Abstract:
Climate change poses an acute threat to the Vietnamese Mekong Delta (VMD). To respond to this threat, the structure of the delta’s agriculture-based economy must transform, becoming more adaptive to changing conditions. One adaptive livelihood option is the use of flood-based farming systems in the upper VMD. The present study examines local perceptions of such a system in Dong Thap Province, a lowland, flood-affected area of the upper VMD. Specifically, we explored lotus farming as a potential flood-based adaptive livelihood model for the region. The study advances the current literature by using historical research, embedded in narrative analysis applying the social construction of technology (SCOT) lens. We collected data through in-depth interviews and field surveys to qualitatively analyze the emergence and status of lotus cultivation in the study area, zooming in on how local society evolved with expansion of this farming model. The findings suggest that as an innovative idea, lotus farming initially emerged due to unfavorable natural conditions, and then was developed as an attractive nature-based livelihood, and thus received increased attention. It has been accepted and modified over time according to the new interests of further stepped-in stakeholders. Our findings echo the social construction of technology perspective as we found lotus farming to be a technological artifact that did not develop on its own, by was driven by different interpretations and re-negotiation process. This made more room for stakeholders to shape and reshape it in a way that fits their interests.

Abstract:
Climate smart agriculture (CSA) has gained considerable attention in Vietnam due to its potential to increase food security and farming system resilience while decreasing greenhouse gas emissions. In recent years, several CSA practices have been introduced in rice production, the most important sub-sector of Vietnam’s agriculture. However, few studies have been done in Vietnam to produce comprehensive assessments of CSA performance in the rice sector. This research proposes a comprehensive approach to assess CSA practices through a new set of evaluation indicators. A case study in An Giang province of the Vietnamese Mekong River Delta was implemented to evaluate the performance of five CSA models versus that of the triple rice crop system (i.e., benchmarking model). Results show that rice-shrimp and rice-lotus rotations are most profitable, low-risk, and applicable at a larger scale. Given that the current study analyzed and calculated only a small number of indicators and types of CSA practices, further research is necessary to test all indicators and diversified types of CSA models.

Abstract:
Recent studies of inner ear development suggest that hair cells and support cells arise within a common equivalence group by cell-cell interactions mediated by Delta and Notch proteins. We have extended these studies by analyzing the effects of a mutant allele of the zebrafish deltaA gene, deltaA(dx2), which encodes a dominant-negative protein. deltaA(dx2/dx2)homozygous mutants develop with a 5- to 6-fold excess of hair cells and a severe deficiency of support cells. In addition, deltaA(dx2/dx2) mutants show an increased number of cells expressing pax2.1 in regions where hair cells are normally produced. Immunohistological analysis of wild-type and deltaA(dx2/dx2) mutant embryos confirmed that pax2.1 is expressed during the initial stages of hair cell differentiation and is later maintained at high levels in mature hair cells. In contrast, pax2.1 is not expressed in support cells. To address the function of pax2.1, we analyzed hair cell differentiation in no isthmus mutant embryos, which are deficient for pax2.1 function. no isthmus mutant embryos develop with approximately twice the normal number of hair cells. This neurogenic defect correlates with reduced levels of expression of deltaA and deltaD in the hair cells in no isthmus mutants. Analysis of deltaA(dx2/dx2); no isthmus double mutants showed that no isthmus suppresses the deltaA(dx2) phenotype, probably by reducing levels of the dominant-negative mutant protein. This interpretation was supported by analysis of T(msxB)(b220), a deletion that removes the deltaA locus. Reducing the dose of deltaA(dx2) by generating deltaA(dx2)/T(msxB)(b220)trans-heterozygotes weakens the neurogenic effects of deltaA(dx2), whereas T(msxB)(b220) enhances the neurogenic defects of no isthmus. mind bomb, another strong neurogenic mutation that may disrupt reception of Delta signals, causes a 10-fold increase in hair cell production and is epistatic to both no isthmus and deltaA(dx2). These data indicate that deltaA expressed by hair cells normally prevents adjacent cells from adopting the same cell fate, and that pax2.1 is required for normal levels of Delta-mediated lateral inhibition.

Birdsfoot trefoil (Lotus corniculatus L.), arrowleaf clover (Trifolium vesiculosum Savi.), berseem clover (Trifolium alexandrinum L.), crimson clover (Trifolium incarnatum L.), barrel medic (Medicago truncatula Gaertn.), winter vetch (Vicia villosa subsp. varia L.), flatpea (Lathyrus tingitanus L.), red clover (Trifolium pratense L.), alfalfa (Medicago sativa L.), sweetclover (Melilotus officinalis Lam.), fababean (Vicia faba L.), field pea (Pisum sativum subsp. arvense L.) and white lupin (Lupinus alba L.) were grown for 2 yr in interior Alaska to assess their potential as forage and green manure crops and to evaluate the effects of N fertilizer on legume performance. Herbage yield, crude protein, and total N yield were the criteria used. In addition to previously identified promising species, berseem clover, arrowleaf clover, winter vetch and flatpea consistently produced over 3 t ha−1 of herbage and accumulated over 67 kg N ha−1. Nitrogen fertilizer (90 kg N ha−1) improved yields of arrowleaf clover and winter vetch at Fairbanks (5344 vs. 3685 kg ha−1), but N fertilization depressed yields at Delta Junction. Forage crude protein for the promising species ranged from 120 to 220 g kg−1. Several species exhibited adequate tolerance of low soil pH to be grown on acid soils without liming. Abstract:
This article introduces five terracotta figurines acquired in Egypt by Vladimir S. Golenishchev and N. G. Ter-Mikaelyan and currently preserved at the Pushkin State Museum of Fine Arts. Three statuettes depict the child god Harpocrates with the attributes of royal power; two figurines demonstrate the identification of Harpocrates with the Greek god Eros. Most of the objects can be dated mainly to the Roman times, one is to be dated to the late Ptolemaic period. The places of finding or manufacturing of figurines are mostly unknown; however, according to a number of specific features, these could be towns of the Fayum Oasis, the Delta, and in one case — probably, Edfu. Terracotta figurines of Harpocrates with royal regalia are rare, especially in comparison with the wide-spread occurrence of terracottaе with Harpocrates holding a pot or cornucopia; all these data bring his functions as patron of fertility and defender of health to the fore. The presence of royal attributes seems to be a kind of secondary, partly decorative elements that only enhance the most popular aspects of terracotta images of Harpocrates. The type of figurines depicting Harpocrates sitting on a throne with the crown of the god Amun reproduces the iconography of small bronze sculpture. In other types of terracotta the royal attributes most frequently found are the double crown and — rarely — a nemes-headdress; the crown is usually surrounded by lotus buds, a favorite motive of Harpocrates’ iconography. The childish image of Harpocrates in the time of interaction between Eastern and Western cultures led to a natural synthesis of images of the child gods of Egyptian and Graeco-Roman worlds — Harpocrates and Eros. Apparently, such terracottaе, which had more Hellenistic than Egyptian features, were in demand by the population of different towns in Ptolemaic and Roman Egypt.

Recently a fourth rearranging gene, the delta chain gene, embedded in the alpha chain locus, has been described. We have determined the structure of the beta, gamma, and delta chain genes in 15 cases of T-cell acute lymphoblastic leukemia (T-ALL) representing stage I (CD7+, CD1-, CD3-) and stage II (CD7+, CD1+, CD3-) of intrathymic T-cell development. The alpha-delta locus was rearranged in 14 of the 15 cases. In three cases the delta constant region was deleted on both chromosomes, suggesting biallelic V-J alpha rearrangement. A limited pattern of rearrangement of the delta locus was observed in the remaining 11 cases. When the alpha-delta region was rearranged, there was rearrangement of the beta and gamma TcR in all cases except two; in these cases the beta chain was in the germline configuration. These findings support the hypothesis that delta chain gene rearrangement is an early event in T- cell development, possibly contemporary to gamma gene rearrangement, and that the delta locus has a limited repertoire.

To analyze the regulation of gene rearrangement at the T cell receptor (TCR) alpha/delta locus during T cell development, we generated transgenic mice carrying a human TCR delta gene minilocus. We previously showed that the presence of the TCR delta enhancer (E delta) within the J delta 3-C delta intron was required to activate a specific step (V-D to J) of transgene rearrangement, and that rearrangement was activated equivalently in the precursors of alpha beta and gamma delta T cells. To further explore the role of transcriptional enhancers in establishing the developmental pattern of gene rearrangement at the TCR alpha/delta locus, we substituted the TCR alpha enhancer (E alpha) in place of E delta within the transgenic minilocus. We found that V-D-J rearrangement of the E alpha+ minilocus was restricted to the alpha beta T cell subset. Further, we found that although V-D-J rearrangement of the E delta+ minilocus was initiated in the fetal thymus by day 14.5, V-D-J rearrangement of the E alpha+ minilocus did not occur until fetal day 16.5. Finally, whereas V-D-J rearrangement of the E delta+ minilocus is essentially completed within the triple negative population of postnatal thymocytes, V-D-J rearrangement of the E alpha+ minilocus is only initiated late within this population. Since the properties of minilocus rearrangement under the control of E delta and E alpha parallel the properties of V delta-D delta-J delta and V alpha-J alpha rearrangement at the endogenous TCR alpha/delta locus, we conclude that these enhancers play an important role in orchestrating the developmental program of rearrangements at this locus.

Abstract:
T cells can be divided into two groups on the basis of the expression of either alpha beta or gamma delta T-cell receptors (TCRs). Because the TCR delta chain locus lies within the larger TCR alpha chain locus, control of the utilization of these two receptors is important in T-cell development, specifically for determination of T-cell type: rearrangement of the alpha locus results in deletion of the delta coding segments and commitment to the alpha beta lineage. In the developing thymus, a relative site-specific recombination occurs by which the TCR delta chain gene segments are deleted. This deletion removes all D delta, J delta, and C delta genes and occurs on both alleles. This delta deletional mechanism is evolutionarily conserved between mice and humans. Transgenic mice which contain the human delta deleting elements and as much internal TCR delta chain coding sequence as possible without allowing the formation of a complete delta chain gene were developed. Several transgenic lines showing recombinations between deleting elements within the transgene were developed. These lines demonstrate that utilization of the delta deleting elements occurs in alpha beta T cells of the spleen and thymus. These recombinations are rare in the gamma delta population, indicating that the machinery for utilization of delta deleting elements is functional in alpha beta T cells but absent in gamma delta T cells. Furthermore, a discrete population of early thymocytes containing delta deleting element recombinations but not V alpha-to-J alpha rearrangements has been identified. These data are consistent with a model in which delta deletion contributes to the implementation of a signal by which the TCR alpha chain locus is rearranged and expressed and thus becomes an alpha beta T cell.

Le travail porte sur l'etude du repertoire t chez la souris, et consiste en l'analyse des genes valpha et vbeta des lymphocytes t ainsi que des facteurs qui influencent leur expression. Nous avons identifie une nouvelle famille, valpha5t constituee de deux membres ; valpha5tpseudo qui est non fonctionnel et valpha5t qui presente toutes les caracteristiques d'un segment fonctionnel. Les transcrits valpha5t sont frequemment episses entre valpha5t et calpha au lieu de jalpha-calpha, en effet un site alternatif d'epissage se situe au niveau de la jonction. Ceci restreint la probabilite pour ce gene de produire un transcrit complet. Ce phenomene pourrait etre un facteur de regulation intervenant dans l'exclusion allelique, pour les chaines alpha. Nous avons egalement determine la structure de l'ensemble des genes de la famille valpha2 chez la souris b10. A dans le but d'etudier la complexite des familles valpha. Cette famille est constituee de dix genes valpha2 et plusieurs genes valpha2 sont identiques. Les differentes substitutions observees dans les regions valpha2, ont ete positionnees sur un modele tridimensionnel. La plupart des changements sont situes dans les regions exposees au solvant. Les genes valpha2 de la souris balb/c ont ete identifies en reference a leurs homologues chez la souris b10. A. Le taux de conservation entre les alleles est tres fort, 1 a 3 substitutions seulement etant observees. L'ensemble de ces donnees indique que des duplications et des deletions successives sont impliquees dans l'evolution des familles valpha multigeniques. Enfin, nous avons analyse la diversite des tcr alpha, beta selectionnes par un antigene homologue au sol, le collagene de type ii. Pour cela, nous avons caracterise les regions valpha et vbeta de clones t specifiques de cet antigene qui, une fois injecte, induit une arthritte au collagene de type ii. La diversite des tcr selectionnes par cet antigene est reduite. Un des clones t etudies a la particularite de prevenir la maladie lorsqu'il est injecte a des souris avant immunisation par le collagene de type ii

<p>Utveckling av olika typer av påbyggnadskod till programvaror blir allt vanligare. Dessa som går under samlingsnamnet plugins skiljer sig från vanliga applikationer då de har en annan struktur samt speciella beroenden till applikationsspecifika moduler. Problem uppstår när denna typ av utveckling inte stöds av de vanliga utvecklingsmiljöer som finns ute på företagen och delar som borde vara automatiserade måste utföras manuellt. Syftet med studien är att undersöka hur utvecklingsmiljön för Java på Sandvik IT Services kan anpassas för att stödja plugin-utveckling för IBM Lotus Notes. I denna studie undersöks skillnaden mellan plugin-utveckling och den vanliga Java-utvecklingen på företaget samt hur detta påverkar verktygen i utvecklingsmiljön. Resultatet beskriver hur utvecklingsmiljön kan anpassas för att stödja plugin-utveckling och en lösning för detta föreslås. Slutligen visas en implementation av lösningen i form av en prototyp där utvecklingsmiljön anpassas för plugins med Maven pluginet Tycho.</p>

VILLA “CAPELLO - MORA”: PROGETTO DI RESTAURO E RIFUNZIONALIZZAZIONE Il restauro è da intendere come un intervento diretto sull’opera, e anche come sua eventuale modifica, condotta sempre sotto un rigoroso controllo tecnico-scientifico e storico-critico, se parliamo di conservazione, intendiamo l’operare in un intento di salvaguardia e di prevenzione, da attuare proprio per evitare che si debba poi intervenire con il restauro, che comprende un evento traumatico per il manufatto. Un seconda parola chiave in questo discorso è la “materia” il restauro interviene sulla materia di un monumento e questa costituisce il tramite dei valori culturali antichi, la sua conservazione e il suo restauro garantisce la trasmissione anche dei significati estetici, storici simbolici del costruito. Ma certamente influisce il tempo sulle cose per cui il progetto di restauro non può astenersi dall’intervenire, in una logica di minimo intervento, di reversibilità, di facile lettura. Il concetto di nuovo in un opera antica, concetto che a parere personale, pare centrare in pieno il problema. Il nuovo infatti “deve avere carattere di autonomia e di chiara leggibilità: come l’<<aggiunta>> di Boito deve essere inequivocabilmente opera nuova, come prodotto figurativo e materiale autonomo, chiara ed inequivocabile espressione <<della nostra cultura e del nostro tempo>>”. Ne deriva riassumendo che oggi l’obbiettivo deve essere quello di conservare da un lato senza non sotrarre altra materia alla fabbrica e di valorizzare, ossia aggiungere, nuove “presenze” di cultura contemporanea. Per questo si parlerà di progetto di restauro e rifunzionalizzazione. La fabbrica ha subito nel corso dell’ultimo decennio una serie di “rovinose” manomissioni, che a differenza di quelle operate in tempi più antichi (coincidenti con esigenze funzionali corrispondenti alla logica dell’adattare) appaiano ben più gravi, e contribuiscono a peggiorare la lettura del fabbricato. Il Veneto e soprattutto la zona intorno a Bassano del Grappa presenta una infinità di dimore padronali casini di caccia, resti di antiche residenze (colombare, oratori, ecc.) risalenti al periodo di maggior fioritura della residenza di “Villa” della Serenissima. Nel caso specifico di studio , quindi della rifunzionalizzazione dell’edificio, la domanda sorge spontanea; Come ristabilire un senso a questi spazi? E nell’ipotesi di poter realmente intervenire che cosa farne di questi oggetti?. E ultimo ma non ultimo in che modo poter ristabilire un “dialogo” con l’edificio in una lettura corretta del suo significato non solo per quel che riguarda la materia ma anche per ciò che ci trasmette nel “viverlo”, nell’usufruirne nel fatto stesso di poterlo vedere, passandoci davanti. tà. Lidea si forma prima ancora da un esigenza del territorio, il comune di Cassola dove ha sede la Villa,pur avendo un discreto numero di abitanti e collocandosi in posizione nevralgica in quanto molto vicino al centro diBasssano (ne è quasi la promulgazione), non possiede uno spazio espositivo /rappresentativo, un edificio a carattere pubblico, sede di “eventi” culturali locali e non. Villa Capello –Mora, potrebbe rispondere bene a questo tipo di utilizzo. Si è deciso di pensare ad un luogo a carattere espositivo che possa funzionare durante tutto l’anno e nei periodi etsivi includa anche il giardino esterno. Il progetto muove da due principi il principio di “reversibilità” e quello del “minimo intervento” sottolinenando volutamente che siano gli ogetti esposti e lo spazio dei locali a fare da protagonisti, Punti chiave nell’interpretazione degli spazi sono stati i percorsi, la “narrazione” degli oggetti esposti avviene per momenti e viene rimarcata nell’allestimento tramite i materiali i colori e le superfici espositive, perché nel momento in cui visito una mostra o un museo, è come se stessi vivendo la narrazione di un qualcosa, oggetto semplice od opera complessa che sia inteso nell’ambito museale-espositivo esso assume una capacità di trasmissione maggiore rispetto a qundo lo stesso ogetto si trova in un contesto differente, la luce e la sua disposizione nello spazio, fanno sì che il racconto sia narrato, bene o male, ancora più importante è lo sfondo su cui si staglia l’opera, che può essere chiuso, come una serie di scatole dentro la scatola (edificio) oppure aperto, con singole e indipendenti pannellature dove l’edificio riveste il carattere proprio di sfondo. Scelta la seconda delle due ipotesi, si è voluto rimarcare nella composizione degli interni i momenti della narrazione, così ad esempio accedendo dall’ingresso secondario (lato nord-ovest) al quale si è dato l’ingresso alla mostra, -superata la prima Hall- si viene, catapultati in uno spazio porticato in origine aperto e che prevediamo chiuso da una vetrata continua, portata a debita distanza dal colonnato. Questo spazio diventa la prima pagina del testo, una prima pagina bianca, uno spazio libero, di esposizione e non di relax e di presentazione dell’evento, uno spazio distributivo non votato solo a questo scopo, ma passibile di trasformazione. A rimarcare il percorso una pavimentazione in cemento lisciato, che invita l’accesso alle due sale espositive a sinista e a destra dell’edificio. Nell’ala a sinistra (rispetto al nord) si apre la stanza con camino seicentesco forse un tempo ad uso cucina? Sala che ospita dei pannelli a sospesi a muro e delle teche espositive appese a soffitto. L’ala di destra, diametralmente opposta, l’unica che al piano terra mantiene la pavimentazione originale, stabiliva il vero atrio d’entrata, nel XVII sec. riconoscibile da quattro colonne tuscaniche centrali a reggere un solaio ligneo, in questa stanza il percorso segnato a terra torna nella disposizione dei pannelli espositivi che si dispongono in un a formare un vero corridoio, tra uno e l’atro di questi pannelli degli spazi sufficienti a intravedere le colonne centrali, i due lati dello stretto percorso non sono paralleli e aprono in senso opposto a due restanti spazi dell’intero salone, La sala suddivisa così in queste tre parti - una di percorrenza due di esposizione - , risulta modificata nella lettura che ne annulla l’originale funzione. Questa nuova “forma”, non vuole essere permanente infatti i pannelli non sono fissi. la narrazione avviene così per momenti, per parti, , che non devono necessariamente far comprendere il “tutto”, ma suggerirlo. Così come per il percorso che non è obbligato ma suggerito. A terminare il piano altre due sale di minore dimensione e di modesto carattere, nelle quali sono presenti pannellature a parete come nella prima sala. L’allestimento prosegue al piano primo a cui si accede tramite due rampe, trattate nel progetto difformemente, in base al loro stato e al loro significato, la rampa più recente (XIX sec) rompe lo spazio al piano nobile stravolgendo l’ingresso a questo salone, che originariamente avveniva salendo dalla scala principale e oltrepassando un disimpegno. La scala ottocentesca introduce direttamente all’ambiente che ora risulta liberato dalle superfetazioni dello scorso secolo, mostrandosi come era stato previsto in origine. Questa rottura dello spazio viene marcata nel progetto da un volume che “imprigiona” la rampa e segna l’inizio della sala espositiva, la sala delle architetture. Al centro della sala in un gioco di pieni e vuoti i pannelli, espositivi, distaccati di poco gli uni dagli altri questa volta non a incorniciare delle colonne ma a richiamo delle pitture murali che raffiguranti una loggia alludono ad una lettura per “parti” e scandiscono lo spazio in una “processione” di eventi. A seguito del “gioco” sono presenti in senso ortogonale ai pannelli sopradescritti ulteriori pannellature appese ad una struttura metallica indipendente dalle capriate lignee. Tale struttura assume ad una duplice funzione, funge sia da irrigidimento della struttura (edificio) che da supporto alla pannellatura, ad essa infatti è collegata una fune, che entrerà in tensione, quando il pannello verrà ad appendersi. Infine questa fune passante da due puleggie permetterà al pannello di traslare da un lato all’altro dell’edificio, bloccandosi nella posizione desiderata tramite freno interno, azionato manualmente.

Denna studie vill ge större inblick och förståelse för individen bakom den kriminella stämpeln, samt bringa insyn i dessa individers egna tankar om sina kriminella handlingar och kampen ut ur kriminaliteten. Avsaknaden av tron på att personliga livshändelser sker utifrån egna handlanden, där personen istället agerar passivt offer i sitt liv är något som återfinns hos interner. Motsatsen till detta är personer som tror på sitt eget handlande, det vill säga internal locus of control. Denna uppsats belyser kriminellas bakgrundshistoria till uppbrottet ur den kriminella livsstilen med fokus på locus of control, vändpunkter och framtidsperspektiv. Tio personer deltog, varav åtta män och två kvinnor i enskilda semistrukturerade intervjuer. Resultatet visade att majoriteten förklarade sin kriminalitet med yttre faktorer. De nådde vändpunkten när rädslan för att förlora sitt liv eller sina närstående blev för stor. Nio av tio ser nu långsiktigt på sin framtid till skillnad från förr.

Chez des lignées de miniporcs homozygotes au CMH, un traitement court par la ciclosporine (CsA)permet d'induire la tolérance aux allogreffes rénales dont le CMH de classe I est allogénique mais dont la classe II est identique au donneur. Cependant, les reins tolérés sont rapidement infiltrés par des lymphocytes T (LT)du receveur. Dans ce travail, nous avons étudié la distribution clonale des LT à la fois chez des animaux tolérants et rejeteurs (non traités par la CsA). Méthode : Dans un premier temps, les séquences nucléotidiques codant pour les 19 régions Vβ, 12 segmetns Jβ de la chaîne b du récepteur à l'antigène des LT (TCR) ont été obtenus. Le polymorphysme de longueur de la région CDR3 des chaînes b incluant ces régions, a été étudié à la fois chez des animaux tolérants et rejeteurs (non traités par la CsA). Résultats : chez les animaux tolérants, nous avons observé 1) une distribution polyclonale parmi les LT circulants mais oligoclonale parmi les LT infiltrant les reins (dominance clonale) 2)la dominance clonale n'affecte pas toujours les mêmes sous familles Vb entre différents animaux 3)sur un même animal la dominance clonale s'approfondit avec le temps après la greffe et est retrouvée identique dans une seconde greffe tolérée, implantée sans CsA 4)une dominance identique à celle du rein est observée après incubation in vitro des cellules d'un receveur tolérant avec des cellules du donneur. Chez les animaux rejeteurs, nous avons trouvé une dominance clonale conservée entre différents animaux aussi bien dans les LT circulants que dans le sang périphérique avant que la dysfonction rénale ne soit détectable. Conclusion : les présents résultats in vivo et in vitro , associés à ceux d'études antérieures, suggèrent que des LT de distribution oligoclonale jouent un rôle actif dans le processus de tolérance aux greffes rénales (ces cellules sont actuellement en cours de caractérisation phénotypique). Enfin, la précocité de la dominance au sein des LT circulants sur des animaux rejeteurs pourrait servir de base à un futur test non invasif de diagnostic précoce de rejet

Dans la plupart des questions écologiques, les phénomènes aléatoires d'intérêt sont spatialement structurés et issus de l'effet combiné de multiples variables aléatoires, observées ou non, et inter-agissant à diverses échelles. En pratique, dès lors que les données de terrain ne peuvent être directement traitées avec des structures spatiales standards, les observations sont généralement considérées indépendantes. Par ailleurs, les modèles utilisés sont souvent basés sur des hypothèses simplificatrices trop fortes par rapport à la complexité des phénomènes étudiés. Dans ce travail, la démarche de modélisation hiérarchique est combinée à certains outils de la statistique spatiale afin de construire des structures aléatoires fonctionnelles "sur-mesure" permettant de représenter des phénomènes spatiaux complexes en écologie des populations. L'inférence de ces différents modèles est menée dans le cadre bayésien avec des algorithmes MCMC. Dans un premier temps, un modèle hiérarchique spatial (Geneclust) est développé pour identifier des populations génétiquement homogènes quand la diversité génétique varie continûment dans l'espace. Un champ de Markov caché, qui modélise la structure spatiale de la diversité génétique, est couplé à un modèle bivarié d'occurrence de génotypes permettant de tenir compte de l'existence d'unions consanguines chez certaines populations naturelles. Dans un deuxième temps, un processus de Poisson composé particulier,appelé loi des fuites, est présenté sous l'angle de vue hiérarchique pour décrire le processus d'échantillonnage d'organismes vivants. Il permet de traiter le délicat problème de données continues présentant une forte proportion de zéros et issues d'échantillonnages à efforts variables. Ce modèle est également couplé à différents modèles sur grille (spatiaux, régionalisés) afin d'introduire des dépendances spatiales entre unités géographiques voisines puis, à un champ géostatistique bivarié construit par convolution sur grille discrète afin de modéliser la répartition spatiale conjointe de deux espèces. Les capacités d'ajustement et de prédiction des différents modèles hiérarchiques proposés sont comparées aux modèles traditionnellement utilisés à partir de simulations et de jeux de données réelles (ours bruns de Suède, invertébrés épibenthiques du Golfe-du-Saint-Laurent (Canada)).

Mature αβ T lymphocytes, defined by expressing functional αβ TCR on the cell surface, are developed in the thymus from precursor cells that pass sequentially through CD4-CD8- double negative (DN), CD4+CD8+ double positive (DP) and CD4+CD8-/CD4-CD8+ single positive (SP) stages. Immature thymocytes can only develop into DP stage, during which TCRα gene recombination occurs, after functional production of TCRβ proteins at the DN stage. Chromatin accessibility to the lymphoid specific recombination activating (RAG1 and RAG2 )gene products (the recombinase) that bind to the recombination signal sequence flanking V, D, and J gene segments has been proposed to be the mechanism underlying this tightly regulated process. However, the precise molecular mechanism remains unclear, especially in the context of wild-type,recombination-active chromatins. The aim of this study is to decipher the factors regulating TCR Vα-to-Jα recombination in wild-type thymocytes. Two approaches were taken; one is to study the epigenetic modifications on histones and RAG1/2-bindings at the TCRα/δ locus in wild-type thymocytes, the other is to search for differentially expressed proteins in DN and DP thymocytes. The first approach would provide information on how epigenetic modifications on chromatins regulate the accessibility to the recombinase. The second approach would provide spectrums of proteins/post-translational modifications potentially involved in regulating the TCRα gene recombination. Results of chromatin immunoprecipitation (ChIP) assays identify the previous unknown roles of histone H3 methylations to the regulation of TCR Vα-to-Jα recombination. Interestingly, our ChIP data provide solid evidence to support the presence of recombination center as one of the many levels in regulating V(D)J recombination. We also identified differentially expressed proteins factors by the 2D gel electrophoresis and 1D LC-MS-MS analysis. We expect further detail analyses on these proteins would provide important molecular information on V(D)J recombination regulation in vivo.

The concept of dignity typically brings to mind an idea of moral status that supposedly belongs to all humans equally, and which serves as the basis of human rights. But this moralized meaning of dignity is historically very young. Until the mid-nineteenth century, dignity suggested an idea about merit: it connoted elevated social rank, of the sort that marked nobility or ecclesiastic preferment. What explains this radical change in meaning? And before this change, did anything like the moralized concept of dignity exist, that is, before it was named by the term “dignity”? If so, exactly how old is the moralized concept of dignity? In this volume, leading scholars across a range of disciplines attempt to answer these questions by clarifying the presently murky history of “dignity,” from classical Greek thought through the Middle Ages and Enlightenment to the present day. In the process, four platitudes about the history of human dignity are undermined: (1) the Roman notion of dignitas is not the ancient starting point of our modern moralized notion; (2) neither the medieval Christian doctrine of imago Dei nor the renaissance speech of Pico della Mirandola, Oration on the Dignity of Man, was a genuine locus classicus of dignity discussion; (3) Immanuel Kant is not the early modern proprietor of the concept; (4) the universalization of the concept of dignity in the postmodern world (ca. 1800–present) is not the result of its constitutional indoctrination by the “wise forefathers” of liberal states like America or France.

The 1970s were marked by dramatic changes in government policies during the governorship of Sir Murray MacLehose. The New Territories became the locus of a planned population shift away from crowded Hong Kong and Kowloon, and the idea of ‘New Towns’, based on Tsuen Wan’s developments, was central. It went hand-in-hand with infrastructural developments, which revolutionized transportation and ended the relative isolation of Tsuen Wan through the MTR. The administration of Tsuen Wan fundamentally changed and the creation of District Boards (later Councils), beginning first in Tsuen Wan, altered the system of political consultation throughout Hong Kong. The 1980s were marked by dramatic changes in economic policies within China, not least in the areas of China adjacent to Hong Kong, which saw industrialization through the Pearl River delta region and de-industrialization in Tsuen Wan. Emergence of a distinctive Hong Kong identity, and a new political culture, emerged as sovereignty over Hong Kong was resumed by China in 1997. Tsuen Wan changed from the working class town that it has been into one dominated by middle class housing developments, much like the rest of Hong Kong, although the villages of the original inhabitants remained.

This chapter discusses ‘grace’ in the context of the Italian Renaissance. During this time, the term became a mark of distinction in the questione della lingua and in the new language of literature. It was reenergised by the recovery of ancient texts that extolled its virtues as an instrument of persuasion in the language and visual arts. And it was the central bone of contention in Reformation and Counter-Reformation discussions about the nature of God's intervention in human salvation. Within each of these contexts, grace became a defining quality that Italians made their own. Grace provides a unique perspective on sixteenth-century Italy, for it rose to prominence in the context of so-called High Renaissance art, yet it also played a pivotal role in its polemical progress towards Mannerism. It was not, therefore, a banner that united artists in their advance towards the full maturity of their discipline, but a locus of encounter and conflict between different ways of conceiving of the visual arts.

O objetivo deste artigo é analisar e estabelecer aproximações entre o atual ensino superior e as reformas educacionais efetivadas pelo Estado brasileiro. Como embasamento teórico utilizou-se algumas ideias de Marx, o qual estabelece críticas contundentes ao Estado que assume a função de garantir, através de seu aparato jurídico e repressivo, a reprodução do capital, em que pese a relação entre exploração e acúmulo de riqueza, geradora das mais diversas formas de desigualdade. A análise às ideias marxianas está contida na primeira parte deste texto. Em seguida, busca-se perceber qual a influência do capital na educação superior brasileira, tendo-se em vista acontecimentos recentes que (re)afirmam o ensino superior como locus produtor de conhecimento a partir dos interesses da classe dominante, a qual acumula riquezas advindas desta produção. Os contemporâneos ataques do governo brasileiro às Ciências Humanas – que possibilitam questionamentos e mudanças na sociedade, confirmam esta lógica produtivista. Nesse contexto, o que se verifica é o entendimento da educação como algo a ser consumido e produtora de riqueza de iniciativa privada, em obediência aos mandos e desmandos do mercado. Conclui-se que, segundo a análise marxiana, o Estado está para o capitalismo como a principal ferramenta que permite a sobreposição do capital sobre o trabalho, já que os interesses da classe burguesa determinam as ações e/ou concessões que o Estado está autorizado, no sentido próprio do poder, a direcionar à classe trabalhadora.

O texto apresenta as mudanças realizadas na disciplina de Urbanismo do curso de graduação em engenharia civil do Centro Universitário do Instituto Mauá de Tecnologia - CEUN-IMT, a partir da revisão das competências esperadas e de alterações na metodologia de ensino. A verificação dos objetivos da disciplina anual de urbanismo indicou a necessidade de se repensar a estrutura cronológica da abordagem de formação das cidades bem como da coleta e análise de dados urbanísticos tradicionalmente empregada. A reestruturação incluiu a forma de realizar a leitura da cidade, a maneira de se incorporar conceitos e utilizar instrumentos urbanísticos com o emprego de recursos tecnológicos mais atuais e de ferramentas BIM buscando aquelas que melhor se ajustavam aos objetivos de cada etapa do desenvolvimento projetual. Diferentes alterações, desde 2017, foram pensadas para a disciplina de urbanismo do curso de engenharia civil do CEUN-IMT. Sua estrutura estava dividida em conhecimentos históricos, instrumentos legais e desenvolvimento de projeto de loteamento. A disciplina de urbanismo possui carga horária de 80 horas em regime anual de oferecimento e faz parte de um conjunto de áreas com inserção de conteúdos BIM conforme projeto pedagógico do curso. O módulo que realizava a abordagem histórica foi substituído por discussões temáticas e análises morfológicas da cidade sob os aspectos da água, circulação, moradia e trabalho com o intuito de mostrar aos alunos da engenharia civil a importância da observação crítica do espaço construído e de suas infraestruturas. Os exercícios práticos, associados a esta etapa inicial, passaram a utilizar o Google Earth Pro na análise da forma das cidades considerando água e meio ambiente, circulação e moradia. Os instrumentos urbanísticos e legislação passaram a ser discutidos de maneira contextualizada e a partir de “cases” e problemas que ilustram de forma concreta a relação entre o regramento e o espaço urbano dele decorrente. A clássica forma de elaboração de mapas temáticos para o planejamento urbano foi substituída pela consulta aos Sistemas de Informação Geográfica – SIG, que permitem hoje a obtenção de informações associadas a bases cartográficas. Os alunos foram incentivados a utilizar estas informações para interpretar e avaliar as características de diferentes locais com a utilização de mapas online e dados abertos de plataformas como “Urban Atlas” da “European Environment Agency”, “NASA - National Aeronautics and Space Administration” e Geosampa que reúne dados georreferenciados da cidade de São Paulo. Os loteamentos eram desenvolvidos em Autocad e Revit com a aplicação da lei de parcelamento do solo e da legislação ambiental e davam ênfase à distribuição de lotes e vias em grandes glebas. A partir de uma avaliação a respeito das competências a serem adquiridas pelos engenheiros civis para trabalhar em equipes multidisciplinares na área do urbanismo e do planejamento urbano foram realizadas sucessivas alterações até que em 2020 foram estruturadas as novas linhas de abordagem e metodologias com o uso de ferramentas BIM que melhor se ajustavam aos objetivos das diferentes etapas do desenvolvimento da disciplina. Paralelamente, e logo após a finalização das análises temáticas, inicia-se o desenvolvimento de propostas para uma gleba urbana, objeto de edital público para a construção de habitações, comércio, equipamentos públicos, edifícios mistos, áreas verdes, galpões logísticos dentre outros. As tentativas anteriores contemplaram a modelagem em REVIT de todo o estudo preliminar e também a modelagem deste diretamente no Infraworks sem passar por outro software. A experiência de 2020 mostrou-se mais adequada ao estudo de viabilidade do empreendimento, simulações com diferentes densidades e posterior estudo de massas: setorização inicial em Autocad, massas no Revit e modelagem no Infraworks. Passar do desenho de vias e lotes para o desenvolvimento de possibilidades de ocupação em glebas utilizando instrumentos legais e ferramentas urbanísticas representou um ganho para a disciplina. A própria disciplina concebia a cidade em duas dimensões ao utilizar apenas o desenho em Autocad. A utilização do Revit no estudo de viabilidade não fez muito sentido, faltava uma etapa de estudo da topografia em Autocad e também uma primeira concepção que permitisse alterações rápidas. A modelagem precisa do Revit não atendia a necessidade de mudanças rápidas, mesmo utilizando os recursos de criação de massas. A utilização direta do Infraworks mostrou-se também inadequada por não permitir o estudo um pouco mais rigoroso e com dimensões precisas. Equilibrar o ensino de conceitos específicos desta área de conhecimento e o ensino de ferramentas tecnológicas aplicadas à prática projetual, dentro do tempo destinado à disciplina, é um desafio e ponto fundamental para a obtenção de resultados positivos e avanços nesta área. A disciplina de projeto de rodovias e vias urbanas, oferecida em paralelo à de urbanismo, passará a contribuir, a partir do segundo semestre de 2021, com o desenvolvimento de parte do projeto com a modelagem em Civil 3D de vias e ciclovias previstas nas diretrizes do edital adotado para a disciplina e que posteriormente será importada no Infraworks. Para os próximos oferecimentos existe a previsão de utilização do Spacemaker, software que utiliza sistema baseado em inteligência artificial, recentemente adquirido pela Autodesk e que permite explorar diferentes soluções de forma mais rápida tornando os processos de planejamento mais eficientes.
