Abstract:

Abstract:
We investigated the taxonomic relationships among Streptomyces hygroscopicus subsp. glebosus , Streptomyces libani subsp. rufus and Streptomyces platensis . The three species formed a single clade in the phylogenetic trees based on 16S rRNA gene sequence and multilocus sequence analyses. Digital DNA–DNA hybridization using whole genome sequences suggested that S. hygroscopicus subsp. glebosus , S. libani subsp. rufus and S. platensis belong to the same genomospecies. Previously reported phenotypic data also supported this synonymy. Therefore, S. hygroscopicus subsp. glebosus and S. libani subsp. rufus should be reclassified as later heterotypic synonyms of S. platensis .

Abstract:
The charcoal disease agents, Biscogniauxia mediterranea and Obolarina persica are two latent, ascomycetous oak pathogens in the Middle Eastern Zagros forests, where they have devastating effects, particularly during drought. Under greenhouse conditions, we investigated the effects of the two charcoal disease agents individually and in combination with drought on survival, growth, foliar gas-exchange, pigment content, oxidative stress and the antioxidant response of Quercus infectoria and Q. libani, two of the dominant tree species in this region. Commonly, the strongest negative effects emerged in the drought–pathogen interaction treatments. Q. infectoria showed less severe lesions, higher survival, more growth, and less leaf loss than Q. libani under combined biotic and abiotic stress. In both oak species, the combination of pathogen infection and drought resulted in more than 50% reduction in foliar gas-exchange parameters with partial recovery over time in Q. infectoria suggesting a superior defense system. Indeed, enhanced foliar anthocyanin, total soluble protein and glutathione concentrations imply an upregulation of the antioxidant defense system in Q. infectoria under stress while none of these parameters showed a significant treatment response in Q. libani. Consequently, Q. infectoria foliage showed no significant increase in superoxide, lower lipoxygenase activity, and less electrolyte leakage compared to the highly elevated levels seen in Q. libani indicating oxidative damage. Our findings indicate greater drought tolerance and pathogen resilience in Q. infectoria compared to Q. libani. Under future climate scenarios, we therefore expect changes in forest community structure driven by a decline in Q. libani and closely associated organisms.

Abstract:
Essential oil composition, antimicrobial activity and flavonoid contents of leaf-fruits of Cedrus libani and Pinus nigra subsp. pallasiana were determined with GC-MS, disc diffusion method and HPLC in three different period. When the essential oil composition of leaf and fruits of P. nigra ssp pallasiana collected in April, July and September, major components were α-pinene, β-pinene, limonene, β-caryophyllene ve germacrene-D. On the other hand, additionally to these contents, myrcene and α–terpineol were also detected in C. libani. According to HPLC analysis, rutin, quercetin, kaempferol, naringin and resveratrol flavonoids were detected in different proportions. While rutin (154.33 µg g-) and resveratrol (20.02 µg g-) has the highest ratio in C. libani, quercetin (9.65 µg g-) and naringin (9.31 µg g-) were detected in P. nigra subsp. pallasiana along with rutin (39.66 µg g-). According to the antimicrobial activity results the essential oils of C. libani obtained in April has produced higher activity than that of July and September. On the contrary, the essential oils from P. nigra subsp. pallasina have produced the best antimicrobial activity on September compared to April and July. As a result, C. libani and P. nigra subsp. pallasiana essential oils have a composition showing antimicrobial activity and their harvesting season should be determined for the best and effective content.

Abstract:
The green oak leaf roller, Tortrix viridana L., is one of the most destructive pests, causing damages to various species of oak, feeding on different host plants including Quercus infectoria Oliv, Q. branti Lindl, and Q. libani Oliv. Nutritional indices, activity of enzymatic and non-enzymatic compounds of T. viridana were studied under laboratory conditions. In addition, chemical components were analysed in the leaves of the three host plants. Fourth instar larvae reared on Q. branti showed the highest values of relative consumption rate (RCR), approximate digestibility, and consumption index (48.73±6.22; 90.45±1.06 and 97.45±12.44 respectively), while the lowest values were observed on Q. libani. Efficiency of conversion of ingested food in the fourth instar larvae was the highest (3.17±0.661) on Q. libani and the lowest (1.53±0.164) on Q. branti. The fifth instar larvae fed on Q. libani had the highest RCR (15.64±2.51). The highest amounts of triglycerides, uric acid, glucose, protein and the lowest activity of alkaline phosphatase were observed in the fifth instar larvae reared on Q. libani. The leaves of Q. libani highlighted the highest amounts of total nitrogen, total protein, water, potassium, magnesium and total carbohydrate. The present research suggested that the nutritional quality of the host plants have crucial effects on T. viridana larvae.

Abstract:

Abstract:
Lebanon cedar (Cedrus libani A. Rich) is an ecologically, economically, and historically important conifer species that currently mainly occurs in the Taurus Mountains in southern Turkey. In former times, extensive forests of this species were also found in Syria and Lebanon. However, because of intensive cutting, burning, and goat grazing, only small populations are left in these countries. Currently, the range of Lebanon cedar covers approximately 600,000 ha in Turkey, including extremely degraded stands and bare karstic land that was previously covered by this species (1). Therefore, efforts to protect existing forests and promote natural regeneration of this endangered tree species were undertaken in recent years. In addition, reforestations were carried out on bare karstic lands to expand the population of Lebanon cedar in Turkey. During disease surveys, carried out in October 2009 in the Mt. Dedegül Region of the western Taurus Mountains (37°36′54″N, 31°20′00″E), a dieback of lower branches and young plants of C. libani was observed at 1,700 to 1,885 m above sea level. The disease often occurred in scattered patches and was most evident near the timberline. Needles, shoots, and twigs of affected trees or entire small trees were covered or completely enmeshed in silky, shining, blackish brown mycelial felts. Symptoms resembled those of brown felt blight, also known as black snow mold, caused by Herpotrichia juniperi and Neopeckia coulteri on various other conifer species (2). For fungal isolation and identification, 18 twig samples from 14 different C. libani trees were collected. Two colonized needles from each twig were transferred to water agar (16 g liter–1 of agar and 0.1 g liter–1 of streptomycin) and incubated at 4°C for at least 8 days in the dark. Single hyphal-tip cultures were then established from only one of the developing colonies per twig and transferred to 1.5-ml microcentrifuge tubes containing 500 μl of potato dextrose broth. DNA extraction, directly from the mycelium, was performed after 20 days (3). DNA was amplified using primer pair ITS1 and ITS4 (4) and sequenced. Sequences of two representative fungal isolates from C. libani were deposited in GenBank (HM853976 and HM853977). Comparison of the 18 internal transcribed spacer sequences obtained from C. libani showed 99 to 100% nucleotide identity with those of reference strains of H. juniperi (2) from GenBank and variation among the 18 sequences was <1%, which is within the limits reported in a previous study (2). To our knowledge, this is the first report of C. libani as a new host of H. juniperi. Thus, brown felt blight is considered to have a significant impact on regeneration of C. libani as well as on the survival and growth of seedlings and young trees in the study area. AbstractGas chromatography–mass spectrometry analysis together with principal component analysis revealed that geographical origin influenced the yield and composition of the essential oils (EOs) extracted by hydrodistillation performed for 3 h using a Clevenger-type apparatus, from the cones of Cedrus libani A. Rich., growing wild at four Lebanese natural reserves and protected areas: Bsharri, Chouf, Ehden, and Tannourine, and from a cultivated cedar growing in Qartaba. Essential oil chemical variability established between the different studied provenances suggested the involvement of abiotic factors such as geographical conditions, cultivation conditions, soil composition, and environmental factors in the chemical polymorphism of C. libani cones EOs. α-Pinene/β-pinene characterized Ehden (β-pinene 35.6%/α-pinene 27.7%), Chouf (α-pinene 37.3%/β-pinene 26.1%), Bsharri (α-pinene 27.7%/β-pinene 21.4%), and Tannourine (α-pinene 25.1%/β-pinene 16.0%) samples, whereas Qartaba EO was distinguished by the dominance of myrcene (30.6%), α-pinene(26%), and limonene (14.1%). Comparison with the existing literature reinforced the chemical variability of C. libani EOs. This current study helped the estimation of a best harvest location for a good EO quality production, resource optimization, and pharmacological properties evaluation, according to the market demand.

Abstract:
Abstract Ancient Taurus cedar (Cedrus libani A. Rich) wood samples from the Tumulus of King Midas at the Gordion archaeological site (about 2700 years old), near Ankara, Turkey, and from the Al-Aksa Mosque (about 1500 to 1900 years old), Jerusalem, Israel, were characterized by studying the sequences of ribosomal DNA (rDNA) internal transcribed spacers (ITS1 and ITS2). After extraction of the DNA, the ITS regions were amplified utilizing the polymerase chain reaction, followed by sequencing, BLAST searches for similar sequences, and phylogenetic analyses. Fifty-six sequences were obtained. In BLAST searches of existing sequence databases, most were closest to those from humans and fungi. However, two sequences exhibited similarities with conifer ITS sequences. One was an ITS1 region from the Gordion wood specimen, and the other one was an ITS2 region from the Al-Aksa wood specimen. Phylogenetic analyses indicated that both were closest to Taurus cedar (C. libani, also known as Lebanon cedar) ITS sequences from three recent samples of Taurus cedar from two sites in Turkey. However, they exhibited many differences from the recent C. libani rDNA ITS sequences from Turkey, probably due to degradation of the DNA in the ancient samples. The implications of the results on future studies are discussed.

Abstract:

Abstract:
We have compared the quality of Ba2YCu3O7−δ (BYCO) films grown by the BaF2 process on (100) substrates of the perovskites LaAlO3, LaGaO3, NdAlO3, NdGaO3, LiBaF3, and SrTiO3. The films were grown by coevaporation of Y, Cu, and BaF2 followed by a two-stage anneal. The high temperature stage of the anneal, the part of the process during which the BYCO crystal structure and morphology develop, has been varied. LaAlO3 and SrTiO3 support much better films, both electrically and structurally, than the other substrates. Of the oxides, NdGaO3 supports the worst films, while films on LiBaF3 are nonconducting. These results emphasize the overriding importance of chemical compatibility in determining the suitability of a potential substrate material. Unless this criterion is satisfied, the issue of lattice matching is unimportant.
