Full textThe barrier function of the skin is related to the unique composition of the stratum corneum (SC) lipids and to their complex arrangement. The aim of this work consists in developing vibrational descriptors allowing to assess the supramolecular organization of the stratum corneum lipids. Two classes of Raman descriptors were identified: conformational descriptors of alkyl chains and organizational descriptors describing the lateral packing of lipid structures: orthorhombic, hexagonal or disordered state. Using these descriptors, we were able to highlight differences between conformations and organization states of SC lipids in vitro, ex vivo and in vivo. This preliminary work gives the possibility to explore new perspectives of in vivo studies. Especially by correlating the supramolecular organization of the SC lipids with the barrier function state in a given population in order to be able to develop personalized transcutaneous formulations

Full textThe objective of this work was to find a lipolytic agent, that could be used as a slimming agent in cosmetic products. Our research aimed at finding mechanisms which had been not extensively studied and where lipolysis was always inhibited. The literature review has helped us to identify a molecule which could match with these requirements: cirsimarine. The initial steps of this work were: finding raw materials, developing extraction and purification methods for cirsimarine. The experiments performed showed that microtea debilis swartz, is the best source of cirsimarine. Then the effects of the vegetal extract and its flavone on lipidic metabolism were investigated. As extracellular matrix changes with adipocytes hypertrophy, the impact of the vegetal extract on components of this matrix was also studied. In addition, the antioxidant effect of cirsimarine was investigated as shown for a lot of flavonoids

Full textThe skin is a frontier between the environment and our organism. The stratum corneum, which is the outmost layer of the epidermis, is composed of dead cells embedded by a compact lipidic matrix. These supramolecular organisation give to the stratum corneum its function of barrier, which protect our body from the penetration of exogeneous molecules and extrernal attacks such as UV irradiation. Uv are very dangerous for health and can be at the origin of Inflammation, photo-ageing and cancers. Our study, thank to different analytic tool, permited to follow th chemical and structural evolution of the stratum corneum lipidic matrix, with natural photo-ageing and photo-ageing induced by UV irradiation. The caracterisation of the chemical composition of the lipidic matrix was realised by HPLC. The caracterisation at both molecular and supramolecular levels was realised by mass spectrometry and FTIR spectroscopy. The use of these techniques lead to a correlation, in vitro on films, between the formation of oxidative entites on unsaturated lipids, and the conformation of the lipids, thank to IR conformational descriptors

Full textMany nanocarriers have been developed to improve the delivery of molecules into the skin. In this PhD thesis, we are interested in lipid-based Janus nanoparticles (JNP), an innovative galenic form characterized by the combination of two compartments of opposite chemical polarity, an aqueous compartment associated to a lipid compartment. The main aim was the characterization of JNP. ATR-FTIR spectroscopy allowed to identify an infrared descriptor to follow the physical stability of JNP in open air and over time. The same descriptor allowed to follow their behavior on the surface of the skin, and to note a significant penetration from 3 hours of application. AFM-IR has been shown to be a promising technique for studying the nanostructure of the human skin. In addition, it has shown that after 24 hours of application, JNP were accumulated in the first layers of the SC with a gradient in the deeper layers of the SC. However, it was not possible to conclude if they have penetrated in the intact or degraded form. JNP seem to have an influence on the cutaneous penetration of the hyaluronic acid, they allowed a significant increase of its penetration flux. The characterization of the lipophilic phase of JNP by different techniques (LC-MS, DLS, Cryo-TEM, X-ray diffraction...) allowed to better understand their instability at high temperatures (32°C - 43°C)

Full textThe aim of this work was to develop and optimize methods for fragrance mix I (FMI) encapsulation into nanoparticles (NPs) of two types of nanoparticles (NPs) : polymeric NPs (poly-ε-caprolactone, PCL) and solid lipid NPs (SLNs) (prepared with petrolatum, shea butter, candelilla wax, C10-18 triglycerides, or cetyl palmitate). Then, these new NPss were evaluated as vectors through a pig skin to analyze the distribution of the FMI molecules in the different skin layers. In parallel, NPs have also been applied as solubilizers for the development of a new in vitro test for the diagnosis of fragrance allergy. Our results show that (i) NPs polymers, mainly anionic NPs, are the most suitable vectors to promote trans-epidermal penetration of fragrance. On the contrary, SLNs were found in the stratum corneum, leading to an accumulation of fragrance in this layer; (ii) whatever the type of NPs, the penetration of the FMI molecules in the deeper layers of the skin depends on their intrinsic partition coefficient; (iii) PCL-NPs significantly increase the FMI solubilization in conventional culture media and, allowing a robust reactivation of circulating specific T cells in patients with allergy to fragrances. All of these results confirm the potential of organic NPs for the development of future strategies (for the skin delivery of several actives in the different skin layers). These new vectors further offer a promising alternative to improve the diagnosis of contact dermatitis induced by fragrances and more generally by hydrophobic allergens
