Thesis (MTech (Biomedical Technology))--Cape Peninsula University of Technology, 2015.<br>Diabetes mellitus (DM) is an endocrine disorder that is characterised not only by severe hyperglycemia but also altered metabolism of glucose and lipids. It is a major health problem worldwide and its impact is greatly noticed in developing countries due to the lack of adequate medical facilities. Oxidative stress remains the principal factor that actively plays major roles in the onset and progression of diabetes mellitus and its complications. The use of medicinal plants in the treatment of DM has undisputedly gained the attention and interest of researchers throughout the globe mainly because plants have established promising outcomes in the treatment of diabetes. It is evident that the plants’ constituents possess therapeutically potent metabolites that have beneficial effects such as antioxidant, antidiabetic, anticancer, anti-inflammatory and antibacterial activities. Hypoxis hemerocallidea is a native plant that grows in the Southern African regions. H. hemerocallidea is well known for its beneficial medicinal values. In South Africa it is known as the African potato. The main aim of this study was to investigate both the beneficial and also the possible toxic effects of H. hemerocallidea in the kidney and liver tissues of streptozotocin-induced diabetic male Wistar rats by assessing the antioxidant status and selected biochemical parameters in the two studied organs. Diabetes was induced in overnight fasted rats by administration of a single intraperitoneal injection of STZ at a dosage of 50mg/kg in citrate buffer (0.1 M at 4.5 pH). Hyperglycemia was confirmed 72 hours after induction of diabetes using STZ in rats with glucose levels > 15 mmol/l. Treatment with the plants extract commenced on the fourth day after STZ administration via gastric gavage that was done once a day over a 6 week period. The effects of H. hemerocallidea on glucose, body weight, liver and kidney weights, liver function, kidney function and the oxidative status were evaluated after the feeding period.

Thesis (PhD(FoodSc))--Stellenbosch University, 2012.<br>ENGLISH ABSTRACT: Potential beneficial and/or adverse modulatory effects of polyphenol-enriched extracts of rooibos (Aspalathus linearis) and honeybush (Cyclopia spp.) on the antioxidant homeostasis in the liver were investigated. Phase II metabolism of aspalathin and mangiferin, the major polyphenols of rooibos and honeybush respectively, was assessed for potential glucuronidation and sulphation. Glucuronidation resulted in a loss of antioxidant activity for aspalathin and mangiferin in post-column HPLC-DAD-DPPH• and HPLC-DAD-ABTS•+ assays and also a decreased activity of iron chelating properties of mangiferin in the FRAP assay. Two independent studies for 28 and 90 days with polyphenol-enriched extracts (PEEs) of unfermented rooibos [Aspalathus linearis (PER)] and honeybush [Cyclopia. subternata (PECsub) and C. genistoides (PECgen)] in male Fischer rats were conducted to assess possible beneficial and/or adverse biological effects. PECgen was only included in the 28 day study. PEEs were characterised by in vitro antioxidant assays and HPLC analysis. The importance of detailed chemical characterization of rooibos and honeybush when investigating biological effects in vivo is clear as distinctive biological effects and major differences in compositions were evident. Biological parameters included were serum chemical parameters, activities of selected antioxidant enzymes, levels of glutathione and the modulation of expression of oxidative stress and antioxidant defense related genes in the liver. Sub-chronic (90 days) exposure of rats to PER and PECsub both adversely affected iron absorption significantly (P<0.05) and significantly (P<0.05) and markedly lowered levels of reduced glutathione (GSH) in the liver. The high levels of polyphenol intake were implicated and interaction with glutathione was postulated to occur via catechol o-quinone conjugations with GSH. This was also implicated in the significantly (P<0.05) increased activity of glutathione reductase (GR) following 28 days. These findings suggest that PEEs from rooibos and honeybush have the potential to alter the glutathione homeostasis, which could contribute to oxidative status in the liver. PECsub resulted in alterations in the liver biliary system which was manifested as significantly (P<0.05) increased serum total bilirubin (Tbili) and alkaline phosphatase (ALP), depending on the age of the rats, level of total polyphenols and duration of exposure. The expression of a number of oxidative stress and antioxidant defense related genes were differentially altered by the PEEs of rooibos and honeybush in rat liver and further indicated potential oxidative stress. Modulatory effects of PEEs on expression of 84 of these genes in rat liver were assessed with a quantitative real-time reverse transcription polymerase chain reaction (RT-PCR) array and provided additional insights into the possible adverse and protective effects of rooibos and honeybush. Further investigation on total polyphenol dose levels and time of exposure in the application of PEEs of rooibos and honeybush as dietary supplements and functional foods is recommended and will also be of value in anticipated regulatory requirements for future substantiation of safety and efficacy.<br>AFRIKAANSE OPSOMMING: Die moontlike voordelige en/of nadelige modulerende effekte van polifenol-verrykde ekstrakte van rooibos (Aspalathus linearis) en heuningbos (Cyclopia spp.) op die antioksidant homeostasis in die lewer is ondersoek. Die fase II metaboliete van aspalatien en mangiferin, die hoof verbindings in rooibos en heuningbos onderskeidelik, is ondersoek t.o.v. glukuronidering en sulfonering. Glukuronidasie het gelei tot n verlies in antioksidant aktiwiteit van aspalatien en mangiferin soos bepaal in post-kolom HPLC-DAD-DPPH• en HPLC-DADABTS•+ toetse, asook verminderde interaksie met yster van mangiferin in die FRAP toets. Twee onafhanklike studies van 28 en 90 dae is onderneem met polifenol-verrykde ekstrakte (PVEs) van ongefermenteerde rooibos [Aspalathus linearis (PVR)] en heuningbos [Cyclopia. subternata (PVCsub) and C. genistoides (PVCgen)] in manlike Fisher rotte om die moontlike voordelige en/of nadelige biologiese effekte te ondersoek. PECgen was slegs in die 28 dae studie ingesluit. PVEs is gekarrakteriseer deur in vitro antioksidant en HPLC analises. Die belang van chemiese karaktirisering van rooibos en heuningbos tydens ondersoeke na biologiese aktiwiteit is duidelik aangesien verskeie en variërende biologiese aktiwiteite en verskille in die komposisie in die huidige studie gesien is. Die biologiese parameters wat ondersoek is om die effek van die PVEs te bepaal het serum kliniese parameters, aktiwiteit van geselekteerde ensieme, glutatioon en evaluering van die ekspressie van oksidatiewe en antioksidant verwante gene in die lewer, ingesluit. Sub-kroniese (90 dae) blootstelling van rotte aan PVR en PVCgen het yster absorpsie negatief beïnvloed. Die beduidende (P<0.05) verlaagde vlak van gereduseerde glutatioon in die lewer was toegeskryf aan die hoë vlakke van polifenole ingeneem tydens die studie en word moontlik veroorsaak deur n spesifieke katekol o-konjugasie van GSH. Hierdie interaksie was ook moontlik die oorsaak van n beduidende (P<0.05) toename in die aktiwiteit van glutatioon reduktase. Dié bevindinge het moontlike implikasies t.o.v die glutatioon homeostase en is n moontlike indikase dat PVEs van rooibos kan bydra tot oksidatiewe stres. PVCsub het veranderinge in die lewer gal-sisteem tot gevolg gehad aangesien daar n beduidende (P<0.05) verhoging in die serum totale bilirubin en alkalien fosfaat was. Hierdie veranderinge is beïnvloed deur die ouderdom, vlakke van die totale polifenole en die periode van blootstelling. Die uitdrukking van oksidatiewe en antioksidant verwante gene is op verskillende maniere beïnvloed deur die PVEs van rooibos en heuningbos in rot lewer and dien as n verdere indikasie van onderliggende oksidatiewe stres. Die modulerende effekte van PVEs op geenuitdrukking het gelei tot additionele insig aangaande die moontlike skadelike of beskermende eienskappe van PVEs vir gebruik as kruie produkte of dieet aanvullings. Die indikasies van moontlike oksidatiewe stres was duidelik van biologiese parameters en modulering van geenuitdrukking in die lewer, en vereis verdere ondersoek na die polifenool dosis en periode van toediening voordat PVEs van rooibos en heuningbos as funksionele voedsel produkte gebruik word. Hierdie ondersoek sowel as toekomstige ondersoeke in hierdie verband sal van waarde wees vir regulatoriese vereistes omtrent die veiligheid en effektiwiteit van rooibos en heuningbos kruie produkte.

Abstract:
In this study a standardized 46 week chronic drinking water toxicity protocol was used to elucidate the toxic potential of Sutherlandia frutescens (S. frutescens) using histopathologic, morphometric and transmission electron microscopic analysis. The histopathologic changes in the duodenum, heart, kidney, liver, lung, pancreas and spleen of male Wistar rats were evaluated. Fifty-four rats were randomly divided into four groups: Group 1 – Normal diet control (ND control), n=7, Group 2 – Normal diet + plant extract (ND + p), n=9, Group 3 – High fat diet control (HFD control), n=19Group 4 – High fat diet + p (HFD + p), n=19In the high fat group male Wistar rats were fed ±55 g/day of a specialised high fat diet over a 46 week period to induce obesity and an insulin resistant state. The treatment groups (groups 2 and 4) received a dose concentration of a tea extract of the S. frutescens plant in their drinking water daily. This study showed that the consumption of S. frutescens significantly reduces weight gain in male Wistar rats on a chronic high fat diet (p≤0.001 vs. HFD control group). S. frutescens appears to propagate periportal and centrilobular glycogen storage in rat hepatocytes in the experimental groups as exemplified by a significantly (p≤0.0001 vs. control groups) increased incidences of Periodic Acid Schiff (PAS) positive staining S. frutescens also reduced intracellular lipid accumulation as made evident by the significantly lower incidence of epicardial adipose tissue (EAT), hepatic steatosis and pancreatic interstitial fat. Obesity was associated with increased fibrotic lesions such as myocardial perivascular fibrosis, centrilobular hepatic fibrosis and pancreatic periductal fibrosis. Obesity associated hypertension contributed to the widespread and significant increase in the average lesion severity of arterial congestion in all organs in the HFD control group. Pulmonary infection was equally prevalent in all rats. Despite the complex histopathology in all groups, differences in the control groups, such as, the presence of a conservative polymorphonuclear leukocyte (PMNL) infiltration, substantial intra-alveolar oedema and focal arterial wall hypertrophy in the control groups was highly suggestive of Sendai viral infection. However histopathologic evidence, in the treatment groups, suggested chronic recurrent viral infection with superimposed Mycoplasma pulmonis (M. pulmonis) bacterial infection. The impact of advanced suppurative pulmonary infection was widespread and exemplified by increased lesion incidences of spontaneous murine progressive cardiomyopathy (MCP) and spontaneous chronic progressive nephropathy (CPN) among others. In conclusion S. frutescens administered for 46 weeks to male Wistar rats significantly lowered intracellular lipid accumulation and obesity associated myocardial, renal, hepatobiliary, pulmonary and pancreatic histopathology. Moreover, duodenal, cardiovascular, hepatobiliary, pulmonary, renal, pancreatic and splenic tissue did not show histopathologic evidence of direct plant extract associated toxicity or carcinogenicity.

Abstract:
  Vad1.3 is an evolutionarily-conserved, testis-specific gene identified from a retinol-treated Vitamin A-deficiency (VAD) rat model. VAD1.3 is expressed throughout spermiogenesis at the acrosome of spermatids and epididymal spermatozoa, suggesting a role in acrosome biogenesis or acrosome reaction. The present study aimed to explore the functional role of VAD1.3 in spermatogenesis and sperm functions by the cellular and gene-knockout approaches.   Double immunofluorescent microscopy confirmed the co-localization of VAD1.3 and syntaxin 1 in mouse spermatids and spermatozoa. Deletion analysis of the Vad1.3 gene in transfected mouse spermatocyte GC2-spd and human cervical cancer HeLa cells revealed a polarized peri-nuclear/Golgi expression pattern for the N-terminal GFP-VAD fusion proteins which contain a bipartite nucleus localization (BNL) motif, but a nuclear expression pattern for the C-terminal GFP-VAD. The N-terminal sequences of VAD1.3 mediated its interaction with syntaxin 1, as demonstrated by both co-localization and co-immunoprecipitation studies. The full-length GFP-VAD co-localized with the Golgi markers and was redistributed into the endoplasmic reticulum after brefeldin A treatment, suggesting that VAD1.3 was recruited through the ER-Golgi-acrosome pathway.   Vad1.3+/- mice was previously generated by the conventional knockout approach. The heterozygous mice had normal spermatogenesis during postnatal days and adulthood (6-8 weeks). At the age of 8-19 months, 6 out of 17 heterozygous mice but no wild-type exhibited a decrease in the epididymal sperm count and testicular weight (p < 0.05). Histological analyses unveiled disarrangement of the seminiferous epithelium and sloughing of germ cells, predominantly spermatids, which was mediated partially by apoptosis as a higher percentage of TUNEL-positive cells were detected in these heterozygous mice (p < 0.05). This phenotype was associated with a decrease in the mRNA (p < 0.05) and protein levels of VAD1.3 in the testis.   Crossing of the Vad1.3+/- mice produced wild-type and heterozygous offspring in a ratio of 1:3, but no Vad1.3-/- mice were found. There was no significant difference between the heterozygous intercrosses and the wild-type intercrosses in the number of oocytes ovulated, the developmental rate of embryos from zygotes to blastocysts, the number of implantation site, resorption site or the offspring could result from defective fertilization between Vad1.3 null gametes rather than developmental lethality. The role of VAD1.3 in fertilization was supported by the inhibitory effects of the anti-VAD1.3 antibody on in vitro fertilization and progesterone-induced acrosome reaction. Immuno-staining revealed that VAD1.3 was present in the acrosome-intact spermatozoa but not in acrosome-reacted spermatozoa, indicating a role of VAD1.3 in ZP-binding or acrosome reaction rather than sperm-egg fusion. In oocytes VAD1.3 was distributed in the cytoplasm near the cortex. litter size. Only a few Vad1.3-/- embryos were found at the zygotic (3.7%) and 2-cell (3%) stages in the heterozygous intercrosses. These findings suggested that the absence of the Vad1.3-/-   In sum, VAD1.3 may play important roles in fertilization and spermatogenesis in mice. The BNL motif of VAD1.3 directs its Golgi expression and the N-terminal sequence of the protein mediates its interaction with syntaxin 1. Abstract:
The structural and functional integrity of a developing and maturing fetal and neonatal lung is critically dependent on carbohydrate metabolism. The energy derived from carbohydrate metabolism is utilized during the processed of cell growth and development. It is reported that maternal nicotine exposure during pregnancy and lactation results in the irreversible inhibition of glycolysis, for which no mechanism is currently proposed and a significant increase in glucose turnover. The principal objective of this thesis was to determine the influence of maternal nicotine exposure during gestation and lactation on the isoenzyme patterns and transcript levels of the selected enzymes in developing neonatal rat lung, in an attempt to elucidate the mechanism of inhibition of glycolysis observed.

Abstract:
Melatonin is a product of the pineal gland and is postulated to play an antigonadotropic role in the reproductive system of mammals. The reproductive system of non-seasonally breeding mammals is believed to be not as responsive to melatonin treatment as that of seasonally breeding mammals. Recently, there has been increasing support from in vivo and in vitro studies, for the hypothesis that melatonin has negative effects on sperm physiology, especially on sperm motility. High and/or low seminal concentrations of melatonin have been associated with abnormalities in human sperm motility and concentration. In this study, I examined the effects of melatonin on the testis, epididymis and sperm physiology, using in vivo and in vitro experiments, in a non-seasonally breeding mammal. Treatment, in vivo, with exogenous melatonin for six weeks did not inhibit testosterone production or spermatogenesis, nor did it affect the mass of the testes and epididymides at dissection, the concentration the morphology of speimatozoa. However, melatonin in vivo had a small, but significant negative effect on sperm motility and sperm motility index. In vitro incubation of spermatozoa Fith melatonin reduced the percentage (%) of forward progressive movement (fpm), increased the % reduction in fpm, reduced the vigor or quality of sperm motility, reduced the sperm motility index, and delayed and/or prolonged the transition of one pattern of sperm motility to the subsequent patterns. Melatonin increased the pH of the culture medium, and the increased pH, and the ethanol utilized as a solvent for melatonin, both negatively affected all the sperm motility parameters that were assessed in my study. The effects of ethanol increased with time, and the effects of pH increased with both time and increasing pH. Melatonin in vitro did not inhibit capacitation and the acrosome reaction, but it delayed the onset and the progression of capacitation and the acrosome reaction. These results suggest that while melatonin did not inhibit spermatogenesis in the Wistar rat, it may influence sperm motility. Therefore, the presence of high concentrations of melatonin in the reproductive fluids may inhibit sperm motility. With further detailed research, melatonin may have a potential use as a contraceptive drug.

Abstract:
Previous work has shown that removal of the midline frontal cortex at seven to ten days of age is followed by recovery of function correlated with apparent spontaneous generation of new tissue in the lesion cavity. The question asked in the present thesis was whether the removal of the regrown tissue in adulthood would block normal function. Rats that received P10 frontal lesions underwent second lesions at P160, and were compared to rats with only P10 or P160 lesions. Rats with P10 + P160 lesions were severely impaired on a spatial learning task, especially relative to the P10 lesion-only rats. In a second experiment, rats with P10 + P160 lesions were given intra-ventricular infusions of a cocktail of three growth factors. The animals with growth factors showed marked behavioral recovery, although there was no cell regeneration. The results of these experiments suggest that filled-in tissue in neonatally lesioned rats is functional.<br>viii, 74 leaves : col. ill. ; 28 cm.

Abstract:
Benefiting from higher SNR as well as better spatial, temporal and spectral resolution, magnetic resonance imaging (MRI) at high field has proved to be a valuable neuroimaging modality which provides comprehensive evaluation of the central nervous system non-invasively. The objectives of this doctoral work were to develop MRI methodologies and to assess the functional, metabolic and structural alterations in rodent brains under normal and manipulated conditions. Firstly, to improve the functional sensitivity and spatial precision, a novel functional MRI (fMRI) method using balanced steady state free precession with intravascular susceptibility contrast agent was proposed and its feasibility was evaluated in rat visual system. This new approach was sensitized to cerebral blood volume (CBV) changes. It provided comparable sensitivity to conventional CBVweighted fMRI using echo planar imaging but with no severe image distortion and signal dropout. Robust negative responses during visual stimulation were observed and activation patterns were in excellent agreement with known neuroanatomy. As a promising alternative to conventional CBV-weighted fMRI, it was particularly suited for fMRI investigation of animal models at high field. Secondly, the relationship between anatomical connections and resting-state fMRI connectivity was explored using a well-controlled animal model of corpus callosotomy. Both complete and partial callosotomy resulted in significant loss of interhemispheric connectivity in the cortical areas whose primary interhemispheric connections via corpus callosum (CC) were severed. Partial restoration of interhemispheric connectivity and increased intrahemispheric connectivity were also observed. The experimental findings directly supported that anatomical connections via CC play a primary and indispensable role in resting-state connectivity, and that resting-state networks could be dynamically reorganized or acquired directly or indirectly through the remaining anatomical connections. Thirdly, proton magnetic resonance spectroscopy (1H MRS) was employed to monitor the longitudinal metabolic alterations elicited by exogenous stimulation and endogenous modification, respectively. Significantly lower hippocampal N-acetylaspartate (NAA) was observed in fear conditioning animals, indicating reduced neuronal dysfunction and/or integrity, which contributed to the trauma-related symptoms. Meanwhile, pregnant animals exhibited prominently higher hippocampal NAA level, reflecting the increased density of neurons in this region, which might facilitate supporting behaviors that involving learning and memory. The 1H MRS detection of ongoing neurochemical changes induced by fear conditioning and pregnancy, especially in the hippocampus, can shed light on the mechanisms of learning and memory and the neurochemical underpinnings of behavioral improvement in pregnant animals. Lastly, manganese-enhanced MRI (MEMRI) was employed to investigate the hypoxic-ischemic (HI) injury in the late phase and the neural response to conditioned fear. Significantly higher enhancement in T1-weighted images was found in the peri-lesional region 24 hours after manganese administration and it colocalized with the increase in glial cell density in histological staining, demonstrating the existence of reactive gliosis in the late phase after HI injury. In fear conditioned animals, higher manganese uptake was observed in amygdala, hippocampus, paraventricular nucleus of hypothalamus and cingulate cortex, which were all highly-involved in the process of fear.
