Abstract:
Les terminaisons axonales sont spécialisées pour coordonner la fusion des vésicules synaptiques (VS) à l’influx calcique lors de l’arrivée d’un potentiel d’action. L’organisation de ces terminaisons est encore mal connue. Nous avons étudié la structure de ces terminaisons par des techniques innovantes de microscopie électronique, afin de comprendre comment les VS peuvent être stabilisées dans le bouton axonal et à la zone active (le site de fusion). Pour éviter les artéfacts induit par les fixateurs aldéhydiques sur l’ultrastructure cellulaire, nous avons immobilisé des tranches d’hippocampes de rongeur par congélation sous haute pression. Nous avons ensuite utilisé la tomographie électronique pour analyser la répartition des VS et le cytosquelette avec une très haute résolution tridimensionnelle (~5 nm). Après congélation sous haute pression, les terminaisons synaptiques sont plus larges et la répartition des VS est moins dense qu’après fixation aldéhydique. L’analyse en tomographie électronique indique que les VS sont prises dans un réseau de filaments, notamment composé de synapsin. A la zone active, les VS ancrées à la membrane ne sont pas en hémifusion avec la membrane mais apposées et connectées avec elle par des filaments latéraux. Dans les souris déficientes pour Munc13, les VS ne sont pas ancrées à la membrane plasmique mais sont à quelques nanomètres de distance. Les protéines Munc13 apparaissent donc nécessaires pour l’ancrage («docking») des VS à la membrane plasmique. Ces résultats montrent l’organisation des VS dans le bouton et à la zone active et posent la question de savoir comment elles bougent dans ce maillage de filaments

Abstract:
This dissertation examines the notion of the female in performance, the latter term applying to occasions that are outside as well as within theatrical venues. I attempt to address the complex and mutually entailed interrelation between the 'normative' as it has, and continues to, govern female behaviour, and those manifestations deemed transgressive of these in some respect. I seek to postulate a conception of the performing female as a phenomenon which owes its force to the presence of both polarities. My preferred term for such a figure is the female bouffon, and after a preliminary definition of associated terms, I discuss the carnivalesque, socially licenced occasions of 'misrule' in pre-modern societies, where norms were temporarily suspended to permit women to 'make a spectacle of themselves'. Some contemporary parallels are furnished. I then address the larger and more discursive issues of the reflexive and self-applied norms of proper female conduct as offered and justified by industrial, scientifically authorized societies. From the above, I turn to the extraordinary creative ferment of the turn of the twentieth century, which witnessed the rebellious re-institution of older performance genres as well as the invention of new ones. I then discuss the associated theatrical theorizing that accompanied this era. After a detailed examination of the work of two contemporary practitioners, who, I consider, gather together past and present themes of bouffonerie in a compelling way, I give examples of my own performance practice, and some analysis of its reciprocal relation with an audience. I conclude with some speculative thoughts as to the future of the bouffonesque female performer.

La premiere partie evoque le cadre geographique et historique puis retrace l'evolution du peuplement de la prehistoire a la fin du moyen age a partir d'un inventaire des sites archeologiques de la commune. La seconde partie constitue le bilan de six campagnes de fouilles a corne, a 400m seulement du village perche actuel. Confirmant la topographie, elles ont montre que le site comprend deux parties distinctes: une zone castrale et une zone d'habitat formant un hameau subordonne. Dans un meandre de l'aurore, la premiere comportain une tour-salle en pierre servant de residence seigneuriale. Sa construction s'est accompagnee d'importants travaux de terrassement : fermeture et remodelage partiel du meandre, mise a l'abri des inondations par exhaussement du terrain autour des fondations du batiement etablies directement sur le sol. Au bas du versant occidental de la vallee. La seconde partie regroupait les habitations en torchis abritant le personnel au service du seigneur. La description du mobilier dans une serie de chapitre traitant des activites agricoles et artisanales suit celle des structures. L'abondance et la diversite de ce materiel autorisant l'etablissement de typologie, notamment pour la quincaillerie et la ceramique
The first part lists the various sites on the territory of l'islebouzon and establishs the successive phases of human occupation from prehistorytto the end of medieval times. The second parti given anccount of the excavati0ns carried out from 1988 to 1993 on the medieval site of corne, 400m the present perched village, as suggested by topography, they revealed two distinct zones: a castle zone including a stone tour-salle, the building of which was accompanied with important works of excavation, and a hamlet consisting of wood and wattle and daud huts. The finds are described in a series of chapters on the domestic, agricultural and craft activities of the inhabitants. Tehir large number and diversity allowed useful classifications. The life-span of the site (c. 1150-1250) makes it likely its creation by a boson de l'isle mentioned in 1176 in the gilont cartulary. It also allows us to confirm it was deserted when the present village (a castelnau which was given a "charte de coutumes" in 1266) was founded. Abstract:
This thesis is about the silver business of Matthew Boulton and John Fothergill at their Soho Manufactory near Birmingham. Their partnership lasted from 1762 until 1782. A rounded discussion of the topic is attempted. Within the contexts of industry elsewhere and Soho's other activities, successive chapters cover the early development, marketing, production, design, and later decline of the partners' silver. Silver plate was prestigious and, untypically for Boulton, he concentrated on sales to the public rather than trade customers. To attract orders he made modest charges. This was viable where mainly machinery was used to make plate, even though sales were not high, since the expense of machinery was substantially covered by the larger sales of non-silver items. However, where Boulton relied to a greater degree upon hand methods, he lacked technical means to compensate for low profit-margins. Moreover, inefficiency and the firm's lack of capital which led to substantial bankers' interest charges on payment for bullion, particularly when customers paid late, caused losses. These problems applied particularly to silver plate and were mainly responsible for the decision to reduce production drastically; however, the manufacture of a large range of small items remained relatively consistent. The thesis includes appendices. Some contain new information about annual totals for the following aspects of the business: the volume of assay silver; each type of article; pieces sold on commission; and sterling silver supplies. Other appendices provide details about the partners' silversmiths and extracts from a Soho inventory. This thesis involves a more detailed use of sources than previous studies of the topic. Apart from the silver itself (which is selectively illustrated), the Matthew Boulton Papers and statistics derived from The Birmingham Assay office provide the main sources. Manuscripts covering silver production elsewhere provide contextual material for understanding the partners' silver business.

Ce travail s'interroge sur la signification et la portée du concept de « souverain Bien » dans la philosophie pratique de Kant. Comment justifier l'affirmation kantienne selon laquelle le souverain Bien est l'objet nécessaire et a priori de la raison pratique pure ? L'objectif de la thèse est de chercher à rendre compte de ce moment téléologique qui surgit dans la « Dialectique de la raison pratique pure » et que la théorie normative exposée dans « l'Analytique » semble pourtant rendre inutile. L'aspiration au souverain Bien est nécessaire a priori parce qu'elle participe de l'inévitable et naturel élan métaphysique de la raison à l'inconditionné. On cherche ensuite à fonder la nécessité pratique du souverain Bien en montrant que si le souverain Bien ne fait pas partie des concepts fondamentaux de l'éthique, il fait cependant partie des concepts fondamentaux de l'application de l'éthique au contexte d'une volonté morale finie. Enfin, la nécessité morale de la tâche de promouvoir de souverain Bien vient ce que celle-ci est à rapporter à l'exigence immanente de la raison pratique de chercher à s'appliquer dans le monde
This study examines the role and the meaning of the concept of the « highest Good » in Kant's practical philosophy. It tries to give a justification for Kant's claim that the highest Good is the necessary and a priori object of pure practical reason, and to give an account for this teleological moment revealed into the « Dialectic of pure practical reason » even though the normative theory founded by the « Analytic » seems to make such a moment useless. At first, the highest Good appears as a necessary object because it refers to the ineluctable and natural striving of reason for the unconditioned. Then, the practical necessity of the highest Good is connected to the fact that this concept, which doesn't belong to the fundamental concepts of ethics, belongs nonetheless to the fundamental concepts of ethic's application to the situation of a finite moral volition. The aspiration to the highest Good is finally related to the moral requirement inherent to practical reason to have an application in the world of nature. For such application is a necessary and a priori duty, the practical task ordering to promote the highest Good inherits of this moral necessity

Ce travail de thèse repose sur l'analyse des processus impliqués dans l'apprentissage de nouveaux comportements moteurs, et sur l'étude de nos capacités à adapter notre comportement à de nouvelles situations. Nous avons réalisé une série de trois expériences où ont été utilisés des dispositifs expérimentaux permettant une analyse de mouvements discrets et continus. L'objectif principal qui a motivé ce travail était d'évaluer : 1) les variables d'apprentissage susceptibles d'influencer l'acquisition d'un geste discret et/ou continu (Expériences 1 et 2), et 2) la nature des représentations sensori-motrices dans l'apprentissage de séquences motrices (Expérience 3). Les données obtenues dans la première étude (Expériences 1 et 2) révèlent que la condition de pratique et/ou le niveau de similarité entre les tâches à apprendre sont susceptibles de moduler les processus intra- et/ou inter-tâches mis en jeu par l'apprenant au cours de la phase de pratique, et donc d'influencer l'apprentissage. De plus, nos résultats indiquent également que les facteurs condition de pratique et similarité des tâches interagissent avec le facteur quantité de pratique ; l'apprentissage de nouveaux comportements moteurs nécessite donc de prendre en considération l'ensemble de ces facteurs. Les résultats de l'Expérience 3 révèlent une prédominance du code visuo-spatial dans la représentation des séquences motrices, associée à une dépendance du système effecteur, quelle que soit la quantité de pratique. Nos résultats ne confirment pas l'hypothèse du passage progressif d'un codage de type visuo-spatial (effecteur indépendant) au début de la pratique (après 1 jour), à un codage de type moteur (effecteur dépendant) avec davantage de pratique (après 2 jours). Les implications théoriques et pratiques qui émergent de ces résultats sont discutées au regard des données et modèles théoriques préexistants
This thesis focuses on the analysis of the cognitive processes underlying learning of new motor behavior, and the study of our faculties to adapt this behavior to new ones. To address this issue, three experiments were designed to analyze discrete and continuous movements. The main purpose of this work was to assess: 1) learning variables that are supposed to influence the acquisition of discrete and/or continuous movements (Experiments 1 and 2), and 2) the way sequential movements are coded (Experiment 3). Experiments 1 and 2 revealed that the schedule of practice and/or task similarity can modulate intra- and/or inter-task processing during practice, and thus, influence subsequent learning. Further, our results also indicated that both schedule of practice and task similarity interact with the amount of practice; thus, learning of new motor behavior supposes to consider all these factors. In Experiment 3, our data argue for the existence of both effectordependent and visual-spatial sequence representation, irrespective of the amount of practice. These findings are not consistent with the notion that the amount of practice is thought to be a determinant factor in the shifting from a visual-spatial (effector-independent) to a motor (effector-dependent) code representation (from 1 to 2 days of practice). Theoretical and practical implications emerging from these results are discussed with regard to pre-existent theoretical data and models

Abstract:
La reproduction fidèle de la locomotion humaine est une problématique d'actualité concernant les robots humanoïdes. Les travaux présentés dans ce mémoire ont pour objectif de définir une procédure permettant l'imitation par les robots humanoïdes du mouvement humain. Dans un premier temps les techniques de l'analyse du mouvement humain sont présentées. Le protocole de mesure adopté est exposé, ainsi que le calcul des angles articulaires. La problématique de la détection des évènements de contact est abordée en proposant l'adaptation des méthodes existantes pour des mouvements complexes. Les algorithmes sont validés par une série de mesures effectuées sur une trentaine de sujets sains. La deuxième partie traite de la transposition du mouvement humain aux robots. Une fois la problématique générale et le processus de transposition définis, le critère d'équilibre des robots marcheurs est présenté. A partir des données du mouvement humain capturé, les trajectoires de référence des pieds et du ZMP sont définies. Une modification de ces trajectoires est ensuite effectuée dans le cas de risque de collision entre les pieds notamment dans le cas de l'exécution d’un slalom. Finallement un algorithme de cinématique inverse, développé pour cette problématique est utilisé pour déterminer les angles articulaires du robot associés aux trajectoires de référence des pieds et du ZMP. Plusieurs applications sur les robots HOAP-3 et HPR-2 sont présentées. Les trajectoires sont validées vis-à-vis du maintien de l'équilibre grâce à des simulations dynamiques du mouvement ainsi que vis-à-vis des limites des actionneurs
The true reproduction of human locomotion is a topical issue on humanoid robots. The goal of this work is to define a process to imitate the human motion with humanoid robots. In the first part, the motion capture techniques are presented. The measurement protocol adopted is exposed and the calculation of joint angles. An adaptation of three existing algorithms is proposed to detect the contact events during complex movements. The method is valided by measurements on thirty healthy subjects. The second part deals with the generation of humanoid trajectories imitating the human motion. Once the problem and the imitation process are defined, the balance criterion of walking robots is presented. Using data from human motion capture, the reference trajectories of the feet and ZMP are defined. These paths are modified to avoid collision between feet, particularly in the case of executing a slalom. Finally an inverse kinematics algorithm developed for this problem is used to determine the joint angles associated with the robot reference trajectories of the feet and ZMP. Several applications on robots HOAP-3 and HRP-2 are presented. The trajectories are validated according to the robot balance through dynamic simulations of the computed motion, and respecting the limits of actuators

De la fin du XVIIème siècle jusqu'à l'abolition de 1848, l'île Bourbon, dans le Sud-Ouest de l'océan Indien, a connu un système esclavagiste marqué par une codification et des pratiques des différents pouvoirs dévalorisant ou niant les structures familiales des esclaves. A côté d'autres moyens de résistances serviles comme le marronnage ou la révolte, il apparaît, grâce à la reconstruction des familles esclaves, que ces formes d'organisation ont permis à une population provenant de razzias en Afrique ou à Madagascar, puis fortement créolisée, de retrouver son humanité, en investissant le champ de la parenté dont les pouvoirs civils ou religieux la privaient. Si un nombre restreint de familles furent reconnues, comme à l'époque de la Compagnie des Indes, la majorité des esclaves vécurent au sein de familles marrons, ignorées et méprisées. Elles apparaitront au grand jour après l'abolition de l'esclavage en 1848. La natalité fut reconnu importante au sein du groupe des esclaves mais accompagnée d'une forte mortalité infantile. Lieu privilégié de transmissions de valeurs culturelles et linguistiques, la famille esclave, quelles qu'en soient les formes, a permis à de très nombreux esclaves de survivre à un système inhumain, les exclus de la parenté appartenant essentiellement aux groupes ayant connu la destruction de leur système familial, à Madagascar ou en Afrique.

La sociologie a proposé une multitude d’explications sur divers phénomènes sociaux. Cependant, plusieurs sociologues, dont Boudon se sont surtout intéressés à l’aspect scientifique de la sociologie. Les questions les plus souvent posées lorsqu’on entreprend de telles études se penchent essentiellement sur le caractère scientifique de la discipline sociologique. Quels sont les critères de scientificité de la sociologie ? Quel est son sens épistémologique dans la littérature scientifique ? Quel est son raisonnement existentiel dans la construction du savoir ? La sociologie pourrait-elle être traitée méthodologiquement parlant au même titre que les autres disciplines des sciences naturelles ou particulières ? Si oui, pourrait-elle expliquer les lois des phénomènes sociaux à partir des observations hypothético-déductives sur la nature de la réalité sociale ? Dans cette thèse, nous tenterons de répondre à ces questions en nous appuyant sur des concepts élaborés par les sociologues classiques et contemporains.

This study aims to understand the divergent positions between the theory of social reproduction of Pierre Bourdieu and the theory of rational choices of Raymond Boudon. Theories involving the sociology of education are often placed at opposite poles in relation individual / society. On one side, the field of action subjectively motivated, rational individual choices, represented by Raymond Boudon, became known as methodological individualism. On the other hand, the inverse perspective, holistic, derived from macrosociological contexts related to the size of the collective structures represented by Bourdieu, known as structuring. In trying to understand the differences of these two theories, research is supported in methodologies related to bibliographic studies, as well as those pertaining to comparative studies of works and authors. To understand the theories of sociology of education, the approaches are preceded by a summary of the general history of western education, contextualized, where relevant, with the history of education in Brazil. Moreover, an itinerary is done in the history of sociology of education to a better understanding of its various currents. With this purpose is focused on the following: a) Identify and analyze the characteristics and contributions of each approach in the construction of modern conceptions of education; b) describe the most relevant findings of the traditions of social science research in education and c) understand the various currents of the field of sociology of education with its various aspects, in seeking to understand the "crisis" of educational paradigms.
O presente trabalho tem por objetivo compreender as posições divergentes entre a teoria da reprodução social de Pierre Bourdieu e a teoria das escolhas racionais de Raymond Boudon. As teorias que envolvem a sociologia da educação são colocadas frequentemente em polos opostos na relação indivíduo/sociedade. De um lado, o campo da ação subjetivamente motivada, das escolhas individuais racionais, representado por Raymond Boudon, tornou-se conhecido como individualismo metodológico. Por outro lado, a perspectiva inversa, holista, oriunda de contextos macrossociológicos relacionada à dimensão das estruturas coletivas, representada por Pierre Bourdieu, conhecida como estruturalismo. Na tentativa de compreender as divergências dessas duas teorias, a investigação está apoiada em metodologias relacionadas aos estudos bibliográficos, assim como àqueles concernentes aos estudos comparativos de obras e autores. Para o entendimento das teorias da sociologia da educação, as abordagens são antecedidas por uma síntese da história geral da educação ocidental, contextualizadas, quando pertinente, com a história da educação no Brasil. Ademais, é feito um itinerário pela história da sociologia da educação para uma melhor percepção de suas diversas correntes. Com esta finalidade é focalizado o seguinte: a) Identificar e analisar as características e contribuições de cada uma das abordagens na construção das concepções modernas sobre educação; b) descrever as descobertas mais relevantes das tradições de pesquisa em ciências sociais na área educacional e c) compreender as diversas correntes do campo da sociologia da educação com suas várias vertentes, na busca do entendimento das “crises” dos paradigmas educacionais.

Le travail présenté dans ce mémoire porte sur le comportement des assemblages métalliques boulonnés avec platine d'about. La première partie décrit deux séries d'essais sur des assemblages métalliques avec platine d'about boulonnée renforcée ou non par raidisseur. La deuxième partie est consacrée au développement d'un modèle numérique 3D non linéaire validé sur la base des résultats d'essais. Les résultats sont aussi comparés aux formulations analytiques de l'EC3. Le modèle a permis d'analyser l'évolution des champs de contraintes et de la plasticité dans différentes zones de l'assemblage. Il a permis aussi de confirmer les observations expérimentales concernant l'influence du raidisseur de platine sur le mécanisme de transfert des efforts à travers son extrémité. Dans une troisième partie, l'influence de la flexion du boulon sur le comportement des tronçons en té est modélisée. Une étude paramétrique est menée pour évaluer les effets de paramètres tels que la dimension du tronçon, la rigidité de la rondelle, la position du boulon et l'épaisseur de la semelle sur la flexion du boulon soumis à un effort normal de traction.

Abstract:
Neurotransmitter release at central synapses is sustained by the synaptic vesicle cycle. It has been assumed that vesicle replenishment operates autonomously at individual presynaptic terminals. In this study the classical model of a compartmentalized synaptic vesicle cycle was tested by using a novel combination of FRAP (fluorescence recovery after photobleaching) and CLEM (correlative light and electron microscopy) in cultured hippocampal neurons. The stability of vesicle clusters labelled with fluorescent styryl dye at individual synapses were assessed by photobleaching using a confocal laser microscope and monitoring fluorescence recovery over time. The observed fluorescence recovery which was abolished by inhibitors of vesicular transport, suggested that synaptic vesicles recycled at sites outside the bleach region were transported along axons into bleached synapses. These newly-imported vesicles could undergo exocytosis upon stimulation, demonstrating that they formed part of the functional recycling pool. The spatial organization of imported vesicles in presynaptic boutons was examined using CLEM and FM dye photoconversion techniques. Imported vesicles were distributed throughout the native vesicle cluster, indicating that they become morphologically integrated into the synapse. The ability of imported vesicles to mix well with native vesicles highlights the dynamic nature of vesicle clusters at resting synapses. The departure of fluorescent packets from boutons into axons was observed by time- lapse microscopy. Ultrastructural analysis confirmed the mature state of donor synapses and showed these mobile packets to be loose aggregates of synaptic vesicles. Mobile vesicle clusters were comprised of vesicles from both the recycling and resting pools of the synapse, thus demonstrating no preference for mobility of any one vesicle population.

De nombreux travailleurs sont exposés à des niveaux de bruit trop élevés qui les mettent en danger de perdre leur audition. Cependant la réduction du bruit à la source, lorsqu'elle est techniquement possible, est souvent difficile à mettre en oeuvre pour des raisons économiques. La protection auditive individuelle reste donc la solution la plus répandue en pratique. Malheureusement, les protecteurs auditifs actuellement disponibles sur le marché sont rarement portés aussi continuellement qu'ils le devraient (car ils sont peu confortables ou doivent être enlevés pour communiquer) et sont généralement mal adaptés aux besoins de protection du travailleur (car leur atténuation individuelle effective est très généralement inconnue). Ce travail de thèse de doctorat, réalisé en étroite collaboration avec la compagnie SONOMAX SANTÉ AUDITIVE INC. avait pour objectif général le développement d'un bouchon d'oreille intelligent qui remédierait à l'ensemble des problèmes des protecteurs auditifs actuellement sur le marché et permettrait ainsi de prévenir efficacement la perte auditive en milieu industriel. Les problématiques associées à ce développement ont été formulées de façon détaillée siys sous l'aspect santé-sécurité au travail (port non continu du protecteur et protection inadaptée), l'aspect technique (développement d'un bouchon confortable du point de vue physique et perceptif et dont les performances sont mesurables) et l'aspect scientifique (modélisation de systèmes physiques, traitement du signal, instrumentation et mesure). Les développements techniques ont conduit à la mise au point d'un bouchon d'oreille sur-mesure obtenu par un procédé d'ajustement instantané au sein même de l'oreille du travailleur, dont les performances acoustiques sont mesurables individuellement grâce à l'utilisation d'une sonde microphonique et dont l'atténuation acoustique peut être adaptée au besoin du travailleur par l'insertion de filtres acoustiques passifs au travers du protecteur. Les développements scientifiques ont permis la mise au point d'une méthode de prédiction de l'atténuation acoustique à partir de la mesure d'un affaiblissement acoustique et la mise au point d'une méthode de prédiction de l'atténuation résultante de l'utilisation des filtres acoustiques.

Com a crescente demanda do mercado mundial de cafés especiais, os produtores brasileiros têm demonstrado interesse em retomar o cultivo do Bourbon, principalmente devido a seu potencial para produzir cafés de qualidade superior de bebida. Dessa forma, os melhoristas têm buscado incorporar acessos de Bourbon nos programas de melhoramento, a fim de obter cultivares para a produção de cafés especiais. Visando dar suporte aos programas de melhoramento da Empresa de Pesquisa Agropecuária de Minas Gerais (Epamig), foi instalado em Patrocínio, MG, um Banco Ativo de Germoplasma (BAG) com 1594 acessos, destes, 140 correspondem a Bourbon. Entretanto, a coleção de germoplasma de Bourbon ainda não foi devidamente caracterizada, fato que interfere no seu aproveitamento para fins comerciais e uso no melhoramento. Deste modo, a caracterização molecular desses genótipos é essencial para ampliar o conhecimento dos materiais genéticos disponíveis e, assim, incorporar os mais adequados nos programas de melhoramento genético do cafeeiro. Nesse sentido, o presente trabalho teve como objetivo caracterizar, por meio de marcadores SSR e AFLP, a coleção de Bourbon da Epamig, como uma ferramenta auxiliar na identificação dos acessos, avaliação da diversidade genética e do perfil molecular (Fingerprinting) dos genótipos. Na identificação dos acessos, apresentado no capítulo 1, os marcadores SSR foram utilizados para discriminar os genótipos de Bourbon juntamente com as técnicas de analise discriminante e redes neurais artificiais. Os locos SSR analisados foram adequados para diferenciar os acessos. Observou-se que as redes neurais artificiais apresentaram maior eficiência na classificação dos acessos de Bourbon do que a análise discriminante e foi possível identificar acessos que apresentaram misturas ou problemas na identificação inicial. Na avaliação da diversidade genética e do perfil molecular (Fingerprinting) dos acessos, apresentados no capítulo 2, os marcadores SSR e AFLP utilizados permitiram separar a maioria dos indivíduos analisados. Esses dados moleculares sugerem que, apesar da reconhecida base genética estreita de C. arabica, o germoplasma de Bourbon pertencente a Epamig apresenta variabilidade genética que pode ser explorada nos programas de melhoramento. A análise de fingerprinting permitiu identificar os perfis moleculares de cada acesso de Bourbon avaliado, os quais podem ser utilizados na identificação dos cafeeiros no banco. Dessa forma, os resultados obtidos nesse estudo fornecem subsídios para a manutenção e conservação da coleção de germoplasma de Bourbon. Além disso, deve auxiliar os melhoristas na escolha de genitores para possíveis hibridações em programas de melhoramento que buscam o desenvolvimento de cultivares com potencial para qualidade de bebida.
With the increasing demand of specialty coffee in the world, the Brazilian coffee producer have demonstrated interest to retake cultivating Bourbon due to its potential of producing high cup quality. So, the breeders are working to incorporate the Bourbon in the breeding programs to develop coffee cultivars with special cup quality. To support the breeding programs, the Agricultural Research Company of Minas Gerais (Epamig) established the active germplasm bank in Patrocínio, MG. This germplasm comprises of 1594 coffee accessions, among these 140 are Bourbons. However, the collection of Bourbon is not well characterized, which interferes with its efficient use for commercial purpose and genetic improvement. This shows the importance of molecular characterization of these genotypes to increase the knowledge about the genetic materials available and incorporated adequately in the genetic improvement of coffee. Thus, the present work was done with the objective of characterizing the Bourbon collections of Epamig using SSR and AFLP molecular markers as a tool to identify the accessions, genetic diversity study and Fingerprinting of the genotypes. In chapter 1, SSR molecular marker combined with discriminant analysis and artificial neural network were used to discriminate the genotypes of Bourbon. The result showed that the artificial neural network is more efficient to classify the accessions of Bourbon than discriminant analysis. It was possible to identify accessions with mixture or with identification problem. In the genetic diversity and fingerprinting analysis presented in chapter 2, SSR and AFLP markers used permitted to separate most of the individuals analyzed. These data suggested that despite the known narrow genetic base of C. arabica, the Bourbon germplasm belonging to Epamig has enough genetic variability that can be exploited in breeding programs. The fingerprinting analysis allowed identifying molecular profile for each accession of Bourbon evaluated that can be used in identification of coffee plants of the germoplasm. Besides this, it can aid the breeders to select the parents for possible hybridization in the breeding programs to develop cultivars with high cup quality potential.

Sleep fragmentation and disturbances of normal circadian rhythms are inherent in the aging human population. The rhesus macaque (Mucaca mulatta) exhibits similar disruption in sleep patterns and is a useful model to study these disturbances. Orexin is an excitatory neuropeptide that has been implicated in the regulation of wakefulness and alertness. Specifically, it has projections from its neuronal bodies in the lateral and perifornical hypothalamus to various forebrain and brainstem regions that control arousal state. One of the regions of high innervation by these orexigenic neurons is the thalamus. This study used a semi-quantitative means to determine if there are age dependent changes in Orexin innervation in the thalamus. There was a general trend of decreasing axon density when analyzed with a novel semi-quantitative method (r = -0.515, p = 0.024, df = 17) and approached significance when assessed with a previously published method (r = -0.454, p = 0.0509, df = 17), indicating a decrease with age. Additionally, there was no significant decrease in unilateral bouton counts iii when examined with age using either method. Although the findings in this study point to an age-related decrease in axon terminals, further research should examine the total orexigenic positive content in the thalamus to explain why bouton counts do not seem to change.

Cette étude est faite dans le but de comprendre comment un assemblage boulonné se desserre ou se dévisse sous des sollicitations dynamiques. Dans un premier temps, nous avons réalisé une étude bibliographique portant sur les différents modes de sollicitations, axiales, transversales ou en rotation, ainsi que sur les normes. Nous avons ensuite abordé une étude expérimentale du dévissage spontané, sous sollicitations axiales. Nous avons défini, calculé et optimisé un premier montage expérimental travaillant en résonance. Nous avons réalisé ensuite un deuxième montage utilisant une masse inertielle, qui nous a permis d'observer les vissages et dévissages spontanés. Nous avons aussi mené des essais sous sollicitations transversales. Des mesures précises nous ont permis de déterminer des seuils de déplacement de la pièce serrée, à partir desquels le dévissage survient de façon certaine. Nous avons obtenu aussi un seuil de glissement de la tête de vis sur son appui et un seuil de glissement de la partie filetée dans son taraudage, sensiblement indépendants du niveau de précharge. Nous avons enfin modélisé le dévissage spontané, en premier temps analytiquement pour une vis courte ensuite par éléments finis pour une vis longue, et nous avons proposé un critère du dévissage spontané des vis courtes
This study is made with the aim of including how a bolted assembly loosens spontaneously under dynamic loadings. In a first time, we made a bibliographical study covering the various methods of loading, axial, transverse or in rotation, as well on the standards. We then approached an experimental study of spontaneous unscrewing under axial loadings. We defined, calculated and optimised a first experimental assembly working in resonance. We carried out then a second assembly using an inertial mass, which enabled us to observe spontaneous screwings and unscrewings. We also carried out tests under transverse loadings. Precise measures enabled us to determine thresholds of displacement of the clamped part from which unscrewing certainly occurs. We also obtained a threshold of slip of the screw head on its bearing surface, and a slip threshold of the threading, appreciably independent of the preload level. We modelled finally spontaneous unscrewing in first time analytically for a short screw then by finite elements for a long screw, and we proposed a criterion of the spontaneous unscrewing of the short screws

Les motoneurones spinaux occupent la place centrale du système moteur. Ils intègrent l’ensemble des informations provenant de système nerveux central et périphérique pour élaborer une commande motrice finale adaptée aux demandes de l’organisme et aux contraintes de l’environnement. En particulier, le seuil de recrutement et la fréquence de décharge des motoneurones sont des paramètres déterminants dans l’élaboration d’un signal approprié à l’intensité de l’effort requis. Il permet de définir l’ordre dans lequel les unités motrices sont recrutées au cours d’une activité physique : des unités motrices de type lent (S) pour le maintien de la posture, aux unités motrices de type rapide pour les efforts d’intensité modérée (FR) et de forte intensité (FF). Cette étude met en évidence l’existence d’un nouvel acteur mis en jeu dans la régulation de l’excitabilité motoneuronale. Il s’agit du canal chlorure activé par le calcium TMEM16F exprimé spécifiquement dans les motoneurones α au niveau des synapses cholinergiques appelées « bouton C ». A l’instar du rôle des boutons C, TMEM16F est nécessaire pour l’exécution d’un effort de forte intensité. En effet, en adéquation avec les enregistrements électrophysiologiques montrant une élévation du seuil de recrutement des motoneurones rapides TMEM16F-/-, la perte de TMEM16F induit des défauts moteurs à l’effort.La sclérose latérale amyotrophique (SLA), est une maladie neurodégénérative conduisant à la mort sélective des motoneurones. Parmi les processus pathologiques décrits, nous savons que l’excitabilité motoneuronale et l’homéostasie calcique constituent des éléments majeurs de la progression de la SLA. Ce sont des facteurs de vulnérabilité qui participent à la dégénérescence séquentielle des motoneurones FF et suivie des motoneurones FR. Étant donné la sensibilité de TMEM16F au calcium et son implication dans la régulation de l’excitabilité motoneuronale, nous avons inhibé l’expression de ce canal dans un modèle murin de SLA SOD1G93A et réalisé une étude longitudinale. Celle-ci met en évidence un effet protecteur de la délétion de TMEM16F qui est dépendant du genre
Spinal motoneurons have a prominent place in motor system. Motoneurons integrate all inputs from the central and peripheral nervous systems to construct a motor output adapted to the organism's demands and environmental constraints. In particular, recruitment threshold and firing frequency are key motoneuronal parameters in developing an appropriate signal regarding task-dependent demands. During muscle activity, motor units are orderly recruited beginning with slow-type (S) motor units for posture maintenance, followed by fast-type motor units for moderate intensity tasks (FR) and high intensity tasks (FF). Our study highlights a new factor involved in the regulation of motoneuron excitability. This refers to a calcium-activated chloride channel called TMEM16F, specifically expressed in α motoneurons at cholinergic C-bouton synapse. Likewise C-boutons, TMEM16F is required for the procution of high intensity effort. Indeed, in accordance with electrophysiological recordings showing an increase in recruitment threshold of fast TMEM16F-/- motoneurons, TMEM16F loss of function induces motor defects during an effort.Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease leading to the selective death of motoneurons. Among the pathological processes already described, we know that motoneuronal excitability and calcium homeostasis are major features in ALS progression. Those are vulnerability factors which contribute to sequential degeneration starting with FF motoneurons and followed by FR motoneurons. Given the TMEM16F sensitivity to calcium and its involvement in regulating motoneuron excitability, we inhibited its expression in a SOD1G93A mouse model of ALS and conducted a longitudinal study. It highlights a gender-dependent protective effect of TMEM16F loss

In this thesis a nerve-bouton preparation of Purkinje cells has been characterised. Mechanically isolated Purkinje cells are shown to retain active afferent nerve terminals. This provides a simplified system where the effects of manipulating the ion channels in nerve boutons can be studied without the potentially confounding influences of the rest of the presynaptic cell or surrounding tissue. Isolated Purkinje cells were initially identified for whole cell patch-clamp recordings by their distinctive size and shape. Vesicular release of neurotransmitter was evident by spontaneous inward synaptic currents with a characteristic time course. Antagonist application established that isolated Purkinje cells receive a mixture of inhibitory GABAergic and excitatory glutamatergic inputs. Changes in the frequency, amplitude, and burst behaviour of these spontaneously occurring synaptic currents were used to infer properties of the afferent boutons. Because rat Purkinje cells can be distinguished by their lack of postsynaptic NMDA receptors the presynaptic effects of NMDA application could be readily investigated. NMDA caused an increase in the frequency of postsynaptic events. The NMDA-induced increase was found to be sensitive to external magnesium and TTX application. NMDA application was found to increase the frequency of both GABAergic events and glutamatergic events. Physiologically, NMDA receptors in afferent inhibitory terminals are thought to be activated by the retrograde release of glutamate. So experiments were performed to determine if retrograde release of glutamate could also increase the frequency of glutamatergic events, however it was found that this process has a much more pronounced influence on the GABAergic events. Properties of afferent boutons were also probed with a range of potassium channel blockers. The relevant topics covered are pharmacology, synaptic transmission, and the role of NMDA receptors in the cerebellum and the main technique used is whole-cell patch clamp recording.

Matthew Boulton (1728-1809) is well known as an eighteenth-century industrialist, the founder of Soho Manufactory and the steam-engine business of Boulton and Watt. Less well known are his scientific and technical abilities in the field of metallurgy and coining, and his role in setting up the Soho Mint. The intention of this thesis is to focus on the coining activities of Matthew Boulton from 1787 until 1809, and to examine the key role he played in the modernisation of money. It is the result of an Arts and Humanities Research Council-funded collaboration with Birmingham Museum and Art Gallery, where, after examination of their extensive collection of coins, medals, tokens and dies produced at the Soho Mint, .research was used to produce a catalogue. A close visual study of the artefacts has been combined with evidence from contemporary archival material, and information from secondary sources, to provide a synthesis of the processes involved in making coins, and Matthew Boulton’s working practices in the eighteenth century. The thesis describes processes involved in making the 600 million coins, medals and tokens made at Soho Mint during Boulton’s lifetime. Chapter one briefly discusses his eighteenth-century background and the reasons for setting up the Soho Mint. In chapter two Boulton’s involvement in the copper and iron industries are discussed, including the importance of his contributions to both industries. He needed large amounts of copper and specialized iron products such as steel to make his coins at Soho Mint. In chapter three the technical aspects of minting are discussed, including Boulton and his team’s contribution to developing new techniques. The final chapter details how materials and products were transported, commissioned, and designed. The second half of the thesis is a sample catalogue of items produced at the Soho Mint prior to Boulton’s death in 1809. Fuller catalogues of Soho Mint products have been produced for several institutions as a result of this research, for use by museum curators, historians and professional and amateur numismatists. Only a selection of this aspect of the research is included, because of the word limitations for PhD theses.

Drosophila melanogaster is an excellent model system to identify genes involved in synaptic growth and function. In Drosophila, the Serrate (Ser) gene encodes a transmembrane protein that is a ligand for Notch receptor. Several previous studies implicated a role for Serrate in normal wing development and patterning. In this study, I demonstrate that Serrate is required for normal synaptic growth and function. I characterized the phenotype of a Serrate mutation (serB936) that was identified by an EMS-induced genetic screen aimed at identifying novel genes that play a role in synaptic growth and function. Co-localization studies show that Serrate protein is expressed at both the pre- and postsynaptic side of larval neuromuscular junctions (NMJs). Mutations in ser impair synaptic transmission at larval NMJs. This defect is entirely presynaptic, as nerve-evoked excitatory junction potentials (EJP) and quantal content (QC) of neurotransmitter release are significantly reduced when compared to wild-type control. Further, mutations in ser also alter the growth of the NMJ and the underlying muscle. Mutations in ser significantly reduce the size of larval body wall muscles (length and surface area) as well as the number and size of synaptic boutons, and the number of secondary axonal branches. Ubiquitous or muscle-specific expression of normal Serrate in serB936 mutants restores a normal muscle size but not a normal size and structure of the innervating NMJ. However, expression of normal Serrate in the motor axon restores a normal number of synaptic boutons and secondary branches at serB936 mutant NMJs. In addition, it restores normal neurotransmitter release. These data suggest that Serrate protein is required presynaptically for normal synaptic growth and function. Interestingly, overexpression of Serrate in a wild type background resulted in similar phenotypes than to those of loss-of-function mutants. In conclusion, these data suggest a new functional role for Serrate in synaptic growth and function.

Cette situation lui conferait en meme temps une importance commerciale evidente, heritiere d'une tradition remontant a l'antiquite. Dans le cadre historique et geographique, le cadastre de mille trois cent neuf, illustre ce que fut l'interet de boulbon, mettant en relief la nature du finage, sa mise en valeur, l'importance du patrimoine, sa physionomie patronymique et sociale. Le village medieval y apparait en effet groupe, au centre d'un finage tres parallise, reconquis sur le terrain marecageux du rhone et portant la triple culture mediterraneenne, ble, vigne, oliviers. Juridiquement, boulbon est le cadre d'une seigneurie dont le pouvoir est partage entre les seigneurs et la communaute des habitants, l'universitas. Le village resiste ra bien aux calamites du siecle, comme le montre le cadastre de mille quatre cent neuf. L'etude demographique laisse apparaitre une relative stabilite de la population en meme temps qu'un certain dynamisme puisqu'il y a progression des espaces cultives. Aujourd'hui, on releve a boulbon des elements de permanence : la physionomie du village, ses toponymes, son activite agricole. Ce qui a change, ce sont les cultures, les taches plus diversifiees de ses habitants, un habitat "extramuros" en voie de developpement. On voit l'interet de l'edition du cadastre de mille trois cent neuf, le plus ancien de provence pour l'etude d'un village d'aujourd'hui dans sa continuite historique.

‘They say very clearly that I am mad, but they are the ones that do mad things’. So wrote Franciscan Friar Manuel Sevane in 1796, in one of many letters to the Viceroy of New Spain. Sevane’s letters catalogue his protest against the treatment he received at the hands of his fellow friars at the Apostolic College of Pachuca in New Spain and contest their plans to return him to Spain. Sevane’s case is an example of the rich archival material that demonstrates how the term madness was used in Bourbon Mexico (1713-1808). His words exemplify how people used madness to mark out individuals, behaviours, and ideas they thought immoral. For the friars living with Sevane, his violence, peculiar noises, and disrespect for religious practices were evidence of his madness. Sevane himself used the same rhetoric to criticise the friars for shunning him, writing reports on him, and ultimately trying to remove him from their community. Historians have yet to fully examine the complex cultural significance of the term and its role in daily life in Bourbon Mexico. This thesis provides a new interpretation of madness. It takes as its starting point Michel Foucault’s concept of ‘technologies of the self’ in order to build upon the growing history of selfhood in Mexico. By exploring the role of madness in constructing selfhood, I explain how elites used the term’s interconnections with sin to delineate the boundaries between acceptable and unacceptable behaviour. The first chapter maps out an innovative historiographical framework for examining madness, introducing a range of categories of historical analysis that have not yet been used in this context. Chapter Two foregrounds a major claim of the thesis, that elite thinkers considered human sin to put all individuals at risk of falling into madness. It explores the relationship between the concepts of sin and madness that underpinned elite uses of madness as a rhetorical tool in a selection of sermons, political tracts, philosophical texts, and poems. Chapter Three analyses underexplored medical texts and shows that madness was understood to be humoral throughout the period. It also argues that madness did not become a secular concept in the eighteenth century; rather, medical texts conceived of madness as a tool for moral regulation. The fourth chapter examines the ways people used their bodies and how others used their sensory perceptions to label individuals as mad. Drawing on histories of the senses and emotions, the chapter analyses exceptionally detailed cases of clerical madness from missionary and Inquisition records. Chapter Five examines how the categories of gender and calidad, a socio-racial term denoting quality or status, interacted with the concept of madness in Inquisition cases. I conclude that an analysis of madness enables us to deepen our understanding of what it was to be human in Bourbon Mexico.
