Conventional slope practice, based on the deterministic factor of safety, cannot address the uncertainty in the input parameters of slope analyses in any explicit way. It relies entirely on the subjective judgment of the designer, which varies substantially among geotechnical engineers. Probabilistic techniques are powerful tools that can be used to quantify and incorporate uncertainty into slope analysis and design. A probabilistic slope analysis methodology based on Monte Carlo simulation using Microsoft® Excel and @Risk software is applied to investigate the Lodalen slide that occurred in Norway in 1954. Starting with field and laboratory data, the study demonstrates the techniques used in quantifying the uncertainties in soil properties and pore-water pressure, conducting a probabilistic assessment, and estimating the probability of unsatisfactory performance. The probability of unsatisfactory performance of the Lodalen slope is estimated to be 0.70, indicating that failure was imminent. The inclination of the Lodalen slope is then flattened, hypothetically, to different angles and the relationships between the slope angle, the factor of safety, and the probability of unsatisfactory performance are investigated.Key words: probabilistic analysis, slope stability, Monte Carlo simulation, spatial variability, Lodalen slide.

Yoga merupakan salah satu aktivitas fisik yang menggunakan berbagai postur untuk mengembangkan kekuatan fisik, kelenturan dan ketahanan dimana dapat digunakan sebagai latihan berintensitas sedang untuk pasien dengan kapasitas latihan terbatas sehingga lebih mudah bagi orang-orang yang inaktif secara fisik. Tujuan Penelitian yaitu untuk mengetahui pengaruh yoga terhadap waist hip ratio (WHR) pada wanita di Desa Lodadi Kecamatan Ngemplak Kabupaten Sleman Yogyakarta. Penelitian ini merupakan penelitian pre–eksperimental dengan One Group Pretest - Postest Design. Subyek penelitian diperoleh dengan teknik purposive sampling di Desa Lodadi. Subyek berjumlah 30 orang. Yoga dilakukan sejak November 2018 hingga Januari 2019 setiap 2x seminggu selama 90 menit. WHR diukur dengan menggunakan meter ukur. Setelah itu data akan dianalisis dengan paired sample t-test. Hasil Penelitian menunjukkan dari 30 subyek penelitian 17 peserta termasuk dalam kriteria eksklusi. Hasil analisis statistik paired sample t-test didapatkan p<0,05 yang menunjukkan perbedaan bermakna antara WHR peserta sebelum dan sesudah pelaksanaan yoga. Kesimpulan pada penelitian ini adalah Yoga dapat mempengaruhi waist hip ratio pada wanita di Desa Lodadi Kecamatan Ngemplak Kabupaten Sleman Yogyakarta.

<p>The research objectives were to identify Dam Lodan rehabilitation and influencing to production and farm income. The research with survey method, and descriptive analysis. The research was carried out Lodan village, Sarang District, Rembang. The research findings indicated that production and farm income to increased, after the Dam Lodan rehabilited. Padi to increased 171,75% and farm income 169,75%. The sensitivity analysis indicated, when the construction cost increasing 10% and the benefit decreasing 10%, EIRR= 12,03, NPV= 872,740 and B/C Ratio= 1,45.</p>

AbstrakPenelitian mengenai perkiraan TDOA dalam pelacakan paus lodan telah dilakukan[1] yang menggunakan Teager-Kaiser operator dalam peningkatan akurasi perkiraan nilai TDOA untuk pelacakan paus lodan menggunakan algoritma LMS adaptif. Namun demikian, belum dilakukan penelitian lebih lanjut mengenai faktor konvergensi (μ) yang tepat (cepat dan akurat) pada perkiraan TDOA menggunakan algoritma LMS adaptif untuk pelacakan paus lodan. Akan dilakukan analisis perkiraan nilai TDOA menggunakan algoritma LMS adaptif pada pelacakan posisi paus lodan dengan variasi nilai μ. Pada penelitian ini, sebagai acuan untuk menentukan nilai μ yang tepat, digunakan hasil pelacakan paus pada penelitian [2].Perkiraan TDOA menggunakan algoritma LMS Adaptif pertama kali dipublikasikan oleh Benesty [3]. Pendekatan yang dilakukan berdasarkan pada perkiraan tanggap impulsional dari sumber ke penerima. Pada algoritma LMS adaptif dilakukan pembaruan nilai koefisien dengan μ sebagai langkah adaptasi. Nilai TDOA diperoleh dari selisih jarak dua tanggap impulsional.Dari hasil penelitian diperoleh hasil bahwa perubahan nilai μ mempengaruhi kecepatan perkiraan TDOA menggunakan algoritma LMS adaptif. Akan tetapi, nilai μ yang tidak tepat mengakibatkan hasil pelacakan paus tidak akurat. Dari penelitian ini diperoleh nilai μ terbaik untuk perkiraan TDOA menggunakan algoritma LMS adaptif pada pelacakan posisi paus lodan adalah 0.01. However, further researches on the right convergence factor (μ) (fast and accurate) in TDOA estimation using adaptive LMS algorithms for sperm whale tracking have not been performed. Will be analyzed the TDOA estimation using adaptive LMS algorithm in sperm whale tracking with the variation of μ. As a reference to determine the right value of μ, the results of sperm whale tracking[2] is used.TDOA estimation using Adaptive LMS algorithm was first published by Benesty [3]. The approach taken is based on estimation of the impulse responses from source to receivers. In the adaptive LMS algorithm function updates the coefficient with μ as adaptation step. TDOA values obtained from the difference between the two impulse responses.From the results, values changing of μ affect the speed of TDOA estimation using the LMS adaptive algorithm. However, the wrong value of μ is lead to inaccurate sperm whale tracking results. The best value of μ for TDOA estimation using the LMS adaptive algorithm for sperm whale tracking is 0.01. AbstractThree tobacco industry based laboratories determined selected mainstream components using their established in-house methods. Machine smoking was done according to the ISO smoking regime. The Test cigarettes smoked for this investigation were manufactured with different amounts of added glycerol, cocoa powder and sucrose. Variability between the three laboratories differed clearly for the analyzed smoke components. No overall effects due to the added ingredients on smoke components could be found. The high ‘tar’ products with the highest lodading of sucrose showed a slight increase in formaldehyde emissions among all three laboratories.

Long-term physical activity causes restructuring in the various functions and systems of the body, which are, as a rule, adaptive in nature and occur at all levels of the structural organization of the body. The question of changes in the functional state of the cardiovascular system and, especially, the microcirculatory section of the circulatory system in normal and under the physical exertion and their relationship with ultrastructural remodelling in tissues, cells and cellular organelles is still remaining unclear. The aim of the study was to assess structural changes in the tissues of the calf muscle and myocardium that cause the development of adaptive mechanisms in response to prolonged physical exertion. We studied the structural components of tissue samples of calf muscle and the apex of the heart taken from mature Wistar male rats weighing 220-250 g in mass exposed to prolonged physical exertion by using electron microscopy. It has been shown that a long and intense physical loading causes the development of a number of compensatory-adaptive reactions directed at improving microcirculation processes, oxygen supply of skeletal muscles and myocardium, as well as maintaining an adequate level of metabolic processes. A detailed assessment of the functional state of the tissues studied is necessary for the early detection of signs of maladjustment under the constant loading in sports and the prevention of pathological states.

Nowadays, people are very facilitated by the existence of various shopping centers, including retail. Because many retailers are close to each other, Alfamart Lodadi must have a good marketing strategy. So far, the strategy used is sometimes inaccurate because it is not based on customer segmentation. Therefore, the purpose of this research is to help retail owners to make decisions regarding the right marketing strategy with three methods so that Alfamart Lodadi can compete and increase sales. The Analytical Hierarchy Process (AHP) is employed to find the priority variables of customer segmentation; meanwhile, the K-Means Clustering is used to group customers based on the similarity of predetermined characteristics. AR-MBA is used to find out the best rules, and products are rarely, sufficient, and frequently purchased. The results of this research, based on AHP, obtained five segmentation priority variables based on the largest eigenvector values. There are income, age, expenditure, distance, and shopping intensity with each eigenvector value of 0.13; 0.16; 0.12; 0.12; 0.17. From clustering, there are three customer clusters with different strategies, including free shipping when shopping, product discounts for certain periods, and providing catalogs and discounts on each transaction and offer notifications. Then, this research also obtained three strategies based on AR-MBA. These include making a catalog by bringing frequently purchased products closer together, choosing a layout for shopping places by bringing frequently purchased products closer together, and making shopping coupons for rarely purchased products. With several strategic choices, companies can make decisions appropriately according to the desired criteria.

Abstract Wnt4, a member of the Wnt family of secreted factors, is essential for kidney organogenesis since the kidney fails to develop in its absence. Besides the kidney, Wnt4 signaling is involved in the control of development of several other organs such as the gonads, adrenal glands and pituitary gland. In the context of the embryonic kidney, Wnt4 signaling induces mesenchymal to epithelial transition of the progenitor cells in the metanephric mesenchyme, an early step in nephrogenesis. Wnt4 signaling may also be relevant in the development of a childhood kidney tumor, the Wilms’ tumor, that involves the function of Wilms’ tumor suppressor protein 1 (WT1). Wilms’ tumor is thought to arise from the early metanephric mesenchymal cells of the embryonic kidney, but the detailed mechanisms are not known. The main aim of this project was to study the mechanisms that regulate expression of the Wnt4 gene by using immortalized embryonic kidney mesenchyme-derived mK4 cells as a model. The Wnt4 gene expression was also analyzed in vivo in the frog embryonic pronephros. Through the use of reporter assays and a two-hybrid screen, Sox11, a member of the SoxC family of transcription factors, was identified as a synergistic protein that interacts with WT1. Immunoprecipitation studies provided further evidence that Sox11 and WT1 may physically interact with each other in the developing embryonic kidney. Indeed, Sox11 and WT1 may regulate the Wnt4 gene expression in vivo since the morpholino-based knock-down of either WT1 or Sox11 led to notable downregulation of the Wnt4 gene expression in the frog embryonic pronephros. The other general aim of this thesis was to develop novel tissue targeting and therapy tools to the cell lineages regulated by the Wnt4 signals, including the podocytes. For this purpose, we utilized mice carrying a floxed expression cassette for the avidin-LDL receptor fusion protein, Lodavin, in the constitutively active Rosa-26 locus. Three Cre driver mice, including the Wnt4-Cre knock-in line, were used to activate Lodavin expression in the respective cells of the embryonic kidney. Moreover, we generated a podocyte injury model by expressing the human receptor for diphtheria toxin specifically in the podocytes. This was achieved by crossing mice containing a floxed expression cassette for this receptor in the Rosa-26 locus with those expressing the Cre recombinase under the nephrin promoter. Administration of diphtheria toxin led initially to podocyte damage only, followed by a progression to glomerular sclerosis. As a summary, Sox11 and WT1 serve as synergistic transcription factors that may regulate expression of the Wnt4 gene in vivo. The transgenic mouse models generated and used provide the basis to generate acute and chronic kidney disease models and the potential to purify the respective cells for developing cell-based therapy avenues for the kidney. Moreover, the Lodavin-based approaches may enable targeted delivery of biotinylated small compounds, proteins, viruses or even cells and novel means for in vivo imaging and functional studies
Tiivistelmä Wnt-4 kuuluu signaloivien proteiinien Wnt-perheeseen ja sen toiminta on välttämätöntä munuaisen kehityksessä. Ilman Wnt-4 proteiinia munuainen ei kehity. Munuaisen lisäksi Wnt4-signalointi on mukana useiden muiden elinten, kuten sukurauhasten, lisämunuaisen ja aivolisäkkeen säätelyssä. Alkion munuaisessa Wnt4-signalointi saa aikaan mesenkymaalisen kantasolukon epitelisoitumisen, edustaen näin ollen nefronin kehityksen varhaisia vaiheita. Wnt4-signaloinnilla on myös merkittävä asema lapsuusiän munuaiskasvaimen, niin kutsutun Wilmsin kasvaimen kehittymisessä. Tämän tyyppisessä kasvaimessa keskeisenä on Wilmsin tuumoriproteiinin WT1:n toiminta, mutta myös Wnt4:n toiminnalla voi olla merkitystä. Wilmsin kasvaimen arvellaan saavan alkunsa varhaisista sikiöaikaisista jälkimunuaisen soluista, mutta yksityiskohtaisia mekanismeja ei vielä tunneta. Tämän projektin tarkoituksena oli tutkia Wnt4-geenin ilmentymistä sääteleviä mekanismeja käyttäen mallina mK4-soluja eli alkion munuaisesta saatuja, immortalisoituja soluja. Wnt4-geenin ilmentymistä analysoitiin myös in vivo sammakon alkion alkumunuaisessa. Tuplahybridi-analysoinnin avulla tunnistettiin transkriptiotekijäperhe SoxC:n jäsen Sox11 samantoimiseksi proteiiniksi transkriptiotekijä WT1:n kanssa Wnt4-geenin ilmentymisen säätelyssä. Immunopresipitaatiotutkimukset tukivat ajatusta, että Sox11 ja WT1 voisivat olla fyysisessä vuorovaikutuksessa säädellessään nefroninmuodostuksen alullepanossa ratkaisevan Wnt4-geenin ilmentymistä. Sox11 ja WT1 voivat mahdollisesti säädellä Wnt4-geenin ilmentymistä myös in vivo, sillä morfoliineihin perustuvissa kokeissa sekä WT1:n että Sox11:n hiljennys laski Wnt4-geenin ilmentymistasoa sammakon alkumunuaisessa. Tämän väitöstutkimuksen toinen yleinen tavoite oli kehittää uusia kudoskohdennus- ja terapiakeinoja Wnt4-signaloinnin säätelemille solulinjoille, kuten podosyyteille. Tätä tarkoitusta varten kloonattiin siirtogeeninen hiiri, jossa floksattu avidiini-LDL -reseptorifuusioproteiini, Lodavin, kohdennettiin jatkuvasti aktiiviseen Rosa-26 -lokukseen. Kolmea eri Cre-hiirilinjaa käytettiin aktivoimaan Lodavinin ilmentyminen kussakin tietyssä alkion munuaisen solupopulaatiossa. Yksi näistä Cre-linjoista oli Wnt4-Cre. Jotta kyettäisiin vahingoittamaan samoja soluja, jotka ilmentävät Lodavinia, hyödynnettiin difteriamyrkyn ihmisen reseptoria (iDTR). IDTR:n ilmentäminen tietyissä hiiren soluissa tekee ne alttiiksi tappavalle difteriamyrkylle. IDTR-perusteisen munuaisvauriomallin kehittämiseksi käytettiin floksattua iDTR-hiirimallia, ja geenin ilmentyminen aktivoitiin Wnt4-indusoiduissa munuaissolulinjoissa, erityisesti podosyyteissä Nephrin Cre -välitteisesti. NephrinCre;R26RiDTR hiiriä altistettiin difteriamyrkylle ja niiden munuaiskerästen muutoksia seurattiin. Tutkimukset antavat viitteitä siitä, että R26R-floksatut iDTR-hiiret toimivat hyvänä mallina kehitettäessä sekä akuutteja että kroonisia munuaistautimalleja. Yhteenvetona voidaan todeta, että Sox-11 ja WT-1 ovat samantoimisia transkriptiotekijöitä, jotka voivat säädellä Wnt4-geenin ilmentymistä in vivo. Tutkimuksessa kehitetyt ja käytetyt siirtogeeniset hiirimallit tarjoavat perustan kehittää sekä akuutteja että kroonisia munuaistautimalleja. Samalla ne mahdollistavat kulloistenkin solujen eristämisen uusien soluperusteisten hoitomenetelmien kehittelemiseksi. Lisäksi Lodavin-perusteiset lähestymistavat voivat mahdollistaa biotinyloitujen pienten yhdisteiden, proteiinien, virusten tai jopa solujen kuljetuksen kohdennetusti sekä avata uusia mahdollisuuksia in vivo -kuvantamiselle ja toiminnallisille tutkimuksille

The aim of this study is to analyse the sexual power relations in contemporary urban novels of Swedish female writers.

I have identified a discourse in these novels as a heteronormative matrix which power is ruled by norms and assumptions about normality in both the heterosexual and lesbian stories. For this purpose I have related to Ivar B. Neumanns theory that individuals always find a way to confront the discourse by three different strategies. These strategies are used in my study as tools to organise these novels, because it is possible to see that the protagonists find at least one of these strategy’s to her protest.

The first strategy I studied was the docile. Here I argument that the heroines in Alla vilda, Jag minns alla mina älskare och hur de brukade ta på mig, Inte enklare än så, Lögner and Att älska henne do not reject the discourse about sexual power relationships. The second strategy, named the oppositionist, implies the protagonists refusing to be left as victims. In one way or another, either by change the rolls of the sexes (Ta vad man vill ha) or by problematising the order of them (Ego girl) or find lesbianism as a way to reject the discourse (both the novels by Mian Lodalen: Smulklubbens skamlösa systrar and Trekant). The reluctant is the last strat-egy who is the creative one, which rejects some things and adapt to other. Here I place the heroines from Stjärnor utan svindel, Andra sidan Alex and Storlek 37.

The result of this study is that despite quite different strategies to oppose against the hetero-sexual power discourse and the normative femininity everyone relates to it and thereby con-firms it.

Syftena med denna uppsats har varit flera. Dels att genomföra en undersökande genrediskussion av svensk chick-lit i stort där det som uppfattats som den huvudsakliga kritiken av genren omformulerats till frågeställningar. Huvudfrågan har därmed varit att ta reda på om svensk chick-lit är intressant, men också om den har några litterära kvaliteter och om den är en feministisk backlash. I uppsatsen arbetas även en definition av svensk chick-lit fram, samt empiriskt grundad teori ur materialet. Avsikten är att detta arbete ska kunna verka som förebild för andra utredningar av nya och marginaliserade genrer. En undersökning av vilka principer biblioteken, här representerade av Umeåregionens bibliotek, utgår från vid inköp av skönlitteratur i stort och chick-lit i synnerhet har också genomförts.

The aims of this paper have been more than one. One aim is to conduct an investigative genre discussion of Swedish chick lit as a whole, where what is conceived as the main critique of the genre is reformulated into questions. The main questions here are if Swedish chick lit is of interest, if it possesses any literary qualities and if it constitutes a feminist backlash. This paper also draws up a definition of Swedish chick lit and empirically grounded theory from the material. The intent is for this paper to serve as a model for other analyses of new and marginalized genres. A study of the principles adopted by libraries, here represented by libraries in the Umeå district, in fiction purchases as a whole and purchases of chick lit in particular, has also been carried out.
