Abstract:
ABSTRACT Sequence analysis of the Lymantria dispar multicapsid nucleopolyhedrovirus (LdMNPV) genome identified an open reading frame (ORF) encoding a 548-amino-acid (62-kDa) protein that showed 35% amino acid sequence identity with vaccinia virus ATP-dependent DNA ligase. Ligase homologs have not been reported from other baculoviruses. The ligase ORF was cloned and expressed as an N-terminal histidine-tagged fusion protein. Incubation of the purified protein with [α-32P]ATP resulted in formation of a covalent enzyme-adenylate intermediate which ran as a 62-kDa labeled band on a sodium dodecyl sulfate-polyacrylamide gel. Loss of the radiolabeled band occurred upon incubation of the intermediate with pyrophosphate, poly(dA) · poly(dT)12–18, or poly(rA) · poly(dT)12–18, characteristics of a DNA ligase II or III. The protein was able to ligate a double-stranded synthetic DNA substrate containing a single nick and inefficiently ligated a 1-nucleotide (nt) gap but did not ligate a 2-nt gap. It was able to ligate short, complementary overhangs but not blunt-ended double-stranded DNA. In a transient DNA replication assay employing six plasmids containing the LdMNPV homologs of the essential baculovirus replication genes, a plasmid containing the DNA ligase gene was neither essential nor stimulatory. All of these results are consistent with the activity of type III DNA ligases, which have been implicated in DNA repair and recombination.

Abstract:
Abstract Liver X receptor (LXR) is a ligand-activated transcription factor that plays important roles in cholesterol and lipid homeostasis. However, ligand-induced posttranslational modification of LXR is largely unknown. Here, we show that ligand-free LXRα is rapidly degraded by ubiquitination. Without ligand, LXRα interacts with an ubiquitin E3-ligase protein complex containing breast and ovarian cancer susceptibility 1 (BRCA1)-associated RING domain 1 (BARD1). Interestingly, LXR ligand represses ubiquitination and degradation of LXRα, and the interaction between LXRα and BARD1 is inhibited by LXR ligand. Consistently, T0901317, a synthetic LXR ligand, increased the level of LXRα protein in liver. Moreover, overexpression of BARD1/BRCA1 promoted the ubiquitination of LXRα and reduced the recruitment of LXRα to the target gene promoters, whereas BARD1 knockdown reversed such effects. Taken together, these data suggest that LXR ligand prevents LXRα from ubiquitination and degradation by detaching BARD1/BRCA1, which might be critical for the early step of transcriptional activation of ligand-stimulated LXRα through a stable binding of LXRα to the promoters of target genes.

Abstract:
The zebrafish gene, mind bomb (mib), encodes a protein that positively regulates of the Delta-mediated Notch signaling. It interacts with the intracellular domain of Delta to promote its ubiquitination and endocytosis. In our search for the mouse homologue of zebrafish mind bomb, we cloned two homologues in the mouse genome: a mouse orthologue (mouse mib1) and a paralogue, named mind bomb-2 (mib2), which is evolutionarily conserved from Drosophila to human. Both Mib1 and Mib2 have an E3 ubiquitin ligase activity in their C-terminal RING domain and interact with Xenopus Delta (XD) via their N-terminal region. Mib2 is also able to ligate ubiquitin to XD and shift the membrane localization of Delta to intracellular vesicles. Importantly, Mib2 rescues both the neuronal and vascular defects in the zebrafish mibta52b mutants. In contrast to the functional similarities between Mib1 and Mib2, mib2 is highly expressed in adult tissues, but almost not at all in embryos, whereas mib1 is abundantly expressed in both embryos and adult tissues. These data suggest that Mib2 has functional similarities to Mib1, but might have distinct roles in Notch signaling as an E3 ubiquitin ligase.

La ligne a ailettes represente beaucoup d'avantages par rapport aux guides d'ondes classiques (guide rectangulaire, guide circulaire, ligne microruban, etc. . . ). Ces avantages sont lies principalement a sa largeur de bande passante, a l'absence des pertes de rayonnement et a sa compatibilite avec les guides rectangulaires. Nous nous sommes specialement interesses aux filtres passe-bande realises sur cette ligne. L'obstacle inductif sur cette ligne permet de concevoir des inverseurs d'impedances qui sont utilises dans la synthese des filtres microondes passe-bande. Pour cela, dans la premiere partie de ce travail, nous avons elabore des formules analytiques du schema electrique equivalent de cet obstacle qui sont indispensables pour la conception assistee par ordinateur (c. A. O. ). Ceci facilite le transfert de cette technologie ligne a ailettes vers l'industrie. Dans la deuxieme partie, nous avons etudie des structures de lignes a ailettes a fentes couplees. Dans un premier temps, nous avons modelise et caracterise la discontinuite (court-circuit) aux extremites des fentes. Cette caracterisation nous a permis de concevoir des inverseurs d'impedances a deux fentes couplees et, par la suite, de proposer une methode de synthese des filtres microondes passe-bande du type tchebyscheff. Les resultats experimentaux nous revelent un bon accord avec la theorie. L'etude de la variation des reponses de ces filtres avec les dimensions de la structure montrent qu'ils sont peu sensibles aux variations de ces dimensions. Ceci represente un grand avantage pour ce type des filtres et les rend plus performants que ceux concus avec des obstacles inductifs

Abstract:
MDM2, murine double minute 2, is a RING type-E3 ligase protein and also an oncogene. MDM2 plays a critical role in determining the steady levels and activity of p53 in cells using two mechanisms. The N-terminal domain of MDM2 binds to the transactivation domain of p53 and inhibits its transcriptional activity. The RING domain of MDM2 plays a role in the ubiquitination (and degradation) of p53. Several proteins are responsible for the ubiquitination mechanism including the ubiquitin-activating enzyme (E1), ubiquitin-conjugating enzyme (E2) and ubiquitin ligase (E3). Since the E2-E3 interaction is essential for ubiquitination, the protein-protein recognition site is a potential drug target. Two different MDM2 RING constructs were expressed and purified: MDM2RING (residues 386-491) and MDM2RING△C (residues 386-478). Both constructs were characterised using dynamic light scattering, size exclusion chromatography, mass spectrometry, NMR and electron microscopy. E3 ligase activity in vitro was also studied. Taken together these results showed that the MDM2RING construct formed a concentration-dependent oligomeric structure. In contrast, the MDM2RING△C construct formed a dimer at all concentrations. Both MDM2RING and MDM2RING △ C retain E3 ligase activity. However, the MDM2RING△C construct is less active. Full length E2 enzyme UbcH5a was also purified. Various biophysical techniques were used to study its interaction with MDM2 as well as with potential small molecule inhibitors as in principle, small molecules which disrupt the interaction between MDM2 and UbcH5a, could prevent/promote ubiquitination of p53. The dimerisation of MDM2 is important for its E3 activity and the C8-binding site potentially provides a second druggable site. In this work, peptide 9, which has the same sequence as the C-terminus of MDMX (an MDM2 homologue) was found to inhibit MDM2 E3 activity. Various biological techniques including NMR, fluorescence anisotropy, and electrospray mass spectrometry were used to investigate the interaction between two inhibitory peptides and MDM2. A major part of project involved virtual screening (VS) to search for small molecules which can affect MDM2-dependent ubiquitination. Three potential targets were considered: (1) the C8-binding site of MDM2; (2) the UbcH5a-binding site of MDM2; and (3) the MDM2-binding site of UbcH5a. Several small molecules were identified using our virtual screening database-mining and docking programs that were shown to affect MDM2-dependent ubiquitination of p53. In terms of understanding the complex biochemical mechanism of MDM2 this work provides two interesting and functionally relevant observations: (i) the MDM2 RING△C construct is a dimer as this would not be expected form the existing studies, and has less E3 ligase activity than MDM2RING; (ii) small molecules that bind MDM2 on the E2 binding site enhanced E3 ligase activity. One model to explain these observations is that binding of small molecule activators family to the RING induces a change in the conformation of the Cterminal tail residues which may enhance E2 binding.

Abstract:
Cette thèse a pour objectif l'étude et l'optimisation de quelques techniques de réduction de bruit impulsif de type asynchrone pour transmission large bande sur courants porteurs en lignes (PLC) en contexte résidentiel. En effet, outre les propriétés physiques assez particulières qui caractérisent ce type de canal de transmission, le bruit impulsif asynchrone demeure la cause principale de perte drastique de débit en PLC. Pour pallier à des dégradations de cette nature, les techniques dites de retransmission sont très souvent utilisées. Bien que robustes aux perturbations de nature impulsive, ces techniques de retransmission conduisent inévitablement à une réduction de débit et à l'introduction de délais de traitement supplémentaires pouvant être critiques pour des applications temps réel. D'un autre côté, plusieurs solutions alternatives sont proposées dans la littérature pour contrecarrer les effets nocifs du bruit impulsif sur les transmissions en PLC. Cependant, seules très peu d'entre elles présentent une complexité acceptable pour un récepteur PLC. En particulier, les techniques d'écrêtage et d'effacement sont d'une efficacité remarquable dans la mesure où elles offrent un très bon compromis entre complexité et amélioration des performances en présence de bruit impulsif. Dans cette thèse, une optimisation de la détermination du seuil de chacune des deux non-linéarités, qui se fait très souvent de manière empirique, est réalisée de manière analytique. Des structures adaptatives de réduction de bruit impulsif asynchrone sont également proposées au niveau du récepteur PLC. D'autre part, compte tenu de ses très bonnes propriétés de sélectivité fréquentielle et de son gain en efficacité spectrale par rapport à l'OFDM avec préfixe cyclique (CP-OFDM), l'OFDM/Offset QAM (OFDM/OQAM) peut être vue comme une modulation multiporteurse alternative pour les transmissions haut débit sur PLC. Cependant, aucune étude ne s'est réellement intéressée aux performances d'une telle modulation en environnement impulsif. Dans cette thèse, une analyse du comportement d'un système OFDM/OQAM en présence de bruit impulsif asynchrone est effectuée. Une évaluation théorique de l'expression du rapport logarithmique de vraisemblance (LLR) pour ce type de modulation, en transmission codée, est également proposée. Les performances en terme de taux d'erreur binaire sont évaluées en environnement PLC réalistes et comparées avec celles obtenues en CP-OFDM codé. Enfin, un nouveau point de vue sur une diversité supplémentaire de l'OFDM/OQAM est présenté
This thesis aims at improving some asynchronous impulsive noise reduction techniques for broadband transmissions over indoor power lines. Indeed, beside the particular physical properties of this type of medium, asynchronous impulsive noise remains one of the main causes of performance degradation in powerline communications (PLC). Such an impairment is usually coped with retransmission strategies at the price of a spectral efficiency loss. Although several other countering strategies are proposed in the literature, only very few of them exhibit a reasonable complexity. In particular, the clipping and the blanking approaches present a very good trade-off between the impulsive noise mitigation and the complexity of the implementation. In this thesis, an optimization of the threshold determination, which is usually empirically done, is performed analytically both for the clipping and the blanking non-linearities. Moreover, some adaptive receiver structures for asynchronous impulsive noise reduction over PLC are proposed. On the other hand, because of its good frequency selectivity features and its spectral efficiency gain compared to cyclic-prefix OFDM (CP-OFDM), the OFDM/Offset QAM (OFDM/OQAM) can be seen as an alternative multicarrier modulation scheme for transmission over power lines. Since no study was dedicated to analyze such a modulation in an impulsive environment, this thesis investigates the performance behavior of an OFDM/OQAM modulated system corrupted by asynchronous impulsive noise. Furthermore, a thorough Log-Likelihood Ratio (LLR) computation is derived for coded OFDM/OQAM transmissions under realistic PLC conditions, and the performance results are compared with those obtained with coded CP-OFDM. Finally, a new diversity of the OFDM/OQAM modulation is pointed out in this work

Cette étude présente sous un angle algorithmique, diverses enquêtes et réflexions concernant la ligne et l'analyse dans la création artistique. Dans la première partie - ligne, nous discutons d'abord la ligne dans différents domaines : mathématiques, ingénierie et beaux-arts. Nous interprétons ensuite le dessin linéaire et le concept de la ligne dans l'art moderne d'un point de vue algorithmique. Enfin, des caractères algorithmiques de la ligne dans la peinture chinoise sont exposés. Dans la deuxieme partie - algorithme, après une brève introduction des principes généraux de l'algorithme et des algorithmes de génération des lignes, nous étudions l'évolution de l'art assiste par ordinateur par rapport au développement des algorithmes. Dans la troisième partie - analyse, nous proposons le langage postscript pour décrire des formes de façon quantitative et algorithmique, et autolisp pour l'analyse interactive des formes et de la composition. En référant à "point et ligne sur plan" de Kandinky, nous écrivons une bibliothèque de prototype des formes élémentaires par postscript, et interprétons des passages de Kandinsky sur les formes dans cet ouvrage. This study presents various surveys and reflections regarding the line and the analysis in artistic creation from an algorithmic viewpoint. In parti - line, we discuss first the line in different domains : mathematics, engineering and fine arts. Then we interpret the line drawing and the concepts of line in modern art from an algorithmic viewpoint. Finally, some algorithmic characters of line in chinese painting are presented. In part ii - algorithm, after a brief introduction of general principles of algorithm and some algorithms for line generation, we study the evolution of the computer art with regard to the development of algorithms. In part III - analysis, we propose the language postscript for describing the forms in the way quantitative and algorithmic, and autolisp for interactive analysis of form and composition. Referring to kandinsky's "point and line to plane", we write a prototype library of elementary forms by postscript, and interpret some passages of kandinsky concerning the forms in this book. We also show how to analyze the composition of painting by use of shape grammars

Abstract:
La foudre par ses effets directs et indirects peut engendrer des dégâts importants sur un réseau de télécommunications. Cette thèse aborde la modélisation des mécanismes de couplage d'une décharge orageuse sur une structure constituée d’un réseau filaire et d’un bâtiment. A l’aide d'un code de calcul basé sur la théorie des lignes de transmission et associé à une approche topologique, l’étude de l'efficacité d'une protection contre la foudre, réalisée grâce à un conducteur nu placé au dessus d'un câble enterré, est effectuée de manière exhaustive en s'appuyant sur des résultats expérimentaux obtenus à l'aide d'éclairs déclenchés. De plus, la thèse aborde, à l’aide de la théorie des antennes dans le domaine temporel (FDTD), le calcul des courants induits simultanément sur un bâtiment et son réseau d'adduction lors d'un impact direct sur un pylône. L'application pratique visée par l'ensemble de ces travaux est l'amélioration de la protection contre la foudre des sites de radiodiffusions
Lightning by its direct and indirect effects may engender important damages on a telecommunications network. This thesis approaches the modeling of the coupling of a lightning stroke with a structure composed of a building associated to a wired network. By means of a formalism based on the transmission line theory associated to a topological approach, a study of the efficiency of a lightning protection device, composed of a shielding wire installed in the ground above the underground cable, is made in a exhaustive way by using experimental data obtained by means of triggered lightning. Furthermore, thanks to the antenna theory in the time domain (FDTD), the thesis also approaches the calculation of the currents generated simultaneously on a building and its wired network when the lighting stroke occurs on the considered structure. The practical application aimed by all these works being the improvement of the lightning protection of the radio-sites

Les technologies offrent des possibilités de rendre l'enseignement et l’apprentissage plus efficaces et plus appropriés et nul ne peut nier les potentiels bénéfices de la révolution numérique dans l'enseignement des langues Une nouvelle forme d’enseignement des langues à distance attire de plus en plus d’apprenants : les MOOCs (Massive Open Online Course) en français, CLOMs (Cours en Ligne Ouverts et Massifs). Sans contrainte de niveau d’études, ni d’assiduité, le MOOC permet d’offrir un apprentissage en ligne et à distance avec un suivi pédagogique spécifique et des ressources pédagogiques (vidéos, documents, devoirs…) accessibles à tout moment. Le MOOC FOFLE (Formation Ouverte en Français Langue Etrangère) est un projet pilote, international et interdisciplinaire, dédié à l'apprentissage en ligne du français. Il a été expérimenté en Tunisie : avant d'être déployé à destination des autres pays francophones présentant la même problématique. Abstract:
The remodeling of the cytoskeleton and focal adhesion distributions for cells on substrates with micro-patterned ligand patches is investigated using a bio-chemo-mechanical model. All the cells have approximately the same area and we investigate the effect of ligand pattern shape on the cytoskeletal arrangements and focal adhesion distributions. The model for the cytoskeleton accounts for the dynamic rearrangement of the actin/myosin stress fibers and entails the highly non-linear interactions between signaling, the kinetics of tension-dependent stress-fiber formation/dissolution and stress dependent contractility. This model is coupled with a focal adhesion (FA) model that accounts for the mechano-sensitivity of the adhesions from thermodynamic considerations. This coupled stress fiber and focal adhesion model is shown to capture a variety of key experimental observations including: (i) the formation of high stress fiber and focal adhesion concentrations at the periphery of circular and triangular, convex–shaped ligand patterns; (ii) the development of high focal adhesion concentrations along the edges of the V, T, Y and U shaped concave ligand patterns; and (iii) the formation of highly aligned stress fibers along the un-adhered edges of cells on the concave ligand patterns. When appropriately calibrated, the model also accurately predicts the radii of curvature of the un-adhered edges of cells on the concave-shaped ligand patterns.

Abstract:
As a useful flotation agent, the xanthate ligand, O-alkyldithiocarbonates, has been used by different countries by its easy and inexpensive synthesis. More recently papers explored many different applications using this ligand within a complex of several metals cation. In order to study the proprieties of the lead (II) complex with such ligand, the object of this work is to provide a better understanding of the Pb-S bond using different theoretical approaches as NBO, EDA and QTAIM analysis and the influence caused by the different alkyl groups of the ligand. By an optimized structure, the NBO showed that the Pb-S is mainly composed by p orbital of the lead and by the p lone pair of the sulfur atom. The calculation with different alkyl groups highlights that the presence of a larger hydrocarbon chain provides a higher contribution of the s orbital of the lead atom to the interaction. Through the EDA analysis, the interaction between ligand and metal has the predominance of an electrostatic character. The size of the alkyl group has an impact on the value of both covalent and electrostatic character, making the interaction more covalent, due to a higher presence of an electronic density on sulfur atom. This density can be evaluated by the topological study of the QTAIM analysis, which enhances the fact that the charge over the sulfur atom gets higher when using a larger alkyl group for the xanthate ligand.

Abstract:
To facilitate analysis of nanoscale heat transfer in nanoparticle systems the thermal properties of ligand layers must be understood. To this end, we use an optical pump-probe technique to study the thermal transport across ligands on gold nanorods and into the solvent. We find that varying properties of the ligand can have large impacts on the thermal decay of a nanorod after exposure to a laser pulse. By raising the concentration of free CTAB from 1 mM and 10 mM in solutions, the CTAB layer’s effective thermal interface conductance increases three fold. The transition occurs near the CTAB critical micelle concentration. Similar results are found for other ligand layers.
