Nowadays, the subjects of external communication are increasingly diversified, and China's new media forms of external communication are rich and varied. In contrast to the authoritative narrative style and tone of official media, short videos have emerged as an important medium for international communication in the new era. With characteristics such as fragmented content, entertainment value, popularity, and colloquial language, they have achieved significant dissemination effects on global social media platforms like YouTube. Therefore, more and more unofficial self-media figures are attempting to share unique Chinese stories and culture through short videos on YouTube, and Li Ziqi is one of them. Li Ziqi has amassed 17.6 million subscribers on YouTube, with a total of 128 videos published and over 2.9 billion views at the time of data collection (2023.07.02). Her short videos showcase her leisurely daily life in natural landscapes, diverse culinary culture, and traditional Chinese crafts culture, among other aspects of traditional culture. At present, Li Ziqi's "Traditional Crafts" series has become one of the important platforms for the external dissemination of Chinese traditional craft culture. This study takes the translation of the subtitles in four short videos from Li Ziqi's "Traditional Crafts" series on the YouTube platform as the research object. From the perspective of the new media environment, it analyzes the external communication of traditional Chinese craft culture in the new era and provides a summary of reflections.

National image is an important part of national identity. «Cultural soft power» can help a country spread national culture values in international relations, and create a good image in the minds of the people of other countries. With the development of Internet technology, new communication methods have emerged, and the scope of the audience has been continuously expanded, which is helpful to the spread of the national culture to the most. Being in vogue with the China cultural specifics in the digital environment will become a new model of communication strategy for shaping the country's image. The study analyzed popular account "李子柒Li Ziqi" (Rus. Ли Цзыци) created by Chinese Internet celebrity on the YouTube video platform and explored ways to shape China's image through cultural videos. We used the Octopus data collection platform to collect 1,000 comments from 20 videos posted on the "李子柒Li Ziqi" account from August 22, 2017 to October 15, 2021. The study content analyzed the comments of Li Ziqi's videos on the YouTube platform. The study found that digital traditional cultural communication has no ideological purpose, but it allows the viewers to identify with the culture. The research revealed: 1. The development of new technologies has expanded the possibility of visual communication, eliminated language barriers, and increased the dissemination of content. 2. The effect of “self-media” surpasses professional media in this sense. 3. Cross-cultural communication in the digital environment helps the audience to form a new identity and create a positive national image.

Abstract Short videos depicting rural China have gained popularity on social media domestically and internationally. Among the genre’s creators, Li Ziqi stands out for her unique style of culinary craft, starting from the most basic materials. I interpret Li Ziqi’s mushroom videos as multimodal “argumentative meshworks” casting a counterstatement to the “involuted” urban life and nature/culture division. To unfold the analyses, I first place videos in the context of urban ills. Built on previous studies of multimodal argumentation and entanglement anthropology, I define “argumentative meshworks” in three aspects: a human-nonhuman entanglement, a simplicity-complexity harmony, and a production-audience interaction. Then I select three mushroom videos as artefacts to unpack the multimodal meshworks. Following the empirical call argumentation studies, I use viewers’ comments to support my points throughout the whole piece. This inquiry explores multimodal argumentation’s new possibility to not only stress things out but create space for harmony and peace of mind.

This paper critically addresses the significant issue of educational opportunity inequality faced by Asian women, exploring its root causes and potential solutions, predicated on principles of justice and social development. The subjects of the study are Asian women, who are encumbered by intersecting societal stereotypes, cultural expectations, and institutional barriers, thereby curtailing their educational prospects. The study deploys an analytical interpretative approach, analyzing various societal stereotypes and prejudices, cultural factors, and systemic constraints, substantiated through a qualitative method. Findings reveal a collective imposition of an educational opportunity ceiling or Bamboo Ceiling on Asian women, proposing potential solutions including policy reforms, awareness initiatives, and educational strategies. Innovative methods are suggested, inspired by the narratives of Asian women, Li Ziqi and Papi Jiang, who overcame such inequalities. Ultimately, the study posits that acknowledging and addressing these entrenched sources of inequality could engender an environment fostering equal educational opportunities for Asian women, aiding in the demolition of the Bamboo Ceiling.

ABSTRACT The genre of Chinese rural “self-” or “we-media” (zi meiti) has drawn ire for “spectacularizing” the “rural lifescape” for urban consumers of China’s rapidly developing platform economy. This article focuses instead on how the videos of Li Ziqi and Qiaofu Jiumei recenter production as a key visual force in global capitalism. By depicting the dynamic reciprocities between traditionally rural, artisanal or craft-based, and immaterial or affective labor, and—on the other hand—more ostensibly commercial farm work and e-commerce, Chinese microcelebrities show that different labor relations can dialectically coexist and overlap under contemporary state capitalism despite developments in productivity and automation that impute a historical directionality to the nature of work. The article discusses this in relation to the historical context and theoretical design of the People’s Republic of China’s recent Rural Revitalization strategic plan, as well as the country’s growing platform economy. Belying Li’s aestheticized portrayal of craftsmanship and self-sufficiency is the notorious non-appearing of alienated labor, private property, and the continuous expropriation of nature—but also their potential reversibility. Jiumei’s candid depictions of rural e-commerce evidence rural platformization’s value-extraction; and yet, perceptible in her comparatively more impoverished images are hints of the return of more conventional forms of work.

Abstract Background: Prostate cancer is the second leading cause of cancer-related death in men in the United States and is dependent on androgens and androgen receptor (AR) activation. Androgen deprivation therapy (ADT) is the mainstay treatment, but it as well as next-generation treatments eventually fail. Disease then progresses as castration-resistant prostate cancer (CRPC), which is driven by reactivation of the androgen-AR axis. 3β-hydroxysteroid dehydrogenase-1 (3βHSD1) catalyzes the rate-limiting step for potent androgen synthesis from adrenal-derived precursors, thereby stimulating CRPC. HSD3B1, which encodes for 3βHSD1, has a common missense-encoding germline variation that enhances conversion from adrenal DHEA to potent androgens. Clinical studies demonstrate that this “adrenal-permissive” HSD3B1 genotype confers more rapid progression on ADT, and recent studies show that overall survival is shorter in patients with the adrenal-permissive genotype who are treated with ADT + enzalutamide. The Etk/BMX kinase may be involved in the development and progression of CRPC. We sought to determine whether phosphorylation of 3βHSD1 is required for its activation and that the necessary kinase could be a potential target. Methods: In prostate cancer lines, mouse xenograft models, and fresh patient prostatectomy tissues, mass spectrometry and HPLC were used to identify steroid metabolites. Genetic and pharmacological methods were used to identify inhibition of BMX-mediated 3βHSD1 phosphorylation in CRPC. Results: BMX inhibition suppressed castration-resistant prostate cancer through targeting 3βHSD1 phosphorylation. 3βHSD1 transcription level and enzyme activity were induced in enzalutamide-resistant prostate cancer cell lines. 3βHSD1 and SF-1 transcription levels were positively correlated in prostate adenocarcinoma (TCGA database). BMX inhibition by zanubrutinib suppressed enzalutamide resistance induced 3βHSD1 enzyme activity. Conclusions: BMX promotes CRPC by regulating 3βHSD1 phosphorylation. BMX inhibition has potential as a new therapy to optimize CRPC treatment. Abstract Introduction Differences in DNA methylation along cell-free DNA (cfDNA) fragments are known to exist between cancer patients and healthy individuals. Profiling these aberrant methylation patterns in urinary cfDNA (ucfNDA) holds great promise for the non-invasive detection and monitoring of bladder tumors, including non-muscle invasive bladder cancer (NMIBC). Here, we analyzed fragment-level DNA methylation patterns in urine samples obtained prior to repeat transurethral resection of a bladder tumor (reTURBT) from a cohort of NMIBC patients. The results were compared to traditional clinical disease assessment and mutation-based tumor fractions. We found a strong concordance between DNA methylation abnormality and clinical diagnosis, as well as tumor fractions, highlighting the potential of epigenetic alterations as a non-invasive indicator of bladder cancer status. Methods Patients with high-risk NMIBC (n = 30) were prospectively enrolled prior to reTURBT. The PredicineEPIC™ genome-wide methylation analysis and PredicineBEACON™ MRD profiling was performed on pre-repeat TURBT urine samples. Abnormally methylated genomic fragments were detected against a background model built from a panel of plasma samples from healthy donors. Results DNA methylation abnormality was significantly higher (p = 4.8 × 10−6) in samples classified as NIMBC positive by a clinical pathologist (n = 19) compared to negative samples (n = 11), as well as compared to hold-out healthy donor samples (n=24; p = 7.0 × 10−9). Classifying the samples based on a DNA methylation score cut-off derived from healthy donor samples, 24/30 samples showed concordant disease status. Furthermore, DNA methylation abnormality scores correlated positively with mutation-based tumor fractions (r = 0.62), with higher grade samples showing a stronger correlation (r = 0.81; n = 13) than lower grade samples (r = 0.54; n = 17). Conclusion We demonstrate the utility of methylation patterns extracted ucfDNA samples obtained prior to reTURBT to detect and monitor NMIBC. Urinary DNA methylation patterns were highly concordant with clinical pathology status and as well as tumor fractions determined from mutation analysis. Abstract Introduction: Tumor endothelial cells (ECs) support the tumor microenvironment and regulate the status of tumor cells. GPRC5B, enriched in ECs, is linked to poor prognosis in colorectal cancer (CRC). Yet, the influence of GPRC5B on ECs and its regulatory mechanism on CRC stemness are still elusive. Therefore, this study aims to clarify the impact of GPRC5B on ECs states, the specific role of GPRC5B-marked ECs (GPRC5B+ ECs) in tumor/stromal interactions and the mechanisms regulating CRC stemness. Methods: We analyzed scRNA-seq data from 18 CRC samples to decode the endothelial heterogeneity. We used an original SVM-based algorithm to find EC subsets linked to CRC stemness, ultimately identifying the GPRC5B+ ECs. By deconvoluted bulk RNA-seq data, we evaluated stemness level in GPRC5B+ EC-enriched samples. Cellular interactions and spatial omics analyses were employed to examine the signaling axis between GPRC5B+ECs and CRC stem cells. ChIP-seq and hdWGCNA algorithm pinpointed upstream regulators in the axis. We also developed a GPRC5B-overexpressing EC line and co-cultured it with CRC cells, whose stemness was then assessed by spheroid formation assays and western blot. Inhibitors targeting this signaling axis were administered, and changes in stemness phenotype and key signaling molecules were measured in co-cultured CRC cells. Finally, we constructed a CT26 subcutaneous tumor mouse model and treated the mice with siRNA targeting GPRC5B (siGPRC5B) to monitor tumor growth. Tumor tissues were collected to detect the expression of stemness markers using IHC and western blot. Results: GPRC5B+ ECs exhibited high angiogenic potential, with significant ERK pathway activation. ScRNA and bulk analyses showed GPRC5B+ ECs enhance CRC stemness, which was further confirmed by the increased spheroid formation ability and enhanced expression of KLF4 and SOX2 in CRC cells after co-culturing with GPRC5B-high ECs. Mouse models showed that high GPRC5B expression in ECs fosters tumor stemness and progression. Mechanistically, multi-omics and in vitro analyses revealed that GPRC5B in ECs activated the ERK-SP1 axis to promote the expression of ADAM17, which cleaved and activated JAG1, ultimately enhancing stemness of CRC cells via JAG1/NOTCH1 interaction. Application of ERK pathway or ADAM17 inhibitors to ECs decreased tumor stemness, respectively. Besides, application of siGPRC5B effectively inhibited tumor growth in vivo. Conclusion: Through single-cell multi-omics analyses, we identified a novel GPRC5B-marked EC subset, promoting angiogenesis and CRC progression, and revealed that GPRC5B+ ECs enhance tumor stemness via the ERK/SP1/ADAM17/JAG1-NOTCH1 axis. The study offers insights into CRC endothelial heterogeneity and tumor/stromal interactions, and future targeted therapy will be conducted to elucidate its clinical application potential. The hydrogenation of organic substances is a central process type in various industries[1]. The usage of gaseous hydrogen, which is primarily sourced from fossil fuels, entails the release of CO2 during its production. In addition, the usage of gaseous hydrogen also carries a safety risk, which should be reduced as much as possible. An alternative approach is direct electrochemical hydrogenation with water or other protic liquids, where the required hydrogen is produced in situ and directly utilized. If this is driven by renewable energy, it represents a sustainable synthesis method that does not require stoichiometric reduction agents or high-temperature conditions[2]. Transition metal chalcogenides, particularly pentlandites (FexCoyNizSaSeb, x+y+z=9, a+b=8), exhibit high conductivity and serve as efficient and affordable electrocatalysts for this purpose. The utilization of hierarchical functional layers in the final electrode greatly enhances their efficiency[3, 4]. A comprehensive approach to analyzing the vast and complex electrode production parameter space, encompassing process parameters and material variables for ink formulation and layer deposition, is critical for understanding the intricate mechanisms within the manufacturing process chain and to maximize the electrochemical conversion efficiency. For this purpose, a time-saving and cost-effective coherent workflow was developed which enables a systematic analysis of several parameters, maximizing the knowledge gained from each experiment while minimizing the total amount of experiments. The workflow can be seen in Figure 1. In step 1, the materials and substrates to be investigated are first selected. Then, in step 2, catalyst inks are formulated, examined, and optimized concerning their sedimentation stability and compatibility with different binders and organic additives. Subsequently, in step 3, substrates are coated with the optimized catalyst inks and coating parameters such as catalyst mass loading are varied at the same time. In the fourth and final step, the resulting electrodes are tested electrochemically and with the help of imaging methods to shed light on process-structure-property relationships. The described procedure enables successive optimization and establishment of efficient membrane-electrode-assemblies for electroorganic reactions, offering new avenues for efficient, green, and sustainable synthesis pathways. Abstract This study illustrates that for the Chinese bare form reflexive ziji ‘self’, feature conflicts can cause both the local binding blocking effect and the long-distance binding blocking effect. To make long-distance binding possible, a feature-matching requirement between the long-distance antecedent and the reindexed local reflexive must be met, as proposed by Tang (1989). In addition to this, a feature-matching requirement between the verb and its subject and object must also be satisfied, with the latter likely having a suppressive effect on the former. Further research is needed to understand the interaction of these two feature-matching requirements and what constraints exist. Furthermore, the blocking effect can be triggered by a variety of factors, including syntax, feature agreement, pragmatics, discourse prominence, animacy and thematic conditions and other combined effects. Ziji’s odd performance might be the result of their joint efforts. We need to further clarify how these factors work together.

Piezoelectric materials are a group of important functional building blocks that interfacing the human body by coupling biomechanical energy and electricity. So far, many technology innovations have advanced piezoelectric materials and composites toward a broad range of biomedical applications, which possess unique biocompatibility and flexibility. Fundamentally, materials design and engineering draw the boundary where this technology may advance. In this talk, I introduce our most recent development of piezoelectric materials and composites that are particularly designed for implantable nanogenerator applications. First, I present our wafer-scale approach to creating piezoelectric biomaterial thin films based on amino acids. The self-assembled sandwich film and truss-like structures enabled both strong piezoelectricity and largely improved flexibility. Then, new ferroelectric composites are presented as a new material used in 3D printing for directly manufacturing of piezoelectric architectures with tunable piezoelectric and mechanical properties. Toward the end, novel applications of implantable piezoelectric materials are introduced, which enable the closed-loop electrostimulations for many biomedical therapeutics. Abstract:
This article discusses the several previously unknown Confucian texts discovered in 1993 in a Warring States tomb at Guodian, near Jingmen, Hubei Province. I believe that these works should be understood as doctrinal material deriving from a single tradition of Confucianism and datable to around 300 B.C. Of the surviving literature from the same period, they are closer to the Xunzi than to any other text, and anticipate several characteristic themes in Xunzi's philosophy. These are: the notion of human nature (xing 性),and the controversy over whether the source of morality is internar or “external”; the role of learning (xue 學)and habitual practice (xi 習) in moral development; the content and origin of ritual (li 禮), by which human beings accord with the Way; the conception of the ruler as the mind (xin 心) of the state; and the psychological utility of music (yue 樂) in inculcating proper values.It is especially important for scholars to take note of these connections with Xunzi, in view of the emerging trend to associate the Guodian manuscripts with Zisi, the famous grandson of Confucius, whom Xunzi bitterly criticized.

U radu se razmatraju dnevnički zapisi dviju hrvatskih autorica, Dragojle Jarnević (1812. – 1875.) i Ivane Brlić-Mažuranić (1874. – 1938.) te autobiografski roman Fališ mi u proljeću, u jeseni, u ljetu, u zimi, Mani Gotovac (1939. – 2019.). Navedena djela svojim poetičkim obilježjima, u širokom smislu riječi, slijede matricu ženskog pisma, nerijetko obilježenog iskustvom traume. Trauma se često manifestira autodestruktivnošću književnog subjekta iskazanom na dvama načinima: specifičnom uporabom jezičnih elemenata i/ili, na sadržajnoj razini teksta, eksplicitnim govorom književnog subjekta o vlastitim autodestruktivnim porivima, potaknutima nemogućnošću uspostavljanja zadovoljavajućeg odnosa s važećim društvenim paradigmama – obuhvatit ćemo ih simbolično nazivom ‘autoriteti’. Uz pretežno subverzivan odnos prema autoritetima, što rezultira autodestruktivnošću književnog subjekta, uočljiv je u Jarnević i Brlić-Mažuranić, obrnuto recipročan odnos između dinamike izražavanja autodestruktivnog i uvažavanja metafizičkog kao jednog oblika autoriteta. Kod Gotovac je odnos prema metafizičkom drukčiji u odnosu na njezine prethodnice, riječ je o svojevrsnom modelu »doživljaja vjere u nevjeri« (Šimundža). Cilj je rada: (1) razmotriti različite manifestacije autodestruktivnoga u književnih subjekata, (2) razmotriti odnos autodestruktivnog i metafizičkog te (3) otvoriti mogućnost drukčijeg iščitavanja subverzivnog elementa ženskog pisma razmatranjem pitanja može li se subverzivan odnos prema autoritetima promatrati kao latentna tendencija prema uspostavljanju dijaloga s njima.

BackgroundOrelabrutinib is an oral, highly-selective, irreversible inhibitor of Bruton’s tyrosine kinase (BTK). Orelabrutinib has been approved for the treatment of B cell malignancies in China. Two distinct lupus animal models showed significant efficacy of orelabrutinib in reducing disease activity, which supported the clinical development of orelabrutinib in Systemic Lupus Erythematosus (SLE).ObjectivesThis phase Ib/IIa, randomized, double-blind, placebo-controlled, dose-finding study aimed to evaluate the safety, tolerability, pharmacokinetics (PK), pharmacodynamics (PD), preliminary efficacy and biomarkers of orelabrutinib in patients with mild to moderate SLE who received standard of care (SoC) therapy.MethodsPatients diagnosed with SLE by the ACR classification criteria for ≥ 6 months, who had a SLEDAI-2K score ≥5 at screening, and were autoantibody-positive, were randomized 1:1:1:1 to receive oral orelabrutinib at 50mg, 80mg, 100mg or placebo once daily for 12 weeks, respectively.ResultsThis study randomized 60 patients with 55 patients who completed 12-week treatment. Age at baseline was 33.7±9.8 years and 96.7% were female. Baseline disease characteristics were generally balanced across treatment groups. Adverse events (AEs) were reported in 80%, 93.3% and 100% of orelabrutinib treated patients at doses of 50mg, 80mg and 100mg QD respectively versus 85.5% in placebo group. AEs were mostly mild or moderate. Treatment-related SAEs were reported in 3 patients treated with orelabrutinib, only 1 of which was grade 3. No deaths were reported. The plasma exposure of orelabrutinib (AUC and Cmax) was proportionally increased with doses. Nearly complete BTK occupancy was achieved at all dose levels, and the occupancy lasted for 24 hours without any decrease compared to that at 4 hour post-dosing. In all evaluable patients, the SLE Response Index (SRI)-4 response rates at week 12 were 50.0%, 61.5% and 64.3% in patients treated with orelabrutinib at 50mg (n=14), 80mg (n=13) and 100mg (n=14) respectively, compared with 35.7% in patients treated with placebo (n=14), which indicated the trend of dose-dependent improvement. Among the subgroup of patients with SLEDAI-2K≥8 at screening, SRI-4 response occurred in 70%, 70% and 66.7% of patients treated with orelabrutinib at 50mg (n=10), 80mg (n=10) and 100mg (n=9), respectively, compared with 30% who received placebo (n=10). Trends of reduced proteinuria, anti-dsDNA and IgG, total B cells and increased complements C4 were also observed following orelabrutinib treatment.ConclusionOrelabrutinib was generally safe and well tolerated in patients with SLE. Abstract Introduction: Aumolertinib, a third generation selective EGFR-TKI, is standard of care for the first-line treatment in advanced or metastatic NSCLC with sensitizing EGFR mutations in China. Evidence has shown that combining EGFR-TKIs with chemotherapy can improve outcomes compared to EGFR-TKIs monotherapy. AENEAS2 (NCT04923906) is a randomized, open-label, multicenter, phase 3 study assessing the efficacy and safety of aumolertinib plus chemotherapy versus aumolertinib alone as first-line treatment in locally advanced or metastatic NSCLC with sensitizing EGFR mutations. Methods: Patients who were naïve to treatment with locally advanced or metastatic NSCLC harboring EGFR mutations (exon 19 deletion or L858R mutation) were randomly assigned in a 1:1 ratio to receive aumolertinib 110 mg QD in combination with pemetrexed (500 mg/m2) plus cisplatin (75 mg/m2) or carboplatin (AUC5), versus to receive aumolertinib 110 mg QD monotherapy. The primary endpoint is progression free survival (PFS) assessed by IRC (Independent Review Committee) per RECIST v1.1. Data cutoff date: 2024/06/18. Results: A total of 624 patients were randomized to aumolertinib plus chemotherapy (n=310) or aumolertinib monotherapy (n=314) and stratified according to EGFR mutation status (Ex19del verse L858R) and CNS metastases at baseline (yes versus no). Baseline characteristics were balanced between treatment arms (aumolertinib plus chemotherapy versus aumolertinib): median age (range), 58.0 (30-84)/59.0 (31-81) years; 55.5/52.5 % female; 49.0/49.0% Ex19del; 51.0/51.0% L858R; 30.0/30.9% CNS metastases. Median follow-up was 23.4 months. Median PFS assessed by IRC was 28.9 months (95% CI, 26.3 to NA) in combination arm versus 18.9 months (95% CI, 17.8 to 21.1) in monotherapy arm; hazard ratio (HR) 0.471 (95% CI, 0.371 to 0.598; p&amp;lt;0.0001). The PFS benefit was consistent across pre-defined subgroups. The overall survival (OS) was immature with event-patient rate 21.6%; HR 0.442 (95% CI, 0.308 to 0.636; p&amp;lt;0.0001). Median cycles of pemetrexed exposure (range) were 20.0 (1-42) cycles, 88.8% of patients completed 4∼6 cycles of platinum therapy. All causality grade ≥3 AEs (aumolertinib plus chemotherapy versus aumolertinib): 75.7%/23.7%; AEs leading to discontinuation of aumolertinib: 3.0%/1.3%. The safety profile of aumolertinib plus pemetrexed and platinum was consistent with the established profiles of the individual agent. Conclusion: Aumolertinib plus chemotherapy as a first-line treatment in advanced EGFR-mutant NSCLC demonstrated a statistically significant and clinically meaningful PFS improvement over aumoletinib monotherapy, with a manageable safety profile. Abstract Tumor prognosis and immunotherapy efficacy coincides with the infiltration ratio and functional status of immune cells in the tumor microenvironment (TME). Signaling lymphocytes activating molecule factors (SLAMFs) has been reported to play a key role in regulating tumor immunity. Herein, using multi cohorts, in vitro and in vivo experiments, we showed that SLAMF9 worsens tumor and suppresses immune therapy responses by remodeling TME. A pan-cancer analysis showed that SLAMF9 had relatively high transcriptional levels within tumor tissues compared with normal tissues across most cancer types. High SLAMF9 expression is associated with poor prognosis and anti-PD-L1 therapy resistance in advanced clear cell renal cell carcinoma cohorts and metastatic urothelial cancer cohorts. SLAMF9 expression was markedly upregulated in anti-CTLA-4-resistant murine B16 melanoma compared with parental tumors. In particular, SLAMF9 expression was generally negatively correlated with CD8+T cell infiltration score in 12159 patients of 40 cancer types even using different immune cell infiltrating algorithms. Although in vitro experiments have shown that SLAMF9 knockdown has almost no effect on the malignant phenotype of tumor cells, subcutaneous melanoma and lung adenocarcinoma with SLAMF9 deficiency grew slowly, which featured with increased infiltration of CD8+T and PD-1+cells. More importantly, SLAMF9-knockdown significantly increases the sensitivity of subcutaneous melanoma to anti-CTLA-4 inhibitor or anti-PD-L1 antibodies and improved the survival benefits of tumor-bearing mice. This work highlights SLAMF9 as a new target in reshaping the tumor immune microenvironment by controlling the functional status of CD+8 T cells and to improve the efficacy of immunotherapy. Abstract Introduction: The molecular pathways involved in the response to radiation therapy (RT) in pancreatic ductal adenocarcinoma (PDAC) remain poorly understood. Attempts to combine RT with immunotherapies in PDAC have yielded inconsistent outcomes, highlighting a critical gap in our understanding of immune dynamics and mechanisms influencing RT response. Our study aims to uncover molecular pathways and cellular subtypes associated with radioresponse. To do so, we developed a novel cell-cell interaction platform, "SpaCCI", to map ligand-receptor (LR) interactions, essential for understanding cellular communication within the TME and its contribution to therapeutic resistance. Methods: Single-cell RNA sequencing (scRNA-seq) was performed on 16 matched pre- and post-treatment PDAC biopsies from a Phase I/II dose-escalation clinical trial of stereotactic body radiation therapy. Additionally, spatial transcriptomics (ST) was conducted on resected PDAC (20 treated with neoadjuvant chemoradiation, 8 untreated). We integrated scRNA-seq and ST datasets using cell type decomposition and developed SpaCCI to enhance LR interaction detection in tissues with complex cell mixtures. Results: ScRNA-seq identified five molecular subpopulations of cancer cells associated to RT response, with PDAC_3 enriched post-RT and marked by genes involved in lipid metabolism and ferroptosis defense. Post-RT depleted subpopulations, such as PDAC_0 and PDAC_prolif, saw upregulation of pathways related to rapid proliferation and oxidative phosphorylation. In a murine model, PDAC_3 was similarly enriched in post-RT tumors, with persister cell lines from these tumors showing overexpression of ferroptosis defense genes and resistance to ferroptosis inducers including Erastin and RSL3. ST analysis also revealed spatial immune changes post-RT, with increased cytotoxic and effector memory T cells balanced by regulatory T cells and M2-like macrophages, indicating immune escape. NK and memory B cells correlated with improved survival post-RT, while plasma IgG cells localized outside the tumor bed, suggesting compartmentalized immune responses. Using SpaCCI, we observed differential enrichment of ligand-receptor interactions in RT-treated tumors, particularly among lipid-associated tumor macrophages, cancer cells, myofibroblast-like cancer-associated fibroblasts, and endothelial cells. Persister PDAC_3 subpopulations showed enriched LR SEMA3 signaling with CAFs, myeloid cells, and endothelial cells consistent with axon guidance pathways. Conclusions: This study presents a molecular and cellular atlas of PDAC response to RT, revealing persister cells populations with adaptations in lipid metabolism and ferroptosis defense that could be targeted to enhance RT efficacy. ST highlighted complex immune dynamics within the TME demonstrating a balance between inflammatory and immunosuppressive populations. Our findings provide a foundation for combinatorial therapeutic strategies and suggest that axon guidance pathways may also play an important role in the response to RT in PDAC. Abstract Background: Androgen receptor (AR) is a critical therapeutic target in prostate cancer (PC) and androgen blockade is known to act synergistically with radiotherapy (RT), however, the mechanisms through which AR modulates RT response is complex and remains poorly elucidated. Aims and Methods: We compared the radiation responses in LNCaP (AR-positive), PC3 (AR-negative), and HEK293T (non-malignant) cells by analyzing DSB induction, cell cycle arrest, and apoptosis. Mechanistic studies, including AR silencing and inhibition, were validated through RNA sequencing and IHC analysis of cell lines and irradiated patient-derived prostate tumors. Results: At 24-hours post-RT, the apoptotic rate increased by 6-fold in HEK293T and PC3 cells versus LNCaP cells. Cell viability at 48-hours post-RT was decreased to less than 10% in HEK293T, remaining at ∼60% in LNCaP cells. Moreover, over 70% of LNCaP cells were arrested in the G1 phase post-RT, accompanied by rapid p53 activation and p21 induction within 3-hours. AR silencing significantly increased radiosensitivity and reduced cell proliferation in LNCaP but did not alter the expression of p53 downstream genes, eg. BAX and p21. Interestingly, AR silencing reduced the phosphorylation of DNA-PKcs, a critical mediator of non-homologous end joining (NHEJ) repair, suggesting that AR facilitates NHEJ. This was further supported by the comet assay results showing that LNCaP repaired RT-induced DSBs more efficiently than PC3 cells. Moreover, AR regulates autophagy post-RT as AR silencing decreased autophagy-related gene (ATG) expression, and macroautophagy was augmented in LNCaP cells post-RT through LC3-II expression and autophagic vacuole formation. Bulk transcriptome analysis revealed significant upregulation of 11 ATGs, including ULK1 and ATG2, at 24-hours post-RT. Moreover, IHC analysis of patient-derived PC ex vivo irradiated tumors consistently showed that RT-induced autophagy supports the survival of AR-positive PC cells. Discussion and Conclusion: Our results demonstrate that AR plays an essential role in mediating radioresistance in AR-positive PC by enhancing both autophagy and DNA repair, with ongoing spatial transcriptome analysis to further explore these mechanisms in patient-derived tumors. Abstract Background: Cyclin-dependent kinase 4/6 inhibitors (CDK4/6i), in combination with endocrine therapy, are established as standard treatment for advanced HR+/HER2- breast cancer (BC). However, response heterogeneity is observed, with approximately one-third of patients exhibiting intrinsic resistance. This study investigates the mechanisms of variable responses to CDK4/6i through single-cell RNA sequencing of metastatic site biopsies, revealing distinct biomarkers for predicting CDK4/6i response. Methods: Single-cell RNA sequencing analyzed metastatic tumors from advanced HR+/HER2- BC patients pre-CDK4/6i treatment at baseline (BL) and/or at disease progression. BL samples were from CDK4/6i responders (n = 8, median progression-free survival [mPFS] = 17 months), while progressors were categorized as early-progressors (EP, n = 3, mPFS = 3 months) and late-progressors (LP, n = 8, mPFS = 10 months). Metastatic sites included liver (10), pleural effusions (6), ascites (2), and bone (1). InferCNV distinguished tumor cells, and functional analysis utilized the Molecular Signatures Database. Results: Both EP and LP tumor cells exhibited reduced estrogen response and increased IFN-γ pathways compared to BL tumors. LP tumors displayed enhanced Myc targets, EMT, TNF-α, and inflammatory pathways compared to EP tumors, revealing differential mechanisms of early versus late progression. Predictive signatures, including TFF3 and MGP, were significantly upregulated in BL tumor cells across different metastatic sites. Metastasis-specific markers included APOA and SERPINA1 (liver), and AGR2 (pleural effusion). BL and LP responders showed increased tumor infiltrating CD8+ T cell and NK cells compared to EP non-responders. Notably, despite a high CD8+ T frequency in responder tumors, functional analysis revealed a significant upregulation of genes associated with stress/apoptosis and exhaustion in proliferative CD4+ and CD8+ T cells from BL compared to LP tumors, including HSP90 and HSPA8, associated with poor response to immune checkpoint inhibitors. These findings suggest potential benefits of immunotherapy for CDK4/6i late progressors. Longitudinal biopsies further revealed dynamic NK cell expansion and enhanced cytotoxic T cell activity alongside the downregulation of stress/apoptosis gene expression at progression compared to BL tumors. Conclusion: This study underscores the significance of metastatic site-specific biomarkers in predicting outcomes to CDK4/6i. Tumor-infiltration lymphocytes may also serve as baseline predictors, while stress/apoptosis gene signatures suggest checkpoint inhibitor usage in CDK4/6i late progressors. These potentially actionable findings emphasize the need to optimize therapeutic approaches based on microenvironment-specific changes following disease progression. Cryopreservation of oocytes and embryos is very useful to conservation of animal genetic resources. Recently, parthenogenesis has received considerable attention as a tool for the production of stem cells. Oocytes and embryos undergo considerable morphological changes and functional damage during cryopreservation, and the survival rate is highly depending on species and developmental stage of oocytes and embryos. The aim of this study was to investigate the survival rate and embryonic quality of bovine parthenogenetic blastocysts post-vitrification cryopreservation. Cumulus-oocyte complexes (COC) from slaughterhouse ovaries were aspirated from 2-mm to 8-mm visible follicles with a 5-mL syringe. The COC were matured in vitro for 22 h in bicarbonate-buffered TCM199 media supplemented with 1 mg mL-1 of FSH, 10 mg mL-1 of LH, 1 mg mL-1 of 17-βiestradiol, and 10% FBS. After in vitro maturation, cumulus cells were removed from COC, oocytes with first polar body were activated by 5 μM ionomycin for 5 min and 2 mM 6-DMAP for 4 h. Subsequently, oocytes were co-cultured with bovine fetal fibroblast cells in SOF media supplemented with amino acids (1% NEAA and 2% EAA), 4 mg mL-1 of BSA, and 10% FBS at conditions of 38.5°C and 5% CO2 for 7 to 9 days. The good expanded blastocysts were selected and refrigerated in different vectors [glass micropipettes (GMP) and straws] and same vitrification solution (VS, 20% EG + 20% DMSO). Blastocysts were exposed to VS, loaded on vectors, and plunged into liquid nitrogen within 25 s. After two days refrigeration, vitrified blastocysts were thawed in air for 10 s and placed into 0.25 M sucrose solution for 1 min and 0.15 M sucrose solution for 5 min. Then, the blastocysts were cultured in the SOF medium same as above. Our results showed that when VS was 20% EG + 20% DMSO, the hatching rate (65%) of blastocysts loaded into GMP was significantly higher (P &lt; 0.05) than that (19%) of blastocysts loaded into straws post-vitrification. Meanwhile, vitrified and nonvitrified blastocysts were fixed and stained for differential cell counting as described by Thouas GA et al. 2001 Reprod. Biomed. Online 3, 25-29). By the differential staining, the total cells of nonvitrified parthenogenetic bovine blastocysts were 102.7, which was higher (P &gt; 0.05) than that (86.7) of vitrified blastocysts. Also, no significant difference (P &gt; 0.05) was seen on ratios between vitrified blastocysts (ICM/TE = 0.22) and nonvitrified blastocysts (ICM/TE = 0.25). Our results indicated that a glass micropipette vector was much better than a straw in vitrification cryopreservation of bovine parthenogenetic blastocysts and caused less damage to blastocyst cells. This study lays the foundation for further research to increase the survival rate of vitrification cryopreservation of bovine embryos. Abstract Background: In the metastatic setting, low HER2 expression is associated with clinical benefit from trastuzumab deruxtecan, a HER2-targeting antibody drug conjugates. However, little is known about the biological significance of low HER2 expression in patients with early stage triple-negative breast cancer (TNBC) receiving neoadjuvant therapy (NAT). Methods: Out of 595 patients with stage I-III TNBC enrolled on the prospective ARTEMIS trial (NCT02276443) from 2015-2021, we identified 367 patients with available HER2 immunohistochemistry (IHC) results on pre-NAT tumor tissue (HER2-zero: n=218; HER2-low [IHC 1+, 2+]: n=149). All patients were treated with anthracycline-based NAT. In cases where sufficient pre-NAT tumor tissue were available, additional IHC and/or RNAseq were performed. Differential gene expression (DGE) and pathway analysis were performed using DEseq2. Gene set enrichment analysis (GSEA) was performed using the Hallmark gene sets. Deconvolution analyses were performed using CIBERSORT. We controlled for multiple hypothesis using a false discovery rate (FDR) threshold with the Benjamini-Hochberg method, accepting as significant genes with at least a 2-fold change and &amp;lt; 5% FDR. Results: Table 1 summarizes baseline clinicopathological features of the 367 patients. Compared to HER2-zero tumors, HER2-low tumors were less likely of metaplastic histology (p=0.001), associated with lower Ki67 (p=0.017) and were more likely to be androgen receptor (AR)-positive (p=0.01). There were no significant differences in tumor-infiltrating lymphocytes (TILs) infiltration and PD-L1 expression between HER2-zero and HER2-low tumors. Among the 226 patients with sufficient pre-NAT tissue for RNAseq, DGE analyses demonstrated upregulation of genes involved in fatty acid metabolism (ACSM1) and steroid hormone metabolism (DHRS2, UGT2B28) in HER2-low tumors compared with HER2-zero tumors. Deconvolution analyses revealed no significant differences between predicted proportions of immune cell subpopulations between HER2-low and HER2-zero tumors. Although rates of pCR were not significantly different between patients with HER2-zero (46%) and HER2-low tumors (40%) (p=0.34), non-pCR in patients with HER2-low tumors was associated with increased expression of EREG, which encodes an EGFR ligand, while non-pCR in patients with HER2-zero tumors was associated with downregulation in genes involved in immune response pathways. GSEA further identified the Hallmark allograft rejection (FDR q=0.001), interferon gamma response (FDR q=0.002), and interferon alpha response pathways (FDR q=0.007) as the 3 most significantly downregulated pathways in HER2-zero tumors from patients experiencing a non-pCR relative to HER2-zero tumors from patients experiencing a pCR. Conclusion: In early stage TNBC, low HER2 expression is associated with increased AR expression and upregulation of genes associated with fatty acid and steroid hormone metabolism. Gene expression analyses suggest that drivers of resistance to NAT differ between HER2-low and HER2-zero tumors. Biological differences between HER2-zero and HER2-low tumors exist and may influence future personalized treatment for patients with early stage TNBC.
