Le récepteur du mannose-6-phosphate cation indépendant (RM6P-CI) permet l'endocytose puis le transfert de molécules porteuses du marqueur M6P vers les lysosomes. Pour améliorer à la fois l'affinité pour le RM6P-CI et la stabilité du résidu M6P, nous avons procédé à la synthèse d'analogues isostères stables et fonctionnalisés en position anomère pour permettre un couplage efficace à des molécules d'intérêt thérapeutique. Tout d'abord un couplage à des enzymes recombinantes humaines a été réalisé. Le remodelage de la partie oligosaccharidique de l'enzyme lysosomale GAA, dont la déficience est responsable de la maladie de Pompe, a permis de mettre en évidence que la néoglycoGAA est reconnue efficacement par les RM6P-CI et que son activité enzymatique est totalement conservée. Deuxièmement, le couplage de ces analogues du M6P à des porphyrines en vue de la thérapie photodynamique des cancers est envisagé. Le modèle mis au point en série du mannose a permis de valider notre approche de ligation de saccharides à des photosensibilisateurs par des méthodes évitant après couplage les étapes classiques de déprotection des saccharides. L'étude biologique menée sur les porphyrines glycosylées préparées a démontré leur cytotoxicité photoinduite<br>The cation independent mannose-6-phosphate receptor (CI-M6PR) allows the endocytosis and the transfer of molecules bearing the M6P marker to lysosomes. To improve both the affinity for the CI-M6PR and stability of the M6P residue, we carried out the synthesis of isosteric M6P analogues functionalized at the anomeric position to allow efficient coupling to molecules of therapeutic interest. First, the coupling on human recombinant enzymes was performed. The remodelling of the oligosaccharide part of the lysosomal enzyme GAA, whose deficiency is responsible for Pompe disease, helped to highlight the neoglycoGAA is recognized efficiently by CI-M6PR and its enzymatic activity is completely preserved. Second, the coupling of these analogues of M6P to porphyrins for photodynamic therapy of cancer was considered. The model developed in the mannose series has validated our strategy of ligation of saccharides to photosensitizers. The employed methods avoid the conventional steps of deprotection of saccharides after coupling. The biological study with the prepared glycosylporphyrins demonstrated the photoinduced cytotoxicity

Les lysosomes sont importants pour la survie et la fonction des cellules du système nerveux central et en particulier des neurones. Le mécanisme de la reformation des lysosomes est crucial pour maintenir une quantité adéquate de lysosomes fonctionnels dans les cellules. La spatacsine, qui joue un rôle dans le ce mécanisme est impliquée dans la paraplégie spastique de type SPG11 ; une maladie caractérisée par des troubles moteurs et cognitifs sévères. L’utilisation de modèles cellulaires de cette pathologie permet d’étudier les mécanismes physiopathologiques à l’origine d’altérations de la reformation des lysosomes. J’ai montré que la perte de fonction de la spatacsine est responsable de l’accumulation de lipides dans les lysosomes. Ces accumulations sont constituées de gangliosides et de cholestérol et sont présentes dans les autolysosomes perturbant leur recyclage en lysosomes, notamment en empêchant le recrutement de protéines impliquées dans le mécanisme. Les accumulations de gangliosides rendent les neurones à l’exposition au glutamate ce qui suggère que ces altérations pourraient avoir un rôle dans la neurodégénérescence. J’ai aussi montré que l’absence de spatacsine provoque une dérégulation de l’import de Ca2+ extracellulaire par le « store-operated calcium entry » ce qui conduit à altération de l’homéostasie calcique. L’inhibition de l’import de calcium par le SOCE permet de réduire les accumulations de lipides et de rétablir partiellement le recyclage des lysosomes. Ainsi, l’absence de spatacsine induit une altération de l’homéostasie calcique qui participe à l’accumulation de lipides dans le système lysosomal ce qui est délétère pour la survie des neurones<br>Lysosomal dysfunctions are involved in a large number of neurodegenerative diseases highlightingthe crucial function of lysosomes in neuron survival and function. The mechanism of lysosomereformation from autolysosomes allow cells to maintain the ool of functional lysosomes.Disruptions of this rocess are involved in athologies affecting the central nervous system. Inparticular, spatacsin that lays a role in lysosome recycling is implicated in hereditary spasticparaplegia type SPG11, a severe disease characterized by motors and cognitive alterations. Thispathology is caused by loss of function mutations in SPG11, encoding spatacsin. The study ofSPG11 cellular models gives the opportunity to decipher the hysiopathological mechanismsunderlying lysosomal reformation disruptions.During my thesis, I showed that loss of spatacsin function induces lipid accumulation in lysosomesand articularly in autolysosomes both in fibroblasts and neurons from Spg11-/- mice. Gangliosidesand cholesterol are among lipids that accumulate in autolysosomes impairing lysosomal membranerecycling by disrupting the recruitment of keys roteins. Neurons with ganglioside accumulationsare more sensitive to glutamate induced neuronal death, suggesting that these accumulations areinvolved in neurodegeneration. These results could be of great importance since accumulations ofgangliosides in lysosomes arise in many diseases.I also showed that loss of spatacsin disrupts extracellular calcium import by the store-operatedcalcium entry (SOCE) leading to an increase in cytosolic calcium levels. Lysosomal calcium contentis also increased in Spg11-/- cells and calcium release from lysosome by TRPML1 is reduced.Inhibiting SOCE and stimulating lysosomal calcium release by TRPML1 reduced lipidsaccumulations in lysosomes and artially restored lysosome reformation.Our data suggest that absence of spatacsin is responsible for a disruption of calcium homeostasisthat contributes to lipid accumulation in autolysosomes, disturbing reformation of lysosomes fromautolysosomes. Inhibiting gangliosides synthesis could be used as a therapeutic strategy. However,understanding how loss of function of spatacsin alters these cellular athways will allow thedevelopment of targeted therapeutic approaches

La réaction inflammatoire est la première étape nécessaire pour restaurer l'homéostasie tissulaire lésée. Elle implique au cours de ses différentes étapes, le système immunitaire, notamment les macrophages qui présentent alors divers remaniements inflammatoires et métaboliques. Les macrophages sont capables de modifier et d'adapter leur métabolisme cellulaire afin de satisfaire leurs besoins énergétiques et réaliser de façon efficace la réaction inflammatoire en fonction des signaux du milieu environnant. Ces adaptations métaboliques influencent la physiologie des mitochondries et les oxydations phosphorylantes (OXPHOS), les sécrétions de molécules pro- et antiinflammatoires, ainsi que la capacité à réaliser la phagocytose (pathogènes, débris cellulaire, autophagie) dépendante de la physiologie des lysosomes. Ces adaptations métaboliques sont sous le contrôle de plusieurs facteurs de transcriptions comme NFkB, TFEB ou HIF1alpha. La compaction et l'accessibilité de la chromatine sont des éléments cruciaux pour la régulation indirecte de l'activité de ces facteurs de transcription. Dans le noyau, la compaction de l'ADN est régulée par les histones mais aussi par d'autres protéines de la famille des High Mobility Group (HMG). Parmi les protéines HMG, la protéine High Mobility Group B1 (HMGB1) principalement localisée dans le noyau est capable de réguler de façon indirecte la transcription de gènes dans de nombreux tissus. En plus de son rôle nucléaire, HMGB1 peut être activement relocalisé dans le cytoplasme puis sécrété par les cellules de l'immunité innée au cours d'inflammation aiguë ou chronique. Une fois dans la circulation sanguine HMGB1 joue le rôle d'alarmine qui initie et maintient l'inflammation. De plus, chez la souris au cours d'une inflammation aiguë ou chronique les concentrations d'HMGB1 circulant sont augmentées par rapport à la condition basale. Tous ces résultats suggèrent un rôle d'HMGB1 dans l'immunométabolisme des macrophages ainsi que dans les processus inflammatoires aiguës ou chroniques. Dans ce contexte, ce travail de thèse s'articule autour de deux objectifs : I/: Étudier le rôle d'HMGB1 dérivé des macrophages et de ses conséquences sur la survenue de la fibrose. II/: Étudier le rôle intracellulaire d'HMGB1 dérivé des macrophages au cours du choc inflammatoire aigu. Ces travaux ont permis de démontrer in vitro et in vivo qu'en tant qu'alarmine HMGB1 dérivé des macrophages n'a pas d'influence sur la survenue de la fibrose suite à une nécro-inflammation chronique. De plus, ces travaux ont permis de démontrer in vitro et in vivo qu'en tant que facteur nucléaire, HMGB1 exerce un puissant effet antiinflammatoire en favorisant la polarisation de type M2, en jouant notamment sur la biogénèse et la fonction des lysosomes. Tous ces résultats pris ensemble ont permis de mieux caractériser et comprendre les fonctions biologiques de la protéine HMGB1 dérivée des macrophages au cours du choc inflammatoire aigu. Ce travail permet aussi de mieux envisager des approches thérapeutiques autour de la fonction d'HMGB1 afin d'éviter toute hyper-réaction inflammatoire aux effets délétères pour l'organisme chez des patients atteints de pathologies inflammatoires aiguës/chroniques<br>The Inflammatory reaction is the first necessary step to combat all pathogens and tissue injuries and restore damaged tissue homeostasis. The immune system is particularly involved during each step, in particular the macrophages which display various inflammatory and metabolic changes. Macrophages can modify and adapt their cellular metabolism to meet their energy needs and efficiently perform the inflammatory reaction according to signals from the surrounding environment. These deep metabolic adaptations influence mitochondria physiology and oxidative phosphorylations (OXPHOS), the secretions of pro and anti-inflammatory molecules, as well as the ability to phagocytize various inflammatory compounds (pathogens, cell debris, and autophagy). These deep metabolic adaptations are under the control of several transcription factors such as NFkB, TFEB or HIF1alpha. The compaction and accessibility of chromatin are crucial for the regulation of the activity of these transcription factors. In the nucleus, DNA compaction is regulated by histones but also by High Mobility Group (HMG) proteins. Among this family of HMG proteins, the High Mobility Group B1 (HMGB1) protein, mainly located in the nucleus, is capable of regulating indirectly the transcription of genes in many tissues. In addition to its nuclear role, HMGB1 can be actively relocated into the cytoplasm and then secreted by innate immune cells during acute or chronic inflammation. Once in the bloodstream, HMGB1 acts as alarmine which initiates and maintains inflammation. Furthermore, during acute or chronic inflammation, concentrations of circulating HMGB1 are increased compared to the basal condition in mice. All these results suggest a role of HMGB1 in the immunometabolism of macrophages as well as in acute or chronic inflammatory processes. In this context, this thesis work has two objectives: I /: To study the role of HMGB1 derived from macrophages and its consequences on the occurrence of tissue fibrosis. II /: To study the intracellular role of HMGB1 derived from macrophages during acute inflammatory shock. This work has demonstrated in vitro and in vivo that, as an alarmine HMGB1 derived from macrophages, does not influence the occurrence of fibrosis following chronic inflammation. Moreover, we demonstrated in vitro and in vivo that as a nuclear factor HMGB1 exerts a potent anti-inflammatory action on macrophages by regulating lysosome biogenesis and function and skewing towards a M2 profile. All these results taken together helped to better characterize and understand the biological functions of HMGB1 proteins during the inflammatory reaction. Boosting these anti-inflammatory functions of HMGB1 may constitute a potential therapeutic approach to counteract the deleterious effect of hyper-inflammation in patients with acute/chronic inflammatory diseases

The amyloidoses are a group of debilitating disorders which include neurodegenerative diseases such as Alzheimer’s disease and systemic diseases such as dialysis-related amyloidosis (DRA). Amyloidoses are associated with the aggregation of proteins into amyloid fibrils with a highly organised cross-β structure. Amyloid fibrils are formed by a variety of proteins and peptides despite differences in sequence and native structure. Amyloid formation occurs by a nucleated growth mechanism and proceeds via oligomeric intermediates into mature fibrils. Despite intense research, the molecular mechanisms involved in disease pathogenesis remain unclear. This thesis discusses the mechanism by which amyloid fibrils cause cellular disruption. Chapter 3 describes work performed to validate the use of β2-microglobulin (β2m), the protein that self-associates into amyloid fibrils found in DRA deposits, as a model to study amyloidosis. Fragmentation of mature β2m fibrils, increased their internalisation and access to intracellular compartments, and were therefore used to investigate mechanisms of cellular disruption. Building on previous work in the laboratory showing trafficking of β2m fibrils to the lysosome, chapter 4 examined the effect of fragmented fibrils on lysosomal function and demonstrates that fragmented fibrils impair lysosome-mediated degradation of endocytosed proteins. Following on from this, chapter 5 discusses the effect of fragmented fibrils on membrane trafficking. Fragmented fibrils perturbed the trafficking of lysosomal membrane proteins and also reduced the trafficking of endocytosed cargo to lysosomes. This may rationalise the impairment in degradation of endocytosed proteins. The molecular chaperone, heat shock protein 70 (Hsp70) has been shown to be protective in amyloid disease. The role of Hsp70 in fibril-mediated cell disruption was investigated in chapter 6. Hsp70 protected fibril-treated cells from impairment in degradation of endocytosed protein but not from membrane trafficking defects. This work demonstrates that fragmented fibrils are nanoparticles which target lysosomes and implicates the lysosome in the pathogenesis of amyloidosis.

Ca2+ is a universal and versatile intracellular messenger, regulating a vast array of biological processes due to variations in the frequency, amplitude, spatial and temporal dynamics of Ca2+ signals. Increases in cytosolic free Ca2+ concentration ([Ca2+]c) are due to influx from either an infinite extracellular Ca2+ pool or from the more limited intracellular Ca2+ stores. Stimulation of the endogenous muscarinic (M3) receptors of human embryonic kidney (HEK) cells with carbachol results in the activation of phospholipase C (PLC) and formation of inositol 1,4,5-trisphosphate (IP3), activation of IP3 receptors (IP3Rs), release of Ca2+ from the endoplasmic reticulum (ER), and activation of store-operated Ca2+ entry (SOCE). Lysosomes are the core digestive compartments of the cell, but their importance as signalling organelles is also now widely appreciated. Accumulating evidence indicates that lysosomal Ca2+ is important for their physiological functions. Lysosomal Ca2+ release triggers fusion during membrane trafficking and, through calmodulin, it regulates lysosome size. Luminal Ca2+ is critical for regulation of lysosomal biogenesis and autophagy during starvation through the transcription factor, TFEB. Furthermore, aberrant lysosomal Ca2+ is associated with some lysosomal storage diseases. Lysosomes in mammalian cells have long been suggested to accumulate Ca2+ via a low-affinity Ca2+-H+ exchanger (CAX). This is consistent with evidence that dissipating the lysosomal H+ gradient increased [Ca2+]c and decreased lysosomal free [Ca2+], and with the observation that lysosomal Ca2+ uptake was followed by an increase in pHly. Furthermore, heterologous expression of Xenopus CAX in mammalian cells attenuated carbachol-evoked Ca2+ signals. However, there is no known CAX in mammalian cells, and so the identity of the lysosomal Ca2+ uptake pathway in mammalian cells is unresolved. Using mammalian cells loaded with a fluorescent Ca2+ indicator, I show that dissipating the pHly gradient pharmacologically or by siRNA-mediated knockdown of an essential subunit of the H+ pump, increases the amplitude of IP3-evoked cytosolic Ca2+ signals without affecting those evoked by SOCE. A genetically encoded low-affinity Ca2+ sensor expressed on the lysosome surface reports larger increases in [Ca2+]c than the cytosolic sensor, but only when the Ca2+ signals are evoked by IP3R rather than SOCE. Using cells expressing single IP3R subtypes, I demonstrate that each of the three IP3R subtypes can deliver Ca2+ to lysosomes. I conclude that IP3Rs release Ca2+ within near-lysosome microdomains that fuel a low-affinity lysosomal Ca2+ uptake system. The temporal relationship between the increase in pHly and reduced Ca2+ sequestration suggests that pHly affects the organization of the microdomain rather than the Ca2+ uptake mechanism. I show that abrogation of the lysosome H+ gradient does not acutely prevent uptake of Ca2+ into lysosomes, but disrupts junctions with the ER where the exchange of Ca2+ occurs. The dipeptide, glycyl-L-phenylalanine 2-naphthylamide (L-GPN), is much used to disrupt lysosomes and release Ca2+ from them. The mechanism is widely assumed to require cleavage of GPN by cathepsin C, causing accumulation of amino acid residues, and osmotic lysis of lysosomal membranes. I show, using LysoTracker Red and Oregon Green-dextran to report pHly, that L-GPN is effective in HEK cells lacking functional cathepsin C, following CRISPR-Cas9-mediated gene disruption. Furthermore, D-GPN, which is resistant to cleavage by cathepsin C, is as effective as L-GPN at increasing pHly, and it is similarly effective in cells with and without cathepsin C. L-GPN and D-GPN increase cytosolic pH, and the effect is similar when the lysosomal V-ATPase is inhibited with bafilomycin A1. This is not consistent with GPN releasing the acidic contents of lysosomes. I conclude that the effects of GPN on lysosomes are not mediated by cathepsin C. Both L-GPN and D-GPN evoke Ca2+ release, the response is unaffected by inhibition or knock-out of cathepsin C, but it requires Ca2+ within the ER. GPN-evoked increases in [Ca2+]c require Ca2+ within the ER, but they are not mediated by ER Ca2+ channels amplifying Ca2+ release from lysosomes. GPN increases [Ca2+]c by increasing pHcyt, which then directly stimulates Ca2+ release from the ER. I conclude that physiologically relevant increases in pHcyt stimulate Ca2+ release from the ER independent of IP3 and ryanodine receptors, and that GPN does not selectively target lysosomes. I conclude that all three IP3R subtypes selectively deliver Ca2+ to lysosomes, and that the low pH within lysosomes is required to maintain the junctions between ER and lysosomes, but not for lysosomal Ca2+ uptake. I suggest that GPN lacks the specificity required to allow selective release of Ca2+ from lysosomes.

Les lysosomes jouent un rôle clé dans divers mécanismes de résistance aux traitements anticancéreux, notamment en piégeant les médicaments à l’intérieur de leur structure et en activant des voies de stress adaptatives. Si cibler la transcription s’est révélée être une stratégie prometteuse contre les cellules cancéreuses, les mécanismes sous-jacents de résistance aux inhibiteurs de la transcription demeurent largement méconnus. Mon projet de thèse vise à explorer le rôle des lysosomes dans les mécanismes de résistance induits en réponse à deux inhibiteurs puissants de l’ARN POL I (composés A et B). Nous avons découvert que le composé A était séquestré dans les lysosomes où il induisait la perméabilisation de la membrane lysosomale (LMP). Mes résultats ont également révélé que l'induction de ce stress lysosomal entraîne l'activation à la fois du facteur de transcription TFEB et de l'autophagie, jouant des rôles cytoprotecteurs. De plus, cibler les lysosomes, en utilisant des dérivés de la chloroquine ou une excitation par lumière bleue, induit une augmentation importante de la LMP, suivie de la libération du composé A des lysosomes. D’un point de vue mécanistique, j’ai montré que l’induction de cette LMP massive amplifie à la fois l’inhibition de la transcription et la mort cellulaire induite par le composé A. Des effets similaires ont été observés quand les dérivés de la chloroquine sont combinés à l’autre inhibiteur de POL I (composé B). Enfin, nous avons confirmé l'effet bénéfique de l'association du composé A et du DC661 (un dérivé de la chloroquine) dans un modèle in vivo de xénogreffe de fibrosarcome chez des souris immunocompétentes. En conclusion, nous avons découvert des mécanismes liés aux lysosomes qui contribuent de manière inattendue à la résistance à deux inhibiteurs de la transcription médiée par la POL I (composés A et B). Cette étude suggère l'utilisation de combinaisons synergiques d'inhibiteurs de la transcription de POL I avec des agents ciblant les lysosomes tels que les dérivés de chloroquine ou avec la photothérapie pour lutter contre la résistance à ces traitements<br>Lysosomes have been known to contribute to the development of drug resistance through a variety of mechanisms that include the sequestration of drugs within their compartments and the activation of adaptive stress pathways. Although targeting RNA Polymerase I (POL I) has shown anticancer effects, the contribution of lysosomes to the efficacy and resistance of RNA POL I inhibitors remains largely unknown. In this study, we investigated this aspect in the context of two potent POL I transcription inhibitors (compounds A and B). We found that they were unexpectedly sequestered in lysosomes, causing lysosomal membrane permeabilization (LMP). This effect activated the transcription factor TFEB resulting in cytoprotective autophagy. Targeting lysosomes using chloroquine derivatives or blue light excitation induced substantial LMP, resulting in the liberation of the compound A from lysosomes. This effect amplified both the inhibition of DNA-to-RNA transcription and cell death induced by both POL I inhibitors. Moreover, combining compound A with the chloroquine derivative DC661 reduced the growth of fibrosarcoma established in immunocompetent mice more efficiently than did monotherapies with each agent. Altogether, our results reveal an unanticipated lysosome- related mechanism that contributes to resistance to POL I transcription inhibitors, as well as a strategy to combat this resistance

Les cellules malignes acquièrent des avantages compétitifs qui permettent une croissance et prolifération anormales, grâce à un large spectre de changements génétiques et épigénétiques conduisant à des changements majeurs dans les profils de transcriptome et protéome et ainsi des modifications dans des voies de signalisation, le trafic intracellulaire et le métabolisme. Des nombreuses voies cellulaires ont été étudiées dans le contexte du cancer, y compris la signalisation, la migration, la perte de la polarité cellulaire apico-basale et l'adhésion cellulaire, cependant très peu est connu sur les altérations au niveau des organelles. Cette thèse a comme objectif d'identifier des altérations dans les compartiments intracellulaires et d'étudier leurs corrélations avec la progression du cancer. Dans la culture cellulaire classique, l'étude systématique de l'organisation du positionnement relatif des organelles est difficile en raison des fortes hétérogénéités morphologiques des cellules. Pour contourner ce problème, nous avons utilisé l’innovante technique des micro-patrons combinée à des cartes de densité des organelles. Après une analyse systématique de différentes lignées cellulaires représentant différents grades du cancer de la vessie, nous avons identifié des changements dans le positionnement de plusieurs organelles. Le changement de position le plus important a été observé pour les lysosomes, dont la distribution était plus périphérique dans les cellules représentant des grades plus avancées du cancer de la vessie. Ceci suggère que le positionnement des lysosomes pourrait être potentiellement important dans la progression du cancer. Par conséquent, nous avons cherché à caractériser l'impact de l’altération des lysosomes sur le comportement des cellules transformées. Nous avons constaté que les changements dans le positionnement des lysosomes jouent un rôle dans l'invasion des cellules cancéreuses de la vessie. En effet, le transport antérograde des lysosomes est en corrélation avec l’invasion 3D, contrairement au transport rétrograde qui corrèle avec une diminution de l’invasion cellulaire. Enfin, nous avons étudié les mécanismes moléculaires par lesquels les altérations du lysosome ont un impact sur l'invasion cellulaire<br>Cancer is a multifactorial disease defined by a rapid development of abnormal cells. Malignant cells acquire competitive advantages for growth and proliferation through a big spectrum of genetic and epigenetic changes leading to major changes in the transcriptome and proteome profiles and thus to alterations in multiple signaling pathways, intracellular trafficking and metabolism. Although many cellular pathways have been studied in the context of cancer, including signaling, migration, loss of apical-basal cell-polarity and cell adhesion, little is known about cancer-related alterations on the sub-cellular, organelle level. This PhD thesis aimed to identify alterations in intracellular compartments and to study how these changes correlate with cancer progression. In classical culture, the systematic study on the organization and relative positioning of organelles is challenging because of the strong morphological cell-to-cell variations. To overcome this problem, we used innovative micro-patterning technique in combination with quantitative, probabilistic mapping of cell organelles. Using a systematic analysis of different cell lines representing different stages of bladder cancer, we identified several changes in the positioning of organelles. The most striking phenotype was revealed by lysosomes, whose distribution was more peripheral in cells representing higher grades of bladder cancer. This suggested that lysosome positioning could be potentially relevant in cancer progression. Therefore, we aimed to characterize the impact of lysosome alteration on cell behavior in transformed cells. We found that changes in lysosome positioning played a role on bladder cancer cell invasion. Indeed, anterograde transport of lysosomes correlate with 3D invasion behavior, contrary to retrograde transport that correlated with decreased cell invasion. De nombreuses fonctions des lysosomes sont impliquées dans le cancer et ils restent donc une cible intéressante pour les thérapies anticancéreuses. Nous avons précédemment étudié le paysage intracellulaire de cet organite dans une collection de lignées cellulaires de cancer de la vessie et de cellules urothéliales humaines normales et nous avons constaté que les lysosomes sont plus dispersés vers la périphérie des cellules dans les cellules agressives de cancer de la vessie. Nous constatons ici une régulation différentielle de la kinase mTORC1 (cible mammalienne du complexe 1 de rapamycine), qui émet des signaux à partir des lysosomes, entre des lignées cellulaires de cancer de la vessie non agressives et agressives, et nous constatons une translocation nucléaire constitutive du facteur de transcription EB (TFEB) dans les cellules de cancer de la vessie agressives. Le silençage de TFEB dans les cellules cancéreuses agressives inverse le phénotype de dispersion des lysosomes, ce qui suggère un rôle de TFEB dans la régulation de la dispersion lysosomale dans ces cellules. De manière cohérente, nous constatons que l'induction de la translocation nucléaire de TFEB après l'inhibition de mTORC1 induit un mouvement périphérique des lysosomes dans les cellules non agressives. Les phosphoinositols étant des régulateurs importants de la fonction et du mouvement des lysosomes, en particulier le phosphatidylinositol-3-phosphate (PI3P) qui est impliqué dans le positionnement des lysosomes, nous avons testé si TFEB régule les niveaux de PI3P dans les cellules cancéreuses agressives de la vessie et donc la dispersion des lysosomes. Nous constatons que la GFP-FYVE qui se lie à PI3P est fortement diminuée après un traitement avec un siTFEB dans les cellules agressives.L'ensemble de nos résultats indique que le positionnement des lysosomes est sous le contrôle de TFEB et que l'hyperactivation de TFEB conduit au phénotype cellulaire caractéristique de dispersion des lysosomes dans les cellules cancéreuses de la vessie agressives. De plus, nos résultats montrent que l'activation de TFEB entraîne une augmentation globale de PI3P au niveau des lysosomes et un fort recrutement des protéines contenant le domaine FYVE. Ainsi, nos résultats révèlent un nouveau rôle de TFEB dans la régulation des niveaux de PI3P. Many functions of lysosomes are implicated in cancer and thus they remain an interesting target for cancer therapies. We had previously investigate the intracellular landscape of this organelle in a collection of bladder cancer cell lines and normal human urothelium cells and found that lysosomes become increasingly scattered towards the cell periphery in aggressive bladder cancer cells. Here we find differential regulation of mTORC1 (mammalian target of rapamycin complex 1) kinase, which signals from lysosomes, between non- aggressive and aggressive bladder cancer cell lines and we find constitutive nuclear translocation of the transcription factor EB (TFEB) in aggressive bladder cancer cells . Silencing of TFEB in aggressive cancer cells reverses the scattered lysosome phenotype, suggesting a role of TFEB in regulation of lysosomal dispersion in these cells. Consistently, we find that inducing nuclear translocation of TFEB after inhibition of mTORC1 induces peripheral movement of lysosomes in non-aggressive cells. Since phosphoinositols are important regulators of lysosomal function and movement, especially phosphatidylinositol-3-phosphate (PI3P) which is involved in lysosomal positioning, we tested whether TFEB regulates levels of PI3P in aggressive bladder cancer cells and thus lysosomal dispersion. We find that GFP-FYVE that binds to PI3P is strongly decreased after siTFEB in aggressive cells.Together our results indicate that lysosome positioning is under the control of TFEB and that hyperactivation of TFEB leads to the characteristic cellular phenotype of lysosome dispersion in aggressive bladder cancer cells. Moreover, our results show that activation of TFEB leads to a global increase of PI3P at lysosomes and strong recruitment of FYVE-domain-containing proteins. Thus, our findings uncover a novel role of TFEB in regulating PI3Ps levels. This conceptually clarifies the double role of TFEB as regulator of endosomal maturation and autophagy, two fundamental but distinct cellular processes that rely and are regulated by PI3P levels. We propose that lysosome positioning changes are a crucial biomarker of alterations in the PI3P pathway in the bladder cancer model

Our understanding of ionizing radiation and its associated biological effects has recently under- gone a paradigm shift from a DNA-centric model to one inclusive of non-targeted effects (NTE), so called for the lack of direct radiation interaction with DNA. Two effects encompassed within the NTE paradigm are termed genomic instability (GI) and bystander effects (BE). GI can be described as an increase in rate of genetic alterations many cell generations after the initial radi- ation exposure. BE can be defined as the manifestation of radiation like effects in un-irradiated cells that have communicated with cells that have been irradiated either through inter-signalling utilising gap junctions or the secretion of a soluble diffusion signalling factor. The exact mechanisms that underlie these processes are still under investigation but a wealth of evidence suggests that a number of mechanisms are involved. These include; cytokine signalling, oxidative stress, inflammation and sub-cellular alterations, in addition to factors such as genetic background and radiation quality/dose. This study was designed to investigate lysosomal involvement in radiation induced NTE, whether it be downstream of one of the above mentioned mechanisms, or independent of their involve- ment. To this end the primary human fibroblast cell line, HF19, was exposed to X-rays at therapeutic and diagnostic doses of 2 and 0.1 Gy respectively. Bystander groups were also established by media transfer techniques. Cells were analysed over the first 24 hours and then at 1 and 20 population doublings, initially for detection of GI and BE and thus confirm the suitability of the system. The lysosomes were then analysed for permeability and their distri- bution within the cell. Oxidative stress was also measured in a bid to correlate this event with lysosomal perturbations. Finally lysosomal contents, in particular DNaseIIα, were analysed for their cellular location along with analysis of nuclear membrane permeability which we surmised would facilitate the redistribution of lysosomal enzymes. The results demonstrated that HF19 cells were susceptible to the induction of GI and BE. The latter was noted within the first hour following irradiation in both 0.1 and 2 Gy bystander groups. High levels of chromosomal instability were also induced in both 0.1 and 2 Gy directly irradiated groups, 1 population doubling after exposure. Chromosomal instability was still noted at 20 population doublings mainly in the 2 Gy although the 0.1 Gy group did show elevated levels. A similar pattern was observed in the bystander group. However we were unable to detect sustained production of the bystander signal at 20 population doublings. Lysosomal properties were also characterised and measured at corresponding time points; large alterations were observed in the first 24 hours following irradiation, furthermore, the lysosomes appeared more permeable at 20 population doublings especially in bystander groups, however, these changes did not correlate with increases in oxidative stress. As a result we further exam- ined cells for changes in the distribution of lysosomal enzymes, in particular DNaseIIα, however no significant changes were observed. Nuclear permeability was additionally investigated as to whether increased permeability facilitated enzyme redistribution; however permeability ap- peared reduced rather than increased. In summary, our investigations have demonstrated and confirmed that HF19 cells are susceptible to the induction of GI and BE following low LET X-ray exposure. The results suggest that the mechanism of radiation induced GI and BE responses can be correlated with alterations in lysosomal membrane permeability which appears independent of oxidative stress. We also demonstrated that if lysosomes are involved in NTE it is unlikely to be through direct action of DNaseIIα but rather from enzymes such as acid sphingomylinase. To conclude, radiation was able to alter lysosomes and nuclear permeability at delayed time points which was correlated with GI , however it appears DNaseIIα is not involved. It also appears that an early effect of the bystander signal may have antioxidant property.

Nous avons étudié l'implication des caractéristiques membranaires et des organites intracellulaires au cours de l'apoptose induite par le benzo[a]pyrène (B[a]P) dans la lignée F258. Nous avons ainsi mis en évidence un effet protecteur du ganglioside GM1, stabilisant membranaire, sur l'apoptose induite par le B[a]P. Cette protection serait liée à une inhibition des dommages mitochondriaux, du stress oxydant et de l'entrée de fer. Nous avons également montré que le B[a]P induisait une augmentation précoce de la fluidité membranaire. Une stabilisation des membranes par le cholestérol a permis d'inhiber l'augmentation de fluidité, l'entrée de fer et l'apoptose. Nous avons de plus observé des modifications de la morphologie et du pH des lysosomes dans les cellules exposées au B[a]P. Ces modifications seraient en partie dues aux désordres mitochondriaux induits par cet hydrocarbure. Nous avons également mis en évidence une implication forte du fer lysosomal dans l'activation de la caspase-3.

Chemotherapy frequently fails to cure cancer patients due to toxicity or resistance to treatment. Variability in toxicity and resistance is influenced by polymorphisms and mutations in the individual's genome (Pharmacogenetics). Zebrafish has extensive evolutionary conservation with human, its genetics is a powerful gene discovery tool, and it has been described as a suitable model in cancer research. To study chemotherapy resistance, we used ENU-mutagenised zebrafish in a forward genetic screen to identify genes that modify responses to cancer chemotherapy. Zebrafish larvae were challenged with two chemotherapeutic drugs and stained with acridine orange (AO) to detect apoptosis and reveal hypo- or hyper-responders to chemotherapy. A mutation, conferring an increased uptake of AO, was identified by genetic mapping as a premature stop codon truncating the ribonuclease T2 (rnaset2) gene. Human RNASET2 encodes a putative lysosomal RNase. Lysosomal storage disorders, due to deficiencies in lysosomal hydrolases and resultant accumulation of macromolecules within lysosomes, are collectively among the commonest genetic diseases. RNASET2 deficiency in man results in a static encephalopathy arising in infancy and characterized by multifocal bilateral white matter lesions, subcortical cysts and focal enlargement of the anterior inferior horn. This doctoral thesis demonstrates that rnaset2 deficient zebrafish embryos suffer from a lysosomal storage disorder accumulating undigested ribosomal RNA (rRNA) in enlarged lysosomes within neurons of the brain. Moreover, high-field intensity μMRI revealed white matter lesions in the brain of adult rnase2 mutant animals. Thus, this zebrafish mutant can be used as a model to study the abnormalities observed in RNASET2 deficient individuals. This model suggests that the leukoencephalopathy results from a lysosomal storage disorder and provides a preclinical model for further elaborating disease mechanisms and evaluating candidate therapeutics.RNASET2 has also been advanced as a candidate tumour suppressor in several solid tumours. Recombinant rnaset2 protein has been tested in the clinic as an anti-cancer cytotoxic agent, with anti-angiogenic properties. By combining the rnaset2 mutant presented here with a transgenic melanoma model developed in the laboratory, the tumour suppressor and angiogenic role for rnaset2 was refuted.

Les lysosomes contiennent environ soixante hydrolases différentes, qui peuvent dégrader une grande variété de macromolécules. L’activité de ces enzymes est dépendante du pH, maintenu dans les lysosomes entre 4.5 et 5.0 par une pompe à protons : la v-ATPase. Les produits de dégradation sont recyclés dans le cytoplasme par des transporteurs actifs secondaires de la membrane des lysosomes.Les maladies de surcharges lysosomales sont causées par des mutations de gènes codant pour des protéines lysosomales, souvent des enzymes. Elles sont caractérisées par un engorgement des lysosomes avec des agrégats ou des cristaux. Les symptômes associés à ces maladies sont variés, mais la moitié d’entre elles induisent des défauts neurologiques. L’étude de ces maladies a permis d’élucider la fonction de nombreuses enzymes, mais la connaissance des transporteurs lysosomaux reste parcellaire. Peu de ces transporteurs sont ainsi caractérisés au niveau moléculaire.Je me suis intéressé à deux gènes dont la mutation provoque une maladie de surcharge particulière : CLN3 et CLN7. Leur mutation provoque des céroïdes lipofuscinoses neuronales, des maladies de surcharge lysosomales caractérisées par une neurodégénérescence précoce et par l’accumulation dans les lysosomes d’un pigment autofluorescent, la lipofuscine. La mutation de 14 gènes différents cause une céroïde lipofuscinose neuronale. J’ai étudié CLN3 et CLN7 car ils codaient pour des protéines membranaires du lysosome, qui pourraient donc être des transporteurs.Sur CLN7, j’ai effectué des tests de transport en utilisant les acides aminés comme substrats potentiels, sans résultats probants. Concernant CLN3, le contenu métabolique de lysosomes a été étudié par spectrométrie de masse dans des souris WT ou de souris où le gène CLN3 était déficient. Les lysosomes des cellules déficientes contenaient moins de produits de la protéolyse, ce qui suggérait que CLN3 était important pour la protéolyse lysosomale. Cela a été confirmé par des mesures plus directes sur des neurones et des fibroblastes primaires, et sur des fibroblastes immortalisés. Ces résultats pourraient aider à comprendre les premières étapes de la physiopathologie dans les cellules où des gènes CLN sont déficients.Pour accroître le nombre de transporteurs lysosomaux potentiels, j’ai participé à la finalisation d’une étude par protéomique de la membrane lysosomale. Elle a révélé 46 potentiels transporteurs de fonction encore inconnue. Dans cette liste, j’ai étudié TMEM104, SPINSTER, MFSD1, SLC37A2, TTYH3 et SNAT7. Pour ce faire, j’ai d’abord muté les motifs d’adressage de ces protéines pour les rediriger, lors de leur synthèse, vers la membrane plasmique, afin de faciliter leur étude. Aucune fonction claire n’a pu être identifiée par cette approche.SNAT7 appartenait cependant à une famille de transporteurs de glutamine, ce qui était suffisamment encourageant pour envisager d’autres approches. Sa fonction a ainsi été étudiée en développement un nouveau test indirect basé sur la détection d’une surcharge artificielle des lysosomes en acides aminés. Un test fonctionnel plus direct a ensuite été mis au point sur des fractions enrichies en lysosomes en utilisant des acides aminés radiomarqués. Ces deux tests ont montré que SNAT7 était un transporteur spécifique de l’asparagine et de la glutamine.J’ai enfin étudié l’hypothèse suggérant que SNAT7 pourrait jouer dans la nutrition de cellules cancéreuses. En effet, certaines utilisent la glutamine comme nutriment principal à la place du glucose ; mais les apports sanguins en glutamine, dans les tumeurs, sont parfois insuffisants. La glutamine est donc obtenue par macropinocytose de protéines extracellulaires et dégradation lysosomale de ces protéines, avant un recyclage vers le cytoplasme. J’ai montré qu’en l’absence de SNAT7, ce phénomène était bloqué. SNAT7 est donc une cible thérapeutique intéressante pour tenter de bloquer l’approvisionnement des cellules cancéreuses en glutamine<br>Within lysosomes, about sixty different hydrolases degrade macromolecules. This degradation is dependent on the acidity of the lysosomal lumen, which pH ranges between 4.5 and 5.0. The lysosomal pH is maintained by the v-ATPase, a proton pump. Lysosomal degradation generates catabolites, which can be recycled to cytosol by secondary active transporters: lysosomal transporters.The dysfunction of lysosomal proteins leads to lysosomal storage disorders (LSDs), rare inherited metabolic diseases characterised by accumulation of material inside lysosomes. Depending on the mutated gene, symptoms of LSDs vary greatly, although about half of LSD patients display some kind of neurodegenerative symptoms. Studying the physiopathology of LSDs has led to a good understanding of the function of lysosomal enzymes, but the knowledge of lysosomal transporters remain poor, since only a few LSDs has been shown to be linked with a mutation in a lysosomal transporter gene.I focused on two proteins which dysfunction causes a special type of LSDs: CLN3 and CLN7. Mutations in CLN3 and CLN7 cause neuronal ceroid lipofuscinoses (NCLs), a special type of LSD which has mostly neurodegenerative symptoms and which is characterized by the accumulation of a specific pigment inside lysosomes: lipofuscin. There are fourteen NCL genes, but CLN3 and CLN7 are the two only proteins of the family which are resident proteins of the lysosomal membrane, suggesting they might be transporters.Amino acids were screened as possible substrates for CLN7, but none could be shown to be transported. For CLN3, the content in metabolites of lysosomes from Cln3-deficient mice and from WT mice were compared by mass spectrometry, revealing a specific decrease in the amount of catabolites of proteins in lysosomes from Cln3-deficient mice. This suggested a lack of lysosomal proteolysis, which was checked in neurons, in primary fibroblasts and in immortalized fibroblasts. These results suggested that CLN proteins could take part to a metabolic pathway important for lysosomal proteolysis and, more generally, for neuronal health. These results could help improve the understanding of the early steps of NCL physiopathology.To extend the number of candidates for lysosomal transporters, I took part to the validation step of an extensive proteomic study of the lysosomal membrane, which revealed forty-six new candidates for lysosomal transporters. I studied in more details TMEM104, SPINSTER, MFSD1, SLC37A2, TTYH3 and SNAT7. Proteins were overexpressed in HeLa cells to check for lysosomal localization. Then, their putative sorting motifs were mutated to misroute their expression to plasma membrane and to enable their functional study. In the last decade, a tremendous gain in knowledge concerning the molecular events of apoptosis signaling and execution has been achieved. The aim of this thesis was to clarify the role of lysosomal membrane permeabilization and lysosomal proteases, cathepsins, in signaling for apoptosis. We identified cathepsin D as an important factor in staurosporine-induced human fibroblast cell death. After release to the cytosol, cathepsin D promoted mitochondrial release of cytochrome c by proteolytic activation of Bid. Cathepsin D-mediated cleavage of Bid generated two fragments with the apparent molecular mass of 15 and 19 kDa. By sequence analysis, three cathepsin D-specific cleavage sites, Phe24, Trp48, and Phe183, were identified. Moreover, we investigated the mechanism by which cathepsins escape the lysosomal compartment, and found that Bax is translocated from the cytosol to lysosomes upon staurosporine treatment. In agreement with these data, recombinant Bax triggered release of cathepsins from isolated rat liver lysosomes. Conceivably, the Bcl-2 family of proteins may govern release of pro-apoptotic factors from both lysosomes and mitochondria. The importance of lysosomal cathepsins in apoptosis signaling was studied also in oral squamous cell carcinoma cells following exposure to the redox-cycling drug naphthazarin or agonistic anti-Fas antibodies. Le cancer est une maladie multifactorielle définie par un développement rapide de cellules anormales. Les cellules malignes acquièrent des avantages compétitifs qui permettent une croissance et prolifération anormales, grâce à un large spectre de changements génétiques et épigénétiques conduisant à des changements majeurs dans les profils de transcriptome et protéome et ainsi des modifications dans des voies de signalisation, le trafic intracellulaire et le métabolisme. Des nombreuses voies cellulaires ont été étudiées dans le contexte du cancer, y compris la signalisation, la migration, la perte de la polarité cellulaire apico-basale et l'adhésion cellulaire, cependant très peu est connu sur les altérations au niveau des organelles. Cette thèse a comme objectif d'identifier des altérations dans les compartiments intracellulaires et d'étudier leurs corrélations avec la progression du cancer. Dans la culture cellulaire classique, l'étude systématique de l'organisation du positionnement relatif des organelles est difficile en raison des fortes hétérogénéités morphologiques des cellules. Pour contourner ce problème, nous avons utilisé l’innovante technique des micro-patrons combinée à des cartes de densité des organelles. Après une analyse systématique de différentes lignées cellulaires représentant différents grades du cancer de la vessie, nous avons identifié des changements dans le positionnement de plusieurs organelles. Le changement de position le plus important a été observé pour les lysosomes, dont la distribution était plus périphérique dans les cellules représentant des grades plus avancées du cancer de la vessie. Ceci suggère que le positionnement des lysosomes pourrait être potentiellement important dans la progression du cancer. Par conséquent, nous avons cherché à caractériser l'impact de l’altération des lysosomes sur le comportement des cellules transformées. Nous avons constaté que les changements dans le positionnement des lysosomes jouent un rôle dans l'invasion des cellules cancéreuses de la vessie. En effet, le transport antérograde des lysosomes est en corrélation avec l’invasion 3D, contrairement au transport rétrograde qui corrèle avec une diminution de l’invasion cellulaire. Malignant cells acquire competitive advantages for growth and proliferation through a big spectrum of genetic and epigenetic changes leading to major changes in the transcriptome and proteome profiles and thus to alterations in multiple signaling pathways, intracellular trafficking and metabolism. Although many cellular pathways have been studied in the context of cancer, including signaling, migration, loss of apical-basal cell-polarity and cell adhesion, little is known about cancer-related alterations on the sub-cellular, organelle level. This PhD thesis aimed to identify alterations in intracellular compartments and to study how these changes correlate with cancer progression. In classical culture, the systematic study on the organization and relative positioning of organelles is challenging because of the strong morphological cell-to-cell variations. To overcome this problem, we used innovative micro-patterning technique in combination with quantitative, probabilistic mapping of cell organelles. Using a systematic analysis of different cell lines representing different stages of bladder cancer, we identified several changes in the positioning of organelles. The most striking phenotype was revealed by lysosomes, whose distribution was more peripheral in cells representing higher grades of bladder cancer. This suggested that lysosome positioning could be potentially relevant in cancer progression. Therefore, we aimed to characterize the impact of lysosome alteration on cell behavior in transformed cells. We found that changes in lysosome positioning played a role on bladder cancer cell invasion. Indeed, anterograde transport of lysosomes correlate with 3D invasion behavior, contrary to retrograde transport that correlated with decreased cell invasion. In this study. I have investigated several aspects of endometrial morphology and function as they relate to endometrial bleeding using human endometrial biopsies. The main emphasis of this study was on endometrial lysosomes and their enzymes. Two models of endometrial bleeding were investigated. The first was a “stable” model involving postpartum breastfeeding women from days 35—210 postpartum. Under the influence of breastfeeding, endometrium was “stable” and breakthrough bleeding was rare. The second was the ''menorrhagic" model involving two groups of women. women suffering from ovulatory dysfunctional uterine bleeding (DUB) and users of copper-bearing intrauterine contraceptive devices (lUCD) in whom heavy menstrual bleeding was common.
Results showed that both lUCD treated endometrium and those from women with dysfunctional bleeding had higher numbers of lysosomes associated with higher lysosomal enzyme levels (acid phosphatase and N-acetyl-B—D-glucosaminidase) when compared with endometrium from women with entirely normal menstrual histories. An active lysosomal system appeared to contribute to menorrhagia in both users of IUCD and women with dysfunctional bleeding. However. this was not the only change present. Both groups showed other evidences of altered cellular functions. This was reflected in alterations of tissue DNA and protein content and morphological evidence of defective secretory function in lUCD-exposed endometrium (see Chapter 5. 7. 10 and 13). There must have been certain triggering mechanisms and other factors at play. Lysosomal disturbances appeared to be part of a common pathway leading to heavy menstrual bleeding.
Previous information available on postpartum endometrium was restricted to a small number of studies from two to three decades ago. In this study. I have examined postpartum endometrium in detail and demonstrated quantitative differences between postpartum and normal proliferative endometrium. The main differences were that postpartum endometrium from days 35—60 in breastfeeding women showed less tissue oedema. stromal cells were densely packed glands were few and glandular dimensions were small compared with normal. These were appearances of an inactive endometrium comparable with menstrual to early proliferative endometrium.
In the first two months postpartum. endometrial re-modelling and regeneration which took place early postpartum was still evident. This was characterised by an initial heavy leucocyte and plasma cell infiltration. abundant glandular mitochondria and endometrial lysosomes. Endometrial lysosomes participated in autophagy or heterophagy as part of normal tissue repair and regeneration. Despite abundant lysosomal activity, postpartum endometrium did not show elevated lysosomal enzyme levels and endometrial bleeding was rare.
After two months postpartum. endometrial repair was complete. Under the influence of breastfeeding, endometrial bleeding was rare and results of endometrial morphometry and ultrastructure were comparable with the mid—proliferative phase. This "stability“ would continue for long periods. a finding in accordance with past literature. The appearance of endometrial lysosomes was similar to those in the mid-proliferative phase. Lysosomal abundance present in the early postpartum period has subsided and lysosomal enzyme activity remained low.

La curcumine, principe actif de Curcuma Longa L. Présente des propriétés inhibitrices sur certains cancers. Nous avons disséqué les conséquences de la déstabilisation du réticulum endoplasmique (RE) et des lysosomes par 25 M de curcumine ce qui impliquent une rupture de l’homéostasie mitochondriale. La curcumine induit un stress du réticulum qui permet la libération de calcium depuis le RE vers la mitochondrie dont l’homéostasie est alors perturbée. Ces évènements sont aussi associés à une perméabilisation lysosomale avec activation de la caspase-8 par les cathepsines et les calpaïnes ce qui donne un clivage de Bid-FL and tBid. Il a été récemment suggéré que l’autophagie puisse jouer un rôle important dans le contexte de la thérapie anticancéreuse. Dans ce travail, une conversation croisée s’engage entre autophagie et apoptose dès lors que le compartiment mitochondrial est perturbé et, produit des anions superoxides et des hydropéroxides délétères. L’induction de l’autophagie est marquée par l’apparition de vacuoles autophagiques marquées à l’acridine orange et la monodansylcadavérine, après exposition à 25 M de curcumine, une dose à laquelle l’apoptose est proéminente. La conversion de LC3-I en LC3-II dans ces conditions est un argument supplémentaire prouvant une exacerbation de l’autophagie. Nous avons également observé que la production de ROS et la formation de vacuoles autophagiques par la curcumine est presque totalement bloqué par l’antioxydant; acétylcystéine et par le MitoQ10, antioxydant ciblé à la mitochondrie, mais aussi par le BAPTA-AM, un chélateur de calcium cytosolique. Cependant l’autophagie induite par la curcumine échoue à protéger les cellules Huh-7 de l’apoptose. De ce fait, par l’induction indirecte d’une production de radicaux libres oxygénés d’origine mitochondriale, la curcumine perturbe les mécanismes de survie cellulaire telle que l’autophagie et conduit un fort taux de mortalité cellulaire<br>Curcumin, a major active component of turmeric Curcuma longa, L. , has been shown to have inhibitory effects on cancers. In vitro studies suggest that curcumin inhibits cancer cell growth by activating apoptosis, but the mechanisms underlying the anticancer effects of curcumin are unclear. We have dissected the mechanistic consequences of endoplasmic reticulum (ER) and lysosomal destabilization involved in a mitochondrially associated apoptosis. Curcumin at 25 M induces an ER stress with calcium release which, in turn, destabilizes the mitochondrial compartment to induce apoptosis. These events are also associated with lysosomal membrane permeabilization and activation of caspase-8 mediated by activation of cathepsins and calpains which induce Bid cleavage. Recently, it has been suggested that enhanced autophagy may play an important role in cancer therapy. In this work, I show that a fine interplay is at work which involves early autophagy as soon as the mitochondria produce superoxide anions and hydrogen peroxide. Induction of autophagy, as shown by autophagic vacuole formation, was detected by acridine orange staining and monodansylcadaverine dye after exposure to curcumin at a concentration of 25 M at which only early events of apoptosis are detectable. Conversion of LC3-I to LC3-II, a marker of active autophagosome formation, was also detectable by Western blotting following curcumin treatment. We also observed that reactive oxygen species production and autophagic vacuole formation following curcumin treatment was almost completely blocked by N-acetylcystein or by the mitochondrial specific antioxidant MitoQ, but also by the mitochondrial calcium uniporter inhibitor ruthenium red and to a lesser extend by the calcium chelators, BAPTA-AM and EGTA-AM. Curcumin-induced autophagy failed to rescue the cell from death and the cells undergo apoptosis after a try for survival. Curcumin successfully induces oxidative stress in Huh-7 cells, perturbs important cell survival mechanisms, and thus achieves high degree of killing of these cancer cells

Nous avons identifié une nouvelle protéine, la Brain And Dendritic cell associated LAMP-like molecule (BAD-LAMP), un nouveau membre de la famille des Lysosomes Associated Membrane Proteins (LAMPs). Par contraste avec les autres LAMPs, l’expression de BAD-LAMP est retreinte au cerveau chez la souris et l’homme. Au cours du développement murin, BAD-LAMP apparait de manière post-natale dans des souspopulations neuronales de couches II/III et V du néocortex. Le début de son expression coïncide avec la synaptogénèse corticale. Dans les neurones corticaux, BAD-LAMP est localisé dans des regroupements vésiculaires définis, s’accumulant le long des neurites. L’assemblage de ces agrégats vésiculaires est lié à l’organisation des microdomaines de la membrane plasmique. Par contraste avec les autres membres de la famille des LAMPs, BADLAMP est endocyté, mais il n’est pas retrouvé dans les compartiments endosomaux/lysosomaux. Chez l’homme, l’expression de BAD-LAMP est également retrouvée dans les Cellules Dendritiques plasmacytoïdes (pDCs) qui expriment spécifiquement les Toll-like receptor (TLR)-7 et TLR-9, et sont spécialisées dans la sécrétion massive d’interférons de type I suite à une stimulation virale. La chaine invariante ii contribue a la specialisation des molecules de classe ii dans la presentation d'antigene exogene. L'expression de molecules de classe ii en presence ou en absence de ii apres transfection dans les cpag non professionnelles nous a permis d'evaluer l'influence de la chaine invariante dans les differentes etapes qui aboutissent a la formation des complexes classe ii-peptide. Nous avons ainsi montre que les molecules de classe ii sont transportees via l'appareil de golgi dans des compartiments intracellulaires de type lysosomial ou elles s'accumulent, et ce meme en l'absence de ii. Par ailleurs, nous avons analyse la stabilite des molecules de classe ii accumulees dans ces compartiments et montre que la presence de ii favorise la formation de dimeres alpha beta de classe ii sous une forme compacte, ces formes correspondent a des molecules de classe ii stabilisees par la presence d'un peptide. Ce modele experimental nous a egalement permis d'etudier l'influence de ii dans la presentation d'antigene endogene. Nous avons montre qu'une forme endogene du hel localisee dans la voie d'exocytose en amont du lieu de formation des complexes classe ii-peptides, est presentee par les molecules de classe ii dans des conditions identiques a celles requises pour la presentation du hel exogene. En corollaire, nous avons montre dans les fibroblastes l transfectes que le transport et la degradation des chaines ii murines dans les endosomes/lysosomes ne dependent pas de leur association aux molecules de classe ii

La n-glycosylation des protéines, modification post-traductionnelle fréquente, s'effectue dans la lumière du réticulum endoplasmique par le transfert en bloc d'un oligosaccharide le glc#3man#9glcnac#2 à partir d'un oligosaccharide-lipide le glc#3man#9glcnac#2-p-p-dol, sur une séquence consensus de la protéine. Dans une lignée dérivant d'hepatocarcinome humain (les cellules hepg2), la n-glycosylation est accompagnée de la libération de quinze à vingt pour cent d'oligosaccharides dont l'origine glc#3man#9glcnac#2-p-p-dol est montrée pour une majeure partie d'entre eux. Les oligosaccharides libérés dans la lumière du réticulum endoplasmique sont directement transportes dans le cytosol ou une endoglucosaminidase et une mannosidase cytosoliques les élaguent jusqu'à la formation d'un produit limite, le man#5glcnac#1 linéaire. Nous avons étudié le devenir des oligosaccharides libres cytosoliques sur la lignée hepg2. Des expériences de pulse-chasse contrôles montrent la disparition des oligosaccharides libres après trois à quatre heures de chasse. Au contraire, la présence de la concanamycine à, qui inhibe les atpases vacuolaires stabilise les oligosaccharides libres dans les cellules. Ceux-ci retrouves dans les compartiments vésiculaires ont des structures compatibles avec une origine cytosolique, ce qui montre un transfert des oligosaccharides libres du cytosol vers un compartiment acide. Alors que le transfert n'est pas modifié par des inhibiteurs de la macro autophagie, il est affecté après l'inhibition de la mannosidase cytosolique et la réduction du niveau d'atp cellulaire, et indique que la dégradation partielle et une source d'énergie sont nécessaires pour assurer le transport. Nous avons montré par des expériences de fractionnement subcellulaire que les oligosaccharides libres cytosoliques et particulièrement le man#5glcnac#1 linéaire sont transportés vers les lysosomes. En conclusion, ces travaux et ceux réalisés précédemment au laboratoire présentent un nouveau trajet intracellulaire, du réticulum endoplasmique vers le cytosol et du cytosol vers les lysosomes.

The vesicle-mediated degradation of low-density lipoprotein (LDL) is an essential cellular function due to its role in cellular biosynthesis of membranes and steroids. Using multi-color single particle tracking fluorescence microscopy, the intracellular degradation of LDL was probed in live, intact cells. Unique to these experiments is the direct observation of LDL degradation using an LDL-based probe that increases fluorescence intensity upon degradation. Specifically, individual LDL particles were labeled with multiple fluorophores resulting in a quenched fluorescent signal. The characteristics of the vesicle responsible for degradation were determined and the vesicle dynamics involved in LDL degradation were quantified. Visualization of early endosomes, late endosomes and lysosomes was accomplished by fluorescently labeling vesicles with variants of GFP. Transient colocalization of LDL with specific vesicles and the intensity of the LDL particle were measured simultaneously. These studies, which are the first to directly observe the degradation of LDL within a cell, strive to completely describe the endo-lysosomal pathway and quantify the dynamics of LDL degradation in cells.

Les maladies lysosomales sont des maladies hereditaires de surcharge liees au deficit de l'activite de certaines hydrolases. Elles se presentent sous des formes cliniques de gravite tres plupart des genes des enzymes lysosomales permet une nouvelle approche de la physiopathologie de ces affections par une etude des relations entre le genotype et le phenotype clinique oubiochimique. Trois lipidoses lysosomales ont ainsi ete abordees : la maladie de sandhoff (gm2-gangliosidose due au defaut de la chaine d ela -hexosaminidase), la maladie de fabry (lipidose a trihexosyl-ceramide, due au deficit en -galactosidase a) et le cesd o polycorie cholesterolique de l'adulte (surcharge en ester de cholesterol par deficit de la lipase acide). Les defauts moleculaires de la maladie de sandhoff ont ete etudies au travers d'une famille consanguine, facilitant ainsi l'etude de larelation genotype-phenotype. La transition 1514g a (arg 505 gln) sur le gene de la chaine de la -hexosaminidase rend l'enzyme thermolabile et donne une forme adulte de la maladie de sandhoff. Une autre transition, 619ag (ile 207 val), a egalement ete mise en evidence sur les deux alleles des patients et d'un des parents. Deja decrite et consideree comme une mutation, cette transition apparait en fait n'etre qu'un polymorphisme relativement frequent. Nous avons decrit deux jumelles monozygotes, heterozygotes pour la maladie de fabry, pathologie liee au chromosome x. Celles-ci presentaient sur le gene de l'-galactosidase a une mutation pontuelle 10182ga (asp 231 asn). Une jumelle etait atteinte, l'autre etait asymptomatique. L'etude du statut d'inactivation du chromosome x chez les deux soeurs a montre un profil d'inactivation en miroir en accord avec la discordance des phenotypes. Les defauts moleculaires du gene codant pour la lipase acide lysosomale (lipa) ont ete etudies chez deux patients presentant un cesd. Une nouvelle mutation 233ct qui introduit un codon stop premature est decrite. L'etude in situ de l'activite residuelle de la lipase acide effectuee en parallele a montre qu'il n'existait pas de correlation directe simple entre les lesions moleculaires du gene lipa, les phenotypes biochimiques et les phenotypes cliniques.

L’ezrine appartient à la famille des protéines ERM. Ces protéines assurent la liaison entre la membrane plasmique et le cytosquelette d’actine. L’ezrine est impliquée dans le trafic de protéines membranaires, cependant sa fonction dans ce processus n’est pas connue à ce jour. Des cribles doubles hybrides utilisant l’ezrine comme appât ont permis l’identification de protéines impliquées dans le transport de protéines membranaires. Nous avons étudié la fonction de l’interaction de l’ezrine avec la sous unité Vps11 du complex HOPS et WWP1, une ubiquitine ligase de type E3. Le complexe HOPS orchestre la fusion de compartiments intracellulaires car il coordonne les activités des protéines Rab et SNARE. Nous avons étudié comment l’interaction de l’ezrine avec Vps11 régule le transport du récepteur de l’EGF vers la voie de dégradation par les lysosomes. Nous avons montré que cette interaction promeut la maturation des endosomes et qu’elle est nécessaire au transport du récepteur à l’EGF des endosomes précoces aux tardifs ainsi participant à la régulation de la dégradation du récepteur à l’EGF. WWP1 a été impliquée dans la régulation et la dégradation des récepteurs aux facteurs de croissance ainsi que des facteurs de transcription. Nous avons montré que l’interaction entre l’ezrine et WWP1 induit l’ubiquitylation de l’ezrine sans toutefois entraîner sa dégradation. Nous avons montré que l’interaction de l’ezrine avec WWP1 stabilise le récepteur c-Met. Nos résultats indiquent que l’ezrine participe au contrôle de la dégradation des récepteurs de type tyrosine kinase en régulant l’assemblage de complexes multiprotéiques impliqués dans le transport et l’ubiquitylation des protéines<br>Ezrin is a member of the ERM protein family, which provides a regulated linkage between the plasma membrane and the actin cytoskeleton. Ezrin has been involved in the trafficking of membrane proteins however its function in this process is as of yet unknown. Two-hybrid screens performed with ezrin as baits led to the identification of proteins involved in membrane protein transport. We analyzed the function of ezrin association with two new interactors: the HOPS complex subunit Vps11 and the E3 ubiquitin ligase WWP1. Vps11 is a member of the tethering HOPS complex that coordinates Rab and SNARE functions during vesicular fusion along the endocytic pathway. We have investigated the role of ezrin/Vps11 interaction in the endocytosis of the EGF receptor. We have shown that the interaction between ezrin and the HOPS complex promotes endosome maturation and is necessary for EGF receptor transport from early to late endosomes, therefore participating in the regulation of EGFR endocytosis and degradation. WWP1 is an E3 ubiquitin ligase of the Nedd4 family. A role for WWP1 was identified in the regulation and degradation of growth factors receptors and transcription factors. We have shown that Ezrin/WWP1 interaction results in ezrin ubiquitylation, but is not involved in regulating ezrin degradation. Rather, we were able to show that this ubiquitylation likely mediates protein interaction and that the binding between ezrin and WWP1 promotes c-Met receptor stabilization. Altogether our results indicate that ezrin participates in the control of tyrosine kinase receptor degradation by regulating the assembly of multimeric complexes involved in protein trafficking and ubiquitylation

La sclérose latérale amyotrophique (SLA ou maladie de Charcot) est la troisième maladie neurodégénérative la plus répandue. La principale cause génétique de la SLA est une expansion de répétitions GGGGCC dans le gène C9ORF72, dont la protéine forme un complexe avec les protéines SMCR8 et WDR41. Afin de mieux comprendre ses fonctions moléculaires, résoudre sa structure était un objectif principal de ma thèse. En parallèle, nous avons découvert que C9ORF72 régule un mécanisme nouvellement décrit de biogenèse de nouveaux lysosomes nommé reformation autophagique des lysosomes (ALR). Ce processus a largement été investigué dans cette thèse afin de mieux comprendre sa régulation, notamment pour la régénération des lysosomes en conditions basales et de privation d’acides aminés. Mon travail révèle un nouveau partenaire du complexe C9ORF72 et une nouvelle fonction de ce complexe dans la biogenèse des lysosomes. Ces résultats pourraient ainsi expliquer le dysfonctionnement des lysosomes et la neurodégénérescence observés dans la SLA, ce qui pourrait ainsi ouvrir de nouvelles voies thérapeutiques pour cette maladie dévastatrice<br>Amyotrophic lateral sclerosis (ALS or Charcot disease) is the third most common neurodegenerative disease. The main genetic cause of ALS is an expansion of GGGGCC repeats in the C9ORF72 gene which protein forms a complex with the SMCR8 and WDR41 proteins. To better understand its molecular functions, solving its structure was a main goal of my thesis. In parallel, we discovered that C9ORF72 regulates a newly described mechanism of biogenesis of newly-formed lysosomes, called autophagic lysosome reformation (ALR). This process has been extensively investigated during my thesis, in order to better understand its regulation, particularly for the regeneration of lysosomes in basal conditions and amino acid deprivation. My work reveals a new partner of the C9ORF72 complex as a novel function in lysosome biogenesis. These results could thus explain the dysfunction of lysosomes and neurodegeneration observed in ALS, which open new therapeutic ways for this devastating disease

Chloracne is a hyperkeratotic acneform skin disease caused by exposure to 2,3,7,8-tetrachlorodibenzo–p-dioxin (TCDD), a potent agonist of the aryl-hydrocarbon receptor (AhR). Despite the known role of TCDD exposure in the pathogenesis of chloracne, the molecular mechanisms mediating the disease remain poorly defined. Using a previously optimised in vitro primary human keratinocyte epidermal equivalent model, we demonstrated the temporal regulation and development of a significantly reduced viable cell layer and compacted stratum corneum over 7 days with 10nM TCDD. These morphological changes were paralleled by cumulative AhR protein degradation and increased mRNA levels of CYP1A1. One of the key findings was a significant increase in the expression of the apoptotic marker, caspase-3 whilst there was no significant effect on Ki67 staining. Furthermore, TCDD treatment caused de-regulated epidermal differentiation as evidenced by increased mRNA expression but decreased protein expression of late differentiation markers. Treatment with TCDD also resulted in an induction of LC3 lipidation and endogenous LC3 protein expression as well as decreased P62 protein expression, well-established markers of autophagy induction. Interestingly, co-treatment of epidermal equivalents with TCDD and the lysosomal inhibitor, bafilomycin or cathepsin D inhibition (by pepstatin A or shRNA knockdown) resulted in restoration of the viable cell layer and reduction in TCDD-induced caspase-3 expression. Similar results were also seen after blockade of the autophagy pathway by ATG7 knockdown. Results also demonstrated lysosomal function and autophagy are required for TCDD-induced AhR degradation, indicating a potential role for chaperone-mediated autophagy. Collectively these data suggest exposure to TCDD results in deregulated epidermal differentiation, induction of autophagy, reduction of a viable cell layer and caspase-3 dependent cell death, likely mediated via lysosomal processing. Results provide an insight into the pathophysiology of chloracne, and demonstrate novel findings including TCDD-induced autophagy and potential role of lysosomal function in TCDD-induced death and AhR degradation.

Bien qu'essentielle à la destruction des agents infectieux par les phagocytes, l'exocytose des lysosomes demeure un événement biologique partiellement caractérisé. L'activation de l'isoforme lysosomale de la tyrosine kinase de la famille Src, p61Hck (Hematopoietic cell kinase), est concomitante à la sécrétion de ces organelles et induit des réarrangements importants du cytosquelette d‘actine. Hck interagit par ailleurs avec des protéines induisant la polymérisation de l'actine. Mon travail de thèse a permis d'établir la capacité de Hck à induire la polymérisation de l'actine de novo, qui, dans le contexte cellulaire, s'organise en comète à la surface des lysosomes associés à Hck. La reconstitution de ce processus in vitro a permis de démontrer que Hck est capable d'induire la biogénèse de ces structures indépendamment d'autres protéines lysosomales. Dans ce contexte, la mise en place des comètes d'actine dépend de l'activité kinase de Hck, de WASp, du complexe Arp2/3, de Cdc42 et de Rho-GDI. Dans la cellule, la biogénèse de comète d'actine induite par l'activation de Hck a été corrélée à une accélération de 35% des lysosomes-p61Hck, qui dépend de la polymérisation de l'actine de novo et de l'intégrité du cytosquelette de microtubules. Ces résultats ont ainsi permis d'identifier p61Hck comme la première protéine lysosomale capable de recruter la machinerie moléculaire responsable de la biogénèse de comète d'actine. L'ensemble de ces données suggère également un nouveau mécanisme de motilité des lysosomes qui implique p61Hck, la formation de comètes d'actine et le cytosquelette de microtubules<br>While essential for the destruction of pathogens by phagocytes, the exocytosis of lysosomes remains a partially characterized biological event. The activation of the lysosomal isoform of the Src-tyrosine kinase p61Hck (Hematopoietic cell kinase), is concomitant to the secretion of these organelles and induces important remodelling of the actin cytoskeleton. Moreover, Hck interacts with proteins that are able to induce actin polymerization. The work I performed demonstrated the ability of Hck to induce de novo actin polymerization. In the cellular context, this process triggered the biogenesis of actin-comet tails at the surface of lysosomes associated with Hck. The in vitro reconstitution of this process indicated that other lysosomal proteins were dispensable in this mechanism. In this context, the actin-comet tails biogenesis was dependant on the kinase activity of Hck, WASp, the Arp2/3 complex, Cdc42 and Rho-GDI. This actin-comet biogenesis was correlated to a 35%-acceleration of p61Hck-lysosomes in cells, that was dependent on de novo actin polymerization and required an intact microtubular network. These results establish p61Hck as the first lysosomal protein able to recruit the molecular machinery responsible for actin tail formation. Altogether, these data suggest a new mechanism for lysosome motility involving p61Hck, actin-comet tail biogenesis and the microtubule network

We investigated whether human lysosomal hydrolases, in common with secretory and plasma membrane glycoproteins, associate with the ER chaperone calnexin. Neither $ alpha$- or $ beta$-chains of $ beta$-hexosaminidase A, cathepsin D, nor the endogenous proteases cathepsins B or L associated with calnexin in COS-I cells. Hex $ alpha$-chains misfolded due to either the incorporation of azetidine-2-carboxylic acid, treatment with dithiothreitol, or the presence of a Tay-Sachs Disease mutation (leading to retention of Hex A $ alpha$-chains in the ER) also did not associate with calnexin. Chemical-crosslinking reagents or long-term labeling also failed to show a Hex A $ alpha$-chain association with calnexin. Lysosomal hydrolases also did not associate with the ER chaperone calreticulin. Surprisingly, $ alpha$-L-iduronidase and Hex A $ alpha$-chains associated with calnexin when overexpressed using a CMV promoter. The segregation of lysosomal hydrolases from secretory proteins thus occurs at an earlier stage than predicted. Hydrolase folding appears to be controlled by a pathway different from that used by secretory and plasma membrane glycoproteins.

Un modèle de dégénérescence rétinienne induite par la lumière (dril) a montré une activation de l'autophagie et des calpaïnes (entre autres) dans les cellules de l'épithélium pigmentaire de la rétine (epr) et dans les photorécepteurs. Le but de ma thèse a été d'étudier la réponse de l'epr face au stress et d'expliquer le rôle des calpaïnes dans la mort des photorécepteurs. Concernant les cellules de l’epr , nous avons montré, dans un modèle de stress sur des cellules en culture , qu’il y a une activation de l'autophagie ; ceci a un effet protecteur pendant les 2 premières heures puis il bascule vers une mort des cellules par autophagie. Hormis l’importance de ce résultat pour la comprehension de la physiologie de l’epr , c'est la première fois qu'on démontre qu'un même stimulus peut faire basculer la réponse protectrice de l'autophagie vers un mécanisme de mort. Concenrnant les calpaïnes, nous avons montré que la calpaïne 1 perméabilise les lysosomes par la dégradation de la lysosomal associated membrane protein 2. Ce clivage est également observé dans la dril. La calpaïne 2, elle, est capable d'activer une endonucléase acide calpaïno-dependante qui pourrait être impliquée dans la dégradation de l'adn des cellules mourantes. Il serait important d'étudier l'effet de l'inhibition de la calpaïne 1 dans le modèle de dril, ainsi que dans d'autres modeles de mort neuronale. La caractérisation complète et implication de l'endonucléase dependante de la calpaïne pourrait apporter plus d'informations sur les mécanismes de mort cellulaiure independnate des caspases très presents dans la rétine<br>In a light induced retinal degeneration model (lird), studied in our laboratory the activation of autophagy and calpaîns have been shown : The aim of my work was to study the response of rpe during stress and to explain the role of calpains in photorecptors death. We have shown that metabolic sterss of rpe cells, in culture, induce autophagy activation. During the first 2 hours; this autophagy protects cells, but, afertwards the same mechanism triggers cell death. We have also shown that calpain 1 permeabilises lysosomes by degradation of the lysosomal associated protein 2. The same protein cleavage has been observed in lird. Moreover, calpain 2 can activate an acidic calpain dependent endonuclease that could be implicated in dna degradation of dying cells. It would be interesting to study the effects of calpain 1 inhibition during lird, as well as in other neural cell death models. The complete caracterisation of the calpain dependent endonuclease could provide more information on caspase-independent cell death mechanisms, highly involved in retinal degeneration

L'uranium étant in fine accumulé au sein des lysosomes des cellules tubulaires proximales, il est probable que l'endocytose soit le processus de transport de cet élément. Nous avons cherché à déterminer, lors de cette étude, quelle pouvait être son implication vis-à-vis de l'absorption de l'actinide et de la toxicité induite. La caractérisation des paramètres de l'absorption de l'uranium par les cellules LLC-PK1, d'origine tubulaire proximale, montre que le système principal de transport est saturable (Vm = 16. 95 ± 0. 92 PMOL/mg prot. /h), l'affinité apparente étant de 1. 105 ± 0. 175 umol/l. De manière similaire au processus endocytique clathrine-dépendant, ce système de transport est un processus actif, dépendant de l'intégrité du cytosquelette et de l'état d'activation de PKC. L'absorption de l'uranium est ainsi respectivement réduite 85. 9, 35. 9, 56. 8 et 46. 6 % lorsque les cellules sont incubées à 4°C pendant 1 heure ou pré-traitées avec 1 umol/l de colchicine, 0. 5 umol/l de cytochalasine D et 10 nmol/l de PMA. Il est également inhibé de 45. 1 % par 10 umol/l de chlorpromazine et de 83. 9 % lorsque les études sont effectuées en milieu hypertonique. Cette inhibition s'accompagne cependant d'une potentialisation de la cytotoxicité de l'actinide. Nos travaux montrent que cet effet résulte de stimulation du transport sodium-dépendant du phosphate (NaPi), et réciproquement, que son inhibition supprime la toxicité de l'uranium. Ces observations sont confirmées lorsque les études sont conduites sur le modèle cellulaire MDCK, d'origine tubulaire distale, transfecté avec l'ADNc codant pour le co-transporteur rNaPi-2. La sur-expression du co-transporteur stimule l'activité de transport du phosphate d'un facteur 4 et la cytotoxicité de l'uranium d'un facteur 3. Nous suggérons donc que l'endocytose clathrine-dépendante est le mécanisme cellulaire principal de l'absorption de l'uranium, et que les co-transporteurs NaPi transportent la fraction cytotoxique de l'actinide.
