBNP ≤8000, and NT-proBNP &gt;8000 were 54.8 months, 11.9 months, and 4.5 months, respectively (p &lt;0.001). The Kaplan-Meier's curve for OS had a clear difference at NT-proBNP 5000 value. On the basis of NT-proBNP, the OS of less than 5000 group was 62 months, and the OS of 5000 or more group was 5.9 months. In analysis of factors affecting the OS, statistically significant results were age at diagnosis (p = 0.018), ECOG (p = 0.002), and NT-proBNP 5000 ng/L or higher (p &lt; 0.001). The dFLC included in the Mayo clinic 2012 was found to have a statistically insignificant on the overall survival (p=0.584). Although disease stage is important in predicting the prognosis of AL amyloidosis, it was revealed that NT-proBNP is the most important factor in predicting survival prognosis. In this study we confirmed that AL amyloid patients with high NT-proBNP of &gt;5000 ng/L may have particularly poor survival rate. Objectives: To assess the potential influence of lifestyle changes during the first month of COVID-19 lockdown on body weight gains (WG) in an Algerian population cohort. Subjects and Methods: A sample survey, carried out using a self-administered questionnaire, sent on social networks to a random sample (172 participants). Anthropometric measurements were obtained as well as lifestyle factors including physical activity, diet habits, sleep, and screen time. Results: The average WG was; 1.02 ± 3.36, 1.18±2.15, and 0.95±3.79 (kg) for the total sample, men and women respectively. Δ-BMI (body mass index difference before and after one month of lockdown period) increased as following; 0.42±1.43, 0.39±0.68, and 0.43±1.66 (Kg/m²) for the total sample, men and women respectively. WG induced slightly changes from the normal BMI category to the overweight category for the total cohort (24.87±6.74 vs 25.28±7.19 kg/m²), women (25.13±7.65 vs 25.56±8.19 kg/m²) whereas no effect was reported in men (24.28±4.03 vs 24.67±4.15 kg/m²). A significant difference (p < 0.001) was revealed in men and women for sport practicing (53.33 vs 40.90 % respectively) and nighttime snacking (56.60% for men against 43.55% for women). Positive correlation between body WG and number of meals/day in men (r=0,341, p=0,012), while for women there was a positive correlation between WG change and both food intake (r=0.170, p=0.066) and nighttime snacking (r=0,228, p=0,013). Furthermore, a negative correlation between WG and sport practicing was found in women (r =-0.221, p=0.016). Conclusions: Significant WG was found during a short COVID-19 lockdown. The WG results presented herein were positively associated with certain lifestyle variables during the COVID-19 lockdown. Dynamics of crop growth and photosynthesis are two main processes that are of major importance for adaptation of plants to their environment. Two experiments were carried out during 2005 and 2006 with sun and shaded barley plants. The results showed that leaf area increased with leaf position on the main stem up to leaf position 5 for sun and leaf position 6 for shaded plants, and then declined towards flag leaves. Air temperature affected leaf appearance, mainly at the beginning of the growing season. A positive correlation between leaf expansion duration (LED) and accumulated temperature sum was measured for both variants, LED linearly increased with temperature sum. Leaf expansion rate (LER) showed a similar dependence on accumulated air temperatures in both light variants. A strong relation was found between soil temperature and LER for the first four leaves of the main stem of shaded plants in 2006. In shaded environment the higher LER was associated with lower accumulated irradiance sum when light restriction supported an increase of leaf elongation and final leaf length.

AbstractLow mass, main sequence stars like our Sun exhibit a wide variety of rotational and magnetic states. Observational and theoretical advances have led to a renewed emphasis on understanding the rotational and magnetic evolution of sun-like stars has become a pressing problem in stellar physics. We use global 3D convection and convective dynamo simulations in rotating spherical shells and with realistic stellar stratification to explore the behavior of “middle-aged” stars. We show that for stars with slightly less rotational influence than our Sun a transition occurs from solar-like (fast equator, slow poles) to anti-solar (slow equator, fast poles) differential rotation. We investigate this transition using two different treatments for the upper boundary of our simulations and we hypothesize that this transition from solar-like to anti-solar differential rotation may be responsible for observations of anomalously rapid rotation for stars older than our Sun.

The Central Park jogger case has returned to news headlines with the 2019 Netflix mini-series When They See Us, a dramatised account of the original trials. It has reignited debate over the injustices faced by the Black community in the United States, and led to lawsuits and job resignations on the part of former police investigators and prosecutors. Since the case’s inception, issues of race, media reporting, economics, and the identity of New York City have influenced the trial and its aftermath and have inspired documentaries, books, and the landmark 1990 essay “Sentimental Journeys” by Joan Didion. In this article, I argue that the creators of two of these works, by testing the boundaries of narrative, demonstrate that the case was inexorably tainted by a pervasive feeling of social precarity and racial prejudice which cost five young men several years of their lives, and offer a productive line of enquiry for acknowledging such factors and their influence, if not resolving them.

Tipping points can be defined as critical ecosystem thresholds that start self-enforced dynamics pushing systems into new stable states. Many lake ecosystems of arid Central Asia are sensitive to hydrological changes as they are located at the intersection of the influence of the dry Siberian Anticyclone and the relatively humid mid-latitude Westerlies, and their sediment records can be used to study past tipping points. We studied subfossil chironomid remains preserved in a ca. 6000-year-long sediment record from the Central Asian lake Son Kol (Central Kyrgyzstan) to reconstruct past ecosystem dynamics. Our results show abrupt transitions from a chironomid fauna dominated by macrophyte-associated, salinity-indicating taxa, to a vegetation-independent fauna, and subsequently to a macrophyte-associated, freshwater-indicating fauna. A comparison of the chironomid-based environmental reconstruction to other proxy indicators from the same record suggests a phase of increased Westerly strength starting about 4900 cal. yr BP. This increase led to enhanced precipitation and sediment fluxes into the lake, which in turn led to high turbidity levels and consequently to a macrophyte collapse causing abrupt changes in the chironomid fauna. At 4300 cal. yr BP, a weakening of the Westerlies in combination with higher lake levels led to lower turbidity and ultimately to the recovery of the macrophyte population and associated changes in the chironomid assemblage. These two sequences of events show how the occurrence of a gradual change in an external trigger (Westerlies) can trigger a cascade of within-lake processes (turbidity, macrophyte density) and may ultimately lead to an abrupt reorganization of the ecosystem (chironomid fauna), providing models for tipping points.

Abstract Introduction Addition of ovarian function suppression to conventional endocrine therapy alone in premenopausal women provides a survival benefit with moderate to high risk hormone-receptor positive breast cancer, especially who received chemotherapy Prediction of menstruation recovery after chemotherapy is important for deciding subsequent endocrine treatment and addressing fertility issues. Methods In the adding OFS after chemotherapy trial (ASTRRA), patients who resumed ovarian function up to 2 years after chemotherapy were randomized to receive either 5 years of tamoxifen or adding 2 years of OFS with tamoxifen. With these 1383 patients, we developed a model that predicts when patients recover menstruation within 3 years after chemotherapy using several variables including age, BMI, chemotherapy regimen and duration, serum E2 and FSH level. Results A total of 1017 patients data were used to develop prediction model and 366 patients data were used for external validation. In development group, 546 (53.6%) patients resumed menstruation during follow up period of 5 years. In multivariable analysis, younger age and AC based regimen without taxane were strong predictive factor for menstruation recovery. However predictive value of chemotherapy regimen was not constant over time. Therefore, we conducted another model with patients (n= 624) who did not recover menstruation within one year. In this patient group, predictive factors for menstruation recovery was age and serum E2 level at 6 months after chemotherapy. We also conducted a simplified scoring system to estimate change of recovery by using risk factors mentioned above. Conclusion Younger age is an important persisting factor predicting menstrual recovery after chemotherapy. Although chemotherapy regimen predicts shor-term menstrual recovery, over time, patient factors have more predictive influence on recovery. Recovery of serum E2 level would be important to predict subsequent menstruation recovery. It is established that the evolution of genetic programming is directly related to the development of the genetic algorithm, it is also determined that with the passage of time a significant improvement in genetic programming has occurred. Since the advent of the genetic algorithm, many modifications and software implementations have appeared. This in turn led to the implementation of the genetic algorithm toolkit in software products, namely: specialized software, applications for mathematical and analytical packages, frameworks and libraries. The article reveals the significant impact of genetic programming in the areas of: quantum computing, electrical circuit design, etc. Not only advantages, but also disadvantages are considered, attention is also paid to methods of eliminating deficiencies by improving optimization methods and applying a genetic algorithm. Results. The analysis of the main directions of the practical use of genetic programming is carried out and tasks that can be effectively solved using this toolkit are outlined. Scientific novelty. It was determined that the improvement of optimization methods and the expansion of the use of genetic algorithms, stimulates the appearance of such software products on the market, simplifies the structure of software tools, designs the interface for working with a specific commercial user community, simplifies the command language, which allows the use of genetic programming tools circle of users with different levels of training.

The structural transformations and magnetic property changes of the Nd16.2FebalCo9.9Ga0.5B7.5 (SG1, SG2) and Nd15.0FebalGa2.0B7.3 (SG3) nanocomposite alloys obtained by melt spinning in the as-quenched state and after annealing at a temperature range of 560–650 °C for 30 min were studied. The methods used were X-ray diffraction analysis, magnetic property measurements, TEM studies, X-ray fluorescence analysis and Mössbauer spectroscopy. Amorphous phase and crystalline phase Nd2Fe14B (P42/mnm) were observed in the alloy after melt spinning. The content of the amorphous phase ranged from 20% to 50% and depended on the cooling rate. Annealing of the alloys resulted in amorphous phase crystallization into Nd2Fe14B and led to the increased coercivity of the alloys up to 1840 kA/m (23.1 kOe) at 600 °C annealing for 30 min. The alloy with the maximum coercivity had a grain size of the Nd2Fe14B phase ≈50–70 nm with an Nd-rich phase between grains.

Multiple representations of this creature, such as regular animals (wolves and dogs) or even fantastic beings (dragons) have led to a mistery concerning the demon's appearance. The werewolf is the embodiment of evil, which disturbs the order of the world, and any deviation from a community can lead to one becoming a werewolf. So, werewolves, like ghouls, can appear from different sources: unnamed children, malformed children, kin-slayers, even an action that is contrary to a tradition can create a werewolf. A man will become a werewolf during an eclipse and most of the time, it's his soul that will ascend to the sky and devour the Sun and the Moon. If the bond between the soul and the body is broken during this transformation, the soul is lost forever.

The Menexenus is also known as Plato's Epitaphios or Funeral Oration. The body of the work is a fictional funeral oration, composed as an example of what should be said at a public funeral for Athenians who have fallen in war. The oration is framed by an encounter between Socrates and a certain Menexenus, an eager young man who thinks he has reached the end of education and philosophy, but who is still rather young to take an active party in the city's affairs. Nevertheless, he is anxious to follow in the tradition of his family, which (Socrates tells us) has always provided someone to look after the Athenians (τινα μν πιμελητν). Menexenus' interest in public affairs has led him to attend a meeting of the Council at which a speaker was to be chosen to compose and deliver the funeral oration at the imminent public funeral. However, no final decision was reached at the meeting, and Menexenus remarks that by the time the choice is made, the speaker will have almost to improvise his speech. Socrates gently mocks Menexenus' respect for public orators, saying that speeches about a dead person follow a predictable pattern; a speaker exaggerates all a dead person's good points and minimizes all the bad, so that one who has died appears a paragon of virtue even if he was not really good for much. Socrates claims that listening to such public orations, when not only individuals but also the state is eulogized, always makes him feel that in living in Athens he is living in the Islands of the Blessed.

Ambient light detection is important for the synchronization of the circadian clock to the external solar cycle. Light signals are sent to the suprachiasmatic nuclei (SCN), the site of the major circadian pacemaker. It has been assumed that cone photoreceptors contribute minimally to synchronization. Here, however, we find that cone photoreceptors are sufficient for mediating entrainment and transmitting photic information to the SCN, as evaluated in mice that have only cones as functional photoreceptors. Using in vivo electrophysiological recordings in the SCN of freely moving cone-only mice, we observed light responses in SCN neuronal activity in response to 60-s pulses of both ultraviolet (UV) (λmax 365 nm) and green (λmax 505 nm) light. Higher irradiances of UV light led to irradiance-dependent enhancements in SCN neuronal activity, whereas higher irradiances of green light led to a reduction in the sustained response with only the transient response remaining. Responses in SCN neuronal activity decayed with a half-max time of ∼9 min for UV light and less than a minute for green light, indicating differential input between short-wavelength–sensitive and mid-wavelength–sensitive cones for the SCN responsiveness. Furthermore, we show that UV light is more effective for photoentrainment than green light. Based on the lack of a full sustained response in cone-only mice, we confirmed that rapidly alternating light levels, rather than slowly alternating light, caused substantial phase shifts. Together, our data provide strong evidence that cone types contribute to photoentrainment and differentially affect the electrical activity levels of the SCN.

Проблемы распространения экстремизма и радикализации общества является одной из первостепенных проблем любого государства. Последствия насильственных преступлений, совершаемых по экстремистским мотивам, всегда поражают своей циничностью, безжалостностью и порождают страх у всего общества. Наши исправительные учреждения тоже столкнулись с проблемами распространения и пропаганды экстремизма среди осужденных со стороны лиц, отбывающих наказание за эти преступления. Однако, как показывает практика, эта проблема актуальна для пенитенциарных учреждений государств всего мира. Поэтому изучение опыта одной из европейских стран представляется интересным с точки зрения заимствования наиболее эффективных профилактических мероприятий при борьбе с экстремизмом. В статье автором анализируются итоги исследования, проведенного в Великобритании, целью которого было изучение масштабов и характера радикализации осужденных в трех тюрьмах строгого режима Англии и Уэльса. В ходе данного исследования был составлен криминологический портрет осужденного, склонного к радикализации, выявлены основные причины распространения экстремизма в тюрьмах, среди которых особенно выделяются проблемы, связанные с сокращением численности сотрудников, увеличением числа вакантных должностей. Кроме того, отсутствие опыта работы именно с осужденными-мусульманами у персонала тюрем, связанного с непониманием ими основ мусульманской религии, ее ценностей, а также отсутствие профессионально подготовленных капелланов, привели к резкому росту экстремизма. Автором изучены все мероприятия, проведенные на уровне уголовно-исполнительной политики государства с целью профилактики экстремизма в тюрьмах. К таким относится создание в 2016 г. нового государственного управления по вопросам безопасности, порядка и борьбы с терроризмом, а также мероприятия, направленные на обучение персонала, координацию деятельности оперативных служб, выявление наиболее «уязвимых» лиц для радикализации, решение кадровых вопросов с подбором персонала. В результате это привело к сокращению роста распространения экстремизма в тюрьмах Великобритании. The problems of the spread of extremism and the radicalization of society is one of the primary problems of any state. The consequences of violent crimes committed for extremist motives are always striking in their cynicism, ruthlessness and generate fear in the whole society. Our correctional institutions have also faced problems with the spread and propaganda of extremism among convicts, on the part of persons serving sentences for these crimes. But as practice shows, absolutely all penitentiary institutions of the world have faced this. Therefore, studying the experience of one of the European countries seems interesting, from the point of view of borrowing positive experience, the most effective preventive measures in the fight against extremism. The author analyzes the results of a study conducted in Great Britain, which sought to examine the extent and nature of radicalization of convicts in three maximum security prisons in England and Wales. In the course of this study, a criminological portrait of a convict prone to radicalization was compiled, the main causes of the spread of extremism in prisons were identified, among which the problems associated with a reduction in the number of employees and an increase in the number of vacant positions are particularly highlighted. In addition, the lack of experience of working with Muslim convicts among prison staff, due to their lack of understanding of the basics of the Muslim religion, its values, as well as the lack of professionally trained chaplains, led to a sharp increase in extremism. The author has studied all the measures carried out at the level of the state's penal policy in order to prevent extremism in prisons. These include the creation in 2016 of a new State Department for security, order and the fight against terrorism, as well as measures aimed at training personnel, coordinating the activities of operational services, identifying the most "vulnerable" persons for radicalization, and resolving personnel issues with recruitment. As a result, this has led to a reduction in the growth of extremism in UK prisons.

В статье представлено авторское видение наиболее значимых парадоксов современной уголовной политики и юридической техники в сфере противодействия преступлениям, посягающим на половую неприкосновенность и половую свободу личности. Фактическое уничтожение сформированных ранее моральных и культурных ценностей привело к существенному снижению уровня нравственности в современном обществе. Это подтверждается ростом посягательств на выделенные нами объекты уголовно-правовой охраны. Реакция законодателя на происходящее повлекла за собой существенное, концептуальное видоизменение уголовного законодательства в этой области. Кардинально изменена уголовно-правовая концепция, ранее сформированная как в восемнадцатой, так и в двадцать пятой главах Уголовного кодекса России, что породило возникновение ряда существенных парадоксов во взятом для исследования секторе правового регулирования. К основным из них можно отнести коллизионность примечания к ст. 131 УК РФ, пробельность положений ст. 134 УК РФ в части ненаказуемости иных действий сексуальной направленности, проблемы в формировании санкционной политики, а также вопросы конкуренции смежных деяний. В статье не только предпринята попытка их демонстрации, но и представлены пути разрешения. The article presents the author's vision of the most significant paradoxes of modern criminal policy and legal technique in the field of combating crimes that infringe on sexual integrity and sexual freedom of the individual. The actual destruction of the previously formed moral and cultural values, entailed a significant decrease in the level of morality among the population of Russia. This is confirmed both by a significant increase in encroachments on the objects of criminal law protection that we have identified, and on public morality. The legislator's reaction to what is happening has led to a significant, conceptual modification of the criminal legislation in this area. The criminal law concept, previously formed both in the eighteenth and twenty-fifth chapters of the Criminal Code of Russia, has been radically changed, which has given rise to a number of significant paradoxes in the area taken for research. The main ones include the conflict of laws of the note to Art. 131 of the Criminal Code of the Russian Federation, the blankness of the provisions of Art. 134 of the Criminal Code of the Russian Federation with regard to the impunity of other sexual acts, problems in the formation of sanctions policy, as well as issues of competition of related acts. The article not only attempts to demonstrate them, but also ways to resolve them.

With the goal of increasing energy efficiency and passenger comfort in an automobile cabin, we developed a regional air-conditioning design to control cold air in specific regions, and an air management strategy that can keep air circulation when the car engine cuts out. According to computational simulations, an air velocity of 2 m/s was adopted, which could form an independent flow field space in the cabin with a temperature gap of 7 °C according to the user’s needs. The designed regional air-conditioning and circulation system could create independent flow field spaces with temperature differences. Inlet air volume demand was also reduced by 60% and blower power by 53 W. In addition, the cabin ventilation system led air exchange rate reached 89% per hour. In 20 min of exposure under sun, the system could lower the cabin temperature by 12.3 °C.

The Neo-Pagan Dimension of Modern Russian Philosophy: Corporality vs RationalityThe article analyses the essence of and reasons for the “neo-pagan turn” of Russian culture and philosophy of the 19th–21st centuries to the theme of “corporality”, which is presented in a wide range of approaches, from Russian Romanticism to modern Russian projects of “new metaphysics” and “progressive archaism”. The author exemplifies the actualization of the corporality theme as the renaissance of neo-paganism in philosophical discourse with phenomena of the Silver Age, Russian philosophy of the first part of the 20th century (represented by V. Rozanov’s, N. Berdyaev’s and L. Shestov’s concepts), Soviet natural philosophy prose of the 1960s–1980s (Ch. Aitmatov, V. Rasputin, V. Astafayev), and modern Russian political philosophy (the National-Bolshevik project led by Aleksandr Dugin). It is concluded that neo-paganism is an invariant cultural phenomenon, an attempt to solve problems of modern society and modern man. Therefore, it functions as a new ideological strategy, new philosophy and new religion.

Данная ситуация сложилась в результате современной государственной политики в сфере противодействия преступности, а именно гуманизации наказания, что привело к изменению количественного и качественного состава осужденных, отбывающих наказание в местах лишения свободы, и осложнило оперативную обстановку. В статье отмечается, что несмотря на незначительное количество действий, дезорганизующих работу исправительного учреждения, в структуре пенитенциарной преступности, данные деликты имеют повышенную общественную опасность, так как совершаются в период отбывания наказания, и высокую латентность в силу как объекта (основного, дополнительного и факультативного), так и способа посягательства. Признавая криминологические данные основой для выработки мер противодействия преступности, в том числе пенитенциарной, авторы исследуют показатели одного из видов преступлений, совершаемых в исправительных колониях, - дезорганизация деятельности учреждений, обеспечивающих изоляцию от общества, а также личность преступника. Проведенное исследование позволило дать криминологическую характеристику преступлений, направленных на дезорганизацию деятельности учреждений, обеспечивающих изоляцию от общества, в частности, исправительных учреждений, а также определить особенности личности осужденных, их совершающих. Результаты исследования могут быть использованы при разработке теории преступного поведения и личности преступника, в частности, пенитенциарного. Currently, the creation of safe conditions for the execution and serving of criminal sentences in the form of deprivation of liberty, strengthening the protection of prison staff from possible threats and dangerous attacks is one of the urgent tasks facing the penal system. This situation has developed as a result of modern state policy in the field of combating crime, namely, the humanization of punishment, which has led to a change in the quantitative and qualitative composition of convicts serving sentences in places of deprivation of liberty, and has complicated the operational situation. The article notes that despite the small number of actions that disorganize the work of a correctional institution, in the structure of penitentiary crime, these torts have an increased public danger, since they are committed during the period of serving a sentence, and high latency due to both the object (both the main and optional) and the method of encroachment. Recognizing criminological data as the basis for developing measures to counteract crime, including penitentiary, the authors study indicators of one of the types of crimes committed in correctional colonies - disorganization of institutions that provide isolation from society, as well as the identity of the criminal. The research made it possible to give criminological characteristics of crimes aimed at disorganizing the activities of institutions that provide isolation from society, in particular correctional institutions, as well as to determine the personality of the convicted person who commits them.The results of the study can be used in the development of a General theory of criminal behavior and the personality of the criminal, and the penitentiary in particular.

Public education is a main target of the assault on the public sector; recently corporate education reformers have expanded their agenda to include the open-access community colleges, which enroll 40 percent of U.S. college students. City College of San Francisco was threatened with closure for resisting this policy agenda. The faculty union, students, and community groups led by people of color waged and won a five-year battle to save it. Although not unscathed, today the college is open, accredited, and free. In the continuing war on working-class community institutions, this struggle offers valuable lessons for coalitions of labor and social movements.

To maximize the efficiency of the system it is necessary to track the path of sun in order to keep the panel perpendicular to the sun. This paper proposes the design and construction of a microcontroller-based solar panel tracking system. The fuzzy controller aims at maximizing the efficiency of PV panel by focusing the sunlight to incident perpendicularly to the panel. The system consists of a PV panel which can be operated with the help of DC motor, four LED sensors placed in different positions and a fuzzy controller which takes the input from sensors and gives output speed to motor. A prototype is fabricated to test the results and compared with the simulation results. The results show the improved performance by using a tracking system

Abstract Background: Hormone-receptor (HR) positive breast cancer is the main subtype of breast cancer. Although overall survival of HR-positive metastatic breast cancer (MBC) patients has improved by various therapies including endocrine therapies, CDK4/6 inhibitors, and cytotoxic chemotherapy, it is still considered incurable. Immune checkpoint inhibitors have rarely been clinically tested in HR-positive breast cancer, despite proving anti-cancer activity in early and metastatic triple-negative breast cancer in various trials. We evaluated efficacy and safety of combined durvalumab and tremelimumab in the HR-positive MBC, which was enriched with high tumor mutational burden (TMB). Methods: HR-positive MBC patients who received prior 1 or more lines of therapy in metastatic setting were prescreened with whole exome sequencing (WES) using metastatic or recurred tumor biopsies. Criterion of high TMB was defined as upper 30%. In the beginning, the criterion of high TMB was 2.1 mutations per Mb, based on the retrospective WES database in Yonsei Cancer Center and this criterion was recalculated every 30 cases. Patients who met upper 30% of TMB were treated with combined durvalumab (1500mg every 4 weeks upto 13 doses) and tremelimumab (75mg every 4 weeks upto 4 doses). Response was evaluated every 2 cycles using RECIST 1.1 and toxicity was evaluated using NCI-CTCAE 4.03. Tumor-infiltrating lymphocyte (TIL) and PD-L1 expression were also analyzed to investigate a correlation with TMB. Results: Biopsies of recurrent or metastatic tumors were taken from a total of 119 patients for WES assay. A median turn-around-time of TMB data was 30.0 days (range, 16~67). Of these 119 patients, a median number of nonsynonymous mutations was 2.0 per Mb (range, 0~21.7) with upper 30% criterion of 3.1. High TMB showed a trend toward old age (P=0.074) and single positivity of estrogen receptor (ER) or progesterone receptor (PR) compared to positivity of both ER and PR (P=0.055). TMB was positively correlated with TILs (r=0.289, P=0.005). Thirty patients with high TMB received study treatment with a median 2 cycles (range, 1~13). A median prior lines of therapies in metastatic setting was 4 (range, 1~9). The objective response and clinical benefit rates were 6.3% (2 PRs of 30) and 20% (2 PRs plus 4 SDs of 30). There was one treatment-related mortality due to pneumonitis. Immune-related adverse events included endocrinopathy (n=3; hypothyroidism in 2, hyperthyroidism in 1), enteritis (n=2), skin rash (n=2), pneumonitis (n=1), and so on. Biomarker analyses are underway. Conclusions: WES-based TMB using metastatic tumor biopsy was a feasible platform to prescreen HR-positive MBC patients. Abstract:
In this work, a detailed microstructural investigation of as-melt-spun and heat-treated Fe67Co20B13 ribbons was performed. The as-melt-spun ribbon was predominantly amorphous at room temperature. Subsequent heating demonstrated an amorphous to crystalline α-(Fe,Co) phase transition at 403 °C. In situ transmission electron microscopy observations, carried out at the temperature range of 25–500 °C and with the heating rate of 200 °C/min, showed that the first crystallized nuclei appeared at a temperature close to 370 °C. With a further increase of temperature, the volume of α-(Fe,Co) crystallites considerably increased. Moreover, the results showed that a heating rate of 200 °C/min provides for a fine and homogenous microstructure with the α-(Fe,Co) crystallites size three times smaller than when the ribbon is heated at 20 °C/min. The next step of this research concerned the influence of both the annealing time and temperature on the microstructure and coercivity of the ribbons. It was shown that annealing at 485 °C for a shorter time (2 s) led to materials with homogenous distribution of α-(Fe,Co) crystallites with a mean size of 30 nm dispersed in the residual amorphous matrix. This was reflected in the coercivity (20.5 A/m), which significantly depended on the volume fraction of crystallites, their size, and distribution.

OBJECT Atlantooccipital dislocation (AOD) in adults cannot be diagnosed with adequate specificity and sensitivity using only CT or plain radiography, and the spine literature offers no guidelines. In children, the most sensitive and specific radiographic measurement for the diagnosis of AOD is the CT-based occipital condyle–C1 interval (CCI). The goal of the current study was to identify the normal CCI in healthy adults and compare it with the CCI in adults with AOD to establish a highly sensitive and specific cutoff value for the neuroimaging diagnosis of AOD. METHODS A total of 81 patients, 59 without AOD and 22 with AOD, were included in this study. Measurements obtained from thin-slice CT scans of the craniovertebral joint to assess atlantooccipital dislocation included the CCI, condylar sum, the Wholey and Harris intervals, Powers and Sun ratios, Wackenheim line, and Lee X-lines. RESULTS The group of patients without AOD included 30 men (50.8%) and 29 women (49.2%) with a mean age of 42.4 ± 16 years (range 19–87 years). The group of patients with AOD included 10 men (45.5%) and 12 women (54.5%) with a mean age of 38.2 ± 9.7 years (range 20–56 years). Interrater reliabilities within a 95% CI were all greater than 0.98 for CCI measurements. A total of 1296 measurements of the CCI were made in 81 patients. The mean CCI for non-AOD patients was 0.89 ± 0.12 mm, the single largest CCI measurement was 1.4 mm, and the largest mean for either right or left CCI was 1.2 mm. The mean condylar sum was 1.8 ± 0.2 mm, and the largest condylar sum value was 2.2 mm. Linear regression with age predicted an increase in CCI of 0.001 mm/year (p < 0.05). The mean CCI in AOD patients was 3.35 ± 0.18 mm (range 1.5 mm–6.4 mm). The shortest single CCI measurements in the AOD patients were 1.1 mm and 1.2 mm. The mean condylar sum for all 22 AOD patients was 6.7 ± 2.7 mm and the shortest condylar sums were 3.0 mm. Cutoff values for AOD were set at 1.5 mm for the CCI and 3.0 mm for the condylar sum, both with a sensitivity of 1 and false-negative rate of 0. Sensitivity for the Powers, Wholey, Harris, Sun, Wackenheim, and Lee criteria were determined to be 0.55, 0.46, 0.27, 0.23, 0.41, and 0.41, respectively. CONCLUSIONS The CCI is shorter in adult patients as opposed to the pediatric population. The revised CCI (1.5 mm) and condylar sum (3.0 mm) cutoff values have the highest sensitivity and specificity for the diagnosis of AOD in the adult population.

Systematic studies have been carried out about the effects of the thermal treatment parameters on melt-spun materials quenched at different cooling rates and based on the 2:14:1 hard magnetic phase. Samples of nominal compositions Dy3Nd10.2Fe79.6B6Si1.2 and Pr3Nd10.2Fe79.6B6Si1.2 were annealed at temperatures above that of crystallization of the amorphous phases present upon quenching, for times ranging from 1 to 30 min and by using different heating rates up to the annealing temperature. It is concluded that best hysteretic properties can be achieved in samples quenched at intermediate cooling rates by means of short-time thermal treatments performed by using high heating rates up to the treatment temperature. Low heating rates and long time anneals led to the deterioration of the hard magnetic behavior, due to the segregation of soft crystalline phases.

ABSTRACT The WalRK two-component system plays important roles in maintaining cell wall homeostasis and responding to antibiotic stress in low-GC Gram-positive bacteria. In the major human pathogen, Streptococcus pneumoniae, phosphorylated WalR Spn (VicR) response regulator positively controls the transcription of genes encoding the essential PcsB division protein and surface virulence factors. WalR Spn is phosphorylated by the WalK Spn (VicK) histidine kinase. Little is known about the signals sensed by WalK histidine kinases. To gain information about WalK Spn signal transduction, we performed a kinetic characterization of the WalRK Spn autophosphorylation, phosphoryltransferase, and phosphatase reactions. We were unable to purify soluble full-length WalK Spn . Consequently, these analyses were performed using two truncated versions of WalK Spn lacking its single transmembrane domain. The longer version (Δ35 amino acids) contained most of the HAMP domain and the PAS, DHp, and CA domains, whereas the shorter version (Δ195 amino acids) contained only the DHp and CA domains. The autophosphorylation kinetic parameters of Δ35 and Δ195 WalK Spn were similar [Km (ATP) ≈ 37 μM; k cat ≈ 0.10 min−1] and typical of those of other histidine kinases. The catalytic efficiency of the two versions of WalK Spn ∼P were also similar in the phosphoryltransfer reaction to full-length WalR Spn . In contrast, absence of the HAMP-PAS domains significantly diminished the phosphatase activity of WalK Spn for WalR Spn ∼P. Deletion and point mutations confirmed that optimal WalK Spn phosphatase activity depended on the PAS domain as well as residues in the DHp domain. In addition, these WalK Spn DHp domain and ΔPAS mutations led to attenuation of virulence in a murine pneumonia model.

Context. Brown dwarfs (BD) are substellar objects intermediate between planets and stars with masses of ~13–80 MJ. While isolated BDs are most likely produced by gravitational collapse in molecular clouds down to masses of a few MJ, a non-negligible fraction of low-mass companions might be formed through the planet-formation channel in protoplanetary discs. The upper mass limit of objects formed within discs is still observationally unknown, the main reason being the strong dearth of BD companions at orbital periods shorter than 10 yr, also known as the BD desert. Aims. To address this question, we aim at determining the best statistics of companions within the 10–100 MJ mass regime and located closer than ~10 au to the primary star, while minimising observation and selection bias. Methods. We made extensive use of the radial velocity (RV) surveys of northern hemisphere FGK stars within 60 pc of the Sun, performed with the SOPHIE spectrograph at the Observatoire de Haute-Provence. We derived the Keplerian solutions of the RV variations of 54 sources. Public astrometric data of the HIPPARCOS and Gaia missions allowed us to constrain the masses of the companions for most sources. We introduce GASTON, a new method to derive inclination combining RVs and Keplerian and astrometric excess noise from Gaia DR1. Results. We report the discovery of 12 new BD candidates. For five of them, additional astrometric data led to a revision of their mass in the M-dwarf regime. Among the seven remaining objects, four are confirmed BD companions, and three others are likely also in this mass regime. Moreover, we report the detection of 42 M-dwarfs within the range of 90 MJ–0.52 M⊙. The resulting M sin i-P distribution of BD candidates shows a clear drop in the detection rate below 80-day orbital period. Above that limit, the BD desert appears rather wet, with a uniform distribution of the M sin i. We derive a minimum BD-detection frequency around Solar-like stars of 2.0 ± 0.5%.

The paper highlights Marie de Gournayʼs considerations, the author of the Renaissance, on the methods of translation, which she set out in her treatise A Letter to Monsignor de Gelas. It is stated that translation, as an intellectual activity, the main purpose of which is to enrich the national culture and improve the native language, has always been a privileged topic of reflection for this creative woman. Notes on this can be found in several treatises, which she collected in her work Les advis ou Les présens de la demoiselle de Gournay (1641). It is in these texts that she best describes her theory of translation. The concept of “theory” should be understood not from the standpoint of modern interpretation, but in the meaning of the original Greek word “theory”, i.e. a coherent presentation of explanations, concepts, ideas, proposals, laws and hypotheses. Thus, it is a question of foregrounding this “theory” in the context of an in-depth analysis of the authorʼs letter to Monsignor de Gelas. The author claims that Marie de Gournауʼs views deserve attention in contemporary discussions on translation and that her ideas about the art of translation still remain constructive and relevant, despite being put forward nearly five centuries ago.

Background: Overconsumption of added sugars, particularly refined sugars, has been shown to be associated with adverse health concerns. Aim: The present study aimed to elaborate calorie-reduced marmalades with nutritional benefits as well as satisfactory sensory properties, in order to reduce sugar intake without compromising consumers' acceptance. Materials and methods: Two formulas of sugar-reduced marmalades were elaborated by substituting 30% of sucrose with different commercial non-nutritive sweeteners: a blend of aspartame-acesulfame-K and sucralose. Physico-chemical, sensory, and microbiological analyses were carried out, in comparison with control sample marmalade. Blood glucose concentrations were determined in 12 healthy volunteers, at 30-min intervals until 120 min after consumption of marmalades. Results: Marmalade quality characterization revealed a significant effect of sucrose substitution on dry extract, Brix, reducing sugars, aw, and CIE Lab color parameters, but not on pH and acidity. The microbiological analysis highlighted that marmalades’ sanitary quality was in accordance with safety standards. Interestingly, sensory analysis by trained panelists showed that the substitution of sucrose by an intense sweetening substance did not impair the sensory properties. Our data also indicate that consumption of calorie-reduced marmalades significantly reduced acute postprandial glycemic responses in healthy volunteers; this effect was more pronounced with sucralose. Conclusions: Taken together, our results showed that the use of sucralose can constitute a relatively healthy choice for food basket of families, in particular for those with high risk of lifestyle-related diseases. Los medios de comunicación social constituyen, sin duda, la cultura principal en la que los niños y los jóvenes se desarrollan y ya nadie duda de que su información es mayor que la suministrada por la escuela.
