Abstract:
The formation of magnetic order in solids is a complex and subtle issue. There are a wide range of different types of magnetisation, all of which may be favoured under different circumstances. In this thesis we consider a novel combination of ideas where the formation of spatially modulated magnetisation is linked to a metamagnetic transition. In this we are inspired by a general principle of modulated phases intervening as intermediate states in phase transitions. In particular we draw analogies with the Fulde-Ferrell-Larkin-Ovchinnikov state of spatially modulated superconductivity. We study a mean-field theory for itinerant magnetism where the crystal lattice drives the formation of a rich phase diagram. A peak in the electronic density of states due to a van Hove singularity creates ferromagnetism and a metamagnetic transition. Furthermore we find that a modulated magnetic phase - a spin-spiral, becomes favoured along the metamagnetic transition line. The appearance of this phase causes the metamagnetic transition to bifurcate to enclose the modulated region. The topology of this reconstructed phase diagram shows remarkable similarity to that observed in experiments on Sr₃Ru₂O₇. This material shows a metamagnetic transition which can be tuned by field angle towards zero temperature. Before this point is reached a new phase with high and anisotropic resistivity appears. We believe that this anomalous phase can be explained by the formation of a phase of modulated magnetisation caused by a peak in the electronic density of states. This mechanism may also apply in a range of other materials as it is driven by rather generic features of the bandstructure.

Abstract:
One of the challenges facing condensed matter physics nowadays is to understand the electronic structure of high temperature superconductors. This dissertation compiles our contribution to the experimental information concerning this subject. Tunneling conductance spectroscopy a technique capable of probing the electronic density of states in hybrid structures was used to study the current and spin transport properties across junctions between metallic counterelectrodes and Bi2Sr2CaCu2O8- (BSCCO) crystals. Since in these structures the transport is mediated by transmission channels depending on superconductive characteristics, the energy resolved density of states is a signature of the mechanism of superconductivity. For instance, one can observe the superconductive energy gap and the behavior of subgap bound states due to phase sensitive Andreev reflections at the junction interface. In particular, tunneling spectroscopy makes possible the observation of the LOFF state characterized by the coexistence of superconductivity and magnetism. Cuprates like BSCCO are highly anisotropic materials and their superconductivity is almost two dimensional, being confined in the CuO2 planes. Therefore, our junctions combine monocrystals of underdoped samples of BSCCO with various thin film counterelectrodes normal metal (Ag), conventional superconductor (Pb) and ferromagnetic metal (Fe) deposited perpendicular onto the cuprate ab-plane (CuO2 plane). We performed measurements on Ag/BSCCO junctions for two current injection directions into the same crystal. We observed that, near the 110 crystal surface, the conductance spectra show a high zero bias peak (ZBCP) which is a manifestation of zero energy Andreev bound states due to an anisotropic superconductive order parameter. Near the 100 surface, the ZBCP is largely suppressed. This is consistent with a predominantly 2 2 x y d - -wave pairing symmetry. In some cases, the ZBCP splits or decreases in amplitude at low temperatures. This is consistent with the existence of a subdominant s-wave (or xy d ) resulting in a mixed d is + state which breaks time reversal symmetry (BTRS). Since we observe this phenomenon in the underdoped case, we do not confirm the possibility of a quantum critical point close to the optimal doping. Our Pb/BSCCO spectra contradict the theory explaining the BTRS by proximity effect. The Fe/BSCCO junctions measure the effect of spin polarization. We explain the recorded 4-peak asymmetric structure by the combined effect of a spin independent BTRS state and a spin filtering exchange energy in the barrier responsible for a large ZBCP splitting. The LOFF state was observed in the proximity region induced on the ferromagnetic side of multilayered-Fe/Ag/BSCCO structures. As expected for the LOFF order parameter, the spectra develops coherent damped oscillations with the Fe layer thickness probing different regions. The magnitude and sign of the oscillation depends on the energy. The conductances at energy zero or equal to the superconductive gap are modulated in antiphase proving that the order parameters takes successively positive and negative values. Changing the junction orientation with 4 p , results in an opposite behavior for the same distance. The maximal amplitudes in one direction is replaced by minima, showing that, besides space, the LOFF state modulation depends on the phase of the high temperature order parameter inducing the proximity

We have cultured the human hepatoblastoma cell line C3A/HepG2 and evaluated its characters. In static culture, the culture medium should be changed every 3 days at least. When nutrients in the culture medium were exhausted, the urea nitrogen synthesis declined faster than albumin synthesis did. But under oxygen deficiency, the albumin synthesis ceased quickly. Loofa sponge was investigated as a three-dimensional scaffold for stationary and perfusion culture of human hepatoblastoma cell line C3A/Hep G2. In stationary culture, C3A/HepG2 cells in loofa cubes produced higher -fetoprotein and albumin secretion rates than those in polyurethane foam (PU). To use loofa cylinders in a packed-bed reactor, immobilization of C3A/HepG2 cells and perfusion culture by re-circulating medium at various flow rates. When the re-circulating medium at 26 mL/min (superficial velocity = 51.7 cm/min) resulted in a cell loading density of (4.59±0.21)×107 cells/cm3-loofa. This cell loading density is higher than values reported in the literature for packed-bed reactor intended for bioartificial liver. During 9 days perfusion culture in the reactor, immobilized C3A/HepG2 increased up to (5.04±0.17)×108 cells/ cm3-loofa and showed steady synthesis of albumin with an average synthesis rate at 42.2 g/106 cells/day. These experimental results and observations by SEM suggested that loofa sponge is a suitable scaffold for high-density culture of human hepatocyte cell line and the immobilized cells could express high levels of liver-specific functions.

In this study, loofa-sponge was as a support, the use of hydroxyethyl cellulose cross-linked to the substrate surface to increase its affinity ligand, the use of chemical synthesis of epichlorohydrin and chelator iminodiacetic acid and transition metal ions, finally after the immobilized metal affinity chromatography adsorption substrate is completed, the transition metal ions could be reacted to coordinate covalent bonds with hexahistidine-Tag on the genetically modified Picrophilus torridus trehalose synthase to purify and immobilize the trehalose synthase.      From the results of this study, we obtained the most suitable bifunctional support for the target protein with both the IMAC-adsorbed enzyme terminus and the epoxy functional group covalently bonded to the enzyme. The kinetics of adsorption and incubation time experiments established the absoption time and the immobilized time. The stability of the physical adsorption and covalent bonding was finally explored to confirm that the covalent bonding is more stable than the physical adsorption.

Every year due to lacking suitable liver for transplanting, many people die. So we try to establish a bio-artificial liver system to replace the functionless liver. In this research, we utilize loofa sponge as a three-dimensional scaffold for stationary and perfusion culture of rat liver cells. In stationary culture, rat liver cells seeding by dynamic method can obtain higher cell immobilized efficiency than those by static method. During 7 days stationary culture, rat liver cells in loofa cubes showed higher metabolic function than those in polyurethane foam (PU). In perfusion culture, to use loofa cylinders in a packed-bed reactor, seeding of 1*108 liver cells by culture medium at 18 mL/min resulted in a better metabolic function. During 7 days perfusion culture, in the reactor the activity of liver cells by perfusion 26 mL/min is better than that by perfusion 18 mL/min. This experimental results and observation by SEM suggested that loofa sponge is a suitable scaffold for culture of rat liver cells and the immobilized liver cells express liver-specific functions.

Loofa sponge has been found by our group to be a 3-D macroporous natural scaffold suitable for hepatocytes culture. Chitinous loofa fibers could be deacetylated by alkaline treatment and followed by conjugating lactobionic acid to the amine groups of chitosan to introduce galactose moieties to the scaffold. Galactosylated loofa sponge is expected to enhance attachment, proliferation, and metabolism of rat hepatocytes due to the specific receptor-ligand interactions between galactose and the asialoglycoprotein receptors on the membranes of the hepatocytes. Successful modification of loofa sponge was first demonstrated by FTIR analysis and the amount of galactose conjugated to the scaffold was measured quantitatively. Both static and perfusion culture were carried out by using galactosylated loofa sponge in dish culture and in a packed bed reactor system under different flow rates. Galactosylated loofa sponge could increase cell seeding efficiencies and up-regulate albumin, urea, and ammonia metabolism activities of hepatocytes when compared to the dish control and the unmodified loofa sponge. Cell aggregates and spheroid-like structures with high viability were observed with the scaffold from Scanning Electron Microscopy examinations and Live/Dead assays with Confocal Laser Scanning Microscopy.

Production of recombinant protein by plant cell expression system have several merits as, for example, (1) post-translational modification of Eukaryotic cell; (2) cheaper culture medium than mammalian expression system; (3) more simple culture medium, less purification cost . The aims of this study are to investigate the fermentation policy of repeat artificial starvation and nature starvation fermentation policy, the utility of modified loofa sponge to immobilize the transgenic rice cell, and the production of high price mGM-CSF. The transgenic rice cell, 1-1 (#14-4 for A1 intron) cell line with mGM-CSF production of 1.0283 mg/L, was selected in this study. In suspension cell culture, the best culture policy in 250 ml flask is high cellular density, N6 medium, and repeat nature starvation, have the mGM-CSF production of 6.96 mg/L. In 2 L airlift culture, the highest production is 5.845 mg/L (day 24). In immobilization cell culture, we had the highest cell viability of 83.43 % by alkali treatment of loofa sponge in 1 L flask; the highest cell dried weight of 1.028g in elimination epidermis of loofa sponge; and the highest production of 0.179 mg/L (day 22) in elimination epidermis of loofa sponge. In TBCR immobilization cell culture, we increased the cell viability up to 84.66 % in elimination epidermis of loofa sponge, but the cell dried weight decreased to 0.8256 g, the production also decreased to 0.0289 mg/L (day 25). At last, the mGM-CSF production of immobilization cell culture for elimination epidermis of loofa sponge in modified 2 L airlift is 0.914 mg/L (day 21).

Immobilization refers to the process of confining microorganisms to a certain area. Compared with traditional free cell fermentation, immobilized cell has more advantages. Cell immobilization offers including cell stability and recycling. Lactic acid is broadly used in food, pharmaceutical, tannery, and textile industries. Furthermore, it can also be polymerized to polylactic acid as biodegradable plastics. The cost of lactic acid products is expensive due to the high-priced nitrogen source which is one of the essential elements during the fermentation. Therefore, many studies use low-cost nitrogen source like by-product from agriculture or forestry. In this experiment, we use soybean hydrolysate as substitutive nitrogen source, glucose as carbon source, and compare the results of the immobilize Lactobacillus casei BCRC 10697 in Ca-alginate base and on loofah sponge fiber to produce lactic acid in batch fermentation. We use repeated batch fermentation to comprehend the stability of immobilization on loofah sponge fiber. Additionally, in order to figure out how Lactobacillus casei uses limited nitrogen source and carbon source effectively to produce lactic acid. The results showed that the production of lactic acid of immobilization on loofah sponge fiber is 70.68 g/L, which is less than immobilization in Ca-alginate base that is 77.60 g/L. But both of their yields could be above 90%. In process, immobilizating on loofah sponge fiber is easier and saving time. Ca-alginate base has low mechanical strength and would be disrupted owing to the build-up of pressure generated during cell division. Immobilization on loofah sponge fiber through physical adsorption can overcome these problems and can use 7 times at least in repeated batch fermentation. According to the results, using 25% soybean hydrolysate as a nitrogen source, the viable count was 9.98 log CFU/mL, the max yield of lactic acid was 2.31±0.15 g/L/h(RSD=6.49%). They were a little higher than 15% nitrogen source which were 9.69 log CFU/mL and 1.49±0.11 g/L/h(RSD=7.38%). The results showed that the less glucose concentration would be consumed more quickly in the condition of low carbon source (glucose concentration:80 g/L, 60 g/L, 40 g/L). Therefore, if using soybean hydrolysate as substitutive nitrogen source, it suggested that keeping above 25% nitrogen source would consume nitrogen source and carbon source (the initial glucose concentration of 40 g/L)effectively in 24 hours to produce lactic acid of 33.35±1.30 g/L(RSD=3.90%).
